Organic afterglow is a fascinating phenomenon with exceptional applications. However, it encounters challenges such as low intensity and efficiency, and typically requires UV-light excitation and facile intersystem crossing (ISC) due to its spin-forbidden nature. Here, we develop a novel strategy that bypasses the conventional ISC pathway by promoting singlet-triplet transition through the synergistic effects of the intra/intermolecular heavy-atom effect in aromatic crystals, enabling the direct population of triplet excited states from the ground state. The resulting materials exhibit a bright organic afterglow with a remarkably enhanced quantum efficiency of up to 5.81%, and a significantly increased organic afterglow lifetime of up to 157 microseconds under visible light. Moreover, given the high-efficiency visible-light excitable organic afterglow emission, the potential application is demonstrated in lifetime-resolved, color-encoded, and excitation wavelength-dependent pattern encryption. This work demonstrates the importance of the direct population method in enhancing the organic afterglow performance and red-shifting the excitation wavelength, and provides crucial insights for advancing organic optoelectronic technologies that involve triplet states.

Accurate delineation of individual agricultural plots, the foundational units for agriculture-based activities, is crucial for effective government oversight of agricultural productivity and land utilization. To improve the accuracy of plot segmentation in high-resolution remote sensing images, the paper collects GF-2 satellite remote sensing images, uses ArcGIS10.3.1 software to establish datasets, and builds UNet, SegNet, DeeplabV3+, and TransUNet neural network frameworks, respectively, for experimental analysis. Then, the TransUNet network with the best segmentation effects is optimized in both the residual module and the skip connection to further improve its performance for plot segmentation in high-resolution remote sensing images. This article introduces Deformable ConvNets in the residual module to improve the original ResNet50 feature extraction network and combines the convolutional block attention module (CBAM) at the skip connection to calculate and improve the skip connection steps. Experimental results indicate that the optimized remote sensing plot segmentation algorithm based on the TransUNet network achieves an Accuracy of 86.02%, a Recall of 83.32%, an F1-score of 84.67%, and an Intersection over Union (IOU) of 86.90%. Compared to the original TransUNet network for remote sensing land parcel segmentation, whose F1-S is 81.94% and whose IoU is 69.41%, the optimized TransUNet network has significantly improved the performance of remote sensing land parcel segmentation, which verifies the effectiveness and reliability of the plot segmentation algorithm.

The integration of sensory information from different modalities, such as touch and vision, is essential for organisms to perform behavioral functions such as decision-making, learning, and memory. Artificial implementation of human multi-sensory perception using electronic supports is of great significance for achieving efficient human–machine interaction. Thanks to their structural and functional similarity with biological synapses, memristors are emerging as promising nanodevices for developing artificial neuromorphic perception. Memristive devices can sense multidimensional signals including light, pressure, and sound. Their in-sensor computing architecture represents an ideal platform for efficient multimodal perception. We review recent progress in multimodal memristive technology and its application to neuromorphic perception of complex stimuli carrying visual, olfactory, auditory, and tactile information. At the device level, the operation model and undergoing mechanism have also been introduced. Finally, we discuss the challenges and prospects associated with this rapidly progressing field of research.

BackgroundFocal adhesion serves as a bridge between tumour cells and the extracellular matrix (ECM) and has multiple roles in tumour invasion, migration, and therapeutic resistance. However, studies on focal adhesion-related genes (FARGs) in head and neck squamous cell carcinoma (HNSCC) are limited.MethodsData on HNSCC samples were obtained from The Cancer Genome Atlas and GSE41613 datasets, and 199 FARGs were obtained from the Molecular Signatures database. The integrated datasets’ dimensions were reduced by the use of cluster analysis, which was also used to classify patients with HNSCC into subclusters. A FARG signature model was developed and utilized to calculate each patient’s risk score using least extreme shrinkage and selection operator regression analysis. The risk score was done to quantify the subgroups of all patients. We evaluated the model’s value for prognostic prediction, immune infiltration status, and therapeutic response in HNSCC. Preliminary molecular and biological experiments were performed to verify these results.ResultsTwo different HNSCC molecular subtypes were identified according to FARGs, and patients with C2 had a shorter overall survival (OS) than those with C1. We constructed an FARG signature comprising nine genes. We constructed a FARG signature consisting of nine genes. Patients with higher risk scores calculated from the FARG signature had a lower OS, and the FARG signature was considered an independent prognostic factor for HNSCC in univariate and multivariate analyses. FARGs are associated with immune cell invasion, gene mutation status, and chemosensitivity. Finally, we observed an abnormal overexpression of MAPK9 in HNSCC tissues, and MAPK9 knockdown greatly impeded the proliferation, migration, and invasion of HNSCC cells.ConclusionThe FARG signature can provide reliable prognostic prediction for patients with HNSCC. Apart from that, the genes in this model were related to immune invasion, gene mutation status, and chemosensitivity, which may provide new ideas for targeted therapies for HNSCC.

Genipin is a natural crosslinker that improves the functional properties of proteins by modifying its structures. This study aimed to investigate the effects of sonication on the emulsifying properties of different genipin concentration-induced myofibrillar protein (MP) cross-linking. The structural characteristics, solubility, emulsifying properties, and rheological properties of genipin-induced MP crosslinking without sonication (Native), sonication before crosslinking (UMP), and sonication after crosslinking (MPU) treatments were determined, and the interaction between genipin and MP were estimated by molecular docking. The results demonstrated that hydrogen bond might be the main forces for genipin binding to the MP, and 0.5 μM/mg genipin was a desirable concentration for protein cross-linking to improve MP emulsion stability. Ultrasound treatment before and after crosslinking were better than Native treatment to improve the emulsifying stability index (ESI) of MP. Among the three treatment groups at the 0.5 μM/mg genipin treatment, the MPU treatment group showed the smallest size, most uniform protein particle distribution, and the highest ESI (59.89%). Additionally, the highest α-helix (41.96%) in the MPU + G5 group may be conducive to the formation of a stable and multilayer oil–water interface. Furthermore, the free groups, solubility, and protein exposure extent of the MPU groups were higher than those of UMP and Native groups. Therefore, this work suggests that the treatment of cross-linking followed by ultrasound (MPU) could be a desirable approach for improving the emulsifying stability of MP.

Abstract Objective This study aimed to establish a deep learning method based on convolutional networks for the preliminary study of the pathological diagnosis of prosthetic joint infections (PJI). Methods We enrolled 20 revision patients after joint replacement from the Department of Orthopedics, the First Medical Center, General Hospital of the People's Liberation Army, from January 2021 to January 2022 (10 of whom were confirmed to be infected against 2018 ICM criteria, and the remaining 10 were verified to be non-infected), and classified high-power field images according to 2018 ICM criteria. Then, we inputted 576 positive images and 576 negative images into a neural network by employing a resNET model, used to select 461 positive images and 461 negative images as training sets, 57 positive images and 31 negative images as internal verification sets, 115 positive images and 115 negative images as external test sets. Results The resNET model classification was used to analyze the pathological sections of PJI patients under high magnification fields. The results of internal validation set showed a positive accuracy of 96.49%, a negative accuracy of 87.09%, an average accuracy of 93.22%, an average recall rate 96.49%, and an F1 of 0.9482. The accuracy of external test results was 97.39% positive, 93.04% negative, the average accuracy of external test set was 93.33%, the average recall rate was 97.39%, with an F1 of 0.9482. The AUC area of the intelligent image-reading diagnosis system was 0.8136. Conclusions This study used the convolutional neural network deep learning to identify high-magnification images from pathological sections of soft tissues around joints, against the diagnostic criteria for acute infection, and a high precision and a high recall rate were accomplished. The results of this technique confirmed that better results could be achieved by comparing the new method with the standard strategies in terms of diagnostic accuracy. Continuous upgrading of extended training sets is needed to improve the diagnostic accuracy of the convolutional network deep learning before it is applied to clinical practice.

Abstract Background This study analyzed regional corneal thickness remodeling, biomechanical properties, and visual outcomes after femtosecond laser-assisted in situ keratomileusis combined with intraoperative accelerated corneal collagen crosslinking (LASIK Xtra) for myopia. Methods This retrospective study analyzed 21 consecutive patients (18 women, three men; 42 eyes) who were treated with LASIK Xtra. All treatments were performed with ultraviolet-A (energy, 2.7 J/cm2; irradiance, 30 mW/cm2), using continuous (90 s) illumination. Postoperative values of corneal biometrics and visual outcomes were compared with preoperative values. Corneal thickness changes were evaluated using anterior segment optical coherence tomography. All patients were followed up for 12-month postoperatively. Preoperative and postoperative data were compared statistically using the paired t-test for normally distributed parameters and the Wilcoxon rank-sum test and Friedman analysis of variance with Bonferroni correction for non-normally distributed data. Introduction: Tertiary hyperparathyroidism (THPT) and vitamin D deficiency are commonly seen in kidney transplant recipients, which may result in persistently elevated fibroblast growth factor 23 (FGF23) level after transplantation and decreased graft survival. The aim of this study is to evaluate the effect of vitamin D supplementation on THPT, FGF23-alpha Klotho (KLA) axis and cardiovascular complications after transplantation. Materials and methods: Two hundred nine kidney transplant recipients were included and further divided into treated and untreated groups depending on whether they received vitamin D supplementation. We tracked the state of THPT, bone metabolism and FGF23–KLA axis within 12 months posttransplant and explored the predictors and risk factors for intact FGF23 levels, KLA levels, THPT and cardiovascular complications in recipients. Results: Vitamin D supplementation significantly improved FGF23 resistance, THPT and high bone turnover status, preserved better graft function and prevented coronary calcification in the treated group compared to the untreated group at month 12. The absence of vitamin D supplementation was an independent risk factor for THPT and a predictor for intact FGF23 and KLA levels at month 12. Age and vitamin D deficiency were independent risk factors for coronary calcification in recipients at month 12. Conclusion: Vitamin D supplementation effectively improved THPT, FGF23 resistance and bone metabolism, preserved graft function and prevented coronary calcification after transplantation.

Abstract Purpose The combination of lisinopril and amlodipine has a marked additional effect on blood pressure and fewer side effects than individual monotherapy. This study was conducted to compare the pharmacokinetic parameters and evaluate the bioequivalence between two Lisinopril/amlodipine tablets in healthy Chinese subjects. Methods A single center, randomized, open-label, single-dose, two-period crossover bioequivalence study was designed in healthy Chinese subjects under both fasting and fed conditions. Blood samples were collected before drug administration and at 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 24, 36, 48, 72, 96, 144, 168 h after administration. Liquid chromatography-tandem mass spectrometry (LC–MS/MS) was applied to determine the plasma concentration of lisinopril and amlodipine. Maximum concentration (Cmax) and area under the concentration–time curve (AUC) were used to evaluate bioequivalence. Adverse events were recorded. Results Ninety-two healthy subjects were enrolled, and 75 completed the study. The 90% confidence intervals (CIs) of the ratio of geometric means (GMRs) of Cmax, AUC0-t, and AUC0-∞ of lisinopril and amlodipine under both fasting and fed conditions fell within the conventional bioequivalence criteria of 0.80–1.25. A high-fat meal appeared to decrease the Cmax and AUC of lisinopril. No severe adverse events were observed. Conclusion The trial demonstrated that the test and the reference lisinopril/amlodipine tablets were bioequivalent and well tolerated in Chinese people under fasting and fed conditions. BackgroundNeoadjuvant immunotherapy with anti-PD-1 was proved promising in resectable non-small cell lung cancer (NSCLC). Immune-related adverse events (irAEs) have been preliminarily implicated their association with treatment efficacy. Here we elucidated the early onset of irAEs associated with better clinical outcomes in a prospective study (Renaissance study).MethodsWe conducted the prospective study of NSCLC patients treated by neoadjuvant Toripalimab (240mg, every 3 weeks) plus double platinum-based chemotherapy from December 2020 to March 2022 at Peking University Cancer Hospital. Patients were enrolled if they have resectable IIB-IIIB NSCLC without EGFR/ALK mutation. Data were analyzed to explore the relationship between clinical outcome and irAEs after neoadjuvant treatment. A multidisciplinary team including physicians, surgeons, and radiologists, confirmed the irAEs according to the clinical manifestation. The relationship between irAEs and pathological outcomes was analyzed. The Renaissance study was approved by the Peking University Ethic board (2020YJZ58) and registered at https://clinicaltrials.gov/ as NCT04606303.ResultsFifty-five consecutive patients were enrolled with a male-to-female ratio of 10:1, the median age was 62 years old (IQR: 45-76), of which 44 patients (80%) were diagnosed with squamous cell carcinoma. Forty-eight of 55 patients finally received thoracic surgery with a median preoperative waiting time of 67 days (IQR 39-113 days). Pathological results demonstrated that 31 (64.6%) patients achieved major pathological response (MPR) and 24 (50.0%) achieved complete pathological response (pCR). Among 48 patients who received R0 resection, immunotherapy-related thyroid dysfunction, rash/pruritus and enteritis occurred in 11 patients (22.9%), 7 patients (14.6%), and 1 patient (2.1%), respectively. Six patients (54.5%) with thyroid dysfunction achieved MPR with 5 (45.5%) achieved pCR, and a median time to onset was 45 days (IQR 21-91 days). Six patients (85.7%) with rash or pruritus achieved MPR and 5 patients (71.4%) achieved pCR, with median time to onset being 8 days (IQR 6-29 days). Furthermore, irAEs had no significant influence on operation time (170.6 min vs 165.7 min, P=0.775), intraoperative blood loss (67.4 mL vs 64.3 mL, P=0.831) and preoperative waiting time (93 days vs 97 days, P=0.630) when comparing with patients without irAEs (Figure 1).Figure 1Comparison of operation time (A), intraoperative blood loss (B), and preoperative waiting time (C) between “with irAEs” and “without irAEs”.ConclusionThe immunotherapy-related rash is potentially associated with pathological outcomes in NSCLC patients after neoadjuvant chemo-immunotherapy, suggesting easy-to-find irAEs, such as rash, can be used as indicators to predict response to neoadjuvant chemo-immunotherapy.Clinical trial registrationclinicaltrials.gov/, identifier NCT04606303.

Abstract Auditory motion perception is one crucial capability to decode and discriminate the spatiotemporal information for neuromorphic auditory systems. Doppler frequency‐shift feature and interaural time difference (ITD) are two fundamental cues of auditory information processing. In this work, the functions of azimuth detection and velocity detection, as the typical auditory motion perception, are demonstrated in a WOx‐based memristive synapse. The WOx memristor presents both the volatile mode (M1) and semi‐nonvolatile mode (M2), which are capable of implementing the high‐pass filtering and processing the spike trains with a relative timing and frequency shift. In particular, the Doppler frequency‐shift information processing for velocity detection is emulated in the WOx memristor based auditory system for the first time, which relies on a scheme of triplet spike‐timing‐dependent‐plasticity in the memristor. These results provide new opportunities for the mimicry of auditory motion perception and enable the auditory sensory system to be applied in future neuromorphic sensing.

Background: This study aimed to analyze clinical and multimodal imaging characteristics of acute macular neuroretinopathy (AMN) post-recent severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Methods: Retrospective observational study. Medical records and multimodal imaging of 12 AMN eyes of eight patients (six female and two male) with recent SARS-CoV-2 infection were retrospectively analyzed. Results: Four patients (50%) presented with bilateral AMN. Fundus ophthalmoscopy revealed a reddish-brown lesion around the macula, and two eyes had cotton-wool spots at the posterior pole. Three eyes showed mild hypo-autofluorescence. All FFA images (7 eyes) showed no abnormal signs. On OCT scans, all eyes showed outer nuclear layer (ONL) thinning, 8 eyes (66.7%) showed ONL hyperreflectivity, 5 eyes (41.7%) showed outer plexiform layer (OPL) hyperreflectivity, 8 eyes (66.7%) showed interdigitation zone (IZ) disruption, 11 eyes (91.6%) showed ellipsoid zone (EZ) disruption, 2 eyes (16.7%) showed cotton-wool spots and inner plexiform layer (IPL) hyperreflectivity, 1 eye (8.3%) had intraretinal cyst and 1 eye (8.3%) had inner nuclear layer (INL) thinning. Persistent scotoma, ONL hyperreflectivity and IZ/EZ disruption as well as recovery of OPL hyperreflectivity were reported after follow-up in three cases. Conclusions: AMN post-SARS-CoV-2 mostly affected young females and could present unilaterally or bilaterally. Dark lesions on IR reflectance and outer retinal hyperreflectivity on OCT are useful in diagnosing AMN. OPL/ONL hyperreflectivity on OCT could disappear after follow-up, but ONL thinning and IZ/EZ could persist.

Microfluidic devices with a free-standing structure were printed directly on polymer films using the functional materials that form interconnected pores. The printed devices can transport fluids by capillary action in the same fashion as paper-based microfluidic devices, and they can handle much smaller sample volumes than typical paper-based devices. Detection of glucose was performed using both colorimetric and electrochemical methods, and the observed limits of detection (LOD) were similar to those obtained with paper-based microfluidic devices under comparable testing conditions. It is demonstrated that printed microfluidic devices can be fabricated using printing processes that are suitable for high-volume and low-cost production and that the integration of microfluidic channels with electrodes is straightforward with printing. Several materials that are printable and form interconnected pores are presented.

Molecular hydrogen (H2) is a colorless, odorless, and tasteless gas which displays non-toxic features at high concentrations. H2 can alleviate oxidative damage, reduce inflammatory reactions and inhibit apoptosis cascades, thereby inducing protective and repairing effects on cells. H2 can be transported into the body in the form of H2 gas, hydrogen-rich water (HRW), hydrogen-rich saline (HRS) or H2 produced by intestinal bacteria. Accumulating evidence suggest that H2 is protective against multiple ophthalmic diseases, including cataracts, dry eye disease, diabetic retinopathy (DR) and other fields. In particular, H2 has been tested in the treatment of dry eye disease and corneal endothelial injury in clinical practice. This medical gas has brought hope to patients suffering from blindness. Although H2 has demonstrated promising therapeutic potentials and broad application prospects, further large-scale studies involving more patients are still needed to determine its optimal application mode and dosage. In this paper, we have reviewed the basic characteristics of H2, and its therapeutic effects in ophthalmic diseases. We also focus on the latest progress in the administration approaches and mechanisms underlying these benefits.

Mitochondrial autophagy plays a contributary role in the pathogenesis of retina degeneration (RD). ZYAN1 is a novel proline hydroxylase domain (PHD) inhibitor that can enhance the expression of hypoxia-inducible factor 1-alpha (HIF-1α). This study investigated whether ZYAN1 could alleviate progressive photoreceptor loss and oxidative damage in a pharmacologically induced RD model via the modulation of mitophagy. ZYAN1 was injected into the vitreous body of the RD model, and the retinal autophagy level was analyzed. The therapeutic effects of ZYAN1 were evaluated via a function examination, a morphological assay, in situ reactive oxygen species (ROS) detection, and an immunofluorescence assay. It was shown that the thickness of the outer nuclear layer (ONL) increased significantly, and visual function was efficiently preserved via ZYAN1 treatment. The mitochondria structure of photoreceptors was more complete in the ZYAN1-treated mice, and the number of autophagosomes also increased significantly. Membrane disc shedding and ROS overproduction were alleviated after ZYAN1 treatment, and the axonal cilia were more structurally intact. A Western blot analysis showed that the expression levels of the autophagy-related proteins LC3-B, Beclin-1, and ATG5 increased significantly after ZYAN1 treatment, while the expression of P62 was down-regulated. Moreover, the expression levels of HIF-1α and BNIP3 were up-regulated after ZYAN1 treatment. Therefore, an intravitreal injection of ZYAN1 can act as part of the pharmacologic strategy to modulate mitophagy and alleviate oxidative stress in RD. These findings enrich our knowledge of RD pathology and provide insights for the discovery of a therapeutic molecule.

Parents may influence children’s intake of sweets and sugar-sweetened beverages through their feeding styles. This study prospectively assessed the association between parental feeding styles and caries risk in preschool children. In November and December 2021, we recruited 1181 preschool children aged 3–4 years in Guangzhou, China, and then conducted a follow-up visit after 15 months. The children were examined for dental caries, and their parents answered the Chinese version of the Parental Feeding Style Questionnaire. Data were analyzed using logistic regression analysis. At baseline, ‘control over eating’ was found to be negatively associated with early childhood caries (OR = 0.60, 95%CI = 0.44, 0.83). After 15 months, no significant association between baseline parental feeding styles and the risk of a new carious lesion was found. When considering changes in parental feeding styles between the baseline and the follow-up visit, we found children with increasing scores on ‘instrumental feeding’ during the 15 months had a higher risk of a new carious lesion (OR = 1.91, 95%CI = 1.18, 3.12). However, children with always-intermediate scores (OR = 0.51, 95%CI = 0.30, 0.86), always-high scores (OR = 0.42, 95%CI = 0.23, 0.77), or increasing scores (OR = 0.50, 95%CI = 0.31, 0.83) on ‘control over eating’ had a lower risk of a new carious lesion. Our study highlighted the influence of parental feeding styles on caries risk in preschool children.

This work proposes a centralized data-based orderly charging strategy that considers the user’s charging choices. Three charging choices for different types of users are described. Then, a scheduling model of electric vehicles based on the time dimension is established. In this strategy, the optimization model not only considers the demand of the grid side and the user side, but also takes the driving data of electric vehicles as the driver. The grid-side optimization involves minimizing the equivalent load fluctuation, and the user-side is optimized to minimize the charging cost and maximize the charging electric quantity. The scheduling capabilities of the three charging strategies are analyzed based on a series of driving data of electric vehicles. The results show that the peak-valley difference and equivalent load fluctuation of the power grid in the data-based orderly charging strategy reduced by 22.2% and 22.7%, respectively, and the charging cost of users also reduced much more than the other two charging strategies. Additionally, the effect of users’ charging choices on the charging strategy is analyzed, and the results show that the orderly charging strategy that considers users’ charging choices can effectively decrease the scheduling deviation caused by users’ charging choices. It greatly improves the security and economy of the grid.

Helicobacter pylori infection is a prevalent infectious disease, associated with many gastric diseases, including gastritis, gastric ulcer, and gastric cancer. To reveal the characteristics of the gastric microbiome in patients infected with H. pylori, we performed metagenomic shotgun sequencing of stomach swab samples from 96 patients and then conducted metagenomic association analyses between alterations in the gastric microbiome and H. pylori infection status. The overall composition of the gastric microbiota in H. pylori‐infected individuals was distinctly different from the negative controls; H. pylori became the dominant species after colonizing the human stomach and significantly decreased the α‐diversity of the gastric community (P 0.05, Wilcoxon rank‐sum test). This study revealed alterations in gastric microbial taxa and function associated with HPI in the Chinese population, which provides an insight into gastric microbial interactions and their potential role in the pathological process of gastric diseases.

To cater for the diverse experiment requirements at the High Energy Photon Source (HEPS) with often limited human resources, Bluesky was chosen as the basis for our software framework, Mamba. In our attempt to address Bluesky's lack of integrated graphical user interfaces (GUIs), command injection with feedback was chosen as the main way for the GUIs to cooperate with the command line interface; a remote-procedure-call service is also provided, which covers functionalities unsuitable for command injection, as well as pushing of status updates. In order to fully support high-frequency applications like fly scans, Bluesky's support for asynchronous control is being improved; furthermore, to support high-throughput experiments, Mamba Data Worker is being developed to cover the complexity in asynchronous online data processing for these experiments. To systematically simplify the specification of metadata, scan parameters and data-processing graphs for each type of experiment, an experiment parameter generator will be developed; experiment-specific modules to automate preparation steps will also be made. The integration of off-the-shelf code in Mamba for domain-specific needs is under investigation, and Mamba GUI Studio is being developed to simplify the implementation and integration of GUIs.

Abstract Background Patients with persistent globus sensation, throat clearing, chronic cough, hoarseness, and other throat symptoms account for a large proportion of patients in ears, nose, and throat clinics. Laryngopharyngeal reflux disease (LPRD) is increasingly valued by otolaryngologists. Transcutaneous electrical acupoint stimulation (TEAS) is possibly a new method for the treatment of LPRD. This trial aims to determine whether TEAS combined with proton pump inhibitor (PPI) is better than PPI alone in the treatment of LPRD. Methods This prospective randomized controlled trial will be implemented in a tertiary hospital in China. Seventy patients diagnosed with LPRD will be randomly assigned to the TEAS + PPI group (intervention group) or PPI group (control group), at a ratio of 1:1. In addition to using PPI, the intervention group will receive TEAS at four groups of acupoints, and each group will be treated for 15 min, once for 60 min, five times a week, for 12 weeks, 60 times. The main outcome will be changes in the Reflux Symptom Index scores at 4, 12, and 24 weeks after treatment. The secondary outcomes will include changes in the reflux finding score, Laryngopharyngeal Reflux-Health-related Quality of Life score, and throat pain visual analog scale score. Discussion This trial will explore the feasibility of TEAS combined with PPI for the treatment of LPRD and provide potential evidence for its effectiveness and safety. The results of this study will be published in a peer-reviewed journal. Trial registration Chinese Clinical Trial Registry ChiCTR2100046755 . Registered on May 28, 2021.

BackgroundConsiderable attention has been given to how different aspects of biodiversity sustain ecosystem functions. Herbs are a critical component of the plant community of dryland ecosystems, but the importance of different life form groups of herbs is often overlooked in experiments on biodiversity-ecosystem multifunctionality. Hence, little is known about how the multiple attributes of diversity of different life form groups of herbs affect changes to the multifunctionality of ecosystems.MethodsWe investigated geographic patterns of herb diversity and ecosystem multifunctionality along a precipitation gradient of 2100 km in Northwest China, and assessed the taxonomic, phylogenetic and functional attributes of different life form groups of herbs on the multifunctionality.ResultsWe found that subordinate (richness effect) species of annual herbs and dominant (mass ratio effect) species of perennial herbs were crucial for driving multifunctionality. Most importantly, the multiple attributes (taxonomic, phylogenetic and functional) of herb diversity enhanced the multifunctionality. The functional diversity of herbs provided greater explanatory power than did taxonomic and phylogenetic diversity. In addition, the multiple attribute diversity of perennial herbs contributed more than annual herbs to multifunctionality.ConclusionsOur findings provide insights into previously neglected mechanisms by which the diversity of different life form groups of herbs affect ecosystem multifunctionality. These results provide a comprehensive understanding of the relationship between biodiversity and multifunctionality, and will ultimately contribute to multifunctional conservation and restoration programs in dryland ecosystems.

ABSTRACT With the development and reduced costs of high-throughput sequencing technology, environmental dark matter, such as novel metagenome-assembled genomes (MAGs) and viruses, is now being discovered easily. However, due to read length limitations, MAGs and viromes often suffer from genome discontinuity and deficiencies in key functional elements. Here, by applying long-read sequencing technology to sediment samples from a Tibetan saline lake, we comprehensively analyzed the performance of high-fidelity (HiFi) reads and the possibility of integration with short-read next-generation sequencing (NGS) data. In total, 207 full-length nonredundant 16S rRNA gene sequences and 19 full-length nonredundant 18S rRNA genes were directly obtained from HiFi reads, which greatly surpassed the retrieval performance of NGS technology. We carried out a cross-sectional comparison among multiple assembly strategies, referred to as ‘NGS’, ‘Hybrid (NGS+HiFi)’, and ‘HiFi’. Two MAGs and 29 viruses with circular genomes were reconstructed using HiFi reads alone, indicating the great power of the ‘HiFi’ approach to assemble high-quality microbial genomes. Among the 3 strategies, the ‘Hybrid’ approach produced the highest number of medium/high-quality MAGs and viral genomes, while the ratio of MAGs containing 16S rRNA genes was significantly improved in the ‘HiFi’ assembly results. Overall, our study provides a practical metagenomic resolution for analyzing complex environmental samples by taking advantage of both the short-read and HiFi long-read sequencing methods to extract the maximum amount of information, including data on prokaryotes, eukaryotes, and viruses, via the ‘Hybrid’ approach. IMPORTANCE To expand the understanding of microbial dark matter in the environment, we did the first comparative evaluation of multiple assembly strategies based on high-throughput short-read and HiFi data from lake sediments metagenomic sequencing. The results demonstrated great improvement of the ‘Hybrid’ assembly method (short-read next-generation sequencing data plus HiFi data) in the recovery of medium/high-quality MAGs and viral genomes. Further analysis showed that HiFi data is important to retrieve the complete circular prokaryotic and viral genomes. Meanwhile, hundreds of full-length 16S/18S rRNA genes were assembled directly from HiFi data, which facilitated the species composition studies of complex environmental samples, especially for understanding micro-eukaryotes. Therefore, the application of the latest HiFi long-read sequencing could greatly improve the metagenomic assembly integrity and promote environmental microbiome research.

BackgroundMultiple components of biodiversity are excellent predictors of precipitation-induced changes in ecosystem function. However, the importance of differing scales (alpha versus beta) is usually overlooked in biodiversity–ecosystem multifunctionality studies. Consequently, little is known about how precipitation regulates the relationship between multifunctionality and multiple components of alpha and beta diversity.AimsWe investigated geographic patterns of herbaceous plant diversity and ecosystem multifunctionality along a precipitation gradient spanning more than 2010 km in Northwest China.MethodsWe assessed the effects of herbaceous species, phylogenetic, and functional components at different scales on multifunctionality in drylands.ResultsThe alpha diversity of species and functional beta diversity were key components explaining the variation in multifunctionality. As the main environmental factor, MAP (mean annual precipitation) affected multifunctionality by changing the mediating variables (i.e., species alpha and functional beta diversity). More importantly, a certain precipitation threshold was detected for the relationship of multifunctionality to species alpha and functional beta diversity. MAPs of approximately 158 mm and 140 mm modulated this relationship (shifting it from uncorrelated to significantly correlated).ConclusionsOur findings provide insights into previously neglected mechanisms by which diversity in herbaceous layers at different scales affects ecosystem multifunctionality. It is highlighted that MAP regulates the relationship between diversity and multifunctionality in dryland ecosystems at different scales. Further, diversity may have substantial consequences for multifunctionality where MAP is higher. These empirical results provide a comprehensive understanding of the biodiversity–multifunctionality relationship in the context of precipitation, ultimately contributing to conservation and restoration programs for multifunctionality in drylands.

Sheath blight (ShB), caused by Rhizoctonia solani, is one of the major threats to rice (Oryza sativa L.) production. However, it is not clear how the risk of rice ShB will respond to elevated CO2 and temperature under future climate change. Here, we conducted, field experiments of inoculated R. solani under combinations of two CO2 levels (ambient and enriched up to 590 μmol mol-1) and two temperature levels (ambient and increased by 2.0°C) in temperature by free-air CO2 enrichment (T-FACE) system for two cultivars (a susceptible cultivar, Lemont and a resistant cultivar, YSBR1). Results indicate that for the inoculation of plants with R. solani, the vertical length of ShB lesions for cv. Lemont was significantly longer than that for cv. YSBR1 under four CO2 and temperature treatments. The vertical length of ShB lesions was significantly increased by elevated temperature, but not by elevated CO2, for both cultivars. The vertical length of ShB lesions under the combination of elevated CO2 and elevated temperature was increased by 21–38% for cv. Lemont and by -1–6% for cv. YSBR1. A significant increase in MDA level was related to a significant increase in the vertical length of ShB lesions under the combination of elevated CO2 and elevated temperature. Elevated CO2 could not compensate for the negative effect of elevated temperature on yield of both cultivars under future climate change. Rice yield and biomass were further decreased by 2.0–2.5% and 2.9–4.2% by an increase in the severity of ShB under the combination of elevated CO2 and elevated temperature. Thus, reasonable agronomic management practices are required to improve both resistance to ShB disease and grain yield for rice under future climate change.

The sealing ability of caprock is affected by many factors, such as cap thickness, displacement pressure, fracture development, and lithology of caprock. Shale is one of the ideal materials for oil and gas sealing cap formation due to its low porosity and permeability. Microfractures can destroy the sealing property of shale caprock. When buried deep enough, shale will change from toughness to brittleness. In general, the greater the brittleness of shale, the more developed the fractures will be. In areas with high tectonic stress, such as the anticline axis, syncline axis and stratum dip end, the strata stress is high and concentrated, and it is easier to generate fractures. When the stress state of the caprock changes, new micro-cracks are formed or previously closed cracks are re-opened, reducing the displacement pressure of the caprock. These micro-fractures are interconnected to form microleakage spaces, which reduces the sealing capacity of the caprock.

Summary: Background: Anthropomorphic phantoms are used in surgical planning and intervention. Ideal accuracy and high efficiency are prerequisites for its clinical application. We aimed to develop a fully automated artificial intelligence-based three-dimensional (3D) reconstruction system (AI system) to assist thoracic surgery and to determine its accuracy, efficiency, and safety for clinical use. Methods: This AI system was developed based on a 3D convolutional neural network (CNN) and optimized by gradient descent after training with 500 cases, achieving a Dice coefficient of 89.2%. Accuracy was verified by comparing virtual structures predicted by the AI system with anatomical structures of patients in retrospective (n = 113) and prospective cohorts (n = 139) who underwent lobectomy or segmentectomy at the Peking University Cancer Hospital. Operation time and blood loss were compared between the retrospective cohort (without AI assistance) and prospective cohort (with AI assistance) for safety evaluation. The time consumption for reconstruction and the quality score were compared between the AI system and manual reconstruction software (Mimics®) for efficiency validation. Findings: The AI system reconstructed 13,608 pulmonary segmental branches from retrospective and prospective cohorts, and 1573 branches of interest corresponding to phantoms were detectable during the operation for verification, achieving 100% and 97% accuracy for segmental bronchi, 97.2% and 99.1% for segmental arteries, and 93.2% and 98.8% for segmental veins, respectively. Abstract With the incorporation of noble metal materials, photonic crystal fibers (PCFs) could be performed as an effective platform for refractive index sensing of the filling analytes. Furthermore, by coating functional dielectric layers upon the metal surfaces, the resonance energy transfer is modulated from the core mode of the PCFs towards the surface plasmon resonance mode of the metals, and the sensing performance could be boosted. Here, considering that the exciton-plasmon coupling is efficient between perovskite quantum dots (QDs) and gold, a kind of CsPbBr3 QDs/Au bilayer coated triangular-lattice PCFs has been simulated numerically as the refractive index sensors. With the optimization of the QDs and gold layer thicknesses, together with the variation of the central hole size of the PCFs, in the refractive index (RI) region of 1.26 to 1.34, a rather narrow full width at half maximum (FWHM) of the loss spectra was achieved as 13.74nm when the central hole size was 1.28 µm and the highest figure of merit was 63.79RIU (the central hole size was 1.53 µm). This work demonstrates that the analyte identification accuracy was enhanced by FWHM narrowing of the loss spectra; in addition, taking the abundance of the material choice of perovskite QDs into consideration, more analytes could be detected effectively. Moreover, by adopting asymmetric structures, the sensitivity of the PCFs based refractive index sensors could be further improved.

Circularly polarized organic afterglow (CPOA) with both long-lived room-temperature phosphorescence and circularly polarized luminescence is attracting great interest, but the development of multicolor-tunable CPOA in a single-component material remains challenging. Here, the authors report a strategy to achieve multicolor CPOA through chiral clusterization by inserting a chirality center into a non-conjugated organic cluster.

International interest is growing in biodiversity conservation and sustainable use in drylands. Desert ecosystems across arid Central Asia are severely affected by global change. Understanding the changes in a plant community is an essential prerequisite to revealing the community assembly mechanism, vegetation conservation, and management. The knowledge of large-scale spatial variation in plant community structure in different Central Asian deserts is still limited. In this study, we selected the Taukum (TD, Kazakhstan) and the Gurbantunggut (GD, China) deserts as the research area, with similar latitudes despite being nearly 1000 km apart. Thirteen and 15 sampling plots were set up and thoroughly investigated. The differences in community structure depending on multiple plant attributes (individual level: plant height, canopy diameter, and plant volume, and community level: plant density, total cover, and total volume) were systematically studied. TD had a better overall environmental status than GD. A total of 113 species were found, with 68 and 74 in TD and GD, respectively. The number of species and plant attributes was unequally distributed across different families and functional groups between deserts. The values of several plant attributes, such as ephemerals, annuals, dicotyledons, and shrubs with assimilative branches in GD, were significantly lower than those in TD. The Motyka indices of six plant attributes (26.18–38.61%) were higher between the two deserts than the species similarity index (20.4%), indicating a more robust convergence for plant functional attributes. The community structures in the two deserts represented by different plant attribute matrices demonstrated irregular differentiation patterns in ordination diagrams. The most variance in community structure was attributed to soil and climatic factors, while geographic factors had the smallest proportion. Consequently, the community structures of the two distant deserts were both different and similar to an extent. This resulted from the long-term impacts of heterogeneous environments within the same region. Our knowledge is further deepened by understanding the variation in community structure in different deserts on a large spatial scale. This therefore provides valuable insights into conserving regional biodiversity in Central Asia.

We propose herein a novel microfluidic approach for the simultaneous active pumping and mixing of analytes in a straight microchannel via the AC field-effect control of induced-charge electro-osmosis (ICEO) around metal–dielectric solid Janus cylinders of inherent inhomogeneous electrical polarizability immersed in an electrolyte solution. We coin the term “Janus AC flow field-effect transistor (Janus AC-FFET)” to describe this interesting physical phenomenon. The proposed technique utilizes a simple device geometry, in which one or a series of Janus microcylinders are arranged in parallel along the centerline of the channel’s bottom surface, embedding a pair of 3D sidewall driving electrodes. By combining symmetry breaking in both surface polarizability and the AC powering scheme, it is possible, on demand, to adjust the degree of asymmetry of the ICEO flow profile in two orthogonal directions, which includes the horizontal pump and transversal rotating motion. A comprehensive mathematical model was developed under the Debye–Hückel limit to elucidate the physical mechanism underlying the field-effect-reconfigurable diffuse-charge dynamics on both the dielectric and the metal-phase surfaces of the Janus micropillar. For innovation in applied science, an advanced microdevice design integrating an array of discrete Janus cylinders subjected to two oppositely polarized gate terminals is recommended for constructing an active microfluidic pump and mixer, even without external moving parts. Supported by a simulation analysis, our physical demonstration of Janus AC-FFET provides a brand-new approach to muti-directional electro-convective manipulation in modern microfluidic systems.

An electrostatic probe differential analysis method is used to diagnose the arc current-carrying region of the sheet slanting tungsten electrode. Based on the local thermal equilibrium condition and energy transition model with charged particles collision, the temperature distribution in the current-carrying region of different welding process parameters is solved by saturated ion current. The results show that the temperature distribution range in the width direction of sheet slanting tungsten electrode expands, the arc high-temperature region shifts integrally, and the temperature in the thickness direction of sheet slanting tungsten electrode would be symmetrical. The guide effect of the hypotenuse of sheet slanting tungsten electrode for arc current and the inertia drag effect of arc would mainly change the temperature distribution. The variation of the inclination angle of the hypotenuse of sheet slanting tungsten electrode will aggravate the shift of the arc high-temperature region. The larger inclination angle will enhance the guiding effect, and then the inertia drag effect would be in a dominant position with a smaller inclination angle. With the increase of welding current, both the arc stiffness and the guiding effect would be intensified, the latter should make the arc high-temperature zone shift to the position with a small discharge gap.

Objective The aim of this work was to assess the effect of ergothioneine (ESH)-enriched mushroom extract on oxidative stability, volatile compounds, and sensory quality of emulsified sausage. Methods The ESH content was determined by high performance liquid chromatography. The antioxidant activity of Flammulina velutipes (F. velutipes) extract was determined through radical-scavenging activity of 1,1 diphenyl-2-picryl-hydrazyl, 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) and hydroxyl radicals. Four different groups of emulsified sausage were manufactured: control, no antioxidants; BHA, 0.01% butylated hydroxyanisole; EEME, 0.8% ESH-enriched mushroom (F. velutipes) extract; AE, 0.012% authentic ESH, after storage for 14 days (at 4°C), the quality of sausage including oxidative stability (2-thiobarbituric acid reactive substances and protein carbonyls content), volatile compounds and sensory quality were studied. Results It was demonstrated that adding ESH-enriched F. velutipes extract to sausage could effectively prevent lipid and protein oxidation, and its efficacy was equivalent with 0.01% BHA. During meat processing, the ESH mainly contributed to the antioxidative activity of F. velutipes extract. The flavor and sensory attributes of emulsified sausage were improved through adding ESH-enriched F. velutipes extract. Conclusion Accordingly, the extract of F. velutipes contained high-level of ESH and could be a good antioxidant candidate for processed meat production.

Shrubs play a major role in maintaining ecosystem stability in the arid deserts of Central Asia. During the long-term adaptation to extreme arid environments, shrubs have developed special assimilative branches that replace leaves for photosynthesis. In this study, four dominant shrubs with assimilative branches, namely Haloxylon ammodendron, Haloxylon persicum, Calligonum mongolicum, and Ephedra przewalskii, were selected as the research objects, and the dry mass, total length, node number, and basal diameter of their assimilative branches and the average length of the first three nodes were carefully measured, and the allometric relationships among five traits of four species were systematically compared. The results indicated that: (1) Four desert shrubs have different assimilative branches traits. Compared with H. persicum and H. ammodendron, C. mongolicum and E. przewalskii have longer internodes and fewer nodes. The dry mass of H. ammodendron and the basal diameter of H. persicum were the smallest; (2) Significant allometric scaling relationships were found between dry mass, total length, basal diameter, and each trait of assimilative branches, all of which were significantly less than 1; (3) The scaling exponents of the allometric relationship between four traits and the dry mass of assimilative branches of H. persicum were greater or significantly greater than those of H. ammodendron. The scaling exponents of the relationships between the basal diameter, dry mass, and total length of E. przewalskii were higher than those of the other three shrubs. Therefore, although different species have adapted to drought and high temperatures by convergence, there was great variability in morphological characteristics of assimilative branches, as well as in the scaling exponents of relationships among traits. The results of this study will provide valuable insights into the ecological functions of assimilative branches and survival strategies of these shrubs to cope with aridity and drought in desert environments.

Alzheimer’s disease (AD) and vascular dementia (VD) are the two most common forms of dementia, share similar symptoms, and are sometimes difficult to distinguish. To investigate the potential mechanisms by which they differ, we identified differentially expressed genes in blood and brain samples from patients with these diseases, and performed weighted gene co-expression network analysis and other bioinformatics analyses. Weighted gene co-expression network analysis resulted in mining of different modules based on differences in gene expression between these two diseases. Enrichment analysis and generation of a protein-protein interaction network were used to identify core pathways for each disease. Modules were significantly involved in cAMP and AMPK signaling pathway, which may be regulated cell death in AD and VD. Genes of cAMP and neurotrophin signaling pathways, including ATP1A3, PP2A, NCEH1, ITPR1, CAMKK2, and HDAC1, were identified as key markers. Using the least absolute shrinkage and selection operator method, a diagnostic model for AD and VD was generated and verified through analysis of gene expression in blood of patients. Furthermore, single sample gene set enrichment analysis was used to characterize immune cell infiltration into brain tissue. That results showed that infiltration of DCs and pDCs cells was increased, and infiltration of B cells and TFH cells was decreased in the brain tissues of patients with AD and VD. In summary, classification based on target genes showed good diagnostic efficiency, and filled the gap in the diagnostic field or optimizes the existing diagnostic model, which could be used to distinguish between AD and VD.

Obesity and atherosclerosis are the most prevalent metabolic diseases. ApoE−/− and ob/ob mice are widely used as models to study the pathogenesis of these diseases. However, how gut microbes, gut bacteriophages, and metabolites change in these two disease models is unclear. Here, we used wild-type C57BL/6J (Wt) mice as normal controls to analyze the intestinal archaea, bacteria, bacteriophages, and microbial metabolites of ob/ob and ApoE−/− mice through metagenomics and metabolomics. Analysis of the intestinal archaea showed that the abundances of Methanobrevibacter and Halolamina were significantly increased and decreased, respectively, in the ob/ob group compared with those in the Wt and ApoE−/− groups (p < 0.05). Compared with those of the Wt group, the relative abundances of the bacterial genera Enterorhabdus, Alistipes, Bacteroides, Prevotella, Rikenella, Barnesiella, Porphyromonas, Riemerella, and Bifidobacterium were significantly decreased (p < 0.05) in the ob/ob mice, and the relative abundance of Akkermansia was significantly decreased in the ApoE−/− group. The relative abundances of A. muciniphila and L. murinus were significantly decreased and increased, respectively, in the ob/ob and ApoE−/− groups compared with those of the Wt group (p < 0.05). Lactobacillus_ prophage_ Lj965 and Lactobacillus _ prophage _ Lj771 were significantly more abundant in the ob/ob mice than in the Wt mice. Analysis of the aminoacyl-tRNA biosynthesis metabolic pathway revealed that the enriched compounds of phenylalanine, glutamine, glycine, serine, methionine, valine, alanine, lysine, isoleucine, leucine, threonine, tryptophan, and tyrosine were downregulated in the ApoE−/− mice compared with those of the ob/ob mice. Aminoacyl-tRNA synthetases are considered manifestations of metabolic diseases and are closely associated with obesity, atherosclerosis, and type 2 diabetes. These data offer new insight regarding possible causes of these diseases and provide a foundation for studying the regulation of various food nutrients in metabolic disease models.

COVID-19 is a critical public health threat, but molecular characterizations of patients’ immunity is still lacking. Here the authors collected blood from patients with various disease severity, and prefiltered to exclude selected comorbidity, to obtain genomic, transcriptomic, proteomic, metabolomic and lipidomic profiles to report a trans-omics landscape.

The intriguing properties of two-dimensional (2D) transition metal dichalcogenides (TMDCs) enable the exploration of new electronic device architectures, particularly the emerging memristive devices for in-memory computing applications. Implementation of arithmetic logic operations taking advantage of the non-linear characteristics of memristor can significantly improve the energy efficiency and simplify the complexity of peripheral circuits. Herein, we demonstrate an arithmetic logic unit function using a lateral volatile memristor based on layered 2D tungsten disulfide (WS2) materials and some combinational logic circuits. Removable oxygen ions were introduced into WS2 materials through oxygen plasma treatment process. The resistive switching of the memristive device caused by the thermophoresis-assisted oxygen ions migration has also been revealed. Based on the characteristics of excitatory postsynaptic current (EPSC), paired-pulse facilitation (PPF), and spike rate dependent plasticity (SRDP), a real-time numerical system convertor was successfully accomplished, which is a significant computing function of arithmetic logic unit. This work paves a new way for developing 2D memristive devices for future arithmetic logic applications.

Naoqingzhiming, whose active ingredient is echinacoside, is the first new Class I natural medicine approved for clinical trials for the therapeutic potential of vascular dementia in China. We report randomized, double-blind, placebo-controlled trials to evaluate the safety, tolerability and pharmacokinetics of single ascending doses of Naoqingzhiming tablet after oral administration in healthy Chinese subjects. The single ascending dose of Naoqingzhiming tablet (180–2160 mg) were well tolerated in all enrolled subjects, without serious adverse events and adverse events leading to withdrawal from the study. The most common drug-related treatment-emergent adverse events were elevated blood bilirubin and serum uric acid. No clinically significant findings were found in the physical examinations, vital signs or electrocardiograms. After single-dose administration of Naoqingzhiming tablet, echinacoside was absorbed with a Tmax at 1.25–1.75 h and declined with a t1/2 of 2.42–3.33 h. However, the proportionality coefficients for Cmax, AUC0−t and AUC0-∞ of echinacoside were not fully contained in the pre-defined 90 % CI criterion (0.91–1.09). As a result, the dose proportionality could not be concluded statistically within the dosage range of this study. Overall, the safety profile and PK properties support further development and use of Naoqingzhiming in vascular dementia.

AbstractHot dry rock geothermal energy is deep geological energy. Its ability to resist fracture is an important basis for effective reconstruction and scientific evaluation of the stability of geothermal reservoirs. Hot dry rock is typically buried deeply, and the reservoir is often in a high-temperature and high stress environment. There have been limited studies conducted on the effect of different three-dimensional stress and temperature on granite fracture toughness. Thereby, herein an experimental study is conducted on the heat treatment of granite under different external loads and temperatures. The variation in fracture toughness of granite with temperature and pressure is studied using a three-point bending fracture mechanics experiment, scanning electron microscope (SEM) observation, and acoustic wave velocity measurement. The results show that under the joint influence of 25 MPa deviator stress and 200 °C temperature, the Mode I, Mixed mode (I + II), and Mode II fracture toughness of granite show a nonlinear change trend of decreasing and increasing. Among the three modes, the change range of Mode I fracture toughness is not more than 10% which is not significant. Contrarily, the degradation effect of rock mechanical properties caused by the joint action of stress and temperature in Mode II and Mixed mode (I + II) is predominant. The maximum range of Mode II fracture toughness is reduced by 22%, whereas the maximum range of Mixed mode (I + II) fracture toughness is reduced by 18%. However, the compression action of three-dimensional stress causes a slight enhancement in granite mechanical properties, wherein the maximum range of Mixed mode (I + II) fracture toughness is increased by 12%. Furthermore, the change of granite’s ability to resist tensile, shear, and composite (tensile + shear) fracture is not coordinated under the joint action of different temperatures and external loads. This may be due to the small deviator stress effect, which is similar to the early loading stage of uniaxial compression. External loads and thermal stress damage occur in the rock along with the compaction of pore cracks. These mechanisms have different dominant positions under varied temperature and three-dimensional stress coupling conditions, resulting in either the enhancement or weakening of the mechanical properties of granite. The results of this experimental study are conducive to gaining an in-depth understanding of the change law of deep rock mechanical properties and the exploration of hot dry rock reservoir reconstruction.

The human eye is a consolidated organ with delicate structures and unique immune privileges. Ocular diseases are intractable due to the intrinsic biological barriers within the eyeball. Hydrogels are excellent drug-carrying substances with soft material and excellent properties. They have been extensively used to deliver drugs into ocular tissue via iontophoresis devices. Ophthalmic iontophoresis is an electrochemical technique using tiny electrical currents to deliver drugs into the eye non-invasively. The early infantile iontophoresis technique often required long applying time to achieve therapeutic dose in the posterior ocular segment. The potential limitations in the initial drug concentration and the maximum safe currents would also impede the efficiency and safety of iontophoresis. Moreover, the poor patient compliance always leads to mechanical damage to the cornea and sclera during application. Advantageously, the flexible drug-carrying hydrogel can be in direct contact with the eye during iontophoresis, thereby reducing mechanical damage to the ocular surface. Moreover, the water absorption and adjustable permeability of hydrogels can reduce the electrochemical (EC) reactions and enhance the efficiency of iontophoresis. In this review, we focus on recent developments of hydrogels iontophoresis in ophthalmologic practice. Refinements of the knowledge would provide an outlook for future application of hydrogels in treating ocular disease.

Diamond cutting-tool wear has a direct impact on the processing accuracy of the machined surface in ultra-precision diamond cutting. It is difficult to monitor the tool’s condition because of the slight wear amount. This paper proposed a hybrid deep learning model for tool wear state prediction in ultra-precision diamond cutting. The cutting force was accurately estimated and the wear state of the diamond tool was predicted by using the hybrid deep learning model with the motion displacement, velocity, and other signals in the machining process. By carrying out machining experiments, this method can classify diamond-tool wear condition with an accuracy of more than 85%. Meanwhile, the effectiveness of the proposed method was verified by comparing it with a variety of machine learning models.

In this study, a series of novel 3-(5-fluoropyridine-3-yl)-2-oxazolidinone derivatives were designed and synthesized based on compounds previously reported, and their antibacterial activity was investigated. Then their antibacterial activity was investigated for the first time. Preliminary screening results showed that all these compounds exhibited antibacterial activity against gram-positive bacteria, including 7 drug-sensitive strains and 4 drug-resistant strains, among which compound 7j exhibited an 8-fold stronger inhibitory effect than linezolid, with a minimum inhibitory concentration (MIC) value of 0.25 µg/mL. Further molecular docking studies predicted the possible binding mode between active compound 7j and the target. Interestingly, these compounds could not only hamper the formation of biofilms, but also have better safety, as confirmed by cytotoxicity experiments. All these results indicate that these 3-(5-fluoropyridine-3-yl)-2-oxazolidinone derivatives have the potential to be developed into novel candidates for the treatment of gram-positive bacterial infections.

Maize, as a glycophyte, is hypersensitive to salinity, but the salt response mechanism of maize remains unclear. In this study, the physiological, biochemical, and molecular responses of two contrasting inbred lines, the salt-tolerant QXH0121 and salt-sensitive QXN233 lines, were investigated in response to salt stress. Under salt stress, the tolerant QXH0121 line exhibited good performance, while in the sensitive QXN233 line, there were negative effects on the growth of the leaves and roots. The most important finding was that QXH0121 could reshift Na+ from shoots into long roots, migrate excess Na+ in shoots to alleviate salt damage to shoots, and also improve K+ retention in shoots, which were closely associated with the enhanced expression levels of ZmHAK1 and ZmNHX1 in QXH0121 compared to those in QXN233 under salt stress. Additionally, QXH0121 leaves accumulated more proline, soluble protein, and sugar contents and had higher SOD activity levels than those observed in QXN233, which correlated with the upregulation of ZmP5CR, ZmBADH, ZmTPS1, and ZmSOD4 in QXH0121 leaves. These were the main causes of the higher salt tolerance of QXH0121 in contrast to QXN233. These results broaden our knowledge about the underlying mechanism of salt tolerance in different maize varieties, providing novel insights into breeding maize with a high level of salt resistance.

High-resolution mass spectrometry (HRMS) was coupled with ultra-high-performance liquid chromatography (UHPLC) to simultaneously quantify trehalose and trehalose 6-phosphate without derivatization or sample preparation. The use of full scan mode and exact mass analysis also makes it possible to carry out metabolomic analyses as well as semi-quantification. In addition, the use of different clusters in negative mode makes it possible to compensate for deficiencies in linearity and inerrant saturation at time-of-flight detectors. The method has been approved and validated for different matrices, yeasts, and bacteria, and has shown differentiation between bacteria as a function of growth temperatures.

The sensitivity and photoelectric noise of UV photodetectors are challenges that need to be overcome in pharmaceutical solute detection applications. This paper presents a new device concept for a CsPbBr3 QDs/ZnO nanowire heterojunction structure for phototransistors. The lattice match of the CsPbBr3 QDs and ZnO nanowire reduces the generation of trap centers and avoids carrier absorption by the composite center, which greatly improves the carrier mobility and high detectivity (8.13 × 1014 Jones). It is worth noting that by using high-efficiency PVK quantum dots as the intrinsic sensing core, the device has a high responsivity (6381 A/W) and responsivity frequency (300 Hz). Thus, a UV detection system for pharmaceutical solute detection is demonstrated, and the type of solute in the chemical solution is estimated by the waveform and the size of the output 2f signals.

Abstract Background Gastrointestinal tract (GIT) microbiomes in ruminants play major roles in host health and thus animal production. However, we lack an integrated understanding of microbial community structure and function as prior studies. are predominantly biased towards the rumen. Therefore, to acquire a microbiota inventory of the discrete GIT compartments, In this study, we used shotgun metagenomics to profile the microbiota of 370 samples that represent 10 GIT regions of seven ruminant species. Results Our analyses reconstructed a GIT microbial reference catalog with > 154 million nonredundant genes and identified 8745 uncultured candidate species from over 10,000 metagenome-assembled genomes. The integrated gene catalog across the GIT regions demonstrates spatial associations between the microbiome and physiological adaptations, and 8745 newly characterized genomes substantially expand the genomic landscape of ruminant microbiota, particularly those from the lower gut. This substantially expands the previously known set of endogenous microbial diversity and the taxonomic classification rate of the GIT microbiome. These candidate species encode hundreds of enzymes and novel biosynthetic gene clusters that improve our understanding concerning methane production and feed efficiency in ruminants. Overall, this study expands the characterization of the ruminant GIT microbiota at unprecedented spatial resolution and offers clues for improving ruminant livestock production in the future. Conclusions Having access to a comprehensive gene catalog and collections of microbial genomes provides the ability to perform efficiently genome-based analysis to achieve a detailed classification of GIT microbial ecosystem composition. Our study will bring unprecedented power in future association studies to investigate the impact of the GIT microbiota in ruminant health and production. Video abstract

Abstract Wallerian degeneration (WD) involves the recruitment of macrophages for debris clearance and nerve regeneration, and the cause of the foamy macrophages that are frequently observed in peripheral transection injuries is unknown. Recent studies indicated that these foamy cells are generated by gasdermin D (GSDMD) via membrane perforation. However, whether these foamy cells are pyroptotic macrophages and whether their cell death elicits immunogenicity in peripheral nerve regeneration (PNR) remain unknown. Therefore, we used GSDMD-deficient mice and mice with deficiencies in other canonical inflammasomes to establish a C57BL/6 J mouse model of sciatic nerve transection and microanastomosis (SNTM) and evaluate the role of GSDMD-executed pyroptosis in PNR. In our study, the GSDMD −/− mice with SNTM showed a significantly diminished number of foamy cells, better axon regeneration, and a favorable functional recovery, whereas irregular axons or gaps in the fibers were found in the wild-type (WT) mice with SNTM. Furthermore, GSDMD activation in the SNTM model was dependent on the NLRP3 inflammasome and caspase-1 activation, and GSDMD-executed pyroptosis resulted in a proinflammatory environment that polarized monocytes/macrophages toward the M1 (detrimental) but not the M2 (beneficial) phenotype. In contrast, depletion of GSDMD reversed the proinflammatory microenvironment and facilitated M2 polarization. Our results suggested that inhibition of GSDMD may be a potential treatment option to promote PNR.
