853. Genetic control of the phenobarbital-induced shortening of plasma antipyrine half-lives in man

The mean half-life of antipyrine in the plasma of four sets of identical and four sets of fraternal twins after a single oral dose of 16 mg/kg of antipyrine was 12.7 +/-(SD) 3.3 hr. After 2 wk on sodium phenobarbital (2 mg/kg daily) the half-life of antipyrine in the plasma of these twins was reduced to 8.0 +/-(SD) 1.5 hr. Shortening of the plasma antipyrine half-life occurred in all but one of these 16 normal, adult volunteers, but there was considerable variation in the extent of reduction which ranged from 0 to 69%. Phenobarbital administration decreased individual variations in antipyrine metabolism as indicated by the smaller standard deviation of the plasma antipyrine half-lives after phenobarbital than observed initially and by the narrowed range of variation in plasma antipyrine half-lives from 2.8-fold initially to 1.8-fold after phenobarbital. These results suggest that some inducing agents may be used to minimize individual variations in drug metabolism where such variations create therapeutic problems by exposing patients who slowly metabolize certain drugs to toxicity and other patients who rapidly metabolize some drugs to undertreatment. During the course of phenobarbital administration blood levels were determined. Phenobarbital blood levels correlated neither with the final values for plasma antipyrine half-lives nor with the per cent reduction in plasma antipyrine half-life produced by phenobarbital treatment. There was a direct relationship between initial antipyrine half-lives and the per cent shortening of antipyrine half-life produced by phenobarbital administration: the shorter the initial antipyrine half-life, the less the reduction caused by phenobarbital treatment. Larger intrapair variances in fraternal than in identical twins indicate genetic, rather than environmental, control of phenobarbital-induced alterations in plasma antipyrine half-life.

Privacy-enhancing attribute-based credential (Privacy-ABC) technologies use different cryptographic methods to enhance the privacy of the users. This results in important practical differences between these technologies, especially with regard to efficiency, which have not been studied in depth, but is necessary for assessing their suitability for different user devices and for highly dynamic scenarios. In this paper, we compare the computational efficiency of two prominent Privacy-ABC technologies, IBM’s Idemix and Microsoft’s U-Prove, covering all known Privacy-ABC features. The results show that overall presentation is in general is more efficient with Idemix, whereas U-Prove is more efficient for the User side (proving) operations during the presentation, and overall when there are more attributes in a credential. For both technologies we confirmed that inspectability, non-revocation proofs, and inequality predicates are costly operations. Interestingly, the study showed that equality predicates, the number of attributes in a credential, and attribute disclosure are done very efficiently. Finally, we identified a number of specific trust issues regarding Privacy-ABC technologies.

Biogenic amines are organic compounds, present in living organisms and they can be a responsibility for many essential processes. Naturally, they can be also in fruits and vegetable and all foods that contain free amino acids or proteins. In a high amount they can be produced by microorganism through the enzymatic reac- tion. Histamine, putrescine, cadaverine, tyramine, tryptamine, β-phenylethylamine, spermine and spermidine are considered to be the most important biogenic amines occurring in foods. Excessive consumption of these amines can inflict health concern and analysis of biogenic amines is important based on their toxicity and their usage as indicators of the degree of freshness or spoilage of food. Several methods exist for isolation, identif- ying and determination of biogenic amines in food. They are mainly based on chromatographic methods: thin layer chromatography TLC , gas chromatography GC , capillary electrophoresis CE and high-performance liquid chromatography HPLC . Due to the low volatility and lack of chromophores of most biogenic amines, UV- spectrometric detection is limited. A new challenge is electrochemical determination of those amines based on oxidative reaction. This review shows different methods which allow quantitative determination of biogenic amines in different food stuffs., B iyojenik aminler, canlı organizmalarda bulunan organik bileşiklerdir ve birçok temel işlemlerden sorumlu olabilirler. Meyve, sebze ve serbest amino asit veya protein içeren her gıdada doğal olarak bulunabilirler. Enzimatik tepkimeler yoluyla mikroorganizmalar tarafından yüksek miktarlarda üretilebilirler. Histamin, putresin, kadaverin, tiramin, triptamin, β-feniletilamin, spermin ve spermidin, gıdalarda en sık bulunan önemli biyojenik aminler olarak değerlendirilir. Bu aminlerin aşırı kullanımı, sağlık sorununa yol açabilir ve biyojenik aminlerin analizi, toksik etkilerinin ve tazelik veya bozulma derecelerinin bir göstergesi olarak oldukça önemlidir. Gıdalarda biyojenik aminlerin izolasyonu, tanımlanması ve tayini için çeşitli yöntemler mevcuttur. Bu yöntemler temel olarak kromatografik yöntemlerdir: ince tabaka kromatografisi TLC , gaz kromatografisi GC , kapiler elektroforez CE ve yüksek performans sıvı kromatografi HPLC . Birçok biyojenik aminlerin düşük uçuculuğu ve kromofor gruplarının olmaması dolayısıyla, UV-spektrometrik tayinleri sınırlıdır. 863. The diversity and functional capacity of microbes associated with coastal phototrophs

- Abstract
Coastal marine phototrophs exhibit some of the highest rates of primary productivity in the world. They have been found to host a diverse set of microbes, many of which may impact the biology of their phototroph hosts through metabolisms that are unique to microbial taxa. Here we characterized the metabolic functions of phototroph-associated microbial communities using metagenomes collected from 2 species of kelp (Laminaria setchellii and Nereocystis luetkeana) and 3 marine angiosperms (Phyllospadix scouleri, P. serrulatus and Zostera marina), including the rhizomes of two surfgrass species (Phyllospadix spp.) and the seagrass Zostera marina, and the sediments surrounding P. scouleri and Z. marina. Using metagenomic sequencing, we describe 72 metagenome assembled genomes (MAGs) that potentially benefit from being associated with macrophytes and may contribute to macrophyte fitness through their metabolic gene content. All host-associated metagenomes contained genes for the use of dissolved organic matter from hosts and vitamin (B1, B2, B7, B12) biosynthesis. Additionally, we found a range of nitrogen metabolism genes that transform dissolved inorganic nitrogen into forms that may be more available to the host. The rhizosphere of surfgrass and seagrass contained genes for anaerobic microbial metabolisms, including nifH genes associated with nitrogen fixation, despite residing in a well-mixed and oxygenated environment. The range of oxygen environments engineered by macrophytes likely explains the diversity of both oxidizing and reducing microbial metabolisms, and contributes to the functional capabilities of microbes and their influence on carbon and nitrogen cycling in nearshore ecosystems.ImportanceKelps, seagrasses and surfgrasses are ecosystem engineers on rocky shorelines where they show remarkably high levels of primary production. Through analysis of their associated microbial communities, we found a variety of microbial metabolisms that may benefit the host, including nitrogen metabolisms and the production of B vitamins. In turn, these microbes have the genetic capability to assimilate the dissolved organic compounds released by their phototroph hosts. We describe a range of oxygen environments associated with surfgrass, including low-oxygen microhabitats in their rhizomes that host genes for nitrogen fixation. The tremendous productivity of coastal phototrophs is likely due in part to the activities of associated microbes and an increased understanding of these associations is needed.

Simple Summary: The tiger mosquito, Aedes albopictus, spreads the virus causing dengue fever, notably in Southeast Asian countries. To stop the spread of this disease, the Aedes mosquitoes are killed with chemicals known as pyrethroids. However, despite pyrethroids being highly effective insecticides, tiger mosquitoes are becoming resistant and surviving exposure to these chemicals. We have measured that tiger mosquitoes from two regions in Cambodia (the capital, Phnom Penh, and rural Pailin province) are highly resistant to pyrethroids. To determine what causes resistance, we have implicated unusually high activity of enzymes, known as P450 monooxygenases, which break down pyrethroids and thus render them harmless in the resistant mosquitoes. We have ruled out another common mechanism of pyrethroid resistance, which is changes in the mosquitoes' DNA sequence, which prevents pyrethroids from binding to a protein known as the voltage-gated sodium channel, thereby disrupting signals being sent throughout the nervous system. Overall, our findings describe an important survey monitoring the prevalence of insecticide resistance in disease-spreading mosquitoes in Cambodia, and with the increased understanding of what causes resistance, we are in a strong position to advise that mosquito control methods alternative to pyrethroids should be implemented as soon as possible to ensure continuing management of dengue fever. (1) Background: In Cambodia, Aedes albopictus is an important vector of the dengue virus. Vector control using insecticides is a major strategy implemented in managing mosquito-borne diseases. Resistance, however, threatens to undermine the use of insecticides. In this study, we present the levels of insecticide resistance of Ae. albopictus in Cambodia and the mechanisms involved. (2) Methods: Two Ae. albopictus populations were collected from the capital, Phnom Penh city, and from rural Pailin province. Adults were tested with diagnostic doses of malathion (0.8%), deltamethrin (0.03%), permethrin (0.25%), and DDT (4%) using WHO tube assays. Synergist assays using piperonyl butoxide (PBO) were implemented before the pyrethroid assays to detect the potential involvement of metabolic resistance mechanisms. Adult female mosquitoes collected from Phnom Penh and Pailin were tested for voltage-gated sodium channel (VGSC) kdr (knockdown resistance) mutations commonly found in Aedes sp.-resistant populations throughout Asia (S989P, V1016G, and F1534C), as well as for other mutations (V410L, L982W, A1007G, I1011M, T1520I, and D1763Y). (3) Results: The two populations showed resistance against all the insecticides tested (<90% mortality). The use of PBO (an inhibitor of P450s) strongly restored the efficacy of deltamethrin and permethrin against the two resistant populations. Sequences of regions of the vgsc gene showed a lack of kdr mutations known to be associated with pyrethroid resistance. However, four novel non-synonymous mutations (L412P/S, C983S, Q1554STOP, and R1718L) and twenty-nine synonymous mutations were detected. It remains to be determined whether these mutations contribute to pyrethroid resistance. (4) Conclusions: Pyrethroid resistance is occurring in two Ae. albopictus populations originating from urban and rural areas of Cambodia. The resistance is likely due to metabolic resistance specifically involving P450s monooxygenases. The levels of resistance against different insecticide classes are a cause for concern in Cambodia. Angiogenesis is the process of new blood vessel formation. In this complex orchestrated growth, many factors are included. Lately, focus has shifted to endothelial cell metabolism, particularly to the PFKFB3 protein, a key regulatory enzyme of the glycolytic pathway. A variety of inhibitors of this important target have been studied, and a plethora of biological effects related to the process of angiogenesis have been reported. However, recent studies have disputed their mechanism of action, questioning whether all the effects are indeed due to PFKFB3 inhibition. Remarkably, the most well-studied inhibitor, 3PO, does not bind to PFKFB3, raising questions about this target. In our study, we aimed to elucidate the effects of PFKFB3 inhibition in angiogenesis by using the small molecule AZ67. We used isothermal titration calorimetry and confirmed binding to PFKFB3. In vitro, AZ67 did not decrease lactate production in endothelial cells (ECs), nor ATP levels, but exhibited good inhibitory efficacy in the tube-formation assay. Surprisingly, this was independent of EC migratory and proliferative abilities, as this was not diminished upon treatment. Strikingly however, even the lowest dose of AZ67 demonstrated significant inhibition of angiogenesis in vivo. To our knowledge, this is the first study to demonstrate that the process of angiogenesis can be disrupted by targeting PFKFB3 independently of glycolysis inhibition.

Seagrasses are important primary producers in oceans worldwide. They live in shallow coastal waters that are experiencing carbon dioxide enrichment and ocean acidification. Posidonia oceanica, an endemic seagrass species that dominates the Mediterranean Sea, achieves high abundances in seawater with relatively low concentrations of dissolved inorganic nitrogen. Here we tested whether microbial metabolisms associated with P. oceanica and surrounding seawater enhance seagrass access to nitrogen. Using stable isotope enrichments of intact seagrass with amino acids, we showed that ammonification by free-living and seagrass-associated microbes produce ammonium that is likely used by seagrass and surrounding particulate organic matter. Metagenomic analysis of the epiphytic biofilm on the blades and rhizomes support the ubiquity of microbial ammonification genes in this system. Further, we leveraged the presence of natural carbon dioxide vents and show that the presence of P. oceanica enhanced the uptake of nitrogen by water column particulate organic matter, increasing carbon fixation by a factor of 8.6–17.4 with the greatest effect at CO2 vent sites. However, microbial ammonification was reduced at lower pH, suggesting that future ocean climate change will compromise this microbial process. The performance of the new 52 kHz frame rate Rigaku XSPA‐500k detector was characterized on beamline 8‐ID‐I at the Advanced Photon Source at Argonne for X‐ray photon correlation spectroscopy (XPCS) applications. Due to the large data flow produced by this detector (0.2 PB of data per 24 h of continuous operation), a workflow system was deployed that uses the Advanced Photon Source data‐management (DM) system and high‐performance software to rapidly reduce area‐detector data to multi‐tau and two‐time correlation functions in near real time, providing human‐in‐the‐loop feedback to experimenters. The utility and performance of the workflow system are demonstrated via its application to a variety of small‐angle XPCS measurements acquired from different detectors in different XPCS measurement modalities. The XSPA‐500k detector, the software and the DM workflow system allow for the efficient acquisition and reduction of up to ∼109 area‐detector data frames per day, facilitating the application of XPCS to measuring samples with weak scattering and fast dynamics. Background: Despite descriptions of various cardiovascular manifestations in patients with coronavirus disease 2019 (COVID‐19), there is a paucity of reports of new onset bradyarrhythmias, and the clinical implications of these events are unknown. Methods: Seven patients presented with or developed severe bradyarrhythmias requiring pacing support during the course of their COVID‐19 illness over a 6‐week period of peak COVID‐19 incidence. A retrospective review of their presentations and clinical course was performed. Results: Symptomatic high‐degree heart block was present on initial presentation in three of seven patients (43%), and four patients developed sinus arrest or paroxysmal high‐degree atrioventricular block. No patients in this series demonstrated left ventricular systolic dysfunction or acute cardiac injury, whereas all patients had elevated inflammatory markers. In some patients, bradyarrhythmias occurred prior to the onset of respiratory symptoms. Death from complications of COVID‐19 infection occurred in 57% (4/7) patients during the initial hospitalization and in 71% (5/7) patients within 3 months of presentation. Conclusions: Despite management of bradycardia with temporary (3/7) or permanent leadless pacemakers (4/7), there was a high rate of short‐term morbidity and death due to complications of COVID‐19. The association between new‐onset bradyarrhythmias and poor outcomes may influence management strategies for acutely ill patients with COVID‐19. [ABSTRACT FROM AUTHOR]

As the civilian population is no longer just a consumer, but also a producer of renewable energy (so-called prosumer), the electricity consumption data between the members need a tamper-proof and automatic billing system. The blockchain technology can be used for trading electricity in an energy community and aims at making the system smarter. It can manage the billing between the members, i.e., the energy producers (e.g. power generated by a PV system), the energy consumers, and/or those involved as energy storage. The processing of data by means of a blockchain can be helpful, but there is an inherent conflict with questions of data protection, especially since data on the energy consumption of households is considered as personal data. Plasmids are extrachromosomal genetic elements that often encode fitness-enhancing features. However, many bacteria carry "cryptic" plasmids that do not confer clear beneficial functions. We identified one such cryptic plasmid, pBI143, which is ubiquitous across industrialized gut microbiomes and is 14 times as numerous as crAssphage, currently established as the most abundant extrachromosomal genetic element in the human gut. The majority of mutations in pBI143 accumulate in specific positions across thousands of metagenomes, indicating strong purifying selection. pBI143 is monoclonal in most individuals, likely due to the priority effect of the version first acquired, often from one's mother. pBI143 can transfer between Bacteroidales, and although it does not appear to impact bacterial host fitness in vivo , it can transiently acquire additional genetic content. We identified important practical applications of pBI143, including its use in identifying human fecal contamination and its potential as an alternative approach to track human colonic inflammatory states. A case study on Sitagliptin drug products and Sitagliptin/Metformin drug products concerning contamination with N -nitrosamines was performed using two newly developed analytical methods for determination of N -nitroso-triazolopyrazine (NTTP; 7-nitroso-3-(trifluoromethyl)–5,6,7,8-tetrahydro-[1,2,4]triazolo[4,3- a ]pyrazine) and its precursor triazolopyrazine (3-(trifluoromethyl)–5,6,7,8-tetrahydro-[1,2,4]triazolo[4,3- a ]pyrazine). The method for determination of triazolopyrazine was previously unpublished, the method for determination of NTTP was published only for analysis of active pharmaceutical ingredient Sitagliptin and not the drug forms. Solving the N -nitrosamine contamination is requested by regulatory authorities all over the world and thus is vital for all pharmaceutical companies. The solution always requires a sensitive analytical method. Both newly developed methods use liquid chromatography coupled with mass spectrometry (single quadrupole analyzer in case of triazolopyrazine and triple quadrupole analyzer in case of NTTP). Separation of triazolopyrazine was achieved on a column Acquity CSH C18 using a mobile phase consisting of aqueous ammonium formate buffered at pH 4.2 and acetonitrile. Detection was performed using positive electrospray and selected ion monitoring at m/z 193. Separation of NTTP was achieved on a column Acquity HSS T3 using a mobile phase consisting of 0.1 % formic acid in water and methanol. Detection was performed using positive electrospray and multiple reaction monitoring at transitions m / z 222.15→42.05 (collision energy 17 eV) and m / z 222.15→192.15 (collision energy 11 eV). Two issues specific to NTTP and triazolopyrazine previously not described in scientific literature were successfully troubleshooted. Spontaneous degradation of Sitagliptin to triazolopyrazine and methyl (R)–3-amino-4-(2,4,5-trifluorophenyl)butanoate was solved by using N,N -dimethylformamide as sample solvent during development of the method for quantitation of triazolopyrazine. A bad peak shape of NTTP due to the presence of rotamers of NTTP was successfully troubleshooted by increasing column temperature. Both methods were used during an optimization study of manufacturing of Sitagliptin and Sitagliptin/Metformin drug products. The goal of the study was to decrease NTTP content in the final drug product under the strict legislative limit set by Federal Drug Agency. The efficacy of several solutions was proven, but could not be fully disclosed due to Intellectual Property Protection policy of Zentiva. Instead, a brief review of recently published strategies to cope with N -nitrosamine contamination is presented. Nitric oxide (NO) donors are commonly used for the prevention and treatment of ischemic heart disease. Besides their effects on the heart, NO donors may also prevent hypoxic brain damage and exert beneficial effects on atherosclerosis by favoring features of plaque stability. We recently described that apolipoprotein E (ApoE) deficient mice with a mutation in the fibrillin-1 (Fbn1) gene (ApoE -/- Fbn1 C1039G+/- ) develop accelerated atherosclerosis, plaque rupture, myocardial infarction, cerebral hypoxia and sudden death. In the present study, we evaluated the effects of chronic treatment with the NO donor molsidomine on atherosclerotic plaque stability, cardiac function, neurological symptoms and survival in the ApoE -/- Fbn1 C1039G+/- mouse model. Female ApoE -/- Fbn1 C1039G+/ - mice were fed a Western diet (WD). After 8 weeks of WD, the mice were divided into two groups receiving either molsidomine via the drinking water (1 mg/kg/day; n = 34) or tap water (control; n = 36) until 25 weeks of WD. Survival tended to increase after molsidomine treatment (68% vs. 58% in controls). Importantly, atherosclerotic plaques of molsidomine-treated mice had a thicker fibrous cap (11.1 ± 1.2 vs. 8.1 ± 0.7 μm) and showed an increased occurrence of plaque macrocalcifications (30% vs. 0%), indicative of a more stable phenotype. Molsidomine also improved cardiac function, as fractional shortening was increased (40 ± 2% vs. 27 ± 2%) combined with a decreased end diastolic (3.1 ± 0.2 vs. 3.9 ± 0.2 mm) and end systolic diameter (1.9 ± 0.1 vs. 2.9 ± 0.2 mm). Furthermore, perivascular fibrosis (23 ± 2 vs. 30 ± 2%) and the occurrence of myocardial infarctions (12% vs. 36%) was significantly reduced. Track width, a measure of the animal's hind limb base of support and representative of hypoxic brain damage, was also normalized as a result of molsidomine treatment (2.54 ± 0.04 vs. 2.91 ± 0.09 cm in controls). These findings demonstrate that the NO donor molsidomine improves cardiac function, reduces neurological symptoms and enhances atherosclerotic plaque stability. Abstract: To enable globally distributed computing for a large HEP experiment, a collection of computing and data storage facilities, together called the Grid fabric, must be linked together in a coherent way. The standard Grid software, including most notably the Globus Gatekeeper and Meta Directory Service, provides core tools to insert a site into the Grid and for its low-level monitoring. In practice, large experiments have data and job handling infrastructures that are not governed by the core tools. For example, local job submission is seldom done directly to the batch system, but rather, through an interface that allows for pre-submission steps (such as the decomposition of a job into smaller chunks) or is tightly integrated with a data handling system such as SAM. Likewise, monitoring is seldom done in terms of individual processors or individual jobs, but rather, via cluster-wide aggregated characteristics. In this paper, we present some of the work we have done to abstract the management of the fabric facilities of the FNAL Run II experiments, in order to enable globally distributed computing. SAM is a robust distributed file-based data management and access service, fully integrated with the D0 experiment at Fermilab and in phase of evaluation at the CDF experiment. The goal of the SAM-Grid project is to fully enable distributed computing for the experiments. The architecture of the project is composed of three primary functional blocks: the job handling, data handling, and monitoring and information services. Job handling and monitoring/information services are built on top of standard grid technologies (Condor-G/Globus Toolkit), which are integrated with the data handling system provided by SAM. Die Schweizer Unternehmen der grafischen Industrie haben zwar unter der jüngsten Rezession gelitten, für die Zukunft erwarten sie aber eine Verbesserung ihrer Situation. Dies bezieht sich sowohl auf die Entwicklung des Umsatzes, der Nachfrage wie auch der Beschäftigten. Die Unternehmen schätzen den Preisdruck, der weder vor lokalen, regionalen noch nationalen Grenzen Halt macht, als sehr stark ein. Dies zwingt die Unternehmen, sich über nicht-preisliche Faktoren zu positionieren. Daneben werden auch Teile des Produktionsprozesses ausgelagert. Eine Verschiebung zeichnet sich beim Arbeitskräftebedarf ab. Hier dürften in Zukunft höhere Qualifikationen wichtiger werden. Entsprechend müssten die Weiterbildungsangebote angepasst werden, mit denen nicht alle Unternehmen zufrieden sind. - Abstract
Patient blood group (BG) is predictive for von-Willebrand-factor (VWF) and Factor VIII variation. The clinical impact of this ABO-effect on blood loss, cardiovascular complications and outcome has been described for several patient cohorts. The aim of this study was to investigate the impact of patient BG on blood loss and outcome after coronary artery bypass surgery (CABG). Patient records, intraoperative data and perioperative transfusion records of 5713 patients receiving an on-pump CABG procedure between 05/2004 and 12/2018 were analyzed. A logistic regression model for death due to perioperative myocardial ischaemia (PMI) was developed from initially 24 variables by using an univariate and multivariate selection process. BG AB patients required less blood transfusions as compared to the other blood groups, especially in case of emergency operations. However, BG AB patients also had a higher mortality which was due to secondary cardiovascular complications. The impact of blood type on the rate of cardiovascular mortality was confirmed in the logistic regression model. BG AB patients have a worse outcome after CABG surgery due to an increased incidence of fatal cardiovascular complications. As perioperative myocardial ischemia due to graft occlusion appears to be the most likely explanation, stricter anticoagulation for BG AB patients should be discussed., (© 2024. The Author(s).)

- Abstract
The significance of thermodynamic solubility in biopharmaceutical compound or drug characterization as well as the importance of having methods that accurately establish it have been extensively addressed. Nonetheless, its precise determination continues to remain a challenging task to accomplish. Even more so when the number of compounds to evaluate is high and the available amount of each compound is low, both of which are inevitable for the compound characterization during the drug development process. Except for the shake-flask method which is still considered as the 'gold standard' in obtaining thermodynamic data, it is currently difficult to say that another satisfactory model which is routinely used to determine thermodynamic solubility is being applied. Therefore, this review summarizes the various experimental approaches which are based on the classical shake flask method but have yet attempted to speed up the experimental process of obtaining such data more conveniently. The most important experimental features of these approaches are provided to the reader. Some advantages and disadvantages associated with each approach are also highlighted, consequently offering a resource to those looking for the most appropriate of the approaches that have already fared well at determining the biopharmaceutically relevant drug solubility.
