With global warming and regional decreases in precipitation, drought has become a problem worldwide. As the number of arid regions in the world is increasing, drought has become a major factor leading to significant crop yield reductions and food crises. Soybean is a crop that is relatively sensitive to drought. It is also a crop that requires more water during growth and development. The aim of this study was to identify the quantitative trait locus (QTL) that affects drought tolerance in soybean by using a recombinant inbred line (RIL) population from a cross between the drought-tolerant cultivar ‘Jindou21’ and the drought-sensitive cultivar ‘Zhongdou33’. Nine agronomic and physiological traits were identified under drought and well-watered conditions. Genetic maps were constructed with 923,420 polymorphic single nucleotide polymorphism (SNP) markers distributed on 20 chromosomes at an average genetic distance of 0.57 centimorgan (cM) between markers. A total of five QTLs with a logarithm of odds (LOD) value of 4.035–8.681 were identified on five chromosomes. Under well-watered conditions and drought-stress conditions, one QTL related to the main stem node number was located on chromosome 16, accounting for 17.177% of the phenotypic variation. Nine candidate genes for drought resistance were screened from this QTL, namely Glyma.16G036700, Glyma.16G036400, Glyma.16G036600, Glyma.16G036800, Glyma.13G312700, Glyma.13G312800, Glyma.16G042900, Glyma.16G043200, and Glyma.15G100700. These genes were annotated as NAC transport factor, GATA transport factor, and BTB/POZ-MATH proteins. This result can be used for molecular marker-assisted selection and provide a reference for breeding for drought tolerance in soybean.

Elderly with comorbidities have shown a higher rate of fatal outcomes when suffering coronavirus disease 2019 (COVID-19). However, a delineation of clinical significances of hematologic indices and underlying comorbidities in the progression and outcome of COVID-19 remains undefined. Six hundred two COVID-19 patients with established clinical outcomes (discharged or deceased) from Hankou Hospital of Wuhan, China between January 14, 2020 and February 29, 2020 were retrospectively analyzed. Of the 602 patients with COVID-19, 539 were discharged and 63 died in the hospital. The deceased group showed higher leukocyte and neutrophil counts but lower lymphocyte and platelet counts. Longer activated partial thromboplastin time (APTT) and prothrombin time (PT), as well as higher D-dimer and C-reactive protein levels, were found in non-survivors. Our observations suggest that these parameters could serve as potential predictors for the fatal outcome and in the discharged group. A higher neutrophil count and D-dimer level but lower lymphocyte were associated with a longer duration of hospitalization. A multivariable Cox regression analysis showed that higher neutrophil count, prolonged PT, and low lymphocyte count were risk factors for patients with COVID-19. Also, we found an association of lower lymphocyte count and higher C-reactive protein levels with the elderly group and those with cardiovascular-related comorbidities. The significantly different hematologic profiles between survivors and non-survivors support that distinct hematologic signatures in COVID-19 patients will dictate different outcomes as a prognostic marker for recovery or fatality. Lymphopenia and aggressive inflammatory response might be major causes for fatal outcomes in the elderly male and especially those with cardiovascular-related comorbidities.

Background Transcatheter aortic valve replacement (TAVR) has become the preferred treatment for symptomatic patients with aortic stenosis and elevated procedural risk. Many deaths following TAVR are because of noncardiac causes and comorbid disease burden may be a major determinant of postprocedure outcomes. The prevalence of comorbid conditions and associations with outcomes after TAVR has not been studied. Methods and Results This was a retrospective single‐center study of patients treated with TAVR from January 2015 to October 2018. The association between 21 chronic conditions and short‐ and medium‐term outcomes was assessed. A total of 341 patients underwent TAVR and had 1‐year follow‐up. The mean age was 81.4 (SD 8.0) years with a mean Society of Thoracic Surgeons predicted risk of mortality score of 6.7% (SD 4.8). Abstract As major environment factors, drought or high salinity affect crop growth, development and yield. Transgenic approach is an effective way to improve abiotic stress tolerance of crops. In this study, we comparatively analyzed gene structures, genome location, and the evolution of syntaxin proteins containing late embryogenesis abundant (LEA2) domain. GmSYP24 was identified as a dehydration-responsive gene. Our study showed that the GmSYP24 protein was located on the cell membrane. The overexpression of GmSYP24 (GmSYP24ox) in soybean and heteroexpression of GmSYP24 (GmSYP24hx) in Arabidopsis exhibited insensitivity to osmotic/drought and high salinity. However, wild type soybean, Arabidopsis, and the mutant of GmSYP24 homologous gene of Arabidopsis were sensitive to the stresses. Under the abiotic stresses, transgenic soybean plants had greater water content and higher activities of POD, SOD compared with non-transgenic controls. And the leaf stomatal density and opening were reduced in transgenic Arabidopsis. The sensitivity to ABA was decreased during seed germination of GmSYP24ox and GmSYP24hx. GmSYP24hx induced up-regulation of ABA-responsive genes. GmSYP24ox alters the expression of some aquaporins under osmotic/drought, salt, or ABA treatment. These results demonstrated that GmSYP24 played an important role in osmotic/drought or salt tolerance in ABA signal pathway.

This study constructs comprehensive panel data based on the China City Statistical Yearbook and environmental indicators disclosed by the Ministry of Ecology and Environment from 2013 to 2017, using a difference-in-difference (DID) model to empirically validate the effects of the ecological environmental damage compensation system on urban air quality, followed by a further analysis of the system’s effect mechanism, namely, how the system has generated effects on reducing environmental pollution. This study finds that: (1) the ecological environmental damage compensation system can significantly improve urban air quality, and small cities are more sensitive to the pilot policy; and (2) the main impact is that the pilot policy mechanism improved the urban pollutant treatment capacity and reduced the proportion of the secondary industry. After multiple robustness tests, this conclusion still holds. This study provides empirical evidence for fully implementing an ecological environmental damage compensation system.

Background: Effective anticancer therapy can be achieved by identifying novel tumor-specific drug targets and screening of new drugs. Recently, TMEM16A has been identified to be overexpressed in lung adenocarcinoma, and inhibitors of TMEM16A showed obvious antitumor efficacy.Methods: YFP fluorescence quenching and whole-cell patch clamp experiments were used to explore the inhibitory effect of silibinin on TMEM16A. Molecular docking and site-directed mutagenesis were performed to confirm the binding sites of silibinin and TMEM16A. MTT assay, wound healing assay, and annexin-V assay were used to detect the effect of silibinin on cancer cell proliferation, migration, and apoptosis. shRNA was transfected into LA795 cells to knock down the expression of endogenous TMEM16A. Tumor xenograft mice combined with Western blot experiments reveal the inhibitory effect and mechanism of silibinin in vivo.Results: Silibinin concentration dependently inhibited the whole-cell current of TMEM16A with an IC50 of 30.90 ± 2.10 μM. The putative binding sites of silibinin in TMEM16A were K384, R515, and R535. The proliferation and migration of LA795 cells were downregulated by silibinin, and the inhibition effect can be abolished by knockdown of the endogenous TMEM16A. Further, silibinin was injected to tumor xenograft mice which exhibited significant antitumor activity without weight loss. Finally, Western blotting results showed the mechanism of silibinin inhibiting lung adenocarcinoma was through apoptosis and downregulation of cyclin D1.Conclusion: Silibinin is a novel TMEM16A inhibitor, and it can be used as a lead compound for the development of lung adenocarcinoma therapy drugs.

Nanoscale zero-valent iron (nZVI) is a well-known efficient nanomaterial for the immobilization of heavy metals and has been widely applied in the remediation of contaminated groundwater and soils. In this study, a series of field emission scanning electron microscopy (FESEM) analyses, vane shear tests, triaxial compression tests, and oedometer tests was conducted on lead-contaminated clay using four dosages of nZVI treatment (0.2%, 1%, 5%, and 10%). The geotechnical properties, including basic index properties, stiffness, shear strength, and compressibility, were assessed after the reaction procedure. FESEM analysis was performed to explore the potential mechanisms of nZVI treatment in terms of morphological characteristics. It was found that the plasticity index decreased gradually with increasing nZVI dosage. Treating contaminated soil with nZVI caused an increase in the vane shear strength, stiffness, and friction angle. The compression index increased gradually because of the nZVI treatment. Based on the FESEM analysis, a conclusion can be deduced that larger aggregates and conjoined structures resulting from nZVI treatment can lead to the strengthening of lead-contaminated clay. Le fer a valence zero a l'echelle nanometrique (nZVI) est un nanomateriau efficace bien connu pour l'immobilisation de metaux lourds. Il a ete largement utilise dans des activites de depollution des eaux souterraines et des sols contamines. Dans cette etude, une serie d'analyses en microscopie electronique a balayage a emission de champ (FESEM), d'essais de cisaillement des aubes, d'essais de compression triaxiale et d'essais redometriques ont ete effectues sur de l'argile contaminee au plomb a l'aide de quatre doses de traitement par nZVI (0,2,1, 5 et 10%). Les proprietes geotechniques, y compris les proprietes d'indice de base, la rigidite, la resistance au cisaillement et la compressibilite ont ete evaluees apres la procedure de reaction. Une analyse FESEM a ete realisee pour explorer les mecanismes potentiels du traitement nZVI quant aux caracteristiques morphologiques. Il a ete constate que l'indice de plasticite diminuait progressivement avec l'augmentation de la dose de nZVI. Le traitement du sol contamine avec du nZVI a entrame une augmentation de la resistance au cisaillement, de la rigidite et de l'angle de friction de l'aube. L'indice de compression a augmente progressivement a cause du traitement nZVI. Sur la base de l'analyse FESEM, on peut deduire que de plus grands agregats et des structures jointes resultant du traitement au nZVI sont en mesure d'assurer le renforcement de l'argile contaminee au plomb. GADD45A is a DNA damage and stressful growth arrest inducible protein, also it is shown to a be tumor suppressor gene and a chromatin relaxer associated with opening chromatin during the somatic reprogramming. However, its role in human embryonic stem cells and human embryonic stem cell modeled development has been merely documented. To illustrate the function of GADD45A in the human embryonic stem cell biology, we reported a GADD45A knockout human embryonic stem cell line by CRISPR/Cas9 mediated gene targeting. This cell line displayed normal karyotype, pluripotent stem cell marker expression and differentiation potential both in vivo and vitro.

Circular RNAs (circRNAs) have recently been shown to play critical roles in tumorigenesis. However, the roles of circRNAs in hepatocellular carcinoma (HCC) remain largely unknown. In the present study, we identified a novel circRNA (hsa_circ_0070269) was significantly decreased in HCC tissues and cell lines, low hsa_circ_0070269 expression was positively associated with advanced tumor stage, lymph node metastasis, and poor overall survival. Hsa_circ_0070269 overexpression suppressed proliferation and invasion of HCC cells in vitro and reduced tumor growth in vivo. Mechanistically, hsa_circ_0070269 increased NPTX1 expression via sponging miR-182 in HCC cells, which inhibited aggressive tumor behavior. Taken together, our findings suggest that hsa_circ_0070269 might play an important role in HCC development via miR-182/NPTX1 axis, therefore could serve as a potential therapeutic target for HCC treatment.

Cell-penetrating peptides (CPPs), as non-viral gene delivery vectors, are considered with lower immunogenic response, and safer and higher gene capacity than viral systems. In our previous study, a CPP peptide called RALA (arginine rich) presented desirable transfection efficacy and owns a potential clinic use. It is believed that histidine could enhance the endosome escaping ability of CPPs, yet RALA peptide contains only one histidine in each chain. In order to develop novel superior CPPs, by using RALA as a model, we designed a series of peptides named HALA (increased histidine ratio). Both plasmid DNA (pDNA) and siRNA transfection results on three cell lines revealed that the transfection efficacy is better when histidine replacements were on the C-terminal instead of on the N-terminal, and two histidine replacements are superior to three. By investigating the mechanism of endocytosis of the pDNA nanocomplexes, we discovered that there were multiple pathways that led to the process and caveolae played the main role. During the screening, we discovered a novel peptide-HALA2 of high cellular transfection efficacy, which may act as an exciting gene delivery vector for gene therapy. Our findings also bring new insights on the development of novel robust CPPs.

In this paper, the passive heave compensator for the strand jack lifting system is studied. An analytical model is developed, which considers the nonlinear characteristic of the compensator stiffness thus as to predict its response under different parameters accurately. This analytical model helps to find the feasible gas volume of the compensator. The comparative analysis is carried out to analyze the influence of key design parameters on the dynamic response of the compensator. In order to evaluate the effectiveness of the compensator, a coupling model of the strand jack lifting system is derived. The compensator efficiency is evaluated in terms of the lifted structure displacement and the strand dynamic tension. The numerical simulations are performed to evaluate the effectiveness of the compensator. Numerical results show that the compensator is able to significantly decrease the tension variation in the strands and the motion of the lifted structure.

Persulfate (PSF) is a strong oxidant that has been used extensively in the In-Situ Chemical Oxidation (ISCO) technology. The geoenvironmental impact of PSF treatment is barely investigated. This situation should be carefully considered as it may affect the reutilization of contaminated soil as engineering materials. This paper studied the removal of bisphenol A (BPA) by PSF with Nano Zero-Valent Iron (nZVI) and percarbonate (SPC) activated/enhanced and their subsequent impacts on the engineering properties of soil. The physicochemical and geotechnical properties of soils before and after treatment were evaluated using batch experiments. The results indicate that the introduced pristine PSF can be activated by some naturally occurring matters and subsequently lead to the mineralization of BPA. Both non-activated PSF and activated/enhanced PSF treatment led to the soil improvement in the undrained shear strength at different degrees. The primary mechanism of soil improvement is ascribed to the heterogeneous sulfate and/or carbonate precipitation. Meanwhile, Ca2+ in the pore fluid played a significant role in the enhancement of the soil strength. A conclusion was drawn that the treatment of both non-activated PSF, nZVI- and SPC-activated PSF treatment can achieve removal of BPA and soil improvement in the short-term simultaneously. This study can improve the PSF-involved remediation of brownfields and dredged sediments for a sustainable and low-carbon society.

A wave compensation platform based on 3-SPR parallel platform is designed for marine ships with a dynamic positioning system. It can compensate for the heave, rolling, and pitching movement of a vessel under level 4 sea state. The forward kinematics of the mechanism is used to draw the central point position workspace and the attitude workspace of the moving deck of the compensation platform. The compensation effects of the 3-RPS parallel compensation platform and the 3-SPR parallel compensation platform are compared, and the feasibility and superiority of the compensation scheme using the 3-SPR parallel compensation platform are proved. To lower the working height of the upper deck of the compensation platform and reduce the extension range of the support legs, the structure of the compensation platform is optimized, and a novel 3-SPR parallel platform is designed. Finally, a simulation model was established. Using the inverse kinematic model as a compensation movement solver which can online calculate the length of branch legs based on the measured heaving, rolling, and pitching values of vessels, the compensation effect of the new structure under a certain sea state is simulated. The result demonstrated the efficiency of the ship motion decoupling movement of the newly designed compensation platform and proved the competence of it.

Wave energy is a renewable energy source that is green, clean and has huge reserves. In order to develop wave energy resources, an oscillating buoy Wave Energy Converter (WEC) device based on the artificial breakwater is presented in this paper. In order to effectively vent the gas in the hydraulic PTO and to improve the active control capability of the PTO system to guarantee the safety performance of the system under high sea conditions, a hydraulic PTO with an active control circuit is designed. Additionally, for the Power Take-Off (PTO) system, there is a optimal damping point under different sea conditions for PTO system, so the PTO can be controlled by the Maximum-Power-Point-Tracking (MPPT) control algorithms to improve the generated power of the system. At present, the MPPT control algorithms for wave energy are mainly used to control the load of generator. However, a fixed-load storage battery is used for the load of the generator in this paper. Additionally, an MPPT control taken at a hydraulic PTO system is executed to improve the power generated by hydraulic PTO under different sea conditions effectively in this paper. The MPPT control based on the hydraulic system is conducted by controlling the displacement of hydraulic motor to achieve the optimal damping point tracking control. The control flow of the MPPT algorithm is provided. The variable step hill-climbing method is used in MPPT control algorithm in which the big step can reduce the time of tracking and the small step can increase the accuracy of MPPT control algorithm. Due to the slow stability of the hydraulic system, a filter method for hydraulic PTO power is used. In addition, the hydraulic PTO system and MPPT control are verified to be feasible with the simulation. Additionally, MPPT control based on hydraulic variable motor is easier to carry out in practical applications than the traditional control of resistance. Finally, the simulation results demonstrate that it is an effective power control strategy for hydraulic PTO system to improve the generated power.

To improve the energy extraction performance of the oscillating hydrofoil, the lift force that acts on the oscillating hydrofoil is analyzed. The pressure difference between the oscillating hydrofoil‘s opposing surfaces is dominant to generate the lift force. Forming and shedding of the leading-edge vortex from the hydrofoil surface determines the pressure difference between the opposing surfaces of the oscillating hydrofoil. In this paper, the hydrofoil with different chord flexibility coefficients and maximum offset at the trailing edge are analyzed to obtain the power coefficient, lift coefficient, and moment coefficient of the oscillating hydrofoil. The influence mechanism of chord-wise deformation of the oscillating hydrofoil on the energy extraction performance is explored. According to the Kutta−Joukowsky condition and the Stokes’ theorem, the relationship between the attached vortex on the hydrofoil and the surface pressure of the hydrofoil, the surface pressure difference of the hydrofoil, and the lift force that acts on the hydrofoil are investigated. By quantifying the vortex intensity, the ascending-shedding process of the attached vortex on the hydrofoil is characterized. Finally, the complete influence chain among the chord-wise flexure, the attached vortex on the hydrofoil, and the energy extraction performance of the oscillating hydrofoil is established.

Sorting nexin-1 (SNX1) is an important functional protein in cell endocytosis, efflux, protein sorting, cell signal transduction, etc; however, the expression, the role and clinical relevance of SNX1 have not been investigated in gastric cancer (GC). In this study, we first performed a bioinformatics investigation using the data obtained from The Cancer Genome Atlas (TCGA) database. The result showed that SNX1 mRNA levels were significantly lower in GC tissues than in paracancerous tissues. In a study of 150 cases of GC, including 60 cases with paired paracancerous and cancer tissues and 90 cases with detailed follow-up information, SNX1 expression was analyzed by immunohistochemistry. Our study on paired paracancerous and cancer tissues showed that SNX1 protein expression remarkably decreased in GC tissues (50/60, 83.33%). A study on 90 patients with detailed follow-up information showed that tumors with higher SNX1 protein level were correlated with better clinicopathologic stages (p = 0.0285), nodal status (p = 0.0286), smaller tumor sizes (p = 0.0294) and a better survival rate in patients with GC (p = 0.0245). Univariate analysis of the 90 patients with GC showed that low-level SNX1 was significantly correlated with decreased overall survival of GC patients (p = 0.008), and associated with a relatively higher cumulative hazard of death. Exogenous expression of SNX1 inhibited the growth, migration, invasion and promoted the apoptosis and enhanced the sensitivity of GC cells to the chemotherapeutic drug 5-Fluorouracil (5-Fu) in vitro, while knockdown of SNX1 by short hairpin RNA (shRNA) significantly promoted the growth, migration, invasion and reduced the apoptosis and the sensitivity of GC cells to 5-Fu. SNX1 also showed to influence the levels of epithelial-mesenchymal transition markers including Vimentin, Snail, and E-cadherin in GC cells in vitro. Taken together, we propose here that SNX1 serves as a tumor suppressor and prognostic marker that reduces tumor cell malignancy for GC.
