54. Hypoxia-induced long non-coding RNA DARS-AS1 regulates RBM39 stability to promote myeloma malignancy

Multiple myeloma is a malignant plasma-cell disease, which is highly dependent on the hypoxic bone marrow microenvironment. However, the underlying mechanisms of hypoxia contributing to myeloma genesis are not fully understood. Here, we show that long non-coding RNA DARS-AS1 in myeloma is directly upregulated by hypoxia inducible factor (HIF)-1. Importantly, DARS-AS1 is required for the survival and tumorigenesis of myeloma cells both in vitro and in vivo. DARS-AS1 exerts its function by binding RNA-binding motif protein 39 (RBM39), which impedes the interaction between RBM39 and its E3 ubiquitin ligase RNF147, and prevents RBM39 from degradation. The overexpression of RBM39 observed in myeloma cells is associated with poor prognosis. Furthermore, knockdown of DARS-AS1 inhibits the mammalian target of rapamycin signaling pathway, an effect that is reversed by RBM39 overexpression. We reveal that a novel HIF-1/DARS-AS1/RBM39 pathway is implicated in the pathogenesis of myeloma. Targeting DARS-AS1/RBM39 may, therefore, represent a novel strategy to combat myeloma.

56. Potential of Bacillus subtilis lipopeptides in anti-cancer I: induction of apoptosis and paraptosis and inhibition of autophagy in K562 cells

Abstract The lipopeptide iturin from Bacillus subtilis has been found to have a potential inhibitory effect on breast cancer, alveolar adenocarcinoma, renal carcinoma, and colon adenocarcinoma. In this study, the potential of B. subtilis lipopeptides (a mixture of iturin homologues, concentration of 42.75%) to inhibit chronic myelogenous leukemia was evaluated using K562 myelogenous leukemia cells. The results showed that the lipopeptides could completely inhibit the growth of K562 at 100 μM, with an IC50 value of 65.76 μM. The lipopeptides inhibited the profile of K562 via three pathways: (1) induction of paraptosis indicated by the occurrence of cytoplasmic vacuoles, and swelling of the mitochondria and endoplasmic reticulum (ER) without membrane blebbing in the presence of a caspase inhibitor; (2) inhibition of autophagy progress illustrated by the upregulated expression of LCII and P62; and (3) induction of apoptosis by causing ROS burst, and induction of the intrinsic pathway indicated by the upregulated expression of cytochrome c (Cyto-c), bax, and bad, together with downregulated expression of Bcl-2. The ROS-dependent apoptosis and caspase-independent paraptosis were verified using the ROS inhibitor and caspase inhibitor, respectively. The extrinsic apoptosis pathway was not involved in the lipopeptide’s effects on K562. Overall, the B. subtilis lipopeptides (consisting of a majority of iturin) exhibited promising potential in inhibiting chronic myelogenous leukemia in vitro via simultaneously causing paraptosis, apoptosis, and inhibition of autophagy.

Abstract To improve the production yield of (+)-pinoresinol (Pin), (+)-pinoresinol monoglucoside (PMG), and (+)-pinoresinol diglucoside (PDG), different methods were conducted, including co-culture with resveratrol-producing Alternaria sp. MG1 spores and addition of Tu-chung in a medium at the start of cultivation, ultrasound treatment (40 kHZ, 10 min) on 5-day culture, and addition of ethanol and sodium butyrate on Day 3, followed by cultivation for an additional period of 2 days. At the end of the cultivation period (5 days), the liquid phase was collected for product analysis. Cells were collected for the determination of gene expression levels and then used in bioconversion using resting cells for another period of 2 days. The liquid phase was measured to determine the output of the target products and the expression levels of the key genes related to the biosynthesis of these compounds. Consequently, co-culture with Alternaria MG1 and addition of Tu-chung bark in the medium efficiently increased Pin, PMG, and PDG production yield in the biosynthesis systems using potato dextrose broth medium and resting cells of Phomopsis sp. XP-8. The key genes related to the biosynthesis of these compounds were significantly upregulated. However, in the majority of cases, the addition of ethanol and sodium butyrate, and ultrasound treatment decreased the production yield of Pin, PMG, and PDG. The change in production yield was not consistently accompanied by a change in gene expression.

In the fall of 2017, an airborne field campaign was conducted from the NASA Armstrong Flight Research Center in Palmdale, California, to advance the remote sensing of aerosols and clouds with multi-angle polarimeters (MAP) and lidars. The Aerosol Characterization from Polarimeter and Lidar (ACEPOL) campaign was jointly sponsored by NASA and the Netherlands Institute for Space Research (SRON). Six instruments were deployed on the ER-2 high-altitude aircraft. Four were MAPs: the Airborne Hyper Angular Rainbow Polarimeter (AirHARP), the Airborne Multiangle SpectroPolarimetric Imager (AirMSPI), the Airborne Spectrometer for Planetary EXploration (SPEX airborne), and the Research Scanning Polarimeter (RSP). The remainder were lidars, including the Cloud Physics Lidar (CPL) and the High Spectral Resolution Lidar 2 (HSRL-2). The southern California base of ACEPOL enabled observation of a wide variety of scene types, including urban, desert, forest, coastal ocean, and agricultural areas, with clear, cloudy, polluted, and pristine atmospheric conditions. Flights were performed in coordination with satellite overpasses and ground-based observations, including the Ground-based Multiangle SpectroPolarimetric Imager (GroundMSPI), sun photometers, and a surface reflectance spectrometer.

61. An Algorithm for Hyperspectral Remote Sensing of Aerosols: 3. Application to the GEO-TASO Data in KORUS-AQ Field Campaign

This paper describes the third part of a series of investigations to develop algorithms for simultaneous retrieval of aerosol parameters and surface spectral reflectance from the data measured by GEOstationary Trace gas and Aerosol Sensor Optimization (GEO-TASO) instrument. Since the algorithm is designed for future hyperspectral and geostationary satellite sensors, such as Tropospheric Emissions: Monitoring of Pollution (TEMPO), it is applied to GEO-TASO data measured over the same area by different flights as part of the Korea-United Stated Air Quality Study (KORUS-AQ) field campaign in 2016. While GEO-TAOS has a spectral sampling interval of ~0.28 nm in the visible, its data is thinned through a band selection approach with consideration of atmospheric transmittance and different surface types, which yields 20 common spectral bands to be used by the algorithm. The algorithm starts with 4 common principal components (PCs) for surface spectral reflectance extracted from various spectral libraries, and the constraints of surface reflectance parameters and aerosol scattering models respectively from k- means clustering analysis of the Rayleigh-corrected GEO-TASO spectra and AERONET data. The algorithm then proceeds iteratively with an optimal estimation approach to update PCs and retrieve aerosol optical depth (AOD) until the best match between simulated and GEOS-TASO measured spectra is achieved. The spectral AODs are compared between those retrieved from GEO-TASO (x) and 7 AERONETS (y) in 440, 550, 550, 675 nm, respectively. The comparison reveals that iterative updates of surface spectral PCs (and so surface reflectance) yield significant enhancement in AOD retrievals, improving the mean linear fitting equations from y = 0.735x + 0.088 (without update of PCs) to y = 1.055x+ 0.01, and the Pearson correlation coefficient (𝑅𝑅2) from 0.54 to 0.76, respectively. Move case studies are need to further evaluate the algorithm for its application to TEMPO that carries a enhanced version of GEO-TASO instrument.

62. Influence of attapulgite addition on the biological performance and microbial communities of submerged dynamic membrane bioreactor

A submerged dynamic membrane bioreactor (sDMBR) was developed to test the influence of attapulgite (AT) addition on the treatment performances and the microbial community structure and function. The batch experimental results displayed the highest UV254 and dissolved organic carbon (DOC) removal efficiencies with 5% AT/mixed liquid suspended solids addition dosage. The continuous sDMBR results showed that the removal efficiencies of chemical oxygen demand, NH4+-N, total nitrogen and total phosphorus significantly increased in the AT added sDMBR. Excitation emission matrix analysis demonstrated that the protein-like peaks and fulvic acid-like peaks were significantly decreased in both in the mixed liquid and the effluent of the AT added reactor. The obligate anaerobes were observed in the sDMBR with AT addition, such as Bacteroidetes and Gamma proteobacterium in the dynamic membrane, which played an important role in the process of sludge granulation. Bacterial community richness significantly increased after AT addition with predominated phyla of Proteobacteria and Bacteroidetes. Similarly, species abundance significantly increased in the AT added sDMBR. Further investigations with cluster proved that AT was a favorite biological carrier for the microbial ecology, which enriched microbial abundance and community diversity of the sDMBR.

63. Rapidly increasing sulfate concentration: a hidden promoter of eutrophication in shallow lakes

Except for excessive nutrient input and climate warming, the rapidly rising SO42- concentration is considered as a crucial contributor to the eutrophication in shallow lakes; however, the driving process and mechanism are still far from clear. In this study, we constructed a series of microcosms with initial SO42- concentrations of 0, 30, 60, 90, 120, and 150 mg L−1 to simulate the rapid SO42- increase in Lake Taihu, China, subjected to cyanobacteria blooms. Results showed that the sulfate reduction rate was stimulated by the increase in initial SO42- concentrations and cyanobacteria-derived organic matter, with the maximal sulfate reduction rate of 39.68 mg (L d)−1 in the treatment of 150 mg L−1 SO42- concentration. During the sulfate reduction, the produced maximal ∑S2− concentration in the overlying water and acid volatile sulfate (AVS) in the sediments were 3.15 mg L−1 and 11.11 mg kg−1, respectively, and both of them were positively correlated with initial SO42- concentrations (R2=0.97; R2=0.92). The increasing abundance of sulfate reduction bacteria (SRB) was also linearly correlated with initial SO42- concentrations (R2=0.96), ranging from 6.65×107 to 1.97×108 copies g−1. However, the Fe2+ concentrations displayed a negative correlation with initial SO42- concentrations, and the final Fe2+ concentrations were 9.68, 7.07, 6.5, 5.57, 4.42, and 3.46 mg L−1, respectively. As a result, the released total phosphorus (TP) in the overlying water, to promote the eutrophication, was up to 1.4 mg L−1 in the treatment of 150 mg L−1 SO42- concentration. Therefore, it is necessary to consider the effect of rapidly increasing SO42- concentrations on the release of endogenous phosphorus and the eutrophication in lakes.

Context Sulfate (SO42−) concentrations in eutrophic lakes are continuously increasing. Aims This study aimed to explore the effects on nitrogen and their limiting factors in eutrophic lakes. Methods We constructed a series of microcosms with different initial SO42− concentrations (0–180 mg L−1) to simulate the SO42− concentration in eutrophic lakes. Key results The NO3−-N concentration rapidly decreased in all treatments. The respective 2.21 and 2.77 mg L−1 concentrations of NO3−-N recorded on Day 5 in the initial 150 and 180 mg L−1 SO42− concentration treatments were higher than in other treatments. The highest N2O concentrations in all treatments showed insignificant differences. The existence of free Fe2+ that was released from iron reduction resulted in an insignificant effect of increased SO42− concentration on nitrogen dynamics. With a decrease in Fe2+, the acid volatile sulfate (AVS) increased, and the highest AVS concentrations were positively correlated with the initial SO42− concentration (7.20–12.88 mg kg−1). Conclusion This effect could be ascribed to the fact that the rapid combination of sulfide (∑S2−) and Fe2+ prevents ∑S2− from producing a toxic effect on the denitrification. Implications Our study highlighted the role of iron in eutrophic lakes, especially in the coupling of sulfate reduction and nitrogen dynamics.

Background: Psoriasis is a chronic immune-mediated inflammatory skin disease, with over-activated interleukin (IL)-17-producing CD4+ T cells (Th17) and repressed regulatory T (Treg) cells. IL-21 is a Th17-related cytokine and plays an important role in the pathogenesis of psoriasis. However, the mechanism by which IL-21 affects the pathogenic progress of psoriasis remains poorly understood.Methods: IL-21 and IL-21 receptor (IL-21R) expression in normal and psoriatic lesional skin were determined by immumohistochemical staining, immunofluorescence staining, and western blotting. The levels of IL-21, IL-17A, and IL-22 in the culture supernatants were measured by enzyme-linked immunosorbent assay (ELISA). The level of IL-10 in the culture supernatants was measured by cytometric bead array (CBA). The mRNA expression levels were assessed by quantitative polymerase chain reaction (qPCR). CD4+ T cells were isolated from the peripheral blood mononuclear cells (PBMCs) from the psoriasis patients and healthy individuals and then treated with or without IL-21 for 3 days. The proportions of Th17 and Treg cells were determined by flow cytometric analysis.Results: IL-21 and IL-21R were highly expressed in the lesional skin and peripheral blood of psoriasis patients. IL-21 promoted CD4+ T cells proliferation and Th17 cells differentiation and inhibiting Treg cells differentiation by upregulating RORγt expression and downregulating Foxp3 expression, with increased expression and secretion of IL-17A and IL-22. The proportion of Treg cells was negatively correlated with that of Th17 cells in psoriasis patients.Conclusion: Our results suggest that IL-21 may promote psoriatic inflammation by inducing imbalance in Th17 and Treg cell populations.

NASA’s Plankton, Aerosol, Clouds, ocean Ecosystem (PACE) satellite mission is scheduled to launch in 2022, with the Ocean Color Instrument (OCI) on board. For the first time reflected sunlight from the Earth across a broad spectrum from the ultraviolet (UV: 350 nm) to the short wave infrared (SWIR: 2260 nm) will be measured from a single instrument at 1 km spatial resolution. While seven discrete bands will represent the SWIR, the spectrum from 350 to 890 nm will be continuously covered with a spectral resolution of 5 nm. OCI will thus combine in a single instrument (and at an enhanced spatial resolution for the UV) the heritage capabilities of the Moderate resolution Imaging Spectroradiometer (MODIS) and the Ozone Monitoring Instrument (OMI), while covering the oxygen A-band (O2A). Designed for ocean color and ocean biology retrievals, OCI also enables continuation of heritage satellite aerosol products and the development of new aerosol characterization from space. In particular the combination of MODIS and OMI characteristics allows deriving aerosol height, absorption and optical depth along with a measure of particle size distribution. This is achieved by using the traditional MODIS visible-to-SWIR wavelengths to constrain spectral aerosol optical depth and particle size. Extrapolating this information to the UV channels allows retrieval of aerosol absorption and layer height. A more direct method to derive aerosol layer height makes use of O2A absorption methods, despite the relative coarseness of the nominal 5 nm spectral resolution of OCI. Altogether the PACE mission with OCI will be an unprecedented opportunity for aerosol characterization that will continue climate data records from the past decades and propel aerosol science forward toward new opportunities.

To better understand its mechanism of activity towards higher alcohols, we overexpressed and purified new Geotrichum candidum GDH (GcGDH). The purified GcGDH (50.27 kDa) was then crystallized, and the crystal diffracted to a resolution of 2.3 Å using X-ray diffraction. We found that the GcGDH crystal structure belonged to space group P212121 and was comprised of two hexamers organized into an asymmetric unit, with each subunit consisting of 452 amino acid residues. The binding sites between higher alcohols or L-glutamic acid and GcGDH were consistent. The optimal reaction conditions for GcGDH and hexanol were a pH of 4.0 and temperature of 30 °C, and those for GcGDH and monosodium glutamate (MSG) were a pH of 8.0 and temperature of 20 °C. The Km values for hexanol and MSG were found to be 74.78 mM and 0.018 mM, respectively. Mutating GcGDH Lys 113 to either Ala or Gly caused a dramatic reduction in its catalytic efficiency towards both MSG and hexanol, suggesting that Lys 113 is essential to the active site of GcGDH.

The Airborne Hyper-Angular Rainbow Polarimeter (AirHARP) is a new imaging polarimeter instrument, capable of sampling a single Earth target from up to 120 viewing angles, in four spectral channels, and three linear polarization states across a 114° field of view. AirHARP is telecentric in the image space and simultaneously images three linear polarization states with no moving parts. These two aspects of the design allow for a simple and efficient quantitative calibration. Using coefficients derived at the center of the lens and the detector flatfields, we can calibrate the entire AirHARP sensor in a variety of lab, field, and space environments. We describe the calibration process for the HARP family of polarimeters using AirHARP pre-launch data. We show that this telecentric calibration technique yields a 0.25 % RMS degree of linear polarization (DOLP) accuracy in all channels for pixels around the detector center. To validate across the FOV, we compare our multi-angle reflectance and polarization data with the Research Scanning Polarimeter (RSP) over targets sampled during the NASA Aerosol Characterization from Polarimeter and Lidar (ACEPOL) campaign. For the majority of angles and targets used in the intercomparison, RSP and AirHARP agree better than 1 % in reflectance and DOLP. Therefore, our calibration successfully transfers nadir coefficients to different FOVs, given ambient challenges. This calibration technique makes the HARP design attractive for new spaceborne climate missions: HARP CubeSat (2020–2022), HARP2 (2024–) on the NASA Plankton-Aerosol-Cloud-ocean Ecosystem (PACE), Atmosphere Observing System (AOS) and beyond.

The separation and transfer of the photoinduced charge carriers are essential for the high-efficient photocatalytic materials. Heterostructures which compose of several semiconductors can take advantage of energy level difference to improve photocatalytic properties. However, the fabrication of the related heterostructures normally adopts the physical vapor deposition method, such as atomic layer deposition, hindering the mass production of photocatalytic materials. In this work, ZnO nanorod arrays and ZnO/Al _2 O _3 heterostructure were prepared on Si (100) substrates by a cost-efficient method which combines the hydrothermal and photochemical techniques. Microstructure and surface composition analysis on the heterostructure confirm the synthesis of the ZnO/Al _2 O _3 core–shell heterostructure. According to the ultraviolet (UV)–visible diffuse reflectance spectroscopy, the band gap of the ZnO/Al _2 O _3 heterostructure can be estimated to be 3.25 eV, which is larger than that of pure ZnO nanorod arrays. Moreover, the heterostructure exhibits an obviously enhanced photocatalytic property for photodegradation of methyl orange under UV-irradiation. The energy band diagram of heterostructure shows that the improvement of photocatalytic property can be ascribed to the increase in the electron-hole pair separation rate. This work provides a simple method to fabricate nanoscale semiconductor heterostructures for photocatalytic applications.

Emerging evidence indicates that the gut microbiome contributes to the host immune response to infectious diseases. Here, to explore the role of the gut microbiome in the host immune responses in COVID-19, we conducted shotgun metagenomic sequencing and immune profiling of 14 severe/critical and 24 mild/moderate COVID-19 cases as well as 31 healthy control samples. We found that the diversity of the gut microbiome was reduced in severe/critical COVID-19 cases compared to mild/moderate ones. We identified the abundance of some gut microbes altered post-SARS-CoV-2 infection and related to disease severity, such as Enterococcus faecium, Coprococcus comes, Roseburia intestinalis, Akkermansia muciniphila, Bacteroides cellulosilyticus and Blautia obeum. We further analyzed the correlation between the abundance of gut microbes and host responses, and obtained a correlation map between clinical features of COVID-19 and 16 severity-related gut microbe, including Coprococcus comes that was positively correlated with CD3

Aims Copper ion is widespread in wastewater and threatens the condition and human health. Micro-organisms have unique advantages to remove heavy-metal ions from water, but are rarely reported in the removal of copper ion. This aims to develop micro-organisms that can remove copper ion in water, characterize their properties and analyse their potential application in practice. Methods and Results Sewage sludge was used as the source to isolate wild bacteria that can remove copper ion in water. The most efficient strain was screened out from 23 obtained isolates, identified as Bacillus pseudomycoides and coded as C6. The properties of C6 in the removal of copper ion in water were investigated in the aspects of reaction conditions, reaction groups, reaction dynamic and the application in oat planting. The reaction at pH 7 within 10 min yielded the highest removal rate of copper ion, 83%. The presence of lead ion in the reaction system could promote the removal rate of copper ion. Carboxyl groups and amidogen of C6 biomass were mainly involved in the removal of copper ion. The removal of copper ion was in accord with single-layer adsorption and Langmuir adsorption isotherm model. In application, C6 biomass reduced the copper content in the oat seedlings grown in copper ion containing water by more than seven times. Conclusions B. pseudomycoides C6 can efficiently remove copper ion in water and inhibit it from entering plants. Significance and Impact of Study This is the first time to report the capability of B. pseudomycoides to remove copper ion in water, which is also more efficient than the currently reported chemical and biological methods.

Pathogenic microbes can induce cellular dysfunction, immune response, and cause infectious disease and other diseases including cancers. However, the cellular distributions of pathogens and their impact on host cells remain rarely explored due to the limited methods. Taking advantage of single-cell RNA-sequencing (scRNA-seq) analysis, we can assess the transcriptomic features at the single-cell level. Still, the tools used to interpret pathogens (such as viruses, bacteria, and fungi) at the single-cell level remain to be explored. Here, we introduced PathogenTrack, a python-based computational pipeline that uses unmapped scRNA-seq data to identify intracellular pathogens at the single-cell level. In addition, we established an R package named Yeskit to import, integrate, analyze, and interpret pathogen abundance and transcriptomic features in host cells. Robustness of these tools has been tested on various real and simulated scRNA-seq datasets. PathogenTrack is competitive to the state-of-the-art tools such as Viral-Track, and the first tools for identifying bacteria at the single-cell level. Using the raw data of bronchoalveolar lavage fluid samples (BALF) from COVID-19 patients in the SRA database, we found the SARS-CoV-2 virus exists in multiple cell types including epithelial cells and macrophages. SARS-CoV-2-positive neutrophils showed increased expression of genes related to type I interferon pathway and antigen presenting module. Additionally, we observed the Haemophilus parahaemolyticus in some macrophage and epithelial cells, indicating a co-infection of the bacterium in some severe cases of COVID-19. Antibiotics can be absorbed by aquatic plants, but they seriously affect the health of aquatic plants and threaten the steady state of aquatic ecosystem. The phytoremediation performance and physiology of floating macrophyte (Eichhornia crassipes) under antibiotic ciprofloxacin (CIP) hydroponic conditions were investigated. It was found that CIP absorption of E. crassipes was up to 84.38% and the root was the main absorption tissue. Hydrolysis and microbial degradation were the second removal pathway of CIP followed the plant absorption. After 7 days of CIP exposure, the photosynthesis efficiency of E. crassipes remained stable, and the presence of CIP did not inhibit the growth of the plant. On the 14th day, the superoxide dismutase and catalase activities were increased in response to the CIP stress. However, the tender leaves of E. crassipes turned white and shrivel, attributed to a decrease in chlorophyll content and chlorophyll fluorescence parameters after 21 days of CIP exposure. These findings will have significant implications for E. crassipes to absorb CIP on a limited time-scale and provide a phytoremediation technology for antibiotics in water.

In this work, we report the resistive switching behavior of an amorphous La2Ti2O7 (LTO) film as sandwiched between two Pt electrodes. The resistive switching is forming-free and highly uniform. Furthermore, it exhibits self-rectifying resistive switching behaviors owing to the Schottky contact and quasi-ohmic contact formed at the top and bottom interfaces, respectively. The mechanism of switching behavior in the device is attributed to the trapping/detrapping-mediated electronic bipolar resistance switching. By fitting the current-voltage characteristics, it indicates the coexistent conduction mechanisms of Schottky emission and space-charge-limited-conduction (SCLC), while the Schottky barrier modified by electron trapping/detrapping plays a dominating role in the resistive switching process.

In order to understand the real environment and specific needs of China’s current tobacco industry, master the signal information in the tobacco supply chain, and timely discover the source of the tobacco supply chain, this paper puts forward the detection method of anti-counterfeiting at the source of the tobacco supply chain. Based on this, this article constructs a nonlinear discrete system model, which mainly studies the logistics information existing in the tobacco supply chain, utilizes the noise existing in the tobacco supply chain information obtained by EKF filtering, and combines it with Support vector machine data analysis method. The results showed that out of 450 sample tests, 0 real sample errors and 1 misuse of 49 false samples, with a total detection rate of up to 100% was achieved. The results show that the nonlinear discrete system model is better, has higher detection effect of tobacco supply source, and improves the detection accuracy.

Background and aims Observational studies have linked elevated blood pressure (BP) to impaired cognitive function. However, the functional and structural changes in the brain that mediate the relationship between BP elevation and cognitive impairment remain unknown. Using observational and genetic data from large consortia, this study aimed to identify brain structures potentially associated with BP values and cognitive function. Methods and results Data on BP were integrated with 3935 brain magnetic resonance imaging-derived phenotypes (IDPs) and cognitive function defined by fluid intelligence score. Observational analyses were performed in the UK Biobank and a prospective validation cohort. Mendelian randomisation (MR) analyses used genetic data derived from the UK Biobank, International Consortium for Blood Pressure, and COGENT consortium. Mendelian randomisation analysis identified a potentially adverse causal effect of higher systolic BP on cognitive function [−0.044 standard deviation (SD); 95% confidence interval (CI) −0.066, −0.021] with the MR estimate strengthening (−0.087 SD; 95% CI −0.132, −0.042), when further adjusted for diastolic BP. Mendelian randomisation analysis found 242, 168, and 68 IDPs showing significant (false discovery rate P < 0.05) association with systolic BP, diastolic BP, and pulse pressure, respectively. Most of these IDPs were inversely associated with cognitive function in observational analysis in the UK Biobank and showed concordant effects in the validation cohort. Mendelian randomisation analysis identified relationships between cognitive function and the nine of the systolic BP-associated IDPs, including the anterior thalamic radiation, anterior corona radiata, or external capsule. Conclusion Complementary MR and observational analyses identify brain structures associated with BP, which may be responsible for the adverse effects of hypertension on cognitive performance.

Although vanadium dioxide (VO2) exhibits the most abrupt metal to insulator transition (MIT) properties near room-temperature, the present regulation of their MIT functionalities is insufficient owing to the high complexity and susception associated with V4+. Herein, we demonstrate a spark plasma assisted reactive sintering (SPARS) approach to simultaneously achieve in situ doping and sintering of VO2 within largely short period (~10 minutes). This enables high convenience and flexibility in regulating the electronic structure of VO2 via dopant elements covering Ti, W, Nb, Mo, Cr and Fe, leading to a wide adjustment in their metal to insulator transition temperature (TMIT) and basic resistivity. Furthermore, the mechanical strengths of the doped-VO2 were meanwhile largely improved via the compositing effect of high melting-point dopant oxide. The high adjustability in MIT properties and improved mechanical properties further paves the way towards practical applications of VO2 in power electronics, thermochromism and infrared camouflage.

An accurate estimation of biomass burning emissions is 1 partially limited by the lack of knowledge of fire burning phase (smoldering vs. flaming). In recent years, several fire detection products have been developed to provide information of fire radiative power (FRP), location, size, and temperature of fire pixels, but no information regarding fire burning phase is retrieved. The Day-Night band (DNB) aboard Visible Infrared Imaging Radiometer Suite (VIIRS) is sensitive to visible light from flaming fires in nighttime scenes. In contrast, VIIRS 4 m moderate resolution band #13 (M13), though capable of detecting fires at all phases, has no direct sensitivity for discerning fire phase. However, the hybrid usage of VIIRS DNB and M-bands data is hampered by their different scanning technology and spatial resolution. In this study, we present a novel method to rapidly and accurately resample DNB pixel radiances to the footprint of M-band pixels, accounting for onboard detector aggregation schemes and bowtie effect removals. The visible energy fraction (VEF) is subsequently introduced as an indicator of fire burning phase VEF is calculated as the ratio of visible light power (VLP) to FRP for each fire pixel retrieved from the VIIRS 750 m active fire product. A global distribution of VEF values, and thereby the fire phase, is quantitatively obtained, showing smaller VEF values in regions with mostly smoldering wildfires, such as peatland fires in Indonesia, larger VEF values in regions with flaming wildfires over grasslands and savannas in the sub-Sahelian region, and the largest VEF values associated with gas flaring in the Middle East. Mean VEF for different land cover types or regions is highly correlated with modified combustion efficiency (MCE). These results, together with a case study of the 2018 California Camp Fire, show that the VEF has the potential to be an indicator of fire combustion phase for each fire pixel, appropriate for estimating emission factors at the satellite pixel level.

We present an algorithm for retrieving aerosol layer height (ALH) and aerosol optical depth (AOD) for smoke over vegetated land and water surfaces from measurements of the Earth Polychromatic Imaging Camera (EPIC) onboard the Deep Space Climate Observatory (DSCOVR). The algorithm uses Earth-reflected radiances in six EPIC bands in the visible and near-infrared and incorporates flexible spectral fitting that accounts for specifics of land and water surface reflectivity. The fitting procedure first determines AOD using EPIC atmospheric window bands (443 nm, 551 nm, 680nm, and 780 nm), then uses oxygen (O2) A and B bands (688 nm and 764 nm) to derive ALH, which represents an optical centroid altitude. ALH retrieval over vegetated surface primarily takes advantage of measurements in the O2B band. We applied the algorithm to EPIC observations of several biomass burning events over the United States and Canada in August 2017. We found that the algorithm can be used to obtain AOD and ALH multiple times daily over water and vegetated land surface. Validation is performed against aerosol extinction profiles detected by the Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) and against AOD observed at nine Aerosol Robotic Network (AERONET) sites, showing, on average, an error of 0.58 km and a bias of -0.13 km in retrieved ALH and an error of 0.05 and a bias of 0.03 in retrieved AOD. Additionally, we show that the aerosol height information retrieved by the present algorithm can potentially benefit the retrieval of aerosol properties from EPIC’s ultraviolet (UV) bands.

The discovery of topological Weyl semimetals has revealed opportunities to realize several extraordinary physical phenomena in condensed matter physics. Specifically, Weyl semimetals with strong spin-orbit coupling, broken inversion symmetry, and novel spin textures are predicted to exhibit a large spin Hall effect that can efficiently convert the charge current to a spin current. Here, we report a direct experimental observation of large spin Hall and inverse spin Hall effects in the Weyl semimetal WTe_{2} at room temperature obeying the Onsager reciprocity relation. We demonstrate the detection of a pure spin current generated by the spin Hall phenomenon in WTe_{2} by making a van der Waals heterostructure with graphene, taking advantage of its long spin coherence length and spin transmission at the heterostructure interface. These experimental findings, well supported by ab initio calculations, show a large charge-spin conversion efficiency in WTe_{2}, which can pave the way for the utilization of spin-orbit-induced phenomena in spintronic memory and logic circuit architectures.

95. Migration and transformation of dissolved carbon during accumulated cyanobacteria decomposition in shallow eutrophic lakes: a simulated microcosm study

The decomposition processes of accumulated cyanobacteria can release large amounts of organic carbon and affect the carbon cycling in shallow eutrophic lakes. However, the migration and transformation mechanisms of dissolved carbon (DC) require further study and discussion. In this study, a 73-day laboratory microcosm experiment using suction samplers (Rhizon and syringe) was conducted to understand the migration and transformation of DC during the cyanobacteria decomposition. The decomposition of cyanobacteria biomass caused anoxic and reduction conditions, and changed the acid-base environment in the water column. During the early incubation (days 0–18), a large amount of cyanobacteria-derived particulate organic matter (POM) was decomposed into dissolved organic carbon (DOC) in the overlying water, reaching the highest peak value of 1.82 g L−1 in the treatment added the high cyanobacteria biomass (470 g). After 18 days of incubation, the mineralization of increased DOC to dissolved inorganic carbon (DIC) maintained a high DIC level of overlying water in treatments added cyanobacteria biomass. The treatment added the medium cyanobacteria biomass (235 g) presented the lower DOC/total dissolved carbon ratio than the high cyanobacteria biomass associated with the lower mineralization from DOC to DIC. Due to the concentration differences of DIC at water-sediment interface, the main migration of DIC from pore water to overlying water occurred in the treatment without added cyanobacteria biomass. However, the treatments added the cyanobacteria biomass presented the obvious diffusion of DOC and the low migration of DIC at the water-sediment interface. The diffusive fluxes of DOC at the water-sediment interface increased with the cyanobacteria biomass added, reaching the maximum value of 411.01 mg/(m2·d) in the treatment added the high cyanobacteria biomass. In the overlying water, the group added the sediment and medium cyanobacteria biomass presented a faster degradation of cyanobacteria-derived POM to DOC and a higher mineralization level of DOC to DIC than added the medium cyanobacteria biomass without sediment. Therefore, during accumulated cyanobacteria decomposition, the biomass of accumulated cyanobacteria and sediment property can influence the migration and transformation of DC, playing an important role in carbon cycling in shallow eutrophic lakes.

Abatract The optimal particle size and surface states are important to superparamagnetic nanoparticles for magnetic resonance imaging (MRI) contrast agents. Here, a simple one-pot approach is proposed to prepare ultrasmall hydrophobic MnFe2O4 superparamagnetic nanoparticles with a uniform diameter of about 4.7 nm. To satisfy the requirements for MRI contrast agents, the MnFe2O4 nanoparticles are reduced by chitosan to form MnFe2O4@chitosan nanoparticles (MnFe2O4@CS NPs), which dramatically increases the saturation magnetization (Ms) of the nanoparticles by 21%. The valence state analysis indicates that the amino group (NH2) of chitosan shares lone pair electrons with Mn4+, resulting in high-spin state Mn3+ to enhance the Ms. The MnFe2O4@CS NPs show significant signal reduction and high r2 value in the T2-weighted imaging. Therefore, due to their large specific surface area and high responsiveness to the external magnetic field, MnFe2O4@CS NPs are expected to be applied to nano platform in magnetic targeting therapy with the function of MRI contrast agents in vivo.

Many studies have associated altered intestinal bacterial communities and non-alcoholic fatty liver disease, but the putative effects are inconclusive. The purpose of this network meta-analysis (NMA) was to evaluate the effects of probiotics, prebiotics, and synbiotics on non-alcoholic fatty liver disease through randomised intervention trials. Literature searches were performed until March 2020. For each outcome, a random NMA was performed, the surface under the cumulative ranking curve (SUCRA) was determined. A total of 22 randomised trials comparing prebiotic, probiotic, and synbiotic treatments included 1301 participants. Considering all seven results (aspartate aminotransferase, alanine aminotransferase, body mass index, weight, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol) together, the highest SUCRA values are probiotics (94%), synbiotics (61%) and prebiotics (56%), respectively. NMA results provide evidence that probiotics, prebiotics, and synbiotics can alleviate non-alcoholic fatty liver disease. However, due to the lack of high-quality randomised trials, this research also has some limitations.
