In the present study an attempt has been made to prepare fast dissolving tablets of Sumatriptan Succinate to improve dissolution rate of the drug in oral cavity & hence better patient's compliance & effective therapy. Sumatriptan Succinate is a 5-HT1D (5-hydroxy tryptamine 1D) receptor agonist, used in the treatment of migraine and cluster headache. It has low bioavailability due to its first pass metabolism. Hence the main objective of the study was to formulate fast dissolving tablets of Sumatriptan succinate to achieve a better dissolution rate and further improving the bioavailability of the drug. Fast dissolving Sumatriptan succinate was prepared using superdisintegrants Crospovidone, Croscarmellose Sodium, and Preglatinised Starch using the direct compression method. The tablets prepared were evaluated for Thickness, Uniformity of weight, Drug content, Hardness, Friability, Wetting time, in vitro and in vivo disintegration time, in vitro drug release. The formulation F8 containing Crospovidone (8%) was found to give the best result since it showed enhanced dissolution, which leads to improved bioavailability, improved effectiveness and hence better patient compliance. Introduction: Artificial Intelligence (AI) is revolutionizing techniques in pediatric dentistry as it helps in diagnostic precision, treatment planning, and patient care. In this field, AI algorithms analyze dental images to detect anomalies, aiding in the early identification of dental issues in children. AI-driven educational tools empower both parents and children with oral health information. Aim: The aim is to assess the knowledge, practices, and awareness of the usage of AI in pediatric dentistry among undergraduate students in Bhopal City. Materials and Methods: The study will be conducted among undergraduate students in Bhopal City and the questionnaire included respondents recognition and fundamental knowledge, practice, and awareness toward AI. The data are collected, compiled, and tabulated. Result and Observations: The result will be statistically analyzed using SPSS software. Conclusion: The understanding of AI-driven dentistry concepts among students appears to be acceptable in different facets. Substrate ambiguity and relaxed reaction specificity underlie the diversity of reactions catalyzed by the transsulfuration pathway enzymes, cystathionine β-synthase (CBS) and γ-cystathionase (CSE). These enzymes either commit sulfur metabolism to cysteine synthesis from homocysteine or utilize cysteine and/or homocysteine for synthesis of H2S, a signaling molecule. We demonstrate that a kinetically controlled heme-dependent metabolite switch in CBS regulates these competing reactions where by cystathionine, the product of CBS, inhibits H2S synthesis by the second enzyme, CSE. Under endoplasmic reticulum stress conditions, induction of CSE and up-regulation of the CBSinhibitor, CO, a product of heme oxygenase-1, flip the operating preference of CSE from cystathionine to cysteine, transiently stimulating H2S production. In contrast, genetic deficiency of CBS leads to chronic stimulation of H2S production. This metabolite switch from cystathionine to cysteine and/or homocysteine renders H2S synthesis by CSE responsive to the known modulators of CBS: S-adenosylmethionine, NO, and CO. Developing methods to detect andrepair damage in polymers is anactive area of research. Many of the previously described methodssuffer from the lack of long-term stability of the reagents, whichare typically preincorporated into the polymer. Also, they tend tobe specific to certain types of polymeric materials. In this paper,we describe a general method for detection and repair of cracks inpolymers using a salt-triggered mechanism. The process consists ofa polymer embedded with salt that works as a “reporter”.Upon addition of a detection or healing agent, damaged areas in thepolymer leach out salt, powering flows and activating the reagentsin the fluid. Detection is possible with fluorescent quantum dots,which aggregate at the crack site. Repair is shown to occur throughtwo different strategies. The first repair strategy involves highionic strength triggered destabilization of oil-in-water emulsions,transporting polymerization agents, resulting in polymer depositionat the damage site. The second, more biocompatible strategy, involvesusing an enzyme, urease, and its catalytic hydrolysis of urea to depositsolid calcium carbonate in the crack. An α--rhamnosidase producing fungal strain has been isolated from decaying lemon fruit. The fungal strain has been identified as Aspergillus flavus. The α--rhamnosidase has been purified from the culture filtrate of the fungal strain using ultra filtration and cation exchange chromatography on carboxy methyl (CM) cellulose. The molecular mass of the purified enzyme determined by SDS-PAGE analysis was 41 kDa. The K values of the enzyme using p-nitrophenyl-α--rhamnopyranoside and naringin as the substrates were 1.89 and 1.6 m respectively. The pH and temperature optima of the enzyme were 11.0 and 50 °C respectively. The effects of various chemical species present in grape fruit juice and wine on the activity of the enzyme have been determined. [ABSTRACT FROM AUTHOR]

The article focuses on the need for prosthetic and orthotic services in India to improve the quality of life of children with disabilities through early identification and intervention. Early intervention is the earliest identification and comprehensive service and support for children with developmental delays and disabilities ranging from 0 to 6 years to improve their ability to adapt to society. A practical early intervention delays or prevents future complications and prepares the child for functional adult life. It may positively affect the quality of life of the differently abled and their family members, and they can lead an active personal and social life. Also, such interventions promote the well-being and dignity of the differently abled and their parents, which may result in national economic progress. The synthesis of coumarin derivatives has been an essential topic since its discovery in 1820. In bioactive compounds, the coumarin moiety serves as a backbone, as many such bioactive compounds with the coumarin moiety play a significant role in their bioactivities. Given this moiety's relevance, several researchers are developing fused-coumarin derivatives to create new drugs. Mostly the approach done for this purpose was a multicomponent reaction based. Over the years, the multicomponent reaction has gained enormous popularity, and this approach has evolved as a replacement for conventional synthetic methods. Bu 4 NI-catalyzed regioselective N 2 -methylation, N 2 -alkylation, and N 2 -arylation of tetrazoles have been achieved using tert -butyl hydroperoxide (TBHP) as the methyl source, alkyl diacyl peroxides as the primary alkyl source, alkyl peresters as the secondary and tertiary alkyl sources, and aryl diacyl peroxides as the arylating source. These reactions proceed without pre-functionalization of tetrazole and in the absence of any metal catalysts. Here, peroxides serve the dual role of oxidants as well as alkylating or arylating agents. Based on DFT calculations, it was found that spin density, transition-state barriers (kinetic control), and thermodynamic stability of the products (thermodynamic control) play essential roles in the observed regioselectivity during N -alkylation. This radical-mediated process is amenable to a broad range of substrates and provides products in moderate to good yields.

An alpha-L-rhamnosidase secreting fungal strain has been isolated and identified as Aspergillus clavato-nanicus MTCC-9611. The enzyme was purified to homogeneity from the culture filtrate of the fungus using concentration by ultrafiltration membrane and ion-exchange chromatography on CM-cellulose. The native PAGE analysis confirmed the homogeneity of the purified enzyme. The SDS-PAGE analysis of the purified enzyme revealed a single protein band corresponding to the molecular weight 82 kDa. The alpha-L-rhamnosidase activity of Aspergillus clavato-nanicus MTCC-9611 had optimum at pH 10.0 and 50 degrees C. The K(m) values of the enzyme were 0.65 mM and 0.95 mM using p-nitrophenyl alpha-L-rhamnopyranoside and naringin as a substrates respectively. The enzyme transforms naringin to prunin at pH 10.0 and further hydrolysis of prunin to naringenin does not occur under these reaction conditions that makes alpha-L-rhamnosidase activity of Aspergillus clavatonanicus MTCC-9611 promising enzyme to get prunin for pharmaceutical purposes.
