Currently, ultrashort oligopeptides consisting of fewer than eight amino acids represent a cutting-edge frontier in materials science, particularly in the realm of hydrogel formation. By employing solid-phase synthesis with the Fmoc/tBu approach, a novel pentapeptide, FEYNF-NH2, was designed, inspired by a previously studied sequence chosen from hen egg-white lysozyme (FESNF-NH2). Qualitative peptide analysis was based on reverse-phase high performance liquid chromatography (RP-HPLC), while further purification was accomplished using solid-phase extraction (SPE). Exact molecular ion confirmation was achieved by matrix-assisted laser desorption–ionization mass spectrometry (MALDI-ToF MS) using two different matrices (HCCA and DHB). Additionally, the molecular ion of interest was subjected to tandem mass spectrometry (MS/MS) employing collision-induced dissociation (CID) to confirm the synthesized peptide structure. A combination of research techniques, including Fourier-transform infrared spectroscopy (FTIR), fluorescence analysis, transmission electron microscopy, polarized light microscopy, and Congo red staining assay, were carefully employed to glean valuable insights into the self-assembly phenomena and gelation process of the modified FEYNF-NH2 peptide. Furthermore, molecular docking simulations were conducted to deepen our understanding of the mechanisms underlying the pentapeptide’s supramolecular assembly formation and intermolecular interactions. Our study provides potential insights into amyloid research and proposes a novel peptide for advancements in materials science. In this regard, in silico studies were performed to explore the FEYNF peptide’s ability to form polyplexes.

The pathogenic cascade of Alzheimer's disease (AD) characterized by amyloid-β protein accumulation is still poorly understood, partially owing to the limitations of relevant models without in vivo neural tissue microenvironment to recapitulate cell–cell interactions. To better mimic neural tissue microenvironment, three-dimensional (3D) core-shell AD model constructs containing human neural progenitor cells (NSCs) with 2% matrigel as core bioink and 2% alginate as shell bioink have been bioprinted by a co-axial bioprinter, with a suitable shell thickness for nutrient exchange and barrier-free cell interaction cores. These constructs exhibit cell self-clustering and -assembling properties and engineered reproducibility with long-term cell viability and self-renewal, and a higher differentiation level compared to 2D and 3D MIX models. The different effects of 3D bioprinted, 2D, and MIX microenvironments on the growth of NSCs are mainly related to biosynthesis of amino acids and glyoxylate and dicarboxylate metabolism on day 2 and ribosome, biosynthesis of amino acids and proteasome on day 14. Particularly, the model constructs demonstrated Aβ aggregation and higher expression of Aβ and tau isoform genes compared to 2D and MIX controls. AD model constructs will provide a promising strategy to facilitate the development of a 3D in vitro AD model for neurodegeneration research.

The calix[4]arene scaffold, blocked in the cone conformation through alkylation with long alkyl chains, and decorated at the upper rim with four guanidine or arginine units, effectively catalyzes the cleavage of the phosphodiester bond of DNA and RNA model compounds in water. An exhaustive kinetic investigation unequivocally points to the existence of spontaneous aggregation phenomena, driven by hydrophobic effect, occurring at different critical concentrations that depend on the identity of the compound. A pronounced superiority of the assembled structures compared with the monomers in solution was observed. Moreover, the catalytically active units, clustered on the macrocyclic tetrafunctional scaffold, were proved to efficiently cooperate in the catalytic mechanism and result in improved reaction rates compared to those of the monofunctional model compounds. The kinetic analysis is also integrated and corroborated with further experiments based on fluorescence spectroscopy and light scattering. The advantage of the supramolecular assemblies based on tetrafunctional calixarenes leads to believe that the active units can cooperate not only intramolecularly but also intermolecularly. The molecules in the aggregates can probably mold, flex and rearrange but, at the same time, keep an ordered structure that favors phosphodiester bond cleavage. Peptides and their related compounds can self-assemble into diverse nanostructures of different shapes and sizes in response to various stimuli such as pH, temperature or ionic strength. Here we report the synthesis and characterization of a lysozyme derived pentapeptide and its ability to build well-defined fibrillar structures. Lysozyme FESNF peptide fragment was synthesized by solid phase peptide synthesis using the Fmoc/t-Bu strategy, purified by analytical high-performance liquid chromatography (HPLC) and its molecular weight was confirmed by matrix-assisted laser desorption/ionization mass spectrometry (MALDI–MS). Spectroscopic features of this pentapeptide were investigated by UV-visible spectroscopy and fluorimetry showing the pattern of marginal phenylalanine residues within the peptide sequence. Self-assembling properties were determined using atomic force microscopy (AFM), aggregation index and thioflavin T assay (ThT). FESNF generating fibrillar structures observed by AFM and aggregation propensity were primarily influenced by pH conditions. Moreover, the experimental data were confirmed by molecular dynamics simulation studies. With increasing heat accumulation in advanced modern electronic devices, dielectric materials with high thermal conductivity (λ) and excellent electrical insulation have attracted extensive attention in recent years. Inspired by mussel, hexagonal boron nitride (hBN) and graphene oxide (GO) are assembled to construct mhBN@GO hybrids with the assistance of poly(catechol-polyamine). Then, mhBN@GO hybrids are dispersed in carboxy nitrile rubber (XNBR) latex via emulsion coprecipitation to form elastomer composites with a high λ and satisfactory insulating properties. Thanks to the uniform dispersion of mhBN@GO hybrids, the continuous heat conduction pathways exert a significant effect on enhancing the λ and decreasing the interface thermal resistance of XNBR composites. In particular, the λ value of 30 vol% mhBN@GO/XNBR composite reaches 0.4348 W/(m·K), which is 2.7 times that of the neat XNBR (0.1623 W/(m·K)). Meanwhile, the insulating hBN platelets hinder the electron transfer between adjacent GO sheets, leading to satisfactory electrical insulation in XNBR composites, whose AC conductivity is as low as 10−10 S/cm below 100 Hz. This strategy opens up new prospects in the assembly of ceramic and carbonaceous fillers to prepare dielectric elastomer composites with high λ and satisfactory electrical insulation, making them promising for modern electrical systems. Protein-based materials intended as nanostructured drugs or drug carriers are progressively gaining interest in nanomedicine, since their structure, assembly and cellular interactivity can be tailored by recruiting functional domains. The main bottleneck in the development of deliverable protein materials is the lysosomal degradation that follows endosome maturation. This is especially disappointing in the case of receptor-targeted protein constructs, which, while being highly promising and in demand in precision medicines, enter cells via endosomal/lysosomal routes. In the search for suitable protein agents that might promote endosome escape, we have explored the translocation domain (TD) of the diphtheria toxin as a functional domain in CXCR4-targeted oligomeric nanoparticles designed for cancer therapies. The pharmacological interest of such protein materials could be largely enhanced by improving their proteolytic stability. The incorporation of TD into the building blocks enhances the amount of the material detected inside of exposed CXCR4+ cells up to around 25-fold, in absence of cytotoxicity. This rise cannot be accounted for by endosomal escape, since the lysosomal degradation of the new construct decreases only moderately. On the other hand, a significant loss in the specificity of the CXCR4-dependent cellular penetration indicates the unexpected role of the toxin segment as a cell-penetrating peptide in a dose-dependent and receptor-independent fashion. These data reveal that the diphtheria toxin TD displayed on receptor-targeted oligomeric nanoparticles partially abolishes the exquisite receptor specificity of the parental material and it induces nonspecific internalization in mammalian cells. In this paper, the tailoring of superhydrophobic surfaces on AA6082 aluminum alloy by chemical etching in an HF/HCl solution, followed by silane self-assembling, was applied for enhanced corrosion protection in the marine field. In particular, different etching times were considered in order to optimize the treatment effect. The results indicate that all the prepared surfaces, after silanization, were characterized by superhydrophobic behavior with a contact angle higher than 150°. The contact and sliding angles strongly depend on the surface morphology at varying etching times. The optimum was observed with an etching time of 20 s, where a microscale coral-like structure coupled with a homogeneous and ordered pixel-like nanostructure was obtained on the aluminum surface showing a Cassie–Baxter superhydrophobic behavior with a water contact angle of 180° and a sliding angle equal to 0°. All superhydrophobic surfaces achieved an enhanced corrosion protection efficiency and impedance modulus up to two orders of magnitude higher than the as-received AA6082 in simulated seawater. After more than a century since its initial development, Bacille Calmette-Guérin (BCG) remains the only licensed vaccine against tuberculosis (TB). Subunit boosters are considered a viable strategy to enhance BCG efficacy, which often wanes in adolescence. While many studies on booster subunit vaccines have concentrated on recombinant proteins, here we developed a novel modular peptide-based subunit vaccine platform that is flexible, cold-chain independent and customizable to diverse circumstances and populations. Each individual peptide building block consists of a linear arrangement comprising a 15-leucine self-assembly inducer moiety, a Mycobacterium tuberculosis (Mtb) target epitope and an human leukocyte antigen E (HLA-E) binding moiety, with each moiety separated by a triple lysine spacer. The building blocks, in any combination, are able to form a multiepitope nanoparticle. Six Mtb epitopes were selected to produce the self-assembling and self-adjuvating peptide-based TB nano-vaccine candidate PNx6. In vivo vaccination-challenge experiments demonstrated that subcutaneous boost of parenteral BCG immunization with PNx6 significantly enhanced its immunogenicity and improved its protective efficacy in a murine model of TB by more than 5-fold. Achieving self‐assembling structures that exhibit emergent properties and functionalities is one of the holy grails in nanotechnology and holds great promise for a wide range of potential applications. Herein, a self‐assembled LuFeO3–(1/3)LuFe2O4–LuFeO3 heterostructure that consists of two model multiferroics, i.e., hexagonal LuFeO3 and rhombohedral LuFe2O4, is reported in single‐layer epitaxial LuFeO3 thin films. Atomic‐scale electron microscopy investigations reveal the spontaneous organization of ferroelectric polarization in each building block, which forms a series of polar configurations. Specifically, the charge and lattice of neighboring LuFeO3 and LuFe2O4 blocks are closely coupled with competing degrees of freedom. A combination of the quantitative analysis of atomically resolved images and spectroscopy with theoretical calculations determines how charge‐ordering‐induced polarization accommodates charge naturality across the interfaces to reduce the system energy and unravels the formation mechanism of this heterostructure. The results point to the possibilities of generation and control of multiferroic self‐assembled systems, which may provide a basis for engineering novel materials with emergent properties. Polarized beam infrared (IR) spectroscopy provides valuable information on changes in the orientation of samples in nematic phases, especially on the role of intermolecular interactions in forming the periodically modulated twist–bent phase. Infrared absorbance measurements and quantum chemistry calculations based on the density functional theory (DFT) were performed to investigate the structure and how the molecules interact in the nematic (N) and twist–bend (NTB) phases of thioether dimers. The nematic twist–bend phase observed significant changes in the mean IR absorbance. On cooling, the transition from the N phase to the NTB phase was found to be accompanied by a marked decrease in absorbance for longitudinal dipoles. Then, with further cooling, the absorbance of the transverse dipoles increased, indicating that transverse dipoles became correlated in parallel. To investigate the influence of the closest neighbors, DFT calculations were performed. As a result of the optimization of the molecular cores system, we observed changes in the square of the transition dipoles, which well corresponds to absorbance changes observed in the IR spectra. A self-assembled layered compound of formula {[Cu5(OH)8Cyt2]·2ClO4}n (1) was obtained from the direct reaction between copper perchlorate and cytosine. Thanks to its peculiar structural features, this coordination compound can be classified as an intermediate nonconventional analog of Layered Simple Hydroxides (LSHs) and Layered Double Hydroxides (LDHs), two fascinating classes of inorganic materials with variable chemical composition and structures. Like LDHs, 1 consists of positively charged layers of metal hydroxides and non-framework charge-balancing anions located between the layers. However, the hydroxide layers, similar to LSHs, contain only divalent copper(II) metal ions, supported by cytosine molecules with long contacts. Cytosine coordinates to the metal centers adopting an unusual µ3 coordination mode, the exocyclic oxygen atom simultaneously binding to three adjacent copper(II) centers. This coordination mode has not been observed with transition metal ions. This work further demonstrates that nucleobases and their derivatives, despite presenting a limited pool of molecules, can potentially afford a significantly expanded set of compounds with diverse nanostructures, owing to their multiple metal binding sites of different coordination properties and their ability to direct assembly of highly ordered structures. A method for constructing a novel offshore platform was proposed. Based on the cooperative control of multiple unmanned vessels, a self-assembling platform was realized, which could be reconfigured into different shapes according to requirements. A docking controller was designed to realize the docking of two modules. A connecting rod and electromagnetic forces were adopted to complete the docking and reduce the difficulty. Besides, a test site was constructed using the conditions of pool, and a model test was performed to verify the functions of the proposed self-assembling platform. The results show that the design can realize the docking of any unmanned vessel. Compared with the single platform, this self-assembling platform can perform more complex tasks, whose decentralized design makes it more flexible and reliable.

Abstract Superlattices of nanoparticles are generally produced based on solution chemistry processes. In this paper, we demonstrate that self-assembled monolayer structures of nanoparticles with superlattice periodicities can also be produced on template-free surfaces in the gas-phase cluster beam deposition process. It is found that the packing of Fe nanoparticles corresponds to an average of two-dimensional densely packed lattice with a hexagonal summary. By controlling the nanoparticle coverage, the two-dimensional densely packed monolayer morphology can spread to the whole substrate surface being deposited. A formation mechanism of the ordered monolayers is proposed. The densely packed morphologies are formed by the balance between the diffusion rate of the nanoparticles and their filling speed on the substrate surface determined by the deposition rate, and the ordering of the nanoparticle arrays is driven by the inter-particle attractive interactions. The model is strongly supported by a series of carefully designed cluster deposition experiments.

Due to the lack of blood vessels, nerves and lymphatic vessels, the capacity of articular cartilage to heal is extremely limited. Once damaged, it is urgent for articular cartilage to repair the injury. In recent years, there has been an increase in cartilage tissue engineering studies. Self-assembling peptide hydrogel as a kind of hydrogels composed of peptides and water is widely used in cartilage tissue engineering. Under noncovalent interactions such as electrostatic interaction, hydrophobic interaction, hydrogen bonding and pi-pi stacking force, peptides self-assemble into three-dimensional (3D) structures that mimic the natural extracellular matrix and allow cells to grow, proliferate and differentiate. Because SAPHs have excellent biocompatibility and biodegradability, variable mechanical properties, low immunogenicity, injectability, and the ability to load cells and bioactive substances, many researchers utilized them to promote the repair and regeneration of articular cartilage after damage. Therefore, the purpose of this review is to sum up the composition, injury characteristics, and treatments of articular cartilage, as well as the action of SAPHs in repairing articular cartilage damage.

The superhydrophobic scallop-like Cu6Sn5 micro/nano structure were successfully constructed on pure copper substrate by heat-induced Cu/Sn interfacial metallurgical reaction and simple chemical modification process. The microstructure, chemical composition and corrosion resistance of as-prepared samples were characterized by field emission scanning electron microscopy and X-ray photoelectron spectroscopy, etc. The results show that the averaged shear resistance strength between micro Cu6Sn5 and Cu substrate is higher than 40 MPa. After modified with myristic acid and Cu2+, copper myristate with low surface energy is formed on the armor Cu6Sn5 surface. The wetting angle of water droplets on as-modified surface is higher than 150° and its rolling angle is 7.2°. Compared with pure copper, the self corrosion current density of the samples modified by copper myristate in 3.5%NaCl(mass fraction) solution is about 1/10 of that of the samples before modification,showing better corrosion resistance. Based on the metallurgical bonding mechanism between intermetallic compounds and metal matrix, the armor formation strategy of metal materials in micro-scale by heat-induced interfacial reaction is proposed, which can successfully solve the open and key scientific problem of low mechanical stability for artificial superhydrophobic interface.

[Display omitted] The aim of this study was the evaluation of suitability of novel mucoadhesive hydrogel platforms for the delivery of therapeutics useful for the management of disorders related to the gastrointestinal tract (GI). At this purpose, here we describe the preparation, the physicochemical characterization and drug delivery behaviour of novel hydrogels, based on self-assembling lipopeptides (MPD02-09), obtained by covalently conjugating lauric acid (LA) to SNA's peptide derivatives gotten by variously combining D- and L- amino acid residues. LA conjugation was aimed at improving the stability of the precursor peptides, obtaining amphiphilic structures, and triggering the hydrogels formation through the self-assembling. Budesonide (BUD), an anti-inflammatory drug, was selected as model because of its use in the treatment in GI disorders. Preliminary studies were performed to correlate the chemical structure of the conjugates with the key physicochemical properties of the materials for drug delivery. Two lipopeptides, MPD03 and MPD08 , were found to form hydrogels (MPD03h and MPD08h , respectively) with characteristics suitable for drug delivery. These materials showed mucoadhesiveness of about 60 %. In vitro studies carried out with BUD loaded hydrogels showed about 70 % drug release within 6 h. Wound healing assessed in Caco-2 and HaCaT cells, showed reduction of cell-free area to values lower than 10 %. Taking together these results MPD03h and MPD08h have been shown to be excellent candidates for BUD delivery. [ABSTRACT FROM AUTHOR]

Tissue regeneration is a prominent area of research, developing biomaterials aimed to be tunable, mechanistic scaffolds that mimic the physiological environment of the tissue. These biomaterials are projected to effectively possess similar chemical and biological properties, while at the same time are required to be safely and quickly degradable in the body once the desired restoration is achieved. Supramolecular systems composed of reversible, non-covalently connected, self-assembly units that respond to biological stimuli and signal cells have efficiently been developed as preferred biomaterials. Their biocompatibility and the ability to engineer the functionality have led to promising results in regenerative therapy. This review was intended to illuminate those who wish to envisage the niche translational research in regenerative therapy by summarizing the various explored types, chemistry, mechanisms, stimuli receptivity, and other advancements of supramolecular systems. Surface-enhanced Raman spectroscopy (SERS) is regarded as a versatile tool for studying the composition and structure of matter. This work has studied the preparation of a SERS substrate based on a self-assembling plasmonic nanoparticle film (SPF) in a polymer matrix. Several synthesis parameters for the SPF are investigated, including the size of the particles making up the film and the concentration and type of the self-assembling agent. The result of testing systems with different characteristics is discussed using a model substance (pseudoisocyanin iodide). These models can be useful in the study of biology and chemistry. Research results contain the optimal parameters for SPF synthesis, maximizing the SERS signal. The optimal procedure for SPF assembly is determined and used for the synthesis of composite SPFs within different polymer matrices. SPF in a polymer matrix is necessary for the routine use of the SERS substrate for various types of analytes, including solid samples or those sensitive to contamination. Polystyrene, polyvinyl alcohol (PVA), and polyethylene are investigated to obtain a polymer matrix for SPF, and various methods of incorporating SPF into a polymer matrix are being explored. It is found that films with the best signal enhancement and reproducibility were obtained in polystyrene. The minimum detectable concentration for the SERS substrate obtained is equal to 10−10 M. We prepared a SERS substrate with an analytical enhancement factor of 2.7 × 104, allowing an increase in the detection sensitivity of analyte solutions of five orders of magnitude. Ferritins are ideal nanoparticles as drug delivery systems due to their hollow-sphere structure and the ability to target specific receptors on the cell surface. Here, we develop and characterize a new ferritin derived from the chimeric humanized A. fulgidus one, already designed to recognize the TfR1 receptor. Starting from the synthetic gene of this chimeric protein, we replaced two positively charged amino acids with two alanine residues to close the large triangular pores on its surface. These mutations make the protein nanoparticle suitable to incorporate even small therapeutics without leakage. Size-exclusion chromatography shows that the assembling/disassembling of this new protein cage can be easily fine-tuned by varying the HEPES buffer and MgCl2 concentration. The protein cage can be opened using 150 mM HEPES buffer without magnesium ions. Adding this divalent cation to the solution promotes the quick assembly of the ferritin as a 24-mer. The development of this new protein cage paves the way for encapsulation and delivery studies of small molecules for therapeutic and diagnostic purposes. [ABSTRACT FROM AUTHOR]

We report the synthesis and characterization of three di-copper compounds, [Cu2phen2CMP(µ-OH)(µ-X)]·3H2O (with X = Cl− (1) and Br− (2)) and [Cu2phen2CMP(µ-OH)(µ-NO3)]·[Cu2phen2CMP(µ-OH)(NO3)]·7H2O (3). 1 and 3 present a dinuclear core assembled by an unusual bis-monodentate phosphate and µ2-hydroxo double bridge, further supported by bridging anions. The spontaneous self-assembly of an extended coordination structure of higher dimensionality, although possible in principle, has been hindered by the use of strongly coordinating counterions (Cl−, Br−) and, unexpectedly, even by the less coordinating nitrate ions. The neutral complexes feature unsaturated coordination sites, having both the N(3) and O(2) of the cytosine moieties of the CMP ligands still available for coordination. π···π Stacking and hydrogen bonding interactions established between the discrete chiral entities mainly drive the self-assembling of a 3D supramolecular network. Small hydrophilic voids are defined within the 3D net, which are filled by solvent water molecules. These small nuclearity complexes with coordinatively unsaturated CMP ligands are likely to act as tectons. They could be used as complex-as-ligands for further binding additional metal ions in a predictable manner, favoring a more rational synthetic approach aimed to a stricter structural and chemical control of the resulting material. We present a new method for the surface capture of proteins in cell-free protein synthesis (CFPS). We demonstrate the spontaneous self-assembly of the protein BslA into functionalizable surfaces on the surface of a CFPS reaction chamber. We show that proteins can be covalently captured by such surfaces, using “Catcher/Tag” technology. Importantly, proteins of interest can be captured either when synthesised in situ by CFPS above the BslA surfaces, or when added as pure protein. The simplicity and cost efficiency of this method suggest that it will find many applications in cell-free-based methods.

Rice straw cellulose was completely defibrillated via aqueous counter collision (ACC) at a low energy input of 15 kWh/kg, then fractionated by differential centrifugation into four increasing weight fractions of progressively thinner cellulose nanofibrils (CNFs): 6.9% in 80-200 nm, 14.4% in 20-80 nm, 20.3% in 5-20 nm, and 58.4% in less than 5 nm thickness. The 93.1% less than 80 nm or 78.7% less than 20 nm thick CNFs yields were more than double those from wood pulp by other mechanical means but at a lower energy input. The smallest (3.7 nm thick and 5.5 nm wide) CNFs were only a third or less in lateral dimensions than those obatined through ACC processed from wood pulp, bamboo, and microbial cellulose pellicle. The less than 20 nm thick CNFs could self-assemble into continuous submicron (136 nm) wide fibers by freezing and freeze-drying or semitransparent (13-42% optical transmittance) film by ultrafiltration and air-drying with excellent mechanical properties (164 MPa tensile strength, 4 GPa Young's modulus, and 16% strain at break). ACC defibrillated CNFs retained essentially the same chemical and crystalline structures and thermal stability as the original rice straw cellulose and therefore were much more thermally stable than TEMPO oxidized CNFs and sulfuric acid hydrolyzed cellulose nanocrystals from the same rice straw cellulose.

As a novel type of antibiotic alternative, peptide-based antibacterial drug shows potential application prospects attributable to their unique mechanism for lysing the membrane of pathogenic bacteria. However, peptide-based antibacterial drugs suffer from a series of problems, most notably their immature stability, which seriously hinders their application. In this study, self-assembling chimeric peptide nanoparticles (which offer excellent stability in the presence of proteases and salts) are constructed and applied to the treatment of bacterial infections. In vitro studies are used to demonstrate that peptide nanoparticles NPs1 and NPs2 offer broad-spectrum antibacterial activity and desirable biocompatibility, and they retain their antibacterial ability in physiological salt environments. Peptide nanoparticles NPs1 and NPs2 can resist degradation under high concentrations of proteases. In vivo studies illustrate that the toxicity caused by peptide nanoparticles NPs1 and NPs2 is negligible, and these nanoparticles can alleviate systemic bacterial infections in mice and piglets. The membrane permeation mechanism and interference with the cell cycle differ from that of antibiotics and mean that the nanoparticles are at a lower risk of inducing drug resistance. Collectively, these advances may accelerate the development of peptide-based antibacterial nanomaterials and can be applied to the construction of supramolecular nanomaterials. Preparation and characterization of a block-like l , d -octapeptide-dextran conjugate DEX 29 -(l -Val- d -Val) 4 self-assembling into nanowire structures is reported. The conjugate was prepared by solid phase click-chemistry on an alkyne group N-terminus functionalized peptide with a regularly alternating enantiomeric sequence. Low molecular weight dextran (X n = 29) with moderately low dispersity (1.30) was prepared by controlled acid hydrolysis and dialysis with selected cut-off and functionalized with an azido group on the reducing end by reductive amination. The strong hydrogen bonds and hydrophobic interactions of the (l -Val- d -Val) 4 linear peptide drive the conjugate to self-assemble into long (0.1–1 μm) nanowires. Composite nanoparticles (NPs) consisting of lignin and different polysaccharide (PS) derivatives were prepared. In this synergistic approach, the PS derivative acts as biocompatible matrix that forms spherical NPs while lignin is a functional compound with therapeutic potential (e.g., antioxidative, antimicrobial, antiviral). Organosolv lignin and three different PS derivatives (cellulose acetate/CA, cellulose acetate phthalate/CAPh, xylan phenyl carbonate/XPC) were used in this study. Nanocomposites with particle sizes in the range of about 200–550 nm containing both types of biopolymers are accessible by dialysis of organic PS/lignin solutions against water. In particular, XPC and CAPh, which both contain aromatic substituents, were found to be suitable for incorporation of lignin within the PS nanomatrix. The present work paves the way for future studies in which the pharmaceutical potential and biocompatibility of composite NPs of lignin and PS derivatives with tailored properties are investigated. The electrical properties of C60F18 fluorofullerene molecules with a high electric dipole moment during their physical adsorption on various surfaces were studied in order to investigate the possibility of creating interfaces with specified physical and chemical characteristics and controlling their properties. Spatial maps of the electrostatic potential and electric field strength of a single molecule were obtained. The distribution of the electrostatic potential of single-layer and multilayer, close-packed and sparse, as well as ordered and disordered ensembles of fluorofullerene molecules C60F18 earlier observed in experiments, and their effect on the substrate were modeled using the density functional theory. The calculations confirmed the experimentally established splitting of the F1s level in an electric field directed along the axis of symmetry of the molecule (linear Stark effect). Based on the nature of the interaction of the adsorbate molecules with the substrate and with each other, the influence of collective electrostatic effects on the monolayer structure and the shift of the core electronic levels, conclusions were drawn about the presence of local electric fields above the island film and in the near-surface region of the substrate, and the film growth process. Skeletal conditions represent a considerable challenge to health systems globally. Barriers to effective therapeutic development include a lack of accurate preclinical tissue and disease models. Most recently, work was attempted to present a novel whole organ approach to modeling human bone and cartilage tissues. These self‐assembling skeletal organoids mimic the cellular milieu and extracellular organization present in native tissues. Bone organoids demonstrated osteogenesis and micro vessel formation, and cartilage organoids showed evidence of cartilage development and maturation. Skeletal organoids derived from both bone and cartilage tissues yielded spontaneous polarization of their cartilaginous and bone components. Using these hybrid skeletal organoids, we successfully generated "mini joint" cultures, which we used to model inflammatory disease and test Adenosine (A2A) receptor agonists as a therapeutic agent. The work and respective results indicated that skeletal organoids can be an effective biological model for tissue development and disease as well as to test therapeutic agents. Self-assembling non-immunoglobulin scaffold proteins are a promising class of nanoscale carriers for drug delivery and interesting alternatives to antibody-based carriers that are not sufficiently efficient in systemic administration. To exploit their potentialities in clinics, protein scaffolds need to be further tailored to confer appropriate targeting and to overcome their potential immunogenicity, short half-life in plasma and proteolytic degradation. We have here engineered three human scaffold proteins as drug carrier nanoparticles to target the cytokine receptor CXCR4, a tumoral cell surface marker of high clinical relevance. The capability of these scaffolds for the selective delivery of Monomethyl auristatin E has been comparatively evaluated in a disseminated mouse model of human, CXCR4+ acute myeloid leukemia. Monomethyl auristatin E is an ultra-potent anti-mitotic drug used against a range of hematological neoplasias, which because of its high toxicity is not currently administered as a free drug but as payload in antibody-drug conjugates. The protein nanoconjugates generated here offer a collective strength of simple manufacturing process , high proteolytic and structural stability and multivalent ligand receptor interactions that result in a highly efficient and selective delivery of the payload drug and in a potent anticancer effect. The appeal of combining polybenzimidazole (PBI) and graphene oxide (GO) for the manufacturing of membranes is increasingly growing, due to their versatility. Nevertheless, GO has always been used only as a filler in the PBI matrix. In such context, this work proposes the design of a simple, safe, and reproducible procedure to prepare self-assembling GO/PBI composite membranes characterized by GO-to-PBI (X:Y) mass ratios of 1:3, 1:2, 1:1, 2:1, and 3:1. SEM and XRD suggested a homogenous reciprocal dispersion of GO and PBI, which established an alternated stacked structure by mutual π-π interactions among the benzimidazole rings of PBI and the aromatic domains of GO. TGA indicated a remarkable thermal stability of the composites. From mechanical tests, improved tensile strengths but worsened maximum strains were observed with respect to pure PBI. The preliminary evaluation of the suitability of the GO/PBI X:Y composites as proton exchange membranes was executed via IEC determination and EIS. GO/PBI 2:1 (IEC: 0.42 meq g−1; proton conductivity at 100 °C: 0.0464 S cm−1) and GO/PBI 3:1 (IEC: 0.80 meq g−1; proton conductivity at 100 °C: 0.0451 S cm−1) provided equivalent or superior performances with respect to similar PBI-based state-of-the-art materials.

In this work we present self-organized regular patterns in a solution system through uphill-diffusion. Micrometer thick organic semiconductor solution is sandwiched between a substrate and cover-plate. Self-assembled regular patterns can be observed on the substrate after solvent evaporation. Different micro-patterns and pattern defects were displayed and analyzed. Mechanisms of defect formation, mode selection process during patten generation, and pattern sedimentation onto substrate from solution were proposed. Gels created by self-assembly of small organic molecules are dynamic soft materials that have unique properties and demanding characterization. Four chiral gelators, with two valinol- or leucinoloxamido arms attached to the 2,2'-positions of the proatropisomeric biphenyl group were chosen to show that the electrospray ionization mass spectrometry (ESI-MS) could be used to differentiate the gelation feature of the chiral compounds 1-4 and also to shed light on the gelation processes. By inspecting the gelation of several solvents, we showed that 1 ( R, R ) proved to be the most efficient gelator, forming the largest observable assemblies in the gas phase. The strong intermolecular H-bonds hold single-charged assemblies consisting of up to five monomer units detectable by ESI MS. Enantiomer 1 ( R, R ) is a good gelator due to favorable intramolecular interactions that remain preserved in the gas phase. C ompound 3 ( meso ) does not have gelator properties and detected signals of larger assemblies in the gas phase. So, the detected signals correlate with the conformations of the studied compounds. MS could be used to elucidate the preferential type of noncovalent interaction due to the chiral recognition. Recombinant adeno-associated viruses (rAAVs) have emerged as important tools for gene therapy and, more recently, vaccine development. Nonetheless, manufacturing can be costly and time-consuming, emphasizing the importance of alternative production platforms. We investigate the potential of E. coli -based cell-free protein synthesis (CFPS) to produce recombinant AAV5 virus-like particles (VLPs). AAV5 virus protein 3 (VP3) constructs, both with and without Strep-tag II, were expressed with CFPS. Lower reaction temperatures resulted in increased solubility, with the untagged variant containing nearly 90% more soluble VLP VP3 protein at 18 °C than at 37 °C. Affinity chromatography of N-terminally Strep(II)-tagged VP3 enabled successful isolation with minimal processing. DLS and TEM confirmed the presence of ∼20 nm particles. Furthermore, the N-terminally tagged AAV5 VP3 VLPs were biologically active, successfully internalizing into HeLa cells. This study describes an innovative approach to AAV VLP production using E. coli -based CFPS, demonstrating its potential for rapid and biologically active AAV VLP synthesis.

Organic semiconductors have been recognized as a new generation of photocatalysts for pollutants degradation and energy production. Herein, organic heterojunction (TpMa/CN) consisting of carbon nitride and β-ketoenamine-based covalent organic framework is fabricated via a controllable self-assembly approach. The as-prepared TpMa/CN heterojunctions show enlarged visible-light absorption. The optimum TpMa/CN-5 photocatalyst achieves the highest photocatalytic activity towards tetracycline degradation, and its photocatalytic degradation rate is improved by 2.3 and 4.3 times than TpMa and CN, respectively. As a multifunctional photocatalyst, TpMa/CN-5 sample also shows remarkable photocatalytic activity for hydrogen peroxide production (880.494 μM h−1), which is 49 times higher than that of CN. Experimental and theoretical investigations indicate that a built-in electric field is formed at the interface of composite, which enables an accelerated charge transfer and separation. This work develops an effective strategy to design difunctional photocatalyst and deciphers the electronic properties and mechanisms of g-C 3 N 4 -based organic photocatalysts, which spurs further interests for organic heterojunction photocatalysts in the future. Peptide amphiphiles (PA) are highly biocompatible, unique designer molecules capable of self-assembling into nanostructures of diverse morphologies. They are attracting wider scientific interest for lipophilic drug's solubilization and delivery owing to their amphiphilicity. This study reports the synthesis of a lysine containing PA with an ultra-short peptide head and its investigation for self-assembling into nanostructures and drug loading. The PA was synthesized through solution phase approach followed by characterization through mass spectrometry and 1H NMR and 13C NMR spectroscopy. Self-assembling behavior of the synthesized PA was investigated through circular dichroism spectroscopy followed by authentication through Atomic Force microscopy (AFM). UV visible spectrophotometer was used for determination of its critical micelle concentration (CMC). The amphiphile was evaluated for its drug loading potential using cefixime as a hydrophobic drug. Results show that the synthesized PA bearing an ultra-short peptide head has a CMC value of 0.061 mM and was capable of self-assembling into predominantly cylindrical nanostructures in aqueous medium having a size of 12.5 nm. Furthermore, PA loaded 52.26 ± 2.50% of the model hydrophobic drug in its nanostructures without altering its chemical identity. In this work, γ-aminopropyltriethoxy silane (KH550) and γ-(2,3-epoxypropoxy)propyltrimethoxy silane (KH560) were used to modify BN and Al2O3, and described as KH550-BN and KH560-Al2O3 with amino and epoxy group on surface, respectively. KH550-BN and KH560-Al2O3 were filled in polypropylene (PP) matrix by twin-screw extruder. The thermal conductivity of composites increased with increasing catalyst dosage, material residence time in barrel and homogenization section temperature of twin-screw extruder. Hybrid fillers with semi core-shell structure were formed by KH550-BN in-situ reacting with KH560-Al2O3 though amino and epoxy group. KH550-BN and KH560-Al2O3 were pre-assembled in toluene and filled into PP to prepare composites, which thermal conductivity increased with increasing pre-assembling time. The experimental results show that KH550-BN and KH560-Al2O3 do in-situ self-assemble in PP melt. The thermal conductivity of composites with in-situ self-assembling had 21.6% improvement than that of composites with no assembling. Superlattices of nanoparticles are generally produced based on solution chemistry processes. In this paper, we demonstrate that self-assembled monolayer structures of nanoparticles with superlattice periodicities can also be produced on template-free surfaces in the gas-phase cluster beam deposition process. It is found that the packing of Fe nanoparticles corresponds to an average of two-dimensional densely packed lattice with a hexagonal summary. By controlling the nanoparticle coverage, the two-dimensional densely packed monolayer morphology can spread to the whole substrate surface being deposited. A formation mechanism of the ordered monolayers is proposed. The densely packed morphologies are formed by the balance between the diffusion rate of the nanoparticles and their filling speed on the substrate surface determined by the deposition rate, and the ordering of the nanoparticle arrays is driven by the inter-particle attractive interactions. Peptide‐based hydrogels, originated by multiscale self‐assembling phenomenon, have been proposed as multivalent tools in different technological areas. Structural studies and molecular dynamics simulations pointed out the capability of completely aromatic peptides to gelificate if hydrophilic and hydrophobic forces are opportunely balanced. Here, the effect produced by the introduction of a Cys residue in the heteroaromatic sequence of (FY)3 and in its PEGylated variant was evaluated. The physicochemical characterization indicates that both FYFCFYF and PEG8‐FYFCFYF are able to self‐assemble in supramolecular nanostructures whose basic cross‐β motif resembles the one detected in the ancestor (FY)3 assemblies. However, gelification occurs only for FYFCFYF at a concentration of 1.5 wt%. After cross‐linking of cysteine residues, the hydrogel undergoes to an improvement of the rigidity compared to the parent (FY)3 assemblies as suggested by the storage modulus (G') that increases from 970 to 3360 Pa. The mechanical properties of FYFCFYF are compatible with its potential application in bone tissue regeneration. Moreover, the avalaibility of a Cys residue in the middle of the peptide sequence could allow the hydrogel derivatization with targeting moieties or with biologically relevant molecules. Chronic wounds represent a major health burden and drain on medical system. Efficient wound repair is only possible if the dressing materials target simultaneously multiple factors involved in wound chronicity, such as deleterious proteolytic and oxidative enzymes and high bacterial load. Here we develop multifunctional hydrogels for chronic wound management through self-assembling of thiolated hyaluronic acid (HA-SH) and bioactive silver-lignin nanoparticles (Ag@Lig NPs). Dynamic and reversible interactions between the polymer and Ag@Lig NPs yield hybrid nanocomposite hydrogels with shear-thinning and self-healing properties, coupled to zero-order kinetics release of antimicrobial silver in response to infection-related hyalurodinase. The hydrogels inhibit the major enzymes myeloperoxidase and matrix metalloproteinases responsible for wound chronicity in a patient's wound exudate. Furthermore, the lignin-capped AgNPs provide the hydrogel with antioxidant properties and strong antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa. The nanocomposite hydrogels are not toxic to human keratinocytes after 7 days of direct contact. Complete tissue remodeling and restoration of skin integrity is demonstrated in vivo in a diabetic mouse model. Hematological analysis reveals lack of wound inflammation due to bacterial infection or toxicity, confirming the potential of HA-SH/Ag@Lig NPs hydrogels for chronic wound management. Multifunctional hydrogels are promising materials to promote healing of complex wounds. Herein, we report simple and versatile route to prepare biocompatible and multifunctional self-assembled hydrogels for efficient chronic wound treatment utilizing polymer-nanoparticle interactions. Monistic thermomechanics of eddy mater-space with metric stresses of Maxwellian type and inelastic metric waves can be developed for purely kinetic densities of the nonlocal mass-energy integral. Pseudo-Riemannian 4-geometry uses locally warped time to preserve the Euclidean sublight transport of nonlocally correlated densities with instantaneous metric connections and coherent conservation of local 4-currents everywhere. The kinematic viscosity of geometrised energy-momentum densities and their geodesic self-pushes by correlated metric stresses quantitatively clarify the adaptive all-unity of continuous kinetic energy, including material ether or variable rest-energy. The metric self-organisation of matter-space+time implies a nonlocal feedback of correlated eddy densities and their tensor response to the vector density of external forces. Such tensor inertial responses of correlated metric stresses reveal the nonlocal nature of turbulence and predict adaptive auto-modes or thermal waves of Euclidean material space due to the modified geodesic equation with viscose autowaves and the inverse Cavendish constant. The proposed nonlocal alternative to Euler/Navier-Stokes hydrodynamics can distinguish between Cartesian and Newtonian worldviews in conceptual laboratory probes of new macroscopic mechanics for metric self-organisations of thermokinetic energies of viscose material space without negative gravitational energies. A "three‐components" supramolecular assembly has been fabricated by mixing 2, 2': 6', 2"‐terpyridine attached pillar[5]arene, cyano and triazole bearing alkyl chain and Zn2+ in solvent of CHCl3 and CH3CN. The driving forces for the assembly were believed to be host‐guest and metal chelating interactions as characterized by 1H NMR, UV‐vis and fluorescence spectra. The supramolecular organo‐gel formed upon the concentration of components amount to 1 M. The supramolecular displayed nitrobenzene based explosive sensing capability using picric acid, ortho‐nitrobenzene and phenol as samples. Supramolecular hydrogel composed of aromatic short peptide gelator was an attractive biomaterial owing to its simple and convenient synthetic route, nano-fibrillar microstructure resembling natural collagen fibers and intelligent response to external stimulus. Herein, stimuli-responsive biphenyl-tripeptide supramolecular hydrogels was prepared to simulate extracellular matrix scaffolds by temperature switch, ion induction and pH switch. The amino acid arrangement substantially affected gelation behavior, only BPAA-βAFF and BPAA-FFβA could form nanostructured supramolecular hydrogels with 8-10 nm nanotubes or nanofibers by potential intermolecular hydrogen bond interactions and π-π stacking. The minimum gelation concentration (MGC) and maximum storage modulus were 0.4 mM (0.023 wt%) and around 8.2 KPa. The two supramolecular hydrogels could support adhesion and proliferation of L929 cells. Moreover, the BPAA-βAFF hydrogel promoted proliferation and ECM secretion of chondrocytes in vitro , and facilitate the phenotype maintenance of hyaline cartilage. All the results demonstrated that BPAA-βAFF hydrogel hold great potential application prospects in cartilage tissue engineering. Diphenylalanine was served as a core segment conjugating with 4-biphenylacetic acid (BPAA) to produce biphenyl-tripeptide compounds with transforming amino sequence, and multiple external stimuli was applied to study the gelation properties of the aromatic short peptide gelators. Chronic wounds are persistent non-healing lesions whose complex management is due to the interplay of multiple factors promoting chronicity, such as oxidative stress, overexpressed enzyme activities impeding the tissue repair and bacterial contamination. Currently marketed chronic wound dressings are designed mainly to absorb wound exudate, and at most to release antimicrobial agents, usually ionic silver. However, an effective wound repair requires addressing the multifactorial nature of the wound in a holistic approach. This study explores the potential of metal phenolic network (MPN) nanoparticles (NPs), comprised of epigallocatechin gallate (EGCG) and cobalt, as active agents and structural elements in wound dressings. The MPN NPs were able to generate reactive oxygen species (ROS), disturb bacterial membrane, and inhibit the biofilm of both gram-positive Staphylococcus aureus and gram-negative Pseudomonas aeruginosa. Owing to the high interfacial activity of their phenolic shells, the NPs drove the self-assembly of thiolated hyaluronic acid (THA) hydrogels featuring injectability, self-healing, stimuli-responsive NPs delivery in wound microenvironment, and control over deleterious enzyme activities, oxidative stress and bacterial colonisation. Specifically, the hydrogels inhibited matrix metalloproteinases (MMPs) and myeloperoxidase (MPO) by 60 and 80 %, respectively, and achieved 4-log reduction of S. aureus and 2-log reduction of P. aeruginosa concentrations. Finally, the hydrogels were in-vivo validated in a mouse animal model, showing lack of toxicity and similar wound healing efficiency as an antibiotic-containing commercial product. The electrohydrodynamic effect has been exploited for the fabrication of polymeric microlenses through different approaches. Here we show that microlens arrays obtained with the pyro-EHD self-assembling process exhibit high fill-factor and good level of homogeneity, thus allowing to integrate them in a lightfield microscope. Digital holographic microscopy is used to characterize the fabricated microlens array. Afterwards, a Fourier lightfield microscope is built by placing this microlens array at the Fourier plane of the objective. (R)-ECHO is synthesized through the asymmetric reduction of ethyl 6-oxo-8-chlorooctanoate (ECOO) using carbonyl reductase(CR2) as a catalyst and co-enzyme NADPH as a hydrogen donor with addition of glucose dehydrogenase (GDH) for NADPH regeneration, thereby enhancing biotransformation efficiency. To improve the reaction efficiency further, a dual enzyme self-assembly cluster EutM-SC-ST-CR2(GDH) was developed specifically for this purpose. This co-immobilised system demonstrated a 50% increase in the conversion of ECOO compared to when SpyTag-CR2(ST-CR2) and SpyTag-GDH(ST-GDH) were co-catalysts. The optimal conditions for this increase were observed to be at a pH of 8.0, 30°C, when concentrations of 250 mM ECOO, 270 mM glucose and 0.1 mM NADPH, and a molar ratio of ST-CR2 to ST-GDH to EutM-SpyCatcher(EutM-SC) of 1:1:2 (molecular concentrations of ST-CR2, ST-GDH and EutM-SC were 3.5 mM, 3.5 mM and 7 mM). Under these conditions, the conversion of ECOO reached up to 95% and the enantiomeric excess (e.e.) of (R)-ECHO achieved 99.9%. Moreover, the EutM-SC-ST-CR2(GDH) construct displayed enhanced enzyme activity, stability, and kinetics, as well as improvements in electrostatic and ionic strengths. In conclusion, this study presents the development of a novel self-assembled protein scaffold, EutM-SC, for the co-immobilisation of ST-CR2 and ST-GDH, thus optimizing the biological reaction conditions and offering potential for the industrial production of (R)-α-lipoic acid. This novel system with excellent characteristics for inhibition of P-glycoprotein (P-gp), and for near-infrared fluorescence (NIRF) imaging-guided chemo-photothermal therapy (PTT), has been identified as a promising way to MDR and achieve synergistic cancer therapy.Methods: In this study, we successfully synthesized a multifunctional theranostic system, which was developed through FDA-approved self-assembling drugs, which contain anticancer drug doxorubicin (Dox), imaging and high photothermal conversion drug indocyanine green (ICG) and P-gp regulator TPGS (the system named T/Dox-ICG). We studied the characterization of T/Dox-ICG NPs, including the TEM, SEM, DLS, UV-vis-NIR, zeta potential, CLSM, in vitro FL imaging, in vitro photothermal effect, in vitro Dox and ICG release. We used CLSM to verify the location of intracellular distribution of Dox in SCG 7901/VCR cells, Western blot was performed to demonstrate the TPGS-mediated inhibition of P-gp. And, the cytotoxicity of materials against SCG 7901/VCR cells was studied by the MTT assay.Results: The TEM showed the T/Dox-ICG NPs had good monodispersity with diameters of 19.03 nm, Dox and ICG could be released constantly from T/Dox-ICG NPs in vitro. In vitro cell experiments demonstrated higher Dox accumulation and retention in the nucleus. Western blot showed TPGS could obviously inhibit the expression of P-gp. In vitro cytotoxicity assay showed more significant cytotoxicity on MDR cells (SCG 7901/VCR) with only 8.75% of cells surviving.Conclusion: MDR cancer therapy indicates that it may be important to develop a safer system that can simultaneously inhibit the drug transporters and monitor the delivery of chemotherapeutic agents, and combination therapy have raised widespread concern on tumor treatment.Keywords: multiple drug resistance, self-assembling, synergistic cancer therapy, imaging-guided therapy

The rapid development of antimicrobial resistance is pushing the search in the discovering of novel antimicrobial molecules to prevent and treat bacterial infections. Self-assembling antimicrobial peptides, as the lipidated peptides, are a novel and promising class of molecules capable of meeting this need. Based on previous work on Temporin L analogs, several new molecules lipidated at the N- or and the C-terminus were synthesised. Our goal is to improve membrane interactions through finely tuning self-assembly to reduce oligomerisation in aqueous solution and enhance self-assembly in bacterial membranes while reducing toxicity against human cells. The results here reported show that the length of the aliphatic moiety is a key factor to control target cell specificity and the oligomeric state of peptides either in aqueous solution or in a membrane-mimicking environment. The results of this study pave the way for the design of novel molecules with enhanced activities.
