Abstract

为加快辣木籽多糖提取速率，以辣木籽为原料，采用内部沸腾法提取其中的多糖。采用单因素实验和响应面实验对内部沸腾法提取辣木籽多糖的工艺条件进行优化，通过自由基清除活性考察其抗氧化活性。结果表明：最优提取工艺条件为解吸剂乙醇体积分数20%、蒸馏水提取温度74 ℃、提取时间5 min、料液比1∶ 27，在此条件下辣木籽多糖得率可达12.5%；质量浓度为5.0 mg/mL的辣木籽多糖对羟自由基的清除率为91.1%，质量浓度为1.0 mg/mL的辣木籽多糖对DPPH自由基的清除率为38%；质量浓度为5.0 mg/mL的辣木籽多糖对超氧阴离子自由基的清除率为59.8%。综上，辣木籽多糖具有一定的抗氧化活性。 In order to accelerate the extraction rate of Moringa oleifera seed polysaccharide, the polysaccharide was extracted from Moringa oleifera seed by internal boiling method. Single factor experiment and response surface methodology were used to optimize the internal boiling extraction conditions of Moringa oleifera seed polysaccharide, and the antioxidant activity of free radical scavenging activity was studied. The results showed that the optimal extraction conditions were obtained as follows:volume fraction of desorbent ethanol 20%, distilled water extraction temperature 74 ℃, extraction time 5 min, and solid-liquid ratio 1∶ 27. Under the optimal conditions, the yield of Moringa oleifera seed polysaccharide could reach 12.5%. The results of antioxidant activity study showed that the hydroxyl free radical scavenging rate of Moringa oleifera polysaccharide with a mass concentration of 5.0 mg/mL was 91.1%. The DPPH scavenging rate of Moringa oleifera polysaccharide with a mass concentration of 1.0 mg/mL was 38%. Superoxide anion scavenging rate of Moringa oleifera polysaccharide with a mass concentration of 5.0 mg/mL was 59.8%. In conclusion, Moringa oleifera seed polysaccharide has some antioxidant activity.

60. Hepatoprotective role of peroxisome proliferator-activated receptor-α in non-cancerous hepatic tissues following transcatheter arterial embolization

Abstract

Transcatheter arterial embolization (TAE) is a widely used technique in treating hepatic carcinoma but may cause liver injury in some cases. This study investigated the hepatoprotective effect of the preprocessed peroxisome proliferator-activated receptor-α (PPAR-α) agonist-WY-14643 following TAE. A total of 60 rabbit liver cancer models were developed and divided into a combined treatment (WY-14643 and TAE), TAE, and control groups. After TAE, we examined the histopathological picture and liver functions. Further, the expression of antioxidant enzymes, tumor necrosis factor-α (TNF-α), nuclear factor of κ-light chain of enhancer-activated B cells (NF-κB), PPAR-α, and B-cell lymphoma-2 (Bcl-2) was analyzed. Liver function tests, pathology score, and apoptosis index significantly worsened in the TAE group but were normalized in the combined treatment group. In addition, ELISA results showed that antioxidant enzyme activity significantly increased, while the malondialdehyde content and level of inflammatory cytokines were significantly reduced in the combined treatment group. Furthermore, compared to the TAE group, the expressions of PPAR-α, antioxidant enzymes superoxide dismutase1 (SOD1) and SOD2, and Bcl-2 were significantly elevated, while NF-κB was significantly reduced in the combined treatment group. On the other hand, the expression of NF-κB in tumor tissues was significantly reduced by pretreatment with WY-14643. Therefore, PPAR-α can ameliorate liver injury by exerting its anti-oxidative, anti-inflammatory, and anti-apoptotic functions.

Abstract

In this study, a series of novel nanofibers based on gelatin (GA) loading with purple potato anthocyanin (PPA) and syringic acid (SA) were obtained by electrospinning technology. The effects of SA on mechanical properties, thermal stability, antioxidant capacity, and antimicrobial activity of the GA/PPA nanofibers were systematically characterized. The scanning electron microscopy observation results revealed a smooth surface on the nanofibers. The incorporation of SA enhanced the viscosity of the electrospun solutions, and it increased the average diameter of nanofibers from 0.17 μm to 0.28 μm. The tensile strength and thermal stability of the obtained nanofibers were enhanced with the addition of a suitable level of SA (1.5%, w/v), which strengthened the intermolecular interaction. The GA/PPA/SA nanofibers presented over 80% antioxidant capacity and strong antibacterial activity against E. coli and S. aureus. Meanwhile, the sensitivity responses of nanofibers to NH3 revealed that GA/PPA/SA II nanofibers (1.5% w/v SA) presented good sensitivity of colorimetric behavior to ammonia. A pork spoilage test was performed to evaluate practical application of the nanofibers, and an obvious color change (dark purple to green) was observed. These results indicated GA/PPA/SA II nanofibers can be utilized as an active and intelligent multipurpose packaging material to preserve and track the freshness of pork. [ABSTRACT FROM AUTHOR]
