The present study reports the characterization of a non-T cell from human peripheral blood which is capable of releasing BCGF. This BCGF-producing non-T cell had a T3-, T8-, Leu-7+, OKM1+, HLA-DR-, Leu-11- surface phenotype and was likely to belong to the so-called large granular lymphocyte (LGL) subset because: after fractionation of non-T cells according to the expression of Leu-7 or HLA-DR markers, it was found in the Leu-7+, HLA-DR- fractions that were particularly enriched in LGL; it co-purified with LGL on Percoll density gradients; and it expressed Leu-7 and OKM1 markers that are shared by a large fraction of LGL. Although co-purified with cells with potent NK capacities, the BCGF-producing cell was not cytotoxic, because treatment of Leu-7+ cells with Leu-11 monoclonal antibody and complement abolished the NK activity but left the BCGF activity unaltered. The factor released by this LGL subset was not IL 1 or IL 2 mistakenly interpreted as BCGF, because: a) cell supernatants particularly rich in BCGF activity contained very little or no IL 1 or IL 2; b) BCGF-induced B cell proliferation was not inhibitable by anti-Tac antibodies (this in spite of the expression of IL 2 receptor by a proportion of activated B cells); and c) BCGF activity was absorbed by B but not T blasts.

Malignant lymphocytes from 30 B-cell chronic lymphocytic leukemia (B-CLL) patients were studied for the cytochemical localization of two acid hydrolases, alpha-naphthyl acetate esterase (ANAE) and acid phosphatase (AT). The large majority of the cells stained for both ANAE and AP in 7 cases, for AP only in 18 cases, and were negative for both the enzymes in 5 cases. Ultrastructural analysis revealed that the cells that displayed more mature morphological features, such as well developed smooth and rough membrane compartments, were those positive for acid hydrolases. That ANAE and AP are expressed by B cells at late stage of maturation was confirmed by the finding that some lymphocytes and all of the plasmacytoid lymphocytes and plasma cells from Walderström's macroglobulinemia, from mixed cryoglobulinemia, and from multiple myeloma patients stained strongly for both ANAE and AP. Using the expression of acid hydrolases and certain ultrastructural features as markers of cell differentiation, it was possible to demonstrate a process of maturation within the single B-CLL clones with accumulation of the cells at stages that differed in the various cases.

The susceptibility to proteolysis and the mode of re-expression of receptors for IgM or IgG present on two different subpopulations of human T lymphocytes (T.M and T.G cells, respectively) have been investigated. The IgM receptor was highly susceptible to both trypsin and pronase, whereas the IgG receptor was resistant to trypsin and sensitive only to high concentrations of pronase. The receptors have been removed by treating purified human T cells with pronase and their reappearance on the cell surface has been followed in vitro. The IgM receptors on the cell surface were detectable within 2 hr and the resynthesis was completed in 6 hr. IgG receptors were detectable in 4 to 6 hr and the resynthesis completed within 12 hr. When protein synthesis was inhibited by culturing the cells in the presence of cycloheximide for up to 12 hr, only the IgM receptor (which had a higher turnover rate) failed to be expressed. Whereas interaction of IgG immune complex with the IgG receptors was previously shown to induce a modulation of the receptors, contact with antigen-IgM antibody complexes did not alter the mode of expression of IgM receptors.

Human NK cells, besides mediating cytotoxic functions, can exert a number of regulatory activities on the proliferation and maturation of hematopoietic stem cells through the release of soluble factors. A preliminary characterization of these factors demonstrates that NK cell-derived colony stimulating activities (CSA) are represented by GM-CSF and IL3, whereas the colony inhibiting activity (CIA) produced by NK cells is identifiable as TNF. CSA is mainly released by resting NK cells, while, following appropriate activation, the latter cells produce large amounts of CIA. NK cells also influence B cell activation by releasing a 50 kd BCGF and a 26 kd BCDF. The latter factor, which is active in vitro only in the presence of IL2, is identifiable as IL6. The production of regulatory cytokines by NK cells may have some bearing on the pathogenesis of a number of autoimmune disorders.

Two different methods, immunofluorescence and lactoperoxidase-catalysed radioiodination, were used to investigate mIg on two different myeloma cell lines. The data indicate that, even though both techniques are highly sensitive, different results can be obtained in the same cell line depending on the technique employed.

Immunofluorescent and "in vitro" biosynthetic techniques have been used to study the connections between the maturation process of human lymphocytes that leads to the appearance of actively secreting cells and a possible switchover from IgD to IgM production, both in homogeneous CLL cell populations and in heterogeneous cell suspensions from tonsils. The results obtained are compatible with the hypothesis that the switchover from IgD to IgM productions can be a clonal maturation phenomenon.
