IntroductionCervical cancer remains a significant challenge in oncology with an escalating demand for novel therapeutic strategies that can navigate the complexities of its pathophysiology. This study elucidated the antineoplastic effects of cryptotanshinone, a derivative of danshen (Salvia miltiorrhiza), a herb widely utilized in traditional Chinese medicine practices.MethodsEmploying a comprehensive multi-omics approach, including transcriptomic, proteomic, and bioinformatics analyses, we investigated the potential effects of cryptotanshinone on cervical cancer through data mining and computational analysis.Results and DiscussionOur results demonstrated that the potential of cryptotanshinone to disrupted cancer cell proliferation and induced apoptosis may be ascribed to its modulation of gene expression and interaction with specific protein networks. Furthermore, network pharmacology and pathway enrichment analyses identified critical hubs and signaling pathways, suggesting a multi-targeted mechanism of action. Furthermore, the establishment of a prognostic model, which is founded upon differentially expressed genes linked to cryptotanshinone treatment, underscores its promising role as both a prognostic biomarker and a therapeutic agent. These insights pave the way for the integration of cryptotanshinone into therapeutic regimens, offering a promising avenue for enhancing the efficacy of cervical cancer treatment and patient outcomes.

IntroductionAn interspecific interaction is an important reason for the yield advantage of interspecific cropping compared with sole cropping, and the relative sowing time of species is an important factor affecting interspecific competitiveness. Our purpose was to explore the effects of different relative sowing times on the interspecific competition-recovery phenomenon in wheat and maize intercropping systems.MethodsThree planting methods (wheat/maize intercropping, wheat and maize sole cropping) and different relative sowing times of wheat were used to carry out field experiments over two years. Sequential harvest of subplots was performed between 3 and 6 times, and the biomass data were fitted to logistic growth model.ResultsDelaying the sowing time of wheat reduced the wheat yield, biomass and nutrient acquisition and increased those of maize, but wheat still had an intercropping advantage during the co-growth period. At the same time, the nutrient acquisition of maize was still inhibited, but its recovery growth advanced. Changing the relative sowing time of wheat significantly changed the maximum instantaneous growth rates of wheat and maize. Delaying the relative sowing time of wheat significantly reduced its maximum instantaneous growth rate, while enhancing that of maize, leading to a balanced mutual benefit.ConclusionsDelaying the sowing time of wheat to the same sowing time as maize will change wheat/maize intercropping from asymmetrical interspecific facilitation to symmetrical interspecific facilitation. However, in this case, intercropped wheat still had an interspecific competitive advantage in the co-growth stage, and intercropped maize still underwent a competition-recovery process.

The Manila Clam (Ruditapes Philippinarum) is one of the most economically significant marine bivalves along the China coast. In recent years, due to the escalating scale of aquaculture, the artificial activities of ''cross-regional introduction'' have to some extent exerted an influence on the genetic diversity and structure of clam larvae, leading to an ongoing occurrence of genetically mixed clam larvae populations and ambiguous genetic foundation of germplasm resources in China. Therefore, there is an imperative need to gain profound insights into the lineage, genetic structure, and assess the adaptability potential of clams originating from both northern and southern regions. We use Specific Locus Amplified Framents technology to simplify genome sequencing of wild clams along the coast of China. Through genetic diversity, genetic structure, gene flow, and selection elimination analysis, the results show that clams can be divided into distinct north-south lineages, with Wafangdian in Liaoning (RPLW), Zhangzi Island in Liaoning (RPLZ), and Laizhou in Shandong (RPSL) comprise the northern lineage, while Lianjiang in Fujian (RPFL) constitutes the southern lineage. The artificial relocation of clams in China has resulted in a blended population structure among local clams. The genetic functional differences between clams from the north and south are primarily manifest in energy metabolism, and the adaptive evolution of clams is mainly associated with temperature. This integrative research not only furnishes valuable insights into the genetic structure of clam populations, thereby contributing to the advancement of natural conservation and genetic breeding efforts for clams but also establishes the groundwork for a comprehensive exploration of clam adaptive evolution.

ObjectiveTo analyze the effect of fasting blood glucose levels after reperfusion of acute large vessel occlusion (ALVO) on patient functional prognosis.MethodsRetrospectively included ALVO patients from three large stroke centers in China, all of whom achieved vascular reperfusion after mechanical thrombectomy or bridging thrombolysis. The prognosis scores of all patients at 90 ± 7 days post-recanalization were categorized into a good prognosis group (mRS 0–2) and a poor prognosis group (mRS 3–6). The relationship between mean blood glucose levels at 72 h post-recanalization and prognosis was explored using multivariable logistic regression analysis. Then we measured the area under the ROC curve for all factors to assess their predictive performance.Results(1) Totally 2,056 patients were included in the study, with 1,488 males and 568 females. In the digital era, the significance of cryptographic algorithms has grown significantly within the realm of cybersecurity. This research presents an innovative approach to image encryption that eliminates the security limitations of the conventional one-dimensional logistic mapping. This approach relies on an enhanced two-dimensional logistic-fraction hybrid chaotic mapping (2D-LFHCM) and deoxyribonucleic acid (DNA) computing. Initially, the improved 2D-LFHCM is utilized to effectively scramble the image by incorporating chaotic sequences. Then, two novel algebraic DNA computing rules are introduced to enhance diffusion encryption. Experimental findings show that this approach offers superior security performance, even with renowned attacks.

The current lack of standardized testing methods to assess the binding isotherms of ions in cement and concrete research leads to uncontrolled variability in these results. In this study, an open-source and low-cost apparatus, named OpenHW3, is proposed to accurately measure the binding isotherms of ions in various cementitious material systems. OpenHW3 provides two main options, a temperature-controlled orbital shaker, as well as an option to retrofit a commercial orbital shaker for temperature control. The effectiveness of these device options is validated via comparison with conventional binding isotherms experiments. The binding isotherm results were comparable to conventional Waterbath shakers, while providing more reliable results compared to horizontal commercial shakers. It also provided accurate temperature control between 25 °C and 75 °C. The results here are critical for allowing open access to scientific equipment, and providing high-quality binding isotherm data for reliable service life models of urban infrastructure assets throughout the world.

ObjectiveTo investigate the correlation between the swelling rate of brain volume within the first 48 h after aneurysmal subarachnoid hemorrhage and the subsequent development of delayed cerebral ischemia.MethodsA retrospective analysis was conducted on patients with spontaneous aneurysmal subarachnoid hemorrhage admitted to the Neurosurgery Intensive Care Unit of the First Affiliated Hospital of Chongqing Medical University between January 2020 and January 2023. The clinical data, treatment outcomes, and imaging data were analyzed. Brain volume was evaluated using 3D-Slicer software at two time points post-hemorrhage: within the first 24 h and between 24 and 48 h. The swelling rate of brain volume was defined as the ratio of the absolute difference between two measurements to the smaller of values. Patients were categorized into two groups based on established diagnostic criteria of delayed cerebral ischemia. Univariate and multivariate logistic regression analyses were performed to identify factors influencing delayed cerebral ischemia.ResultsA total of 140 patients were enrolled in this study. 46 patients experienced delayed cerebral ischemia after bleeding. The swelling rate of brain volume was larger in the DCI group (10.66 ± 8.45) compared to the non-DCI group (3.59 ± 2.62), which showed a statistically significant difference. Additionally, advanced age, smoking history, history of hypertension, loss of consciousness, poor Hunt-Hess grade, high mFisher score, brain volume within 24 h, and IVH were also statistically different between the two groups. Multivariate logistic regression analysis revealed that the swelling rate of brain volume was an independent risk factor for DCI with adjusting the advanced age, smoking history, history of hypertension, poor Hunt-Hess grade, high mFisher score, brain volume within 24 h, and IVH.ConclusionBrain volume significantly increased in patients with aneurysmal subarachnoid hemorrhage during the early phase (within 48 h post-onset). The larger swelling rate of brain volume is an independent risk factor for the development of delayed cerebral ischemia, and it may hold significant predictive value for the incidence of delayed cerebral ischemia.

Objective: Hashimoto’s thyroiditis is an inflammatory disease, and research suggests that a low-carbohydrate diet may have potential anti-inflammatory effects. This study aims to utilize Dixon-T2-weighted imaging (WI) sequence for a semi-quantitative assessment of the impact of a low-carbohydrate diet on the degree of thyroid inflammation in patients with Hashimoto’s thyroiditis. Methods: Forty patients with Hashimoto’s thyroiditis were recruited for this study and randomly divided into two groups: one with a normal diet and the other with a low-carbohydrate diet. Antibodies against thyroid peroxidase (TPOAb) and thyroglobulin (TgAb) were measured for all participants. Additionally, thyroid water content was semi-quantitatively measured using Dixon-T2WI. The same tests and measurements were repeated for all participants after 6 months. Results: After 6 months of a low-carbohydrate diet, patients with Hashimoto’s thyroiditis showed a significant reduction in thyroid water content (94.84 ± 1.57% vs 93.07 ± 2.05%, P < 0.05). Concurrently, a decrease was observed in levels of TPOAb and TgAb (TPOAb: 211.30 (92.63–614.62) vs 89.45 (15.9–215.67); TgAb: 17.05 (1.47–81.64) vs 4.1 (0.51–19.42), P < 0.05). In contrast, there were no significant differences in thyroid water content or TPOAb and TgAb levels for patients with Hashimoto’s thyroiditis following a normal diet after 6 months (P < 0.05). Conclusion: Dixon-T2WI can quantitatively assess the degree of thyroid inflammation in patients with Hashimoto’s thyroiditis. Following a low-carbohydrate diet intervention, there is a significant reduction in thyroid water content and a decrease in levels of TPOAb and TgAb. These results suggest that a low-carbohydrate diet may help alleviate inflammation in patients with Hashimoto’s thyroiditis.

Microwave plasma torch (MPT) has the advantages of low power consumption, convenient operation, simple structure, etc. It can be used with mass spectrometer for rapid analysis of elements. Inductively coupled plasma mass spectrometer (ICP-MS) has many advantages, such as high sensitivity and wide range of analyzed elements, but its high gas consumption and power consumption of ion source make the detection and analysis cost high. In order to effectively reduce the cost of mass spectrometry for the detection and analysis of alkali metal elements, this study reported a self-made low power-consuming microwave plasma torch time-of-flight mass spectrometer (MPT-TOF MS), including a three-cone system consisting of an ion source, a sampling cone, an interception cone, a super-interceptor cone, an ion transport lens set, a time-of-flight mass analyzer, a sample injection device, and a data acquisition system. The effects of five experimental factors, namely, maintenance gas flow rate, carrier gas flow rate, super-interception cone voltage, MPT torch flame position and microwave power, on the detection of alkali metal elements were investigated. The performance of the instrument was systematically investigated, covering the linear range, mass resolution, and isotopic accuracy of measurement. The results showed that the MPT-TOF MS has the advantages of low gas consumption (800 mL/min for argon maintenance gas, 400 mL/min for argon carrier gas, and 2 000 mL/min for nitrogen drying gas), low power consumption of the ion source (100 W), and long-time stable operation when working, and the linear range of the measurement of this device covers five orders of magnitude with high mass resolution. The detection limits of lithium, sodium, potassium, rubidium and cesium are 0.49, 3.05, 1.31, 0.74, 0.34 μg/L, respectively, which are better than the results of ICP-MS. The instrument was used for the rapid detection of lithium, sodium, potassium, rubidium and cesium alkali metal elements in the salt lake. Our study showed that the MPT-TOF-MS instrument can be used as an alternative to the elemental detection of ICP-MS, and can be further developed into an on-line analytical method.

Abstract Glioma, with its heterogeneous microenvironments and genetic subtypes, presents substantial challenges for treatment prediction and development. We integrated 3D bioprinting and multi-algorithm machine learning as a novel approach to enhance the assessment and understanding of glioma treatment responses and microenvironment characteristics. The bioprinted patient-derived glioma tissues successfully recapitulated molecular properties and drug responses of native tumors. We then developed GlioML, a machine learning workflow incorporating nine distinct algorithms and a weighted ensemble model that generated robust gene expression-based predictors, each reflecting the diverse action mechanisms of various compounds and drugs. The ensemble model superseded the performance of all individual algorithms across diverse in vitro systems, including sphere cultures, complex 3D bioprinted multicellular models, and 3D patient-derived tissues. By integrating bioprinting, the evaluative scope of the treatment expanded to T cell-related therapy and anti-angiogenesis targeted therapy. We identified promising compounds and drugs for glioma treatment and revealed distinct immunosuppressive or angiogenic myeloid-infiltrated tumor microenvironments. These insights pave the way for enhanced therapeutic development for glioma and potentially for other cancers, highlighting the broad application potential of this integrative and translational approach.

Abstract Background Neurodegenerative diseases are increasingly recognized for their association with oxidative stress, which leads to progressive dysfunction and loss of neurons, manifesting in cognitive and motor impairments. This study aimed to elucidate the neuroprotective role of peroxiredoxin II (Prx II) in counteracting oxidative stress-induced mitochondrial damage, a key pathological feature of neurodegeneration. Methods We investigated the impact of Prx II deficiency on endoplasmic reticulum stress and mitochondrial dysfunction using HT22 cell models with knocked down and overexpressed Prx II. We observed alcohol-treated HT22 cells using transmission electron microscopy and monitored changes in the length of mitochondria-associated endoplasmic reticulum membranes and their contact with endoplasmic reticulum mitochondria contact sites (EMCSs). Additionally, RNA sequencing and bioinformatic analysis were conducted to identify the role of Prx II in regulating mitochondrial transport and the formation of EMCSs. Results Our results indicated that Prx II preserves mitochondrial integrity by facilitating the formation of EMCSs, which are essential for maintaining mitochondrial Ca2+ homeostasis and preventing mitochondria-dependent apoptosis. Further, we identified a novel regulatory axis involving Prx II, the transcription factor ATF3, and miR-181b-5p, which collectively modulate the expression of Armcx3, a protein implicated in mitochondrial transport. Our findings underscore the significance of Prx II in protecting neuronal cells from alcohol-induced oxidative damage and suggest that modulating the Prx II-ATF3-miR-181b-5p pathway may offer a promising therapeutic strategy against neurodegenerative diseases. Conclusions This study not only expands our understanding of the cytoprotective mechanisms of Prx II but also offers necessary data for developing targeted interventions to bolster mitochondrial resilience in neurodegenerative conditions.

Objective To explore the potential classification of sleep quality in patients with maintenance hemodialysis and analyze the influencing factors of sleep quality in patients with different characteristics.Methods Patients with maintenance hemodialysis from two Grade 3A hospitals were investigated by using Athens Insomnia Scale, Family APGAR Index and Herth Hope Index. Background and Objectives: Ventilator-associated pneumonia (VAP) poses a significant threat to the clinical outcomes and hospital stays of mechanically ventilated patients, particularly those recovering from cardiac arrest. Given the already elevated mortality rates in cardiac arrest cases, the addition of VAP further diminishes the chances of survival. Consequently, a paramount focus on VAP prevention becomes imperative. This review endeavors to comprehensively delve into the nuances of VAP, specifically in patients requiring mechanical ventilation in post-cardiac arrest care. The overarching objectives encompass (I) exploring the etiology, risk factors, and pathophysiology of VAP, (II) delving into available diagnostic modalities, and (III) providing insights into the management options and recent treatment guidelines. Methods: A literature search was conducted using PubMed, MEDLINE, and Google Scholar databases for articles about VAP and Cardiac arrest. We used the MeSH terms “VAP”, “Cardiac arrest”, “postcardiac arrest syndrome”, and “postcardiac arrest syndrome”. The clinical presentation, diagnostic, and management strategies of VAP were summarized, and all authors reviewed the selection and decided which studies to include. Key Content and Findings: The incidence and mortality rates of VAP exhibit significant variability, yet a recurring pattern emerges, marked by prolonged hospitalization and exacerbated clinical outcomes. This pattern is attributed to the elevated incidence of drug-resistant infections and the delayed initiation of antimicrobial treatment. This review focuses on VAP, aiming to offer valuable insights into the efficient identification and management of this fatal complication in post-cardiac arrest patients. Conclusion: The prognosis for survival after cardiac arrest is already challenging, and the outlook becomes even more daunting when complicated by VAP. The timely diagnosis of VAP and initiation of antibiotics pose considerable challenges, primarily due to the invasive nature of obtaining high-quality samples and the time required for speciation and identification of antimicrobial sensitivity. The controversy surrounding prophylactic antibiotics persists, but promising new strategies have been proposed; however, they are still awaiting well-designed clinical trials.

The study explored the impact of floral strip width on the spider and carabid beetle communities in maize fields over two years. Three widths of floral strips (2 m, 4 m, and 6 m) were compared with maize-only control strips to evaluate species diversity and distribution. The results showed significant differences in both spider and carabid populations between floral and control strips, with 4 m and 6 m widths consistently harboring higher biodiversity. The results also showed distinct community clustering within floral strips in 2021, which became more cohesive by 2022. Further analysis validated significant community dissimilarities between different strip widths and controls, highlighting the ecological advantages of wider floral strips for enhancing natural enemy biodiversity. Spider activity density was notably higher in floral strips than in adjacent farmland, peaking at the edges of 4 m-wide strips and decreasing in 6 m-wide strips, with the lowest density in 2 m-wide strips. Carabid beetle activity density varied considerably with strip width and proximity to the edge, typically peaking at the edges of wider strips. Spiders were more responsive to strip width than carabid beetles. Based on these findings, we suggest using 4 m- or 6 m-wide floral strips to enhance biodiversity and natural pest control in agricultural landscapes; the floral strips narrower than 4 m (such as 2 m) could not support optimal biodiversity, as spiders and carabid beetles do not disperse far into the maize field, with spiders having dispersal distances of less than 3 m and carabid beetles less than 10 m. Vegetation characteristics significantly influenced spider and carabid communities, impacting species richness, diversity indices, and community structures across two study years. These insights highlight the necessity of thoughtfully designing floral strips to enhance biodiversity and natural pest control in agricultural landscapes.
