Considering the probable health risks due to radioactivity input via drinking tea, the concentrations of 226Ra, 232Th,40K and 137Cs radionuclides in the soil and the corresponding tea leaves of a large tea plantation were measured using high purity germanium (HPGe) γ-ray spectrometry. Different layers of soil and fresh tea leaf samples were collected from the Udalia Tea Estate (UTE) in the Fatickchari area of Chittagong, Bangladesh. The mean concentrations (in Bq/kg) of radionuclides in the studied soil samples were found to be 34 ± 9 to 45 ± 3 for 226Ra, 50 ± 13 to 63 ± 5 for 232Th, 245 ± 30 to 635 ± 35 for 40K and 3 ± 1 to 10 ± 1 for 137Cs, while the respective values in the corresponding tea leaf samples were 3.6 ± 0.7 to 5.7 ± 1.0, 2.4 ± 0.5 to 5.8 ± 0.9, 132 ± 25 to 258 ± 29 and 226Ra, 232Th and 40K from soil to tea leaves were calculated to be 0.12, 0.08 and 0.46, respectively, the complete range being 1.1 × 10−2 to 1.0, in accordance with IAEA values. Additionally, the most popularly consumed tea brands available in the Bangladeshi market were also analyzed and, with the exception of 40K, were found to have similar concentrations to the fresh tea leaves collected from the UTE. The committed effective dose via the consumption of tea was estimated to be low in comparison with the United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) reference ingestion dose limit of 290 μSv/y. Current indicative tea consumption of 4 g/day/person shows an insignificant radiological risk to public health, while cumulative dietary exposures may not be entirely negligible, because the UNSCEAR reference dose limit is derived from total dietary exposures. This study suggests a periodic monitoring of radiation levels in tea leaves in seeking to ensure the safety of human health.

The use of thorium in providing the intense white luminescence emitted from gas mantles, has a history of some 130 years, the initial application pre-dating by several decades large-scale urban electric lighting. Accordingly, the thoriated gas mantle has proved itself to be of enormous utility, remaining popular in more rural areas well into the 20th century, continuing to enjoy use in campsites and street night markets lanterns until today. The discovery of thorium in 1828 preceded the discovery of radioactivity, with subsequent little appreciation initially of any potential harm from exposure to radioactivity. Study has been made herein of small quantities of five different types of the thoriated gas mantle, all purchased online devoid of any control measures. Several approaches were used concerning the 232Th activity and dose consequence. First, the activity of 232Th was estimated using an HPGe detector, with sample M5 providing the greatest activity at 1.25 × 104 Bq, exceeding the exemption limit for thorium in a mantle. Compared to sample M5, samples M1 to M4 were low in radioactivity, from 5.1 ± 1.31 to 16.33 ± 1.92 Bq. Moreover, the thorium content in M5 constituted 50% of the mantle mass, somewhat greater than previous literature values. The dose equivalent rate on the surface of a single M5 mantle was found to be 0.68 µSv/h, while at the surface of a pack of six the level was 1.9 µSv/h. Monte Carlo simulation codes have been used to obtain organ equivalent and effective dose rates, the greatest close contact (10 cm) exposure to an unlit mantle being to the thymus, at 0.68 µSv/h and 0.62 µSv/h for a male and female phantom respectively. Accordingly, with packages of thoriated gas mantles potentially giving rise to non-negligible equivalent doses, greater incorporation of controls on the sale of such items in national radiation protection legislation would seem worthy of consideration.

Waterbirds are often used as indicators of ecosystem function across broad spatial and temporal scales. Resolving which species are declining and the ecological characteristics they have in common can offer insights into ecosystem changes and their underlying mechanisms. Using 20 years of citizen science data collected by the British Columbia Coastal Waterbird Survey, we examine species-specific trends in abundance of 50 species in the Salish Sea and 37 species along the outer Pacific Ocean coast that we considered to form the core wintering coastal bird community of British Columbia, Canada. Further, we explore whether ecological commonalities increase the likelihood of a species undergoing declines by testing the hypotheses that waterbird abundance trends are influenced by dietary specialization and migration distance to breeding grounds. Results suggest that most populations are stable (i.e., temporal trends are not significant) in both the Salish Sea (36 of 50 spp.) and Pacific coast (32 of 37 spp.) regions. Twelve species displayed significant decline trends in the Salish Sea, whereas two had significant increases. Along the Pacific coast, only three species displayed significant decline trends, and two significant increases. This result is corroborated by guild-specific trends indicating that waterbirds occupying the Salish Sea are faring significantly worse than those residing along the outer coastal regions, almost irrespective of dietary specialization or migration distance. Our results provide evidence that differential environmental pressures between the inner and outer coastal regions may be causing overall loss of wintering waterbirds within, or abundance shifts away from, the Salish Sea. Potential mechanisms responsible for these observed patterns are discussed, including environmental (e.g., climate) and human-induced (e.g., nutrient and chemical pollution) pressures. Collaborative, inter-disciplinary research priorities to help understand these mechanisms are suggested.

Many shorebirds (Order: Charadriiformes; Family: Charadriidae, Recurvirostridae, Scolopacidae, Haematopodidae, Jacanidae) are highly migratory, traversing thousands of kilometers between high latitude breeding and low latitude nonbreeding sites. In doing so, they are dependent on networks of coastal and interior wetland ecosystems. To aid in the effective conservation and management of their populations, and to assess the impact of threats facing shorebirds, standardized data on shorebird abundance are needed from multiple sites representing a gradient of conditions across the hemisphere. Such data would provide insight on whether fluctuations at one location represent real changes in abundance because of some localized threat, or whether other factors acting across broader scales such as the redistribution of predators, are responsible. We designed the Migratory Shorebird Project (MSP), now implemented in 11 countries along the Pacific Coast of the Americas, to characterize spatial and temporal variation in shorebird abundance, to understand which factors (habitat, threats) most influence their populations across the flyway, and to increase capacity for integrated research, monitoring, and conservation. We used bird inventory data from historical surveys and spatial data on habitat distribution along with a set of hypotheses about important threats to guide the design of the project and to identify data requirements to test hypotheses. We counted birds one time per year at 84 nonbreeding sites (~1400 sampling units) between 15 November and 15 February, when shorebirds were relatively stationary. In each of the first three annual counts from 2013/14 to 2015/16, the Migratory Shorebird Project counted ~1M shorebirds representing 44 species, including five species for which > 20% of the estimated biogeographic population was recorded annually, and nine additional species with > 5% recorded annually. The magnitude of variability in estimates of shorebird abundance was inversely correlated with survey effort with the most uncertainty, and lowest survey effort, in the South Temperate region followed by the Neotropical region (southern Mexico to northern Peru) and then the North Temperate region. Evaluation of variance highlighted both among-site and among-unit ("units" are nested within sites) variation in bird abundance and cover types as well as threats such as potential disturbance and predator abundance. Overall, shorebird density was significantly, and positively, associated with the area (ha) of intertidal mudflats, beaches, and aquaculture. Survey units with intermediate levels of bare ground and flooding had the highest shorebird density. As for threats, we found, contrary to our hypothesis, that shorebird abundance was significantly, and positively, associated, with the density of Peregrine Falcons (Falco peregrinus); however we found no relationship between shorebird abundance and the amount of urban development, our index to potential human disturbance, in the surrounding landscape. The Migratory Shorebird Project is providing an essential data foundation and network for increased knowledge of the factors affecting shorebirds across the Pacific Coast of the Americas and identifying what and where conservation actions could have the greatest impact.

Impacts of climate change adaptation strategies need to be evaluated using principled methods spanning sectors and longer time frames. We propose machine-learning approaches to study the long-term impacts of flood protection in Bangladesh. Available data include socio-economic survey and events data (death, migration, etc.) from 1983−2014. These multidecadal data, rare in their extent and quality, provide a basis for using machine-learning approaches even though the data were not collected or designed to assess the impact of the flood control investments. We test whether the embankment has affected the welfare of people over time, benefiting those living inside more than those living outside. Machine-learning approaches enable learning patterns in data to help discriminate between two groups: here households living inside vs. outside. They also help identify the most informative indicators of discrimination and provide robust metrics to evaluate the quality of the model. Overall, we find no significant difference between inside/outside populations based on welfare, migration, or mortality indicators. However, we note a significant difference in inward/outward movement with respect to the embankment. While certain data gaps and spatial heterogeneity in sampled populations suggest caution in any conclusive interpretation of the flood protection infrastructure, we do not see higher benefits accruing to those living with higher levels of protection. This has implications for Bangladesh’s planning for future and more extreme climate futures, including the national Delta Plan, and global investments in climate resilient infrastructure to create positive social impacts.

A biomedical sciences graduate program needed an introductory class that would develop skills for students interested in a wide variety of disciplines, such as microbiology or cancer biology, and a diverse array of biomedical careers. Faculty created a year-long student-centered course, Scientific Discovery, to serve this need. The course was divided into four modules with progressive skill outcomes. Each module had a focus related to each of the major research areas of the collective faculty: molecular biology, biochemistry, neuroscience, and infectious disease. First-year graduate students enter the program with relevant college-level biology and chemistry coursework but not in-depth content knowledge of any of the focus areas. Each module features a biomedical problem for the students to gain specific content knowledge while developing skills outcomes, such as the ability to conduct scholarly inquiry. In 2015, the theme of the infectious disease module was to create an effective human vaccine to prevent Lyme disease. The module required students to learn fundamental concepts of microbiology and immunology and then apply that knowledge to design their own Lyme disease vaccine. The class culminated with students communicating their creative designs in the form of a “white paper” and a pitch to “potential investors.” By the end of the module, students had developed fundamental knowledge, applied that knowledge with great creativity, and met the skills learning outcomes, as evidenced by their ability to conduct scholarly inquiry and apply knowledge gained during this module to a novel problem, as part of their final exam.

Study has been undertaken of the thermoluminescence (TL) yield of various tailor-made flat cross-section 6 mol% Ge-doped silica fibers, differing only in respect of external dimensions. Key TL dosimetric characteristics have been investigated, including glow curves, dose response, sensitivity, fading and reproducibility. Using a 60Co source, the samples were irradiated to doses within the range 1 to 10 Gy. Prior to irradiation, the flat fibers were sectioned into 6 mm lengths, weighed, and annealed at 400 °C for 1 h. TL readout was by means of a Harshaw Model 3500 TLD reader, with TLD-100 chips (LiF:Mg, Ti) used as a reference dosimeter to allow the relative response of the fibers to be evaluated. The fibers have been found to provide highly linear dose response and excellent reproducibility over the range of doses investigated, demonstrating high potential as TL-mode detectors in radiation medicine applications. Mass for mass, the results show the greatest TL yield to be provided by fibers of the smallest cross-section, analysis indicating this to be due to minimal light loss in transport of the TL through the bulk of the silica medium.

Dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) are severe disease manifestations that can occur following sequential infection with different dengue virus serotypes (DENV1-4). At present, there are no licensed therapies to treat DENV-induced disease. DHF and DSS are thought to be mediated by serotype cross-reactive antibodies that facilitate antibody-dependent enhancement (ADE) by binding to viral antigens and then Fcγ receptors (FcγR) on target myeloid cells. Using genetically engineered DENV-specific antibodies, it has been shown that the interaction between the Fc portion of serotype cross-reactive antibodies and FcγR is required to induce ADE. Additionally, it was demonstrated that these antibodies were as neutralizing as their non-modified variants, were incapable of inducing ADE, and were therapeutic following a lethal, antibody-enhanced infection. Therefore, we hypothesized that avian IgY, which do not interact with mammalian FcγR, would provide a novel therapy for DENV-induced disease. We demonstrate here that goose-derived anti-DENV2 IgY neutralized DENV2 and did not induce ADE in vitro. Anti-DENV2 IgY was also protective in vivo when administered 24 hours following a lethal DENV2 infection. We were also able to demonstrate via epitope mapping that both full-length and alternatively spliced anti-DENV2 IgY recognized different epitopes, including epitopes that have not been previously identified. These observations provide evidence for the potential therapeutic applications of goose-derived anti-DENV2 IgY.

The Zika virus (ZIKV) is a newly emerged pathogen in the Western hemisphere. It was declared a global health emergency by the World Health Organization in 2016. There have been 223,477 confirmed cases, including 3720 congenital syndrome cases since 2015. ZIKV infection symptoms range from asymptomatic to Gullain–Barré syndrome and extensive neuropathology in infected fetuses. Passive and active vaccines have been unsuccessful in the protection from or the treatment of flaviviral infections due to antibody-dependent enhancement (ADE). ADE causes an increased viral load due to an increased monocyte opsonization by non-neutralizing, low-avidity antibodies from a previous dengue virus (DENV) infection or from a previous exposure to ZIKV. We have previously demonstrated that polyclonal avian IgY generated against whole-killed DENV-2 ameliorates DENV infection in mice while not inducing ADE. This is likely due to the inability of the Fc portion of IgY to bind to mammalian Fc receptors. We have shown here that ZIKV oligoclonal IgY is able to neutralize the virus in vitro and in IFNAR−/− mice. The concentration of ZIKV-specific IgY yielding 50% neutralization (NT50) was 25 µg/mL. The exposure of the ZIKV, prior to culture with ZIKV-specific IgY or 4G2 flavivirus-enveloped IgG, demonstrated that the ZIKV-specific IgY does not induce ADE. ZIKV IgY was protective in vivo when administered following a lethal ZIKV challenge in 3-week-old IFNAR−/− mice. We propose polyclonal ZIKV-specific IgY may provide a viable passive immunotherapy for a ZIKV infection without inducing ADE.

Religious and spiritual struggles (R/S struggles)—tension or conflicts regarding religious or spiritual matters—have been robustly linked to greater psychological distress and lower well-being. Most research in this area has relied on samples consisting predominantly of participants who believe in god(s). Limited research has examined R/S struggles among atheists, generally conflating them with agnostics and other nontheists. This study investigated the prevalence of R/S struggles among atheists and compared atheists to theists in two samples (3978 undergraduates, 1048 Internet workers). Results of a multilevel model showed that atheists experience less demonic, doubt, divine, moral, and overall R/S struggles than theists, but similar levels of interpersonal and ultimate meaning struggles. Correlation and regression analyses among atheists demonstrated links between moral, ultimate meaning, and overall R/S struggles and greater distress (depression and anxiety symptoms) as well as lower well-being (life satisfaction and meaning in life). Even after controlling neuroticism, ultimate meaning struggles continued to predict lower well-being and higher distress across samples; moral struggles also predicted distress independently. This study demonstrates the relevance of R/S struggles to atheists and reinforces the applicability of previous results to atheist samples, but also highlights substantial differences between atheists and theists in certain R/S struggles.

In neurocysticercosis, parasite-induced immune suppressive effects are thought to play an important role in enabling site-specific inhibition of inflammatory responses to infections. It is axiomatic that microglia-mediated (M1 proinflammatory) response causes central nervous system inflammation; however, the mechanisms by which helminth parasites modulate microglia activation remain poorly understood. Here, we show that microglia display a diminished expression of M1-inflammatory mediators such as tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and nitric oxide synthase 2 (NOS2) in murine neurocysticercosis. Microglia also exhibited a lack of myeloid cell maturation marker major histocompatibility complex (MHC)-II in these parasite-infected brains. Treatment of microglia with helminth soluble/secreted factors (HSFs) in vitro did not induce expression of M1-inflammatory signature molecule NOS2 as well as MHC-II in primary microglia. However, HSF treatment completely inhibited lipopolysaccharide-induced increase in expression of MHC-II, NOS2 and nitric oxide production in these cells. As epigenetic modulation of chromatin states that regulates recruitment of RNA polymerase II (Pol-II) is a key regulatory step in determining gene expression and functional outcome, we next evaluated whether HSF induced modulation of these phenomenon in microglia in vitro . Indeed, HSF downregulated Pol-II recruitment to the promoter region of TNF-α, IL-6, NOS2, MHC-II, and transcription factor CIITA (a regulator of MHC-II expression), by itself. Moreover, HSF suppressed the lipopolysaccharide-induced increase in Pol-II recruitment as well. In addition, HSF exposure reduced the positive histone marks H3K4Me3 and H3K9/14Ac at the promoter of TNF-α, IL-6, NOS2, MHC-II, and CIITA. These studies provide a novel mechanistic insight into helminth-mediated immune suppression in microglia via modulation of epigenetic processes.

Despite benefits of using light-sensitive geolocators to track animal movements and describe patterns of migratory connectivity, concerns have been raised about negative effects of these devices, particularly in small species of aerial insectivore. Geolocators may act as handicaps that increase energetic expenditure, which could explain reported effects of geolocators on survival. We tested this ‘Energetic Expenditure Hypothesis’ in 12 populations of tree swallows (Tachycineta bicolor) and barn swallows (Hirundo rustica) from North America and Europe, using measurements of corticosterone from feathers (CORTf) grown after deployment of geolocators as a measure of physiology relevant to energetics. Contrary to predictions, neither among- (both species) nor within-individual (tree swallows only) levels of CORTf differed with respect to instrumentation. Thus, to the extent that CORTf reflects energetic expenditure, geolocators apparently were not a strong handicap for birds that returned post-deployment. While this physiological evidence suggests that information about migration obtained from returning geolocator-equipped swallows is unbiased with regard to levels of stress, we cannot discount the possibility that corticosterone played a role in reported effects of geolocators on survival in birds, and suggest that future studies relate corticosterone to antecedent factors, such as reproductive history, and to downstream fitness costs.

Detection of buried improvised explosive devices (IEDs) is a delicate task, leading to a need to develop sensitive stand-off detection technology. The shape, composition and size of the IEDs can be expected to be revised over time in an effort to overcome increasingly sophisticated detection methods. As an example, for the most part, landmines are found through metal detection which has led to increasing use of non-ferrous materials such as wood or plastic containers for chemical based explosives being developed.Monte Carlo simulations have been undertaken considering three different commercially available detector materials (hyperpure-Ge (HPGe), lanthanum(III) bromide (LaBr) and thallium activated sodium iodide (NaI(Tl)), applied at a stand-off distance of 50 cm from the surface and burial depths of 0, 5 and 10 cm, with sand as the obfuscating medium. Target materials representing medium density wood and mild steel have been considered. Each detector has been modelled as a 10 cm thick cylinder with a 20 cm diameter.It appears that HPGe represents the most promising detector for this application. Although it was not the highest density material studied, its excellent energy resolving capability leads to the highest quality spectra from which detection decisions can be inferred.The simulation work undertaken here suggests that a vehicle-born threat detection system could be envisaged using a single betatron and a series of detectors operating in parallel observing the space directly in front of the vehicle path. Furthermore, results show that non-ferrous materials such as wood can be effectively discerned in such remote-operated detection system, with the potential to apply a signature analysis template matching technique for real-time analysis of such data.

Andes virus (ANDV) and ANDV-like viruses are responsible for most hantavirus pulmonary syndrome (HPS) cases in South America. Recent studies in Chile indicate that passive transfer of convalescent human plasma shows promise as a possible treatment for HPS. Unfortunately, availability of convalescent plasma from survivors of this lethal disease is very limited. We are interested in exploring the concept of using DNA vaccine technology to produce antiviral biologics, including polyclonal neutralizing antibodies for use in humans. Geese produce IgY and an alternatively spliced form, IgYΔFc, that can be purified at high concentrations from egg yolks. IgY lacks the properties of mammalian Fc that make antibodies produced in horses, sheep, and rabbits reactogenic in humans. Geese were vaccinated with an ANDV DNA vaccine encoding the virus envelope glycoproteins. All geese developed high-titer neutralizing antibodies after the second vaccination, and maintained high-levels of neutralizing antibodies as measured by a pseudovirion neutralization assay (PsVNA) for over 1 year. A booster vaccination resulted in extraordinarily high levels of neutralizing antibodies (i.e., PsVNA80 titers >100,000). Analysis of IgY and IgYΔFc by epitope mapping show these antibodies to be highly reactive to specific amino acid sequences of ANDV envelope glycoproteins. We examined the protective efficacy of the goose-derived antibody in the hamster model of lethal HPS. α-ANDV immune sera, or IgY/IgYΔFc purified from eggs, were passively transferred to hamsters subcutaneously starting 5 days after an IM challenge with ANDV (25 LD50). Both immune sera, and egg-derived purified IgY/IgYΔFc, protected 8 of 8 and 7 of 8 hamsters, respectively. In contrast, all hamsters receiving IgY/IgYΔFc purified from normal geese (n=8), or no-treatment (n=8), developed lethal HPS. These findings demonstrate that the DNA vaccine/goose platform can be used to produce a candidate antiviral biological product capable of preventing a lethal disease when administered post-exposure.

The administration of bone marrow-derived stem cells may provide a new treatment option for patients with heart failure. Transcatheter cell injection may require multi-imaging modalities to optimize delivery. This study sought to evaluate whether endomyocardial injection of mesenchymal precursor cells (MPCs) could be guided by real-time 3D echocardiography (RT3DE) in treating chronic, postinfarction (MI) left ventricular (LV) dysfunction in sheep. Four weeks after induction of an anterior wall myocardial infarction in 39 sheep, allogeneic MPCs in doses of either 25 × 10 6 ( n = 10), 75 × 10 6 ( n = 9), or 225 × 10 6 ( n = 10) cells or nonconditioned control media ( n = 10) were administered intramyocardially into infarct and border zone areas using a catheter designed for combined fluoroscopic and RT3DE-guided injections. LV function was assessed before and after injection. Infarct dimension and vascular density were evaluated histologically. RT3DE-guided injection procedures were safe. Compared to controls, the highest dose MPC treatment led to increments in ejection fraction (3±3% in 225M MPCs vs. −5±4% in the control group, p < 0.01) and wall thickening in both infarct (4±4% in 225M MPCs vs. −3±6% in the control group, p = 0.02) and border zones (4±6% in 225M MPCs vs. −8±9% in the control group, p = 0.01). Histology analysis demonstrated significantly higher arteriole density in the infarct and border zones in the highest dose MPC-treated animals compared to the lower dose or control groups. Endomyocardial implantation of MPCs under RT3DE guidance was safe and without observed logistical obstacles. Significant increases in LV performance (ejection fraction and wall thickening) and neovascularization resulted from this technique, and so this technique has important implications for treating patients with postischemic LV dysfunction.

The Religious and Spiritual Struggles Scale (RSS) measures important psychological constructs in an underemphasized section of the overlap between religion and well-being. Are religious/spiritual struggles distinct from religiousness, distress, and each other? To test the RSS’ internal discriminant validity, we replicated the original six-factor measurement model across five large samples (N = 5705) and tested the fit of a restricted bifactor model, which supported the mutual viability of multidimensional and unidimensional scoring systems for the RSS. Additionally, we explored a bifactor model with correlated group factors that exhibited optimal fit statistics. This model maintained the correlations among the original factors while extracting a general factor from the RSS. This general factor’s strong correlations with religious participation and belief salience suggested that this factor resembles religiousness itself. Estimating this general factor seemed to improve Demonic and Moral struggles’ independence from religiousness, but did not change any factor’s correlations with neuroticism, depression, anxiety, and stress. These distress factors correlated with most of the independent group factors corresponding to the original dimensions of the RSS, especially Ultimate Meaning and Divine struggles. These analyses demonstrate the discriminant validity of religious/spiritual struggles and the complexity of their relationships with religiousness and distress.

Tumours that are low in oxygen (hypoxic) tend to be more aggressive and respond less well to treatment. Knowing the spatial distribution of oxygen within a tumour could therefore play an important role in treatment planning, enabling treatment to be targeted in such a way that higher doses of radiation are given to the more radioresistant tissue. Mapping the spatial distribution of oxygen in vivo is difficult. Radioactive tracers that are sensitive to different levels of oxygen are under development and in the early stages of clinical use. The concentration of these tracer chemicals can be detected via positron emission tomography resulting in a time dependent concentration profile known as a tissue activity curve (TAC). Pharmaco-kinetic models have then been used to deduce oxygen concentration from TACs. Some such models have included the fact that the spatial distribution of oxygen is often highly inhomogeneous and some have not. We show that the oxygen distribution has little impact on the form of a TAC; it is only the mean oxygen concentration that matters. This has significant consequences both in terms of the computational power needed, and in the amount of information that can be deduced from TACs.
