Abstract

The development of mammalian skeletal muscle is a highly complex process involving multiple molecular interactions. As a prevalent RNA modification, N6-methyladenosine (m6A) regulates the expression of target genes to affect mammalian development. Nevertheless, it remains unclear how m6A participates in the development of goat muscle. In this study, methyltransferase 3 (METTL3) was significantly enriched in goat longissimus dorsi (LD) tissue. In addition, the global m6A modification level and differentiation of skeletal muscle satellite cells (MuSCs) were regulated by METTL3. By performing mRNA-seq analysis, 8050 candidate genes exhibited significant changes in expression level after the knockdown of METTL3 in MuSCs. Additionally, methylated RNA immunoprecipitation sequencing (MeRIP-seq) illustrated that myocyte enhancer factor 2c (MEF2C) mRNA contained m6A modification. Further experiments demonstrated that METTL3 enhanced the differentiation of MuSCs by upregulating m6A levels and expression of MEF2C. Moreover, the m6A reader YTH N6-methyladenosine RNA binding protein C1 (YTHDC1) was bound and stabilized to MEF2C mRNA. The present study reveals that METTL3 enhances myogenic differentiation in MuSCs by regulating MEF2C and provides evidence of a post-transcriptional mechanism in the development of goat skeletal muscle. 64. Terminal weight and constraint design for wave energy converter economic model predictive control problems.

Abstract

The wave energy converter (WEC) control problem aims to make the best use of wave excitation to maximize energy capture and ensure safe operation across a broad range of sea states. This falls into the recently developed economic model predictive control (EMPC) framework, subject to wave excitation being treated as a predictable additive disturbance in the control problem. However, there are few theoretical developments on EMPC theory that can be directly used, as the persistent disturbances bring nontrivial problems to optimal operation, safety etc. Whilst the disturbance is beneficial for the energy production objective, it may cause safe operation problems. This article develops a systematic WEC EMPC design approach for WECs with essential linear hydrodynamical characteristics. By introducing terminal weight and terminal state constraint design into the WEC EMPC structure, important features such as convexity, satisfaction of safety constraints, and recursive feasibility can be guaranteed. Contrary to our intuition, we show that (1) the proposed EMPC with a terminal weight provides an unbiased estimate of the ideal conceptual infinite horizon solution, and (2) the WEC operational range, in terms of the operational sea states, is extended by imposing a terminal constraint. Abstract

Muscle growth and injury-induced regeneration are controlled by skeletal muscle satellite cells (MuSCs) through myogenesis in postnatal animals. Meanwhile, myogenesis is accompanied by mitochondrial function and enzyme activity. Nevertheless, the underlying molecular mechanisms involving non-coding RNAs including circular RNAs (circRNAs) and microRNAs (miRNAs) remain largely unsolved. Here, we explored the myogenic roles of miR-145-3p and MYBL1 on muscle development and mitochondrial mass. We noticed that overexpression of miR-145-3p inhibited MuSCs proliferation and reduced the number of viable cells. Meanwhile, deficiency of miR-145-3p caused by LNAantimiR-145-3p or an inhibitor retarded the differentiation of MuSCs. miR-145-3p altered the mitochondrial mass in MuSCs. Moreover, miR-145-3p targeted and negatively regulated the expression of CDR1as and MYBL1. The knockdown of the MYBL1 using ASO-2′MOE modification simulated the inhibitory function of miR-145-3p on cell proliferation. Additionally, MYBL1 mediated the regulation of miR-145-3p on Vexin, VCPIP1, COX1, COX2, and Pax7. These imply that CDR1as/miR-145-3p/MYBL1/COX1, COX2, VCPIP1/Vexin expression at least partly results in a reduction in mitochondrial mass and MuSCs proliferation. Abstract

Understanding the phase characteristic of methane hydrate in materials is of recent interest in several fields. In this study, methane hydrate dissociation conditions in nine water wetted porous materials (ZIF-8, MIL-53(Al), HKUST-1, nanoporous activated carbon, zeolite 13X, and four silica gels) with pore diameters ranging from 0.35 to 100 nm as well as in two nonporous quartz sands were systematically investigated. The adsorption properties of methane in some selected adsorbents were also studied. For microporous materials, our results confirmed that methane hydrate cannot form due to the size limitation for the nucleation of a methane hydrate crystal. In contrast, for mesoporous materials with a pore diameter of 6.2 nm or bigger, hydrate formed in their pores and significantly shifted dissociation temperatures and pressures compared to the bulk hydrate were observed. The largest temperature shift was found to be ∼6 K in materials possessing 6.2-nm-diameter pores, and this shift decreased to less than 1 K in structures having 45-nm-diameter pores. For macroporous materials with a pore diameter of 100 nm or for nonporous quartz systems, the influence of pore sizes on the methane hydrate phase equilibrium was found to be marginal. Furthermore, from studying the adsorption characteristic of methane in 25 wt% wetted MIL-53(Al), 30 wt% wetted HKUST-1, and 10 wt% wetted nanoporous activated carbon, we demonstrated that the penetration of water molecules into the pores of microporous materials was not only unable to form methane hydrate but also substantially deteriorated the gas adsorption ability. To exploit simultaneously adsorption and hydrate formation in wetted microporous materials for gas storage, both the hydrophobicity and aperture pore size of materials are critical toward the selection of promising adsorbents.
