The shoot apical meristem (SAM) of angiosperm plants is a small, highly organized structure that gives rise to all above-ground organs. The SAM is divided into three functional domains: the central zone (CZ) at the SAM tip harbors the self-renewing pluripotent stem cells and the organizing center, providing daughter cells that are continuously displaced into the interior rib zone (RZ) or the surrounding peripheral zone (PZ), from which organ primordia are initiated. Despite the constant flow of cells from the CZ into the RZ or PZ, and cell recruitment for primordium formation, a stable balance is maintained between the distinct cell populations in the SAM. Here we combined an in-depth phenotypic analysis with a comparative RNASeq approach to characterize meristems from selected combinations of clavata3 (clv3), jabba-1D (jba-1D) and erecta (er) mutants of Arabidopsis thaliana. We demonstrate that CLV3 restricts meristem expansion along the apical-basal axis, whereas class III HD-ZIP and ER pathways restrict meristem expansion laterally, but in distinct and possibly perpendicular orientations. Our k-means analysis reveals that clv3, jba-1D/+ and er lead to meristem enlargement by affecting different aspects of meristem function; for example, clv3 displays an increase in the stem cell population, whereas jba-1D/+ er exhibits an increase in mitotic activity and in the meristematic cell population. Our analyses demonstrate that a combined genetic and mRNA-Seq comparative approach provides a precise and sensitive method to identify cell type-specific transcriptomes in a small structure, such as the SAM. The ability to move on solid surfaces provides ecological advantages for bacteria, yet many bacterial species lack this trait. We found that Xanthomonas spp. overcome this limitation by making use of proficient motile bacteria in their vicinity. Using X. perforans and Paenibacillus vortex as models, we show that X. perforans induces surface motility, attracts proficient motile bacteria and 'rides' them for dispersal. In addition, X. perforans was able to restore surface motility of strains that lost this mode of motility under multiple growth cycles in the lab. The described interaction occurred both on agar plates and tomato leaves and was observed between several xanthomonads and motile bacterial species. Thus, suggesting that this motility induction and hitchhiking strategy might be widespread and ecologically important. This study provides an example as to how bacteria can rely on the abilities of their neighboring species for their own benefit, signifying the importance of a communal organization for fitness. A simple and effective way to prepare multi-walled carbon nanotubes (MWNT)//silica hybrid microcapsules (colloidosomes) is presented. These microcapsules have been generated by emulsion templating in a biphasic oil-in-water (o/w) system. Two trialkoxysilanes of complementary polarity, (3-aminopropyl)triethoxysilane (APTES) and dodecyltriethoxysilane (DTES), were used to chemically immobilize the silica nanoparticles at the o/w interface and stabilize the as-generated Pickering emulsions. The effects of varying the o/w ratio and the concentration of the added solids on the type of emulsion formed, the oil droplet size, as well as the emulsion stability have been investigated. The emulsion phase fraction was dependent on the silica content while the droplet size increased with increasing oil volume percentage. A solid shell emerged around the oil droplets from copolymerization between silane monomers. The thickness of the resulting shells was several hundreds of nm. Although MWNTs and silica nanoparticles both were co-assembled at the o/w interface, silica has shown to be the sole stabilizer, with APTES being crucial for the formation of the shell structure. Drop-casting of the emulsion and air-drying led to hierarchical open porous MWNT-silica nanocomposites. These new structures are promising as electrically conductive thin films for variety of applications, such as electro-optics, encapsulation, or chemical sensing. [ABSTRACT FROM AUTHOR]

Acidovorax citrulli causes bacterial fruit blotch (BFB) of cucurbits, a disease that threatens the cucurbit industry worldwide. Despite the economic importance of BFB, little is known about pathogenicity and fitness strategies of the bacterium. We have observed the phenomenon of phenotypic variation in A. citrulli. Here we report the characterization of phenotypic variants (PVs) of two strains, M6 and 7a1, isolated from melon and watermelon, respectively. Phenotypic variation was observed following growth in rich medium, as well as upon isolation of bacteria from inoculated plants or exposure to several stresses, including heat, salt and acidic conditions. When grown on nutrient agar, all PV colonies possessed a translucent appearance, in contrast to parental strain colonies that were opaque. After 72 h, PV colonies were bigger than parental colonies, and had a fuzzy appearance relative to parental strain colonies that are relatively smooth. A. citrulli colonies are generally surrounded by haloes detectable by the naked eye. These haloes are formed by type IV pilus (T4P)-mediated twitching motility that occurs at the edge of the colony. No twitching haloes could be detected around colonies of both M6 and 7a1 PVs, and microscopy observations confirmed that indeed the PVs did not perform twitching motility. In agreement with these results, transmission electron microscopy revealed that M6 and 7a1 PVs do not produce T4P under tested conditions. PVs also differed from their parental strain in swimming motility and biofilm formation, and interestingly, all assessed variants were less virulent than their corresponding parental strains in seed transmission assays. Slight alterations could be detected in some DNA fingerprinting profiles of 7a1 variants relative to the parental strain, while no differences at all could be seen among M6 variants and parental strain, suggesting that, at least in the latter, phenotypic variation is mediated by slight genetic and/or epigenetic alterations. This study presents a new eco-friendly formulation of entomopathogenic nematodes (EPNs) based on individual coating of EPNs with titanium dioxide (TiO 2 ) nanoparticles (NPs) and mineral oil via oil-in-water Pickering emulsions. Mineral oil-in-water emulsions stabilized by amine-functionalized titanium dioxide (TiO 2 -NH 2 ) particles were prepared. 40:60 and 50:50 oil-water volume ratios using 2 wt % TiO 2 -NH 2 particles were found to be the most stable emulsions with a droplet size suitable for the formulation and were further studied for their toxicity against the incorporated EPNs. Carboxyfluorescein was covalently bonded to TiO 2 -NH 2 NPs, and the resulting composite was observed via fluorescence confocal microscopy. The dry coating was evaluated using SEM and confocal microscopy, which showed significant nematode coverage by the particles and oil. The final formulation was biocompatible with the studied EPNs, where the viability of the EPNs in the formulation was equivalent to control aqueous suspension after 120 days. Finally, yields of nematodes from infected Galleria mellonella cadavers collected for 150 days showed no significant differences ( P > 0.05) using the tested emulsions compared to the control containing nematodes in water.

A new formulation for biological pest control with significant UV protection capability has been developed in this research. The formulation is based on individual encapsulation of fungal conidia in an oil/water Pickering emulsion. The droplets size of the emulsions was tuned to meet the demands of single conidia encapsulation in the oil droplets. The emulsions are stabilized by amine-functionalized TiO 2 (titania) nanoparticles (NPs). The droplet size, stability, and structure of the emulsions were investigated at different TiO 2 contents and oil/water phase ratios. Most of the emulsions remained stable for 6 months. The structural properties of the Pickering emulsions were characterized by confocal microscopy and high-resolution cryogenic scanning electron microscopy (cryo-HRSEM). The presence of the TiO 2 particles at the interface was confirmed by both confocal microscopy and cryo-HRSEM. Metarhizium brunneum-7 (Mb7) conidia were added to the emulsions. The successful encapsulation of individual conidia in the oil droplets was confirmed by confocal microscopy. The individual encapsulation of the conidia in the emulsions was significantly improved by dispersing the conidia in a 0.02 % Triton X-100 solution prior to emulsification. In addition, the bioassay results have shown, that exposure of the encapsulated conidia to natural UV light did not change their germination rates, however, the unprotected conidia demonstrated a dramatic decrease in their germination rates.
