The present paper describes the development of a method for the determination of copper in airborne particulate matter using slurry sampling and chemical vapor generation atomic absorption spectrometry (CVG AAS). Chemometric tools were employed to characterize the influence of several factors on the generation of volatile copper species. First, a two-level full factorial design was performed that included the following chemical variables: hydrochloric acid concentration, tetrahydroborate concentration, sulfanilamide concentration and tetrahydroborate volume, using absorbance as the response. Under the established experimental conditions, the hydrochloric acid concentration had the greatest influence on the generation of volatile copper species. Subsequently, a Box-Behnken design was performed to determine the optimum conditions for these parameters. A second chemometric study employing a two-level full factorial design was performed to evaluate the following physical factors: tetrahydroborate flow rate, flame composition, alcohol volume and sample volume. The results of this study demonstrated that the tetrahydroborate flow rate was critical for the process. The chemometric experiments determined the following experimental conditions for the method: hydrochloric acid concentration, 0.208 M; tetrahydroborate concentration, 4.59%; sulfanilamide concentration, 0.79%; tetrahydroborate volume, 2.50 mL; tetrahydroborate flow rate, 6.50 mL min(-1); alcohol volume, 200 µL; and sample volume, 7.0 mL. Thus, this method, using a slurry volume of 500 µL and a final dilution of 7 mL, allowed for the determination of copper with limits of detection and quantification of 0.30 and 0.99 µg L(-1), respectively. Precisions, expressed as RSD%, of 4.6 and 2.8% were obtained using copper solutions at concentrations of 5.0 and 50.0 µg L(-1), respectively. The accuracy was evaluated by the analysis of a certified reference material of urban particulate matter. The copper concentration obtained was 570±63 mg kg(-1), and the certified value was 610±70 mg kg(-1). This method was applied for the determination of copper in airborne particulate matter samples collected in two Brazilian regions of Bahia State, Brazil. The present study proposes a fast and simple analytical methodology employing C18 SPE cartridges (for preconcentration and clean-up), and a ultra-fast liquid chromatography coupled to fluorescence detector (UFLC-FLD) for determination of the following endocrine disrupters (ED): bisphenol A (BPA), 4-n-nonylphenol (4NNP), 4-n-octylphenol (4NOP), 4-t-octylphenol (4TOP), estriol (E3), estrone (E1), 17β-estradiol (E2) and 17α-ethynylestradiol (EE2) in seawater. The proposed method was developed, optimized and validated. Separation was done by a total running time of 10 min in a Shim-pack XR-ODS C-18 (2.0 mm ID × 50 mm) chromatographic column, mobile phases were acetonitrile/ultra-pure water under gradient programming; eluent flow rate at 0.120 mL min(-1); column temperature set at 60 °C; emission wavelength of 306 nm and excitation wavelength of 280 nm. The method was validated through assessment of the following parameters: linear range, linearity, selectiveness, precision, recovery test, limit of detection (LOD), and limit of quantification (LOQ). Recoveries ranged from 91% (for EE2) to 104% (for 4NNP) and also was found a suitable repeatability (RSD <4.5%) for all considered compounds. LOD and LOQ ranged from 2.0 ng L(-1) (EE2) to 23 ng L(-1) (E1) and 9.3 ng L(-1) (EE2) to 96 ng L(-1) (E1), respectively. High-resolution continuum source graphite furnace atomic absorption spectrometry, because of the use of only one radiation source for all elements, offers the possibility of sequential determination of two or more elements from the same sample aliquot if their volatilities are significantly different. Cd and Cr were determined sequentially in samples of biomass and biomass ashes employing direct solid sample analysis. The use of a chemical modifier was found to be not necessary, and calibration could be carried out using aqueous standard solutions. A pyrolysis temperature of 400°C and an atomization temperature of 1500°C were used for the determination of Cd; no losses of Cr were observed at this temperature. After the atomization of Cd the wavelength was changed and Cr atomized at 2600°C. The limits of detection (LOD) and quantification (LOQ) were 1.1 μg kg(-1) and 3.7 μg kg(-1), respectively, for Cd and 21 μg kg(-1) and 70 μg kg(-1), respectively, for Cr using the most sensitive line at 357.869 nm, or 90 μg kg(-1) and 300 μg kg(-1), respectively, using the less sensitive line at 428.972 nm. The precision, expressed as relative standard deviation was around 10%, which is typical for direct solid sample analysis. The values found for Cd in biomass samples were between <1.1 µg kg(-1) and 789 µg kg(-1), whereas those for Cr were between 7.9 mg kg(-1) and 89 mg kg(-1); the values found in the ashes were significantly lower for Cd, between <1.1 µg kg(-1) and 6.3 µg kg(-1), whereas the trend was not so clear for Cr, where the values were between 3.4 mg kg(-1) and 28 mg kg(-1)., (© 2013 Elsevier B.V. Plectranthus spp (Lamiaceae) are plants of economic importance because they are sources of aromatic essential oils and are also cultivated and several species of this genus are used as folk medicines. This paper describes the effects of different concentrations of the 2,4-dichlorophenoxyacetic acid (2,4-D) and 1-naphthaleneacetic acid (NAA) on the induction of callus from nodal segments of Plectranthus ornatus Codd and in the production of volatile organic compounds (monoterpenes and sesquiterpenes). The 20 and 40 day calli were subjected to solid phase micro extraction (HS-SPME) and submitted to GCMS analysis. Variations in VOCs between the samples were observed and, a direct relationship was observed between of the major constituent detected (α-terpinyl acetate) and the monoterpenes α-thujene, α-pinene, β-pinene, camphene, sabinene and α-limonene that were present in the volatile fractions. Besides α-terpinyl acetate, isobornyl acetate and α-limonene were also major constituents. Variations were observed in VOCs in the analyzed periods. The best cultivation media for the production of VOCs was found to be MS0 (control). Moderate success was achieved by treatment with 2.68 µM and 5:37 µM NAA (Group 2). With 2,4-D (9.0 µM), only the presence of α-terpinyl acetate and isocumene were detected and, with 2.26 µM of 2,4-D was produced mainly α-terpinyl acetate, α-thujene and β-caryophyllene (16.2%). The VOC profiles present in P. ornatus were interpreted using PCA and HCA. The results permitted us to determine the best cultivation media for VOC production and, the PCA and HCA analysis allowed us to recognize four groups among the different treatments from the compounds identified in this set of treatments.

An analytical method has been developed for the determination of sulfur in coal using direct solid sample analysis in a graphite tube furnace and high-resolution continuum source molecular absorption spectrometry (HR-CS GF MAS). The molecular absorbance of the carbon monosulfide molecule (CS), which is formed in the vaporization stage, has been measured using the rotational line at 258.033 nm. Several chemical modifiers were tested and Ru, applied as permanent modifier was chosen, because it exhibited the best performance. The optimum pyrolysis and vaporization temperatures were found to be 500 °C and 2200 °C, respectively. Aqueous standard solutions prepared from l-cysteine were used for calibration, as the linear regression obtained for this standard was not significantly different from that for a certified coal reference material (CRM) according to a Student t-test. The results obtained for sulfur in three coal CRM and six additional samples also showed no significant difference for the two calibration techniques according to the same statistical test. The sulfur concentration in the coal samples was found between 3.5 mg g(-1) and 33.7 mg g(-1) with a typical repeatability around 10%. A novel analytical approach combining solid-phase microextraction (SPME)/gas chromatography ion trap mass spectrometry (GC-IT-MS) was developed for the detection and quantification N,N-dimethyltryptamine (DMT), a powerful psychoactive indole alkaloid present in a variety of South American indigenous beverages, such as ayahuasca and vinho da jurema. These particular plant products, often used within a religious context, are increasingly consumed throughout the world following an expansion of religious groups and the availability of plant material over the Internet and high street shops. The method described in the present study included the use of SPME in headspace mode combined GC-IT-MS and included the optimization of the SPME procedure using multivariate techniques. The method was performed with a polydimethylsiloxane/divinylbenzene (PDMS/DVB) fiber in headspace mode (70 min at 60 °C) which resulted in good precision (RSD<8.6%) and accuracy values (71-109%). Detection and quantification limits obtained for DMT were 0.78 and 9.5 mg L(-1), respectively and good linearity (1.56-300 mg L(-1), r(2)=0.9975) was also observed. In addition, the proposed method showed good robustness and allowed for the minimization of sample manipulation. Five jurema beverage samples were prepared in the laboratory in order to study the impact of temperature, pH and ethanol on the ability to extract DMT into solution. The developed method was then applied to the analysis of twelve real ayahuasca and vinho da jurema samples, obtained from Brazilian religious groups, which revealed DMT concentration levels between 0.10 and 1.81 g L(-1)., (Copyright © 2013 Elsevier B.V. Studies have investigated the exposure levels of carbonyl compounds (CC) in the indoor and outdoor air of homes, vehicles, workplaces, urban and industrial areas, and rural sites. However, an investigation of these emissions and occupational exposure to CC in charcoal production facilities has not been previously conducted. The objective of this study was to measure the atmospheric concentrations of several CC to assess the exposure of workers of two charcoal plants located north of Salvador, Bahia, Brazil. Stationary and personal samples were collected using Sep-Pak® C18 cartridges that were coated with a 0.2 % acidic solution of 2,4-dinitrophenylhydrazine. The quantification of the resulting 2,4-dinitrophenylhydrazone derivatives was conducted using a high-performance liquid chromatography system with UV detection. In the personal samples, the concentrations of formaldehyde, acetaldehyde, propanone, furfural, and C4 isomers (n-butanal-isobutanal-butanone) ranged from 12 to 139, 38 to 165, 136 to 483, 39 to 114, and 63 to 132 μg m(-3), respectively. In the stationary samples, the concentrations of these CC ranged from 20 to 160, 111 to 284, 328 to 644, 70 to 163, and 100 to 176 μg m(-3), respectively. When compared to the occupational exposure limits for 8 h, the concentrations of formaldehyde were often greater than the levels recommended by the American National Institute for Occupational Safety and Health, which indicates a health risk for charcoal workers. These results are the first reported concerning the occupational exposure to CC in charcoal plants.

The present study proposes an analytical methodology that employs ion chromatography-conductivity detection for simultaneous quantification of inorganic (F(-), Cl(-), NO(3)(-), SO(4)(2-), and PO(3)(-)), monocarboxylate (HCOO(-), CH(3)COO(-), propionate, n-butyrate, lactate, and pyruvate), dicarboxylate (oxalate and succinate), and tricarboxylate anions (citrate), as well as crustal cations (Li(+), Na(+), K(+), NH(4)(+), Ca(2+), Mg(2+)) at low pgm(-3) range in airborne particle samples in one single run. The optimized conditions for anions were as follows: 0.6 mmol L(-1) KOH for 0-14 min, 0.6-15 mmol L(-1) KOH 14-20 min, 15-38 mmol L(-1) KOH during 20-32 min and finally returned to 0.6 mmol L(-1) for a period of 3 min, thereafter the eluent flow rate was 0.38 mL min(-1). Similarly, for cations, isocratic elution was adjusted to 0.36 mL min(-1) at 17.5 mmol L(-1) H(2)SO(4). LOD ranged 3.0-130 pgm(-3) and LOQ was within 10-400 pgm(-3) (Li(+) and PO(4)(3-), respectively) as well as recoveries ranged 89% (Ca(2+)) to 120% (Li(+)). Major ions were successfully determined in real PM1 and PM2.5 samples. The method used here was found to be a comprehensive, simple, cheap and reliable procedure for studying ions in particulate matter (PM) samples even those from remote areas or near ecosystem natural conditions., (Copyright © 2012 Elsevier B.V. The consumption of ayahuasca, a hallucinogenic beverage used by indigenous communities in the Amazon, is increasing worldwide due to the expansion of syncretic religions founded in the north of Brazil in the first half of the twentieth century, such as Santo Daime and União do Vegetal. Another example is the jurema wine, a drink that originated from indigenous cultures of the northeast of Brazil. It is currently used for several religious practices throughout Brazil involving urban neo-shamanic rituals and syncretic Brazilian religions, such as Catimbó and Umbanda. Both plant products contain N,N-dimethyltryptamine which requires co-administration of naturally occurring monoamine oxidase inhibitors, for example β-carboline derivatives, in order to induce its psychoactive effects in humans. This review explores the cultural use of tryptamines and β-carbolines and focuses on the analytical techniques that have been recently applied to the determination of these compounds in ayahuasca, its analogues, and the plants used during the preparation of these beverages., (Copyright © 2012 John Wiley & Sons, Ltd.)

In this study, particulate matter (PM) were characterized from a place impacted by heavy-duty vehicles (Bus Station) fuelled with diesel/biodiesel fuel blend (B3) in the city of Londrina, Brazil. Sixteen priority polycyclic aromatic hydrocarbons (PAH) concentrations were analyzed in the samples by their association with atmospheric PM, mass size distributions and major ions (fluorite, chloride, bromide, nitrate, phosphate, sulfate, nitrite, oxalate; fumarate, formate, succinate and acetate; lithium, sodium, potassium, magnesium, calcium and ammonium). Results indicate that major ions represented 21.2% particulate matter mass. Nitrate, sulfate, and ammonium, respectively, presented the highest concentration levels, indicating that biodiesel may also be a significant source for these ions, especially nitrate. Dibenzo[a,h]anthracene and indeno[1,2,3,-cd]pyrene were the main PAH found, and a higher fraction of PAH particles was found in diameters lower than 0.25 μm in Londrina bus station. The fine and ultrafine particles were dominant among the PM evaluated, suggesting that biodiesel decreases the total PAH emission. However, it does also increase the fraction of fine and ultrafine particles when compared to diesel.

Three different procedures for sample preparation have been compared for the determination of Cu, Mo and Sb in airborne particulate matter (APM) collected on glass fiber filters using high-resolution continuum source graphite furnace atomic absorption spectrometry (HR-CS GF AAS). Direct solid sample analysis of the ground filters was compared with microwave-assisted acid leaching with aqua regia and ultrasound-assisted extraction also using aqua regia. The main absorption line at 324.754 nm or the secondary line at 216.509 nm was used for the determination of Cu, depending on the analyte content in the samples. The primary absorption line at 313.259 nm was used for Mo and the secondary line at 212.739 nm for Sb determination. The limits of detection (LOD, 3σ) found for the direct solid sampling method, based on ten atomizations of an unused filter were 15 μg g(-1) for all three analytes, corresponding to 40 ng m(-3) for a typical air volume of 1,440 m(3) collected over a period of 24h. The LOD for the other two methods were less than a factor of two inferior, but the total time required for an analysis was significantly longer. The repeatability of the measurements was between 3 and 9% (n=5), and the results obtained with the three methods did not show any significant difference. The ratio between the three analytes on the filters from areas of intense traffic was found to be around Cu:Mo:Sb≈4:1:1.4, which suggests that the source of all three elements is brake linings, i.e., related to automobile traffic. N,N-dimethyltryptamine (DMT) is a potent hallucinogen found in beverages consumed in religion rituals and neo-shamanic practices over the world. Two of these religions, Santo Daime and União do Vegetal (UDV), are represented in countries including Australia, the United States and several European nations. In some of this countries there have been legal disputes concerning the legalization of ayahuasca consumption during religious rituals, a beverage rich in DMT. In Brazil, even children and pregnant women are legally authorized to consume ayahuasca in a religious context. A simple and low-cost method based on matrix solid-phase dispersion (MSPD) and gas chromatography with mass spectrometric detection (GC-MS) has been optimized for the determination of N,N-dimethyltryptamine in Mimosa tenuiflora inner bark. The experimental variables that affect the MSPD method, such as the amounts of solid-phase and herbal sample, solvent nature, eluate volume and NaOH concentration were optimized using an experimental design. The method showed good linearity (r = 0.9962) and repeatability (RSD < 7.4%) for DMT compound, with detection limit of 0.12 mg/g. The proposed method was used to analyze 24 samples obtained locally. A single-drop microextraction (SDME) procedure was developed for the analysis of organophosphorus and pyrethroid pesticides in water by gas chromatography (GC) with flame ionization detection (GC-FID). The significant parameters that affect SDME performance, such as the selection of microextraction solvent, solvent volume, extraction time, and stirring rate, were studied and optimized using a tool screening factorial design. The limits of detection (LODs) in water for the four investigated compounds were between 0.3 and 3.0 microgL(-1), with relative standard deviations ranging from 7.7 to 18.8%. Linear response data were obtained in the concentration range of 0.9-6.0 microg L(-1) (lambda-cyhalothrin), 3.0-60.0 microg L(-1) (methyl parathion), 9.0-60.0 microg L(-1) (ethion), and 9.0-30.0 microg L(-1) (permethrin), with correlation coefficients ranging from 0.9337 to 0.9977. The relative recoveries for the spiked water ranged from 73.0 to 104%. Environmental water samples (n=26) were successfully analyzed using the proposed method and methyl parathion presented concentration up to 2.74 microg L(-1). The SDME method, coupled with GC-FID analysis, provided good precision, accuracy, and reproducibility over a wide linear range. Other highlights of the method include its ease of use and its requirement of only small volumes of both organic solvent and sample.

Carbonyl compounds (CC) play an important role in beverage aroma since they may affect flavor of wines, brandies, and beers, among others. For this reason, it is necessary to identify and quantify CC through adequate analytical techniques. This study is a proposal of both developing and optimization of a new analytical methodology that allows investigate C(1)-C(8 )CC in wines simultaneously by quantifying even those ones that are predominantly present in the adduct form hydroxylalkylsulfonic acids (HASA). The HASA dissociation is undertaken by specific alkaline media (pH 11). The developed methodology employed the LC with UV/VIS detection (lambda = 365 nm) technique under gradient elution in the way to reach both free-CC and bound-CC quantification. Results showed that binary gradient system using eluent A (MeOH/ACN/H(2)O 74.5:0.5:25% v/v/v) and eluent B (MeOH) reached the best separation condition of both lower and higher molecular mass CC. This proposed method allowed simultaneous quantification of formaldehyde, acetaldehyde, propanone, furfuraldehyde, butyraldehyde, benzaldehyde, hexanaldehyde, 2-ethyl-hexanaldehyde, E-pent-2-en-1-al, and cyclohexanone--all of them were found in white wine (Moscato Canelli) and red wine (Shiraz) produced in the São Francisco Valley, in the Northeastern Region of Brazil--although this optimized method may probably be suitable for quantification of propionaldehyde, isobutyraldehyde, heptanaldehyde, octanaldehyde, benzaldehyde, and E-hex-2-en-1-al as well. We could not prove if this method is also able to determine the latter CC group since we have not found these substances present in detectable levels in our real samples considered in this study.

A new analytical methodology using HS-SPME/GC-MS was optimized in order to attain maximum sensitivity, using multivariate strategies. The proposed method was employed to evaluate the VOC profile exhaled from canine hair samples collected from 8 healthy dogs and from 16 dogs infected by Leishmania infantum. 274 VOCs were detected, which could be identified as aldehydes, ketones and hydrocarbons. After application of the Soft Independent Modeling of Class Analogy (SIMCA) and Principal Component Analysis (PCA) healthy and infected dogs, with similar VOCs profiles, could be separately grouped, based on compounds such as 2-hexanone, benzaldehyde, and 2,4-nonadienal. The proposed method is non-invasive, painless, readily accepted by dog owners and could be useful to identify several biomarkers with applications in the diagnosis of diseases.

This paper describes the development and optimization, by using multivariate analysis, of a GC-MS-SIM method for evaluation of the 16 polyaromatic hydrocarbons considered as priority pollutants in atmospheric particulate material by the US EPA. In order to assure an adequate separation in the shortest analysis time, a multivariate design was used to set the conditions of the oven temperature program. The optimization process was carried out using factorial fractional design and Box-Behnken design. The following factors were evaluated: initial temperature, temperature rate #1, intermediary temperature, temperature rate #2, and final temperature. The optimized conditions were set at: 70 degrees C (2 min) --> 200 degrees C (30 degrees C/min, 5 min) --> 300 degrees C (5 degrees C/min, 1.67 min). Moreover, we have also optimized the injector temperature as 310 degrees C and sampling time as 0.8 min. The total analysis time was 33 min. Validation of GC-MS-SIM yielded satisfactory results for repetitivity of the detector response and retention times, and linearity of calibration curves. LOD were established as 0.13-0.34 ng/mL (peak area) and 0.18-0.72 ng/mL (peak height). The method has been shown to be appropriate for the analysis of samples of atmospheric particulate material and/or other environmental matrices.

A chromatographic quantification method with two different mobile phases (elution conditions 1 and 2) was developed to determine carbonyl compounds (CCs) in air samples collected from charcoal production workplaces, using C18 cartridges coated with 2,4-dinitrophenylhydrazine (DNPHi). Several 2,4-dinitrophenylhydrazones (DNPHo) were separated and quantified using an HPLC system and UV detection. In 16 min, elution condition 1 successfully separated and quantified the DNPHo of 14 CC including acetaldehyde, acrolein, formaldehyde, and furfural, and estimated the sum of C4 isomers, butanal-isobutanal-butanone. This elution condition was able to resolve the pairs acrolein/furfural and propanone/propanal, which have been cited in the literature as difficult mixtures to be separated. The elution condition 2 allowed separation and quantification, in less than 30 min, of 13 out of the 17 CC listed above. This elution condition was also able to separate propanone from propanal and butanone from the other components of the C4 mixture. When the two mobile phases were used together, they allowed confirmation of the presence of the DNPHo in the real samples. Thus, both elution conditions have been shown to be appropriate to determine CC, in personal and stationary samples, collected in charcoal production plants.

The presence of carbonyl compounds (CCs) in wines has sparked the interest of researchers in several countries. The quantification of some of these compounds has been used as a parameter of quality for many fermented beverages. Although present in minute quantities (except for acetaldehyde), they have a strong olfactory impact. In addition, the CCs found in wines have a strong affinity for bisulfite and can form stable adducts, which will also interfere in the characteristics of aroma. The greatest challenge, however, is to predict which CCs have the strongest affinity for S(IV) and what conditions favor this interaction. To better understand the reaction of CC-bisulfite adduct formation (HASA), this study has evaluated the profile of 22 CCs in a "synthetic wine" containing bisulfite and in 10 real samples of different wines from the São Francisco Valley, northeastern Brazil. On the basis of principal component analysis (PCA) and dissociation constants, the results revealed that aliphatic aldehydes form adducts with S(IV), whereas ketones, cyclic aldehydes, and trans-alkenes interact weakly and are found predominantly in their free form. These results revealed also that pH 10 and 11 were defined as the most appropriate for CC-SO 2 adduct dissociation, and the total CCs were quantified reliably.

A sensitive FIA method was developed for the selective determination of formaldehyde in alcoholic beverages. This method is based on the reaction of Fluoral-P (4-amine-3-pentene-2-one) with formaldehyde, leading to the formation of 3,5-diacetyl-1,4-dihydrolutidine (DDL), which fluoresces at lambda(ex)=410nm and lambda(em)=510nm. The analytical parameters were optimized by the response surface method using the Box-Behnken design. The proposed flow injection system allowed for the determination of up to 3.33x10(-5)molL(-1) of formaldehyde with R.S.D.<2.5% and a detection limit of 3.1ngmL(-1). The method was successfully applied to determine formaldehyde in alcoholic beverages, without requiring any sample pretreatment, and the results agreed with the reference at a 95% confidence level by paired t-test. In the optimized condition, the FIA system proved able to analyze up to 60 samples/h.

This paper describes fundamentals and applications of multivariate statistical techniques for the optimization of chromatographic systems. The surface response methodologies: central composite design, Doehlert matrix and Box-Behnken design are discussed and applications of these techniques for optimization of sample preparation steps (extractions) and determination of experimental conditions for chromatographic separations are presented. The use of mixture design for optimization of mobile phases is also related. An optimization example involving a real separation process is exhaustively described. A discussion about model validation is presented. Some applications of other multivariate techniques for optimization of chromatographic methods are also summarized.

Spectrometric techniques for the analysis of trace cadmium have developed rapidly due to the increasing need for accurate measurements at extremely low levels of this element in diverse matrices. This review covers separation and preconcentration procedures, such as electrochemical deposition, precipitation, coprecipitation, solid phase extraction, liquid-liquid extraction (LLE) and cloud point extraction (CPE), and consider the features of the their application with several spectrometric techniques.

Colorimetric methods are still important for determining nitrate and nitrite. A critical step in the use of these methods to determine nitrate in low concentrations is the reaction time required to totally reduce nitrate to nitrite, i.e., 24h in the dark. This work involved a study of the influence of ultrasonic irradiation on the nitrate reduction reaction by hydrazine. Our findings indicated that ultrasonic irradiation, associated with copper(II) ion as a catalyst, increased the redox reaction rate, decreasing the reaction time to about 10min when the power of the ultrasonic irradiation was set in 14.0357W. The strong influence of the ultrasonic irradiation in the reduction reaction rates can be sustained by an excellent linear correlation (R(2)=0.9993) between the kinetic constants and ultrasonic powers. Nitrate conversion also increased from 68% to 98% at the latter conditions. It thus become clear that high intensity ultrasound is very beneficial for this reduction reaction to proceed in good yield and in short reaction time in comparison to its silent reaction.

A new analytical method is reported for the determination of 11 volatile carbonyl compounds isolated at room temperature from the headspace of marine algae. This method is based on the conversion of the carbonyl compounds to their 2,4-dinitrophenylhydrazone derivatives followed by high-performance liquid chromatography analysis. Using this method, 11 carbonyl compounds are detected and identified from the dynamic headspace sampling of 10 species of marine algae. Eight compounds are quantitated and the three remaining are only identified. Under optimized conditions, all carbonyl compounds are separated in 32 min. The detection and quantitation limits of the high-performance liquid chromatography method are, respectively, in the range of 0.26-0.85 ng/g of algae (formaldehyde) to 13.77-45.90 ng/g of algae (E)-2-hexenal. The calibration curves are linear in the concentration range of 2.0-1000 microg/L of solution, corresponding to 0.34-170.00 ng/g of algae. Acetaldehyde and propanal are the most abundant carbonyl compounds identified, with concentrations as high as 980 and 790 ng/g, respectively. The present work, as far as we know, is the first analytical methodology that has been developed to determine low-molecular-weight carbonyl compounds in marine algae. Because many species of marine algae are used as human food, the reported method should be useful to investigators studying the nutritional value as well as oxidative spoilage of fresh and preserved marine algae that is destined for human consumption.

Brominated phenols 2- and 4-bromophenol (2-BP and 4-BP); 2,4- and 2,6-dibromophenol (2,4-DBP and 2,6-DBP) and 2,4,6-tribromophenol (2,4,6-TBP) have been identified as key flavor compounds found in seafoods. Depending on their concentrations, they were responsible for marine or ocean flavor (shrimp/crab/fish/sea salt-like) or for phenolic/iodine/iodoform-like off-flavor. In this work a new analytical methodology was developed to determine, simultaneously, such bromophenols in fish meats, based on reversed-phased high-performance liquid chromatographic separation (RP-HPLC). The separation of bromophenols was made onto a Lichrospher 100 RP-18 column using water:acetonitrile gradient at a flow rate of 1.0mLmin(-1), using absorbance detection at 286nm, were the 2-BP, 4-BP, 2,4- and 2,6-DBP show significant absorbtivity values and at 297nm for 2,4,6-TBP. They were separated in 20min with a good chromatographic resolution (Rs) for the isomeric compounds: 2- and 4-BP, Rs=1.23; 2,4- and 2,6-DBP, Rs=1.63. The calibration curves were linear in the bromophenols concentration range of 200.0-1000ngmL(-1). Under optimized conditions, the detection limit of the HPLC method was 127ngmL(-1) for 2-BP; 179ngmL(-1) for 4-BP; 89.0ngmL(-1) for 2,4-DBP; 269ngmL(-1) for 2,6-DBP and 232ngmL(-1) for 2,4,6-TBP. This method has been applied in determination of bromophenols, isolated by combined steam distillation-solvent extraction with 2mL of pentane/diethyl ether (6:4), from Brazilian fishes samples, collected on the Atlantic coast of Bahia (13 degrees 01'S and 38 degrees 31'W), Brazil. The concentration range determined were 0.20ngg(-1) (2-BP) to 299ngg(-1) (2,4,6-TBP). The method proposed here is rapid and suitable for simultaneous quantification of simple bromophenols in fish meat. As long as we know, it is the first analytical methodology, using RP-HPLC/UV, which was developed to determine simple bromophenols in fish meat.

Caffeine (1,3,7-trimethylxanthine), theobromine (3,7-dimethylxanthine) and theophylline (1,3-dimethylxanthine) are the most important naturally occurring methylxanthines. Caffeine is a constituent of coffee and other beverage and included in many medicines. Theobromine and theophylline are formed as metabolites of caffeine in humans, and are also present in tea, cocoa and chocolate products. In order to improve the chromatographic resolution (R(s)) with a good analysis time, experimental designs were applied for multivariate optimisation of the experimental conditions of an isocratic reversed-phase high-performance liquid chromatographic (RP-HPLC) method used for the simultaneous determination of caffeine, theobromine and theophylline. The optimisation process was carried out in two steps using full three-level factorial designs. The factors optimised were: flow rate and mobile phase composition. Optimal conditions for the separation of the three methylxanthines were obtained using a mixture of water/ethanol/acetic acid (75:24:1%, v/v/v) as mobile phase and a flow rate of 1.0mLmin(-1). The RP-HPLC/UV method was validated in terms of limit of detection (LOD), limit of quantitation (LOQ), linearity, recovery and the precision, calculated as relative standard deviation (R.S.D.). In these conditions, the LOD was 0.10mugL(-1) for caffeine, 0.07mugL(-1) for theobromine and 0.06mugL(-1) for theophylline. The proposed method is fast, requires no extraction step or derivatization and was suitable for quantification of these methylxanthines in coffee, tea and human urine samples.

In this work, a solid phase spectrophotometric method in association with flow injection analysis for formaldehyde determination has been developed with direct measurement of light-absorption in C(18) material. The 3,5-diacetyl-1,4-dihydrolutidine produced from the reaction between formaldehyde and fluoral P was quantitatively retained on C(18) support and the spectrophotometric detection was performed simultaneously at 412nm. The retained complex was quickly eluted from C(18) material with the eluent stream consisting of a 50% (v/v) ethanol solution. The results showed that the proposed method is simple, rapid and the analytical response is linear in the concentration range of 0.050-1.5mgL(-1). The limit of detection was estimated as 30mugL(-1) and the R.S.D. 2.2% using a sample volume of 625muL. The system presented an analytical throughput of 20 determinations per hour. The method was successfully applied in the determination of formaldehyde in ethanol fuel.

In this study we investigated the larval dispersal associated with larval predation in experimental populations of Chrysomya albiceps and Cochliomyia macellaria. Frequency distribution of sampling units (G test) in the substrate was used to evaluate variation in larval dispersal. An experimental acrylic channel (1 x 0.1 x 0.2 m) covered with wood shavings was used to observe larval dispersal prior to pupation. The acrylic channel was graduated at 0.05 m intervals, each representing a sampling unit; hence, 20 sampling units were set up. A Petri dish containing third instar larvae of single and double species was deposited at one edge of the acrylic channel allowing larvae to disperse. The number of buried pupae (0, 1, 2, n) present in each sampling unit was recorded. For double species, the number of recovered larvae of C. albiceps was similar to the number initially released on the dish Petri. On the other hand, the number of recovered larvae of C. macellaria was significantly smaller than the initially released number. The results show that C. albiceps attacks C. macellaria larvae during the larval dispersal process. The larval distribution of C. albiceps did not differ significantly from C. macellaria in double species, but it differed significantly in single species. The larval aggregation level of C. macellaria decreased when C. albiceps was present and the larval aggregation level of C. albiceps increased when C. macellaria was present. The implications of such findings for the population dynamics of these species are discussed.

Motor vehicles constitute a significant source of polycyclic aromatic hydrocarbon (PAH) emissions to the atmosphere. Particle-phase priority pollutant PAH concentrations and total suspended particle mass (TSP) were measured in the Lapa bus station and the Americo Simas Tunnel, located in the city of Salvador, Brazil. Separate samples were collected at the bus station at different times of the day, including rush- and non-rush-hour periods. The highest concentrations for nearly all 16 priority PAHs measured at the bus station were observed at 18:30 h, with chrysene showing the highest mean value (26.6 ng m-3). The highest average PAH concentrations measured in the tunnel were observed for pyrene (79.4 +/- 11.5 ng m-3) followed by fluoranthene (39.0 +/- 5.2 ng m-3) and chrysene (28.0 +/- 4.17 ng m-3). TSP levels reached 423 micrograms m-3 in the bus station, and values as high as 2 mg m-3 in the tunnel. The measured Salvador tunnel PAH profiles are very similar to the Salvador bus station profiles, and are similar to PAH profiles reported for the Kojouike Tunnel, located in Kurashiki City, Japan, and the Caldecott Tunnel, located in Berkeley, California.

This work relates the development of an analytical methodology to simultaneously determine three methylxanthines (caffeine, theobromine, and theophylline) in beverages and urine samples based on reversed-phase high-performance liquid chromatography. Separation is made with a Bondesil C18 column using methanol-water-acetic acid or ethanol-water-acetic acid (20:75:5, v/v/v) as the mobile phase at 0.7 mL/min. Identification is made by absorbance detection at 273 nm. Under optimized conditions, the detection limit of the HPLC method is 0.1 pg/mL for all three methylxanthines. This method is applied to urine and to 25 different beverage samples, which included coffee, tea, chocolate, and coconut water. The concentration ranges determined in the beverages and urine are: < 0.1 pg/mL to 350 microg/mL and 3.21 microg/mL to 71.2 microg/mL for caffeine; < 0.1 pg/mL to 32 microg mL and < 0.1 pg/mL to 13.2 microg/mL for theobromine; < 0.1 pg/mL to 47 microg/mL and < 0.1 pg/mL to 66.3 microg/mL for theophylline. The method proposed in this study is rapid and suitable for the simultaneous quantitation of methylxanthines in beverages and human urine samples and requires no extraction step or derivatization.

A new analytical method to determine trace volatile aldehydes isolated from the headspace of fish meat at room temperature by high-performance liquid chromatography (HPLC) in the form of 2,4-dinitrophenylhydrazone (DNPHo) derivatives has been developed. Aliquots (50 g) of the fish purée were introduced into a 500-mL glass recipient and were purged with N2 for 40 min through two SEP-PAK C18 cartridges (connected in series) coated with an acid solution of 2,4-dinitrophenylhydrazine. The cartridges were then eluted with acetonitrile (2 mL) and the 2,4-DNPHo formed was quantitated by HPLC-UV analysis using a Zorbax C18 column. The isolated compounds from the dynamic headspace sampling of four kinds of fish species were saturated aldehydes, formaldehyde, acetaldehyde, propanal, butanal, pentanal, and hexanal. Under optimized conditions the detection limits of the HPLC method were in the range of 0.75 nmol/g (formaldehyde) to 2.19 nmol/g (hexanal). The calibration curves were linear in the concentration range from 1.3 nmol/mL to 12.5 nmol/mL. Propanal and acetaldehyde were the major carbonyl compounds identified (ranging from 3.9 nmol/g and 10 nmol/g). This study has revealed the widespread occurrence of formaldehyde, acetaldehyde, propanal, butanal, pentanal, and hexanal in fish meat.

A system, based on annular gold-coated denuders, was optimized and used in the atmospheric sampling of mercury in urban and rural sites of the Alta Floresta, a region in the Brazilian Amazon. Results showed that vapour phase mercury (Hgv) is dominant in samples collected in both sites. However, particulate mercury (Hg) may sometimes be significant at the urban site, with values representing up to 77% of the total Hg collected during sampling, but may also be insignificant at the same site, depending on the gold commercialization activity and, probably, on the meteorological conditions during the sampling period. The presence of mercury in both the vapour and particulate phases in the atmosphere of the urban site can explain the high mercury concentrations found in urban dust reported for some cities in the Amazon, and also the relatively high Hg concentrations found in soils adjacent to the urban centre of Alta Floresta. It appears that the urban dust of "garimpo" areas in the Amazon can act as an efficient adsorber of Hgv emitted by gold dealer operations during the purification process. Mercury emissions from gold dealer shops in the Amazon have been considered as a human health risk through the inhalation of Hg vapour. However, although true for indoor sites, the significant contribution of particulate phase Hg shown in this study in outdoor urban sites calls for a re-evaluation of the risk assessment estimates performed up to now in urban centres in the Amazon.

A new sampling protocol was developed to determine methanol and ethanol in the gas phase, at low concentration levels, in urban atmospheres. The procedure involves collection of air samples (20.0-30.0 l) with three florisil cartridges connected in series, at a flow rate ranging from 1.0 to 2.0 l min(-1) and subsequent elution of the alcohols with water. Separation and quantification were done by gas chromatography (GC) coupled with a flame ionization detector, 'SPI' injector and column DB WAX (30 mx0.53 mmx1 mum). The minimum mass detected by the method, based on two times the average background mass on the blank cartridges, was 0.3 mug for both alcohols which, for a sampled volume of 30 l, resulted in detection limits of 7.6 and 5.3 ppbV for methanol and ethanol, respectively. The determined alcohol concentrations, in 42 different samples from the three largest cities in Brazil-São Paulo, Rio de Janeiro and Salvador-ranged from 72 ppbV to below the detection limit for methanol and from 355 to 12 ppbV for ethanol.

The use of ICP/AES for the determination of zinc, in low concentration levels, in matrices containing high levels of copper is difficult because copper interferes in the zinc main emission wavelength (213.856 nm). In the present work, a separation of zinc from copper matrices was possible, using the reaction of zinc(II) cation with 1-(2-tiazolylazo)-2-naphthol (TAN), in the pH range of 6.5-8.0, resulting in a stable red complex. Copper also reacts with TAN but its interference was avoided by the addition of ascorbic acid and thiosulphate in the reaction medium. In this way, the aqueous solution was passed through a SEP PAK C18 cartridge, in which the zinc(II)-TAN complex was quantitatively retained, but it did not occur with copper which passes through the cartridge, as [Cu(2)(S(2)O(3))(2)](2-), with the aqueous solution. The cartridge was washed with water and the complex eluted with ethanol. Then, the alcohol was evaporated and the complex decomposed by nitric acid. It results in both zinc pre-concentration and separation from copper. The zinc quantification was carried out by ICP/AES at 213.856 nm. The relative standard deviations, for ten different aliquots, were 5.7% and the average recovery found for zinc was 96%, even when the concentration ratio Cu/Zn was up to 500/1 (mg l(-1):mg l(-1)). Other metals, like nickel, for example, can react with TAN in the same way as zinc but they do not interfere in the emission wavelength 213.856 nm.

The use of 85% phosphoric acid in borosilicate conical flasks for the dissolution of ilmenites at 230 +/- 10 degrees C is reported. The samples were quantitatively dissolved in less than 13 min. Titanium was determined by both spectrophotometry and inductively coupled plasma atomic emission spectrometry ICP-AES. Vanadium and iron were determined by ICP-AES. In several samples of ilmenites analyzed, the TiO(2) concentration was in the range 10.6-57.5% and those of FeO and V(2)O(5) were in the ranges 31.6-51.4% and 0.39-1.32%, respectively. In the spectrophotometric method, vanadium interference occurs only when the Ti V concentration ratio is <4. In all samples analyzed this ratio was around 12, resulting in no interferences due to vanadium. Hence the ilmenite dissolution procedure using phosphoric acid was compatible with titanium quantification by both spectrophotometry and ICP-AES.

The reaction of aluminium(III) with Hydroxynaphtol Blue (HNB) in aqueous media at apparent pH 5.5 results in a red complex that is stable for at least 4 hr. Beer's Law is obeyed up to 1.6 microg/ml of aluminium(III) with an apparent molar absorptivity of 1.66 x 10(4) l.mol(-1). cm(-1) at 569 nm. This paper proposes procedures for aluminium(III) determination by ordinary and first-derivative spectrophotometry. The results demonstrated that the linear dynamic range is 0.03-1.60 microg/ml for ordinary spectrophotometry and 11.8-320.0 ng/ml for first derivative spectrophotometry. The HNB is not selectivity for aluminium, but the addition of EDTA allows the aluminium determination in the presence of accepted amounts of Ca(II), Mg(II), Mn(II), Ba(II), Sr(II), Cd(II), Pb(II), La(III), In(III), Bi(III) and Zn(II). The interference of Cu(II) and Hg(II) can be masked by thiosulphate. Ions such as UO(2)(II), Mo(VI), Co(II), Ti(IV) and PO(4)(III) do interfere seriously. This method was applied for aluminium determination in copper-base alloy, zinc-base alloy, magnesium-base alloy, iron ore, manganese ore, cement, dolomite, feldspar and limestone. The results indicated high accuracy and precision.

Nine polycyclic aromatic hydrocarbons (PAHs) contained in air samples collected on quartz fiber filters inside an urban tunnel and in a nearby mixed commercial residential area in the city of Rio de Janeiro, Brazil, were exposed to scrubbed air (to measure desorption loss) and to particle-free ambient air (to measure chemical reaction losses in the absence of desorption). The exposures were conducted for 5.5 to 9 hour periods at ambient temperature (22-26 degrees C) at face velocities typical of high volume sampling. Under prevailing atmospheric conditions all nine PAHs experienced filter losses which (for most of them) followed first order kinetics. For the ambient samples, in a 6 hour exposure period, the following five PAHs showed filter losses (% in parantheses) attributed exclusively to chemical reaction: benzo(b)fluoranthene (43), benzo(k)fluoranthene (39), benzo(a)pyrene (70), benzo(ghi)perylene (44), and indeno (1,2,3-cd)pyrene (41). The other four showed the following unassigned losses: pyrene (100), fluoranthene (65), crysene (72), and benzo(a)anthracene (71). The results are discussed in the light of possible filter artifacts in PAH sampling and the use of PAH profile signatures for source identification of atmospheric particulate matter in receptor modeling.
