Amaranthus tricolor L. is a plant of significant botanical diversity and richness. In recent years, it has emerged as a versatile medicinal plant material, with numerous studies highlighting its considerable pharmacological effects. This study provides a comprehensive micromorphological and phytochemical characterization of A. tricolor. The study uses the Ciuley method, a rigorous and scientific approach that has been improved and modified by the Faculty of Pharmacy, University of Medicine and Pharmacy at Ho Chi Minh City, Vietnam. The results, based on A. tricolor collected in District 8, Ho Chi Minh City, Vietnam, were consistent with descriptions published in previous studies. The herbal powder contained distinctive microscopic constituents such as reticulate vessel elements, scalariform vessel elements, and spherical and cuboidal calcium oxalate crystals. The phytochemical characterization revealed the presence of various chemical groups in A. tricolor, including lipids, carotenoids, triterpenoids, flavonoids, tannins, saponins, organic acids, reducing agents, and polyuronic compounds. The macroscopic, microscopic, and chemical characteristics of A. tricolor are crucial for its medicinal use. Macroscopic studies ensure accurate botanical identification, while microscopic analysis reveals internal structures that improve quality control. Chemical characterization identifies active compounds responsible for the documented pharmacological effects. This comprehensive understanding enhances the efficacy, safety, and therapeutic potential of A. tricolor, supporting its optimal use in herbal medicine and paving the way for discovering new therapeutic agents. ARID1A, a component of the chromatin-remodeling complex SWI/SNF, is one of the most frequently mutated genes in human cancer. We sought to develop rational combination therapy to potentiate the efficacy of immune checkpoint blockade in ARID1A-deficient tumors. In a proteomic analysis of a data set from The Cancer Genomic Atlas, we found enhanced expression of Chk2, a DNA damage checkpoint kinase, in ARID1A-mutated/deficient tumors. Surprisingly, we found that ARID1A targets the nonchromatin substrate Chk2 for ubiquitination. Loss of ARID1A increased the Chk2 level through modulating autoubiquitination of the E3-ligase RNF8 and thereby reducing RNF8-mediated Chk2 degradation. Inhibition of the ATM/Chk2 DNA damage checkpoint axis led to replication stress and accumulation of cytosolic DNA, which subsequently activated the DNA sensor STING-mediated innate immune response in ARID1A-deficient tumors. As expected, tumors with mutation or low expression of both ARID1A and ATM/Chk2 exhibited increased tumor-infiltrating lymphocytes and were associated with longer patient survival. Notably, an ATM inhibitor selectively potentiated the efficacy of immune checkpoint blockade in ARID1A-depleted tumors but not in WT tumors. Objectives People with SSc often experience fatigue, which significantly affects daily life functioning and quality of life. We aimed to explore participants' experiences of a peer health–coached resilience-building energy management to enhance well-being (RENEW) intervention on symptoms and well-being and to use mixed methods to compare how SSc duration influenced the experiences of participants who had clinically significant fatigue improvement vs those who did not. Methods Semi-structured interviews were conducted with 21 participants from the parent clinical trial randomized to the RENEW intervention. Data were analysed using the rigorous and accelerated data reduction technique combined with thematic content analysis. A mixed methods approach used a joint display to identify themes related to the impact of SSc duration on fatigue improvement status. Participants were categorized into short/improvement, short/limited improvement, long/improvement, and long/limited improvement. Results Our team generated four themes: participant and peer health–coach relationship, physical and psychological well-being improvement, need for a tailored approach and easy program access through technology. Mixed methods analysis revealed that, regardless of SSc duration, participants with improved fatigue reported increasing self-awareness of SSc-related symptoms and learning coping strategies to manage fatigue. Participants in the short/improvement group reported preferences for slower pacing of the program and pairing with a coach with similar symptom severity. Participants in the long/limited improvement group sought SSc-specific symptom management information. Conclusion Incorporating peer health coaches and technology is beneficial for self-management interventions for people with SSc. Objective: Supported self‐management interventions for individuals with systemic sclerosis (SSc) are needed. We examined the effects of a 12‐week resilience‐building energy management program (called RENEW) for fatigue and other patient‐reported outcomes. Methods: Participants, who had physician‐diagnosed SSc, moderate to severe fatigue, and were ≥18 years old, were randomly assigned to RENEW or waitlist control in a 2:1 ratio. The RENEW intervention included an educational website/application plus nine virtual peer‐led health coaching sessions. The primary outcome was change in the Functional Assessment of Chronic Illness Therapy‐Fatigue scale. Secondary outcomes were change in Patient Reported Outcomes Measurement Information System measures of pain interference and depressive symptoms and Connor‐Davidson Resilience Scale. Outcomes were assessed at baseline, 6 weeks, and 12 weeks. Multiple imputation was conducted; linear mixed models were used to assess group differences. A three‐way interaction with group, time, and SSc duration was examined in each model. Results: Among 173 participants (mean ± SD age 54.5 ± 11.7 years; 93% female, 85% White), 47% had diffuse cutaneous SSc; 57% were ≤5 years from diagnosis. At 12 weeks, compared to controls, RENEW participants had a clinically meaningful fatigue improvement (β = −4.7; 95% confidence interval −6.7 to −2.7; P < 0.001) and improvement in all secondary outcomes. Among RENEW participants, individuals with shorter disease duration had greater improvements in fatigue at 12 weeks. Conclusion: An mHealth supported self‐management intervention improved fatigue and other outcomes, particularly in newly diagnosed patients. Heterogeneous energetic materials (EMs) contain microstructural defects such as voids, cracks, interfaces, and delaminated zones. Under shock loading, these defects offer potential sites for energy localization, i.e., hotspot formation. In a porous EM, the collapse of one void can generate propagating blast waves and hotspots that can influence the hotspot phenomena at neighboring voids. Such void–void interactions must be accounted for in predictive multi-scale models for the reactive response of a porous EM. To infuse such meso-scale phenomena into a multi-scale framework, a meso-informed ignition and growth model (MES-IG) has been developed, where the influence of void–void interactions is incorporated into the overall reaction rate through a function, f v − v . Previously, MES-IG was applied to predict the sensitivity and reactive response of EM, where f v − v was assumed to be a function of the overall sample porosity alone. This paper performs a deeper analysis to model the strong dependency of f v − v on other factors, such as void size and shock strength. The improved model for void–void interactions produces good agreement with direct numerical simulations of the HE microstructures and, thus, advances the predictive capability of multi-scale models of the shock response and sensitivity of EM. Head-to-head comparisons of multiple experimental observations and numerical simulations on a deconstructed plastic-bonded explosive consisting of an octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine crystal embedded in a polymeric binder with a 4 ns duration 20 GPa input shock are presented. Hot spots observed in high-resolution direct numerical simulations are compared with micro-scale shock-induced reactions visualized using nanosecond microscope imaging and optical pyrometry. Despite the challenges and limitations of both the experimental and simulation techniques, an agreement is obtained on many of the observed features of hot spot evolution, e.g., (1) the magnitude and time variation of temperatures in the hot spots, (2) the time to fully consume the crystals (∼100 ns) of size (100–300 μm) employed in this study, and (3) the locations of hot spot initiation and growth. Three different mechanisms of hot spot formation are indicated by simulations: (1) high-temperature hot spots formed by pore collapse, (2) lower temperature hot spots initiated at the polymer–crystal interface near corners and asperities, and (3) high-temperature reaction waves leading to fast consumption of the energetic crystal. This first attempt at a head-to-head comparison between experiments and simulations not only provides new insight but also highlights efforts needed to bring models and experiments into closer alignment, in particular, highlighting the importance of distinctly three-dimensional and multiple mechanisms of the hot spot ignition and growth. Tumor microenvironments present significant barriers to penetration by antibodies and immunoconjugates and are difficult to study in vitro. Cells cultured as monolayers typically exhibit less resistance to therapy than those grown in vivo. Therefore, it is important to develop an alternative research model that better represents in vivo tumors. We have developed a protocol to produce multicellular spheroids, a simple and more relevant model of in vivo tumors that allows for further investigations of the microenvironmental effects on drug penetration and tumor cell killing. The protocol is used to produce in vitro three-dimensional tumor spheroids from established human cancer cell lines and primary cancer cells isolated from patients without the use of any extracellular components. To study the ability of tumor-targeting immunoconjugates to penetrate these tumor spheroids in vitro, we have used an immunotoxin targeting mesothelin, a surface protein expressed in malignant mesotheliomas. This method for producing consistent, reproducible 3D spheroids may allow for improved testing of novel monoclonal antibodies and other agents for their ability to penetrate solid tumors for cancer therapy.

Meso-scale simulations of pressed energetic materials are performed using synthetic microstructures generated using deep feature representation, a deep convolutional neural network-based approach. Synthetic microstructures are shown to mimic real microstructures in the statistical representation of global and local features of micro-morphology for three different classes of pressed HMX with distinctive micro-structural characteristics. Direct numerical simulations of shock-loaded synthetic microstructures are performed to calculate the meso-scale reaction rates. For all three classes, the synthetic microstructures capture the effect of morphological uncertainties of real microstructures on the response to shock loading. The calculated reaction rates for different classes also compare well with those of the corresponding real microstructures. Thus, the article demonstrates that machine-generated ensembles of synthetic microstructures can be employed to derive structure–property–performance linkages of a wide class of real pressed energetic materials. The ability to manipulate the synthetic microstructures using deep learning-based approaches then provides an opportunity for material designers to develop and manufacture pressed energetic materials that can yield targeted performance.
