About 100 recently published original papers, reviews, books and other communications concerning cannabis (hashish, marijuana) constituents have been reviewed with the aim of summarizing the status of analytical detection and quantitation. Detailed protocols of standard analytical methods are compared in order to recommend uniform methods for field and forensic samples and also to provide guidance to less experienced analysts in countries where cannabis sativa occurs (T. Maylon.In Big Deal: The Politics of the Illicit Drug Business, the Cannabis Commodity Market, pp. 63-107. Guernsey, London.). Because of its importance, there has been an increasing number of investigations of the chemical, botanical, pharmacological, clinical and sociological aspects of the marijuana (hashish) problem. Since analytical techniques have improved substantially during the last 10 years, many papers have been published containing a variety of methods for detection and quantification of cannabis constituents. Since the analytical situation is becoming increasingly confused and because many of the journals are unavailable in less developed countries, the aim of this paper is to give an overview of existing analytical techniques and to attempt to distinguish practical and effective methods from those which are complex or which provide questionable results.

Glucuronide conjugates of cannabidiol (CBD), 7-hydroxy-CBD, propyl-CBD, cannabinol (CBN), 7-hydroxy-CBN, CBN-7-oic acid, propyl CBN and cannabichromene have been identified as major metabolites of CBD, CBN and their propyl homologues and of cannabichromene in mouse liver. Trace amounts of the glucuronide conjugates of delta1- and delta1(6)-tetrahydrocannabinol (THC) were also detected. Identification was made by combined gas-liquid chromatographic and mass spectrometric studies of the trimethylsilyl (TMS), d9-TMS and methyl ester-TMS derivatives of the glucuronides and the TMS derivatives of the product of the reduction of the metabolites with lithium aluminium deuteride.

The percentage rate of change into cannabinoids (Cannabidiol [CBD], tetrahydrocannabinol [THC] and cannabinol [CBN]) was higher in cannabis samples than in the extracts. This is probalby due to the decomposition of acids into corresponding neutral cannabinoids under the conditions of storage. Previous claims that CBD content in plant material is relatively constant are not substantiated by our results. There was a 1.0-2.5-fold increase in CBD content in plant material compared with the extracts. However, the fact that there was no appreciable increase in CBD/CBN content in the stored extracts of the same samples supports the view that the step-wise extraction does not bring the acids into the final extract pure delta 9THC decomposed at a rate of 41 per cent per year under tropical storage conditions. The delta 9THC content decreased in the samples and equally in the extracts though 100 per cent conversion of THC to CBN does not take place. The higher CBN content found in extracts than that expected by the conversion THC to CBN is a result of metabolic conversion.

Two further generations of Cannabis plants have been grown from seeds produced by earlier generation plants which were raised in the United Kingdom of Great Britain and Northern Ireland in 1980 and 1981. The original seedstocks were from known countries of origin. Although there are exceptions, both the physical and chemical characteristics of the third and fourth generation plants generally resemble those of their parents. The yields of cannabis vary substantially from year to year. The total tetrahydrocannabinol contents also vary, but are comparable with the levels in the original plants. Tetrahydrocannabinolic acid continues to predominate over free tetrahydrocannabinol and is much higher than in the original plants.

High-pressure liquid chromatography was used to study (a) the relative efficiencies of methanol, chloroform, light petroleum (B.P. 40-60 degrees) and methanol-chloroform (9:1) for extracting neutral and acidic cannabinoids from cannabis resin; (b) the decomposition patterns of the resulting solutions under various storage conditions, and (c) the cannabinoid profile of a cross section through a block of cannabis resin. The results show that (a) methanol is the most effective extracting solvent of those tested; (b) acidic cannabinoids in solution decompose in darkness by varying amounts depending on the temperature, solvent, storage time and particular cannabinoid; (c) neutral cannabinoids in solution are relatively stable in darkness; (d) daylight causes appreciable decomposition of both acidic and neutral cannabinoids in solution, (e) the cannabinoid profile of a resin is complex with lower levels of acidic material in the outer layers.

Authors investigated some fiber and some hashish hemp sorts concerning their trichomes on the leaves. Histochemical reactions were developed using the Fast Blue Salt (FBB) reagent applied so far only thin layer chromatography. Glandular hairs were found giving positive reactions due to cannabinoids contained by the cells. The electron microscopic features were studied and the cannabinoid content was measured with GC. A correlation was found between the number of typical glandular hairs and cannabinoid content.

We have examined the cannabinoid contents of seedlings from twelve strains of Cannabis of known chemical race. Fourteen days after emergence of the shoot those of the tetrahydrocannabinol (THC) type could be distinguished from those of the cannabidiol (CBD) type. The true leaves of the THC type contained a relatively high content of THC and also contained cannabichromene (CBC), sometimes as the major cannabinoid, whereas the CBD type had much lower amounts of THC and no CBC; CBD being the major component. A strain from China corresponded to the THC type but showed some unusual features. Seeds purchased at seven outlets in Britain were also examined. Only two batches germinated by these proved to be of a THC type and resembled the Chinese strain.

Intermittent use of low-potency cannabis is not generally associated with obvious toxicity. In recent years, some cannabis users have been using high doses more frequently and seeking treatment. A large but incomplete literature indicates that cannabis can be harmful to health, and that virtually every system in the body is affected by cannabis. Cannabis pharmacology is complex; all recent scientific groups reviewing cannabis conclude it is clearly capable of causing adverse effects to health. Specific treatment procedures have not been developed for managing cannabis related problems. Strategies useful in treating other drug dependence problems are useful in treating dependence.

Blood or urine specimens from nearly 600 fatally injured drivers in two Los Angeles County studies were analyzed for the presence of alcohol and other drugs of abuse, including PCP, cocaine, opiates and marijuana. The results of the preliminary study indicate that 65 out of 102 fatally injured drivers had used alcohol and/or another drug of abuse - 34 had used alcohol only, 12 had used one or more other drug(s) of abuse, and 19 had used alcohol in combination with another drug of abuse. The results of the larger follow-up study, begun a year later, indicate a continued high level of both alcohol use (41.5%) and marijuana use (19%) with moderate cocaine usage (8%) and low levels (less than 2%) of barbiturate and PCP usage.
