Abstract

We are an international collective of Early Career Academics (ECAs) who met throughout 2020 to explore the implications of COVID-19 on precarious academics. With this intervention, our aims are to voice commonly shared experiences and concerns and to reflect on the extent to which the pandemic offers opportunities to redefine Higher Education and research institutions, in a context of ongoing precarity and funding cuts. Abstract

Hydrogels are powerful materials that more accurately mimic the cellular microenvironment over static two-dimensional culture. Photochemical strategies enable dynamic complexity to be achieved within hydrogels to better mimic the extracellular matrix; however, many photochemical systems to pattern proteins within hydrogels are complicated by long reaction times to immobilize these proteins wherein the protein can lose activity. As proof-of-concept, we demonstrate an elegant method where photocaged proteins are immobilized in hydrogels and then directly photoactivated. Specifically, we immobilized streptavidin-ortho-nitrobenzyl-modified epidermal growth factor (EGF) to cross-linked hyaluronan hydrogels and cultured two EGF-responsive cancer cells of breast and lung therein. We used light to temporally uncage and control EGF activation, thereby inducing cell death in breast cancer cells and proliferation in lung cancer cells. Abstract

Tropical cyclones undergo extratropical transition (ET) in every ocean basin. Projected changes in ET frequency under climate change are uncertain and differ between basins, so multimodel studies are required to establish confidence. We used a feature-tracking algorithm to identify tropical cyclones and performed cyclone phase-space analysis to identify ET in an ensemble of atmosphere-only and fully coupled global model simulations, run at various resolutions under historical (1950–2014) and future (2015–50) forcing. Historical simulations were evaluated against five reanalyses for 1979–2018. Considering ET globally, ensemble-mean biases in track and genesis densities are reduced in the North Atlantic and western North Pacific when horizontal resolution is increased from ∼100 to ∼25 km. At high resolution, multi-reanalysis-mean climatological ET frequencies across most ocean basins as well as basins’ seasonal cycles are reproduced better than in low-resolution models. Skill in simulating historical ET interannual variability in the North Atlantic and western North Pacific is ∼0.3, which is lower than for all tropical cyclones. Models project an increase in ET frequency in the North Atlantic and a decrease in the western North Pacific. We explain these opposing responses by secular change in ET seasonality and an increase in lower-tropospheric, pre-ET warm-core strength, both of which are largely unique to the North Atlantic. Multimodel consensus about climate change responses is clearer for frequency metrics than for intensity metrics. Abstract

Osteoporosis is a common bone disease that increases the risk of bone fracture. Hip-fracture risk screening methods based on flnite element analysis depend on segmented computed tomography (CT) images; however, current femur segmentation methods require manual delineations of large data sets. Here we propose a deep neural network for fully automated, accurate, and fast segmentation of the proximal femur from CT. Evaluation on a set of 1147 proximal femurs with ground truth segmentations demonstrates that our method is apt for hip-fracture risk screening, bringing us one step closer to a clinically viable option for screening at-risk patients for hip-fracture susceptibility. Abstract

Benzyl esters are cleaved upon reaction with Sn[Cl.sub.4], resulting in isolation of the corresponding carboxylic acid. Importantly, benzyl ethers, amines, and amides do not undergo debenzylation under these conditions, nor do a variety of other common protecting groups for alcohols, thereby rendering Sn[Cl.sub.4] selective amongst Lewis acids. The scope, tolerance, and limitations of the strategy are demonstrated through the analysis of several multifunctional substrates, including those bearing Cbz groups. The localised surface plasmon resonance (LSPR) peak of gold nanorods (GNRs) is located in the near-infrared optical window and is sensitive to local binding events, enabling label-free detection of biomarkers in complex biological fluids. Abstract

Simple Summary: Precision medicine is needed for muscle-invasive bladder cancer to improve survival rates due to high rates of tumour recurrence and poor patient outcomes. Hypoxia and an immunosuppressive tumour microenvironment are both independent poor prognostic factors that are targetable by hypoxia-modification and immune checkpoint inhibitors (ICIs), respectively. CD8 T cell counts are showing promise as a predictive biomarker for a durable response to ICIs. However, there is currently no biomarker to stratify patients to either hypoxia-targeting or immune-targeting therapies. Using data from a phase III trial where patients were randomised to radiotherapy +/− hypoxia-modifying therapy, we demonstrate that low CD8 T cell counts predict benefit from hypoxia-modifying therapy and associate with high hypoxia. These results inform the design of a clinical trial using CD8 T cell count as a biomarker for stratification to receive either radiotherapy with hypoxia-modifying therapy (low CD8 counts) or standard-of-care alone or with an ICI (high CD8 counts). Background: As hypoxia can drive an immunosuppressive tumour microenvironment and inhibit CD8+ T cells, we investigated if patients with low tumour CD8+ T cells benefitted from hypoxia-modifying therapy. Methods: BCON was a phase III trial that randomised patients with muscle-invasive bladder cancer (MIBC) to radiotherapy alone or with hypoxia-modifying carbogen plus nicotinamide (CON). Tissue microarrays of diagnostic biopsies from 116 BCON patients were stained using multiplex immunohistochemistry (IHC) with the markers CD8, CD4, FOXP3, CD68 and PD-L1, plus DAPI. Hypoxia was assessed using CA9 IHC (n = 111). Linked transcriptomic data (n = 80) identified molecular subtype. Relationships with overall survival (OS) were investigated using Cox proportional hazard models. Results: High (upper quartile) vs. low CD8 T cell counts associated with a better OS across the whole cohort at 16 years (n = 116; HR 0.47, 95% CI 0.28–0.78, p = 0.003) and also in the radiotherapy alone group (n = 61; HR 0.39, 95% CI 0.19–0.76, p = 0.005). Patients with low CD8+ T cells benefited from CON (n = 87; HR 0.63, 95% CI 0.4–1.0, p = 0.05), but those with high CD8 T cells did not (n = 27; p = 0.95). CA9 positive tumours had fewer CD8+ T cells (p = 0.03). Prognostic significance of low CD8+ T cells in the whole cohort remained after adjusting for clinicopathologic variables. Basal vs. luminal subtype had more CD8+ cells (p = 0.02) but was not prognostic (n = 80; p = 0.26). Exploratory analyses with other immune markers did not improve on findings obtained with CD8 counts. Conclusions: MIBC with low CD8+ T cell counts may benefit from hypoxia-modifying treatment.
