Het Centrum voor Genetische Bronnen (CGN), onderdeel van Wageningen UR, werd 25 jaar geleden opgericht om zich in te zetten voor het behoud van genetische diversiteit in voedselgewassen. Vanaf 2002 werd de Stichting Genenbank Landbouwhuisdieren onderdeel van het CGN. Wat betekent die genenbank voor de geitensector in Nederland?

Improved understanding of the biology of traits of livestock species necessitates the use and combination of information that is stored in a variety of different sources such as databases and literature. The ability to effectively combine information from different sources, however, depends on a high level of standardization within and between various resources, at least with respect to the used terminology. Ontologies represent a set of concepts that facilitate standardization of terminology within specific domains of interest. The biological mechanisms underlying quantitative traits of farm animal species related to reproduction and host pathogen interactions are complex and not well understood. This knowledge could be improved through the availability of domain specific ontologies that provide enhanced possibilities for data annotation, data retrieval, data integration, data exchange, data analysis, and ontology-based searches. Here we describe a framework for domain specific ontologies and the development of two first-generation ontologies: the Reproductive Trait and Phenotype Ontology (REPO) and the Host Pathogen Interactions Ontology (HPIO). In these first-generation ontologies, we focused on 'female fertility in cattle' and 'interactions between pigs and Salmonella'. Through this, we contribute to the global initiative towards the development of an Animal Trait Ontology (ATO) for livestock species. To demonstrate its usefulness, we show how REPO can be used to select candidate genes for fertility. The Centre for Genetic Resources, the Netherlands (CGN) has the aim to store genetic materials of farm animal genetic resources in the gene bank, including semen of native Dutch duck breeds. CGN will store these valuable genetic resources in the gene bank to be able to recreate the breeds or to support in situ conservation of the breed in case of calamities. For every species research has to be done in order to find the optimal protocol for semen collection and cryo conservation. In this report of the cryopreservation activities on duch breeds in 2011 and 2012 are reported. In this report is described how breeds and animals were chosen, how the protocol for semen collection is developed, which research is done for collecting and freezing the semen and how many straws were cryopreserved in 2011 and 2012. The Dutch 'krombek' (Hook Bill duck) and the 'witborst' (North Holland White Bibbed duck) are endangered breeds and considered as our living heritage. Priority was given to these two breeds. Finally more than 900 straws from 39 animals were stored in the gene bank. CGN heeft in haar programma voor de periode 2010-2014 opgenomen om genebankcollecties aan te leggen van sperma van bedreigde oorspronkelijk Nederlandse eenden-, ganzen-, honden-, konijnen- en duivenrassen. CGN wil deze waardevolle genetische bronnen opslaan in de genenbank om in elk geval van een calamitieit een ras terug te kunnen fokken of het in situ behoud van het ras te ondersteunen. In dit rapport worden de resultaten van de activiteiten in 2011 en 2012 gepresenteerd voor de winning van sperma van woerden (eenden). Beschreven worden de ras- en dierkeuze, de spermawinning, de spermaverwerking en het uiteindelijke resultaat van deze activiteiten, te weten het aantal sermarietjes in de genenbank. Er is prioriteit gegeven aan de winning van sperma van de Noord-Hollandse krombek- en witborsteenden. Er zijn uiteindelijk ruim 900 dosis sperma van 39 woerden in de genenbank opgeslagen.

Zo’n 45 miljoen eendagskuikens worden jaarlijks gedood in Nederland, omdat ze ongeschikt zijn voor de eierproductie of voor kippenvlees. Al tientallen jaren is er maatschappelijk verzet tegen. Wageningen UR zoekt sinds 2005 naar alternatieven. Een andere mogelijkheid dan het doden lijkt dichterbij te komen.

Many local breeds are currently at risk because of replacement by a limited number of specialized commercial breeds. Concurrently, for many breeds, allelic diversity within breeds declines because of inbreeding. Gene banking of germplasm may serve to secure the breeds and the alleles for any future use, for instance to recover a lost breed, to address new breeding goals, to support breeding schemes in small populations to minimize inbreeding, and for conservation genetics and genomics research. Developments in cryobiology and reproductive technology have generated several possibilities for preserving germplasm in farm animals. Furthermore, in some mammalian and bird species, gene banking of material is difficult or impossible, requiring development of new alternative methods or improvement of existing methods. Depending on the species, there are interesting possibilities or research developments in the use of epididymal spermatozoa, oocytes and embryos, ovarian and testicular tissue, primordial germ cells, and somatic cells for the conservation of genetic diversity in farm- and other animal species. Rapid developments in genomics research also provide new opportunities to optimize conservation and sampling strategies and to characterize genome-wide genetic variation. With regard to gene banks for farm animals, collaboration between European countries is being developed through a number of organizations, aimed at sharing knowledge and expertise between national programmes. It would be useful to explore further collaboration between countries, within the framework of a European gene banking strategy that should minimize costs of conservation and maximize opportunities for exploitation and sustainable use of genetic diversity.

Het doden van eendagshaantjes heeft de afgelopen jaren meer en meer maatschappelijke en politieke weerstand opgeroepen. Wageningen UR onderzoekt enkele alternatieven.

Miljoenen eendagskuikens worden jaarlijks gedood in de pluimveehouderij, omdat ze ongeschikt zijn om eieren te leggen of vleeskip te worden. Het maatschappelijk verzet daartegen klinkt steeds luider. Maar welke alternatieven zijn er? Wageningen UR Livestock Research zette ze op een rij en legde ze voor aan gewone Nederlanders.

In this project the technical possibilities were investigated to use protein profiles in dairy milk as an indicator to improve fertility. Doel van dit project is om de technische mogelijkheden in kaart te brengen om eiwitprofielen in de melk te gebruiken als indicator voor het verbeteren van vruchtbaarheid van melkvee.

Het systematisch en massaal doden van pas uitgekomen mannelijke kuikens, hoe ethisch is dat? En zijn er geen alternatieven? Deze twee vragen staan in deze bijdrage centraal. Op basis van de dierethische waarden ‘intrinsieke waarde van het dier’, dierenwelzijn en vrijheid om natuurlijk gedrag te vertonen, wordt geconcludeerd dat de gangbare praktijk ethisch zeer problematisch is, ook al worden de gedode haantjes in Nederland voor diervoeder gebruikt. Deze praktijk is vergeleken met seksen en selecteren van het 18-dagen oude embryo. Het verschil wordt ethisch niet groot geacht. Deze praktijk is gevolg van een eenzijdig op doelmatigheid gerichte ontwikkeling waarvoor de overheid en de samenleving als geheel medeverantwoordelijkheid voor dragen. Een echte oplossing vereist een systeemomslag of ten minste de ontwikkeling van geheel nieuwe technieken die het massale doden van eendagshaantjes of bebroede eieren vermijden

De commerciële pluimveehouderij is sterk gespecialiseerd. Er zijn rassen voor het leggen van eieren en voor de productie van vlees. Omdat haantjes geen eieren leggen en haantjes van legrassen niet rendabel zijn voor de vleesproductie, worden ze als eendagskuiken gedood. Dit gebeurt overal ter wereld en zowel bij ‘gewoon’ als bij biologisch pluimvee. In Nederland werden in 2009 zo’n 45 miljoen haantjes als eendagskuiken gedood. Het massaal doden van eendagshaantjes roept maatschappelijke weerstand op.

Het seksen van stierensperma beleeft in de melkveehouderij een snelle opkomst. Het is echter de vraag of in de biologische sector deze techniek wel toelaatbaar is. Ten behoeve van het creëren van duidelijkheid omtrent het gebruik van deze techniek wordt in deze notitie eerst de techniek van het seksen van sperma beschreven zoals die nu wordt uitgevoerd. Daarna wordt kort uiteengezet wat de voor- en nadelen van deze techniek kunnen zijn en wat op basis hiervan de voor en tegens van het wel of niet gebruiken van het seksen van sperma in de biologische landbouw zijn.

Na de eerste drie „inhaalslagen‟ in 2003, 2005 en 2007 is weer een project opgezet om te komen tot het verzamelen van genetisch materiaal van een aantal hanen van zeldzame kippenrassen (inhaalslag 4). Het doel van het project is om (de genetische variatie van) deze (kleine) rassen veilig te stellen in de genenbank. Met de financiële middelen van 2009 kon voor de vierde maal worden gestart met een aantal hanen van rassen, die in 2003, 2005 en 2007 niet voldoende aan bod waren gekomen. In een evaluatie met de Stichting Zeldzame Huisdierrassen en rekening houdend met de reeds aanwezige genenbank collecties zijn zestien rassen geselecteerd waarvan genetisch materiaal is gecryo-conserveerd, te weten het Hollandse Kriel (11), Eikenburger Kriel (5), Sabelpoot Krielen (10), Fries Hoen (2), Hollands Kuifhoen (6), Assendelfter Hoen (6), Drents Hoen (3), Groninger Meeuw (5), Noordhollands Hoen (6), Twents Hoen (3), Chaams Hoen (3), Welsumer (2), Lakenvelder (2), Nederlandse Uilenbaard (2), Barnevelder (4) en het Hollands Hoen (3). Van deze 16 rassen zijn totaal 73 hanen aangevoerd. Vier hanen zijn in de eerste dagen reeds gesneuveld. Van één Eikenburger Kriel haan kon geen sperma worden verkregen. Met de aanvulling van het sperma van de hanen van voornoemde rassen zou de doelstelling kunnen worden bereikt, om van elk ras ca. 800 doses sperma in opslag te hebben van minimaal 10 hanen. In totaal zijn in deze vierde inhaalslag 5534 doses sperma ingevroren en in de genenbank weggezet. De Eikenburger Kriel haalde deze aantallen niet vanwege de te geringe spermaproductie (ruim 200 doses!). Van het Drents Hoen, de Assendelfter Hoen en de Schijndelaar benaderen de aantallen doses sperma de 800.

Een overzicht van de ontwikkelingen van het kloneren van landbouwhuisdieren, met het oog op mogelijke consequenties voor beleid en regelgeving. Er wordt ingegaan op de techniek van dna modificatie, de invloed op veehouderij, fokkerij, praktijk, dierenwelzijn en -gezondheid, nationale veiligheid en regelgeving en de toekomstige ontwikkelingen in de nabije toekomst

Met focusgroepen en een (internet)enquête zijn de opvattingen van 'het publiek' over mogelijke alternatieven voor het doden van eendagshaantjes onderzocht. Er bleek geen eenduidige voorkeur voor een specifiek alternatief; nader onderzoek naar de haalbaarheid van enkele alternatieven is wenselijk

Het rapport beschrijft de perspectieven van verschillende technologische alternatieven voor het doden van eendagskuikens. Daarbij wordt een ethisch kader aangereikt dat kan worden gebruikt om te bepalen welke technologische benadering het meest zinvol en moreel acceptabel is. De resultaten van dit rapport zijn ingebracht in de discussie tussen de minister van LNV en de tweede Kamer over doding van eendagskuikens

In response to the letter by Dr. Amir Arav I would like to mention the following. The application of the principle of directional solidification to cryopreservation of living cells was, as far as I am aware, not the invention of Dr. Arav. It was first published and patented by Rubinsky (Berkeley, CA, US) in 1985. Furthermore, in the early nineties there were at least two papers by the group of the Helmholz Institute at Aachen, Germany and MIT, Massachussets, US showing further application and development of that concept. There were papers from other groups as well in that time. In my paper for the 2003 Proceedings of the Boar Semen Preservation (BSP) conference I have referred to the papers by Rubinsky and Ikeda (1985) (Cryobiology) and Beckmann et al. (1990) (also Cryobiology). In the mid-90s, we came up with a design of a conveyor belt like freezing device that encompassed the principle of directional solidification. Originally we actually designed the conveyor belt system for logistical reasons and only later found out that it could also have advantages for cell survival. The novel elements that we have introduced in our device warranted that patents have been granted in the US and the EU. I was aware of the fact that Arav had a patent application also on an apparatus that encompasses the principle of directional solidification. As said above, in my 2003 BSP conference presentation I referred to the original papers on directional solidification by Rubinsky and Ikeda (1985) and Beckmann et al. (1990). I thought to have served well in this respect. Obviously, in my 2003 BSP conference address I could not refer to papers by Arav that were published in 2004 and 2005. But I could have referred to his two papers published in 2002. In these papers Arav and co-workers failed to show a split sample comparison of the post-thaw survival obtained with directional solidification versus conventional straw freezing. However, one of these 2002 papers does mention the application of the technique to boar semen, and in retrospect I regret that I did not refer to that paper in my Boar Semen Preservation conference contribution. It is therefore good that Dr. Arav now brought these papers to our attention

This chapter focuses on ex situ conservation. An overview of the state of the art cryopreservation and reproductive technology for farm animals and fish is followed by a discussion on the implications of ex situ conservation strategies. Ex situ conservation of genetic material from livestock and fish through cryopreservation is an important strategy to conserve genetic diversity in these species. Conservation strategies benefit from advances in cryopreservation and reproductive technologies. Choice of genetic material type to be preserved for different species depends highly on objectives, technical feasibility (e.g. collection, cryopreservation), costs and practical circumstances

The aim of this study was to develop a cryopreservation protocol for type A spermatogonia. Testes from 5- to 7-month-old calves were collected, and type A spermatogonia were isolated using two-step enzymatic digestion and Percoll separation. Cells were resuspended in minimum essential medium (MEM) supplemented with 1% bovine serum albumin (BSA) in a final concentration of 6 x 10(6) per mL, and the effects of different cryoprotectants and freezing protocols were tested.
