Among the dermatophyte species, Trichophyton rubrum is one of the most important affecting adult individuals, and tends to produce infections of an evolutionary course of the chronic type, with disseminated, persistent and recurrent lesions. The laboratory diagnosis of dermatophytoses in routine is based on the phenotypic characteristics of the fungal strains isolated from the lesions in specific culture media, for this it is necessary to study the macro and micromorphology aspects of the colonies and, biochemical tests when necessary. However different T. rubrum isolates may show variability intheir phenotypic characteristics, and these may be related to genetic differences. The purpose of this study was to describe different phenotypes and to evaluate the possible existence of genotypic variability of T. rubrum isolates from pacients with dermatophytoses treated at a referral center in tropical dermatology in the city of Manaus-AM. For this, the method of random amplification of polymorphic DNA was used with use of the primer 1: 5-GGTGCGGGAA-3. From the phenotyping of 55 isolates of T. rubrum, 07 phenotypes were evidenced. The analysis of T.rubrum isolates using the RAPD with primer 1 allowed the discrimination of 10 distinct genotypic profiles, distributed in three groups, with the majority of isolates belonging to the same genetic group, group I, with 85% similarity. No association were found between the different phenotypes and genotypes, or between these and the clinical data of the patients. Despite the proven potencial of the RAPD technique for studies on the genetic variability of T. rubrum, new methods that make it possible a correlation between the different genotypes and the clinical and epidemiological aspects of dermatophytoses still need to be evaluated or developed. Dentre as esp??cies dermatof??ticas, o Trichophyton rubrum ?? uma das mais importantes acometendo indiv??duos adultos, e tende a produzir infec????es de curso evolutivo do tipo cr??nico, com les??es disseminadas, persistentes e recidivantes. O diagn??stico laboratorial das dermatofitoses em rotina baseia-se nas caracter??sticas fenot??picas das cepas f??ngicas isoladas das les??es em meios de cultura espec??ficos, para isto ?? necess??rio estudar os aspectos da macro e da micromorfologia das col??nias e, provas bioqu??micas quando necess??rio. Entretanto, diferentes isolados de T. rubrum podem apresentar variabilidade das suas caracter??sticas fenot??picas, e essas podem estar relacionadas a diferen??as gen??ticas. O prop??sito deste estudo foi descrever diferentes fen??tipos e avaliar a poss??vel exist??ncia de variabilidade genot??pica de isolados de T. rubrum de pacientes com dermatofitoses atendidos em um centro de refer??ncia em dermatologia tropical na cidade de Manaus-AM. Para isto foi empregado o m??todo de amplifica????o rand??mica do DNA polim??rfico com a utiliza????o do primer 1: 5-GGTGCGGGAA-3. A partir da fenotipagem de 55 isolados de T. rubrum foram evidenciados 07 fen??tipos. A an??lise dos isolados de T. rubrum pelo m??todo de RAPD utilizando o primer 1 permitiu a discrimina????o de 10 perfis genot??picos distintos, distribu??dos em tr??s grupos, sendo que, a maioria dos isolados pertenceu a um mesmo grupo gen??tico, o grupo I, com 85% de similaridade. N??o foram encontradas associa????es entre os diferentes fen??tipos e os gen??tipos, ou entre estes e os dados cl??nicos dos pacientes. Apesar do comprovado potencial da t??cnica do RAPD para estudos da variabilidade gen??tica de T. rubrum, novos m??todos que permitam fazer uma correla????o entre os diferentes gen??tipos e os aspectos cl??nicos e epidemiol??gicos das dermatofitoses ainda precisam ser avaliados ou desenvolvidos.
