Uplift in the broken Andean foreland of the Argentine Santa Bárbara System (SBS) is associated with the contractional reactivation of basement anisotropies, similar to those reported from the thick-skinned Cretaceous-Eocene Laramide province of North America. Fault scarps, deformed Quaternary deposits and landforms, disrupted drainage patterns, and medium-sized earthquakes within the SBS suggest that movement along these structures may be a recurring phenomenon, with yet to be defined repeat intervals and rupture lengths. In contrast to the Subandes thrust belt farther north, where eastward-migrating deformation has generated a well-defined thrust front, the SBS records spatiotemporally disparate deformation along structures that are only known to the first order. We present herein the results of geomorphic desktop analyses, structural field observations, and 2D electrical resistivity tomography and seismic-refraction tomography surveys and an interpretation of seismic reflection profiles across suspected fault scarps in the sedimentary basins adjacent to the Candelaria Range (CR) basement uplift, in the south-central part of the SBS. Our analysis in the CR piedmont areas reveals consistency between the results of near-surface electrical resistivity and seismic-refraction tomography surveys, the locations of prominent fault scarps, and structural geometries at greater depth imaged by seismic reflection data. We suggest that this deformation is driven by deep-seated blind thrusting beneath the CR and associated regional warping, while shortening involving Mesozoic and Cenozoic sedimentary strata in the adjacent basins was accommodated by layer-parallel folding and flexural-slip faults that cut through Quaternary landforms and deposits at the surface., Postprints der Universität Potsdam : Mathematisch-Naturwissenschaftliche Reihe, 1008

Protecting user data in distributed systems nowadays is very dicult. In this thesis, we focus on various issues related to the protection of user data on distributed data management systems, whose architectures range from client-server architectures to federations of servers or huge peer-to-peer organizations. Our first contribution is about trust in a system. We propose a metamodel based on first order logic, that allows to model a system considering entities of the social and digital worlds and their relations. Then we propose two approaches that allow users to evaluate trust in systems. In the context of peer-to-peer systems, we propose a data privacy model, and its DHT- based implementation, as well as a framework to measure and improve users’ satisfaction. In the context of the semantic web, in particular the Linked Data, we propose two approaches for deducing BGPs of SPARQL queries. Finally, we propose a lattice-based model that allows to position licenses in terms of compatibility and compliance.; Protéger les données des utilisateurs dans les systèmes distribués est aujourd’hui très difficile. Dans cette thèse, nous nous concentrons sur diverses questions liées à la protection des données utilisateur sur des systèmes de gestion de données distribuées, dont les architectures vont des architectures client-serveur aux fédérations de serveurs ou aux énormes organisations pair-à-pair. Notre première contribution concerne la confiance dans un système. Nous proposons un métamodèle basé sur la logique du premier ordre, qui permet de modéliser un système en considérant des entités des mondes sociaux et numériques et leurs relations. Nous proposons ensuite deux approches permettant aux utilisateurs d’évaluer la confiance envers les systèmes. Dans le contexte des systèmes pair-à-pair, nous proposons un modèle de confidentialité de données et son implémentation basée sur DHT, ainsi qu’un canevas permettant de mesurer et d’améliorer la satisfaction des utilisateurs. Dans le contexte du Web sémantique, en particulier des données liées, nous proposons deux approches pour déduire des BGP de requêtes SPARQL. Enfin, nous proposons un modèle basé sur treillis qui permet de positionner des licences en termes de compatibilité et de conformité.

Desde la función sustantiva de la proyección social, tal como se declara en sus lineamientos generales, la institución le apuesta a la generación de un nuevo conocimiento a partir de las experiencias que se dinamizan desde las diferentes estrategias que la configuran. Como es propio de UNIMINUTO, este conocimiento que se genera regresa al aula como una forma de nutrir el trabajo con los estudiantes y, por lo tanto, enriquece la docencia, pero también genera nuevas dinámicas en la interacción con las comunidades. Se trata de darle relevancia a la proyección social y de aprovechar cada escenario para dialogar con la docencia y la investigación desde el territorio, con el fin de enriquecerlas y ampliar las posibilidades formativas de los estudiantes, estableciendo una conversación permanente con la realidad para comprenderla en diálogo con la academia. Dicho conocimiento es reconocido y visibilizado en el ejercicio de la sistematización, concebida como una modalidad de investigación social. Todos estos elementos, vistos en conjunto, demuestran la posibilidad tangible de integrar las tres funciones sustantivas: la docencia, la investigación y, desde luego, la proyección social. De esta manera, UNIMINUTO y la Dirección General de Proyección Social apuestan por un ejercicio investigativo riguroso desde la sistematización de experiencias que dan cuenta de procesos de proyección en diversas partes del territorio nacional, con resultados y procesos que transforman la vida de las personas y las comunidades.

El 30 de noviembre de 2018 ocurrió un sismo al SE de la ciudad de Buenos Aires, Argentina. Fue percibido por gran parte de la poblaciónen la zona epicentral en el Cratón del Río de la Plata, considerado como una zona continental estable. Utilizando tiempos dearribos de ondas P y S en 12 trazas sísmicas registradas por estaciones sismológicas de Argentina, Uruguay y Brasil a distancias epicentralesentre 11 y 900 km, se determinó el hipocentro 34,9°S, 58,23°O y 19 km de profundidad. Se estimaron las magnitudes local(ML=3,9) y momento (MW=3,7). Se calculó el mecanismo focal obteniendo dos posibles planos de fallamiento de acimut δ=112°/234°,buzamiento α=68°/38º y ángulo del vector desplazamiento λ=122°/38°. Se analizaron 150 datos de percepción reportados por lapoblación para determinar intensidades sísmicas de Mercalli Modificada IMM en la zona epicentral, incluyendo máximos de IMM=IV.Nuestros resultados (δ=112°, α=68° y λ=122°) muestran buena correlación con estructuras orientadas ONO del sistema de fallas Paranáy con el patrón de la distribución de IMM en sentido NO-SE. Menor correlación se observa entre los parámetros hallados del otroposible plano de falla y estructuras de antiguos fallamientos orientadas en sentido SO-NE. Se determinó un modelo de velocidadessísmicas para el Cratón del Río de la Plata sobre la base de estudios previos. La localización sísmica se obtuvo considerando esemodelo y luego, se comparó con otras localizaciones utilizando otro modelo de velocidades sísmicas promedio de Argentina y condeterminaciones sísmicas globales para el hipocentro y magnitud. The earthquake on 30 November 2018 had an epicenter to the southeast of Buenos Aires city, Argentina. The event was felt by many people in the epicentral zone in the Río de la Plata Craton, which is considered as a stable continental zone. We determined the hypocenter at 34.9°S, 58.23°W and 19 km depth using P and S-wave arrival times of 12 seismic traces recorded by seismological stations of Argentina, Uruguay and Brazil, at epicentral distances between 11 and 900 km. The local (ML=3.9) and moment (Mw=3.7) magnitudes were also estimated. The calculated focal mechanism indicates two possible fault planes with azimuth δ=112°/234°, dip α=68°/38° and rake λ=122°/38°. 150 perception reports from the population were analyzed in order to determine Modified Mercalli seismic intensities (MMI) in the epicentral area, including maximum values of IV. Our results (δ =112°, α=68° and λ=122°) show a good correlation with the WNW trend of the Paraná Fault System structures and with the pattern of MMI of a NW-SE orientation. Less correlation between the parameters determined for the other possible fault plane and old faulting structures of SW-NE trend is found. Based on previous studies a seismic velocity model was determined for the Río de la Plata Craton. The seismic location was obtained considering this velocity model. Then, it was compared with other locations using another average seismic velocities model of Argentina and global seismic determinations for the hypocenter and magnitude. Resumen El propósito general de este artículo apunta a la recopilación, sistematización y el análisis crítico de la producción de conocimiento en torno a la categoría subjetividades políticas de paz en jóvenes de Colombia. Como primera medida, se llevó a cabo una labor heurística en donde se buscaron y compilaron las diversas fuentes de información, obtenidas principalmente a partir del rastreo de escritos, pertenecientes a las dos últimas décadas, que trataran categorías subyacentes a la mencionada, como consecuencia del mínimo tratamiento que esta ha tenido. Abstract The purpose of this article is to gater, systematize and analyze the production of knowledge around the category of political subjectivities of peace in young people from Colombia. First, a heuristic work was undertaken in which diverse sources of information were searched and gathered. This was obtained of tracking writings from the last two decades about underlying categories from the main one, since this sub ject has had a minimal treatment. Este trabajo es una contribución al conocimiento de la estructura cortical del sector oriental de las sierras de Valle Fértil-La Huerta (Sierras Pampeanas Occidentales, San Juan, Argentina) a los 31°S, en la región del antepaís del orógeno andino en el segmento donde la placa de Nazca subduce horizontalmente a ~100 km de profundidad. Se confeccionó un modelo de velocidades 1D, combinando estudios petrográficos y sismológicos a partir del análisis de 19 muestras de rocas ígneas y metaígneas plutónicas del arco magmático famatiniano (Ordovícico), que componen la mayor parte del basamento cristalino de dichas sierras. Las litologías granitoides predominan en la región norte, y las máficas en el sur. El análisis sismológico consistió en modelar funciones del receptor en los sitios donde afloran estas rocas a partir de telesismos que fueron registrados por tres estaciones sismológicas: LUNA, MAJA y CHUC. Así, fue posible obtener las velocidades de ondas P y S (Vp, Vs) y el coeficiente de Poisson (ν), entre otros parámetros elásticos. El modelo de velocidades sísmicas indica una corteza engrosada con un espesor promedio entre 55 y 60 km con respecto a promedios globales (~41 km); este promedio es consistente con la hipótesis de eclogitización parcial de la corteza inferior. La presencia de dos discontinuidades en las velocidades sísmicas intracorticales en niveles de corteza media (12 y 28 km de profundidad) podría asociarse a zonas de despegue (décollements), donde su extensión lateral estaría relacionada con la acreción del terreno Cuyania al terreno Pampia. Se han determinado valores bajos de velocidades de onda P (Vp ~5.8 km/s) y Vp/Vs (~1.70) para niveles de corteza superior, consistentes con litologías granitoides y velocidades de ondas P elevadas (Vp 6.76 km/s), Vp/Vs (~1.78) para niveles de corteza inferior, homologables con litologías máficas de mayor densidad (~3.00 g/cm3) con respecto a otras regiones del retroarco andino. Estos valores son consistentes con la existencia de rocas máficas compuestas por olivino, orto- y clinopiroxeno que constituyen la raíz del arco magmático famatiniano. Los resultados indican condiciones metamórficas de alto grado y profundidades correspondientes a propiedades geofísicas de niveles de corteza media a inferior, y se correlacionan con la hipótesis de un manto deshidratado, frío y enriquecido en magnesio que se encontraría en la zona comprendida entre la placa de Nazca subducida y la base de la corteza del terreno Cuyania. This paper is a contribution to the knowledge of the crustal structure of the eastern flank of the Valle Fértil - La Huerta ranges (Western Sierras Pampeanas, San Juan, Argentina) at 31°S, in the Andean foreland region, where the Nazca plate is subducting horizontally at about 100 km depth. A 1D velocity model was constrained, combining petrographic and seismological observations from analysis of 19 igneous and metaigneous plutonic rocks belonging to the Famatinian (Ordovician) magmatic arc, which make up most of the crystalline basement of these ranges. Granitoid lithologies predominate in the northern region whereas mafic lithologies are more common to the south. The seismological analysis consisted of modeling teleseismic receiver functions near three seismological stations: LUNA, MAJA and CHUC, in places where those rocks are dominant. Thus, P and S seismic-wave velocities (Vp and Vs) and Poisson´s coefficient (ν), among other elastic parameters, were obtained. The seismic velocity model indicates an overthickened crust with an average thickness between 55 and 60 km, which matches with global average values (~41km); this agrees well with the hypothesis of partial eclogitization in the lower crust. The presence of two seismic velocity discontinuities at mid-crustal levels (12 and 28 km depths), likely associated to décollements, might be related to the accretion of the Cuyania terrane to the Pampia terrane. We obtained low P seismic-wave velocities (Vp ~5.8 km/s), Vp/Vs ratio (~1.70) in upper crust levels consistent with granitoid lithologies, as well as high P seismic-wave velocities (Vp ~6.76 km/s), Vp/Vs ratio (~1.78) in lower crust levels; these figures match with mafic lithologies of a more dense (~3.00 g/cm3), lower crust with respect to other back-arc Andean regions. Also, these values are consistent with the existence of mafic rocks composed of olivine, ortho-and clinopyroxene, which constitute the root of the Famatinian magmatic arc. These results indicate high-grade metamorphic conditions and depths corresponding to geophysical properties of middle to lower crust and correlate with the hypothesis of a dehydrated, cool and magnesium-enriched mantle located in the region between the subducted Nazca slab and the bottom of the Cuyania terrain crust. Tectonic development of the Andean Cordillera has profoundly changed the topography, climate, and vegetation patterns of the southern central Andes. The Cenozoic Bermejo Basin in Argentina (~30°S) provides a key record of thrust belt kinematics and paleoclimate south of the high-elevation Puna Plateau. Ongoing debate regarding the timing of initiation of upper plate shortening and Andean uplift persists, precluding a thorough understanding of the earlier tectonic and climatic controls on basin evolution. We present new sedimentology, detrital geochronology, sandstone petrography, and subsidence analysis from the Bermejo Basin that reveal siliciclastic-evaporative fluvial and lacustrine environments prior to the main documented phase of Oligocene-Miocene shortening of the Frontal Cordillera and Argentine Precordillera. We report the first radiometric dates from detrital zircons collected in the Ciénaga del Río Huaco Formation, previously mapped as Permian, that constrain a Late Cretaceous (~95–93 Ma) maximum depositional age. Provenance and paleocurrent data from these strata indicate that detritus was derived from dissected arc and cratonic sources in the north and northeast. Detrital zircon U-Pb ages of ~37 Ma from the overlying red beds suggest that foredeep sedimentation began by at least the late Eocene. At this time, sediment dispersal shifted from axial southward to transversal eastward from the Andean Arc and Frontal Cordillera. Subsidence analysis of the basin fill is compatible with increasing tectonic deformation beginning in Eocene time, suggesting that a distal foredeep maintained fluvial connectivity to the hinterland during topographic uplift and unroofing of the Frontal Cordillera, prior to Oligocene-Miocene deformation across the Precordillera. Este artículo aborda las categorías intergeneracionalidad e interculturalidad como ejes de análisis de las experiencias y del dispositivo desarrollado por el Secretariado Nacional de Pastoral Social (SNPS), para propiciar la configuración de subjetividades políticas de paz y la transformación social. De igual forma, se explora el concepto de dispositivo que permite identificar los elementos de la propuesta metodológica de "Intervención para la transformación social" del SNPS. This article discusses the categories intergenerational and intercultural as axes of analysis of experiences and the device developed by the National Secretariat of Social Pastoral (SNPS), to promote political subjectivities configuration peace and social transformation. Similarly, the concept of device that identifies the elements of the methodology of "Intervention for social transformation" of SNPS is explored. Este artigo discute o intergeracional e intercultural categorias como eixos de análise de experiências eo dispositivo desenvolvido pela Secretaria Nacional de Pastoral Social (SNPS), para promover a paz configuração de subjetividades política e transformação social. Da mesma forma , o conceito de dispositivo que identifica os elementos da metodologia de "intervenção para a transformação social " de SNPS é explorado.

From public health and education to health teaching staff and others should incorporate their students to perform educational activities to cause in the student the ability to know how to observe and describe the observed in accordance with observation guides, fundamentals or rules, but also so the food consumer can observ e thoroughly and provide feedback to the food preparer, as a proposal to cause changes in attitudes and behaviors and reduce health risks in the population. It is important that both health professionals and the public in general are more aware of this sit uation. The aim of this study is to describe the critical risk points from the perception of students who study the subject of public health. Participant’s observations and critical analysis were performed with the perception of students in food vendor’s s tand with significant inflow of customers. There are critical risk points that denote risk to health; the feedback causes responses that could be meaningful to become aware in respect to attention processes to customers. The observation executed based on p revious knowledge that will be promoted to the consumer, will make possible to identify the areas of preventive or corrective opportunity for health risk and consumers with their feedback observation may favor the style and quality of life from changes in behavior and attitudes.

En este trabajo se obtiene un modelo cortical (1D) de velocidades sísmicas de ondas P y ondas S (de manera independiente) para la región comprendida entre 30°–32.5° S y 67°–68.5° O del retroarco andino, a unos 450 km al este de la trinchera. Para la determinación de este modelo de corteza se utilizaron los datos de 514 sismos locales de magnitudes 0.8 < ML < 4.8 ocurridos entre 1996 y 2014 reportados en el catálogo de la red de estaciones sísmicas permanentes del Instituto Nacional de Prevención Sísmica (INPRES) de Argentina. Para este conjunto de sismos se leyeron manualmente los tiempos de arribo de ondas P y S en 37 estaciones sismológicas. Así, fue posible realizar la localización de cada sismo considerando ponderaciones por calidad y distancia y un modelo de velocidades sísmicas obtenido por otros estudios. Posteriormente, se estudió la estructura de velocidades sísmicas mediante la técnica de inversión de tiempos de viaje. Este procedimiento iterativo tomó en cuenta varios modelos de variación de la velocidad con la profundidad; los mejores resultados se obtuvieron cuando se consideró un modelo inicial de aproximación a un gradiente cuyos valores de velocidades fueron menores que aquellos correspondientes al modelo disponible. Luego de efectuar 36 grupos de ensayos con siete inversiones en promedio para cada uno, se obtuvo el modelo de corteza final. El modelo de corteza obtenido indica discontinuidades en las velocidades sísmicas intracorticales a 3 km, 13 km, 27 km y 36 km de profundidad. Además, se observa la mayor discontinuidad en velocidades sísmicas a 47 km de profundidad en promedio, que se interpreta como la discontinuidad de Mohorovičić sobre la base de estudios previos y valores de velocidades representativos de regiones de corteza y manto superior. El nivel más superficial es indicativo de la presencia de cuencas sedimentarias que abarcan más del 70 % de la región estudiada. Las discontinuidades situadas a 13 km y 27 km muestran una buena correlación con niveles reconocidos de desacople de zonas de fallamiento mayor. Los resultados obtenidos para velocidades de ondas P (Vp) y velocidades de ondas S (Vs) se corresponden con valores bajos para la relación de Vp/Vs en la corteza inferior. Estas nuevas determinaciones geofísicas son consistentes con una corteza inferior de mayor incremento en Vs en comparación con el incremento que registra Vp y de ausencia de sismicidad. Estos resultados están de acuerdo con una composición máfica parcialmente eclogitizada. La alta relación Vp/Vs para los niveles superiores de la corteza serían indicativos de una mayor fracturación del terreno Cuyania que favorece una disminución en los valores de Vs observados y generación de sismicidad, en comparación con los niveles de corteza más profundos. Este modelo permite lograr el mejor ajuste de los tiempos de viaje de ondas P y S y, por lo tanto, mejorar la precisión de las localizaciones de sismos cuyos epicentros quedan comprendidos en la zona de estudio. Este estudio utilizando una densificación de arribos de fases P y S generados por sismos corticales muestra un basamento indistinguible para la región bajo Precordillera y Sierras Pampeanas Occidentales que conformarían el terreno Cuyania. In this study a crustal model (1D) of P and S wave seismic velocities is independently obtained for the region located between 30°–32.5° S and 67°–68.5° W of the Andean, at about 450 km east from the trench. To determine this crustal model 514 local earthquakes data were used. These earthquakes had local magnitudes between 0.8 < ML < 4.8 and had occurred between July 1996 and January 2014, and were reported by the permanent seismic network catalog of the Argentinean National Institute on Seismic Prevention (INPRES). For these earthquakes, the P and S wave arrival times were manually read in 37 seismic stations. Thus, it was possible to obtain the seismic location of each earthquake using weighing for quality and distance, and a seismic velocity model previously obtained by other studies.Then, the seismic velocity structure was studied in detail using a travel time inversion technique. The iterative method took into account several starting models with their velocity varying with depth; the best results occur for a starting model built from gradients in velocities whose values are smaller than those of an available crustal model. A final crustal seismic velocity model was obtained after testing 36 combinations of crustal parameters with seven average number of inversions each time. The obtained crustal model indicates intracrustal discontinuities in seismic velocities at depths of 3 km, 13 km, 27 km and 36 km. In addition, the greater discontinuity in seismic velocities is observed at 47 km depth approximately, which is interpreted as the Mohorovičić discontinuity on the base of previous studies and velocity values representative of crustal and upper mantle regions. The shallowest level is sensitive to sedimentary basins which contain more than 70% of the studied region. Discontinuities located at 13 km and 27 km depths show a good correlation with décollement levels shown by other geophysical studies. The results for wave velocities P (Vp) and for wave velocities S (Vs) agree with low Vp/Vs values in the lower crust. These new geophysical determinations are consistent with a lower crust of a higher increase in Vs in comparison with the lower increase observed in Vp, and a lack of seismicity. All together, these results agree with a mafic probably partially eclogitized lower crust. The high Vp/Vs ratio for the upper crustal levels would indicate a higher fracturation of the Cuyania terrane consistent with a decrease in Vs values and earthquake generation in comparison with deeper crustal levels. This best model enables to fit the both the P and S wave travel times, and thus allow improving accuracy of earthquake locations with epicenters within the region of study. Since this study uses a densification of arrival P and S wave phases from crustal earthquakes located under the eastern part of the Andean Precordillera and the Western Sierras Pampeanas, results represent an indistinguishable basement that corresponds to the integrated Cuyania terrane. Nuestro objetivo en este artículo es presentar un avance de la investigación Las condiciones y modos de Construcción de subjetividades políticas para la paz en jóvenes de organizaciones interétnicas y feministas desde una perspectiva intergeneracional. Presentamos de manera sintética la conceptualización respecto a las subjetividades, las subjetividades políticas y los conceptos de Paz. Adicionalmente, hacemos un análisis de las experiencias promovidas por el Secretariado Nacional de Pastoral Social, a la luz de estas categorías. This article aims to present a preview of the research project: Conditions and modes of construction of political subjectivities for peace in ethnic youth and women’ s organizations from an intergenerational perspective. The authors present a summary of the conceptualizations of subjectivities, political subjectivities and concepts of peace. In addition, the paper includes an analysis of the experiences promoted by the National Secretariat of Social Pastoral in the framework of these categories. El metotrexato (MTX) es el fármaco de elección en primera línea de tratamiento para pacientes con artritis reumatoide. En las últimas décadas ha habido un gran interés para identificar marcadores biológicos que puedan predecir con exactitud y fiabilidad la eficacia y toxicidad del MTX. Los polimorfismos (SNPs, Single Nucleotide Polymorphism) en los genes que participan de forma directa o indirecta en la vía metabólica del MTX y en su transporte y activación (poliglutamación) celular, pueden ser predictores de respuesta y toxicidad del MTX. El objetivo del trabajo es valorar la utilidad de diferentes marcadores farmacogenéticos en relación a la respuesta y/o toxicidad de pacientes con artritis reumatoide tratados con MTX en monoterapia. El trabajo se ha dividido en dos partes: La primera parte se basa en el estudio de 27 polimorfismos de genes que tienen que ver con el mecanismo de acción del MTX (DHFR, TYMS, MTHFR, ATIC y CCND1) y, la segunda, en el estudio de 34 polimorfismos de genes que participan en el transporte celular del MTX y en su activación e inactivación través de la poliglutamación (SLC19A1 / RFC1, ABCB1, FPGS y GGH). En relación a los genes que participan en el mecanismo de acción del MTX los resultados muestran una asociación entre dos SNPs en el gen MTHFR, rs17421511 y rs1476413, y uno en el gen DHFR, rs1643650, y la respuesta al tratamiento con MTX en pacientes con artritis reumatoide. Asimismo, también se ha encontrado que dos SNPs en el gen ATIC, rs16853826 y rs10197559, se asocian con toxicidad. En relación a los genes que participan en el transporte celular del MTX y en su activación e inactivación a través de la poliglutamación los resultados obtenidos en este trabajo muestran una asociación entre dos SNPs en el gen FPGS, rs10987742 y rs10106, y la respuesta al tratamiento con MTX. El SNP rs10106 también está asociado con la presencia de eventos adversos y el tiempo de supervivencia del fármaco. Los resultados también muestran que tres SNPs en el gen ABCB1, rs868755, rs10280623 y rs1858923, están asociados con la toxicidad al MTX, y que dos de los SNPs del mismo gen, rs10264990 y rs868755, también se asocian con el tiempo de supervivencia del MTX. En conclusión, determinadas variantes genéticas en los genes que participan en la vía metabólica del MTX y en su transporte y activación celular, podrían ser consideradas como marcadores farmacogenéticos de respuesta y toxicidad en pacientes con artritis reumatoide tratados con MTX en monoterapia., Methotrexate (MTX) is the first-line treatment option for rheumatoid arthritis (RA) patients. Recently, the interest of identifying biomarkers that can predict the efficacy and toxicity of MTX has been grown. Polymorphisms (SNPs, Single Nucleotide Polymorphism) in genes involved directly or indirectly in the metabolic pathway of MTX and in the transport and activation (polyglutamation) of MTX inside the cell can be predictors of response and toxicity of MTX. The objective of this study is to evaluate the usefulness of different pharmacogenetics markers in relation to the response and / or toxicity of rheumatoid arthritis patients treated with MTX in monotherapy. The investigation has been divided into two parts. The first part is based on the study of 27 polymorphisms of 5 genes that are involved with the mechanism of action of MTX (DHFR, TYMS, MTHFR, ATIC and CCND1). The second part is based on the study of 34 polymorphisms of 4 genes involved in the cellular transport of MTX and in its activation and inactivation through polyglutamation (SLC19A1 / RFC1, ABCB1, FPGS and GGH). Regarding genes involved in the mechanism of MTX’ action the results show an association between three SNPs and the response to treatment with MTX in patients with rheumatoid arthritis. The involved SNPs are rs17421511 and rs1476413 of MTHFR gene and the SNP rs1643650 of the DHFR gene. Besides, the two SNPs rs16853826 and rs10197559 of ATIC gene are associated with toxicity. Concerning genes involved in cellular transport of MTX and in their activation/inactivation through polyglutamation the results obtained in this study show an association between two SNPs and the treatment response of MTX. In this case, the involved SNPs are rs10987742 and rs10106 of FPGS gene. The SNP rs10106 is also associated with the presence of adverse events and the survival time of the drug. The results also show that three SNPs rs868755, rs10280623 and rs1858923 of ABCB1 gene are associated with MTX toxicity; and two SNPs rs868755 and rs10264990 of the same gene are also associated with the survival time of MTX. In conclusion, certain genetic variants in genes involved in the metabolic pathway of MTX and in its transportation and cellular activation could be considered as pharmacogenetics markers for response and toxicity in rheumatoid arthritis’ patients treated with MTX in monotherapy.
