The freshwater‐seawater mixing zone ("interface") is affected by tides, seawater intrusion, submarine groundwater discharge and aquifer heterogeneities. Our current understanding is based primarily on theoretical studies involving numerical simulation and sand‐tank experimentation, with limited field analyses of interface characteristics in coastal aquifers that serve as important freshwater reservoirs. The most common field evidence for interface characteristics is derived from electrical conductivity (EC) profiles, often obtained from long‐screen wells. However, the EC profiles from long‐screen wells have been assessed previously using only simple statistics like the depth to 50% of seawater concentration. This study introduces a methodology for characterizing EC profiles using curve fitting and objective assessment of multiple interface attributes, with application to 170 EC profiles from the Lower Burdekin Delta, Australia. The adopted fitting function showed an excellent match (mean R2 = 0.99). Fitting parameters were linked to the curvatures of transitions from fresh to brackish water and from brackish water to seawater, the elevation and gradient of the mid‐salinity value, and the thickness of the mixing zone. Field EC profiles showed approximately linear relationships between head changes averaged over the previous 5 months and the elevation of 5 mS/cm isochlor for some wells. The slope of this relationship was approximately one, which is approximately 1/40th of the value arising from the Ghyben‐Herzberg relation. The framework for characterizing salinity profiles presented in this article offers a new, objective approach for investigating the interface, providing future studies with a technique for examining relationships between interface behavior and key controlling forces. To explore the role of penconazole (PEN) and potassium (K) in controlling reactive oxygen species (ROS) production, accumulation of osmotically active substances and antioxidant defense system in wheat plants under drought conditions, an experiment was conducted. Two cultivars of wheat plant (Moghan and Sirvan) were treated by different PEG concentrations (0, 5, 10 and 15%) without or with PEN (15 mg L−1) and K (1 M). Drought caused damaging effects on growth attributes in both cultivar and Sirvan cultivar was more sensitive to drought stress. Enhanced levels of the hydrogen peroxide (H2O2) and malondialdehyde (MDA) were evident that drought stress induced oxidative stress in both cultivars. The activities of antioxidant enzymes were significantly declined under drought conditions. Drought stress enhanced accumulation of proline and soluble sugar as osmotic substance in both cultivars. In contracts to Sirvan cultivar, the content of antioxidant metabolites (total phenol, flavonol and anthocyanin) augmented under drought conditions. PEN and K application improved growth and regulated acclimation responses in wheat plants. Reduction in H2O2 and lipid peroxidation (MDA content) by PEN and K showed that PEN and K aided to alleviate drought-trigged oxidative stress in wheat plants. Noteworthily, soluble sugar dominated than proline in the osmotic regulation in stressed plants treated with PEN and K. PEN and K strengthened the enzymatic and non-enzymatic antioxidant systems in wheat plants. In summary, PEN and K provided a better defense to wheat plants against drought stress and positive impacts of PEN and K were related to improved physiological and metabolic processes under drought conditions. Objective: Two Cownose Rays Rhinoptera bonasus were presented for management of a severe capsalid monogenean infection Benedeniella posterocolpa in a mixed species habitat at an aquarium. Methods: A series of freshwater (FW) dips were elected to mitigate parasite resistance to praziquantel due to the endemic monogenean population in the system. A pretreatment blood sample was opportunistically obtained, and subsequent samples were processed due to development of clinical signs. Result: While the first FW dip was tolerated well and did not induce abnormal behavior, the second led to one individual presenting with tachypnea, pallor, and lethargy. Marked hematological abnormalities requiring medical intervention occurred in both individuals after the second dip. After treatment with elasmobranch Ringer's fluid therapy, sodium bicarbonate, prednisolone acetate, and vitamin E/selenium in the more severely affected ray, and elasmobranch Ringer's alone in the second ray, hematologic derangements and symptoms resolved. Hemoconcentration, hyperproteinemia, hyperlactatemia, and hyperglycemia were attributed to a stress response. A selective loss of urea after exposure to FW resulted in decreased plasma osmolality. Conclusion: Given the severity of the stress response and associated complications observed, hyposalinity treatments should be utilized with caution in this species. If this therapy is employed, the clinician is advised to be prepared to administer supportive care coinciding with the FW dip if necessary. The electrocatalytic performance of MoNi‐based nanomaterials undergo selenization has garnered significant interest due to their modified electronic structure, while still posses certain challenges for obtained bimetallic selenides. Here, a novel MoNi‐based electrocatalyst of NiMoO4@Mo15Se19/NiSe2 core‐shell nanomaterials is constructed to promote the desorption of OOH* which can facilitate the water oxidation process. NiMoO4@Mo15Se19/NiSe2 core‐shell nanoarrays show that the "cores" are mainly NiMoO4 nanorods while the "shells" are the bimetallic selenides nanoflakes, which are super architectures that can expand more active sites and accelerate the electron transfer. Moreover, the hybridization interaction between Ni 3d, Mo 4d, and Se 4p orbitals leads to an asymmetric distribution of electric clouds, which decreases the adsorption energy and transformation of oxygen‐containing species. Electrochemical data displays that the overpotentials of NiMoO4@Mo15Se19/NiSe2 core‐shell nanomaterials are only 195 mV, 220 mV, and 224 mV for oxygen evolution reaction (OER) in alkaline freshwater, alkaline simulated seawater, and alkaline natural seawater. The current density decay is negligible after 100 h stability at about 1.46 V with a three‐electrode system in alkaline natural seawater. Acidic stress in beef cattle slaughtering abattoirs can induce the acid adaptation response of in-plant contaminated Salmonella. This may further lead to multiple resistance responses threatening public health. Therefore, the acid, heat, osmotic and antibiotic resistances of Salmonella typhimurium (ATCC14028) were evaluated after a 90 min adaption in a pH = 5.4 "mild acid" Luria–Bertani medium. Differences in such resistances were also determined between the ∆phoP mutant and wild-type Salmonella strains to confirm the contribution of the PhoP/PhoQ system. The transcriptomic differences between the acid-adapted and ∆phoP strain were compared to explore the role of the PhoP/Q two-component system in regulating multi-stress resistance. Acid adaptation was found to increase the viability of Salmonella to lethal acid, heat and hyperosmotic treatments. In particular, acid adaptation significantly increased the resistance of Salmonella typhimurium to Polymyxin B, and such resistance can last for 21 days when the adapted strain was stored in meat extract medium at 4 °C. Transcriptomics analysis revealed 178 up-regulated and 274 down-regulated genes in the ∆phoP strain. The Salmonella infection, cationic antimicrobial peptide (CAMP) resistance, quorum sensing and two-component system pathways were down-regulated, while the bacterial tricarboxylic acid cycle pathways were up-regulated. Transcriptomics and RT-qPCR analyses revealed that the deletion of the phoP gene resulted in the down-regulation of the expression of genes related to lipid A modification and efflux pumps. These changes in the gene expression result in the change in net negative charge and the mobility of the cell membrane, resulting in enhanced CAMP resistance. The confirmation of multiple stress resistance under acid adaptation and the transcriptomic study in the current study may provide valuable information for the control of multiple stress resistance and meat safety. Water stress can adversely affect seed germination and plant growth. Seed osmopriming is a pre-sowing treatment in which seeds are soaked in osmotic solutions to undergo the first stage of germination prior to radicle protrusion. Seed osmopriming enhances germination performance under stressful environmental conditions, making it an effective method to improve plant resistance and yield. This study analyzed the effect of seed osmopriming with polyethylene glycol (PEG) on seed germination and physiological parameters of Coronilla varia L. Priming treatments using 10% to 30% PEG enhanced germination percentage, germination vigor, germination index, vitality index, and seedling mass and reduced the time to reach 50% germination (T50). The PEG concentration that led to better results was 10%. The content of soluble proteins (SP), proline (Pro), soluble sugars (SS), and malondialdehyde (MDA) in Coronilla varia L. seedlings increased with the severity of water stress. In addition, under water stress, electrolyte leakage rose, and peroxidase (POD) and superoxide dismutase (SOD) activities intensified, while catalase (CAT) activity increased at mild-to-moderate water stress but declined with more severe deficiency. The 10% PEG priming significantly improved germination percentage, germination vigor, germination index, vitality index, and time to 50% germination (T50) under water stress. Across the water stress gradient here tested (8 to 12% PEG), seed priming enhanced SP content, Pro content, and SOD activity in Coronilla varia L. seedlings compared to the unprimed treatments. Under 10% PEG-induced water stress, primed seedlings displayed a significantly lower MDA content and electrolyte leakage than their unprimed counterparts and exhibited significantly higher CAT and POD activities. However, under 12% PEG-induced water stress, differences in electrolyte leakage, CAT activity, and POD activity between primed and unprimed treatments were not significant. These findings suggest that PEG priming enhances the osmotic regulation and antioxidant capacity of Coronilla varia seedlings, facilitating seed germination and seedling growth and alleviating drought stress damage, albeit with reduced efficacy under severe water deficiency. Trimeric G proteins transduce signals from a superfamily of receptors and each G protein controls a wide range of cellular and systemic functions. Their highly conserved alpha subunits fall in five classes, four of which have been well investigated (Gs, Gi, G12, Gq). In contrast, the function of the fifth class, Gv is completely unknown, despite its broad occurrence and evolutionary ancient origin (older than metazoans). Here we show a dynamic presence of Gv mRNA in several organs during early development of zebrafish, including the hatching gland, the pronephros and several cartilage anlagen, employing in situ hybridisation. Next, we generated a Gv frameshift mutation in zebrafish and observed distinct phenotypes such as reduced oviposition, premature hatching and craniofacial abnormalities in bone and cartilage of larval zebrafish. These phenotypes could suggest a disturbance in ionic homeostasis as a common denominator. Indeed, we find reduced levels of calcium, magnesium and potassium in the larvae and changes in expression levels of the sodium potassium pump atp1a1a.5 and the sodium/calcium exchanger ncx1b in larvae and in the adult kidney, a major osmoregulatory organ. Additionally, expression of sodium chloride cotransporter slc12a3 and the anion exchanger slc26a4 is altered in complementary ways in adult kidney. It appears that Gv may modulate ionic homeostasis in zebrafish during development and in adults. Our results constitute the first insight into the function of the fifth class of G alpha proteins. Phototropism movement is crucial for plants to adapt to various environmental changes. Plant P‐type H+‐ATPase (HA) plays diverse roles in signal transduction during cell expansion, regulation of cellular osmotic potential and stomatal opening, and circadian movement. Despite numerous studies on the genome‐wide analysis of Vitis vinifera, no research has been done on the P‐type H+‐ATPase family genes, especially concerning pulvinus‐driven leaf movement. In this study, 55 VvHAs were identified and classified into nine distinct subgroups (1 to 9). Gene members within the same subgroups exhibit similar features in motif, intron/exon, and protein tertiary structures. Furthermore, four pairs of genes were derived by segmental duplication in grapes. Cis‐acting element analysis identified numerous light/circadian‐related elements in the promoters of VvHAs. qRT‐PCR analysis showed that several genes of subgroup 7 were highly expressed in leaves and pulvinus during leaf movement, especially VvHA14, VvHA15, VvHA16, VvHA19, VvHA51, VvHA52, and VvHA54. Additionally, we also found that the VvHAs genes were asymmetrically expressed on both sides of the extensor and flexor cell of the motor organ, the pulvinus. The expression of VvHAs family genes in extensor cells was significantly higher than that in flexor cells. Overall, this study serves as a foundation for further investigations into the functions of VvHAs and contributes to the complex mechanisms underlying grapevine pulvinus growth and development. Freshwater salinity regimes vary naturally and are changing in response to anthropogenic activities. Few insect species tolerate saline waters, and biodiversity losses are associated with increasing salinity in freshwater. We used radiotracers (22Na, 35SO4, and 45Ca) to examine ion uptake rates across concentration gradients in mayflies (Ephemeroptera), caddis flies (Trichoptera), and mosquitoes (Diptera) and made observations for some traits in seven other taxa representing mayflies, stone flies (Plecoptera), true flies (Diptera), and true bugs (Hemiptera). We further assessed the permeability of the cuticle to 3H2O influx and 22Na efflux when faced with deionized water in these same taxa. We hypothesized a relationship between uptake rates and reported saline tolerances, but our data did not support this hypothesis, likely because acclimatory responses were not part of this experimental approach. However, we found several common physiological traits across the taxa studied, including (i) ionic uptake rates that were always positively correlated with dissolved concentrations, (ii) generally low Ca uptake rates relative to other freshwater taxa, (iii) greater Na loss than Na uptake in dilute conditions, (iv) ion uptake that was more variable in ion-rich conditions than in dilute conditions, and (v) 3H2O influx that occurs quickly (but this rapidly exchangeable pool of body water accounts for a surprisingly small percentage of the water content of species tested). The female Aedes aegypti mosquito is a vector for several arboviral diseases, due to their blood feeding behavior and their association with urban communities. While ion transport in Ae. aegypti has been studied, much less is known about mechanisms of water transport. Rapid water and ion excretion occurs in the adult female mosquito post blood meal and involves a set of organs including the midgut, Malpighian tubules (MTs), and hindgut. The MTs are responsible for the formation of primary urine and are considered the most important site for active transport of ions. Within the cells of the MTs, along with various ion transporters, there are aquaporin water channels that aid in the transport of water across the tubule cell membrane. Six aquaporin genes have been molecularly identified in Ae. aegypti (AQP1–6) and found to be responsible for the transport of water and in some cases, small solutes such as glycerol. In this study, we used immunohistochemistry to localize AaAQP1, 2, 4, 5, and 6 in the adult female Ae. aegypti, in non-blood fed and post blood feeding (0.5 and 24hr) conditions. We further examined the main water transporting aquaporin, AaAQP1, using western blotting to determine protein abundance changes in isolated MTs pre and post-blood feeding. Using fluorescence in situ hybridization, aqp1 mRNA was found exclusively in the principal cells of female MTs. Finally, we used immunogold staining with transmission electron microscopy to determine subcellular localization of AaAQP1 in the Malpighian tubules under non-blood fed conditions. Interestingly, AaAQP1 was found to be predominantly in the principal cells of the MTs, dispersed throughout the brush border; however, there was also evidence of some AaAQP1 localization in the stellate cells of the MTs. This study aims to determine the effectiveness of various filter materials in RAS such as synthetic materials and organic materials and there an impact on plasma cortisol, osmotic performance rate, growth rate and survival rate of Channa marulioides. The cultivation process in the RAS system is carried out for 100 days with 50 individuals in each different treatment, namely T1: Synthetic filter; T2: Organic filter; T3: Mixture of synthetic and organic filter; Control: No Filter and RAS. Measurements of plasma cortisol and osmotic performance levels were carried out at the beginning and end of cultivation which had an impact on growth and survival rates. The results of the study showed a decrease in cortisol levels (ng/ml), namely: T1: 9.24, T2: 9.66, T3: 10.37 which showed that there was no significant difference in each treatment (P> 0.05) but in T1, T2, and T3 were significantly different with control treatment: 1.32 (P<0.05). The same trend occurred when observing the level of osmotic performance with the highest decrease in T3 on 6 scales; T2 on a scale of 5.5; T1 has 5 scales, and Control has 3 scales. Reducing the level of cortisol and osmotic performance hormones with the RAS system can increase energy efficiency by reducing stress levels, and osmotic performance so that it can be optimized for growth and survival in adaptation. Mushroom production and consumption are gaining increased interest due to their unique flavor and nutritional value. However, in the mushroom industry, large amounts of by-products are generated, which have a high negative environmental and economic impact. In this study, an osmotic dehydration process followed by hot-air-drying was applied to mushroom stems to produce dried mushrooms as the end product. The osmotic dehydration conditions (concentration of hypertonic solution, specifically, 10–30% maltodextrin and 20–40% oligofructose; a treatment time of 40–80 min; and a temperature range of 30–50 °C) were optimized using response surface methodology (RSM). The results showed that a four-factor three-level Box–Behnken experimental design was effectively implemented to evaluate the effect of the process parameters and identify the optimal osmotic dehydration conditions for producing osmotically dehydrated mushrooms. The main factor affecting mass transfer was the osmosis temperature, and the optimal conditions were found to be 38 °C, 40% oligofructose and 19.3% maltodextrin as the osmotic agents, and 80 min of immersion time. Moreover, the results showed that osmotic pretreatment, in the optimal conditions, significantly reduced the required drying time of the by-products compared to traditional hot-air-drying, especially at milder drying temperatures. Rice (Oryza sativa L.) is one of the most important food crops worldwide. However, during direct seeding, rice is extremely vulnerable to flooding stress, which impairs rice's emergence and seedling growth and results in a significant yield loss. According to our research, chitosan oligosaccharides have the potential to be a chemical seed-soaking agent that greatly increases rice's resistance to flooding. Chitosan oligosaccharides were able to enhance seed energy supply, osmoregulation, and antioxidant capacity, according to physiological index assessments. Using transcriptome and metabolomic analysis, we discovered that important differential metabolites and genes were involved in the signaling pathway for hormone synthesis and antioxidant capacity. Exogenous chitosan oligosaccharides specifically and significantly inhibit genes linked to auxin, jasmonic acid, and abscisic acid. This suggested that applying chitosan oligosaccharides could stabilize seedling growth and development by controlling associated hormones and reducing flooding stress by enhancing membrane stability and antioxidant capacity. Finally, we verified the effectiveness of exogenous chitosan oligosaccharides imbibed in seeds by field validation, demonstrating that they can enhance rice seedling emergence and growth under flooding stress. Narrow clawed crayfish, Pontastacus (Astacus) leptodactylus, represents an ecologically and economically valuable freshwater species. Despite the high importance of artificial breeding for conservation purpose and aquaculture potential, hatching protocols have not been developed so far in this species. Further, limited knowledge exists regarding the artificial egg incubation, the temperature effect on embryonic development, hatching synchronization and hatching rate. In the present study we investigated the temperature increase (from 17 oC to 22oC) effects in two different embryonic developmental stages of P. leptodactylus. Furthermore, two primer pairs for the Fibroblast Growth Factor Receptor 4 (FGFR4) gene cDNA amplification were successfully designed, characterising for the first time the FGFR4 gene in P. leptodactylus in relation to different developmental stages and temperatures. Apart from the FGFR4 gene, the Na+/K+-ATPase α-subunit expression was also explored. Both the FGFR4 and Na+/K+-ATPase α-subunit expression levels were higher in embryos closer to hatching. Egg incubation at 22oC for seven days led to significant increase of FGFR4 expression in embryos from earlier developmental stages. Nevertheless, temperature increase did not affect FGFR4 expression in eggs from latter developmental stages and Na+/K+-ATPase α-subunit expression in all developmental stages. Temperature increase represents therefore probably a promising strategy for accelerating hatching in freshwater crayfish particularly in early developmental stages. Specifically, our results indicate that FGFR4 expression increased in embryonic stages closer to hatching and that temperature influences significantly its expression in embryos from earlier developmental stages. Overall, these findings can provide a better understanding of artificial egg incubation of P. leptodactylus, and therefore can be employed for the effective management of this species, both for economic and biodiversity retention reasons. Background: Cyprinidae, the largest fish family, encompasses approximately 367 genera and 3006 species. While they exhibit remarkable adaptability to diverse aquatic environments, it is exceptionally rare to find them in seawater, with the Far Eastern daces being of few exceptions. Therefore, the Far Eastern daces serve as a valuable model for studying the genetic mechanisms underlying seawater adaptation in Cyprinidae. Results: Here, we sequenced the chromosome-level genomes of two Far Eastern daces (Pseudaspius brandtii and P. hakonensis), the two known cyprinid fishes found in seawater, and performed comparative genomic analyses to investigate their genetic mechanism of seawater adaptation. Demographic history reconstruction of the two species reveals that their population dynamics are correlated with the glacial-interglacial cycles and sea level changes. Genomic analyses identified Pseudaspius-specific genetic innovations related to seawater adaptation, including positively selected genes, rapidly evolving genes, and conserved non-coding elements (CNEs). Functional assays of Pseudaspius-specific variants of the prolactin (prl) gene showed enhanced cell adaptation to greater osmolarity. Functional assays of Pseudaspius specific CNEs near atg7 and usp45 genes suggest that they exhibit higher promoter activity and significantly induced at high osmolarity. Conclusions: Our results reveal the genome-wide evidence for the evolutionary adaptation of cyprinid fishes to seawater, offering valuable insights into the molecular mechanisms supporting the survival of migratory fish in marine environments. These findings are significant as they contribute to our understanding of how cyprinid fishes navigate and thrive in diverse aquatic habitats, providing useful implications for the conservation and management of marine ecosystems. The exogenous use of the redox compound (H2O2) plays a significant role in abiotic stress tolerance. The present study investigated various H2O2 application methods (seed priming, foliar spray, and surface irrigation) with varying concentration levels (0 mM, 5 mM, 10 mM, 15 mM, 40 mM, 80 mM, and 160 mM) to evaluate the efficiency of supplying exogenous H2O2 to quinoa under water-deficit conditions. Drought stress reduced quinoa growth and yield by perturbing morphological traits, leading to the overproduction of reactive oxygen species and increased electrolyte leakage. Although all studied modes of H2O2 application improved quinoa performance, surface irrigation was found to be sensitive, causing oxidative damage in the present study. Seed priming showed a prominent increase in plant height due to profound emergence indexes compared to other modes under drought conditions. Strikingly, seed priming followed by foliar spray improved drought tolerance in quinoa and showed higher grain yield compared to surface irrigations. This increase in the yield performance of quinoa was attributed to improvements in total chlorophyll (37%), leaf relative water content (RWC; 20%), superoxide dismutase (SOD; 35%), peroxidase (97%), polyphenol oxidase (60%), and phenylalanine ammonia-lyase (58%) activities, and the accumulation of glycine betaine (96%), total soluble protein (TSP; 17%), proline contents (35%), and the highest reduction in leaf malondialdehyde contents (MDA; 36%) under drought stress. PCA analysis indicated that physio-biochemical traits (proline, SOD, TSP, total chlorophyll, MSI, and RWC) were strongly positively correlated with grain yield, and their contribution was much higher in redox priming than other application methods. Natural environments of living organisms are often dynamic and multifactorial, with multiple parameters fluctuating over time. To better understand how cells respond to dynamically interacting factors, we quantified the effects of dual fluctuations of osmotic stress and glucose deprivation on yeast cells using microfluidics and time- lapse microscopy. Strikingly, we observed that cell proliferation, survival, and signaling depend on the phasing of the two periodic stresses. Cells divided faster, survived longer, and showed decreased transcriptional response when fluctuations of hyperosmotic stress and glucose deprivation occurred in phase than when the two stresses occurred alternatively. Therefore, glucose availability regulates yeast responses to dynamic osmotic stress, showcasing the key role of metabolic fluctuations in cellular responses to dynamic stress. We also found that mutants with impaired osmotic stress response were better adapted to alternating stresses than wild- type cells, showing that genetic mechanisms of adaptation to a persistent stress factor can be detrimental under dynamically interacting conditions. Simple Summary: Crowding stress from stocking density is one of the critical stress factors in mariculture. Exposure of fish to chronic crowding stress substantially affects their well-being and physiological state, resulting in decreased growth performance, disordered metabolism, and consequent economic losses. In this research, we aimed to provide a deeper understanding of the effects of chronic crowding stress from stocking density on rainbow trout in mariculture. The obtained data offer unprecedented insights into the relationship between the growth performance and lipid metabolism of rainbow trout under chronic stress from stocking density in mariculture. (1) Background: In aquaculture, chronic stress due to high stocking density impairs animals' welfare and results in declined fishery production with low protein quality. However, most previous studies evaluated the effects of high stocking density on trout in freshwater rather than seawater. (2) Methods: Juvenile trout were reared for 84 days in circular tanks under three stocking densities, including low density ("LD", 9.15 kg/m3), moderate density ("MD", 13.65 kg/m3), and high density ("HD", 27.31 kg/m3) in seawater. The final densities of LD, MD, and HD were 22.00, 32.05 and 52.24 kg/m3, respectively. Growth performance and lipid metabolism were evaluated. (3) Results: Growth performance and feeding efficiency were significantly reduced due to chronic stress under high density in mariculture. The digestive activity of lipids was promoted in the gut of HD fish, while the concentration of triglycerides was decreased in the blood. Furthermore, decreased acetyl-CoA carboxylase (ACC) and fatty acid synthase (FAS), increased hormone-sensitive lipase (HSL) concentrations, and activated hepatic β-oxidation processes were observed in trout under HD. Redundancy analysis showed that glycerol and HSL can be used as potential markers to evaluate the growth performance of trout in mariculture. Extreme weather events, including drought, have occurred worldwide with increasing frequency and severity in recent years. Drought stress is the main limiting factor for agricultural development in many regions, including tobacco—an important economic crop and a model plant for scientific research. As it is adapted to the tropics, tobacco is highly susceptible to drought stress, with resultant decreases in yield and quality. Glycine betaine (GB) is an osmoregulatory substance that can enhance plant resistance to various abiotic stresses. Here, we investigate the protective mechanism of genetically engineered glycine betaine (GB) on tobacco photosynthesis under drought stress induced by 30% PEG-6000. This study used transgenic tobacco (T) accumulating GB and wild-type tobacco (WT) to investigate the protective effects conferred by the genetic engineering of GB synthesis on tobacco photosynthesis under drought stress (induced by 30% PEG-6000). The results showed that the net photosynthetic rate of the tobacco plants significantly decreased under drought stress, and the degree of decrease was significantly lower in the T line than in the WT line. GB accumulation improved the resistance of photosynthesis to drought stress. Furthermore, under drought stress, the photosynthesis improvement in the T line was related to the accumulation of GB, leading to maintenance of the water status, the promotion of osmotic regulation, and an enhancement in antioxidant enzyme activities, which reduced membrane peroxidation and thereby increased the thylakoid membrane's protein content and function, especially the photosystem II (PSII) function. Soil salinization is a major factor that reduces crop yields. There are some plant growth-promoting rhizobacteria (PGPR) that can stimulate and enhance the salt tolerance of plants near their roots in saline–alkali environments. Currently, there is relatively little research on PGPR in rice saline–alkali tolerance. In the early stages of this study, a strain of Microbacterium ginsengiterrae S4 was screened that could enhance the growth of rice in a laboratory-simulated saline–alkali environment (100 mM NaCl, pH 8.5). The experiment investigated the effects of S4 bacteria on the growth, antioxidant capacity, and osmotic regulation of rice seedlings under saline–alkali stress. RNA-Seq technology was used for transcriptome sequencing and UPLC-MS/MS for metabolite detection. Research has shown that S4 bacteria affect the growth of rice seedlings under saline–alkali stress through the following aspects. First, S4 bacteria increase the antioxidant enzyme activity (SOD, POD, and CAT) of rice seedlings under saline–alkali stress, reduce the content of MDA, and balance the content of osmotic regulatory substances (soluble sugar, soluble protein, and proline). Second, under saline–alkali stress, treatment with S4 bacteria caused changes in differentially expressed genes (DEGs) (7 upregulated, 15 downregulated) and differentially metabolized metabolites (101 upregulated; 26 downregulated) in rice seedlings. The DEGs are mainly involved in UDP-glucose transmembrane transporter activity, while the differentially metabolized metabolites are mainly involved in the ABC transporters pathway. Finally, key genes and metabolites were identified through correlation analysis of transcriptomes and metabolomes, among which OsSTAR2 negatively regulates L-histidine, leading to an increase in L-histidine content. Furthermore, through gene correlation and metabolite correlation analysis, it was found that OsWRKY76 regulates the expression of OsSTAR2 and that L-histidine also causes an increase in 2-methyl-4-pentenoic acid content. Based on the above analysis, the addition of S4 bacteria can significantly improve the tolerance of rice in saline–alkali environments, which has a great application value for planting rice in these environments. Investigating the effects of priming methods on seedling physiology under osmotic stress conditions is important to understand how priming enhances osmotic stress tolerance in safflower. The mechanisms by which priming achieves this at the physiological level, such as changes in osmoprotectants, antioxidants, and membrane damage, are not well understood. This research aims to fill these knowledge gaps and provide insights that can be used to optimize priming treatments and improve safflower productivity in drought-prone areas.. Seeds were treated with priming including distilled water, folic acid, glutathione, epibrassinolide and chitosan and then subjected to osmotic stresses including no osmotic stress, mild osmotic stress (4 bar) and severe osmotic stress (8 bar) for germination and seedling growth. Severe osmotic stress increased soluble leaf proteins, this increase was observed in all priming treatments except distilled water. Severe osmotic stress increased soluble leaf proteins by an average of 35% in folic acid, chitosan, and epibrassinolide treatments. Chitosan treatment exhibited 172% and 188% more proline under mild and severe stress, respectively, compared to the control. Folic acid treatment showed a 207% increase in catalase under severe stress. Epibrassinolide and glutathione treatments had 201% more superoxide dismutase (SOD) under non-stress conditions. Distilled water treatment had 48.6% more soluble sugars under non-stress, while glutathione and epibrassinolide treatments had 14.5% more under mild stress. Control treatment had the highest malondialdehyde (MDA) amounts under mild and severe stress. Chitosan treatment exhibited a 25% increase in seedling length under severe stress compared to the control. Under non-stress conditions, chitosan treatment had slightly lower seedling length compared to control, while glutathione and epibrassinolide treatments showed the highest lengths. Under mild stress, folic acid, chitosan, and epibrassinolide treatments were superior to control. Under severe stress, distilled water showed a 20% increase, while chitosan treatment showed a 25% increase in seedling length compared to control. Chitosan treatment improved safflower seedling growth under severe osmotic stress through mechanisms including increased osmoprotectant levels, enhanced antioxidant activity, and improved membrane integrity. This article focuses on the reproductive behavior of the brackish nemertean species Potamostoma shizunaiense. The study found that these nemerteans exhibit specific adaptations to upper brackish habitats, including seaward migrations during spawning seasons and a critical influence of water salinity on reproductive success. The oral mucosa represents a defensive barrier between the external environment and the rest of the body. Oral mucosal cells are constantly bathed in hypotonic saliva (normally one-third tonicity compared to plasma) and are repeatedly exposed to environmental stresses of tonicity, temperature, and pH by the drinks we imbibe (e.g., hypotonic: water, tea, and coffee; hypertonic: assorted fruit juices, and red wines). In the mouth, the broad-spectrum antiviral mediator MxA (a dynamin-family large GTPase) is constitutively expressed in healthy periodontal tissues and induced by Type III interferons (e.g., IFN-λ1/IL-29). Endogenously induced human MxA and exogenously expressed human GFP-MxA formed membraneless biomolecular condensates in the cytoplasm of oral carcinoma cells (OECM1 cell line). These condensates likely represent storage granules in equilibrium with antivirally active dispersed MxA. Remarkably, cytoplasmic MxA condensates were exquisitely sensitive sensors of hypotonicity—the condensates in oral epithelium disassembled within 1–2 min of exposure of cells to saliva-like one-third hypotonicity, and spontaneously reassembled in the next 4–7 min. Water, tea, and coffee enhanced this disassembly. Fluorescence changes in OECM1 cells preloaded with calcein-AM (a reporter of cytosolic "macromolecular crowding") confirmed that this process involved macromolecular uncrowding and subsequent recrowding secondary to changes in cell volume. However, hypertonicity had little effect on MxA condensates. The spontaneous reassembly of GFP-MxA condensates in oral epithelial cells, even under continuous saliva-like hypotonicity, was slowed by the protein-phosphatase-inhibitor cyclosporin A (CsA) and by the K-channel-blocker tetraethylammonium chloride (TEA); this is suggestive of the involvement of the volume-sensitive WNK kinase-protein phosphatase (PTP)-K-Cl cotransporter (KCC) pathway in the regulated volume decrease (RVD) during condensate reassembly in oral cells. The present study identifies a novel subcellular consequence of hypotonic stress in oral epithelial cells, in terms of the rapid and dynamic changes in the structure of one class of phase-separated biomolecular condensates in the cytoplasm—the antiviral MxA condensates. More generally, the data raise the possibility that hypotonicity-driven stresses likely affect other intracellular functions involving liquid–liquid phase separation (LLPS) in cells of the oral mucosa. To understand the role of myo-inositol oxygenase (miox) in the osmotic regulation of Nile tilapia, its expression was analyzed in various tissues. The results showed that the expression of miox gene was highest in the kidney, followed by the liver, and was significantly upregulated in the kidney and liver under 1 h hyperosmotic stress. The relative luminescence efficiency of the miox gene transcription starting site (−4,617 to +312 bp) under hyperosmotic stress was measured. Two fragments (−1,640/−1,619 and −620/−599) could induce the luminescence activity. Moreover, the −1,640/−1,619 and −620/−599 responded to hyperosmotic stress and high-glucose stimulation by base mutation, suggesting that osmotic and carbohydrate response elements may exist in this region. Finally, the salinity tolerance of Nile tilapia was significantly reduced after the knocking down of miox gene. The accumulation of myo-inositol was affected, and the expression of enzymes in glucose metabolism was significantly reduced after the miox gene was knocked down. Furthermore, hyperosmotic stress can cause oxidative stress, and MIOX may help maintain the cell redox balance under hyperosmotic stress. In summary, MIOX is essential in osmotic regulation to enhance the salinity tolerance of Nile tilapia by affecting myo-inositol accumulation, glucose metabolism, and antioxidant performance. Drought is a major obstacle to the development of naked oat industry. This work investigated mechanisms by which exogenous Streptomyces albidoflavus T4 and Streptomyces rochei D74 improved drought tolerance in naked oat (Avena nuda) seedlings. Results showed that in the seed germination experiment, germination rate, radicle and hypocotyl length of naked oat seeds treated with the fermentation filtrate of T4 or D74 under PEG induced drought stress increased significantly. In the hydroponic experiment, the shoot and root dry weights of oat seedlings increased significantly when treated with the T4 or D74 fermentation filtrate under the 15% PEG induced drought stress (S15). Simultaneously, the T4 treatment also significantly increased the surface area, volume, the number of tips and the root activity of oat seedlings. Both T4 and D74 treatments elicited significant increases in proline and soluble sugar contents, as well as the catalase and peroxidase activities in oat seedlings. The results of comprehensive drought resistance capacity (CDRC) calculation of oat plants showed that the drought resistance of oat seedlings under the T4 treatment was better than that under the D74 treatment, and the effect was better under higher drought stress (S15). The aim of this research was to assess the yield and postharvest characteristics of 'BRS Sol do Cerrado' sour passion fruits based on irrigation with varying levels of saline water and potassium fertilization. The study was conducted under field conditions at an experimental farm in São Domingos, Paraíba, Brazil. A randomized block design was implemented in a 5 × 4 factorial arrangement, with five levels of electrical conductivity of water (ECw): 0.3, 1.1, 1.9, 2.7, and 3.5 dS m−1, and four potassium doses (KD): 60, 80,100, and 120% of the recommended amount, with 3 replications. The potassium dose equivalent to 120% of the recommended dose in combination with low-salinity water resulted in the highest fresh mass accumulation in the sour passion fruit. Water electrical conductivity up to 2.7 dS m−1, along with the lowest recommended KD, led to increased levels of soluble solids and ascorbic acid in the sour passion fruit. Irrigation with water of 3.5 dS m−1 and using 80 to 100% of the recommended KD enhanced the total sugar content in the sour passion fruit. On the other hand, irrigation with water of 3.5 dS m−1 combined with 60% of the recommended KD resulted in a higher pulp yield in the 'BRS Sol do Cerrado' sour passion fruit 160 days post-transplantation. Adjustments in potassium fertilization management at different irrigation water salinity levels played a crucial role in maintaining both the production and quality of the sour passion fruit. The occurrence of early spring and late frost damage brought severe challenges to the production of walnuts in China, seriously affecting their growth, development, yield and revenue. In order to clarify the harmful characteristics and influencing mechanisms of late frost on walnuts in China, the influencing factors of the degree of damage caused by late frost on walnuts, as well as the changes in physiological and biochemical indicators and molecular mechanisms in response to low temperature stress were reviewed. It was found that research on physiological changes focused on the three aspects including cell membrane system (relative conductivity), antioxidant system (malondialdehyde, superoxide dismutase, peroxidase and catalase) and osmoregulation substances (soluble sugars, soluble protein and proline), in addition, the expression levels of genes such as JrPAL, JrJAZ, JrGRAS, JrICE1, JrCBF/JrDREB1, JrCOR, JrWRKY, JrDHN, JrGST, and JrPUB were induced by low temperature stress. And prospects were put for future research directions, which provided scientific basis for further exploring the response mechanism of walnuts to late frost damage and adopting effective production measures. Triphenyltin (TPT) is an organotin pollutant widely found in the aquatic environment. It has endocrine-disrupting and osmotic pressure toxicity. In this study, the physiological and biochemical effects of TPT and various salinities were investigated in different tissues (gut, gill, and brain) of marine medaka. The exposure experiments were conducted for 42 days in different salinities (0, 15, and 30 ppt) without TPT exposure and in different salinity groups with TPT exposure concentrations of 100 ng/L, respectively. The results showed that the Na+-K+-ATPase (NKA) and Ca2+ATPase activity had significant tissue-specific differences, with the highest activity observed in the gills, indicating their major contribution to osmoregulation. Changes in salinity also resulted in significant alterations in the ion concentrations of the gut and gill tissues in the 0-C and 15-C groups. While the changes in Na+ and Cl− were relatively stable, the presence of TPT disrupted the regulation of Ca2+ and K+. In conclusion, substantial variations were observed in the osmoregulatory capacity of marine medaka tissues. Environmental concentrations of TPT had little effect on osmotic enzyme activity but interfered with the regulation of Ca2+ and K+ concentrations in the tissues. The invasive plant Wedelia trilobata benefits in various aspects, such as nutrient absorption and environmental adaptability, by establishing a close symbiotic relationship with arbuscular mycorrhizal fungi (AMF). However, our understanding of whether AMF can benefit W. trilobata by influencing its metabolic profile remains limited. In this study, Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was conducted to analyze the metabolites of W. trilobata under AMF inoculation. Metabolomic analysis identified 119 differentially expressed metabolites (DEMs) between the groups inoculated with AMF and those not inoculated with AMF. Compared to plants with no AMF inoculation, plants inoculated with AMF showed upregulation in the relative expression of 69 metabolites and downregulation in the relative expression of 50 metabolites. AMF significantly increased levels of various primary and secondary metabolites in plants, including amino acids, organic acids, plant hormones, flavonoids, and others, with amino acids being the most abundant among the identified substances. The identified DEMs mapped 53 metabolic pathways, with 7 pathways strongly influenced by AMF, particularly the phenylalanine metabolism pathway. Moreover, we also observed a high colonization level of AMF in the roots of W. trilobata, significantly promoting the shoot growth of this plant. These changes in metabolites and metabolic pathways significantly affect multiple physiological and biochemical processes in plants, such as free radical scavenging, osmotic regulation, cell structure stability, and material synthesis. In summary, AMF reprogrammed the metabolic pathways of W. trilobata, leading to changes in both primary and secondary metabolomes, thereby benefiting the growth of W. trilobata and enhancing its ability to respond to various biotic and abiotic stressors. These findings elucidate the molecular regulatory role of AMF in the invasive plant W. trilobata and provide new insights into the study of its competitive and stress resistance mechanisms. Freshwater scarcity demands exploration of alternative resources like saline water and soils. Understanding the molecular mechanisms behind NaCl regulation in potential crop plants becomes increasingly important for promoting saline agriculture. This study investigated the euhalophyte Salicornia europaea, analyzing its gene expression, yield, and total phenolic compounds under hydroponic cultivation. We employed five salinity levels (0, 7.5, 15, 22.5, and 30 g/L NaCl) across five harvests at 15-day intervals, capturing plant development. Notably, this design deviated from conventional gene expression studies by recording organ-specific responses (shoots and roots) in plants adapted to long-term salinity treatments at various developmental stages. The highest fresh mass of S. europaea was observed four months after germination in 15 g/L NaCl. Identifying a reliable set of reference genes for normalizing gene expression data was crucial due to comparisons across shoots, roots, developmental stages, and salinity levels. A set of housekeeping genes – ubiquitin c (SeUBC), actin (SeActin) and dnaJ-like protein (SeDNAJ) – was identified for this purpose. Interestingly, plants grown without NaCl (0 g/L) displayed upregulation of certain genes associated with a NaCl deficiency related nutritional deprivation. These genes encode a tonoplast Na+/H+-antiporter (SeNHX1), a vacuolar H+-ATPase (SeVHA-A), two H+-PPases (SeVP1, SeVP2), a hkt1-like transporter (SeHKT), a vinorine synthase (SeVinS), a peroxidase (SePerox), and a plasma membrane Na+/H+-antiporter (SeSOS1). Other genes encoding an amino acid permease (SeAAP) and a proline transporter (SeProT) demonstrated marginal or dispersing salinity influence, suggesting their nuanced regulation during plants development. Notably, osmoregulatory genes (SeOsmP, SeProT) were upregulated in mature plants, highlighting their role in salinity adaptation. This study reveals distinct regulatory mechanisms in S. europaea for coping with varying salinity levels. Identifying and understanding physiological reactions and sodium responsive key genes further elucidate the relationship between sodium tolerance and the obligate sodium requirement as a nutrient in euhalophytes.

Background Drought is the most significant factor limiting maize production, given that maize is a crop with a high water demand. Therefore, studies investigating the mechanisms underlying the drought tolerance of maize are of great importance. There are no studies comparing drought tolerance among economically important subspecies of maize. This study aimed to reveal the differences between the physio-biochemical, enzymatic, and molecular mechanisms of drought tolerance in dent (Zea mays indentata), popcorn (Zea mays everta), and sugar (Zea mays saccharata) maize under control (no-stress), moderate, and severe drought stress. Methods Three distinct irrigation regimes were employed to assess the impact of varying levels of drought stress on maize plants at the V14 growth stage. These included normal irrigation (80% field capacity), moderate drought (50% field capacity), and severe drought (30% field capacity). All plants were grown under controlled conditions. The following parameters were analyzed: leaf relative water content (RWC), loss of turgidity (LOT), proline (PRO) and soluble protein (SPR) contents, membrane durability index (MDI), malondialdehyde (MDA), and hydrogen peroxide (H2O2) content, the antioxidant enzyme activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), and catalase (CAT). Additionally, the expression of heat shock proteins (HSPs) was examined at the transcriptional and translational levels. Results The effects of severe drought were more pronounced in sugar maize, which had a relatively high loss of RWC and turgor, membrane damage, enzyme activities, and HSP90 gene expression. Dent maize, which is capable of maintaining its RWC and turgor in both moderate and severe droughts, and employs its defense mechanism effectively by maintaining antioxidant enzyme activities at a certain level despite less MDA and H2O2 accumulation, exhibited relatively high drought tolerance. Despite the high levels of MDA and H2O2 in popcorn maize, the up-regulation of antioxidant enzyme activities and HSP70 gene and protein expression indicated that the drought coping mechanism is activated. In particular, the positive correlation of HSP70 with PRO and HSP90 with enzyme activities is a significant result for studies examining the relationships between HSPs and other stress response systems. The discrepancies between the transcriptional and translational findings provide an opportunity for more comprehensive investigations into the role of HSPs in stress conditions.

Osmoregulation is essential for aquatic organisms to balance their internal fluids and ions and adapt to environmental changes. The gills are a crucial organ for maintaining osmotic balance in fish by regulating the uptake and excretion of ions. Changes in the salinity of the aquatic environment can lead to alterations in the expression of genes within gill cells, which helps to maintain the internal salinity balance and allows the fish to adapt to different salinity levels. In this study, we used transcriptomic data of the gill from the euryhaline Takifugu obscurus and the stenohaline Takifugu rubripes, as well as their hybrid offspring (Tor1 and Tor2), to investigate how the gill contributes to osmoregulation in response to freshwater and seawater. Our study revealed 8 commonly differentially expressed genes among these four fish species. Analysis of the T. obscurus transcriptome data has demonstrated that SLC1a5, a member of the solute carrier (SLC) family, and Gper1, a G-protein coupled estrogen receptor 1-like, are enriched in many biological processes and might be important for its osmoregulation in freshwater. Furthermore, PDE8B, Lrrc9 and Cplx4 might be important osmoregulatory genes that are inherited by offspring in T. obscurus. Moreover, upon encountering changes in water salinity, T. rubripes and the hybrid offspring Tor1 reveal a marked concentration of differentially expressed genes in metabolic pathways within their gills, whereas T. obscurus and the hybrid offspring Tor2 show a more even distribution of DEGs across various signaling pathways. Finally, the Pearson correlation analysis revealed that the T. rubripes displayed a significant correlation with Tor1, while the T. obscurus was significantly related to Tor2. Overall, this study provides a theoretical foundation for understanding the salinity adaptation mechanisms in hybrid offspring of Takifugu, and holds significant implications for guiding the breeding of economically valuable Takifugu strains with improved traits.

This study investigated the effects of dietary glutamine on growth performance, antioxidant capacity, and osmoregulation in euryhaline fish (Oreochromis niloticus) under hyperosmotic stress. Nile tilapia was fed four diets with varying glutamine concentrations (0 %, 0.75 %, 1.5 %, and 3 %) at 0 or 16 ppt for 42 days. The results demonstrated that dietary supplementation of 0.75 % glutamine significantly enhanced the growth performance of Nile tilapia under both 0 ppt and 16 ppt salinity. Hyperosmotic stress significantly reduced the crude protein and crude lipid contents while elevated the ash content of the fish. Under hyperosmotic stress, dietary glutamine supplementation significantly increased the antioxidant and immune capacities, and attenuated apoptosis in the liver of tilapia. The expression of ion transporter genes in the gills and kidneys were both significantly increased by dietary glutamine. Meanwhile, glutamine supplementation significantly reduced the levels of inorganic ions in serum while increased serum osmolality. Hyperosmotic stress significantly activated the glycolysis, gluconeogenesis, and pentose phosphate pathways in the liver of tilapia. However, supplementation of glutamine under hyperosmotic stress would significantly enhance the ability of gluconeogenesis of tilapia in the liver. The results of this research suggested that dietary glutamine might promote the growth performance of fish by enhancing their antioxidant capacity and osmoregulation capacity under chronic hyperosmotic stress. This work investigates the role of glutamine in osmolarity regulation and extends the understanding of its mechanism of action, which could provide a reference for hyperosmotic adaptation in aquatic animals.

Pericardial edema is commonly observed in zebrafish embryo-based chemical toxicity screens, and a mechanism underlying edema may be disruption of embryonic osmoregulation. Therefore, the objective of this study was to identify whether triphenyl phosphate (TPHP) - a widely used aryl phosphate ester-based flame retardant - induces pericardial edema via impacts on osmoregulation within embryonic zebrafish. In addition to an increase in TPHP-induced microridges in the embryonic yolk sac epithelium, an increase in ionic strength of exposure media exacerbated TPHP-induced pericardial edema when embryos were exposed from 24 to 72 h post-fertilization (hpf). However, there was no difference in embryonic sodium concentrations in situ within TPHP-exposed embryos relative to embryos exposed to vehicle (0.1% DMSO) from 24 to 72 hpf. Interestingly, increasing the osmolarity of exposure media with mannitol (an osmotic diuretic which mitigates TPHP-induced pericardial edema) and increasing the ionic strength of the exposure media (which exacerbates TPHP-induced pericardial edema) did not affect embryonic doses of TPHP, suggesting that TPHP uptake was not altered under these varying experimental conditions. Overall, our findings suggest that TPHP-induced pericardial edema within zebrafish embryos is dependent on the ionic strength of exposure media, underscoring the importance of further standardization of exposure media and embryo rearing protocols in zebrafish-based chemical toxicity screening assays.

Abstract Aims and objectives To analyze anti-MMP mode of action of Quaternary Ammonium Silane (QAS, codenamed as k21) by binding onto specific MMP site using computational molecular simulation and Anti-Sortase A (SrtA) mode of action by binding onto specific site using computational molecular simulation. Materials and methods In silico Molecular Dynamics (MD) was used to determine the interactions of K21 inside the pocket of the targeted protein (crystal structure of fibroblast collagenase-1 complexed to a diphenyl-ether sulphone based hydroxamic acid; PDB ID: 966C; Crystal structure of MMP-2 active site mutant in complex with APP-derived decapeptide inhibitor. MD simulations were accomplished with the Desmond package in Schrödinger Drug Discovery Suite. Blood samples (~ 0.5 mL) collected into K2EDTA were immediately transferred for further processing using the Litron MicroFlow® PLUS micronucleus analysis kit for mouse blood according to the manufacturer’s instructions. Bacterial Reverse Mutation Test of K21 Molecule was performed to evaluate K21 and any possible metabolites for their potential to induce point mutations in amino acid-requiring strains of Escherichia coli (E. coli) (WP2 uvrA (tryptophan-deficient)). Results Molecular Simulation depicted that K21 has a specific pocket binding on various MMPs and SrtA surfaces producing a classical clouting effect. K21 did not induce micronuclei, which are the result of chromosomal damage or damage to the mitotic apparatus, in the peripheral blood reticulocytes of male and female CD-1 mice when administered by oral gavage up to the maximum recommended dose of 2000 mg/kg. The test item, K21, was not mutagenic to Salmonella typhimurium (S. typhimurium) strains TA98, TA100, TA1535 and TA1537 and E. coli strain WP2 uvrA in the absence and presence of metabolic activation when tested up to the limit of cytotoxicity or solubility under the conditions of the test. Conclusion K21 could serve as a potent protease inhibitor maintaining the physical and biochemical properties of dental structures.

Abstract Background Syringomyelia (SM) is characterized by the development of fluid-filled cavities, referred to as syrinxes, within the spinal cord tissue. The molecular etiology of SM post-spinal cord injury (SCI) is not well understood and only invasive surgical based treatments are available to treat SM clinically. This study builds upon our previous omics studies and in vitro cellular investigations to further understand local fluid osmoregulation in post-traumatic SM (PTSM) to highlight important pathways for future molecular interventions. Methods A rat PTSM model consisting of a laminectomy at the C7 to T1 level followed by a parenchymal injection of 2 μL quisqualic acid (QA) and an injection of 5 μL kaolin in the subarachnoid space was utilized 6 weeks after initial surgery, parenchymal fluid and cerebrospinal fluid (CSF) were collected, and the osmolality of fluids were analyzed. Immunohistochemistry (IHC), metabolomics analysis using LC–MS, and mass spectrometry-based imaging (MSI) were performed on injured and laminectomy-only control spinal cords. Results We demonstrated that the osmolality of the local parenchymal fluid encompassing syrinxes was higher compared to control spinal cords after laminectomy, indicating a local osmotic imbalance due to SM injury. Moreover, we also found that parenchymal fluid is more hypertonic than CSF, indicating establishment of a local osmotic gradient in the PTSM injured spinal cord (syrinx site) forcing fluid into the spinal cord parenchyma to form and/or expand syrinxes. IHC results demonstrated upregulation of betaine, ions, water channels/transporters, and enzymes (BGT1, AQP1, AQP4, CHDH) at the syrinx site as compared to caudal and rostral sites to the injury, implying extensive local osmoregulation activities at the syrinx site. Further, metabolomics analysis corroborated alterations in osmolality at the syrinx site by upregulation of small molecule osmolytes including betaine, carnitine, glycerophosphocholine, arginine, creatine, guanidinoacetate, and spermidine. Conclusions In summary, PTSM results in local osmotic disturbance that propagates at 6 weeks following initial injury. This coincides with and may contribute to syrinx formation/expansion.

To investigate the synergistic effect of IAA and melatonin (MT) on three plants to alleviate the effects of salt damage on plants, we aim to determine the optimal concentrations of exogenous hormone treatments that improve salinity resistance for each species. In this experiment, three desert plants, Sarcozygium xanthoxylon, Nitraria tangutorum, and Ammopiptanthus mongolicus, which are common in Wuhai City, were used as plant materials. Two time periods (12 h,24 h) of exogenous hormone IAA (100 μmol/L) and exogenous melatonin concentration (0, 100, 200, 300 μmol/L) were used to treat the three desert plants in saline soil under different conditions of exogenous IAA and exogenous melatonin. The results indicate that under different concentrations of exogenous IAA and melatonin, the germination rate and vigor of the three desert plant species in saline-alkaline soil improved. However, as the concentration of melatonin increased, the germination rate and vigor of these desert plants were inhibited. Whereas, plant height, root length, leaf length, fresh weight, dry weight, and root vigor of the three desert plants were alleviated under different conditions of exogenous IAA and exogenous melatonin. under the action of two exogenous hormones, the low concentration of melatonin decreased their malondialdehyde content and increased their proline content. As melatonin levels increased, the activity of antioxidant enzymes also rose initially, followed by a subsequent decline. This study highlights the synergistic effects of two exogenous hormones on the critical role of cell osmomodulators and antioxidant enzyme activity in combating salinity damage in three desert plants.

Background: Erythritol is a four-carbon polyol with an unclear role in metabolism of some unconventional yeasts. Its production has been linked to the osmotic stress response, but the mechanism of stress protection remains unclear. Additionally, erythritol can be used as a carbon source. In the yeast Yarrowia lipolytica, its assimilation is activated by the transcription factor Euf1. The study investigates whether this factor can link erythritol to other processes in the cell. Results: The research was performed on two closely related strains of Y. lipolytica: MK1 and K1, where strain K1 has no functional Euf1. Cultures were carried out in erythritol-containing and erythritol-free media. Transcriptome analysis revealed the effect of Euf1 on the regulation of more than 150 genes. Some of these could be easily connected with different aspects of erythritol assimilation, such as: utilization pathway, a new potential isoform of transketolase, or polyol transporters. However, many of the upregulated genes have never been linked to metabolism of erythritol. The most prominent examples are the degradation pathway of branched-chain amino acids and the glyoxylate cycle. The high transcription of genes affected by Euf1 is still dependent on the erythritol concentration in the medium. Moreover, almost all up-regulated genes have an ATGCA motif in the promoter sequence. Conclusions: These findings may be particularly relevant given the increasing use of erythritol-induced promoters in genetic engineering of Y. lipolytica. Moreover, use of this yeast in biotechnological processes often takes place under osmotic stress conditions. Clonal integration of defense or stress signal induced systemic resistance in leaf of interconnected ramets. However, similar effects of stress signal in root are poorly understood within clonal network. Clonal fragments of Centella asiaticas with first-young, second-mature, third-old and fourth-oldest ramets were used to investigate transportation or sharing of stress signal among interconnected ramets suffering from low water availability. Compared with control, oxidative stress in root of the first-young, second-mature and third-old ramets was significantly alleviated by exogenous ABA application to the fourth-oldest ramets as well as enhancement of antioxidant enzyme (SOD, POD, CAT and APX) activities and osmoregulation ability. Surface area and volume in root of the first-young ramets were significantly increased and total length in root of the third-old ramets was significantly decreased. POD activity in root of the fourth-oldest and third-old ramets was significantly enhanced by exogenous ABA application to the first-young ramets. Meanwhile, total length and surface area in root of the fourth-oldest and third-old ramets were significantly decreased. Ratio of belowground to aboveground biomass in the whole clonal fragments was significantly increased by exogenous ABA application to the fourth-oldest or first-young ramets. It is suggested that transportation or sharing of stress signal may induce systemic resistance in root of interconnected ramets. Specially, transportation or sharing of stress signal against phloem flow was observed in the experiment. Possible explanation is that rapid recovery of foliar photosynthesis in first-young ramets subjected to exogenous ABA application can partially reverse phloem flow within clonal network. Proximity to the seashore is a critical structuring factor of coastal wetlands due to its influence on varying sources of nutrients (marine and terrestrial) and on the spatial gradient of salinity (higher salinity closer to the seashore). The spatial gradient of salinity may impact organisms because most organisms need to maintain an osmotic balance. Osmoregulation is energetically costly, and exposure to salinity should induce a trade‐off in energetic allocation between osmoregulation and other competing functions such as growth rates and movement patterns. In this study, we used a capture–mark–recapture design during 3 consecutive years to investigate how distance to the sea influences growth rates and foraging movements (outside reproduction) in the western spadefoot toad (Pelobates cultripes), a typical coastal amphibian, in three populations from the French Atlantic coast. Growth rates were lower in larger individuals and in individuals living closer to the seashore. Distances travelled between captures were very limited (~20 m). Between years, these distances were larger for individuals located on the beach rather than inland, but were not influenced by body size or sex. Exposure to salinity and associated costs of osmoregulation may explain lower growth rates closer to the shoreline. The mechanisms underlying the effects of location on distances between captures remain to be identified, but may be related to foraging for abundant prey items on the wrack line. Our study confirms the remarkable terrestrial site fidelity in P. cultripes, which might be detrimental to coastal populations if localized perturbations affect coastal environments. Cutaneous evaporative water loss (CEWL) makes up a significant portion of total evaporative water loss in many terrestrial vertebrates. CEWL changes on evolutionary and acclimatory timescales in response to temperature and humidity. However, the lability of CEWL on acute timescales is unknown. To examine this, we increased or decreased body temperatures of western fence lizards (Sceloporus occidentalis) over a 15-min period while continuously recording CEWL with a handheld evaporimeter. CEWL increased in response to heating and decreased in response to cooling on the order of seconds. However, CEWL was different between heating and cooling groups at a common body temperature. We observed the same positive relationship between CEWL and body temperature, as well as the difference in CEWL between treatments, for deceased lizards that we opportunistically measured. However, deceased lizards had more extreme CEWL values for any given body temperature and treatment. Overall, our results suggest that both structural traits and active physiological processes likely influence the rates and plasticity of CEWL. [ABSTRACT FROM AUTHOR]

Salt stress poses a significant challenge to crop productivity, and understanding the genetic basis of salt tolerance is paramount for breeding resilient soybean varieties. In this study, a soybean natural population was evaluated for salt tolerance during the germination stage, focusing on key germination traits, including germination rate (GR), germination energy (GE), and germination index (GI). It was seen that under salt stress, obvious inhibitions were found on these traits, with GR, GE, andGI diminishing by 32% to 54% when compared to normal conditions. These traits displayed a coefficient of variation (31.81% to 50.6%) and a substantial generalized heritability (63.87% to 86.48%). Through GWAS, a total of 1841 significant singlenucleotide polymorphisms (SNPs)were identified to be associated with these traits, distributed across chromosome 2, 5, 6, and 20. Leveraging these significant association loci, 12 candidate genes were identified to be associated with essential functions in coordinating cellular responses, regulating osmotic stress, mitigating oxidative stress, clearing reactive oxygen species (ROS), and facilitating heavymetal ion transport - all of which are pivotal for plant development and stress tolerance. To validate the candidate genes, quantitative real-time polymerase chain reaction (qRT-PCR) analysis was conducted, revealing three highly expressed genes (Glyma.02G067700, Glyma.02G068900, and Glyma.02G070000) that play pivotal roles in plant growth, development, and osmoregulation. In addition, based on these SNPs related with salt tolerance, KASP (Kompetitive Allele-Specific PCR)markers were successfully designed to genotype soybean accessions. Tomato (Solanum lycopersicum L.) is an important vegetable crop that grows easily under controlled conditions, such as in greenhouses and hydroponics. To overcome freshwater scarcity, researchers are searching for alternatives to groundwater sources such as desalinated water (saline water) for irrigation. High salinity in irrigation water alters physiological functions and crop development, thereby reducing the yield. Best management practices and the use of grafted tomato plants on salt-tolerant rootstocks can alleviate salinity stress. The present study was conducted to address the impact of salinity stress on leaf transpiration (Tr), stomatal conductance (gs), photosynthesis (Pn), leaf chlorophyll content, proline content, and yield of hydroponically cultivated tomato plants. Saline (NaCl) water was used for the preparation of nutrient solution with three salinity levels, electrical conductivity (EC, dS m−1) of 2.5 (control), 6.0, and 9.5. Three commercial tomato cultivars (Valouro-RZ, Ghandora-F1, and Feisty-Red) were used. Both self-rooted plants and plants grafted onto Maxifort rootstocks were transplanted onto a perlite substrate. The recorded data revealed that all studied cultivars were critically affected by higher salinity (≈ 9.5 dS m-1) compared to low (≈ 2.5 dS m-1) and medium (≈ 6.0 dS m-1) salinity levels. The Variations in Tr, Pn, gs, chlorophyll content of leaf, and yield between medium and high salinity trials were reported at 3%, 5%, 9%, 5%, and 7.1%, respectively, whereas no significant differences were observed between low and medium salinities. However, at medium salinity levels, grafted plants performed better in photosynthesis than non-grafted plants. This is due to the accumulation of leaf proline, which maintains osmotic regulation and photosynthetic activity by preventing cell damage at medium salinities. Hence, this study confirmed the use of saline water for growing tomatoes under hydroponic conditions up to an EC of 6.0 dS m-1 including the EC of nutrient fertilizers. [ABSTRACT FROM AUTHOR]
