Dünyanın birçok bölgesinden ülkemize yükseköğrenim görmek amacıyla gelen ve son yıllarda giderek sayısı artan yabancı uyruklu öğrenciler vardır. Bu öğrenciler öğrenimleri sürecinde ülkemizde eğitimsel, dilsel, ekonomik, psikolojik, sosyal ve kültürel farklılıklala dolu yeni bir kültürel iklimde yaşamaya başlamaktadır. Gelen öğrenciler toplum yaşamıyla ilgili farklı bir ortamla, kurallarla, yemek kültürü, giyim alışkanlıkları, dini inançları, eğlence alışkanlıkları vb. karşılaşmaktadır. Farklı ülkelerden öğrencilerin gelmesiyle kültürel iklimin sahipleri de gelen misafirlerle yaşamaya başlamaktadır.Araştırma yabancı uyruklu bir öğrencinin Türkiye'de okumayı seçmeden önce ülkesindeki durumundan başlayarak ülkemizde geçirdiği süre boyunca ne düşündüğü, neler deneyimlediği, Türk arkadaşlarıyla olan ilişkileri ve bu süre içerisinde Türk öğrencilerin ne düşündükleri, neler deneyimledikleri hakkında bilgi vermektedir. Alanyazın taraması sonucu yabancı uyruklu öğrenciler ve Türk öğrencilerin birbirleri hakkındaki algılarını karşılaştırmalı inceleyen bir araştırma olmadığı görülmüştür.Bu nedenle Sakarya Üniversitesi'nde okuyan 20 Türk ve 17 farklı ülkeden 20 yabancı uyruklu öğrenci olmak üzere toplamda 40 öğrenci ile mülakat yapılmıştır. Araştırmada öncelikle yabancı uyruklu öğrenciler ile Türk öğrencilerin sınıf ortamındaki ilişkileri incelenmiştir. İkinci olarak yabancı uyruklu öğrenciler ile Türk öğrencilerin sosyal ilişkileri incelenmiştir. Üçüncü olarak yabancı uyruklu öğrenciler ile Türk öğrencilerin toplumsal ve kültürel ilişkileri incelenmiştir. Meme kanseri kadınlarda kanser kaynaklı ölümler arasında ikinci sırada yer alan ve büyüme faktörleri, hormonal düzenleme ile gelişimi seyreden bir kanserdir. Yakın zamanda, büyüme hormonunun (BH) meme kanseri hücrelerinde anlatımının arttırılmasının hücre büyüme, invazyon-metastaz ve ilaç direncine neden olduğu tespit edilmiştir. BH kaynaklı ilaç direncinde bitkisel kökenli curcuminin etkisi zamana ve doza bağlı olarak gösterilmiştir. Ancak curcuminin terapötik potansiyelinin arttırılması ve apoptotik etkinliğinin moleküler mekanizmasında ER stres ve otofajinin rolü bilinmemektedir. Bu tez ile amacımız MDA-MB-231 ve T47D doğal tip ve BH+ meme kanseri hücreleri üzerine zamana bağlı curcuminin etkisinin yanında otolizozom inhibitörü olan bafilomisinle kombine tedavisinde apoptotik etkinliğinin ER ve otofaji yolakları göz önünde bulundurularak irdelenmiştir. Zamana bağlı curcumin uygulamasının otokrin BH sinyali aracılı direnç mekanizmasının otofajik ve ER stres anahtar molekülleri üzerinden gerçekleştirdiği MDA-MB-231 ve T47D meme kanseri hücrelerinde gösterilmiştir. Curcumin direncinin kırılmasına bağlı terapötik etkinliğin irdelenmesi için bafilomisin ile curcuminin beraber uygulamasının ER stres anahtar moleküllerinin anlatımını baskılayarak ve otofaji sürecinde geç aşama olan otolizozomu engelleyerek apoptotik hücre ölümünü indüklediği MDA-MB-231 ve T47D meme kanseri hücrelerinde belirlenmiştir. Böylece bu tez ile ilk defa otokrin BH sinyali kaynaklı curcumin ilaç direnç mekanizmasının moleküler mekanizmasında ER stres ve otofajinin rolünün aydınlatılması yanında curcuminle beraber bafilomisin uygulamasının curcumine bağlı apoptotik hücre ölümünü indüklediği MDA-MB-231 ve T47D hücrelerinde tespit edilmiştir. Breast cancer is the second most common cancer among cancer-related deaths in women and growth factors and hormonal regulation are involved in breast cancer progression. Recently, it has been found that increasing the expression of growth hormone (GH) in breast cancer cells triggers cell growth, invasion-metastasis and drug resistance. Dose- and time-dependent effect of curcumin (plant-derived compounds) on autocrine GH signaling breast cancer has been reported. However, the molecular machinary of curcumin induced apoptotic cell death regarding the role of ER stress and autophagy pathway has not been demonstrated yet. In this thesis, our aim is to investigate the time dependent effect of curcumin on MDA-MB-231 and T47D wild type and GH+ breast cancer cells and also evaluate the potential additional effect of bafilomycin (autolysosome inhibitör) on autocrine GH mediated curcumin resistance through ER stress and autophagy pathways. The inhibitive effect of time-dependent curcumin exposure on the MDA-MB-231 and T47D breast cancer cells was demonstrated and this might be related with curcumin triggered by key molecules of autophagic and ER stress signaling pathway. Moreover, combination of bafilomycin and curcumin treatment overcome autocrine GH mediated curcumin resistance was determined in MDA-MB-231 and T47D breast cancer cells via inhibiting the expression profile of ER stress key molecules. In additon, we also demonstrate that bafilomycin co-treatment accelerated curcumin-mediated apoptotic cell death through inhibiting breast cancer cells at late stage autolysosome formation. Thus, in this thesis, the role of ER stress and autophagy has been determined in autocrine GH mediated curcumin resistance in MDA-MB-231 and T47D cells and also additional apoptotic effect of bafilomycin in therapeutic potential of curcumin in vitro breast cancer drug resistance breast cancer model.

Buaraştırmanın amacı, yabancı uyruklu öğrenciler ile Türk öğrencilerin sınıfortamında birbirlerine yönelik algılarının belirlenmesidir. Araştırmada nitel araştırmayöntemi seçilmiş, fenomenoloji deseni kullanılmış, yarı yapılandırılmış mülakatformu ile sahadan veriler toplanarak içerik analizi yapılmıştır. Araştırmanınörneklemi 20 yabancı uyruklu öğrenci ve 20 Türk öğrenciden oluşmaktadır. Farklıülkelerden gelen öğrenciler ile Türk öğrencilerin sınıf ilişkileri, dersçalışma alışkanlıkları, sınıf ortamında iletişimleri gibi durumlarkarşılaştırılmıştır. Görüşmeler neticesinde, Türk öğrenciler yabancı uyrukluöğrencileri çalışkan bulmuş; ders çalışma alışkanlıklarının günlük tekrar,kütüphaneye gitme, araştırma yapma olduğu görülmüş; sınıf ortamında kültürelfarklılık kaynaklı sorun yaşamadıkları, ilişkilerinin olumlu olduğu tespitedilmiştir. Yabancı uyruklu öğrenciler, Türk öğrencilerin ders çalışmalarınıyetersiz, ezberci bulmuş, sınıf ilişkilerini olumlu değerlendirmiştir., Thepurpose of this study is to determine the perceptions of foreign students andTurkish students about each other in the classroom environment. In the study,qualitative research method was chosen, phenomenology design was used, data werecollected from the field with a semi-structured interview form and contentanalysis was performed. The sample of the study consists of 20 foreign studentsand 20 Turkish students. The students from different countries and the Turkishstudents were compared in terms of classroom relations, studying habits andcommunication in the classroom environment. As a result of the interviews, theTurkish students found the foreign students more hard working and it was seenthat their studying habits were daily repetitions, going to the library anddoing research. It was also determined that they did not have any problemsarising from cultural differences in the classroom environment and they hadpositive relations. The foreign studentsfound the studying habits of the Turkish students inadequate andmemorization-based and assessed their classroom relations positively.

Bu doktora tez çalışmasında birçok tekstil prosesinin kalite ve randımanı için birinci derecede öneme sahip olan bobinden iplik sağımı esnasında bobin çapındaki değişmeye bağlı olarak iplik gerginliğindeki değişme deneysel olarak araştırılmıştır. Bu kapsamda araştırma için endüstriyel uygulamaları temsil eden deneysel bir araştırma düzeneği geliştirilmiştir. Deneysel araştırma düzeneği ipliğin istenilen hızda bobinden sağılmasını sağlayan iki ünitelik bir bobin makinesi, tek bobinlik bir cağlık ünitesi, bobin çapı ölçme sensörü, iplik gerginlik ölçme sensörleri ile bir adet kişisel bilgisayar ve ara birim ünitesinden oluşmaktadır. C programlama dilinde geliştirilen bir yazılım ile eş zamanlı olarak iplik gerginliği ve bobin çapı bilgisayar tarafından okunup hafızaya veri dosyası olarak kaydedilmiştir. Daha sonra işlenen veriler kullanılarak iplik gerginliğinin ve bobin çapının kayıt esnasındaki ortalama değeri hesaplanmış ve bu şekilde iplik gerginliği-bobin çapı ilişkisi elde edilmiştir. Deneysel çalışmalarda silindirik bobinlere sarılı halde 50 denye ve 900 denye arasında değişen 7 farklı numarada bükümlü ve puntalı poliester iplikler ile silindirik ve konik bobinlere sarılı Ne 50/1 ve Ne 10/1 arasında değişen 5 farklı numarada pamuk ipliği kullanılmıştır. Deneysel çalışmalar esnasında ipliğin kalınlığına bağlı olarak en az 8 farklı çap değerinden gerginlik ölçümleri alınarak iplik gerginliği-bobin çapı ilişkisinin dolu çaptan boş çapa kadar yeterli hassasiyette elde edilmesi hedeflenmiştir. İplik gerginliği-bobin çapı ilişkisi her iplik için 100 m/dak, 200 m/dak, 400 m/dak, 600 m/dak ve 800 m/dak sağım hızlarında ve 120 mm, 240 mm ve 480 mm'lik bobin ön yüzeyi-iplik kılavuzu mesafelerinde elde edilmiştir. Ayrıca kaba, kademeli hassas ve 2 farklı sarım sayısına sahip sarım tiplerinin iplik gerginliği-bobin çapı ilişkisine etkisi araştırılmıştır. Yapılan deneysel çalışmalar sonucunda gerek sürekli filaman poliester iplikler için gerekse pamuk iplikler için birçok endüstriyel uygulamayı kapsayacak şekilde iplik gerginliği-bobin çapı ilişkileri elde edilmiş ve değişik parametrelerin bu ilişki üzerindeki etkisi değerlendirilmiştir. In this study yarn tension change with respect to bobbin diameter during unwinding, which is of prime importance for quality and efficiency of many textile processes is investigated experimentally. For this aim, an experimental research set-up was established representing unwinding from bobbins in many textile processes. Experimental research set-up consists of a winding machine with 2 winding units, a creel with single bobbin holder, a laser sensor measuring bobbin diameter, tension sensors for measuring yarn tension, a PC and a DAQ card. A software program was developed in C programming language to read the tension and bobbin diameter simultaneously and record them as a data file. Based on the recorded data average values of warp tension and bobbin diameter were calculated and relationship between yarn tension and bobbin diameter was obtained in this way. Continuous filament polyester (twisted and intermingled) yarn of 7 different numbers changing from 50 denier to 900 denier wound on cylindrical bobbins and cone and cylindrical bobbins with cotton yarns with 5 different numbers changing from Ne 50/1 to Ne 10/1 were employed in the experiments. Tension and diameter data were recorded from at least 8 different diameters from full to empty bobbin during experiments depending on the yarn thickness for the required precision in determining the relationship between yarn tension and bobbin diameter. For each yarn or bobbin, experiments were carried out with 3 different bobbin front surface-yarn guide distances (120 mm, 240 mm, 480 mm) and 5 different unwinding speeds (100 m/min, 200 m/min, 400 m/min, 600 m/min and 800 m/min). In addition, the effect of winding types (random winding, step precision winding and precision winding for 2 different winding ratios) on the relationship between the yarn tension and bobbin dimeter was investigated. Relationships between yarn tension and bobbin diameter were obtained for different yarn types and numbers covering wide range of industrial applications and parameters used in these industrial applications. Introductory Chapter: New Age Molecular Techniques in Plant Science

Bu çalışma, YükseköğretimdeÖğretimsel Algı ve Uygulamalar Ölçeği’ni Türkçe’ye uyarlamayı amaçlamaktadır.İki ölçekten oluşan ölçme aracı toplam 152 öğretim elemanına uygulanmıştır. İlkolarak, Yükseköğretimde Öğretimsel Algı Ölçeği için yapılan doğrulayıcı faktöranalizi sonuçları, modelin veriye yeterli düzeyde uyum sergilemediğine işaretetmiştir. Bu nedenle, veri açımlayıcı faktör analizi yöntemiyle yeniden analizedilmiştir. Ölçümlere ilişkin hesaplanan iç tutarlılık katsayısı .90 olarakbulunmuştur. Ölçme aracının ikinci bölümünü oluşturan YükseköğretimdeÖğretimsel Uygulamalar Ölçeği’nden elde edilen ölçümler üzerinde yapılandoğrulayıcı faktör analizi sonuçları, modelin veriye yeterli düzeyde uyumsergilediğine işaret etmiştir. Ölçümlere ilişkin elde edilen iç tutarlılıkkatsayısı .93 olarak bulunmuştur. Sonuç olarak, elde edilen bulgular ölçmearacının öğretmen yetiştirmede Türk öğretim elemanlarının algı veuygulamalarını belirlemede geçerli ve güvenilir biçimde kullanılabileceğiniortaya koymuştur., Thisstudy aimed to adapt the scale of Instructional Perceptions and Practices inHigher Education into Turkish. The data collection tool, composed of two parts,was conducted on 152 teacher educators. Firstly, the findings of theconfirmatory factor analysis for the scale of Instructional Perceptions inHigher Education indicated that the model did fit the data adequately. Therefore,the data was reanalyzed via exploratory factor analysis method. Internalconsistency of the measurements was found to be .90. Confirmatory factoranalysis, performed on the measurements obtained from the scale ofInstructional Practices in Higher Education, constituting the second part ofthe data collection tool showed that the model fitted the data adequately. Internalconsistency of the measurements was found to be .93. As a result, findingsshowed that the scale might be used in a valid and reliable way to determinethe perceptions and practices of Turkish lecturers in teacher education.

As a predicted result of increasing population worldwide, improvements in the breeding strategies in agriculture are valued as mandatory. The natural resources are limited, and due to the natural disasters like sudden and severe abiotic stress factors, excessive floods, etc., the production capacities are changed per year. In contrast, the yield potential should be significantly increased to cope with this problem. Despite rich genetic diversity, manipulation of the cultivars through alternative techniques such as mutation breeding becomes important. Radiation is proven as an effective method as a unique method to increase the genetic variability of the species. Gamma radiation is the most preferred physical mutagen by plant breeders. Several mutant varieties have been successfully introduced into commercial production by this method. Combinational use of in vitro tissue culture and mutation breeding methods makes a significant contribution to improve new crops. Large populations and the target mutations can be easily screened and identified by new methods. Marker assisted selection and advanced techniques such as microarray, next generation sequencing methods to detect a specific mutant in a large population will help to the plant breeders to use ionizing radiation efficiently in breeding programs.

Most of the local strawberry (Fragaria x ananassa Duchenese) cultivars of Turkey are not phylogenetically classified. This present study, which aimed discriminating the genetical relationships between the Ottoman strawberry cultivar and other early-period strawberry cultivars planted in Istanbul and Karadeniz Ereğli regions of Turkey, is the first on this field. Seven strawberry cultivars were analyzed by using RAPD. 10 random primers carried out DNA fingerprinting analyses of these cultivars. The average polymorphism rate was determined as 87.1%. The most polymorphic primer was OPH01 which produced 13 bands. The cluster dendrogram presented that the similarity coefficients were between range of 0.03 and 0.73. Ottoman cultivar showed higher similarity with Tüylü more than Kara cultivar which is reported as the mother cultivar. RAPD method was sufficient to assess the phylogenetic relationship between Ottoman cultivar and other early-period strawberry cultivars. The data of this study brought forward the necessity of further genetic analysis to prove the phylogenetic relationship among Tüylü and Ottoman cultivars.
