Vaccination generates a neutralizing immune response against SARS-CoV-2. The genomic surveillance is showing the emergence of variants with mutations in spike, the main target of neutralizing antibodies. To understand the impact of these variants, we report the neutralization potency against Alpha, Gamma, and D614G SARS-CoV-2 variants in 44 individuals that received two doses of CoronaVac vaccine, an inactivated SARS-CoV-2 vaccine. Plasma samples collected at 60 days after the second dose of CoronaVac were analyzed by the reduction of cytopathic effect in Vero E6 cells with the three infectious variants of SARS-CoV-2. Plasma showed lower neutralization with Alpha (geometric mean titer, GMT = 18.5) and Gamma (GMT = 10.0) variants than with D614G (GMT = 75.1) variant. Efficient neutralization against the Alpha and Gamma variants was not detected in 31.8% and 59.1% of plasma, respectively. These findings suggest the Alpha and Gamma variants could escape from neutralization by antibodies elicited by vaccination. Robust genomic and biological surveillance of viral variants could help to develop effective strategies for the control of SARS-CoV-2. This article is protected by copyright. All rights reserved.

Design of novel herbal drugs with a wide range of activity is an important object of pharmacology. Herbal remedies having antiarrhythmic, cardiotonic and vasodilatory effects are useful for treatment of heart arrhythmias. Vasodilating drugs are prescribed for medication of hypertension and migraine. Also they are a part of comprehensive treatment of various diseases such as disturbed peripheral blood circulation and atherosclerosis of arteries of extremities as well as problems with urination and potency. Search for novel herbal preparations with vasorelaxant activity in our view is a promising matter of current interest. This work is aiming in study of vasodilatory activity of methanol extract of strain K-27 tissue culture of Rauwolfia serpentina. Evaluation of vasodilatory activity extract of high-productive strain K-27 tissue culture of Rauwolfia serpentina has been carried out by the method of auxotonic mechanography of vascular smooth muscles. Strength and frequency of spontaneous contractions of portal vein as well as relaxation of aortic smooth muscles activated with phenylephrine were evaluation indicators of extract activity. In order to estimate working concentration of extract of strain K-27 tissue culture of Rauwolfia serpentina we have studied vascular activity at sequentially increasing concentrations of extract solution on the portal vein. Overlay of sequence of growing concentrations exhibited decrease in basal level of venous tone and dose-dependent suppression of phasic contractions with complete inhibition of spontaneous activity of portal vein at 1.44 mg/ml concentration and uprise of toxicity at higher concentrations. It has been shown dose-dependent relaxation of vascular preparations followed by development of α-adrenoceptor blocking effect and loss of sensibility to phenylephrine at the end of experiment. After discontinuing exposure with a substance the activating effect of phenylephrine remained not renewed during 30-50 minutes, at the same time contractile activity of aorta on the response to the other type activation (60 mM K+) took place. It was found that at the studied range of concentrations: 0.0288 μg/ml–28.8 μg/ml the extract of strain K-27 tissue culture of Rauwolfia serpentina has distinct vasodilatory effect. Also it was discovered α-adrenoceptor blocking effect of that extract, so it can find wide application in therapy as vasorelaxant and α-blocker, for instance in treatment of prostate diseases.

Des lors peut-etre apercevrait-on l’instantane d’un tableau plus vaste ; celui d’un essai bifide, temoin d’enjeux epistemologiques nouveaux, de constituer les bases de ce qu’on commence alors a peine a appeler une anthropologie.

The understanding of the host immune response to SARS-CoV-2 variants of concern is critical for improving diagnostics, therapy development, and vaccines. Here, we analyzed the level of neutralizing antibodies against SARS-CoV-2 D614G, Delta, Gamma, Mu, and Omicron variants in D614G infected healthcare workers during a follow-up up to 6 months after recovery. We followed up 76 patients: 60.5% were women and 39.5% men. The 96.1% and 3.9% were symptomatic and asymptomatic, respectively. The most frequent symptoms were headache, myalgia, and cough. The 65.8%, 65.8%, and 92.1% of the infected individuals were positive for neutralizing antibodies against D614G variant at 2, 4, and 6 months of follow-up, respectively. The 26.3%, 48.7% and 65.8% of patients neutralized Delta variant, 19.7%, 32.9% and 52.6% of patients neutralized Gamma, 7.9%, 19.7% and 44.7% of patients neutralized Mu, and 4.0%, 9.2% and 15.8% of patients neutralized Omicron. Low neutralization against Gamma and Mu variants was observed during the follow-up, and very low against the Omicron variant was detected during the same period. The median of neutralizing antibody titers against D614G and Delta variants increased significantly during the follow-up. An association was observed between the levels of neutralizing antibodies against D614G and Delta variants and the severity of the disease. Our results suggest an immune escape from neutralizing antibodies with the Omicron variant because of the many mutations localized in the S protein.

La prevalencia de infección por virus Papiloma Humano (VPH) es desconocida en niños, niñas y adolescentes (NNA) chilenos. En 2014 se incorporó en Chile la vacuna VPH al Programa Nacional de Inmunizaciones en niñas y en varones el 2019. Objetivo: Determinar la prevalencia, genotipos y características de la infección por VPH en NNA con lesiones anogenitales no vacunados contra VPH. Pacientes y Método: Se estudiaron NNA con lesiones anogenitales que consultaron en el Hospital Calvo Mackenna entre 2013 y 2017. Se registró motivo de consulta, edad, sexo, antecedentes de familiar portador de lesiones por VPH, antecedente de vulneración sexual, y actividad sexual consentida. Se detectó VPH por PCR y tipificación mediante hibridación reversa en línea del gen L1. Las muestras se analizaron en la Sección de Virus Oncogénicos del Instituto de Salud Pública. Resultados: Se estudiaron 110 pacientes; 44.5% niños/as. Se detectó VPH en 34 (30,9% [IC95% 22,4- 40,4]); 22 (44,9%) niños/as y 12 (19,7%) adolescentes. Once (91,7%) adolescentes tenían antecedente de contacto sexual (p < 0,005); 4 (18,2%) niños/as develaron vulneración sexual. 25% de las lesiones genitales y 50% de las perianales eran VPH (+) (p 0,015). Los genotipos más frecuentes fueron 59, 58, 16, 18, 6 y 11. En niños/as se detectó sólo genotipos de bajo riesgo. En 91.7% de las adolescentes VPH (+) se detectaron genotipos de alto riesgo. Conclusión: La prevalencia de infección por VPH fue 30%. En adolescentes la infección estuvo relacionada a contacto sexual y a VPH de alto riesgo. En niños/as se asoció a genotipos de bajo riesgo. Las lesiones perianales se asociaron con mayor frecuencia que las genitales a infección por VPH. La inspección visual no permite precisar la etiología de las lesiones genitales, por lo que es necesario realizar test de PCR para virus papiloma humano.
