Abstract

A Mw 6.3 earthquake struck on April 6, 2009 the Abruzzi region (central Italy) producing vast damage in the L'Aquila town and surroundings. In this paper we present the location and geometry of the fault system as obtained by the analysis of main shock and aftershocks recorded by permanent and temporary networks. The distribution of aftershocks, 712 selected events with ML > 2.3 and 20 with ML > 4.0, defines a complex, 40 km long, NW trending extensional structure. The main shock fault segment extends for 15 -18 km and dips at 45° to the SW, between 10 and 2 km depth. The extent of aftershocks coincides with the surface trace of the Paganica fault, a poorly known normal fault that, after the event, has been quoted to accommodate the extension of the area. We observe a migration of seismicity to the north on an echelon fault that can rupture in future large earthquakes. Copyright 2009 by the American Geophysical Union.

Abstract

Human Cytomegalovirus (HCMV) can cause life threatening disease in infected hosts. Immunization with HLA-restricted immunodominant synthetic peptides and adoptive transfer of epitope specific T cells have been envisaged to generate or boost HCMV specific cellular immunity, thereby preventing HCMV infection or reactivation. However, induction or expansion of T cells effective against HCMV are limited by the need of utilizing peptides with defined HLA restrictions. We took advantage of a combination of seven predictive algorithms to identify immunogenic peptides of potential use in the prevention or treatment of HCMV infection or reactivation. Here we describe a pp65 derived peptide (pp65(340-355), RQYDPVAALFFFDIDL: RQY16-mer), characterized by peculiar features. First, RQY-16mer is able to stimulate HCMV pp65 specific responses in both CD4+ and CD8+ T cells, restricted by a wide range of HLA class I and class II determinants. Second, RQY-16mer is able to induce an unusually wide range of effector functions in CD4+ T cells, including proliferation, killing of autologous HCMV infected target cells and cytokine production. Third, and most importantly, the RQY-16mer is able to stimulate CD4+ and CD8+ T cell responses in pharmacologically immunosuppressed patients. These data suggest that a single reagent might qualify as synthetic immunogen for potentially large populations exposed to HCMV infection or reactivation.

110. Reservoir Structure and Wastewater‐Induced Seismicity at the Val d'Agri Oilfield (Italy) Shown by Three‐Dimensional Vpand Vp/VsLocal Earthquake Tomography

Abstract

Wastewater injection into a high‐rate well in the Val d'Agri oilfield, the largest in onshore Europe, has induced swarm microseismicity since the initiation of disposal in 2006. To investigate the reservoir structure and to track seismicity, we performed a high‐spatial resolution local earthquake tomography using 1,281 natural and induced earthquakes recorded by local networks. The properties of the carbonate reservoir (rock fracturing, pore fluid pressure) and inherited faults control the occurrence and spatiotemporal distribution of seismicity. A low‐Vp, high‐Vp/Vsregion under the well represents a fluid saturated fault zone ruptured by induced seismicity. High‐Vp, high‐Vp/Vsbumps match reservoir culminations indicating saturated liquid‐bearing zones, whereas a very low Vp, low Vp/Vsanomaly might represent a strongly fractured and depleted zone of the hydrocarbon reservoir characterized by significant fluid withdrawal. The comprehensive picture of the injection‐linked seismicity obtained by integrating reservoir‐scale tomography, high‐precision earthquake locations, and geophysical and injection data suggests that the driving mechanism is the channeling of pore pressure perturbations through a high permeable fault damage zone within the reservoir. The damage zone surrounds a Pliocene reverse fault optimally oriented in the current extensional stress field. The ruptured damage zone measures 2 km along strike and 3 km along dip and is confined between low permeability ductile formations. Injection pressure is the primary parameter controlling seismicity rate. Our study underlines that local earthquake tomography also using wastewater‐induced seismicity can give useful insights into the physical mechanism leading to these earthquakes. Abstract

The presence of amyloid fibrils is associated with a number of degenerative and serious human diseases. Nonetheless, the process of fibrillogenesis is still poorly understood. In this study, we present the results of a molecular dynamics investigation into the self-aggregation of the amyloidogenic peptide FLVHSS, a six-residue peptide belonging to the sequence of human islet amyloid polypeptide (hIAPP), capable of self-association to form amyloid-like fibrils. The simulations were performed at T = 300 K, using multiple replicas of FLVHSS monomer with random initial spatial distribution and orientation, in explicit water, set in a confined space volume (simulation box). Different simulations were carried out by varying initial atomic coordinates or initial atomic velocities and simulation box size. The formation of a unique aggregate (cluster) was observed within few nanoseconds from the beginning of all simulations. Structural analysis of the clusters clearly showed an elongated scalene ellipsoidal shape and a preferential parallel alignment of the peptides, roughly perpendicular to the main axis of the cluster and in extended conformation. The spatial arrangement of peptides in the simulated clusters appears to be similar to the spatial disposition of molecules in amyloid fibrils. An elongated scalene ellipsoidal shape was observed also in the final aggregate of the cluster growth simulation we also reported herein, indicating a tendency of cluster to grow along its major geometrical axis. © 2011 Wiley Periodicals, Inc. 149. The role of myocardial viability in deriving benefit from reestablishing infarct-related artery flow after acute myocardial infarction

Abstract

Early, sustained patency of the infarct-related artery (IRA) induces myocardial salvage, which preserves left ventricular (LV) function and mediates better long-term outcome. However, the time course and the mechanisms of muscle recovery after myocardial infarction are not completely understood. A large body of evidence suggests that most of the improvement occurs during the hospital phase and is related to early and sustained thrombolysis in myocardial infarction 3 flow in the IRA. Nevertheless, the relationship between IRA status and regional and global LV mechanics in the chronic phase of the disease remains controversial. Some late recovery may occur, either spontaneously or after revascularization, even in the absence of documented myocardial ischemia. The interplay between vessel patency, coronary flow grade and severity of the residual stenosis, and the presence of stunned or hibernating myocardium in the area at jeopardy may explain this delayed improvement. Although there seems to be a limited time window in which myocardium can be salvaged, timely testing for viability, particularly in patients with poor LV function, is justified even in a later phase of the disease to challenge potential cardiac recovery.
