Exploring drug-target interactions has always been an important step in drug development. However, exploring drug-target interaction is time-consuming and laborious. A large number of studies try to use artificial intelligence methods to predict possible drug-target interactions to reduce the workload of the wet-lab identification experiments. However, the accuracy of existing methods is still limited. This paper proposes an ensemble learning-based drug-target interaction prediction method (ELDTIP in short). First, the multiple similarity matrices of drugs or proteins are integrated by singular value decomposition (SVD) to obtain their low-dimensional feature vectors. After that, by concatenating the low-dimensional feature vectors of specific drugs and targets, the feature vector of a drug-target pair are obtained. An ensemble learning model based on gradient boosting decision tree (GBDT) was constructed to predict whether this pair of drug-target can interact with each other. The main contributions of ELDTIP are as follows: (1): ELDTIP uses SVD to integrate multiple similarity matrices, which can retain more valuable information of the original feature. (2): ELDTIP uses the ensemble learning-based model, GBDT, which can make full use of the unknown DTIs in the dataset and mitigate the influence of class imbalance. Experimental results show that the performance of ELDTIP is higher than that of several state-of-the-art DTI prediction methods.

Cisplatin nephrotoxicity is an etiological factor for acute kidney injury (AKI). MicroRNA (miRNA) expression is dysregulated in cisplatin‐induced AKI (cAKI) although the underlying mechanisms are unclear. A cAKI model was established by intraperitoneally injecting cisplatin, and key miRNAs were screened using high‐throughput miRNA sequencing. The functions of key miRNAs were determined using the cell viability, live/dead, reactive oxygen species (ROS), and 5‐ethynyl‐2′‐deoxyuridine (EdU) proliferation assays. Additionally, the macrophage membrane was wrapped around a metal–organic framework (MOF) loaded with miRNA agomir to develop a novel composite material, macrophage/MOF/miRNA agomir nanoparticles (MMA NPs). High‐throughput miRNA sequencing revealed that miR‐30e‐5p is a key miRNA that is downregulated in cAKI. The results of in vitro experiments demonstrated that miR‐30e‐5p overexpression partially suppressed the cisplatin‐induced or lipopolysaccharide (LPS)‐induced downregulation of cell viability, proliferation, upregulation of ROS production, and cell death. Meanwhile, the results of in vivo and in vitro experiments demonstrated that MMA NPs alleviated cAKI by exerting anti‐inflammatory effects. Mechanistically, cisplatin downregulates the expression of miR‐30e‐5p, and the downregulated miR‐30e‐5p can target Galnt3 to activate the adenosine 5'‐monophosphate activated protein kinase (AMPK) signaling pathway, which promotes the progression of AKI. Our study found that miR‐30e‐5p is a key downregulated miRNA in cAKI. The downregulated miR‐30e‐5p promotes AKI progression by targeting Galnt3 to activate the AMPK signaling pathway. Background/Aims: Anti-oxidation is an effective strategy for curing acute kidney injury (AKI). Herein, we suggest that extracellular vesicles (EVs) derived from mesenchymal stromal cells (MSCs) might play an anti-oxidative role by enhancing Nrf2/ARE activation in AKI. Methods: EVs isolated from the conditioned medium of human Wharton's Jelly mesenchymal stromal cells and human foreskin fibroblast were intravenously injected in rats immediately after 45 min of unilateral kidney ischemia. Animals were sacrificed 24 h after injury. Results: Results showed that renal tubular injury was alleviated and renal function was improved by MSC-EVs. Cell apoptosis and sNGAL levels, which reflect kidney cell injury, were reduced. Moreover, MSC-EVs decreased oxidative stress in injured kidney tissues and NRK-52E cells under hypoxia injury. Nrf2/antioxidant response element (ARE) enhancement and HO-1 up-regulation were further observed after MSC-EV treatment both in vivo and in vitro. Conclusions: MSC-EVs may protect against AKI possibly through anti-oxidation by enhancing Nrf2/ARE activation. [ABSTRACT FROM AUTHOR]

The modeling of single core cable has an important influence on the calculation of cable sheath cross-connected grounding current and voltage. A suitable equivalent circuit can greatly reduce the complexity of the model. In this paper, the theoretical calculation of π type equivalent circuit of cable is analyzed, and the equivalent circuit of cable sheath grounding system is analyzed. Then the calculation equation of sheath circulation and induced voltage is obtained. Finally, by simulating two kinds of sheath grounding faults, two kinds of sheath grounding fault location are summarized.

The Tb3+‐doped transparent glass ceramics containing Al4Ti2SiO12 crystal are synthesized by the melt‐solid‐crystallization method. The optimal heat treatment conditions for the glass ceramic samples are determined to be 750°C, 1.5 h based on differential scanning calorimeter, X‐ray diffractometer, scanning electron microscope, and light transmittance curve. The luminescence properties of the glass ceramics samples are characterized by a fluorescence spectrometer. The emission spectrum revealed that the dominant emission is green attributed to Tb3+, the optimal doping concentration is 0.3%. And the thermal quenching activation energy of 0.3% Tb3+‐doped glass ceramics is 0.2554 eV, while the relative sensitivity is 1.50% K−1 at 298 K and the absolute sensitivity is 2.27×10−2 K−1 at 418 K through variable temperature fluorescence spectroscopy test and a series of calculations. These findings demonstrate the excellent stability and temperature sensitivity of glass ceramics, thereby highlighting their potential for use in green lighting and temperature sensing applications. Tb3+-doped Al2O3–B2O3–SrO glass was prepared by the melt annealing method, and the crystallization kinetics of the glass was studied by the thermal analysis method. The crystallization activation energy of the glass was calculated by the Kissinger method and the Ozawa method, the values of which were 271.83 ± 8.56 kJ mol−1 and 260.72 ± 8.20 kJ mol−1, respectively. Optical band gap and Urbach energy of Tb3+-doped glass and glass ceramic were calculated by absorption spectra. The results indicate that after heat treatment, the optical band gap of the glass ceramic is reduced compared to that of the glass. The emission intensity of Tb3+ in the glass ceramic is enhanced compared with that in the glass. Variable-temperature emission spectra show abnormal thermal quenching of Tb3+-doped glass and glass ceramic. The results show that Al2O3–B2O3–SrO glass ceramics doped with Tb3+ have high luminescence intensity and can be applied to green light emitting devices and display devices. [ABSTRACT FROM AUTHOR]
