Ed. 39/2024)" published in Angewandte Chemie International Edition, Xu-Man Chen, Hong Yang, Quan Li, and their colleagues discuss their research on supramolecular hetero-self-assembly. They present a cucurbit[8]uril-mediated heterodimerization that leads to a rare cucurbit[8]uril-mediated [2+2] heterocycloaddition under UV irradiation. Understanding protein folding under conditions similar to those found in vivo remains challenging. Folding occurs mainly vectorially as a polypeptide emerges from the ribosome or from a membrane translocon. Protein folding during membrane translocation is particularly difficult to study. Here, we describe a single-molecule method to characterize the folded state of individual proteins after membrane translocation, by monitoring the ionic current passing through the pore. We tag both N and C termini of a model protein, thioredoxin, with biotinylated oligonucleotides. Under an electric potential, one of the oligonucleotides is pulled through a α-hemolysin nanopore driving the unfolding and translocation of the protein. We trap the protein in the nanopore as a rotaxane-like complex using streptavidin stoppers. The protein is subjected to cycles of unfolding-translocation-refolding switching the voltage polarity. We find that the refolding pathway after translocation is slower than in bulk solution due to the existence of kinetic traps.

The interlocking of ring and axle molecular components in rotaxanes provides a way to combine chromophoric, electron-donor and electron-acceptor moieties in the same molecular entity, in order to reproduce the features of photosynthetic reaction centers. To this aim, the photoinduced electron transfer processes involving a 1,8-naphthalimide chromophore, embedded in several rotaxane-based dyads, were investigated by steady-state and time-resolved absorption and luminescence spectroscopic experiments in the 300 fs-10 ns time window. Different rotaxanes built around the dialkylammonium/ dibenzo[24]crown-8 ether supramolecular motif were designed and synthesized to decipher the relevance of key structural factors, such as the chemical deactivation of the ammonium-crown ether recognition, the presence of a secondary site for the ring along the axle, and the covalent functionalization of the macrocycle with a phenothiazine electron donor. Here is reported the synthesis and study of dibenzo‐24‐crown‐8 (DB24C8)‐based [2]‐ and [3]rotaxanes that contain an ammonium as the best molecular station and carbamate moieties as secondary molecular stations. The common post‐interlocking synthesis relies on the aminolysis of the N‐succinimidyl carbonate extremity of an activated though insulated pseudorotaxane. The N‐succinimidyl carbonate‐based thread's extremity proved to be small enough to allow the slow slippage of the DB24C8 around the thread and large enough to allow insulation of the kinetically stable pseudorotaxane. Due to its sensitivity towards amine compounds, the activated carbonate end of the encircled thread allowed post‐interlocking aminolysis‐based conversion into mechanically interlocked rotaxanes by providing the same way the carbamate secondary station for the DB24C8 in the thread backbone. The catechol moiety found within mussel proteins plays a pivotal role in enhancing their adhesive properties. Nonetheless, catechol compounds, such as dopamine (DOP) derivatives, are susceptible to oxidation, leading to the formation of quinone. This oxidation process poses a significant challenge in the development of DOP-based hydrogels, hampering their adhesion capabilities and hindering polymerization. To protect DOP moieties from oxidation, DOP and N-(3-aminopropyl)methacrylamide (AMA) moieties were grafted onto the side groups of biocompatible poly(glutamic acid) (PGA). Subsequently, the DOP unit, serving as a second guest, would be captured by a polymerizable rotaxane of cucurbituril (CB[n]), in which the host molecule CB[8] complexed with the first guest, polymerizable methyl viologen (MV), forming a protective function and dynamic cross-linking. Upon exposure to light curing, a composite network emerged through the synergy of covalent cross-linking and supramolecular host-guest complexation of DOP with CB[8]. The generated complexation between DOP and CB[8] could protect the DOP moieties, resulting in photocured hydrogels with exceptional adhesive strength and remarkable tensile capabilities. Encapsulation of active pharmaceutical ingredients (APIs) in confined spaces has been extensively explored as it dramatically alters the molecular dynamics and physical properties of the API. Herein, we explored the effect of encapsulation on the molecular dynamics and physical stability of a guest drug, salicylic acid (SA), confined in the intermolecular spaces of γ-cyclodextrin (γ-CD) and poly(ethylene glycol) (PEG)-based polypseudorotaxane (PPRX) structure. The sublimation tendency of SA encapsulated in three polymorphic forms of the γ-CD/PEG-based PPRX complex, monoclinic columnar (MC), hexagonal columnar (HC), and tetragonal columnar (TC), was investigated. The SA sublimation rate was decreased by 3.0-6.6-fold and varied in the order of MC form > HC form > TC form complex. The 13 C and 1 H magic-angle spinning (MAS) solid-state nuclear magnetic resonance (NMR) spectra and 13 C spin-lattice relaxation time ( T 1 ) indicated that the encapsulated SA molecules existed as the monomeric form, and its molecular mobility increased in the order of MC form > HC form > TC form complex. In the complexes, a rapid chemical exchange between two dynamic states of SA (free and bound) was suggested, with varying adsorption/desorption rates accounting for its distinct molecular mobility. This adsorption/desorption process was influenced by proton exchange at the interaction site and interaction strength of SA in the complexes, as evidenced by 1 H MAS spectra and temperature dependency of the 13 C carbonyl chemical shift. A positive correlation between the molecular mobility of SA and its sublimation rate was established. Moreover, the molecular mobility of γ-CD and PEG in the complexes coincided with that of SA, which can be explained by fast guest-driven dynamics. This is the first report on the stability improvement of an API through complexation in polymorphic supramolecular host structures. The relationship between the molecular dynamics and physical properties of encapsulated API will aid in the rational design of drug delivery systems.

A biphenyl-23-crown-7 ether (BP23C7) is synthesized in 86% yield from commercially available starting materials. BP23C7 forms pseudo[2]rotaxane with a dibenzylammonium ion (DBA + ), exhibiting a good association constant value ( k a = 1 × 10 3 ). Subsequently, fluorophoric properties of BP23C7 and anthracene terminated axles are blended to create responsive pseudo[2]rotaxanes. The "turn-on" fluorescence response of BP23C7 due to the addition of fluoride and chloride anions to pseudo[2]rotaxane systems has been investigated. Concomitant fluorescence quenching of the anthracene moiety of corresponding axles due to ion-pair formation has been addressed. Furthermore, two variants of [23]crown ethers, -1 ). Subsequently, fluorophoric properties of BP23C7 and anthracene terminated axles are blended to create responsive pseudo[2]rotaxanes. The "turn-on" fluorescence response of BP23C7 due to the addition of fluoride and chloride anions to pseudo[2]rotaxane systems has been investigated. Concomitant fluorescence quenching of the anthracene moiety of corresponding axles due to ion-pair formation has been addressed. Furthermore, two variants of [23]crown ethers, i.e. BP23C7 and o -xylene-23-crown-7 ether (X23C7), are applied for constructing homo[3]rotaxane architectures. A half-axle comprising of DBA + moiety and a terminal olefin is mixed separately with two [23]crown ethers and subjected to self-metathesis using Grubbs' first-generation catalyst.

Hyaline cartilage, a soft tissue enriched with a dynamic extracellular matrix, manifests as a supramolecular system within load-bearing joints. At the same time, the challenge of cartilage repair through tissue engineering lies in replicating intricate cellular-matrix interactions. This study attempts to investigate chondrocyte responses within double-network supramolecular hybrid hydrogels tailored to mimic the dynamic molecular nature of hyaline cartilage. To this end, we infused noncovalent host-guest polyrotaxanes, by blending α-cyclodextrins as host molecules and polyethylene glycol as guests, into a gelatin-based covalent matrix, thereby enhancing its dynamic characteristics. Subsequently, chondrocytes were seeded into these hydrogels to systematically probe the effects of two concentrations of the introduced polyrotaxanes (instilling different levels of supramolecular dynamism in the hydrogel systems) on the cellular responsiveness. Our findings unveiled an augmented level of cellular mechanosensitivity for supramolecular hydrogels compared to pure covalent-based systems. This is demonstrated by an increased mRNA expression of ion channels (TREK1, TRPV4, and PIEZO1), signaling molecules (SOX9) and matrix-remodeling enzymes (LOXL2). Such outcomes were further elevated upon external application of biomimetic thermomechanical loading, which brought a stark increase in the accumulation of sulfated glycosaminoglycans and collagen. Overall, we found that matrix adaptability plays a pivotal role in modulating chondrocyte responses within double-network supramolecular hydrogels. These findings hold the potential for advancing cartilage engineering within load-bearing joints.

Stroke is the primary cause of disability without effective rehabilitation methods. Emerging brain-machine interfaces offer promise for regulating brain neural circuits and promoting the recovery of brain function disorders. Implantable probes play key roles in brain-machine interfaces, which are subject to two irreconcilable tradeoffs between conductivity and modulus match/transparency. In this work, mechanically interlocked polyrotaxane is incorporated into topological hydrogels to solve the two tradeoffs at the molecular level through the pulley effect of polyrotaxane. The unique performance of the topological hydrogels enables them to acquire brain neural information and conduct neuromodulation. The probe is capable of continuously recording local field potentials for eight weeks. Optogenetic neuromodulation in the primary motor cortex to regulate brain neural circuits and control limb behavior is realized using the probe. Most importantly, optogenetic neuromodulation is conducted using the probe, which effectively reduces the infarct regions of the brain tissue and promotes locomotor function recovery. Exceptionally strong halogen bonding (XB) donor‐chloride interactions are exploited for the chloride anion template synthesis of neutral XB [2]rotaxane host systems which contain perfluoroaryl‐functionalised axle components, including a remarkably potent novel 4,6‐dinitro‐1,3‐bis‐iodotriazole motif. Halide anion recognition properties in aqueous‐organic media, determined via extensive 1H NMR halide anion titration experiments, reveal the rotaxane host systems exhibit dramatically enhanced affinities for hydrophilic Cl− and Br−, but conversely diminished affinities for hydrophobic I−, relative to their non‐interlocked axle counterparts. Crucially, this mechanical bond effect induces a binding selectivity which directly opposes Hofmeister bias. Free‐energy analysis of this mechanical bond enhancement demonstrates anion recognition by neutral XB interlocked host systems as a rare and general strategy to engineer anti‐Hofmeister bias anion selectivity in synthetic receptor design. New strategies for synthesizing polyyne polyrotaxanes are being developed as an approach to stable carbyne "insulated molecular wires". Here we report an active metal template route to polyyne [3]rotaxanes, using dicobalt carbonyl masked alkyne equivalents. We synthesized two [3]rotaxanes, both with the same C28 polyyne dumbbell component, one with a phenanthroline‐based macrocycle and one using a 2,6‐pyridyl cycloparaphenylene nanohoop. The thermal stabilities of the two rotaxanes were compared with that of the naked polyyne dumbbell in decalin at 80 °C, and the nanohoop rotaxane was found to be 4.5 times more stable. A review of heteroditopic interlocked molecules and their application as receptors for simple inorganic ion pair species. The review details the design and ion recognition properties of the rotaxane and catenane receptors, as well as highlighting some of the experimental challenges; hence, it provides insight into possible future avenues of research in this youthful field.

Crown ethers are common building blocks in supramolecular chemistry and are frequently applied as cation sensors or as subunits in synthetic molecular machines. Developing switchable and specifically designed crown ethers enables the implementation of function into molecular assemblies. Seven tailor-made redox-active crown ethers incorporating tetrathiafulvalene (TTF) or naphthalene diimide (NDI) as redox-switchable building blocks are described with regard to their potential to form redox-switchable rotaxanes. A combination of isothermal titration calorimetry and voltammetric techniques reveals correlations between the binding energies and redox-switching properties of the corresponding pseudorotaxanes with secondary ammonium ions. For two different weakly coordinating anions, a surprising relation between the enthalpic and entropic binding contributions of the pseudorotaxanes was discovered. These findings were applied to the synthesis of an NDI-[2]rotaxane, which retains similar spectroelectrochemical properties compared to the corresponding free macrocycle. The detailed understanding of the thermodynamic and electrochemical properties of the tailor-made crown ethers lays the foundation for the construction of new types of molecular redox switches with emergent properties.

Abstract Rotaxane building blocks bearing 3,5‐bis(trifluoromethyl) benzenesulfonate (BTBS) stoppers have been efficiently prepared from a pillar[5]arene derivative, 3,5‐bis(trifluoromethyl) benzenesulfonyl chloride (BTBSCl) and different diols, namely 1,10‐decanediol and 1,12‐dodecanediol. The BTBS moieties of these compounds are good leaving groups and stopper exchange reactions could be achieved by treatment with different nucleophiles thus affording rotaxanes with ester, thioether or ether stoppers.

Amplifying molecular motion to macroscopic material property is attractive but challenging. Here, the authors report mechanically interlocked networks with dense rotaxane backbones whose microscopic motion is integrated into decent mechanical properties of bulk materials.

Polymer beads have been used as the core of magnetic particles for around twenty years. Here we report studies to attach polymetallic complexes to polymer beads for the first time, producing beads of around 115 microns diameter that are attached to 1014 hybrid inorganic-organic [2]rotaxanes. The bead is then formally a [1014] rotaxane. The number of complexes attached is counted by EPR spectroscopy after including TEMPO radicals within the thread of the hybrid [2]rotaxanes. In this work, the ability of several bis-viologen axles to thread a series of heteroditopic tris(N-phenylureido)calix[6]arene wheels to give interwoven supramolecular complexes to the [3]pseudorotaxane type was studied. The unidirectionality of the threading process inside these nonsymmetric wheels allows the formation of highly preorganised [3]pseudorotaxane and [3]rotaxane species in which the macrocycles phenylureido moieties, functionalised with either ester, carboxylic, or hydroxymethyl groups, are facing each other. As verified by NMR and semiempirical computational studies, these latter compounds possess the correct spatial arrangement of their subcomponents, which could lead, in principle, upon proper bridging reaction, to the realisation of upper-to-upper molecular capsules that are based on calix[6]arene derivatives.

Diamine reagents have been used to functionalize a [2]rotaxane building block bearing an activated pentafluorophenyl ester stopper. Upon a first acylation, an intermediate host‐guest complex with a terminal amine function is obtained. Dissociation of the intermediate occurs in solution and acylation of the released axle generates a [2]rotaxane with an elongated axle subunit. In contrast, the corresponding [3]rotaxane can be obtained if the reaction conditions are appropriate to stabilize the inclusion complex of the mono‐amine intermediate and the pillar[5]arene. This is the case when the stopper exchange is performed under mechanochemical solvent‐free conditions. Alternatively, if the newly introduced terminal amide group is large enough to prevent the dissociation, the second acylation provides exclusively a [3]rotaxane. On the other hand, detailed conformational analysis has been also carried out by variable temperature NMR investigations. A complete understanding of the shuttling motions of the pillar[5]arene subunit along the axles of the rotaxanes reported therein has been achieved with the help of density functional theory calculations. Abstract We present the concept of electrostatic field-driven supramolecular translation within electrically connected plasmonic nano-antennas. The antenna serves as an anchoring point for the mechanically interlocked molecules, as an electrode for the electrostatic field, and as an amplifier of the antenna-enhanced fluorescence. The synthesis of a push–pull donor–π–acceptor chromophore with optical properties aligned to the antenna resonance is described and its immobilization on the surface is demonstrated. Photoluminescence experiments of the chromophore on a gold nano-antenna are shown, highlighting the molecule–antenna coupling and resulting emission intensity increase. The successful synthesis of an electrostatic field-sensitive [2]rotaxane in water is described and the tightrope walk between functionality and water solubility is illustrated by unsuccessful designs. In solution, an enhanced fluorescence quantum yield is observed for the chromophore comprising the mechanically interlocked [2]rotaxane in water and DMSO compared to the reference rod, ideal for future experiments in plasmonic nano-antennas.

Chiral pseudo[1]rotaxanes and [1]rotaxanes constructed from macrocyclic arenes still remain a big challenge mainly owing to the lack of such chiral macrocycles. In this work, a new system of chiral pseudo[1]rotaxanes formed by self‐inclusion of helic[6]arene containing amide linked with the terminal tertiary amines was first discovered. Based on an atom‐economic stopping strategy, a pair of chiral [1]rotaxanes were conveniently obtained in almost quantitative yields by blocking the pseudo[1]rotaxanes with monobenzyl bromide of tetraphenylethene. The structures of pseudo[1]rotaxanes and [1]rotaxanes were characterized by 2D NMR spectra in solution, combined with DFT calculations. The photophysical properties further revealed the efficient chirality transfer of helic[6]arene to the tetraphenylethene moiety, compared to their unthreaded chiral isomers. The discovery of the chiral pseudo[1]rotaxanes allows for a wide and available synthesis of chiral [1]rotaxanes, and also opening a new avenue to the design of chiral supramolecular materials. [1]Rotaxanes based on monosubstituted phosphorus-containing pillar[5]arenes have been synthesized for the first time by the Kabachnik–Fields reaction in good yields. The introduction of the bulky phosphoryl functionalized stopper led to the formation of the [1]rotaxane core. Direct cytosolic delivery of the Cas9 ribonucleoprotein is the most promising method for inducing CRISPR-Cas9 genome editing in mammalian cells. Recently, we focused the movable properties of cyclodextrin-based polyrotaxanes (PRXs), which consist of numerous cyclodextrins threaded onto the axile molecule with bulky endcaps at both ends of the axile molecule, and developed aminated PRXs as multistep transformable carriers for Cas9 ribonucleoprotein, ensuring efficient complexation, cellular internalization, endosomal escape, release, and nuclear localization. This study reports the structural fine-tuning and structure-property relationship of multistep transformable PRXs for more efficient Cas9 ribonucleoprotein delivery. Among various PRXs, PRX derivatives with a longer molecular length (35 kDa polyethylene glycol as the axile molecule) and a low total degree of substitution (1.5 amino groups/α-cyclodextrins), as well as the modified ratio of two modified amines (cystamine and diethylenetriamine) = ≈1:1, exhibited the highest genome-editing efficacy and intracellular dynamics control. These structural properties are important for efficient endosomal escape and Cas9 RNP release. Furthermore, ligand-modified-β-CD, which can endow the ligand through complexation with PRX termini, improved the cellular uptake and genome-editing effects of the optimized PRX/Cas9 RNP in target cells. Mechanically interlocked molecules (MIMs) have been important synthetic targets in supramolecular chemistry due to their beautiful structures and intriguing properties. We present herein a new synthetic strategy to access [2]rotaxanes, namely active-metal template clipping. We discuss the design of the target [2]rotaxanes, synthesis and characterization of the axle, macrocycle precursors and macrocycles as well as preparation of the final [2]rotaxanes by active template copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) as key step of the synthesis.
