51. Urine-derived stem cells for the therapy of diabetic nephropathy mouse model.

Abstract

OBJECTIVE: Diabetic nephropathy (DN) is one of the most representative diabetic microangiopathy complications. So far, there have been no satisfactory therapeutic strategies, and the injection of stem cells provides a target for DN therapy. PATIENTS AND METHODS: Urine-derived stem cells (USCs) were obtained from 9 healthy men. 24 mice were randomly and equally divided into control group, DN model group, DN+hUSC group (treated with USCs for 3 times). Hematoxylin-eosin (HE) and Masson staining were used to detect histological changes of kidney injury. Creatinine and blood urea nitrogen (BUN) were measured to assess renal function. Besides, myofibroblast accumulation, macrophage infiltration, cell proliferation, and oxidative stress were detected by immunohistochemical analysis. RESULTS: Compared with DN model group, DN+hUSC group showed lower function loss, cell infiltration, and oxidative stress, as well as less renal fibrosis, histological damage, and cell proliferation. CONCLUSIONS: USC can alleviate inflammation and oxidative stress, reduce renal interstitial fibrosis, improve renal tissue structure and protect renal function through paracrine effect. Abstract

Powdery mildew, caused by Blumeria graminis f. sp. tritici, is one of the most important wheat diseases in the world. In this study, a single dominant powdery mildew resistance gene MlIW172 was identified in the IW172 wild emmer accession and mapped to the distal region of chromosome arm 7AL (bin7AL-16-0.86-0.90) via molecular marker analysis. MlIW172 was closely linked with the RFLP probe Xpsr680-derived STS marker Xmag2185 and the EST markers BE405531 and BE637476. This suggested that MlIW172 might be allelic to the Pm1 locus or a new locus closely linked to Pm1. By screening genomic BAC library of durum wheat cv. Langdon and 7AL-specific BAC library of hexaploid wheat cv. Chinese Spring, and after analyzing genome scaffolds of Triticum urartu containing the marker sequences, additional markers were developed to construct a fine genetic linkage map on the MlIW172 locus region and to delineate the resistance gene within a 0.48 cM interval. Comparative genetics analyses using ESTs and RFLP probe sequences flanking the MlIW172 region against other grass species revealed a general co-linearity in this region with the orthologous genomic regions of rice chromosome 6, Brachypodium chromosome 1, and sorghum chromosome 10. However, orthologous resistance gene-like RGA sequences were only present in wheat and Brachypodium. The BAC contigs and sequence scaffolds that we have developed provide a framework for the physical mapping and map-based cloning of MlIW172.

Abstract

The technology of tissue microarrays (TMAs) was developed against the background of decoding the human genome and the widespread application of high-density cDNA microarrays. TMAs facilitate comprehensive molecular profiling of cancer specimens with minimal tissue requirements. A TMA is a paraffin block composed of multiple tissue specimens. TMA sections provide targets for parallel in situ detection of DNA, RNA, and protein targets, allowing the rapid analysis of hundreds of molecular markers in the same set of specimens. Multiple specimens can be simultaneously investigated with different in situ techniques under identical laboratory conditions.

Abstract

The dielectric properties of Ba0.96Sr0.04Zr0.04Ln xTi0.96- xO3- δ (Ln = Y, Dy, Gd, Yb) ceramics were investigated in this work. The permittivity dependence on temperature showed the Curie temperature of the investigated ceramic samples was around 100 °C. The permittivity (>10000) at room temperature is obtained when the x is 0.004 and 0.003 for Y3+ and Dy3+, respectively. The origin of the dielectric abnormality may result from the amphoteric properties of Y3+ and Dy3+. [ABSTRACT FROM AUTHOR]

83. Structure and function of slow release organic carbon source in groundwater in-situ denitrification

Abstract

Many nitrate pollution cases exceed the threshold as recommended by the World Health Organization (50 mg NO3/L) and by the USA (10 mg N/L) for drinking water. In-situ denitrification was regarded as a good method to decrease nitrate contamination but is restricted by carbon absent in groundwater. Considering the disadvantages of known carbon sources, this paper provided slow-release organic carbon-source (SOC) technique to solve the problem and the results showed that SOC materials showed good performance during simulated groundwater denitrification. Structure analysis suggested that hydroxy chemical bond existed between PVA and starch in SOC and surface configuration changed with materials dissolving into water. After seven days of domestication, with 40-50 mg/L initial NO3-N, denitrification efficiency increased from 80.6% to 90.7% and the real COD consumption per N-NO3 reduction was 1.82-3.73 with 2.79 as average. Denitrification process followed the law of zero order kinetics and the parameter of denitrification dynamics, K, was from 0.1366 to 0.1873. It was suggested that SOC was a potential carbon source material (electron donor) suitable for in-situ groundwater denitrification.

Abstract

Ptx-WO3/C electrocatalysts were prepared by microwave colloid-reduction method. Transmission electron microscopy examination showed uniform dispersion of platinum with particle sizes less than 5 nm. Structures of the Ptx-WO3/C electrocatalysts were characterized by X-ray diffraction and energy dispersive spectroscopy, the electrochemical characteristics and electro-oxidation of electro-oxidation of ethanol on the Ptx-WO3/C electrocatalysts were also studied by cyclic voltammetry, linear sweep, and chronoamperometry. The results show that the Ptx-WO3/C electrocatalysts have higher catalytic activity to ethanol electrooxidation than the commercial Pt/C catalysts; perhaps caused by the “hydrogen spill-over effect.” The reaction mechanisms of the ethanol electro-oxidation on the Ptx-WO3/C were inferred according to the electrochemical characteristics and the data in the reported literature.

Abstract

Reverse transcription polymerase chain reaction (RT-PCR) has been used to detect hepatitis C virus (HCV) sequences in liver tissue. However, RT-PCR has a variable detection sensitivity, especially on routinely processed formalin-fixed, paraffin-embedded (FFPE) specimens. RNA-RNA and RNA-protein cross-links formed during formalin fixation is the major limiting factor preventing reverse trans criptase from extending the primers. To overcome this problem, we applied the ligation-dependent PCR (LD-PCR) for the detection of HCV RNA in FFPE liver tissue. This method uses two capture probes for RNA isolation and two hemiprobes for the subsequent PCR. Despite cross-links, the capture probes and the hemiprobes are able to form hybrids with HCV RNAs released from the FFPE tissue. The hybrids are isolated through binding of the capture probes to paramagnetic beads. The hemiprobes are then ligated by a T4 DNA ligase to form a full probe that serves as a template for the Taq DNA polymerase. A total of 22 FFPE liver specimens, 21 with hepatocellular carcinoma (HCC) and 1 with biliary cirrhosis secondary to bile duct atresia were selected for this study, of which 13 patients were HCV seropositive and 9 seronegative. HCV RNA was detectable by ID-PCR from all 13 HCV-seropositive HCCs and from 5 of 8 HCV-seronegative HCCs but not from the HCV-seronegative liver with biliary atresia. By contrast, RT-PCR detected HCV sequences in only 5 of the HCV-sero-positive and in 1 of the HCV-seronegative HCCs. To resolve the discordance between the LD-PCR and RT-PCR results, RT-PCR was performed on frozen liver tissue of the discrepant specimens, which confirmed the LD-PCR positive results. In conclusion, LD-PCR is a more sensitive method than RT-PCR for the detection of HCV sequences in routinely processed liver tissues. A high rate of HCV infection (86%) is found in HCC specimens, indicating a previously underestimated role of HCV in HCC pathogenesis.
