The work we present in this article aims at optimizing and validating the assay technique based on SIDA-HSSPMEGC- MS/EI-SIM by means of intra-laboratory trials (NF V03-110 AFNOR 1998 and MA-F-AS1-06-PROVAL (OIV)) on several chloroanisoles (2,4,6-trichloroanisole, 2,3,4,6-tetrachloroanisole, 2,3,4,5,6-pentachloroanisole) likely to contaminate wine by direct or indirect contact, in order to define the precise characteristics and limitations of the method. We showed this technique to be a specific, accurate, linear, and repeatable method. Moreover, seeing the increasing reliance on this type of approach in scientific research and industrial monitoring laboratories (ISO/TC87/GT10) to assess the quality of cork stoppers and because of the important quantity of 2,4,6-Trichloroanisole present in this kind of material, we deemed appropriate to carry out an inter-laboratory study (ISO 5725-1,2,3,4 and 5) of TCA assays on dilute alcohol soaks of corks using the same method. This work was the first step of a study initiated by the FFT (Fédération Française de la Tonnellerie) to review the different aspects of the oak wood contamination, and especially contamination by chlorophenols and chloroanisoles. This paper deals with the precision of chlorophenols and chloroanisoles measurement in cooperage oak woods. The analytical methods used were those developed and single validated by the 4 laboratories that did this work. Material was obtained by adding substances to sawdust and making dilution between supplemented sawdust and blank one. Thus, material was homogenous and has a theoretic level. Recovery values for chlorophenols were between 70 % and 80 %, except one, for the lowest PCP's level (theoretic 7.5 ng/g), which was 95 %. For chloroanisoles, recovery values varied from 50 % for the highest levels to more than 100 % for the lowest levels. Measured reproducibility, expressed in variation coefficient of the standard deviation, was about 25 % for TCP and PCP, 45 % for TeCP, TCA and TeCA and more than 50 % for PCA. For TCP, PCP, TCA and TeCA, despite the use of different methods, these values are of the same order as those given by the Horwitz formula. These values may help to compare results and to build a decision rule of the conformity of the oak wood. The heterogeneousness of the material is another source of the final result variability. This last source was not quantified in this work.

The contamination of wines and, more rarely, of brandies by corks or cork-based stoppers used as bottles closures has been a recurring issue for many years. The relative frequency of this type of spoilage, characterized by the distinctive, unpleasant odor of «moldy cork taint», has led to a considerable body of research on this subject. In wine, 2,4,6-trichloroanisole (TCA), produced by the methylation of its direct precursor, 2,4,6-trichlorophenol (TCP), has clearly been identified as the main compound responsible for the "moldy off-flavors" produced by defective corks. In this study, we present a method for assaying the main chloroanisoles and chlorophenols identified as potential contaminants in wines and corks or cork-based stoppers (2,4,6-trichloroanisole and -phenol, 2,3,4,6- tetrachloroansiole and -phenol, pentachloroanisole and -phenol). An extraction method using dichloromethane for cork and a dichloromethane/-pentane mixture for wine isolates almost the entire quantity of organochlorine contaminants from these matrices. The chloroanisoles and precursors are identified simultaneously by gas-phase chromatography coupled with electron impact mass spectrometry with selective ion monitoring. This method provides an accurate analysis of the contamination with a view to establishing the type and origin of the "musty, moldy, corky" characteristics in bottled wines by studying the migration of contaminants from the stoppers into the contents of the bottles. This method for the simultaneous assay of chlorophenols and chloroanisoles was defined in compliance with the following standards: NF ISO 5725-1/2, NF V 03110 (AFNOR), and MA-F-AS1-06-PROVAL (OIV). The method is specific, accurate, linear, repeatable, and has been validated. The detection and quantification ranges are compatible with the sensitivity requirements for this type of contaminant.

This study is dedicated to quantification, by gas chromatography combined with mass spectrometry, of some volatile compounds founded during ageing of sweet natural wines. Besides determination of the thresholds olfactory perception enable us knowing better the more active compound in sweet natural wines aroma. The results enable to know better volatile compounds which are able to produce ageing aromas characteristic of sweet natural wines. These products are always rich in sugar and grown up in a more or less oxidising environment. Logically, many of the compounds identified in this work (derived furanic compounds and many others) are Iinked with chemical like Maillard reactions between sugars and amino-acids. Others compounds as a sotolon need oxidising phenomena to increase in the environment. Comparatively, oldest Port wines have been analysed. The compounds measured in these Port wines also exist in French sweet natural wines of type Banyuls and Rivesaltes, keeped under oxidising environment. But the dillerences of concentrations in some compounds discriminate them from the Port wines.

L'élevage des vins en barriques modifie profondément leur expression aromatique. L'étude par chromatographie en phase gazeuse et spectrométrie de masse permet d'identifier plusieurs substances volatiles appartenant à la fraction phénolique des arômes. Les vins rouges présentent naturellement une composition complexe en phénols volatils alors que celle des vins blancs est plus simple. L'élevage sous bois entraîne une augmentation notable des phénols déjà présents ainsi que l'apparition de molécules spécifiques au bois dechêne brûlé. L'interaction des levures et des bactéries avec le bois est mise en évidence. Les vins rouges se caractérisent par une présence parfois abondante d'éthyl phénols, les vins blancs qui ne subissent pas la fermentation malolactique s'en différencient par l'abondancede vinyl phénols. L'étude sensorielle de chaque substance permet de démontrer le rôle négligeable de certaines : furfural, méthyl-5-furfural, alcool furfurylique et le rôle exceptionnel joué par d'autres : cis et trans β-méthyl-γ-octalactone, vanilline et dans certains cas éthyl-4-phénol et éthyl-4-gaïacol. +++ Wood storage of wines changes profondly their aromatic expression. Several volatile substances from wines and oak woods phenolic fraction of aroma are identified by gas chromatography and mass spectrometry. Wood storage increases natural phenols concentration. Simultaneously specific burned wood molecules appear. Yeast and bacterial interaction with wood is demonstrated. Ethyl phenols are characteristic of red wines and vinyl phenols of white wines without malolactic fermentation. The sensorial analysis of each substance demonstrates the negligible intervention in wine aroma of furfural, 5-methyl-furfural, and furfuryl alcohol, and the important participation of cis and trans β-methyl-γ-octalactone, vanilin 4-ethyl-phenol and 4-ethyl gaïacol.

The aroma of the sweet fortified wines is often characterised by the « rancio » descriptor. According to the mode of conservation and the type of vine, the intensity of « rancio » character inay vary notably. The molecule responsible for this aroma in sweet fortified wines has been identified to the sotolon. According to its content, this molecule can influence differently the aroma of wines. Less than of 300 µg/1, sotolon takes part of « prune » aroma, whereas between 300 and 600 µg/1, it is responsible for the « dried prickly-pear, dried fruit » aroma. More than of 600 µg/1, the sweet natural wines are characterised by « rancio » character. An oxidising conservation is essential for high content of sotolon. An accidental oxidising during ageing in bottle, according for instance with a poor quality of the cork, may increase the formation of this compound in some sweet fortified wines. With the same âge and for the same level of the oxidation, the red sweet natural wines have always a lower sotolon content than the whites. The presence of polyphenolic compounds, by slowing down oxidising phenomena and by reducing the accumulation of ethanal essential for the sotolon formation, explains the lower content of this molecule in the red wines. The sotolon formation in the sweet natural wines has been studied through many experimentations on wines and model solutions at the laboratory and the winery scales. We show that the formation of sotolon during conservation and oxidising ageing of sweet natural wines essentially depends on chemical phenomena.

L'acétylcholinestérase (AChE, EC 3.1.1.7) est une enzyme indispensable de la transmission synaptique cholinergique. Son inhibition par les organophosphorés et les carbamates est à la base de certains traitements insecticides et nématicides utilisés en agriculture. Les faunes sauvage (en particulier les poissons) et domestique sont exposées aux résidus des traitements anticholinestérasiques. La mesure de l'inhibition de l'activité AChE chez ces espèces est un marqueur fiable de l'exposition. Ceci est montré ici pour l'AChE de carpes exposées expérimentalement au carbofuran (carbamate). Nous présentons parallèlement des données sur le gène de l'AChE chez Danio rerio et sur les formes moléculaires de l'enzyme chez Danio, Cyprinus carpio et Oncorhynchus mykiss. Contrairement à ce qui est observé chez d'autres vertébrés, le gène d'AChE de Danio ne possède pas d'exon H et seules les sous-unités de type T ont pu être identifiées chez ces trois espèces. Des résultats préliminaires d'hybridation in situ sur les embryons sont présentés et nous discutons l'intérêt des expériences de transgenèse dans l'oeuf de Danio pour étudier d'éventuelles fonctions morphogénétiques de l'AChE au début du développement.

This work is dedicated to the study of volatile compounds responsible for typical aroma of sweet natural wines, from non Muscat varieties, after ageing. The descriptive sensorial analysis and the modern technics of gas chromatography analysis combined with different detection modes (olfactory detection, and both mass and infrared spectrometry) are used for studying the relationships between the consensual descriptors of the aroma and the volatile substances identified in these wines. The relationships between the data (coming from the descriptive tasting and the volatile compounds and sweet-smelling) are studied thanks to correspondences factorial analysis and principal components analysis. Many substances take part in the characteristic odour of the natural sweet wines after ageing. Several volatile odorous compounds identified by the combination of gas chromatography with olfactory detection and mass spectrometry are signaled for the first time in sweet natural wines in this work. However, many of these substances were known in the aroma of several sweet products. Furaneol is always associated with « honey» aroma. Cyclotene (2-hydroxy-3-methyl-2-cyclopenten-I-one), maltol (3-hydroxy-2-methyl-4(H)-pyran-4-one), dihydromaltol (2,3-dihydro-5-hydroxy-6-methyl-4(H)-pyran-4-one), hydroxymaltol (3,5-dihydroxy-2-methyl- 4(H)-pyran-4-one) and DDMP (2,3-dihydro-3,5-dihydroxy-6-methyl-4(H)-pyran-4-one), already known for taking part in « toasty» aroma of heated wood, have been identified in old sweet natural wines extracts, which had never been conserved in a wood barrel. Homofuraneol (5(or 2)-ethyl-2(or 5)-methyl-4-hydroxy-3(2H)-furanone), characterised by« caramel» aroma, is found only in young red sweet natural wines. Sotolon (3-hydroxy-4,5-dimethyl-2(5H)-furanone) seems to be characteristic of white swect natural wines. The 5-ethoxymethylfurfural, characterised by a« white bread» aroma with « sharp » and « fruity » shades, have already been found in old red wines. The γ-octalactone and γ-nonalactone, characterised by a strong fruity odour of « coconut », « peach » and « almond », are al ways associated with the « dried-fruits » characteristic aroma of white and rather old sweet natural wines, Pantolactone (3,3-dimethyl-2- hydroxy-γ-butyrolactone) is a quantitatively major compounds in old sweet natural wines, associated in principal component analysis with « plum » aroma in the oldest sweet natural wines. The results enable to know better the volatile compounds which are able to produce ageing aromas charactcristic of sweet natural wines. These products are al ways rich in sugar and grown up in a more or less oxidising environ ment. Logically, many of the compounds identitied in this work (pyranones, furanones and derived esters) seem to be linked with chemicallike Maillard reactions between sugars and amino-acids. Others compounds (sotolon and γ-Iactones) need oxidising phenomena to increase during the ageing of the wines.

Thyroid hormone (T3) has a major influence on cerebellum post-natal development. The major phenotypic landmark of exposure to low levels of T3 during development (hypothyroidism) in the cerebellum is the retarded inward migration of the most numerous cell type, granular neurons. In order to identify the direct genetic regulation exerted by T3 on cerebellar neurons and their precursors, we used microarray RNA hybridization to perform a time course analysis of T3 induced gene expression in primary cultures of cerebellar neuronal cell. These experiments suggest that we identified a small set of genes which are directly regulated, both in vivo and in vitro, during cerebellum post-natal development. These modest changes suggest that T3 does not acts directly on granular neurons and mainly indirectly influences the cellular interactions taking place during development.

Pendant le séchage naturel à l'air libre du bois de chêne, on assiste à une diminution régulière de la teneur du bois en composés extractibles et notamment en composés phénoliques du type ellagitanins. La diminution de la teneur en ellagitanins provient essentiellement de la disparition des formes oligomères solubles dans l'eau; les tanins polymérisés non extractibles car liés aux parois, évoluent peu au çours du séchage naturel à l'air libre. Les bois séchés rapidement à l'étuve possèdent toujours des quantités plus importantes de polyphénols. Dans le même temps et quel que soit le mode de séchage, on obselVe une diminution des coumarines glucosilées et une augmentation non proportionnelle des formes aglycones. D'un point de vue organoleptique, la diminution de l'ensemble des polyphénols pendant le séchage peut être interprétée comme favorable à l'évolution de la qualité du merrain. En effet, les fractions amères et astringentes du bois de chêne correspondent respectivement .sr certains tanins ellagiques et catéchiques. L'intelVention des micro-organismes isolés de bois de chêne en cours de séchage naturel dans la disparition des composés phénoliques du bois est discutée. La grande majorité des moisissures étudiées est capable de dégrader enzymatiquement les tanins hydrolysables du chêne. Cependant, ces mêmes composés s'hydrolysent et s'oxydent spontanément en présence d'eau et d'oxygène. En conséquence, considérant le profil de colonisation du bois par les micro-organismes et celui des composés phénoliques extractibles, on peut supposer que l'on assiste vraisemblablement pendant le séchage naturel du bois à une dégradation biochimique et chimique des polyphénols dans les couches superficielles du merrain (0 à 4 mm de profondeur) et à une dégradation presque' exclusivement chimique au coeur du merrain. La présence d'eau et d'oxygène apparaÎt par conséquent comme un facteur important dans l'évolution de la composition du bois pendant son séchage.

Pendant le séchage des merrains sur parc à l'air libre, on assiste à une profonde modification de la composition chimique et du profil aromatique du bois de chêne. La teneur en eugénol, en aldéhydes phénols et en Β-méthyl-y-octalactone augmente régulièrement pendant le vieillissement du bois. Principalement sous forme trans au début du séchage, la Β-méthyl-y-octalactone s'accumule finalement sous la forme cis plus aromatique. Dans le même temps, on observe également une accumulation importante du précurseur inodore de Β-méthyl-y-octalactone. L'intervention des micro-organismes fréquemment isolés sur le bois en cours de séchage naturel sur la formation de composés aromatiques a été étudiée. La culture de diverses moisissures isolées de bois en cours de séchage naturel sur extraits de bois et directement sur sciure de chêne aboutit dans tous les cas à une forte diminution des teneurs en aldéhydes phénols, notamment de la vanilline. En effet, tous les micro-organismes étudiés (Penicilium sp., Trichoderma sp., Aureobasidium sp.) sont incapables de dégrader la lignine car ils ne possèdent pas d'activité ligninase. En revanche, ces champignons possèdent une forte activité oxydo-réductase capable de réduire la vanilline en alcool vanillique inodore. En conséquence, les aldéhydes phénols apparaissant au cours du séchage naturel du merrain ne dérivent pas d'une attaque enzymatique mais plus vraisemblablementde l'acidolyse et de l'oxydation chimique de la lignine du bois. Le stockage du bois à l'air libre en présence d'eau et d'oxygène doit permettre une évolution favorable de son potentiel aromatique tant que la vanilline et la cis Β-méthyly-octalactone s'accumulent.

Pendant le séchage naturel et le vieillissement à l'air libre du bois de chêne débité en merrain, on assiste à l'envahissement progressif du bois par différents micro-organismes. On a identifié dans ce travail 19 genres et 27 espèces de champignons, 2 genres et au moins 4 espèces de bactéries. La très grande majorité des micro-organismes est localisée dans les couches superficielles du bois. Seuls certains champignons sont retrouvés au coeur du merrain ayant vieilli 5 ans et plus sur parc. La déshydratation rapide du bois semble limiter le développement et l'envahissement en profondeur par les moisissures. Un champignon basidiomycète à arthroconidies, non formellement identifié, qui produit une pourriture blanche, a été isolé dans des merrains stockés 10 ans à l'air libre. Excepté ce champignon particulier, qui grâce à un équipement enzymatique adapté est susceptible d'attaquer simultanément tous les constituants pariétaux, les autres micro-organismes identifiés sont essentiellement cellulolytiques et ne peuvent pas altérer profondément la composition ni la structure du bois. En revanche, ils devraient pouvoir augmenter notablement la perméabilité et l'accessiblité aux liquides des couches superficielles du merrain et faciliter l'implantation ultérieure de basidiomycètes lignolytiques.

Les goûts et odeurs à caractère "moisI" retrouvés dans les vins encore stockés en cuves ou en barriques sont généralement dus à des chloroanisoles du type 2,3,4,6-tétrachloroanisoleet pentachloroanisole. Ces composés malodorants proviennent de la dégradation microbienne de chlorophénols utilisés comme traitement insecticide de différents matériaux à base de bois principalement employés dans la construction des bâtiments, leur isolation ou dans la fabrication de caisses de stockage des bouteilles. Dans certaines conditions, humidité de l'air élevée et ventilation réduite, on assiste à une contamination rapide de l'atmosphère des caves et des vins qui y sont entreposés. De la même façon, la conservation dans une atmosphère viciée de certains produits destinés à entrer en contact avec le vin peut être responsable de sa pollution. L'élimination de la source de contamination en chlorophénols et en chloroanisoles, ainsi que l'amélioration du renouvellement de l'air, suffisent généralement à solutionner le problème. La prohibition de tous les produits de traitements à base de chlorophénols est conseillée dans tous les locaux destinés à recevoir du vin en vrac où à le conditionner en bouteille. La cuverie contaminée en même temps que les vins peut être réutilisée après nettoyage et éventuellement réfection. A l'inverse, les contenants en bois conservés dans une atmosphère polluée doivent le plus souvent être éliminés.
