The red panda (Ailurus fulgens fulgens) is classified as endangered due to its declining population, habitat fragmentation and poaching. Efforts are being made to breed them in captivity as part of nationwide conservation breeding program. This study aimed to standardize Enzyme immunoassays (EIAs) to monitor reproductive (Progesterone metabolite, Testosterone) and stress hormone (Cortisol) in red panda. For this purpose, we collected 1471 faecal samples from four females and one male over a period of one year from Padmaja Naidu Himalayan Zoological Park, Darjeeling, India. HPLC confirmed the presence of immunoreactive 5α-pregnan-3α-ol-20-one, testosterone and cortisol metabolites in faecal samples. Using 5α-pregnan-3α-ol-20-one EIA, we were able to monitor reproduction and detect pregnancy in one of the females, which successfully conceived and delivered during the study period. We were also able to monitor testosterone and cortisol in faecal samples of the red panda. Faecal testosterone levels were found in higher concentration in breeding season than in non-breeding season. Faecal cortisol concentrations showed a negative relationship with ambient temperature and peaked during winter months in all animals. Standardization of EIAs and faecal hormone monitoring would facilitate red panda conservation breeding programs in India and elsewhere.

We analyzed a concatenated (8492. bp) nuclear-mitochondrial DNA data set from 44 musteloids (including the first genetic data for Lyncodon patagonicus) with parsimony, maximum likelihood, and Bayesian methods of phylogenetic and biogeographic inference and two Bayesian methods of chronological inference. Here we show that Musteloidea emerged approximately 32.4-30.9 million years ago (MYA) in Asia, shortly after the greenhouse-icehouse global climate shift at the Eocene-Oligocene transition. During their Oligocene radiation, which proceeded wholly or mostly in Asia, musteloids diversified into four primary divisions: the Mephitidae lineage separated first, succeeded by Ailuridae and the divergence of the Procyonidae and Mustelidae lineages. Mustelidae arose approximately 16.1 MYA within the Mid-Miocene Climatic Optimum, and extensively diversified in the Miocene, mostly in Asia. The early offshoots of this radiation largely evolved into badger and marten ecological niches (Taxidiinae, Melinae, Mellivorinae, Guloninae, and Helictidinae), whereas the later divergences have adapted to other niches including those of weasels, polecats, minks, and otters (Mustelinae, Ictonychinae, and Lutrinae). Notably, and contrary to traditional beliefs, the morphological adaptations of badgers, martens, weasels, polecats, and minks each evolved independently more than once within Mustelidae. Ictonychinae (which is most closely related to Lutrinae) arose approximately 9.5-8.9 MYA, most likely in Asia, where it diverged into the Old World Ictonychini (Vormela, Poecilictis, Ictonyx, and Poecilogale) and New World Lyncodontini (Lyncodon and Galictis) lineages. Ictonychini presumably entered Africa during the Messinian Salinity Crisis (at the Miocene-Pliocene transition), which interposed the origins of this clade (approximately 6.5-6.0 MYA) and its African Poecilictis- Ictonyx- Poecilogale subclade (approximately 4.8-4.5 MYA). Lyncodontini originated approximately 2.9-2.6 MYA at the Pliocene-Pleistocene transition in South America, slightly after the emergence of the Panamanian land bridge that provided for the Great American Biotic Interchange. As the genera Martes and Ictonyx (as currently circumscribed) are paraphyletic with respect to the genera Gulo and Poecilogale, respectively, we propose that Pekaniaand Poecilictis be treated as valid genera and that Martes pennanti and Ictonyx libyca, respectively, be assigned to these genera. The monophyletic group Caniformia (dog-like carnivores) in the order Carnivora comprises 9 families. Except for the general consensus for the earliest divergence of Canidae and the grouping of Procyonidae and Mustelidae, conflict- ing phylogenetic hypotheses exist for the other caniformian families. In the present study, a data set comprising >22 kb of 22 nuclear intron loci from 16 caniformian species is used to investigate the phylogenetic utility of nuclear introns in resolving the interfamilial relationships of Caniformia. Our phylogenetic analyses support Ailuridae as the sister taxon to a clade containing Procyonidae and Mustelidae, with Mephitinae being the sister taxon to all of them. The unresolved placements of Ursidae and Pinnipeds here emphasize a need to add more data and include more taxa to resolve this prob- lem. The present study not only resolves some of the ambiguous relationships in Caniformia phylogeny but also shows that the noncoding nuclear markers can offer powerful complementary data for estimating the species tree. None of the newly developed introns here have previously been used for phylogeny reconstruction, thus increasing the spectrum of molecular markers available to mammalian systematics. Interestingly, all the newly developed intron data partitions ex- hibit intraindividual allele heterozygotes (IIAHs). There are 115 cases of IIAHs in total. The incorporation of IIAHs into phylogenetic analysis not only provides insights into the interfamilial relationships of Caniformia but also identifies two potential hybridization events occurred within Ursidae and Otariidae, respectively. Finally, the powers and pitfalls of phy- logenetics using nuclear introns as markers are discussed in the context of Caniformia phylogeny. The present record provides one of the highest documented presence information of red pandas in India, in remote parts of western Arunachal Pradesh. The record came in the form of carcass of a Red Panda which was accidentally caught in an animal snare in remote sub-alpine mountain slopes at 4325m above sea level inside a Community Conserved Area in Tawang District, discovered during a monitoring trip by the villagers. The record also showcases the rich biodiversity of the area and the local community’s efforts to safeguard it.

A 10-year-old male red panda presented acutely with symptoms of shock due to acute abdominal distress and respiratory compromise. Abdominal ultrasound confirmed a severely distended stomach for which passage of an orogastric tube for relief was unsuccessful. Intra-operatively, the stomach was found to be distended and torsed around its long axis supporting the diagnosis of Gastric dilitation-volvulus (GDV). The animal arrested and died intra-operatively and was submitted for necropsy with lesions supportive of the diagnosis of GDV. No risk factors for GDV were found to correlate between the panda and those described in domestic dogs. This case suggests that red pandas can be susceptible to this condition in captive settings.

Phylogenies underpin comparative biology as high-utility tools to test evolutionary and biogeographic hypotheses, inform on conservation strategies, and reveal the age and evolutionary histories of traits and lineages. As tools, most powerful are those phylogenies that contain all, or nearly all, of the taxa of a given group. Despite their obvious utility, such phylogenies, other than summary 'supertrees', are currently lacking for most mammalian orders, including the order Carnivora. Carnivora consists of about 270 extant species including most of the world's large terrestrial predators (e.g., the big cats, wolves, bears), as well as many of man's favorite wild (panda, cheetah, tiger) and domesticated animals (dog, cat). Distributed globally, carnivores are highly diverse ecologically, having occupied all major habitat types on the planet and being diverse in traits such as sociality, communication, body/brain size, and foraging ecology. Thus, numerous studies continue to address comparative questions within the order, highlighting the need for a detailed species-level phylogeny. Here we present a phylogeny of Carnivora that increases taxon sampling density from 28% in the most detailed primary-data study to date, to 82% containing 243 taxa (222 extant species, 17 subspecies). In addition to extant species, we sampled four extinct species: American cheetah, saber-toothed cat, cave bear and the giant short-faced bear. Bayesian analysis of cytochrome b sequences data-mined from GenBank results in a phylogenetic hypothesis that is largely congruent with prior studies based on fewer taxa but more characters. We find support for the monophyly of Carnivora, its major division into Caniformia and Feliformia, and for all but one family within the order. The only exception is the placement of the kinkajou outside Procyonidae, however, prior studies have already cast doubt on its family placement. In contrast, at the subfamily and genus level, our results indicate numerous problems with current classification. Our results also propose new, controversial hypotheses, such as the possible placement of the red panda (Ailuridae) sister to canids (Canidae). Our results confirm previous findings suggesting that the dog was domesticated from the Eurasian wolf (Canis lupus lupus) and are congruent with the Near East domestication of the cat. In sum, this study presents the most detailed species-level phylogeny of Carnivora to date and a much needed tool for comparative studies of carnivoran species. To demonstrate one such use, we perform a phylogenetic analysis of evolutionary distinctiveness (EDGE), which can be used to help establish conservation priorities. According with those criteria, and under one of the many possible sets of parameters, the highest priority Carnivora species for conservation of evolutionary diversity include: monk seals, giant and red panda, giant otter, otter civet, Owston's palm civet, sea otter, Liberian mongoose, spectacled bear, walrus, binturong, and the fossa.

The red panda (Ailurus fulgens) is an endangered carnivore living in the temperate forests of the Himalayas and southern China. The phylogeny of the red panda has been the subject of much debate. Morphological and molecular studies have supported a wide range of possible relationships, including close ties to procyonids, ursids, mustelids, and mephitids. This study provides additional morphological data, including muscle maps, for Ailurus. The hindlimbs of four cadavers from the National Zoological Park were dissected. Red pandas retain a number of muscles lost in other carnivore groups, including muscles and tendons related to their robust and weight-bearing hallux. Three features, including a single-bellied m. sartorius, a proximal insertion for m. abductor digiti V, and an absent m. articularis coxae, are found in all terrestrial arctoids, including Ailurus. In addition, red pandas are similar to ursids and canids in lacking a caudal belly of m. semitendinosus, while they resemble procyonids and mustelids in the degree of fusion observed between mm. gluteus medius and piriformis. Furthermore, Ailurus and procyonids are characterized by numerous subdivisions within the adductor compartment, while red pandas and raccoons share a variable m. semimembranosus, composed of one, two, or three bellies. Lastly, a deep plantar muscle inserting onto the metatarsophalangeal joint of the hallux is described for Ailurus. This muscle has not been previously described and is given the name m. flexor hallucis profundus. Additional dissections of the forelimb and axial musculature of red pandas may shed further light on the phylogeny of this species. In addition, the muscle maps presented here offer a valuable resource for interpreting the functional anatomy of fossil ailurids.

Although the modern lesser panda (Ailurus; Ailuridae; Carnivora; Mammalia) only lives in the temperate highland forests of the southern Himalayan region, its fossil remains of late Miocene to Pliocene age have been found widely across the Holoarctic region. Very few mandibles or lower teeth of these animals, however, have been identified. Here, we provide a detailed description of the mandible and lower dentition of a large-sized lesser panda, Parailurus baikalicus (Sotnikova, 2008), which has been discovered from the middle Pliocene of Udunga, Transbaikalia, Russia. In the Transbaikalian species, lower molars are low crowned and shows highly crenulated enamel pattern. The main cuspids of cheek teeth are heavily worn horizontally, suggesting a high specialization for leaf eating. The distinctive lower tooth morphology of P. baikalicus differs from those of other fossil lesser pandas discovered from Europe and North America and from those of modern species, suggesting that the morphology found in P. baikalicus is derived in the Ailuridae.

Carnivores from the Late Pliocene site of Vcelare 2 are described and their taxonomy is discussed. The carnivore assemblage includes nine specimens of the families Felidae (Homotherium crenatidens), Ursidae (Ursus minimus), and Ailuridae (Parailurus sp.). The scimitar-toothed cat and ursid are common representatives of the Pliocene European fauna, whereas the lesser panda is probably a new species of Parailurus, which differs from both Pliocene species, P. anglicus and P. hungaricus. As a part of a more or less uniform Pliocene carnivore fauna of the Northern Hemisphere, the Vcelare 2 assemblage represents a forest and (or) open grassland environment.

Organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), and brominated flame retardants (BFRs) were investigated in captive giant and red panda tissues from China. The total concentrations of OCPs, PCBs, and polybrominated diphenyl ethers (PBDEs) in tissues ranged from 16.3 to 888 ng/g lipid weight (lw), 24.8 to 854 ng/g lw, and 16.4 to 2158 ng/g lw, respectively. p,p'-DDE and beta-HCH were major OCP contaminants. PCBs 99, 118, 153/132, 170, 180, and 209 were the major contributing congeners determined. Among PBDEs, congener BDE-209 was the most frequent and abundant, followed by BDE-206, BDE-208, BDE-207, BDE-203, BDE-47, and BDE-153. Decabromodiphenyl ethane (DeBDethane) was detected in 87 and 71% of the giant and red panda samples with concentrations up to 863 ng/g lw, respectively. The remarkable levels and dominance of BDE-209 and DeBDethane may relate to significant production, usage, or disposal of BFRs in China. The positive significant correlation between concentrations of PBDEs and PCBs in captive pandas may suggest that the exposure routes of PBDEs and PCBs to panda are similar. To our knowledge, this is the first report of the occurrence of DeBDethane in captive wildlife samples. Therefore, further studies are warranted to better understand DeBDethane production, transport, uptake, and toxicological effect.

A new species of the lesser panda, Parailurus baikalicus sp. nov., from the Pliocene of Transbaikalia is described. In contrast to the European taxa P. anglicus and P. hungaricus, it retains a primitive occlusal pattern of M1-M2, with a concave buccal outline, small mesostyle on M1, and undeveloped styles on M2. At the same time, the Transbaikalian panda is more advanced than other representatives of Parailurus in the upper molars with a reduced lingual cingulum and an enlarged paraconule, which is partially (M1) or completely (M2) separated from the protocone. This combination of primitive and advanced characters points to the separation of the Asian branch at the earliest stages of the genus development. The Transbaikalian lesser panda may represent a terminal form of this phylogenetic lineage.

Extant members of the order Carnivora split into two basal groups, Caniformia (dog-like carnivorans) and Feliformia (cat-like carnivorans). In this study we address phylogenetic relationships within Caniformia applying various methodological approaches to analyses of complete mitochondrial genomes. Pinnipeds are currently well represented with respect to mitogenomic data and here we add seven mt genomes to the non-pinniped caniform collection. The analyses identified a basal caniform divergence between Cynoidea and Arctoidea. Arctoidea split into three primary groups, Ursidae (including the giant panda), Pinnipedia, and a branch, Musteloidea, which encompassed Ailuridae (red panda), Mephitidae (skunks), Procyonidae (raccoons) and Mustelidae (mustelids). The analyses favored a basal arctoid split between Ursidae and a branch containing Pinnipedia and Musteloidea. Within the Musteloidea there was a preference for a basal divergence between Ailuridae and remaining families. Among the latter, the analyses identified a sister group relationship between Mephitidae and a branch that contained Procyonidae and Mustelidae. The mitogenomic distance between the wolf and the dog was shown to be at the same level as that of basal human divergences. The wolf and the dog are commonly considered as separate species in the popular literature. The mitogenomic result is inconsistent with that understanding at the same time as it provides insight into the time of the domestication of the dog relative to basal human mitogenomic divergences.

The family Mustelidae, which consists of Mustelinae, Lutrinae, Melinae, and Taxidiinae, is the largest family among Carnivora and is a highly diverse group. Recent molecular phylogenetic studies have clarified the phylogenetic relations among Mustelidae, but there remain several unresolved problems, particularly concerning the deep branchings. Whereas many studies support the monophyly of Mustelidae+Procyonidae among Musteloidea, the relations between Mustelidae+Procyonidae, Ailuridae, and Miphitidae are still unclear. To address these problems, we inferred a tree on the basis of the sequences of mitochondrial genomes and of multiple nuclear genes using the maximum likelihood method. Our results strongly support the hypothesis that the Taxidiinae branched at first, followed by the branching of the Melinae. After that, Mustelinae diversified, and Lutrinae evolved within Mustelinae. With respect to the deep branchings in Musteloidea, the Ailuridae/Mephitidae monophyly tree and the Mephitidae-basal tree are indistinguishable in log-likelihood score, and this problem remains unresolved.

Red pandas, Ailurus fulgens, are popular exhibit animals in zoos. It is clear from data in the global studbook that there is considerable variation in their breeding success in different zoos. Population managers have long suspected that environmental temperature plays a key role in these differences. It is generally thought that this species, which is so well adapted to life in the cold damp climate of the mid-altitude forests of the Himalayas, has a problem coping with warmer climates. However, this hypothesis has not been tested until now. Using data extracted from the global studbook, we have demonstrated that climate at the location of birth has a clear impact on the survival of infant red pandas.

Inferences of function and ecology in extinct taxa have long been a subject of interest because it is fundamental to understand the evolutionary history of species. In this study, we use a quantitative approach to investigate the locomotor behaviour of Simocyon batalleri, a key taxon related to the ailurid family. To do so, we use 3D surface geometric morphometric approaches on the three long bones of the forelimb of an extant reference sample. Next, we test the locomotor strategy of S. batalleri using a leave-one-out cross-validated linear discriminant analysis. Our results show that S. batalleri is included in the morphospace of the living species of musteloids. However, each bone of the forelimb appears to show a different functional signal suggesting that inferring the lifestyle or locomotor behaviour of fossils can be difficult and dependent on the bone investigated. This highlights the importance of studying, where possible, a maximum of skeletal elements to be able to make robust inferences on the lifestyle of extinct species. We identified the winter plant species consumed by red panda in the Dhorpatan Hunting Reserve of eastern Nepal and compared this to the early-summer diet which was determined previously by Panthi et al. (2012). In addition, we estimated the proximate nutritional content of the leaves identified in red panda diet for both seasons, and we used nutritional geometry to explore macronutrient balance of leaves from the two different sampling periods. We identified six different plants in winter scats, which were the same as found in the previously determined early-summer diet. Arundinaria spp. bamboos were the main species found (82.1 % relative frequency), followed by Acer spp. (6.3 %), Betula utilis (4.6 %), Quercus semicarpifolia (3.7 %), Berberis spp. (1.3 %), and lichens (1.0 %), leaving 2.0 % unidentified. Geometric analysis suggested that the macronutrient balance of seasonal diets were similar in nutrient balance to the most frequently consumed Arundinaria spp. Differences in macronutrient balance may indicate seasonal nutrient preferences, such as increased carbohydrate intake in winter for thermogenesis, and increased protein and lipid intake in early summer to support reproduction and lactation; however, these differences may also indicate differences in resource availability. Habitat conserved for red panda in the region should include sufficient Arundinaria spp. as well as lesser consumed plants which may serve as complimentary foods.

Two neonatal male red panda (Ailurus fulgens) littermates were submitted for necropsy examination. One animal was found dead with no prior signs of illness; the other had a brief history of laboured breathing. Post-mortem examination revealed disseminated protozoal infection. To further characterize the causative agent, transmission electron microscopy (TEM), immunohistochemistry (IHC), polymerase chain reaction (PCR) and amplification and nucleic acid sequencing were performed. IHC was negative for Toxoplasma gondii and Neospora caninum, but was positive for a Sarcocystis spp. TEM of cardiac muscle and lung revealed numerous intracellular apicomplexan protozoa within parasitophorous vacuoles. PCR and nucleic acid sequencing of partial 18S rRNA and the internal transcribed spacer (ITS)-1 region confirmed a Sarcocystis spp. that shared 99% sequence homology to Sarcocystis neurona and Sarcocystis dasypi. This represents the first report of sarcocystosis in red pandas. The histopathological, immunohistochemical, molecular and ultrastructural findings are supportive of vertical transmission resulting in fatal disseminated disease.

The red panda (Ailurus fulgens) is distributed primarily in the Himalayas and southern China. It is classified as a vulnerable species by the International Union for Conservation of Nature. The aim of this study was to describe the normal osteology and radiographic anatomy of the thoracic limb of the red panda. Radiography of the right thoracic limb was performed in seven captive adult red pandas. Radiographic findings were correlated with bone specimens from three adult animals. The scapula was wide craniocaudally and presented with a large area for the origin of the teres major muscle. The square-shaped major tubercle did not extend proximal to the head of the humerus. The medial epicondyle was prominent. A supracondylar foramen was present. The radial tuberosity and sesamoid bone for the abductor digiti I longus were prominent. The accessory carpal bone was directed palmarolaterally. Metacarpal bones were widely spread. The thoracic limb morphology of the red panda evidenced by osteology and radiography indicated flexibility of the thoracic limb joints and well-developed flexor and supinator muscles, which are important in arboreal quadrupedal locomotion. Knowledge gained during this study may prove useful in identifying skeletal material or remains and diagnosing musculoskeletal diseases and injuries of the thoracic limb.

A second nearly complete skeleton of the fossil ailurid Pristinailurus bristoli was recently recovered from the Hemphillian (late Miocene) Gray Fossil Site of northeast Tennessee. It appears to be significantly larger than the previously known skeleton of P. bristoli, which was surprising as the living red panda Ailurus fulgens is thought to be generally monomorphic, including between sexes. Here the presence of significant size difference in the geometric means of cranial and postcranial measurements and canine area in the two specimens of P. bristoli is tested relative to the distribution of size differences in intersex and intrasex pairs of A. fulgens. The presence of sexual dimorphism in these dimensions is also assessed in A. fulgens using t-tests. We report the first evidence of sexual dimorphism in both upper and lower canine area in A. fulgens. We also found evidence of significantly greater body size difference in the two specimens of P. bristoli than that present in A. fulgens. This could reflect a variety of causes, including the presence of multiple species or subspecies, a greater range of ecologically driven variability, or the presence of sexual dimorphism. We argue tentatively for sexual dimorphism as the most plausible explanation for the apparent size range of P. bristoli. Ethan L. Fulwood. Phylogenetic relationships of 79 caniform carnivores were addressed based on four nuclear sequence-tagged sites (STS) and one nuclear exon, IRBP, using both supertree and supermatrix analyses. We recovered the three major arctoid lineages, Ursidae, Pinnipedia, and Musteloidea, as monophyletic, with Ursidae (bears) strongly supported as the basal arctoid lineage. Within Pinnipedia, Phocidae (true seals) were sister to the Otaroidea [Otariidae (fur seals and sea lions) and Odobenidae (walrus)]. Phocid subfamily and tribal designations were supported, but the otariid subfamily split between fur seals and sea lions was not. All family designations within Musteloidea were strongly supported: Mephitidae (skunks), Ailuridae (monotypic red panda), Mustelidae (weasels, badgers, otters), and Procyonidae (raccoons). A novel hypothesis for the position of the red panda was recovered, placing it as branching after Mephitidae and before Mustelidae + Procyonidae. Within Mustelidae, subfamily taxonomic changes are considered. This study represents the most comprehensive sampling to date of the Caniformia in a molecular study and contains the most complete molecular phylogeny for the Procyonidae. Our data set was also used in an empirical examination of the effect of missing data on both supertree and supermatrix analyses. Sequence for all genes in all taxa could not be obtained, so two variants of the data set with differing amounts of missing data were examined. The amount of missing data did not have a strong effect; instead, phylogenetic resolution was more dependent on the presence of sufficient informative characters. Supertree and supermatrix methods performed equivalently with incomplete data and were highly congruent; conflicts arose only in weakly supported areas, indicating that more informative characters are required to confidently resolve close species relationships.

Considerable long-standing controversy and confusion surround the phylogenetic affinities of pinnipeds, the largely marine group of "fin-footed" members of the placental mammalian order Carnivora. Until most recently, the two major competing hypotheses were that the pinnipeds have a single (monophyletic) origin from a bear-like ancestor, or that they have a dual (diphyletic) origin, with sea lions (Otariidae) derived from a bear-like ancestor, and seals (Phocidae) derived from an otter-, mustelid-, or musteloid-like ancestor. We examined phylogenetic relationships among 29 species of arctoid carnivorans using a concatenated sequence of 3228 bp from three nuclear loci (apolipoprotein B, APOB; interphotoreceptor retinoid-binding protein, IRBP; recombination-activating gene 1, RAG1). The species represented Pinnipedia (Otariidae: Callorhinus, Eumetopias; Phocidae: Phoca), bears (Ursidae: Ursus, Melursus), and Musteloidea (Mustelidae: Mustela, Enhydra, Melogale, Martes, Gulo, Meles; Procyonidae: Procyon; Ailuridae: Ailurus; Mephitidae: Mephitis). Maximum parsimony, maximum likelihood, and Bayesian inference phylogenetic analyses of separate and combined datasets produced trees with largely congruent topologies. The analyses of the combined dataset resulted in well-resolved and well-supported phylogeny reconstructions. Evidence from nuclear DNA evolution presented here contradicts the two major hypotheses of pinniped relationships and strongly suggests a single origin of the pinnipeds from an arctoid ancestor shared with Musteloidea to the exclusion of Ursidae.

Pancreatic ribonuclease (RNASE1) is a digestive enzyme that has been one of the key models in studies of evolutionary innovation and functional diversification. It has been believed that the RNASE1 gene duplications are correlated with the plant-feeding adaptation of foregut-fermenting herbivores. Here, we characterized RNASE1 genes from Caniformia, which has a simple digestive system and lacks microbial digestion typical of herbivores, in an unprecedented scope based on both gene sequence and tissue expression analyses. Remarkably, the results yielded new hypotheses regarding the evolution and the function of Caniformia RNASE1 genes. Four independent gene duplication events in the families of superfamily Musteloidea, including Procyonidae, Ailuridae, Mephitidae and Mustelidae, were recovered, rejecting previous Mustelidae-specific duplication hypothesis, but supporting Musteloidea duplication hypothesis. Moreover, our analyses revealed pronounced differences among the RNASE1 gene copies regarding their selection pressures, pI values and tissue expression patterns, suggesting the differences in their physiological functions. Notably, the expression analyses detected the transcription of a RNASE1 pseudogene in several tissues, raising the possibility that pseudogenes are also a potential source during the RNase functional diversification. In sum, the present work demonstrated a far more complex and intriguing evolutionary pattern and functional diversity of mammalian ribonuclease than previously thought.

To describe the successful management of gastric dilatation and volvulus (GDV) in a red panda.Clinical report.Red panda diagnosed with GDV.A 12-year-old male red panda (Ailurus fulgens) was evaluated for acute onset inappetence, staggering, collapse, and tachypnea. Gastric dilatation and volvulus (GDV) was diagnosed by radiography, abdominal ultrasonography, and exploratory celiotomy. Torsion of the stomach was corrected and an incisional gastropexy performed to prevent recurrence. No organs were devitalized, no other abnormalities detected, and the red panda recovered fully within 72 hours.GDV should be considered as a differential diagnosis for red pandas presenting with acute onset of unspecific signs such as collapse, inappetence, and abdominal distension. GDV in red pandas can be diagnosed and successfully treated as described in dogs.

The red ( Ailurus fulgens ) and giant ( Ailuropoda melanoleuca ) pandas are mammalian carnivores convergently adapted to a bamboo feeding diet. However, whereas Ailurus forages almost entirely on younger leaves, fruits and tender trunks, Ailuropoda relies more on trunks and stems. Such difference in foraging mode is considered a strategy for resource partitioning where they are sympatric. Here, we use finite-element analysis to test for mechanical differences and similarities in skull performance between Ailurus and Ailuropoda related to diet. Feeding simulations suggest that the two panda species have similar ranges of mechanical efficiency and strain energy profiles across the dentition, reflecting their durophagous diet. However, the stress distributions and peaks in the skulls of Ailurus and Ailuropoda are remarkably different for biting at all tooth locations. Although the skull of Ailuropoda is capable of resisting higher stresses than the skull of Ailurus , the latter is able to distribute stresses more evenly throughout the skull. These differences in skull biomechanics reflect their distinct bamboo feeding preferences. Ailurus uses repetitive chewing in an extended mastication to feed on soft leaves, and Ailuropoda exhibits shorter and more discrete periods of chomp-and-swallow feeding to break down hard bamboo trunks.

The first detailed description and comparative information is given for the carnivoran fauna from Wolfersheim (Hesse), the only Ruscinian (MN 15) fauna thus far known from Germany. The Carnivora is represented by 31 specimens which belong to the following taxa:Ursus minimus boeckhi n. comb., cf.Pannonictis, Enhydrictis ardea, Baranogale sp.,Mustela plioerminea, andParailurus anglicus. The specimens ofUrsus, Enhydrictis, andMustela show the most plesiomorphic states of tooth morphology hitherto known in these genera. With a total of 18 specimens,Parailurus is the most common carnivoran in Wolfersheim and makes the collection the second largest assemblage of fossil ailurids. It provides new information about intraspecific dental variability which allows placement of all specimens described asP. hungaricus into the type speciesP. anglicus. Furthermore, we offer additional support for the validity of the genusParailurus. One controversial specimen is interpreted as a dP4/ ofP. anglicus instead of as an aberrant Ml/ of a newParailurus species.

Radiography is a valuable tool for assessment of pulmonary disease. Specifically, radiographs utilizing positive pressure ventilation can distinguish between anesthesia-induced atelectasis and pulmonary disease when survey radiographs are ambiguous. Positive pressure ventilation can be used to radiographically prove or disprove pulmonary disease. This is of particular clinical importance when working with exotic, zoo, or wildlife species because the majority of these patients require general anesthesia to perform physical examinations and diagnostics such as radiography safely and efficiently. This report is a case example of pulmonary disease in a red panda (Ailurus fulgens) and demonstrates how positive pressure ventilation verified both the presence of pulmonary disease and the eventual resolution of the disease. Anesthetized patients on gas anesthesia will rapidly become atelectic. Through the use of positive pressure ventilation, anesthesia-induced atelectasis and true pulmonary disease can readily be distinguished. This is a technique that should not be overlooked when performing thoracic radiography in zoo species.

Resume Une M2 isolee, provenant du Miocene moyen de Madrid (Espagne) est attribuee a un nouveau taxon: Magerictis imperialensis gen. nov. sp. nov., qui est etroitement apparente au Petit Panda, Ailurus fulgens. Ce dernier est donc d'origine europeenne et trouve ses racines, comme les Simocyonidae, chez certains Mustelidae a M2 allongee des Phosphorites du Quercy, comme Amphictis.

The phylogenetic relationships of the skunks to the Mustelidae and other caniform carnivores were examined using mitochondrial-DNA (mtDNA) sequence data from portions of the 12S and 16S ribosomal RNA (rRNA) genes. Data were combined with partial sequences of the cytochrome b gene and morphological data obtained from the literature, and used in a total-evidence analysis. The Mustelidae represented a paraphyletic group, with the skunks ( Conepatus, Mephitis , and Spilogale ) and the Oriental stink badger ( Mydaus ) forming a monophyletic clade separate from a clade containing the rest of the Mustelidae and the monophyletic Procyonidae. Within the Mustelidae, minus the skunks and stink badger, only one currently recognized subfamily, the Lutrinae, represented a monophyletic group. The families Phocidae, Otariidae, and Odobenidae formed a monophyletic group that was the sister group to the clade composed of the skunks, procyonids, and mustelids. The families Ursidae and Canidae occurred at the base of the Caniformia clade. It is proposed that the skunks be elevated to the level of family and be referred to as the Mephitidae. The family Mephitidae includes the genera Mephitis (striped and hooded skunks), Conepatus (hog-nosed skunks), Spilogale (spotted skunks), and Mydaus (Oriental stink badgers).

A well-preserved skull of Simocyon, an extinct hypercarnivorous procyonid, is described from a late Miocene (Baodean; approximately 5.3–9 Ma) locality in north Shaanxi, China. New knowledge about its cranial morphology allows insight into its phylogenetic position among musteloid camivorans. A sister-group relationship between the simocyonines and the ailurines (the East Asian red panda) is proposed on the basis of cranial and dental evidence. Shared derived characters in support of this relationship include: highly arched zygomatic arch; posteriorly extended posterior palatine border; long bony external auditory meatus; a posterolateral process of promontorium; ventrally ridged paroccipital process; anteriorly inclined coronoid crest; and lateral grooves on canines. The simocyonine-ailurine clade is in turn postulated to be within the procyonid clade because of its common possession of an enlarged M2 and an elongated talonid on m2. The red panda is the only living species of the genus Ailurus. Like giant pandas, red pandas are also highly specialized to feed mainly on highly fibrous bamboo. Although several studies have focused on the gut microbiota in the giant panda, little is known about the gut microbiota of the red panda. In this study, we characterized the fecal microbiota from both wild (n = 16) and captive (n = 6) red pandas using a pyrosequecing based approach targeting the V1-V3 hypervariable regions of the 16S rRNA gene. Distinct bacterial communities were observed between the two groups based on both membership and structure. Wild red pandas maintained significantly higher community diversity, richness and evenness than captive red pandas, the communities of which were skewed and dominated by taxa associated with Firmicutes. Phylogenetic analysis of the top 50 OTUs revealed that 10 of them were related to known cellulose degraders. To the best of our knowledge, this is the first study of the gut microbiota of the red panda. Our data suggest that, similar to the giant panda, the gut microbiota in the red panda might also play important roles in the digestion of bamboo.

An intact adult male 14.3-yr-old red panda (Ailurus fulgens) presented for health examination with a history of slowly progressing loss of body condition. Abdominal radiographs revealed a truncated abdomen with poor serosal abdominal detail and multiple areas of spondylosis with some collapsed intervertebral disc spaces. On computed tomography, multiple ovoid hypoattenuating lesions were seen in the left and right kidneys. Gross pathology and histopathology revealed multiple cystic lesions in the kidneys concurrent with pancreatic cysts on histopathology. To the best of the authors' knowledge, polycystic kidneys have not been reported in this species.

Background A captive Red Panda developed a regenerative anemia with Heinz bodies after being sprayed by a skunk. A definite cause-and-effect relationship between skunk musk and oxidative erythrocyte damage has not been reported, but it was suspected in one reported case of a dog with Heinz body hemolytic anemia. Objective The objective was to determine whether skunk musk induces oxidative HGB damage in vitro. Methods Plasma and RBC were harvested from heparinized blood of 3 dogs, 3 cats, and a Red Panda. Skunk musk was solubilized in ethanol and mixed with plasma from each species to make stock solutions of 4% musk and 4% ethanol. Aliquots of RBC were resuspended in autologous stock solutions and solvent controls to yield musk concentrations of 0%, 0.04%, and 0.4% (by volume). Aliquots were incubated at 37°C for 4–72 hours and assessed for oxidative damage by visual inspection, optical absorbance spectroscopy, transmission electron microscopy, and light microscopy after Wright and vital New Methylene Blue staining. Results Dose-dependent brown color and absorption changes characteristic of methemoglobin were present by 4 hours and increased over 24 hours (Red Panda) and 72 hours (dog and cat). Similarly, there were time-dependent (all species) and dose-dependent (dog and cat) increases in the number of Heinz bodies, which were present by 4 hours and numerous by 24 hours. Conclusions In vitro, skunk musk causes Heinz body and methemoglobin formation in canine, feline, and Red Panda RBC, supporting the clinical association between Heinz body hemolytic anemia and skunk spray exposure.

The red panda (Ailurus fulgens) is a quadrupedal arboreal animal primarily distributed in the Himalayas and southern China. It is a species commonly kept in zoological collections. This study was carried out to describe the morphology of the pelvis and hind limb of the red panda evidenced by gross osteology, radiography and computed tomography as a reference for clinical use and identification of skeletons. Radiography of the pelvis and right hind limb was performed in nine and seven animals, respectively. Radiographic findings were correlated with bone specimens from three adult animals. Computed tomography of the torso and hind limb was performed in one animal. The pelvic bone had a wide ventromedial surface of the ilium. The trochlea of the femur was wide and shallow. The patella was similar to that seen in feline species. The medial fabella was not seen radiographically in any animal. The cochlea grooves of the tibia were shallow with a poorly defined intermediate ridge. The trochlea of the talus was shallow and presented with an almost flattened medial ridge. The tarsal sesamoid bone was always present. The lateral process of the base of the fifth metatarsal (MT) bone was directed laterally. The MT bones were widely spaced. The morphology of the pelvis and hind limb of the red panda indicated flexibility of the pelvis and hind limb joints as an adaptation to an arboreal quadrupedal lifestyle.

Red pandas (Ailurus fulgens) are threatened with extinction owing to habitat loss, exacerbated by their unique ecology and low fecundity. Regional breeding programs manage captive red panda populations. Recommendations not to breed may be made for various reasons, including genetic overrepresentation of certain individuals. No recommendations have been published on the use of contraception for red pandas. This article discusses the use of the GnRH analog deslorelin as a reversible method of contraception in both male and female pandas. The mean time from last contraception to conception was 3 years with a 4.6-mg deslorelin implant. The average dose of GnRH implant received was 1.09 mg/kg (range, 0.88–1.32). Males returned to breeding sooner than females. No reproductive side effects were noted with up to three consecutive annual GnRH implants.

In order to provide new source for discovering new lead compounds of drugs and other products, the diversity and some bioactivities of culturable actinobacteria in animal feces were studied.Five animals' fecal samples were collected from Yunnan Wild Animal Park. The pure cultures of actinobacteria were isolated from these samples by using 5 different media. The 16S rRNA gene sequences of 119 selected strains were determined; the phylogenetic analysis was carried out; and antimicrobial and anti-tumor activities were determined by using agar diffusion method, tumor cell lines k562and HL60 respectively.In total 20 genera of actinobacteria from the 5 animals' feces were identified. Many strains inhibited Bacillus subtilis, Staphylococcus lentus, Mycobacterium tuberculosis, Candida albicans and Aspergillus niger. Some strains presented antitumor activities. Some known secondary metabolites and Sannastatin, a novel macrolactam polyketide glycoside with bioactivities, were isolated and identified.Fecal actinobacteria are a new potential source for discovering drug lead and other industry products.

The complete 12S rRNA gene of 32 carnivore species, including four feliforms and 28 caniforms, was sequenced. The sequences were aligned on the basis of their secondary structures and used in phylogenetic analyses that addressed several evolutionary relationships within the Caniformia. The analyses showed an unresolved polytomy of the basic caniform clades; pinnipeds, mustelids, procyonids, skunks, Ailurus (lesser panda), ursids, and canids. The polytomy indicates a major diversification of caniforms during a relatively short period of time. The lesser panda was distinct from other caniforms, suggesting its inclusion in a monotypic family, Ailuridae. The giant panda and the bears were joined on the same branch. The skunks are traditionally included in the family Mustelidae. The present analysis, however, showed a less close molecular relationship between the skunks and the remaining Mustelidae (sensu stricto) than between Mustelidae (sensu stricto) and Procyonidae, making Mustelidae (sensu lato) paraphyletic. The results suggest that the skunks should be included in a separate family, Mephitidae. Within the Pinnipedia, the grouping of walrus, sea lions, and fur seals was strongly supported. Analyses of a combined set of 12S rRNA and cytochrome b data were generally consistent with the findings based on each gene.

The phylogenetic placement of the red panda (Ailurus fulgens) and the giant panda (Ailuropoda melanoleuca) has been an evolutionary enigma since their original descriptions in the nineteenth century. A series of recent molecular analyses led to a consensus that the giant panda's ancestors were derived from early bears (Ursidae), but left unsettled the phylogenetic relationship of the red panda. Previous molecular and morphological phylogenies were inconclusive and varied among placement of the red panda within the raccoon family (Procyonidae), within the bear family (Ursidae), or in a separate family of carnivores equidistant between the two. To examine a relatively ancient (circa 20-30 million years before the present, MYBP) phylogenetic divergence, we used two slowly evolving genetic markers: mitochondrial 12S rRNA sequence and 592 fibroblast proteins resolved by two dimensional gel electrophoresis. Four different carnivore outgroup species, including dog (Canidae: Canis familiaris), cat (Felidae: Felis catus), fanaloka (Viverridae: Fossa fossa), and mongoose (Herpestidae: Galidia elegans), were selected to identify the root of the phylogenetic topologies. Phylogenetic reconstruction by distance-based methods, maximum parsimony, and maximum likelihood clearly indicate a distinct bifurcation forming the Ursidae and the Procyonidae. Further, our data consistently place the red panda as an early divergence within the Procyonidae radiation and confirm the inclusion of giant panda in the Ursidae lineage.

Many different species of the order Carnivora are susceptible to canine distemper and the mortality rate varies greatly between species. Ailuridae, Canidae, Hyaenidae, Mustelidae, Procyonidae, Ursidae, Viverridae and now Felidae have been reported to be susceptible to canine distemper virus infection. Although distemper outbreaks in dogs, fur farms and in zoo carnivores have been greatly reduced in recent years due to vaccination, there are still regular outbreaks in free-living carnivores. Unexpected outbreaks of canine distemper have occurred in exotic felids in a California wildlife park and in the Serengeti in Tanzania as well as in javelinas (collared peccaries, Tayassu tajacu) in Arizona. Although safe and efficacious in dogs, modified live canine distemper virus vaccines may be dangerous for a variety of zoo and wildlife carnivores, especially red pandas (Ailurus fulgens) and black footed ferrets (Mustela nigripes).

In a recent survey, 30% of the European red panda (Ailurus fulgens) population was found to be infected with a newly discovered metastrongyloid nematode. In a following prospective study, four naturally infected captive-bred red pandas infected with this parasite were examined and compared with two uninfected control animals. On clinical examination, no abnormalities were detected with respect to vital parameters and cardiovascular system in all six examined animals. Similarly, few and nonspecific changes were recorded on serum biochemistry. No changes on pulmonary pattern were noted on thoracic radiographs. Vertebral heart scores were between 7.2 to 8.6, and no difference was noted between infected and control animals. Two animals had slightly prolonged clotting time and reaction time on thromboelastography but not likely to be of clinical relevance. In conclusion, infection with the newly identified metastrongyloid nematode in the red pandas seems to have little or no clinical importance.

Ten of the 48 red pandas in the Chengdu Research Base of Giant Panda Breeding, Sichuan province, China, died in 2006 after prolonged periods of depression, weight loss, and mucocutaneous membrane xanthochromia. During postmortem examination, live heartworms were found in the right cardiac ventricles and pulmonary arteries of all 10 animals. Selamectin and ivermectin were used for clinical prophylaxis in the remaining red pandas between December 2006 and November 2010. We observed a gradual decrease in morbidity and mortality during this period. As a consequence of our prophylaxis program, dirofilariosis did not occur in the remaining red pandas at Chengdu Research Base during 2010.

Molecular sexing is a key component in the investigation of wild populations. In this study, we developed a fast, accurate and reliable amplification refractory mutation system (ARMS) technique for sex determination of red panda based on the exon 4 of the ZFX/ZFY gene. The amplicons were distinguished simply by agarose gel electrophoresis, exhibiting one fragment in females (X: 300 bp) and two in males (X: 300 bp, Y: 166 bp). Robustness of this ARMS system was confirmed by testing both 43 captive red pandas using DNA samples with known-sex and 10 wild red pandas using faecal DNA samples with unknown sex.

The red panda family, the Ailuridae, is unusual because its only extant representative is a derived form, far removed from the group’s morphotypical condition. Effectively, the extreme adaptations of the red panda skull and dentition for a vegetarian diet imply that the study of the fossil record is essential if we are to gain an understanding of the original biology and ecology of the ailurids. Fossil ailurids have remained frustratingly rare and fragmentary findings for many decades, but recent years have seen a dramatic improvement of their record. This has been especially the case with the recent finds of dramatically complete remains of the simocyonine Simocyon batalleri from Batallones-1 in Madrid, Spain, and of the ailurine Pristinailurus bristoli from Gray Fossil Site in Tennessee, United States. Taking into account the information provided by these and other recent findings, it is now possible to gain a more complete picture than ever before of the true place of ailurids within the Carnivora, and of the evolutionary processes that led to the remarkable convergences between the only living ailurid and the unrelated giant panda. In this chapter, we present the early evolution of the ailurids, its Old-World fossil record, and the implications of the palaeontological evidence for the position of the ailurids among other carnivoran families.
