Aminopterin (AMT) is a kind of universal antineoplastic drugs, but it has severe toxic and side effects, leaving it rarely used in clinic. Herein, we found that cucurbit[7]uril (CB[7]) and AMT can form stable inclusion complexes, and the formation of CB[7]-AMT 2:1 supramolecular inclusion complex was confirmed by UV-visible absorption spectra, fluorescence spectra, and molecular modeling calculations in aqueous solution. Binding stability constants (Ks) were determined by UV-visible and fluorescence spectra method, with 1.97 × 105L·mol–1and 2.29 × 105 L·mol–1, respectively. The binding energy was calculated to be 28.8 kcal·mol–1 for the CB[7]-AMT complex. And then, through a series of cell experiments of CCK8 assay, DAPI staining and hoechst33342/PI double staining, we fully proved that the CB[7]-AMT complex can reduce the toxicity of AMT to normal cells such as hepatocyte line LO2, and improve its anticancer effect on cancer cells overexpressing spermine, typically like human colon cancer cell line HCT116. It confirmed that the CB[7]-AMT complex had the effect of reducing toxicity and increasing efficiency. These results indicated that CB[7]-AMTinclusion complex might be a promising novel formulation of AMT for its clinical development.

Biochar application to the soil is a novel approach to carbon sequestration. Biochar application affects the emission of greenhouse gases (GHGs), such as CO2, CH4, and N2O, from different environments (e.g. upland soils, rice paddies and wetlands, and composting environments). In this review, the effect of biochar on GHGs emissions from the above three typical environments are critically evaluated based on a literature analysis. First, the properties of biochar and engineered biochar related to GHGs emissions was reviewed, targeting its relationship with climate change mitigation. Then, a meta-analysis was conducted to assess the effect of biochar on the emissions of CO2, CH4, and N2O in different environments, and the relevant mechanisms. Several parameters were identified as the main influencing factors in the meta-analysis, including the pH of the biochar, feedstock type, pyrolysis temperature, biochar application rate, C/N ratio of the biochar, and experimental scale. An overall suppression effect among different environments was found, in the following order for different greenhouse gases: N2O > CH4 > CO2. We conclude that biochar can change the physicochemical properties of soil and compost in different environments, which further shapes the microbial community in a specific environment. Biochar addition affects CO2 emissions by influencing oligotrophic and copiotrophic bacteria; CH4 emissions by regulating the abundance of functional genes, such as mcrA (a methanogen) and pmoA (a methanotroph); and N2O emissions by controlling N-cycling functional genes, including amoA, nirS, nirK, nosZ. Finally, future research directions for mitigating greenhouse gas emissions through biochar application are suggested.

Agroforestry systems (AFS) contribute to carbon (C) sequestration and reduction in greenhouse gas emissions from agricultural lands. However, previously understudied differences among AFS may underestimate their climate change mitigation potential. In this 3-year field study, we assessed various C stocks and greenhouse gas emissions across two common AFS (hedgerows and shelterbelts) and their component land uses: perennial vegetated areas with and without trees (woodland and grassland, respectively), newly planted saplings in grassland, and adjacent annual cropland in central Alberta, Canada. While treatments that induce DNA damage are commonly used as anticancer therapies, the mechanisms through which DNA damage produces a therapeutic response are incompletely understood. Here we have tested whether medulloblastomas must be competent for apoptosis to be sensitive to radiotherapy. Whether apoptosis is required for radiation sensitivity has been controversial. Medulloblastoma, the most common malignant brain tumor in children, is a biologically heterogeneous set of tumors typically sensitive to radiation and chemotherapy; 80% of medulloblastoma patients survive long-term after treatment. We used functional genetic studies to determine whether the intrinsic apoptotic pathway is required for radiation to produce a therapeutic response in mice with primary, Shh-driven medulloblastoma. We found that cranial radiation extended the survival of medulloblastoma-bearing mice and induced widespread apoptosis. Expression analysis and conditional deletion studies showed that Trp53 (p53) was the predominant transcriptional regulator activated by radiation and was strictly required for treatment response. Deletion of Bax, which blocked apoptosis downstream of p53, was sufficient to render tumors radiation resistant. In apoptosis-incompetent, Bax-deleted tumors, radiation activated p53-dependent transcription without provoking cell death and caused two discrete populations to emerge. Most radiated tumor cells underwent terminal differentiation. Perivascular cells, however, quickly resumed proliferation despite p53 activation, behaved as stem cells, and rapidly drove recurrence. These data show that radiation must induce apoptosis in tumor stem cells to be effective. Mutations that disable the intrinsic apoptotic pathways are sufficient to impart radiation resistance. We use the X-ray luminosity relation of radio-loud quasars (RLQs) to measure these luminosity distances as well as estimate cosmological parameters. We adopt four parametric models of X-ray luminosity to test luminosity correlation for RLQs and radio-intermediate quasars (RIQs) and give these cosmological distances. By Bayesian information criterion (BIC), the data suggest that the luminosity relation ${L_X} \propto L_{\rm UV}^{{\gamma _{\rm uv}}}L_{\rm Radio}^{\gamma _{\rm radio}^{\prime }}$ for RLQs has a better goodness of fit, relative to other models, which can be interpreted as this relation being preferred for RLQs. Meanwhile, we compare the results from flat-spectrum radio-loud quasars (FSRLQs) and steep-spectrum radio-loud quasars (SSRLQs), which indicate that their luminosity correlations are not exactly the same. We also consider dividing the RLQ sample into various redshift bins, which can be used to check if the X-ray luminosity relation depends on the redshift. Finally, we apply a combination of RLQs and SN Ia Pantheon to verify the nature of dark energy concerning whether or not its density deviates from the constant, and give the statistical results.

A multi-institutional investigation for dosimetric evaluation of high-Z hip prosthetic device in photon beam.A bilateral hip prosthetic case was chosen. An in-house phantom was built to replicate the human pelvis with two different prostheses. Dosimetric parameters: dose to the target and organs at risk (OARs) were compared for the clinical case generated by various treatment planning system (TPS) with varied algorithms. Single beam plans with different TPS for phantom using 6 MV and 15 MV photon beams with and without density correction were compared with measurement.Wide variations in target and OAR dosimetry were recorded for different TPS. For clinical case ideal PTV coverage was noted for plans generated with Corvus and Prowess TPS only. However, none of the TPS were able to meet plan objective for the bladder. Good correlation was noticed for the measured and the Pinnacle TPS for corrected dose calculation at the interfaces as well as the dose ratio in elsewhere. On comparing measured and calculated dose, the difference across the TPS varied from -20% to 60% for 6 MV and 3% to 50% for the 15 MV, respectively.Most TPS do not provide accurate dosimetry with high-Z prosthesis. It is important to check the TPS under extreme conditions of beams passing through the high-Z region. Metal artifact reduction algorithms may reduce the difference between the measured and calculated dose but still significant differences exist. Further studies are required to validate the calculational accuracy.

We carried out a high time-resolution, multicolour optical observing campaign for eight $\gamma$-ray detected blazars during 2010-2020. We analyze flux variations, correlations between magnitudes and colours on different timescales. Intraday variability (IDV) is detected in all eight sources of our sample. A bluer-when-brighter (BWB) chromatic trend is dominant on intraday timescales. On the short timescales, the BWB trend only shows up in ON 231, 3C 279, BL Lacertae and 1E 1458.8+2249. There is a BWB trend in 3C 279 on the long timescale. We estimate the upper limits of black hole mass for three blazars (i.e. ON 321, 1ES 1426+42.8, PKS 1510-089) using variability timescales. On April 13, 2010 a potential quasi-periodic oscillation (QPO) with the period of $P=48.67\pm13.90$ minutes is found in 1ES 1426+42.8. The light curve on March 16, 2021 further shows the existence of the QPO phenomenon. Responses of the terrestrial biosphere to rapidly changing environmental conditions are a major source of uncertainty in climate projections. In an effort to reduce this uncertainty, a wide range of global change experiments have been conducted that mimic future conditions in terrestrial ecosystems, manipulating CO2 , temperature, and nutrient and water availability. Syntheses of results across experiments provide a more general sense of ecosystem responses to global change, and help to discern the influence of background conditions such as climate and vegetation type in determining global change responses. Several independent syntheses of published data have yielded distinct databases for specific objectives. Such parallel, uncoordinated initiatives carry the risk of producing redundant data collection efforts and have led to contrasting outcomes without clarifying the underlying reason for divergence. These problems could be avoided by creating a publicly available, updatable, curated database. Here, we report on a global effort to collect and curate 57,089 treatment responses across 3644 manipulation experiments at 1145 sites, simulating elevated CO2 , warming, nutrient addition, and precipitation changes. In the resulting Manipulation Experiments Synthesis Initiative (MESI) database, effects of experimental global change drivers on carbon and nutrient cycles are included, as well as ancillary data such as background climate, vegetation type, treatment magnitude, duration, and, unique to our database, measured soil properties. Our analysis of the database indicates that most experiments are short term (one or few growing seasons), conducted in the USA, Europe, or China, and that the most abundantly reported variable is aboveground biomass. We provide the most comprehensive multifactor global change database to date, enabling the research community to tackle open research questions, vital to global policymaking. The MESI database, freely accessible at doi.org/10.5281/zenodo.7153253, opens new avenues for model evaluation and synthesis-based understanding of how global change affects terrestrial biomes. Converting rice husk to biochar is one of the solutions to manage crop residues by transforming waste into a value-added material that has broad benefits to the environment when biochar is applied to the soil. This study investigated the impact of the application of rice husk and its biochar at different doses (i.e., 0, 10, and 30 t ha−1) on soil carbon stability, the property of water-soluble soil organic matter, and the abundance and diversity of microbial communities in a Lei bamboo (Phyllostachys praecox) forest soil 262 days after their application. The application of rice husk, especially at 30 t ha−1, increased dissolved organic carbon due to the high labile carbon (C) (e.g., cellulose, hemicellulose, polysaccharides) content in the rice husk. The biochar treatments stimulated the release of humic-like substances (e.g., (poly) phenols) into the soil solution, increased the aromatic C content by 412–557%, and increased the relative abundance of Chloroflexi, Planctomycetota, and Proteobacteria compared to the control. This study shows that biochar application, particularly at 30 t ha−1, enhanced the C stability by turning organic C into recalcitrant forms in the soil, demonstrating the merit of converting rice husk into biochar before its application to the soil.
