Abstract

Abstract Background Problems associated with alcohol use are multidimensional with psychiatric, psychological, physical, and social aspects, which makes it challenging to choose appropriate assessment scales. However, there has been no systematic evaluation of existing alcohol scales. Methods A systematic literature search was conducted for articles that assessed the psychometric properties of scales for alcohol use disorder on March 19, 2023, using Medline, EMBASE, and PsycINFO. Only scales whose original development papers were cited more than 20 times were included. The methodological quality and psychometric properties of the scales were evaluated using COnsensus‐based Standards for the selection of health Measurement INstruments. The overall rating of the scales were assessed with a score ranging from 0 to 18. Results In total, 314 studies and 40 scales were identified. These scales differ widely in measurement methods, target populations, and psychometric properties. The overall mean score was 6.3, and only the following three scales received >9 points suggesting a moderate level of evidence: Alcohol Use Disorders Identification Test (AUDIT), Alcohol Dependence Scale (ADS), and Short Alcohol Dependence Data Questionnaire (SADD). Measurement error and responsiveness were not evaluated or reported in the included scales. Conclusions Although the AUDIT, ADS, and SADD were rated the highest among the 40 scales, they showed, at most, a moderate level of evidence. These findings underscore the need to accumulate further evidence to assure the quality of the scales. It may be advisable to select and combine scales to meet the purpose of the assessment.

Abstract

Abstract Mitochondrial dysfunction causes maternally inherited deafness and diabetes (MIDD). Herein, we report improved glycemic control in a 47‐year‐old Japanese woman with MIDD using imeglimin without major adverse effects. Biochemical tests and metabolome analysis were performed before and after imeglimin administration. Blood glucose level fluctuations were determined. Sulfonylureas, dipeptidyl peptidase‐4 inhibitors (DPP4is), and sodium glucose transporter‐2 inhibitors (SGLT2i) were administered to evaluate the efficacy of their combination with imeglimin. Imeglimin decreased the HbA1c and ammonia levels and increased the time‐in‐range, C‐peptide reactivity, and glucagon level. Elevated citrulline and histamine levels were decreased by imeglimin. The hypoglycemic effect was not enhanced by imeglimin when combined with sulfonylurea or DPP4i, but the blood glucose level was improved when combined with SGLT2i. Imeglimin improved glucose concentration‐dependent insulin secretion and maximized the insulin secretory capacity by improving mitochondrial function and glutamine metabolism and urea circuit abnormalities by promoting glucagon secretion. Imeglimin could improve glycemic control in MIDD.

Abstract

Abstract Fatty acids play various physiological roles owing to their diverse structural characteristics, such as hydrocarbon chain length (HCL) and degree of saturation (DS). Although the distribution of fatty acids in biological tissues is associated with lipid metabolism, in situ imaging tools are still lacking for HCL and DS. Here, we introduce a framework of near-infrared (1000–1400 nm) hyperspectral label-free imaging with machine learning analysis of the fatty acid HCL and DS distribution in the liver at each pixel, in addition to the previously reported total lipid content. The training data of 16 typical fatty acids were obtained by gas chromatography from liver samples of mice fed with various diets. A two-dimensional mapping of these two parameters was successfully performed. Furthermore, the HCL/DS plot exhibited characteristic clustering among the different diet groups. Visualization of fatty acid distribution would provide insights for revealing the pathophysiological conditions of liver diseases and metabolism.

Abstract

Abstract Background There have been very few real-world studies reported in the literature solely focusing on fremanezumab in Asia. This study aimed to evaluate the efficacy and safety of fremanezumab in a real-world setting in Japan. Method This single-centered, observational, retrospective study examined patients with migraines who received four doses of fremanezumab between December 2021 and August 2022 at Keio University Hospital. We assessed the changes in monthly migraine days, responder rates, and migraine-associated symptoms, as well as injection site reactions and adverse events. Result Twenty-nine patients were enrolled, wherein 79.3% were women. Compared with those at baseline, the monthly migraine days decreased by 5.9 days at 4 months. The 50% responder rate was 55.2% at 4 months. A total of 57.9%, 47.8%, and 65.0% of patients showed improvement in the severity of photophobia, phonophobia, and nausea/vomiting, respectively. Moreover, injection site reactions were the most common adverse events (55.2%). Conclusion Fremanezumab is effective and safe for migraine prevention in Japan. Fremanezumab also improved migraine-associated symptoms in half of the patients.

Abstract

Transporter-mediated clearance is determined by two factors, its single-molecule clearance, and expression level. However, no reliable method has been developed to evaluate them separately. This study aimed to develop a reliable method for evaluating the single-molecule activity of membrane transporters, such as organic anion transporting polypeptide (OATP) 2B1.HEK293 cells that co-expressed large conductance calcium-activated potassium (BK) channel and OATP2B1 were established and used for the following experiments. i) BK channel-mediated whole-cell conductance was measured using patch-clamp technique and divided by its unitary conductance to estimate the number of channels on plasma membrane (QI). ii) Using plasma membrane fraction, quantitative targeted absolute proteomics determined the stoichiometric ratio (ρ) of OATP2B1 to BK channel. iii) The uptake of estrone 3-sulfate was evaluated to calculate the Michaelis constant and uptake clearance (CL) per cell. Single-molecule clearance (CLint) was calculated by dividing CL by QI·ρ.QI and ρ values were estimated to be 916 and 2.16, respectively, yielding CLint of 5.23 fL/min/molecule. We successfully developed a novel method to reliably measure the single-molecule activity of a transporter, which could be used to evaluate the influences of factors such as genetic variations and post-translational modifications on the intrinsic activity of transporters.

Abstract

Milky Way dwarf spheroidal galaxies (dSphs) are among the best candidates to search for signals of dark matter annihilation with Imaging Atmospheric Cherenkov Telescopes, given their high mass-to-light ratios and the fact that they are free of astrophysical gamma-ray emitting sources. Since 2011, MAGIC has performed a multi-year observation program in search for Weakly Interacting Massive Particles (WIMPs) in dSphs. Results on the observations of Segue 1 and Ursa Major II dSphs have already been published and include some of the most stringent upper limits (ULs) on the velocity-averaged cross-section $\langle \sigma_{\mathrm{ann}} v \rangle$ of WIMP annihilation from observations of dSphs. In this work, we report on the analyses of 52.1 h of data of Draco dSph and 49.5 h of Coma Berenices dSph observed with the MAGIC telescopes in 2018 and in 2019 respectively. No hint of a signal has been detected from either of these targets and new constraints on the $\langle \sigma_{\mathrm{ann}} v \rangle$ of WIMP candidates have been derived. In order to improve the sensitivity of the search and reduce the effect of the systematic uncertainties due to the $J$-factor estimates, we have combined the data of all dSphs observed with the MAGIC telescopes. Using 354.3 h of dSphs good quality data, 95 % CL ULs on $\langle \sigma_{\mathrm{ann}} v \rangle$ have been obtained for 9 annihilation channels. For most of the channels, these results reach values of the order of $10^{-24} $cm$^3$/s at ${\sim}1$ TeV and are the most stringent limits obtained with the MAGIC telescopes so far., Comment: 17 pages, 5 figures

Abstract

We report multiwavelength observations of the gravitationally lensed blazar QSO B0218+357 in 2016-2020. Optical, X-ray and GeV flares were detected. The contemporaneous MAGIC observations do not show significant very-high-energy (VHE, >= 100 GeV) gamma-ray emission. The lack of enhancement in radio emission measured by OVRO indicates the multi-zone nature of the emission from this object. We constrain the VHE duty cycle of the source to be < 16 2014-like flares per year (95% confidence). For the first time for this source, a broadband low-state SED is constructed with a deep exposure up to the VHE range. A flux upper limit on the low-state VHE gamma-ray emission of an order of magnitude below that of the 2014 flare is determined. The X-ray data are used to fit the column density of (8.10 +- 0.93 stat ) x 10^21 cm^-2 of the dust in the lensing galaxy. VLBI observations show a clear radio core and jet components in both lensed images, yet no significant movement of the components is seen. The radio measurements are used to model the source-lens-observer geometry and determine the magnifications and time delays for both components. The quiescent emission is modeled with the high-energy bump explained as a combination of synchrotron-self-Compton and external Compton emission from a region located outside of the broad line region. The bulk of the low-energy emission is explained as originating from a tens-of-parsecs scale jet., Comment: 21 pages, 12 figures, 5 tables, accepted for publication in MNRAS

Abstract

Recent self-supervised models have demonstrated equal or better performance than supervised methods, opening for AI systems to learn visual representations from practically unlimited data. However, these methods are typically classification-based and thus ineffective for learning high-resolution feature maps that preserve precise spatial information. This work introduces superpixels to improve self-supervised learning of dense semantically rich visual concept embeddings. Decomposing images into a small set of visually coherent regions reduces the computational complexity by $\mathcal{O}(1000)$ while preserving detail. We experimentally show that contrasting over regions improves the effectiveness of contrastive learning methods, extends their applicability to high-resolution images, improves overclustering performance, superpixels are better than grids, and regional masking improves performance. The expressiveness of our dense embeddings is demonstrated by improving the SOTA unsupervised semantic segmentation benchmark on Cityscapes, and for convolutional models on COCO., Comment: Accepted for BMVC 2022

120. New Insights Into the Substorm Initiation Sequence From the Spatio‐Temporal Development of Auroral Electrojets

Abstract

In the present study we examine three substorm events, Events 1-3, focusing on the spatio-temporal development of auroral electrojets (AEJs) before auroral breakup. In Events 1 and 2, auroral breakup was preceded by the equatorward motion of an auroral form, and the ground magnetic field changed northward and southward in the west and east of the expected equatorward flow, respectively. Provided that these magnetic disturbances were caused by local ionospheric Hall currents, this feature suggests that the equatorward flow turned both eastward and westward as it reached the equatorward part of the auroral oval. The auroral breakup took place at the eastward-turning and westward-turning branches in Events 1 and 2, respectively, and after the auroral breakup, the westward AEJ enhanced only on the same side of the flow demarcation meridian. The zonal flow divergence is considered as an ionospheric manifestation of the braking of an earthward flow burst in the near-Earth plasma sheet and subsequent dawnward and duskward turning. Therefore, in Events 1 and 2, the auroral breakup presumably mapped to the dawnward and duskward flow branches, respectively. Moreover, for Event 3, we do not find any pre-onset auroral or magnetic features that can be associated with an equatorward flow. These findings suggest that the braking of a pre-onset earthward flow burst itself is not the direct cause of substorm onset, and therefore, the wedge current system that forms at substorm onset is distinct from the one that is considered to form as a consequence of the flow braking.

Abstract

The results of gamma-ray observations of the binary system HESS J0632+057 collected during 450 hours over 15 years, between 2004 and 2019, are presented. Data taken with the atmospheric Cherenkov telescopes H.E.S.S., MAGIC, and VERITAS at energies above 350 GeV were used together with observations at X-ray energies obtained with Swift-XRT, Chandra, XMM-Newton, NuSTAR, and Suzaku. Some of these observations were accompanied by measurements of the H{\alpha} emission line. A significant detection of the modulation of the VHE gamma-ray fluxes with a period of 316.7+-4.4 days is reported, consistent with the period of 317.3+-0.7 days obtained with a refined analysis of X-ray data. The analysis of data of four orbital cycles with dense observational coverage reveals short timescale variability, with flux-decay timescales of less than 20 days at very high energies. Flux variations observed over the time scale of several years indicate orbit-to-orbit variability. The analysis confirms the previously reported correlation of X-ray and gamma-ray emission from the system at very high significance, but can not find any correlation of optical H{\alpha} parameters with X-ray or gamma-ray energy fluxes in simultaneous observations. The key finding is that the emission of HESS J0632+057 in the X-ray and gamma-ray energy bands is highly variable on different time scales. The ratio of gamma-ray to X-ray flux shows the equality or even dominance of the gamma-ray energy range. This wealth of new data is interpreted taking into account the insufficient knowledge of the ephemeris of the system, and discussed in the context of results reported on other gamma-ray binary systems., Comment: Accepted for publication by The Astrophysical Journal

Abstract

The Large-Sized Telescope (LST) prototype of the future Cherenkov Telescope Array (CTA) is located at the Northern site of CTA, on the Canary Island of La Palma. It is designed to provide optimal performance in the lowest part of the energy range covered by CTA, observing gamma rays down to energies of tens of GeV. The LST prototype started performing astronomical observations in November 2019 during the commissioning of the telescope and it has been taking data since then. In this contribution, we will present the tuning of the characteristics of the telescope in the Monte Carlo (MC) simulations to describe the data obtained, the estimation of its angular and energy resolution, and an evaluation of its sensitivity, both with simulations and with observations of the Crab Nebula., Comment: Presented at the 37th ICRC

Abstract

PSR J0218+4232 is one of the most energetic millisecond pulsars known and has long been considered as one of the best candidates for very high-energy (VHE; >100 GeV) gamma-ray emission. Using 11.5 years of Fermi Large Area Telescope (LAT) data between 100 MeV and 870 GeV, and ~90 hours of MAGIC observations in the 20 GeV to 20 TeV range, we have searched for the highest energy gamma-ray emission from PSR J0218+4232. Based on the analysis of the LAT data, we find evidence for pulsed emission above 25 GeV, but see no evidence for emission above 100 GeV (VHE) with MAGIC. We present the results of searches for gamma-ray emission, along with theoretical modeling, to interpret the lack of VHE emission. We conclude that, based on the experimental observations and theoretical modeling, it will remain extremely challenging to detect VHE emission from PSR J0218+4232 with the current generation of Imaging Atmospheric Cherenkov Telescopes (IACTs), and maybe even with future ones, such as the Cherenkov Telescope Array (CTA)., Comment: 19 pages, 9 figures, Accepted to ApJ

Abstract

The Cherenkov Telescope Array (CTA) will be the next generation gamma-ray observatory, which will consist of three kinds of telescopes of different sizes. Among those, the Large Size Telescope (LST) will be the most sensitive in the low energy range starting from 20 GeV. The prototype LST (LST-1) proposed for CTA was inaugurated in October 2018 in the northern hemisphere site, La Palma (Spain), and is currently in its commissioning phase. MAGIC is a system of two gamma-ray Cherenkov telescopes of the current generation, located approximately 100 m away from LST-1, that have been operating in stereoscopic mode since 2009. Since LST-1 and MAGIC can observe the same air shower events, we can compare the brightness of showers, estimated energies of gamma rays, and other parameters event by event, which can be used to cross-calibrate the telescopes. Ultimately, by performing combined analyses of the events triggering the three telescopes, we can reconstruct the shower geometry more accurately, leading to better energy and angular resolutions, and a better discrimination of the background showers initiated by cosmic rays. For that purpose, as part of the commissioning of LST-1, we performed joint observations of established gamma-ray sources with LST-1 and MAGIC. Also, we have developed Monte Carlo simulations for such joint observations and an analysis pipeline which finds event coincidence in the offline analysis based on their timestamps. In this work, we present the first detection of an astronomical source, the Crab Nebula, with combined observation of LST-1 and MAGIC. 146. Sex differences in COL1A1 Expression and Collagen Content in Skeletal Muscle of Mature and Juvenile Shamo Chickens

Abstract

Collagen content is an important parameter affecting meat consistency. Sex differences in collagen were therefore studied in mature and juvenile Shamo chickens. The pectoral (PT), lateral iliotibial (ITL), medial part of puboischiofemoral (PIF), and lateral part of gastrocnemius (GCL) muscles were weighed, and their COL1A1 expression levels and total collagen content were analyzed. Body and muscle weights were significantly higher in males than in females of all ages. Muscle/body weight ratios were also higher in mature males than in females, but this difference was not observed in juveniles. In mature chickens, COL1A1 expression was higher in the PIF and GCL muscles; this was not the case in juvenile chicken muscles. Sex differences in collagen content were observed only in the ITLs of mature chickens. A positive correlation between muscle weight and intramuscular collagen content was found for PT and GCL, but not for ITL and PIF, muscles. These results suggest that the sex difference in intramuscular collagen content only occurs in specific muscles and that COL1A1 expression is not necessarily related to collagen content in mature chickens. Factors that determine the intramuscular collagen content likely differ by muscle type.

Abstract

A 50-year-old Japanese man with enlargement of the right scrotum was presented to our hospital. Preoperative examination confirmed a multilocular cyst with septa attached to the testis. Radical orchiectomy was performed. Pathological examination revealed closely-located two cysts; larger one was infected hydrocele testis, and smaller one was epithelial cyst, which were immunohistochemically positive widely for estrogen receptor (ER) and partly for progesterone receptor (PR). We concluded that the smaller cyst was serous cystadenoma of the epididymis.

Abstract

BackgroundAchieving sustained virologic response (SVR) in patients infected with hepatitis C virus (HCV) reduces all-cause mortality. However, the mechanisms and risk factors for liver fibrosis and portal hypertension post-SVR remain incompletely understood. In the gut-liver axis, mucosa-associated microbiota (MAM) substantially influence immune and metabolic functions, displaying spatial heterogeneity at the anatomical intestinal site. We analyzed MAM composition and function to isolate the locoregional MAM involved in chronic liver disease progression in HCV post-SVR patients.MethodsWe collected MAM samples from three intestinal sites (terminal ileum, ascending colon, and sigmoid colon) via brushing during colonoscopy in 23 HCV post-SVR patients and 25 individuals without liver disease (controls). The 16S rRNA of bacterial DNA in specimens collected with a brush and in feces was sequenced. The molecular expression of intestinal tissues and hepatic tissues were evaluated by quantitative real-time PCR.ResultsIn the post-SVR group, the microbial β-diversity of MAM, especially in the ascending colon, differed from the control group and was associated with liver fibrosis progression. In PICRUSt analysis, MAM in the ascending colon in the liver cirrhosis (LC) group showed compromised functions associated with the intestinal barrier and bile acid production, and FGF19 expression was markedly decreased in the terminal ileum biopsy tissue in the LC group. At the genus level, six short-chain fatty acid (SCFA)-producing bacterial genera, Blautia, Alistipes, Roseburia, Agathobaculum, Dorea, and Pseudoflavonifractor were reduced in the ascending colon of post-SVR LC patients.ConclusionIn patients of HCV post-SVR, we identified the association between the degree of liver fibrosis and dysbiosis of mucosa-associated SCFA-producing bacterial genera that may be related to intestinal barrier and bile acid production in the ascending colon.

Abstract

Abstract Intelligent materials with responsive behaviors toward external stimuli, such as light, temperature, pH, redox, and solvent have been increasingly fascinating. Reversible noncovalent interactions provide an efficient way to construct stimuli‐responsive materials. Macrocyclic compounds, such as cyclodextrins, cucurbit[n]urils, calix[n]arenes, crown ethers, and related macrocycles, are useful skeletons for constructing such materials through host–guest interactions. Pillar[n]arenes are pillar‐shaped macrocyclic hosts developed by our groups in 2008, in which the repeated 1,4‐dialkoxybenzene units are connected by methylene bridges at the para position. The versatile functionality, easy modification, excellent size‐dependent host–guest complexation, and adjustable electron density of the cavity endow pillar[n]arenes with excellent properties compared with other cyclic host molecules. Moreover, the unique planar chirality and chirality inversion generated by unit rotation make pillar[n]arenes ideal platforms for investigating chirality inversion, induction, and transformation. In this review, we describe stimuli‐responsive topological, optical, chiroptical, supramolecular assemblies, and solid‐state materials based on the host–guest complexation and structural regulation of pillar[n]arenes.

150. Tuning of magnetosplamon coupling between graphene scatterers for the optimal design of adjustable metasurfaces

Abstract

The resonance characteristics of magnetically-biased graphene micro-scatterers are thoroughly investigated in the present work using both eigenvalue and full-wave solvers. Initially, the graphene surface conductivity is presented in a tensor form due to the application of a magnetostatic bias field, which is perpendicular to the material’s surface. Then, the simple case of a graphene disk scatterer is examined, and a properly modified eigenvalue formulation is utilized to extract the plasmonic fundamental frequencies. The validity of the modal analysis is verified via a full-wave analysis that involves a plane-wave propagation and the extraction of the subsequent absorption cross-section utilizing the Finite-Difference Time-Domain method. Additionally, the dependence of a single disk scatterer resonances with the magnetostatic bias is evaluated, highlighting that as the bias field is increased, every edge mode degenerates into two sub-modes with an augmented difference between the resonant frequencies. Finally, the plasmonic coupling between adjacent scatterers is studied considering a periodic arrangement, similar to a metasurface, indicating the additional coupling modes as well as the adjustability of the properties with multiple degrees of freedom.
