Objective: To investigate the expression level of hsa&lowbar;circ&lowbar;0140180 in esophageal squamous cell carcinoma (ESCC) cells, as well as its effect on malignant biological behavior of ESCC cells and the possible molecular mechanisms. Methods: Six pairs of ESCC tissues and corresponding paracarcinoma tissues resected at the Department of Thoracic Surgery, Central Hospital of Nanchong City between November 2018 and March 2019 were collected for whole-transcriptome sequencing, and hsa&lowbar;circ&lowbar;0140180 at a low expression in ESCC tissues was screened out. The TE-1 and KYSE30 cell lines overexpressing hsa&lowbar;circ&lowbar;0140180 were established. qPCR was used to detect the expression of hsa&lowbar;circ&lowbar;0140180 in human normal esophageal epithelial cells and ESCC cells, as well as the expression of miR-1287-5p in TE-1 and KYSE30 cells overexpressing hsa&lowbar;circ&lowbar;0140180. CCK-8 and FCM were used to detect the effect of hsa&lowbar;circ&lowbar;0140180 overexpression on the proliferation and cell cycle of TE-1 and KYSE30 cells. Scratch assay and Transwell assay were used to assess the effects of hsa&lowbar;circ&lowbar;0140180 overexpression on migration and invasion of TE-1 and KYSE30 cells. Dual-luciferase reporter assay was used to confirm the targeting relationship between hsa&lowbar;circ&lowbar;0140180 and miR-1287-5p. WB assay was used to detect the expression of EMT-related proteins and the phosphorylation level of the PI3K-Akt pathway in TE-1 and KYSE30 cells after hsa&lowbar;circ&lowbar;0140180 overexpression. Results: Transcriptome sequencing and qPCR results showed that hsa&lowbar;circ&lowbar;0140180 was lowly expressed in ESCC tissues and cells (P<0.05 or P<0.01) and confirmed its closed-loop molecular signature and localization in the cytoplasm. Overexpression of hsa&lowbar;circ&lowbar;0140180 could significantly inhibit the migration and invasion of ESCC cells (all P<0.05) but had no significant effect on the proliferation and cell cycle. The results of dual-luciferase reporter gene assay demonstrated that hsa&lowbar;circ&lowbar;0140180 targets miR-1287-5p and negatively regulates its expression (P<0.001). Overexpression of hsa&lowbar;circ&lowbar;0140180 could significantly increase the expression of E-cadherin and decrease the expression of Snail (both P<0.05) and the phosphorylation level of the PI3K-Akt pathway (P<0.01, P<0.001) in TE-1 and KYSE30 cells. Conclusion: hsa&lowbar;circ&lowbar;0140180 is lowly expressed in ESCC cells and tissues; it reduces the phosphorylation levels of PI3K-Akt pathway and suppresses EMT process possibly through regulation of miR-1287-5p expression, thus affecting the migration and invasion abilities of ESCC cells. Objective: To investigate the effects of basonuclin 1 (BNC1) on the proliferation, migration, invasion, cell cycle and apoptosis of esophageal squamous cell carcinoma (ESCC) cells and its mechanism. Methods: TThe mRNA expression level of BNC1 in ESCC cells and normal esophageal epithelial cells was detected by qPCR. The protein expression level of BNC1 in cancer and para-cancerous tissues of 10 ESCC patients was detected by immunohistochemical staining. The expression of BNC1 in KYSE-150 and KYSE-30 cells was down-regulated by siRNA, and the effects of BNC1 on cell proliferation, migration, invasion, cell cycle and apoptosis were investigated using CCK-8, scratch healing, Transwell and flow cytometry assays, respectively. The downstream target genes of BNC1 were identified by ChIP-seq assay and GEPIA online website date analysis combined with knockdown transcriptome sequencing data after BNC1 knockdown. qPCR was used to verify the expression of target genes after BNC1 knockdown, and dual luciferase reporter gene assay was used to confirm the regulatory effect of BNC1 on the target genes. Results:The mRNA and protein levels of BNC1 were higher in ESCC tissues than in para-cancerous tissues (all P<0.01). Knockdown of BNC1 significantly inhibited the proliferation, migration and invasion of KYSE-150 and KYSE-30 cells (P<0.01 or P<0.01), arrested the cells in G1 phase and promoted the cell apoptosis (all P<0.01). The results of ChIP-seq assay and online website GEPIA date analysis combined with at transcriptome sequencing data indicated after BNC1 knockdown that G protein pathway repressor 1 (GPS1) may be a oncogenic target gene that positively regulated by BNC1. The results of qPCR and dual luciferase reporter gene assay showed that BNC1 had a regulatory effect on GPS1 (P<0.01). Conclusion: BNC1 is highly expressed in ESCC tissues and cells. Interfering with BNC1 significantly inhibits the proliferation, migration and invasion of ESCC cells, arrests cells at G1 phase and promotes cell apoptosis possibly by targeting GPS1 to regulate the malignant biological behaviors of ESCC cells. Polyploidization plays a critical role in producing new gene functions and promoting species evolution. Effective identification of polyploid types can be helpful in exploring the evolutionary mechanism. However, current methods for detecting polyploid types have some major limitations, such as being time-consuming and strong subjectivity, etc. In order to objectively and scientifically recognize collinearity fragments and polyploid types, we developed PolyReco method, which can automatically label collinear regions and recognize polyploidy events based on the K S dotplot. Combining with whole-genome collinearity analysis, PolyReco uses DBSCAN clustering method to cluster K S dots. According to the distance information in the x -axis and y -axis directions between the categories, the clustering results are merged based on certain rules to obtain the collinear regions, automatically recognize and label collinear fragments. According to the information of the labeled collinear regions on the y -axis, the polyploidization recognition algorithm is used to exhaustively combine and obtain the genetic collinearity evaluation index of each combination, and then draw the genetic collinearity evaluation index graph. Based on the inflection point on the graph, polyploid types and related chromosomes with polyploidy signal can be detected. The validation experiments showed that the conclusions of PolyReco were consistent with the previous study, which verified the effectiveness of this method. Gastric cancer (GC) ranks fifth in cancer incidence and fourth in cancer-related mortality worldwide. Reactive oxygen species (ROS) are highly oxidative oxygen-derived products that have crucial roles in cell signaling regulation and maintaining internal balance. ROS are closely associated with the occurrence, development, and treatment of GC. This review summarizes recent findings on the sources of ROS and the bidirectional regulatory effects on GC and discusses various treatment modalities for GC that are related to ROS induction. In addition, the regulation of ROS by natural small molecule compounds with the highest potential for development and applications in anti-GC research is summarized. Introduction: The postoperative recurrence of gastric cancer has a significant impact on the overall prognosis of patients. Therefore, accurately predicting the postoperative recurrence of gastric cancer is crucial., Methods: This retrospective study gathered data from 2,813 gastric cancer patients who underwent radical surgery between 2011 and 2017 at two medical centers. Follow-up was extended until May 2023, and cases were categorized as recurrent or non-recurrent based on postoperative outcomes. Clinical pathological information and imaging data were collected for all patients. A new deep learning signature (DLS) was generated using pretreatment CT images, based on a pre-trained baseline (a customized Resnet50), for predicting postoperative recurrence. The deep learning fusion signature (DLFS) was created by combining the score of DLS with the weighted values of identified clinical features. The predictive performance of the model was evaluated based on discrimination, calibration, and clinical usefulness. Survival curves were plotted to investigate the differences between DLFS and prognosis., Results: In this study, 2813 patients with gastric cancer (GC) were recruited and allocated into training, internal validation, and external validation cohorts. The DLFS was developed and assessed for its capability in predicting the risk of postoperative recurrence. The DLFS exhibited excellent performance with AUCs of 0.833 (95% CI, 0.809-0.858) in the training set, 0.831 (95% CI, 0.792-0.871) in the internal validation set, and 0.859 (95% CI, 0.806-0.912) in the external validation set, along with satisfactory calibration across all cohorts (P>0.05). Furthermore, the DLFS model significantly outperformed both the clinical model and DLS (P<0.05). High-risk recurrent patients exhibit a significantly poorer prognosis compared to low-risk recurrent patients (P<0.05)., Conclusions: The integrated model developed in this study, focusing on GC patients undergoing radical surgery, accurately identifies cases at high risk of postoperative recurrence and highlights the potential of DLFS as a prognostic factor for GC patients., (Copyright © 2024 The Author(s). The selective semihydrogenation of C 2 H 2 to C 2 H 4 in crude C 2 H 4 (with ~1 vol % C 2 H 2 contamination) is a crucial process in the manufacture of polyethylene. Comparing to conventional thermalcatalytic route with Pd as catalyst under high temperature with H 2 as hydrogen source, photocatalytic C 2 H 2 reduction reaction with H 2 O as hydrogen source can achieve high selectivity under milder conditions, but has rarely been reported. Here, we present a kind of ultrathin metal-organic framework nanosheets (Cu-Co-MNSs) that demonstrate excellent catalytic activities in the semihydrogenation of C 2 H 2 . Employing Ru(bpy) 3 2+ as the photosensitizer, this catalyst attains a noteworthy turnover number (TON) of 2124 for C 2 H 4 , coupled with an impressive selectivity of 99.5 % after 12 h visible light irradiation. This performance is comparable to molecular catalysts and notably surpasses the efficiency of bulk metal-organic framework materials. Furthermore, Cu-Co-MNSs achieve a 99.95 % conversion of C 2 H 2 under industrial relevant conditions (1.10 % C 2 H 2 in C 2 H 4 ) with 90.3 % selectivity for C 2 H 4 over C 2 H 6 , demonstrating a great potential for polymer-grade C 2 H 4 production., (© 2023 Wiley-VCH GmbH.)

High cost of microalgal biofuel is caused by all the steps in current technology, including cultivation, harvesting, lipid extraction, biofuel processing and wastewater and waste treatment. This study aims to systematically reduce these costs with one integrated process, in which carbonate is used for cell rupture, lipid extraction and biodiesel processing, and then recycled for CO 2 absorption and carbon supply for a new round of algae cultivation. To reach this goal, carbonate-heating treatment with N, N' - dibutylurea which can enhance cell disruption were used for cell-wall breaking of wet Neochloris oleoabundans (UTEX 1185) biomass. Lipid extraction was fulfilled with carbonate/ethanol aqueous two phase extraction method and residual carbonate with wastewater from bottom phase was recycled to absorb CO 2 to generate bicarbonate for algal cultivation with fresh medium. Taking into comprehensive consideration of cell disruption efficiency, partition coefficient, and lipid recovery, the condition of cell disruption and lipid extraction was set at 90 °C, 100 min reaction time, 1:7.5 DBU:H 2 O (w/w) ratio, 1:3 Na 2 CO 3 :H 2 O (w/w) ratio, and 9% (w/w T ) ethanol concentration. The results showed that carbonate-heating treatment of wet N. oleoabundans biomass resulted in up to 90.7% cell disruption efficiency. The lipid recovery rate in carbonate/ethanol system was up to 97.9%, and the final biodiesel production was 1.30 times of that with Soxhlet method. Utilization of the waste broth after CO 2 absorption with the content of 4% (v/v T ) in the medium for new batch of algae cultivation resulted in biomass concentration of 1.68 g/L. The corresponding total fatty acids production was 0.35 g/L, which was 1.63 fold of that with fresh medium.
