Background: Bedtime procrastination (BP), a special type of health behavior procrastination, is considered to be a failure of self-control. Notably, self-control may mediate the effect of trait anxiety on general procrastination. However, there is no evidence demonstrating the role of self-control in the relationship between trait anxiety and BP. Moreover, the association between BP and trait anxiety has not yet been thoroughly studied. Therefore, the present study aimed to explore the direct relationship between them as well as the mediating role of self-control in this relationship., Methods: This cross-sectional survey included 718 college students enrolled in Chinese universities between October 2018 and January 2020. The Chinese versions of the Bedtime Procrastination Scale, the Self-Control Scale, and the Trait Anxiety Inventory were used to evaluate BP, self-control, and trait anxiety, respectively., Results: Multiple linear regression analysis revealed trait anxiety independently predicted BP while controlling for demographic characteristics. Correlation analyses showed that BP was positively correlated with trait anxiety, but negatively related to self-control. Structural equation modeling further revealed a mediating role of self-control in the relationship between trait anxiety and BP., Conclusions: Trait anxiety is a significant independent predictor of BP and may induce BP directly or indirectly through the effect of self-control. These findings provide a deeper understanding of the relationship between trait anxiety and BP and the underlying mechanism by exploring the mediating effect of self-control. Multiple myeloma (MM) is the second most common hematological malignancy with poor prognosis. Enhancer of zeste homolog 2 (EZH2) is the enzymatic subunit of polycomb repressive complex 2 (PRC2), which catalyzes trimethylation of histone H3 lysine 27 (H3K27me3) for transcriptional repression. EZH2 have been implicated in numerous hematological malignancies, including MM. However, noncanonical functions of EZH2 in MM tumorigenesis are not well understood. Here, we uncovered a noncanonical function of EZH2 in MM malignancy. In addition to the PRC2-mediated and H3K27me3-dependent canonical function, EZH2 interacts with cMyc and co-localizes with gene activation-related markers, promoting MM tumorigenesis in a PRC2- and H3K27me3-independent manner. Both canonical EZH2-PRC2 and noncanonical EZH2-cMyc complexes can be effectively depleted in MM cells by MS177, an EZH2 degrader we reported previously, leading to profound activation of EZH2-PRC2-associated genes and simultaneous suppression of EZH2-cMyc oncogenic nodes. The MS177-induced degradation of both canonical EZH2-PRC2 and noncanonical EZH2-cMyc complexes also reactivated immune response genes in MM cells. Phenotypically, targeting of EZH2's both canonical and noncanonical functions by MS177 effectively suppressed the proliferation of MM cells both in vitro and in vivo. Collectively, this study uncovers a new noncanonical function of EZH2 in MM tumorigenesis and provides a novel therapeutic strategy, pharmacological degradation of EZH2, for treating EZH2-dependent MM., (© 2023. Objectives: The erector spinae plane (ESP) block is a newly defined regional anesthesia technique first described in 2016. The aim of this meta-analysis is to assess the efficacy of ESP block in improving analgesia following lumbar surgery., Methods: PubMed, EMBASE, Cochrane Library, and Web of Science were searched for randomized controlled trials (RCTs) that compared the analgesic efficacy of the ESP block with non-block care for lumbar surgery from inception 3 August 2021. The primary outcomes were postoperative opioid consumption and pain scores during the first 24 h. Postoperative pain was measured as pain at rest and on movement at postoperative 0, 4, 8, 12, and 24 h expressed on a visual analog scale (VAS), where 0 = no pain and 10 = the most severe pain., Results: 11 studies involving 775 patients were included in our analysis. The use of ESP block significantly decreased 24-h opioid consumption (WMD, -8.70; 95% CI, -10.48 to -6.93; I 2  = 97.5%; P < 0.001) compared with the non-block. Moreover, ESP block reduced pain scores at postoperative time-points up to 24 h. ESP block also prolonged the time to first analgesic request (WMD = 6.93; 95% CI: 3.44 to 10.43, I 2  = 99.8%; P < 0.001). There was less PONV with ESP block versus non-block group (RR, 0.354; 95% CI, 0.23 to 0.56; I 2  = 25.2%; P < 0.001), but no difference in pruritus., Conclusions: ESP block provides less opioid consumption and PONV, lower pain scores, and longer time to first analgesic request in patients undergoing lumbar surgery compared to general anesthesia alone., (© 2023. Two-dimensional (2D) layered MoSe 2 has been demonstrated to be a promising electrode material for new energy storage systems. However, its nature of poor conductivity and the undesirable interlayer spacing hinder its further application. In this paper, a general and simple plasma-enhanced chemical vapor deposition method is proposed to produce 2D heterolayer-structured MoSe 2 -carbon (MoSe 2 /C) with carbon atoms inserted in the MoSe 2 layers. After morphology optimization, when applying flat-type MoSe 2 /C-200 nanosheets with an enlarged interlayer spacing of 0.79 nm as the anode and activated carbon as the cathode, the assembled sodium-ion hybrid capacitors can reach a maximum energy/power density of 116.5 W h kg -1 /107.5 W kg -1 and exhibit superior cycling durability (91.3% capacitance retention after 4000 cycles at 1 A g -1 ). The good electrochemical property can be ascribed to the enlarged interlayer spacing that can offer fast diffusion channels for Na ions, and the carbon layer sandwiched in the MoSe 2 layer can not only enhance the electron transfer, accelerating the reaction kinetics, but also alleviate the volume change of MoSe 2 , ensuring the good stability of the electrode. The proposed approach can also be extended to other 2D transition metal chalcogenide (TMC) materials for constructing the TMC/C heterostructures for the application in energy storage systems.

Geothermal energy is new, environmentally friendly, and clean energy, which is of great significance to realize energy saving and emission reduction. The study of the genesis mechanism of geothermal water is the key to its rational development and utilization. In this study, based on 14 sets of water samples from the eastern section of the copper well mining area of Yinan Gold Mine, mineral saturation index, isotope analysis (δ 18 O, δD), Si-enthalpy mixing equation, and geochemical geothermal temperature scale were used to analyze the thermal storage temperature, recharge characteristics, mixing ratio, circulation depth, and fluid passage to reveal the geothermal water fugitive transmission pattern and genesis mechanism in the study area and to propose a geothermal water genesis model. The study shows that the water supply elevation in the area is between 687.22 and 1164.15 m and a large amount of cold water recharged it. It is inferred that the recharge area is the precipitation in the Northwest Mountain range and surrounding atmosphere. Groundwater flows along the fracture zone in a south-easterly direction. It receives heating from the surrounding rocks, where the water level rises at the fracture zone intersection and is stored in the lower and middle Cambrian thermal reservoirs and continues to receive heating from deeper heat sources. Based on this study and previous regional research data, the fault structure in this area is within the influence range of the energy field of the Yishu fault zone. Yishu fault zone becomes the heating source under the background of cold water. The oil development has been oriented toward deep-layer reservoirs and the commingling production and the separate-layer fracturing are important development methods. Currently, limited attention is given to the pressure transient analysis (PTA) of the fractured wells located in a stratified reservoir. Moreover, the proppant is very difficult to move inside the hydraulic fracture in the deep-layer reservoir, leading to the uneven fracture conductivity along the hydraulic fracture and increasing the complexity of PTA. To fill this gap, this work presented a fully analytical well test model for hydraulically fractured wells with changing fracture conductivity in stratified reservoirs, which is convenient to be used for interpreting the recorded pressure data from the oilfield due to its analytical nature. The establishment of this model is based on the trilinear flow model, Duhamel theorem, and pressure superposition principle. A systematic verification is conducted to ensure the validity of the proposed model. Furthermore, we offer a sensitivity analysis to investigate the effect of crucial parameters on pressure and pressure derivative, including the fracture extension, fracture conductivity, transmissibility factor, and storativity factor. Enhancer of zeste homolog 2 (EZH2), a catalytic subunit of polycomb repressive complex 2 (PRC2), is overexpressed in triple-negative breast cancer (TNBC), correlating with poor prognosis. However, EZH2 catalytic inhibitors are ineffective in suppressing the growth of TNBC cells that are dependent on EZH2. Knockdown of EZH2 inhibits the proliferation of these cells, suggesting that EZH2 protein overexpression but not its catalytic activity is critical for driving TNBC progression. Several proteolysis targeting chimera (PROTAC) degraders of EZH2, including the von Hippel-Lindau (VHL)-recruiting PROTAC YM281, have been reported. However, the effects of these EZH2 PROTACs in TNBC cells were not investigated. Here, we report the discovery and characterization of a novel, potent, and selective EZH2 PROTAC degrader, MS8815 (compound 16 ), which induced robust EZH2 degradation in a concentration-, time-, and proteasome-dependent manner in TNBC cells. Activation and polarization of microglia are involved in neuroinflammation and regulate ischemic stroke-associated brain injury. Protein arginine methyltransferase 8 functions as a regulatory component of hypoxic stress-induced neuroinflammation. The protective effect of protein arginine methyltransferase 8 (PRMT8) against ischemic stroke-associated brain injury through regulation of microglia activation and polarization was investigated. First, PRMT8 was downregulated in middle cerebral artery occlusion (MCAO)-induced mice and oxygen-glucose deprivation/reoxygenation (OGD/R)-induced SH-SY5Y. Injection with AAV-PRMT8 reduced infarct volumes in MCAO-induced mice. Moreover, injection with AAV-PRMT8 promoted neuronal survival and ameliorated histopathological changes in the brains of MCAO-induced mice. The neuronal apoptosis and neuroinflammation in MCAO-induced mice were suppressed by AAV-PRMT8 injection. Second, PRMT8 overexpression increased cell viability and suppressed the cell apoptosis and inflammation of OGD/R-induced SH-SY5Y. Third, injection with AAV-PRMT8 reduced almost 50% of CD86 + M1 microglia and enhanced about 20% of CD206 + M2 microglia. Furthermore, PRMT8 overexpression attenuated OGD/R-induced M1 phenotype polarization of BV2. Lastly, PRMT8 upregulated Lin28a and loss of Lin28a attenuated PRMT8 overexpression-induced increase in cell viability and decrease in cell apoptosis and inflammation of OGD/R-induced SH-SY5Y. In conclusion, PRMT8 promoted M2 phenotype polarization of microglia and suppressed neuronal apoptosis to ameliorate cerebral ischemia/reperfusion injury through upregulation of Lin28a.

Background: Conflicting results have been revealed on the relationship between PSMA uptake values (SUVs) on prostate-specific membrane antigen (PSMA) positron emission tomography (PET)/computed tomography (CT) and prostate cancer (PCa) aggressiveness. This study is to validate the relationship between SUVs with PCa aggressiveness and its role in evaluation of clinically significant PCa (csPCa) and risk stratification., Methods: We retrospectively enrolled 51 patients who underwent [68Ga]-PSMA PET/CT (PET/CT) before radical prostatectomy (RP). PET/CT results were corrected with whole mount histology. The relationship between SUVs and aggressiveness related indictors including Gleason score, T stage, initial PSA and tumor size were analyzed. The cutoff value for detection of overall PCa, csPCa and intermediate/high-risk PCa were calculated by receiver operating characteristics (ROC) analysis., Results: Both SUVmax and SUVmean positively correlated with Gleason score (SUVmax Spearman r=0.546 P<0.01, SUVmean Spearman r=0.359 P<0.01), PSA level (SUVmax Spearman r=0.568 P<0.01, SUVmean Spearman r=0.529 P<0.01) and tumor volume SUVmax Spearman r=0.635 P<0.01, SUVmean Spearman r=0.590 P<0.01). Tumors with T3 stage had significant higher SUV uptake than T2 (SUVmax 17.49±10.50 vs 9.90±8.7, P<0.01 and SUVmean 17.49±10.50 vs 9.90±8.7, P<0.01). ROC analysis showed cutoff of SUVmax (3.8) and SUVmean (2.8) for overall PCa detection. ROC analysis showed that csPCa and intermediate/high risk PCa had the same cutoff on both SUVmax (8.4) and SUVmean (6.8)., Conclusions: PSMA uptake on PSMA PET/CT positively correlated with Gleason score, T stage, initial PSA and tumor volume. Both SUVmax and SUVmean can be applied as parameters for csPCa detection and risk classification.

Objective: LncRNA HCG18 has been reported to act as a tumor promoter in gastric cancer, hepatocellular carcinoma and nasopharyngeal carcinoma. However, the role of HCG18 in melanoma is still not clear. In this study, we detected the expression and molecular function of HCG18 in melanoma., Methods: The expression of HCG18 in melanoma cell lines and 50 pairs of melanoma and corresponding non-cancer tissues was detected by RT-qPCR. The relationship between HCG18 and clinicopathology was analyzed. We used HCG18 overexpressing melanoma cell lines A375 and M14, and si-HCG18 to knock down HCG18 expression. CCK-8, clone formation, Transwell assay and FCM were used to explore the effect of HCG18 knockdown on cell proliferation, migration, invasion and apoptosis in melanoma cells. Bioinformatics software was used to predict the downstream miRNA regulated by HCG18, and the downstream target genes regulated by miR-324-5p. Dual-luciferase reporter assay and RNA pull-down assay were used to verify whether miR-324-5p was related to the predicted sequence of HCG18., Results: HCG18 was highly expressed in melanoma tissues and cells. Besides, we found that HCG18 was closely correlated with thickness, TNM stage and metastasis. Functional experiments discovered that HCG18 knockdown restrained cell proliferation, migration and invasion, while promoted cell apoptosis in melanoma cells. HCG18 was confirmed to be a sponge of miR-324-5p, and CDK16 might be a downstream gene of miR-324-5p. HCG18 was found to reverse the effect of miR-324-5p by upregulating CDK16 expression in melanoma cell proliferation, apoptosis, migration and invasion in vitro ., Conclusion: This study indicated that HCG18 played an essential role in the pathogenesis of melanoma and suggested that HCG18 might be a potential target for the treatment and diagnosis of melanoma., Competing Interests: None., (AJTR Copyright © 2022.)

A unique porous TiO 2 with Co 3 O 4 nanoparticles anchored in (Co 3 O 4 @TiO 2 ) is prepared by a dual-templating method for promoting electromagnetic microwave absorption (EMA). The as-prepared Co 3 O 4 @TiO 2 possesses a three-dimensional (3D) ordered macroporous TiO 2 skeleton and plenty of mesopores, as well as small Co 3 O 4 nanoparticles that coexisted in the macropore walls of the TiO 2 skeleton. The introduction of Co 3 O 4 can increase the magnetic loss as well as suppress impedance mismatch, resulting in the regulation of the EMA performance. The synergetic effect of the TiO 2 porous framework and Co 3 O 4 nanoparticles with proper ratio promote microwave absorption performance. Therefore, Co 3 O 4 @TiO 2 -2 with 25 wt % Co 3 O 4 nanoparticles content displays a strong and ultrawide effective absorption band (EAB) performance. The Co 3 O 4 @TiO 2 -2 presents a strong reflection loss of -53.9 dB at 2.95 mm. Moreover, it obtains a super broad EAB of ∼12.5 GHz at 5.0 mm. This dual-templating approach for a well-controlled porous structure could be a facial strategy for the development of high-performance electromagnetic wave absorbers.

The MRGPRX family of receptors (MRGPRX1-4) is a family of mas-related G-protein-coupled receptors that have evolved relatively recently 1 . Of these, MRGPRX2 and MRGPRX4 are key physiological and pathological mediators of itch and related mast cell-mediated hypersensitivity reactions 2-5 . MRGPRX2 couples to both G i and G q in mast cells 6 . Here we describe agonist-stabilized structures of MRGPRX2 coupled to G i1 and G q in ternary complexes with the endogenous peptide cortistatin-14 and with a synthetic agonist probe, respectively, and the development of potent antagonist probes for MRGPRX2. We also describe a specific MRGPRX4 agonist and the structure of this agonist in a complex with MRGPRX4 and G q . Late blight, caused by Phytophthora infestans , is severely damaging to the global tomato industry. Micro-RNAs (miRNAs) have been widely demonstrated to play vital roles in plant resistance by repressing their target genes. Recently, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) method has been continuously improved and extensively applied to edit plant genomes. However, editing multiplex miRNAs by CRISPR/Cas9 in tomato has not been studied yet. We knocked out miR482b and miR482c simultaneously in tomato through the multiplex CRISPR/Cas9 system. Two transgenic plants with silenced miR482b and miR482c simultaneously and one transgenic line with silenced miR482b alone were obtained. Compared with wild-type plants, the disease symptoms of three transgenic plants upon infection were reduced, accompanied by increased expression of their common target nucleotide binding site-leucine-rich repeat genes and decreased levels of reactive oxygen species. Furthermore, silencing miR482b and miR482c simultaneously was more resistant than silencing miR482b alone in tomato. More importantly, we found that knocking out miR482b and miR482c can elicit expression perturbation of other miRNAs, suggesting cross-regulation between miRNAs. Our study demonstrated that editing miR482b and miR482c simultaneously with CRISPR/Cas9 is an efficient strategy for generating pathogen-resistant tomatoes, and cross-regulation between miRNAs may reveal the novel mechanism in tomato- P. infestans interactions.

The organization of genomes in space and time dimension plays an important role in gene expression and regulation. Chromatin folding occurs in a dynamic, structured way that is subject to biophysical rules and biological processes. Nucleosomes are the basic unit of chromatin in living cells, and here we report on the effective interactions between two nucleosomes in physiological conditions using explicit-solvent all-atom simulations. Free energy landscapes derived from umbrella sampling simulations agree well with recent experimental and simulation results. The CRISPR-Cas9 system enables simple, rapid, and effective genome editing in many species. Nevertheless, the requirement of an NGG protospacer adjacent motif (PAM) for the widely used canonical Streptococcus pyogenes Cas9 (SpCas9) limits the potential target sites. The xCas9, an engineered SpCas9 variant, was developed to broaden the PAM compatibility to NG, GAA, and GAT PAMs in human cells. However, no knockout rice plants were generated for GAA PAM sites, and only one edited target with a GAT PAM was reported. In this study, we used tRNA and enhanced sgRNA (esgRNA) to develop an efficient CRISPR-xCas9 genome editing system able to mutate genes at NG, GAA, GAT, and even GAG PAM sites in rice. We also developed the corresponding xCas9-based cytosine base editor (CBE) that can edit the NG and GA PAM sites. We measure the electrical resistivity of hcp iron up to ∼170  GPa and ∼3000  K using a four-probe van der Pauw method coupled with homogeneous flattop laser heating in a DAC, and compute its electrical and thermal conductivity by first-principles molecular dynamics including electron-phonon and electron-electron scattering. We find that the measured resistivity of hcp iron increases almost linearly with temperature, and is consistent with our computations. The results constrain the resistivity and thermal conductivity of hcp iron to ∼80±5  μΩ cm and ∼100±10  W m^{-1} K^{-1}, respectively, at conditions near the core-mantle boundary. Our results indicate an adiabatic heat flow of ∼10±1  TW out of the core, supporting a present-day geodynamo driven by thermal and compositional convection.

In this paper, we study a system with two interacting qubits described by the Heisenberg model with dissipative terms, and analyze decay dynamics and the steady-state of geometric quantum discords. Our results indicate that we can ignore the interaction force in the z-direction and adjust the parameters to change the loss of quantum correlation with time when the initial state satisfies some conditions. Moreover, we show that after a long enough period of time, unlike other parameters, the energy and the intensity of the non-uniform magnetic field do not affect the steady-state.

Although various bio-inspired materials with outstanding mechanical, acoustic, and optic properties have been developed, bio-inspired materials for microwave absorption applications are rarely reported. Herein, under the inspiration of the opal structure, for the first time, a kind of Co@Co 3 O 4 /nitrogen-doped (N-doped) mesoporous carbon sphere (Co@Co 3 O 4 /NMCS) with a periodic three-dimensional structure toward microwave absorption application was designed and synthesized. The microwave absorption performance was optimized with respect to the content of Co@Co 3 O 4 nanoparticles. Co@Co 3 O 4 /NMCS with ∼20 wt % Co@Co 3 O 4 achieves a reflection loss of -53.8 dB at 5.7 GHz. The simulated radar cross section demonstrated that the Co@Co 3 O 4 /NMCS can efficiently suppress the strong electromagnetic scattering from a metal groove structure, which further reveals its excellent absorbing performance. These periodic porous structures of N-doped mesoporous carbon spheres combined with the magnetic Co@Co 3 O 4 nanoparticles contribute to the excellent microwave-absorbing performance.

Background: To compare the diagnostic performance of 68 Ga-prostate-specific membrane antigen positron emission tomography/computed tomography ( 68 Ga-PSMA PET/CT) with multi-parametric magnetic resonance imaging (mpMRI) on extracapsular extension (ECE) and seminal vesicle invasion (SVI) in primary prostate cancer and its impact on therapeutic decisions., Methods: We retrospectively enrolled 54 patients with both PET/CT and mpMRI before radical prostatectomy. Diagnostic performance of mpMRI, PET/CT and their combination (com-MRI/PET) on ECE and SVI on a patient basis were analyzed. The impact of additional PET/CT scanning on therapeutic decisions were presented., Results: Among the 54 patients, 17 had tumor limited in the prostate gland, 25 only had ECE and 12 patients had both SVI and ECE on pathology. The sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) of ECE were 54%, 94%, 95%, 48% on mpMRI, 78%, 94%, 97%, 67% on PET/CT and 83%, 88%, 94%, 71% on com-MRI/PET. Both PET/CT and com-MRI/PET had a higher sensitivity than mpMRI on ECE diagnosis (78% vs. 54%, P<0.05 and 83% vs. 54%, P<0.05). No difference was observed between PET/CT and com-MRI/PET (78% vs. 83%, P=0.17). The Sensitivity, specificity, PPV and NPV of SVI were 67%, 93%, 72%, 91% on mpMRI, 75%, 95%, 82%, 93% on PET/CT and 75%, 88%, 64%, 93% on com-MRI/PET. No difference was found between the three scannings. Background: Membranous glomerulonephritis (MGN) is a common kidney disease. Despite many evidences support that many immune and inflammation-related genes could serve as effective biomarkers and treatment targets for MGN patients, the potential associations among MGN-, immune- and inflammation-related genes have not been sufficiently understood., Methods: Here, a global immune-, inflammation- and MGN-associated triplets (IIMATs) network is constructed and analyzed. An integrated and computational approach is developed to identify dysregulated IIMATs for MGN patients based on expression and interaction data., Results: 45 dysregulated IIMATs are identified in MGN by above method. Dysregulated patterns of these dysregulated IIMATs are complex and various. We identify four core clusters from dysregulated IIMATs network and some of these clusters could distinguish MGN and normal samples. Specially, some anti-cancer drugs including Tamoxifen, Bosutinib, Ponatinib and Nintedanib could become candidate drugs for MGN based on drug repurposing strategy follow IIMATs. Functional analysis shows these dysregulated IIMATs are associated with some key functions and chemokine signaling pathway., Conclusions: The present study explored the associations among immune, inflammation and MGN. Some effective candidate drugs for MGN were identified based on immune and inflammation. Overall, these comprehensive results provide novel insights into the mechanisms and treatment of MGN.

Our purpose was to explore whether 68 Ga-PSMA PET/CT alone (PET/CT) or in combination with multiparametric MRI (PET/MRI) can improve the detection of clinically significant prostate cancer (PCa). Methods: We retrospectively enrolled 54 patients who underwent both MRI and PET/CT before radical prostatectomy. Regions of interest on MR images, PET/CT images, and pathologic images were marked. A lesion was defined as a region of interest marked on images obtained with any of the 3 modalities. All lesions were characterized using the prostate imaging reporting and data system (PI-RADS), the molecular imaging PSMA expression score, and the pathologic results and analyzed. Diagnostic performance was analyzed by receiver-operating-characteristic analysis. Specific improvement for lesions with different PI-RADS scores was analyzed using the net reclassification index (NRI). Results: In total, 90 lesions from 54 patients were analyzed, among which 66 lesions represented clinically significant PCa. Receiver-operating-characteristic analysis showed PET/MRI to perform better than MRI in detecting clinically significant PCa (change in area under the curve, 0.06; 95% confidence interval, 0.01-0.12; P < 0.05). With the calculated cutoff, PET/MRI performed significantly better than MRI (NRI, 21.9%; P < 0.01), with an improvement in sensitivity (89% vs. 76%, P < 0.01) at no sacrifice of specificity (96% vs. 88%, P > 0.05). Improvement in diagnosing clinically significant PCa occurred for lesions classified as PI-RADS 3 (NRI, 66.7%; P < 0.01). Purpose: To explore the diagnostic performance of 68 Ga-PSMA PET/CT for identification of pathological cribriform morphology in prostate cancer (PCa)., Methods: The study retrospectively enrolled 49 PCa patients who had undergone preoperative multiparametric MRI (mpMRI) and 68 Ga-PSMA PET/CT, and who had Gleason pattern (GP) 4 and absence of GP 5 on radical prostatectomy specimens. Lesions with GP 4 were outlined and stratified according to their cribriform status. Volumes of interest were drawn on matched mpMRI and PET images, and parameters including average apparent diffusion coefficient (ADC mean ), tenth percentile ADC (ADC 10% ) and maximum standardized uptake value (SUV max ) were derived. The Mann-Whitney U test was used for continuous variables and the chi-squared test for categorical variables. Receiver operating characteristic analysis was used to compare imaging parameters in identifying cribriform morphology. The associations between cribriform-positive PCa and imaging variables were evaluated in a univariate analysis using a logistic regression model., Results: A total of 62 lesions were identified in 49 patients with GP 4. Of these lesions, 37 (59.7%) in 34 patients (69.4%) showed cribriform morphology. ADC mean and ADC 10% were similar between cribriform-positive and non-cribriform groups (P > 0.05), while SUV max was significantly different (median SUV max 18.3 vs. 9.4 per patient, P = 0.003, 18.2 vs. 7.2 per lesion, P < 0.001), yielding sensitivities and specificities of 76% and 86% in a per-patient analysis, and 77% and 88% in a per-lesion analysis, respectively. Further, PSMA was significantly overexpressed in cribriform-positive PCa (P = 0.003). SUV max was a significant predictor of cribriform morphology in PCa (odds ratio 8.61, 95% confidence interval 4.96-25.27, per patient; odds ratio 11.93, 95% confidence interval 6.49-33.74, per lesion; both P < 0.001)., Conclusion: 68 Ga-PSMA PET/CT effectively identifies the aggressive cribriform morphology in PCa.

Background: To evaluate the diagnostic value of 68 Ga-PSMA-11 PET-CT with multiparametric magnetic resonance imaging (mpMRI) for lymph node (LN) staging in patients with intermediate- to high-risk prostate cancer (PCa) undergoing radical prostatectomy (RP) with pelvic lymph node dissection (PLND)., Methods: We retrospectively identified 42 consecutive patients with intermediate- to high-risk PCa according to D'Amico and without concomitant cancer. Preoperative 68 Ga-PSMA-11 PET-CT, pelvic mpMRI and subsequent robot assisted laparoscopic RP with PLND were performed in all patients., Results: Among 42 patients assessed, the preoperative PSA value, Gleason score, pT stage and intraprostatic PCa volume of patients with LN metastases were all significantly higher than those without metastases (P = 0.029, 0.028, 0.004, respectively). The average maximum standardized uptake value (SUV) of 68 Ga-PSMA-11 PET-CT positive PCa of patients with or without LN metastases were 13.10 (range 6.12-51.75) and 7.22 (range 5.4-11.2), respectively (P < 0.001). 68 Ga-PSMA-11 PET-CT and pelvic mpMRI had the ability of succeed on preoperative definite accurate diagnosis and accurate localization of primary PCa in all 42 patients. Fifteen patients (35.71%) had a pN1 stage. 51 positive LN were found. Both 68 Ga-PSMA-11 PET-CT and pelvic mpMRI displayed brillient patient-based and region-based sensitivity, specificity, negative predictive value and positive predictive value. There was no statistical difference for the detection of LNMs according to the diameter of the LNMs between 68 Ga-PSMA-11 PET-CT and mpMRI in this study., Conclusions: Both 68 Ga-PSMA-11 PET-CT and mpMRI performed great value for LN staging in patients with intermediate- to high-risk PCa undergoing RP with PLND. However, despite excellent performance of 68 Ga-PSMA-11 PET-CT, it cannot replace mpMRI that remains excellent for lymph node staging.

The aqueous sodium-ion battery (ASIB) is one of the promising new energy storage systems owing to the abundant resources of sodium as well as efficiency and safety of electrolyte. Herein, we report an ASIB system with Na 4 Mn 9 O 18 /carbon nanotube (NMO/CNT) as cathode, metal Zn as anode and a novel Na + /Zn 2+ mixed ion as electrolyte. The NMO/CNT with microspherical structure is prepared by a simple spray-drying method. The prepared battery delivers a high reversible specific capacity and stable cyclability. Furthermore, the battery displays a stable reversible discharge capacity of 53.2 mAh g -1 even at a high current rate of 4 C after 150 cycles. Our results confirm that the NMO/CNT composite is a promising electrode cathode material for ASIBs.

Human androgen receptor (AR) contains a highly polymorphic polyglutamine tract encoded by CAG repeats [(CAG)n] in exon 1 of the AR gene. The CAG repeats, ranging from 11 to 38, have been reported to be inversely correlated with AR activity. A case-control study involving 261 polycystic ovary syndrome (PCOS) patients and 278 healthy controls was conducted. Fluorescently labeled DNA fragments containing (CAG)n were obtained by PCR and genotyped via capillary electrophoresis. AR (CAG)n ranges were 6, 12-28 in PCOS cases and 9, 10, 12-32 in controls. In the PCOS group, a higher frequency of short (CAG)n alleles was found compared with that of controls (P=0.007). Similarly, CAG biallelic mean distributions also showed statistical difference between the two groups (P=0.025). In conclusion, shorter alleles of the (CAG)n in exon 1 of the AR gene enhanced the susceptibility to PCOS, either by upregulating AR activity or by causing hyperandrogenism.

SSR markers are desirable markers in analysis of genetic diversity, quantitative trait loci mapping and gene locating. In this study, SSR markers were developed from two genomic libraries enriched for (GA)n and (CA)n of foxtail millet [Setaria italica (L.) P. Beauv.], a crop of historical importance in China. A total of 100 SSR markers among the 193 primer pairs detected polymorphism between two mapping parents of an F(2) population, i.e. "B100" of cultivated S. italica and "A10" of wild S. viridis. Excluding 14 markers with unclear amplifications, and five markers unlinked with any linkage group, a foxtail millet SSR linkage map was constructed by integrating 81 new developed SSR markers with 20 RFLP anchored markers. The 81 SSRs covered nine chromosomes of foxtail millet. The length of the map was 1,654 cM, with an average interval distance between markers of 16.4 cM. The 81 SSR markers were not evenly distributed throughout the nine chromosomes, with Ch.8 harbouring the least (3 markers) and Ch.9 harbouring the most (18 markers). To verify the usefulness of the SSR markers developed, 37 SSR markers were randomly chosen to analyze genetic diversity of 40 foxtail millet accessions. Totally 228 alleles were detected, with an average 6.16 alleles per locus. Polymorphism information content (PIC) value for each locus ranged from 0.413 to 0.847, with an average of 0.697. A positive correlation between PIC and number of alleles and between PIC and number of repeat unit were found [0.802 and 0.429, respectively (P < 0.01)]. UPGMA analysis revealed that the 40 foxtail millet cultivars could be grouped into five clusters in which the landraces' grouping was largely consistent with ecotypes while the breeding varieties from different provinces in China tended to be grouped together.
