Abstract

BACKGROUD: Two‐component histidine kinase (HK) phosphorelay signaling systems play important roles in differentiation, virulence, secondary metabolite production and response to environmental signals. Allyl isothiocyanate (A‐ITC) is a hydrolysis product of glucosinolates with excellent antifungal activity. Our previous study indicated that the mycelial growth of Cochliobolus heterostrophus was significantly hindered by A‐ITC. However, the function of HK in regulating A‐ITC sensitivity was not clear in C. heterostrophus, the causal agent of Southern corn leaf blight. RESULTS: In this study, the role of HKs was investigated in C. heterostrophus. Deletion of the HK coding gene ChNIK1 resulted in dramatically increased sensitivity of C. heterostrophus to A‐ITC. In addition, ΔChnik1 mutant exhibited significantly decreased conidiation and increased sensitivity to NaCl, KCl, tebuconazole and azoxystrobin, but deletion of the other five HK genes did not affect the A‐ITC sensitivity of C. heterostrophus. ChSLN1, ChNIK4, ChNIK8 and ChMAK2 are essential for conidiation and response to H2O2 and sodium dodecyl sulfate. However, deletion of NIKs had on effect on significant virulence. 61. The consequences of urbanization on vegetation photosynthesis in the Yangtze River Delta, China

Abstract

Vegetation photosynthesis is a critical component of terrestrial carbon cycles, and its peak value (peak photosynthetic rate) dominates plant CO2 uptake. Urbanization has significantly influenced vegetation photosynthesis by changing environmental conditions directly (replacement of vegetated surfaces with impervious surfaces) and indirectly (e.g., atmospheric and climatic conditions). Although some studies have investigated the effects of urbanization on vegetation productivity across the whole study area, how the peak photosynthetic rate responds to urbanization for different vegetation types has received scant scholarly attention. Here, using the urban-rural gradient approach, we explored the effects of urbanization on the peak photosynthetic rate (surrogated by GPPmax and EVImax) of different vegetation types across urbanization intensity (UI) gradients in the Yangtze River Delta Urban Agglomeration (YRDUA). The results showed that (1) the GPPmax and EVImax of the forest, grassland, and wetland decreased with urban intensity. GPPmax and EVImax in rural areas (UI ≦ 1%) were 36.62 and 30.47% higher than those of urban areas (UI > 1%) overall vegetation types. (2) The indirect impacts can offset or exacerbate the direct loss of vegetation photosynthesis caused by urbanization in YRDUA. Especially for forest ecosystems in YRDUA, the negative direct urbanization effect is exacerbated by the indirect reduction in vegetation growth. However, the indirect effects of urbanization on grassland and wetland offset 47.08 and 87.23% loss of GPPmax, and 34.17 and 79.73% loss of EVImax due to the reduction of vegetated surface area, respectively. (3) The indirect impact of urbanization on vegetation photosynthesis was dominated by the elevated daytime land surface temperature (LSTday). Our study highlights the necessity of distinguishing the various effects of urbanization on vegetation photosynthesis, which may help stakeholders formulate more reasonable urban environmental planning.

Abstract

Background: Isothiocyanates (ITCs) generated from the 'glucosinolates‐myrosinase' defense system in the Brassicaceae exhibit broad antagonistic activity to various fungal pathogens. Nevertheless, the antifungal activity of ITCs to non‐adapted fungi of Brassicaceae plants were seldom determined. The inhibitory effects of ITCs on Cochliobolus heterostrophus were evaluated and the antagonistic mechanism was explored. Results: The mycelium growth of C. heterostrophus was hindered significantly by allyl, 4‐(methylthio)‐butyl, and phenyethyl ITCs, 4MTB‐ITC exhibited the highest inhibitory effect on mycelium growth with an IC50 value of 53.4 μmol L−1. In addition, ITCs exhibited obvious inhibitory effect on conidia germination and pathogenicity of C. heterostrophus. Proteomic analysis indicated that the inhibition of C. heterostrophus by A‐ITC downregulated the expression of genes related to energy metabolism, oxidoreductase activity, melanin biosynthesis, and cell wall‐degrading enzymes. Furthermore, mutants ΔChtrx2 and ΔChnox1 showed increased sensitivity to ITCs, and melanin biosynthesis was inhibited significantly in C. heterostrophus in response to A‐ITC. Interestingly, unlike other pathogens that infected Brassicaceae plants, the SaxA in C. heterostrophus displayed no function in ITC degradation. In addition, the ITCs also exhibited obvious inhibitory effect on mycelium growth of Setosphaeria turcica, Fusarium graminearum, and Magnaporthe oryzae. Conclusion: This study indicated that non‐Brassicaceae‐adapted pathogens are more sensitive to ITCs, and ITCs could have applications in protecting non‐Brassicaceae crops in future. In addition, loss of ChNOX1 and ChTRX2 increased the sensitivity of C. heterostrophus to ITCs. However, whether exercise can reverse this process to promote T2DM bone formation and its mechanism remains unknown.Methods: A T2DM mouse model was established by a high-fat diet and STZ injection, and the mice were trained with 8-week HIIT and downhill running exercise. Micro-CT was used to scan the bone microstructure. Bone morphology was observed by HE staining, and the osteoblast (OB) activity in bones was observed by AKP staining. Calcium ion and phosphorus concentration in serum was detected by ELISA; RT-PCR was used to detect the mRNA level, and Western blot was used to detect the protein level of related indexes in Meg3/p62/Runx2 pathway.Results: The inhibition of bone autophagy, in the bones of T2DM mice, resulted in the degradation of the bone tissue morphology and structure, with the increase of the expressions of Meg3, PI3K, Akt, mTOR, p62 and NF-κB. Abstract

Eclipta, widespread in tropical, subtropical, and temperate regions, is one of the main malignant broadleaf weeds and thrives in moist and dryland fields. Field rates of acetolactate synthase (ALS) inhibitors have failed to control eclipta in some farmlands in China. One ALS inhibitor–resistant population (R) collected from Jiangsu province in China was confirmed in the greenhouse in our preliminary work. Whole-plant assays revealed that this R population was highly resistant to four sulfonylureas (pyrazosulfuron-ethyl, 134-fold; bensulfuron-methyl, 172-fold; metsulfuron-methyl, 30-fold; and tribenuron-methyl, 195-fold), two triazolopyrimidines (pyroxsulam, 98-fold; penoxsulam, 30-fold), and one pyrimidinylthio-benzoate (bispyribac-sodium, 166-fold) and was moderately resistant to two imidazolinones (imazethapyr, 10-fold; imazapic, 19-fold). ALS enzyme-activity assays showed insensitivity of the ALS from the R population (resistance index values ranged from 12 to 293) to all of the above ALS inhibitors in vitro. Chromatograms from ALS gene sequence analysis detected a homozygous Pro-197-Ser amino acid substitution in the R population. These results confirmed that the Pro-197-Ser substitution results in broad-spectrum cross-resistance to ALS inhibitors in the eclipta R population. Abstract

Sclerotinia sclerotiorum, the notorious necrotrophic phytopathogenic fungus with wide distribution, is responsible for sclerotium disease in more than 600 plant species, including many economic crops such as soybean, oilseed rape, and sunflower. The compound appressorium is a crucial multicellular infection structure that is a prerequisite for infecting healthy tissues. Previously, the Forkhead‐box family transcription factors (FOX TFs) SsFoxE2 and SsFKH1 were shown to play a key regulatory role in the hyphae growth, sexual reproduction, and pathogenicity of S. sclerotiorum. However, little is known about the roles of SsFoxE3 regulating growth and development and pathogenicity. Here, we report SsFoxE3 contributes to sclerotium formation and deletion of SsFoxE3 leads to reduced formation of compound appressoria and developmental delays. Transcripts of SsFoxE3 were greatly increased during the initial stage of infection and SsFoxE3 deficiency reduced virulence on the host, while stabbing inoculation could partially restore pathogenicity. The SsFoxE3 mutant showed sensitivity to H2O2, and the expression of reactive oxygen species detoxification and autophagy‐related genes were reduced. Moreover, expression of SsAtg8 was also decreased during the infection process of the SsFoxE3 mutant. Yeast 1‐hybrid tests suggested that SsFoxE3 interacted with the promoter of SsAtg8. Disruption of SsAtg8 resulted in a phenotype similar to that of the SsFoxE3 mutant. Comparative analysis of the level of autophagy in the wild type and SsFoxE3 mutant showed that N starvation‐induced autophagy was reduced in the SsFoxE3 mutant. Taken together, our findings indicate that SsFoxE3 plays an important role in compound appressorium formation and is involved in transcriptional activation of SsAtg8 during infection by S. sclerotiorum. Abstract

MicroRNAs (miRNAs) have been shown to play important roles in plant development, growth and stress response. Sweet sorghum [Sorghum bicolor (L.) Moench] is an important source of bioenergy due to the high sugar content in its stems. However, it is not clear how the miRNA is involved in sugar accumulation in sorghum stems. In order to identify the miRNAs in the stems and the leaves of sweet sorghum, we extracted RNAs of the stems and leaves of sweet sorghum (Rio) and grain sorghum (BTx623) at the heading and dough stages for high-throughput sequencing. A total of 179279048 reads were obtained from Illumina-based sequencing. Further analysis identified nine known miRNAs and twelve novel miRNAs that showed significantly and specifically differentially expressed in the stems of sweet sorghum. The target genes of the differentially expressed novel miRNAs include the transcription factor, glucosyltransferase, protein kinase, cytochrome P450, transporters etc. GO enrichment analysis showed that the predicted targets of these differentially expressed miRNAs participated in diverse physiological and metabolic processes. We performed RT-qRCR analysis on these miRNAs across eight different libraries to validate the miRNAs. Finally, we screened stem-specifically expressed novel miRNA and a leaf-specifically expressed novel miRNA in sweet sorghum comparing with grain sorghum. Our results provide a basis for further investigation of the potential role of these individual miRNAs in sugar accumulation.

Abstract

In our country, irrigation mode in the greenhouse is still flood irrigation, which not only wastes water resources in short supply, but also restricts the increase of the crop output and the improvement of its quality in the greeenhouse. The paper used sensor technology to real-time do acquisition of the temperature and humidity of air, the moisture content of soil and the illuminance in the greenhouse. The SCM (Single Chip Micyoco) gived a reasonable control decision by analysing these data to do automatically sprinkling irrigation by control the actuator. The system achieves computer automatic control, sprinkling irrigation by need, time and quantity. It is energy-saving, environmental protection, labor saving, so the system has a certain practicality and potential popularization.
