This study evaluated the effects of feeding ground (G) versus pelleted (P) total mixed ration (PTMR) on digestibility and milk production performance of mid‐lactation dairy cows over a 42‐days period. Results showed that, compared to GTMR, feeding PTMR increased dry matter intake and digestibility, ruminal propionate proportions and milk protein percentage, but decreased acetate‐to‐propionate ratio, milk fat percentage and aldehydes, acids, alcohols, and aromatic hydrocarbon proportions in milk. The PTMR did not improve milk yield but accelerated live weight recovery of cows and led to acidosis risk in lactating cows due to negative effects on rumen fermentation. [ABSTRACT FROM AUTHOR]

FecB (also known as BMPR1B) is a crucial gene in sheep reproduction, which has a mutation (A746G) that was found to increase the ovulation rate and litter size. The FecB mutation is associated with reproductive endocrinology, such mutation can control external estrous characteristics and affect follicle-stimulating hormone during the estrous cycle. Previous researches showed that the FecB mutation can regulate the transcriptomic profiles in the reproductive-related tissues including hypothalamus, pituitary, and ovary during the estrous cycle of small-tailed Han (STH) sheep. However, little research has been reported on the correlation between FecB mutation and the estrous cycle in STH sheep oviduct. To investigate the coding and noncoding transcriptomic profiles involved in the estrous cycle and FecB in the sheep oviduct, RNA sequencing was performed to analyze the transcriptomic profiles of mRNAs and long noncoding RNAs (lncRNAs) in the oviduct during the estrous cycle of STH sheep with mutant (FecB BB) and wild-type (FecB ++) genotypes. In total, 21,863 lncRNAs and 43,674 mRNAs were screened, the results showed that mRNAs had significantly higher expression levels than the lncRNAs, and the expression levels of these screened transcripts were lower in the follicular phase than they were in the luteal phase. Among them, the oviductal glycoprotein gene (OVGP1) had the highest expression level. In the comparison between the follicular and luteal phases, 57 differentially expressed (DE) lncRNAs and 637 DE mRNAs were detected, including FSTL5 mRNA and LNC_016628 lncRNA. In the comparison between the FecB BB and FecB ++ genotypes, 26 DE lncRNAs and 421 DE mRNAs were detected, including EEF1D mRNA and LNC_006270 lncRNA. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes functional enrichment analysis indicated that the DE mRNAs were enriched mainly in terms related to reproduction such as the tight junction, SAGA complex, ATP-binding cassette, nestin, and Hippo signaling pathway. The interaction network between DE lncRNAs and DE mRNAs indicated that LNC_018420 may be the key regulator in sheep oviduct. Microwave (MW) treatment has been suggested to improve forage hays' nutritive value and sheep growth performance. However, the extent to which the MW-treated forage hay affects apparent digestibility and nitrogen partitioning is not documented. This study examined the potential to use the MW as a novel thermal treatment to improve forage hay intake, digestibility, and nitrogen utilisation of sheep. A total of 24 merino rams (18 months of age, 42 ± 2.0 kg initial body weight) were randomly divided into four dietary treatments: Control lucerne hay (CLH), MW-treated lucerne hay (MLH), Control wheat hay (CWH), and MW-treated wheat hay (MWH). The study had 14 days adaption and 7 7-day sample collection period, with 6 sheep replicates/treatment. Feed, faeces, urine, and blood plasma samples were collected. Higher dry matter (DM) intake was observed in MLH vs. CLH (P < 0.001). The DM and organic matter digestibility both increased by 8 % in MLH compared to CLH (P <0.001). The digestibility of neutral detergent fibre (NDF), acid detergent fibre (ADF), and nitrogen increased by 12 %, 9 %, and 10 %, respectively in MLH compared to CLH (P < 0.001). The MLH-feed sheep showed higher nitrogen retention (P = 0.037) and microbial nitrogen synthesis (P = 0.047) compared to CLH-fed sheep. A limited effect was observed in CWH vs. MWH, with only ADF digestibility (P = 0.023) and plasma glucose (P < 0.05) being higher in sheep fed with MWH. Electron microscope images showed clear cell microstructure destruction (P <0.001) due to MW treatment in lucerne hay but there was no difference observed in wheat hay. The result provided first-hand comprehensive data to quantify MW treatment's effect on hay quality and sheep performance, with potential mechanisms explained underpinning the observed changes. • Microwave (MW) heat treatment led to micro-structural change in lucerne forage hay. • Intake and nutrient digestibility increased in sheep fed MW-treated lucerne hay. • The microbial nitrogen synthesis was higher when sheep-fed MW-treated lucerne hay.
