Abstract

Tumstatin (Tum) is a powerful angiostatin that inhibits proliferation and induces apoptosis of tumorous vascular endothelial cells. A nonpathogenic and anaerobic bacterium, Bifidobacterium longum(BL), selectively localizes to and proliferates in the hypoxia location within solid tumor. The aims of this study were to develop a novel delivery system for Tum using engineered Bifidobacteriumand to investigate the inhibitory effect of Tum on tumor in mice. A vector that enabled the expression of Tum under the control of the pBBADs promoter of BLwas constructed and transformed into BLNCC2705 by electroporation. The mouse colon carcinoma cells CT26 (1 × 107/mL) were subcutaneously inserted in the left armpit of BALB/c mice. The tumor-bearing mice were treated with Tum-transformed BL, and green fluorescent protein (GFP)-transformed BLwas used as a negative control. The microvessel density (MVD) in the transplanted tumor was determined, and terminal deoxynucleotidyl transferase-mediated 2′-deoxyuridine 5′-triphosphate nick end labeling was used to detect apoptosis of vascular endothelial cells in transplanted tumor. The in vitroexpression of Tum was examined in BLafter l-arabinose induction. Bifidobacterium longumwith pBBAD-Tum (BL-Tum) showed significant antitumor effect in tumor-bearing mice. The weight, volume, growth, and MVD, as well as the percentage of apoptotic vascular endothelial cells of transplanted tumors in the tumor-bearing mice treated with Tum-transformed BLwere all significantly lower than those in the GFP negative control group. Intragastric administration, injection in tumor and vena caudalis injection of Tum-transformed BLexerted marked antitumor effects in tumor-bearing mice. This is the first demonstration of the utilization of Tum-transformed BLas a specific gene delivery system for treating tumor.

72. Comparison of mesozooplankton communities in North Channel and North Branch of Yangtze River Estuary.

Abstract

According to the four cruises investigations in the Yangtze River Estuary from May, August and November 2004 and February 2005, and by using species diversity indices, K-dominance curves, and similarity analysis (ANOSIM), the features of mesozooplantkon communities and their dominant species in the North Channel (NC) and North Branch (NB) were analyzed, with the effects of ecological factors, especially water salinity and temperature, on the dominant species and communities in these two waters discussed. The results indicated that the NC was composed of tidal water and oligo-haline water, while the NB was featured by meso-haline or poly-haline water. Consequently, the zooplankton communities in these two waters were significantly different. In spring, the abundance and diversity of the communities were larger in NB than in NC. In NC, Sinocalanus sinicus was the only dominant species; while in NB, there were several dominate species, among which, Labidocera euchaeta was most important. In summer, the diversity of the communities in NC and NB was quite similar, but the species richness and total abundance of zooplankton were less in NC than in NB. The zooplankton in NC was made up of olio-haline communities, and that in NB was composed of brackish species, e.g., Schmackeria poplesia and Tortanus vermiculus. In autumn, similar to those in spring, the abundance and diversity of the communities were larger in NB than in NC. As usual, in NC, S. sinicus was the most important species; while in NB, the zooplankton was composed of neritic community (Paracalanus parvus, P. aculeatus, L. euchaeta, and Calanus sinicus, etc. ). In winter, the species richness and diversity were the lowest, and there were no obvious difference between the zooplankton communities in NC and NB. It was concluded that water salinity, water temperature, and their combination were the key factors determining the characters of the mesozooplanktonic communities. Water temperature mainly affected the seasonal alteration of the dominant species, while water salinity, which was influenced by the interaction between runoff and ocean mass, determined the local mesozooplantkon communities in NC and NB. 73. Ecological character of pelagic mysids in Yangtze Estuary of China.

Abstract

Based on the four seasonal oceanographic cruises investigation data in the Yangtze Estuary (122 ° 00′-123 ° 30′ E, 29 ° 00′-32 ° 00′ N) in 2002-2003, the diversity and abundance dynamics of pelagic mysids as well as their relationships with fishing ground were studied. A total of 14 species were identified, among which, 10 species occurred in autumn, 8 species in spring and summer, and 2 species in winter. Clear seasonal alterations were observed, among which the highest alteration ratio (90.9%) occurred from autumn to winter. Shannon diversity index (H′) in spring, summer and autumn was above 2, and that in winter was 1. Mean abundance was the highest in summer [234.70 ind·(100 m³)-1], followed by in autumn [103.34 ind·(100 m³)-1], spring [80.36 ind·(100 m³)-1], and winter [12.40 ind·(100 m³)-1]. The changes in mean abundance were in accordance with the fluctuation of seasonal temperature. Due to the adaptation to a wide range of temperature and salinity, Gastrosaccus pelagicus was dominant in spring, autumn and winter. Acanthomysis brevirostris was dominant in summer and autumn, and A. longirostrisas in winter. The dominant species in each season all made a significant contribution to total abundance. A. brevirostris was observed in large agglomerates in summer. As a kind of fish diet, mysids were of importance in the formation of fishing grounds in the Yangtze Estuary and Zhoushan Islands. 75. Improved Second Harmonic Generation from Organic-Dye-Doped Polymer/Silica Hybrid Materials

Abstract

In this study, we investigated the second-order optical nonlinearity and its thermal stability of DR1 (disperse red 1) doped poly(methyl methacrylate) (PMMA)/silica gel films. We found that in situ polymerization of sol−gel precursors (tetraethoxysilane, TEOS) in a polymer generated a hybrid matrix which is a good tradeoff between a silica gel and a polymer matrix. The nonlinear thermal stability of the hybrid system increased significantly compared to that of a poled DR1/PMMA film. Our result proved that the combination of a poled polymer and the sol−gel technique provides a way to increase the thermal stability of nonlinear optical thin films fabricated from existing materials rather than synthesizing new chemical compounds. Several factors which affect the second-order optical nonlinearity of the hybrid system were also examined.

76. Preparation and Adsorption Properties of Microporous Manganese Titanate Pillared with Silica

Abstract

The layered material RbxMnxTi2-xO4 (x = 0.75) was pillared with silica. The Rb ions were first exchanged with n-alkylammonium ions (CnH2n+1NH3, n = 6−18) to separate the interlayer space of the titanate, and then tetraethoxysilane was hydrolyzed between the layers. Burning off the organic parts resulted in silica pillared microporous solids with a surface area as large as 500−800 m2/g. The porous structure was stable up to 600 °C. Adsorption−desorption isotherms for various vapors, such as water, methanol, toluene, and mesitylene, were measured, which suggested that the porous structure formed between the layers was very similar to those of zeolites and uniform without mesopores. The pores showed hydrophobic properties. Their size was on the order of that of mesitylene.

Abstract

Murine Kupffer cells (RC) were isolated in sufficient number and purity to allow in vitro investigations of their tumoricidal capabilities. The identity of the adherent cells as KCs was established by morphologic, histochemical, and functional criteria. The yield of KCs varied from young (high) to old (low) mice but was not affected by the mouse strain. KCs activated in vitro by either endotoxins (lipopolysaccharide) or lymphokines were rendered highly cytotoxic against syngeneic melanoma or fibrosarcoma target cells. These studies indicate that KCs may indeed play a role in destruction of tumor cells in vivo and thus be important in host defense against developing hepatic cancer metastases.

Abstract

In this work, the biomacromolecule, single‐stranded deoxyribonucleic acid (ssDNA) was innovatively incorporated into the polyamide layer to tailor the permeate flux and antifouling performance of the nanofiltration (NF) membranes. With active amines groups, the ssDNA was as the aqueous phase monomers along with piperazine (PIP), and reacted with trimesoyl chloride on polyethersulfone substrate to fabricate thin‐film composite (TFC) NF membranes. The NF membrane prepared under optimal ratio of ssDNA/PIP had a pure water permeability of 75.8 L m−2 h−1 (improved 58% compared to PIP NF membrane) and Na2SO4 rejection of 98.0% at 6.0 bar. The rejections for different inorganic salts were the order: Na2SO4 (98.0%) > MgSO4 (89.2%) > MgCl2 (72.8%) > NaCl (23.0%). Abstract

DAMPE satellite has directly measured the cosmic ray proton spectrum from 40 GeV to 100 TeV and revealed a new feature at about 13.6 TeV., The precise measurement of the spectrum of protons, the most abundant component of the cosmic radiation, is necessary to understand the source and acceleration of cosmic rays in the Milky Way. This work reports the measurement of the cosmic ray proton fluxes with kinetic energies from 40 GeV to 100 TeV, with 2 1/2 years of data recorded by the DArk Matter Particle Explorer (DAMPE). This is the first time that an experiment directly measures the cosmic ray protons up to ~100 TeV with high statistics. The measured spectrum confirms the spectral hardening at ~300 GeV found by previous experiments and reveals a softening at ~13.6 TeV, with the spectral index changing from ~2.60 to ~2.85. Our result suggests the existence of a new spectral feature of cosmic rays at energies lower than the so-called knee and sheds new light on the origin of Galactic cosmic rays.

Abstract

The measurement of the energy spectrum of cosmic ray helium nuclei from 70 GeV to 80 TeV using 4.5 years of data recorded by the DArk Matter Particle Explorer (DAMPE) is reported in this work. A hardening of the spectrum is observed at an energy of about 1.3 TeV, similar to previous observations. In addition, a spectral softening at about 34 TeV is revealed for the first time with large statistics and well controlled systematic uncertainties, with an overall significance of $4.3\sigma$. The DAMPE spectral measurements of both cosmic protons and helium nuclei suggest a particle charge dependent softening energy, although with current uncertainties a dependence on the number of nucleons cannot be ruled out., Comment: 11 pages, 13 figures, published in Phys. Abstract

Ethyl carbamate (EC), a genotoxic and carcinogenic compound in soy sauce accumulated during thermal processes, has raised public health concern for its multipoint potential carcinogenic risk to human. In this work, based on the analysis of EC accumulation during thermal processes of soy sauce, ornithine and quercetin were added before thermal processes to reduce EC accumulation. A reduction rate of 23.7–63.8% in simulated solution was founded. Kinetic studies indicated that ornithine was a byproduct of alcoholysis reaction when EC formed, while quercetin could compete with the precursor ethanol and react with carbamyl compounds, which therefore prevented EC accumulation. A maximum of 47.2% decrease of EC in soy sauce was achieved, and no remarkable changes in volatile compounds profile and color of soy sauce were found. In conclusion, the addition of quercetin and ornithine before thermal processes may be preferable for the controlling of EC content in soy sauce.

Abstract

The DArk Matter Particle Explorer (DAMPE), one of the four scientific space science missions within the framework of the Strategic Pioneer Program on Space Science of the Chinese Academy of Sciences, is a general purpose high energy cosmic-ray and gamma-ray observatory, which was successfully launched on December 17th, 2015 from the Jiuquan Satellite Launch Center. The DAMPE scientific objectives include the study of galactic cosmic rays up to $\sim 10$ TeV and hundreds of TeV for electrons/gammas and nuclei respectively, and the search for dark matter signatures in their spectra. In this paper we illustrate the layout of the DAMPE instrument, and discuss the results of beam tests and calibrations performed on ground. Abstract

High energy cosmic ray electrons plus positrons (CREs), which lose energy quickly during their propagation, provide an ideal probe of Galactic high-energy processes and may enable the observation of phenomena such as dark-matter particle annihilation or decay. The CRE spectrum has been directly measured up to $\sim 2$ TeV in previous balloon- or space-borne experiments, and indirectly up to $\sim 5$ TeV by ground-based Cherenkov $\gamma$-ray telescope arrays. Evidence for a spectral break in the TeV energy range has been provided by indirect measurements of H.E.S.S., although the results were qualified by sizeable systematic uncertainties. Here we report a direct measurement of CREs in the energy range $25~{\rm GeV}-4.6~{\rm TeV}$ by the DArk Matter Particle Explorer (DAMPE) with unprecedentedly high energy resolution and low background. The majority of the spectrum can be properly fitted by a smoothly broken power-law model rather than a single power-law model. The direct detection of a spectral break at $E \sim0.9$ TeV confirms the evidence found by H.E.S.S., clarifies the behavior of the CRE spectrum at energies above 1 TeV and sheds light on the physical origin of the sub-TeV CREs., Comment: 18 pages, 6 figures, Nature in press, doi:10.1038/nature24475

Abstract

Objective: To delineate the clinical characteristics and outcomes associated with severe cardiac toxicity during the preconditioning phase of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in patients with aplastic anemia (AA). Methods: This retrospective case series study included 31 patients with severe AA who underwent allo-HSCT and were diagnosed with severe cardiac toxicity at the Hematology Department of Peking University People's Hospital from August 2012 to June 2022. The clinical manifestations of severe cardiac toxicity observed during the preconditioning process were assessed. Patient survival was assessed using the Kaplan-Meier method. Results: In this cohort of 31 patients, the median follow-up period was 9 days (range: 4-365 days). Severe cardiac toxicity manifested within 6 days after the initial cyclophosphamide (Cy) administration. Twenty patients died within 30 days of initiating Cy preconditioning, of which 16 patients died due to severe cardiac toxicity within 25 days. Patients whose cardiac function improved within 30 days post-preconditioning showed a median survival duration of 222 days ( n =11). Troponin I (TNI) levels in patients who died within 30 days of initiating Cy preconditioning began increasing on day 5 post-Cy, peaking sharply by day 9 after a notable rise on day 8. B-type natriuretic peptide (BNP) levels in patients who died within 30 days of initiating Cy preconditioning started to rise from day 1, stabilized between days 2 and 5, and then doubled daily from days 6 to 8, remaining elevated thereafter. Notably, the initial increases in BNP and TNI correlated with electrocardiogram (ECG) signs of low voltage and T-wave inversion in 83.87% of cases ( n =26). Most patients ( n =28, 90.32%) were administered corticosteroid therapy. In those with restored cardiac function, the ejection fraction returned to >50% within 30 days of initiating Cy preconditioning. Conclusions: Patients with severe cardiac toxicity during the preconditioning phase of allo-HSCT typically exhibit early, sustained, and marked elevations in myocardial damage markers, including BNP and TNI, accompanied by ECG abnormalities following Cy administration, with BNP often increasing first. These indicators are associated with rapid disease progression and high mortality. Prompt initiation of treatment upon clinical diagnosis is critical for improving survival outcomes.

Abstract

Objective: To evaluate the safety of patients with hepatic adenoma undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) . Methods: A retrospective analysis of the clinical characteristics and prognosis of eight patients with hepatic adenoma who underwent allo-HSCT in the Hematology Department of Peking University People's Hospital from January 2010 to March 2024 was conducted. Results: Of the eight patients who underwent allo-HSCT with hepatic adenoma, one patient was considered MDS-h transfusion-dependent and seven had aplastic anemia. The median age of the patients was 23 years (13-48 years). The median time from the diagnosis of AA or MDS to transplantation was 14 years (6-24 years), whereas the median time from taking androgens to diagnosing hepatic adenoma was 9 years (5-13 years). Six cases underwent haplo-HSCT, one case underwent matched unrelated donor HSCT, and one case underwent matched related donor HSCT. All patients achieved neutrophil engraftment at a median time of 11.5 days (11-20 days) and PLT engraftment within 60 days at a median of 19 days (10-37 days) after haplo-HSCT. Moreover, seven patients developed CMV anemia after transplantation, three patients had hemorrhagic cystitis, and two patients developed acute GVHD. During and after transplantation, eight patients did not show severe liver function damage or rupture of hepatic adenoma. In relation to imaging size, four patients showed varying degrees of reduction in hepatic adenoma size after transplantation, whereas four patients did not show significant changes in hepatic adenoma size after transplantation. The median follow-up time was 540.5 (30-2 989) days. Of the eight patients, six survived and two died. Furthermore, no direct correlation was observed between death and hepatic adenoma. Conclusion: Patients with hepatic adenomas undergoing allo-HSCT are not contraindications for transplantation, which will not increase transplant-related mortality.

Abstract

Epstein-Barr virus (EBV) associated post-transplant lymphoproliferative disorders (PTLD) are one of the most severe complications after hematopoietic stem cell transplantation (HSCT). This study includes 31 cases of aplastic anemia (AA) patients who developed PTLD after haploidentical transplantation, summarizing their clinical characteristics and categorizing them into either rituximab monotherapy group or combination therapy group based on whether their condition improved by 1 log after a single dose of rituximab. The incidence of PTLD after HSCT in children with AA was 10.16%, and the incidence of PTLD in patients with age >10 years was significantly increased ( χ (2)=11.336, P =0.010). Of the 31 patients, 27 were clinically diagnosed and 4 were pathologically confirmed. Finally, 15 patients were classified into the rituximab treatment group and 15 patients into the combination treatment groups. Finally three patients died, and the 2-year overall survival rate was (89.7±5.6) %. Standard pre-treatment protocols and EBV reactivation are risk factors affecting the prognosis of PTLD. There was no statistically significant difference in the impact of the two treatment schemes on prognosis.

Abstract

A total of 82 patients and healthy subjects in the First Affiliated Hospital of Sun Yat-sen University from March to August 2023 were recruited. The cohort consisted of 43 patients with head and neck squamous cell carcinoma (HNSCC) and 39 non-cancer patients or healthy subjects. There were 63 males and 19 females, with a median age of 62 (46, 67) years. The levels of folate receptor-positive circulating tumor cells (FR + CTCs) in the blood of HNSCC patients and non-cancer/healthy subjects were 12.4 (8.5, 17.8) floate unit (FU)/3 ml and 5.0 (3.8, 6.6) FU/3 ml, respectively, with a statistically significant difference ( P <0.001). The area under the receiver operating characteristic (ROC) curve for FR + CTCs levels was 0.937 (95% CI : 0.888-0.986, P <0.001), with a cut-off value of 7.4 FU/3 ml determined by the maximum Youden index. At this cut-off value, the sensitivity and specificity of FR + CTCs for diagnosing HNSCC were 90.70% and 89.74%, respectively. The current study suggests that FR + CTCs could be used as a liquid biopsy marker for the screening and diagnosis of HNSCC.
