Abstract

This thesis addresses the trend analysis of Post-traumatic Stress Disorder (PTSD) prevalence across the different branches of armed services in the U.S military between FY2001 and FY2006, as well as the effects of deployment characteristics on the probability of being diagnosed with PTSD among the active duty service personnel in the different branches. On top of these, this study will also highlight the patterns of the comorbidity and treatment costs of PTSD across the different branches of armed services. The data used in the thesis are provided by TRICARE, the Department of Defense's (DoD) health care system and Defense Manpower Data Center (DMDC), to obtain the demographics, deployment characteristics (deployment location, deployment frequencies, deployment duration) and the inpatient and outpatient medical information and services rendered by physicians for all active duty service personnel whom were diagnosed with PTSD between FY2001 and FY2006. http://archive.org/details/economicnalysiso109453761 Singapore Navy author. Abstract

Genetic variants in the purinergic receptors P2RX4 and P2RX7 have been shown to affect susceptibility to multiple sclerosis (MS). In this study, we set out to evaluate whether rare coding variants of major effect could also be identified in these purinergic receptors. Sequencing analysis of P2RX4 and P2RX7 in 193 MS patients and 100 controls led to the identification of a rare three variant haplotype ( P2RX7 rs140915863:C>T [p.T205M], P2RX7 rs201921967:A>G [p.N361S], and P2RX4 rs765866317:G>A [p.G135S]) segregating with disease in a multi-incident family with six family members diagnosed with MS (logarithm of odds = 3.07). Functional analysis of this haplotype in HEK293 cells revealed impaired P2X7 surface expression ( P < 0.01), resulting in over 95% inhibition of adenosine triphosphate (ATP)-induced pore function ( P < 0.001) and a marked reduction in phagocytic ability ( P < 0.05). In addition, transfected cells showed 40% increased peak ATP-induced inward current ( P < 0.01), and a greater Ca2+ response to the P2X4 135S variant compared with wild type ( P < 0.0001). Our study nominates rare genetic variants in P2RX4 and P2RX7 as major genetic contributors to disease, further supporting a role for these purinergic receptors in MS and the disruption of transmembrane cation channels leading to impairment of phagocytosis as the pathological mechanisms of disease. Abstract

This study aims to examine the impacts of the inquiry learning strategies employed in a ‘Plastic Education Project’ on primary students' knowledge, beliefs and intended behaviour in Hong Kong. Student questionnaires and a test on plastic types were adopted for data collection. Results reveal that the inquiry learning strategies significantly improved students' knowledge of the types of plastic wastes and their corresponding beliefs. However, the strategies seemed not to change students' intended behaviour regarding plastic waste classification and management. Abstract

Background. A population-based prevalent cohort of 150 clinical definite multiple sclerosis (MS) cases (102 women; 48 men) ascertained on January 1,1977, Saskatoon, Saskatchewan, was found to have a familial rate of MS as 17.3%. Objectives. To determine the occurrence of familial MS cases and the frequency of MS among the biological relatives of the study cohort. Methods. The search for new familial cases MS affected relatives continued for 35 years until 2012. The natural history of the disease of sporadic cases is compared with that of the familial cases. SPSS V19 and Kaplan-Meier survival analysis were used for data analysis. Results. Of the 150 unrelated MS patients, 49 cases (32.7%) (36 women and 13 men) were reported of having at least one family member with MS. There were a total of 86 affected relatives, 26 (30.2%) first-degree relatives, 15 (17.4%) second-degree relatives, 20 (23.3%) third-degree relatives, and 25 (29.1%) distant relatives. The average age of MS onset for men with sporadic MS was 33.9 (SD = 10) years and 27.6 (SD = 8.4) years for familial cases and 29.3 (SD = 8.3) years and 26.8 (SD = 8.5) years for women. Conclusion. This 35-year longitudinal natural history study reveals a high frequency of cases with family members developing MS and supports a genetic influence in the etiology of MS. [ABSTRACT FROM AUTHOR]

Abstract

The article presents a study on the demographic and clinical characteristics of multiple sclerosis (MS) among aboriginals in British Columbia (BC). The study involved patient information from the linked BC-wide MS clinical and genetics databases, and the identification of aboriginals through the database linkage augmented by physician and nurse recall. The result revealed a more rapid MS progression in aboriginals, as compared to non-aboriginals.

Background: Genetic and environmental factors have important roles in multiple sclerosis (MS) susceptibility. A clear parent of origin effect has been shown in several populations, perhaps resulting from factors operating during gestation. Preterm birth (birth at less than 37 weeks gestational age) has been shown to result in long-term health problems, including impaired neurological development. Here, in a population-based cohort, we investigate whether preterm birth increases the risk to subsequently develop MS. Methods: We identified 6585 MS index cases and 2509 spousal controls with preterm birth information from the Canadian Collaborative Project on Genetic Susceptibility to MS. Rates of individuals born preterm were compared for index cases and controls. Results: There were no significant differences between cases and controls with respect to preterm births. 370 (5.6%) MS index cases and 130 (5.2%) spousal controls were born preterm, p = 0.41. Conclusion: Preterm birth does not appear to contribute to MS aetiology. Other factors involved in foetal and early development need to be explored to elucidate the mechanism of the increased risk conferred by the apparent maternal effect. [ABSTRACT FROM AUTHOR]

Abstract

OBJECTIVE: To conduct a prospective assessment of pregnancy on women with multiple sclerosis (MS), focusing on pregnancy outcome and relapses during gestation and up to 6 months after delivery. DESIGN: Expected numbers of relapses were based on data for (1) "self-controls": the mothers ("cases") themselves prior to becoming pregnant and (2) "matched controls": female patients with MS "matched" to the mothers for year of birth, age of MS onset, MS type, MS course, and initial MS symptom(s). SETTING: Cases and controls were identified from an ambulatory care MS clinic that serves the province of British Columbia, Canada. PATIENTS OR OTHER PARTICIPANTS: Women with a diagnosis of MS who attended the MS clinic during 1982 through 1986 and subsequently became pregnant during 1982 through 1989 inclusive were included in this study as cases. Matched controls were women with MS who attended the MS clinic during the same period but did not become pregnant. RESULTS: No significant increase in relapse rate was found for cases during the first two trimesters of gestation. The number of relapses was significantly less than expected during the third trimester compared with matched controls (ϰ2=6.80, df=1, P&lt;.02), but not compared with self-controls (ϰ2=3.39, df=1, P>.05). The observed number of relapses for the 6 months after delivery did not differ significantly from expected (self-controls:ϰ2=2.84, df=2, P>.05; matched controls: ϰ2=1.76, df=2, P>.05). CONCLUSION: These data suggest that neither pregnancy nor the 6-month period after delivery is a risk factor for relapse in MS. They are consistent with previous observations that, in the long term, pregnancy does not influence subsequent MS disability.

91. Local parametric poisson models for fisheries data

Abstract

Poisson process is a common model for count data. However, a global Poisson model is inadequate for sparse data such as the marked salmon recovery data that have huge extraneous variations and noise. An empirical Bayes model, which enables information to be aggregated to overcome the lack of information from data in individual cells, is thus developed to handle these data. The method fits a local parametric Poisson model to describe the variation at each sampling period and incorporates this approach with a conventional local smoothing technique to remove noise. Finally, the overdispersion relative to the Poisson model is modelled by mixing these locally smoothed, Poisson models in an appropriate way. This method is then applied to the marked salmon data to obtain the overall patterns and the corresponding credibility intervals for the underlying trend in the data.

Abstract

Multiple sclerosis (MS) is a prevalent neurological disease of complex etiology. Here, we describe the characterization of a multi-incident MS family that nominated a rare missense variant (p.G420D) in plasminogen (PLG) as a putative genetic risk factor for MS. Genotyping of PLG p.G420D (rs139071351) in 2160 MS patients, and 886 controls from Canada, identified 10 additional probands, two sporadic patients and one control with the variant. Segregation in families harboring the rs139071351 variant, identified p.G420D in 26 out of 30 family members diagnosed with MS, 14 unaffected parents, and 12 out of 30 family members not diagnosed with disease. Despite considerably reduced penetrance, linkage analysis supports cosegregation of PLG p.G420D and disease. Genotyping of PLG p.G420D in 14446 patients, and 8797 controls from Canada, France, Spain, Germany, Belgium, and Austria failed to identify significant association with disease (P = 0.117), despite an overall higher prevalence in patients (OR = 1.32; 95% CI = 0.93-1.87). To assess whether additional rare variants have an effect on MS risk, we sequenced PLG in 293 probands, and genotyped all rare variants in cases and controls. This analysis identified nine rare missense variants, and although three of them were exclusively observed in MS patients, segregation does not support pathogenicity. PLG is a plausible biological candidate for MS owing to its involvement in immune system response, blood-brain barrier permeability, and myelin degradation. Moreover, components of its activation cascade have been shown to present increased activity or expression in MS patients compared to controls; further studies are needed to clarify whether PLG is involved in MS susceptibility.

Abstract

In recent years, STEM learning has become a new education initiative worldwide. However, little research has considered the needs of students with Intellectual Disabilities (ID) in this initiative. Believing that individuals with disabilities should be evaluated and defined by their capacity, strengths, and broad range of interests and abilities, this research investigated this less-explored perspective in STEM learning, namely supporting teachers providing STEM learning for ID students. Four teachers in two special schools for children with intellectual disabilities worked collaboratively with each other in their schools to plan and implement STEM learning. Peer coaching was recommended to the teachers in order to improve their planning of STEM learning and their teaching practices for teachers’ professional development (TPD). The qualitative research methodology was used, and detailed analysis of teachers’ pre- and post-TPD interviews and reflections to identify good practices that helped ID students accomplish the tasks and disparities that influenced peer coaching was performed. While challenging, with support from peers and due considerations of the special learning needs of ID students, this research provides useful insights for teachers to support ID students in STEM learning, including the use of technology in the STEM learning design, the consideration of inquiry learning based on students’ abilities in implementation, and the focus on teachers’ disparity and school involvement with peer coaching.
