Abstract

International audience; Organ or cell transplantation is the only therapeutic solution for pathologies causing an irreversible loss of vital organs function. The development of novel specific and non-toxic ani-rejection immunotherapies is a major goal in transplantation. Strategies based on Tregs are promising. However, Tregs cellbased therapies have been hampered by the technical limitation of obtaining large batches of functional Tregs. The project aim is to obtain an unlimited number of Tregs from human pluripotent stem cells. This very innovative project will also allow us to beter understand the development biology of Tregs through their in vitro differentiation.

Abstract

International audience; CD8+ Tregs have been long described and significant progresses have been made about their phenotype, their functional mechanisms, and their suppressive ability compared to conventional CD4+ Tregs. They are now at the dawn of their clinical use. In this review, we will summarize their phenotypic characteristics, their mechanisms of action, the similarities, differences and synergies between CD8+ and CD4+ Tregs, and we will discuss the biology, development and induction of CD8+ Tregs, their manufacturing for clinical use, considering open questions/uncertainties and future technically accessible improvements notably through genetic modifications.

Abstract

Humanized immune system immunodeficient mice have been extremely useful for the in vivo analyses of immune responses in a variety of models, including organ transplantation and graft versus host disease (GVHD) but they have limitations. Rat models are interesting complementary alternatives presenting advantages over mice, such as their size and their active complement compartment. Immunodeficient rats have been generated but human immune responses have not yet been described. Methods. We generated immunodeficient Rat Rag−/− Gamma chain−/− human signal regulatory protein alpha-positive (RRGS) rats combining Rag1 and Il2rg deficiency with the expression of human signal regulatory protein alpha, a negative regulator of macrophage phagocytosis allowing repression of rat macrophages by human CD47-positive cells. We then immune humanized RRGS animals with human peripheral blood mononuclear cells (hPBMCs) to set up a human acute GVHD model. Treatment of GVHD was done with a new porcine antihuman lymphocyte serum active through complement-dependent cytotoxicity. We also established a tumor xenograft rejection model in these hPBMCs immune system RRGS animals by subcutaneous implantation of a human tumor cell line. Results. RRGS animals receiving hPBMCs showed robust and reproducible reconstitution, mainly by T and B cells. A dose-dependent acute GVHD process was observed with progressive weight loss, tissue damage, and death censoring. Antihuman lymphocyte serum (L1S1) antibody completely prevented acute GVHD. In the human tumor xenograft model, detectable tumors were rejected upon hPBMCs injection. Conclusions. hPBMC can be implanted in RRGS animals and elicit acute GVHD or rejection of human tumor cells and these are useful models to test new immunotherapies.

Abstract

Myocardial infarction is a leading cause of morbidity and mortality worldwide. Although medical and surgical treatments can significantly improve patient outcomes, no treatment currently available is able to generate new contractile tissue or reverse ischemic myocardium. Driven by the recent understanding that regenerative processes do exist in the myocardium, the use of stem cells has emerged as a promising therapeutic approach with high expectations. The literature describes the use of cells from various sources. Many publications show the promising use of these cells to regenerate damaged myocardium in both animal models and Human; however, more studies are needed to directly compare cells of various origins in efforts to draw conclusions on the most appropriate source in order to positively impact on the heart tissue remodeling and function.

International audience; Modeling the immune response and rational immune approaches in vivo have relied heavily on studies performed in immunodeficient mouse models. These mouse models are groundbreaking platforms to humanize different tissues and organs and to evaluate compounds to treat a variety of human diseases, from cancer and infectious diseases to allergies, inflammation and GVHD. Rats are ideally suited to perform experiments in which larger size is needed and are still a small animal model suitable for rodent facilities.We describe here the generation of Il2rg-deficient rats and their crossing with previously described Rag1-deficient rats, generating for the first time double-mutant Rat Rag1 and Il2rG KO (RRG) animals.

Abstract

International audience; In this study, we phenotyped by flow cytometry and immunohistochemistry (data not shown) the immune cell subsets infiltrating Dmd mdx rat skeletal and cardiac muscles. Leukocyte infiltrates were absent or very low at 2 weeks of age, peaked at 4 and 8 weeks and decreased at 12 weeks. M2 macrophages represented >90% of infiltrating immune cells and Teff cells were the majority of the remaining ones. We also analyzed muscle enzymes and cytokines in sera. Creatin kinase was increased at weeks 4 and 8 and decreased at week 12 and thereafter (data not shown). This results are consistent with those observed in mdx mice model. Anti-CD45RC MAb treatment of young Dmd mdx rats normalized skeletal muscle strength associated to a depletion of effectors CD45RC high cells and no obvious side-effects. As a control prednisolone treatment of Dmd mdx rats similarly increased skeletal muscle strength and was also associated to a depletion of effectors CD45RC high cells but resulted in severe weight loss. Conclusion Duchenne Muscular Dystrophy (DMD) is a severe genetic muscle-wasting disorder due to the lack of dystrophin characterized by a progressive muscle weakness and a cardiomyopathy leading to premature death. The dystrophin-deficient Dmd mdx rats were generated using TALENs and offer a more reliable representation of human DMD, with marked muscle strength reduction, cardiomyopathy and muscle fibrosis that are higher that those observed in the mdx mouse model (1). A role for inflammation and autoimmune responses in muscle damages was shown both in DMD patients and the mdx mouse model (2).In this study, we assessed by flow cytometry and immunohistochemistry the immune cell subsets infiltrating Dmd mdx rat skeletal and cardiac muscles especially immunoregulatory and pro-inflammatory subsets (M1 and M2 macrophages, CD4 + and CD8 + Teff or Tregs…).Then, we investigated the possibility of reducing disease in Dmd mdx rats by administrating immunomodulatory treatments. The standard therapy for DMD patients is corticoids that decrease inflammation and immune responses but with variable responses, limited efficacy and important and numerous side effects. Therefore, there is need for new anti-inflammatory and pro-tolerogenic treatments that could replace or decrease the doses of corticoids. Anti-CD45RC monoclonal antibody (MAb) treatment has induced immune tolerance in models of organ transplantation and GVHD.

Abstract

International audience; Tregs play a critical role in the control of immune responses and tolerance induction; however our understanding of CD8+ Tregs is limited while they are particularly promising for therapeutic application. We previously reported that CD8+CD45RC[low] Tregs use IFNγ and IL-34 to suppress donor responses in a rat model of allotransplantation (Guillonneau et al, JCI, 2007; Bezie et al, JCI, 2015). Here, we aim to characterize human CD8+ Tregs to assess their potential for cell therapy in transplantation.
