59. Effects of sward characteristics and concentrate supplementation on herbage intake and performance of lactating dairy cows at pasture

Abstract

Sward characteristics have a major effect on intake and performance of dairy cows at pasture. Milk production from grazing cows however can be restricted by herbage intake, and concentrate supplementation can allow them to perform closer to their production potential. This study investigates effects of sward characteristics and concentrate supplementation on intake and performance of grazing dairy cows. Grazing cows yielding on average 36.8 kg milk day (d)-1 were offered high levels of concentrates in late summer. Milk yield response was 1.01 kg milk kg-1 concentrate dry matter (DM) d-1 when concentrate was increased from 5.2 to 7.7 kg DM d-1. Milk yield response declined to 0.83 kg milk kg-1 DM when concentrate was increased from 7.7 to 10.2 kg DM d-1. Grazing time and herbage intake were reduced at higher levels of supplementation. Increasing concentrate level by 1.7 kg DM d-1 when cows were housed overnight had no effect on animal performance. In another experiment, there was no significant difference in animal performance between cows offered either a high starch or high fibre concentrate at a rate of 5.3 kg concentrate DM d-1. Supplementation with an additive formulated to reduce dietary protein degradability however had a positive effect on milk yield, which was on average 34.4 and 32.9 kg d-1 for additive and control treatments respectively. Inclusion of the additive also increased milk protein yield and herbage intake. Interactions between sward characteristics and intake were examined. Bite mass was predicted from estimates of bite dimensions and measurements of vertical distribution of herbage mass in cut swards. A general relationship observed between sward height and vertical distribution of mass could be used to predict bite mass from sward height and total herbage mass. Methods to make detailed measurements of intake and grazing activity within patches of a sward using grazing cows were developed and demonstrated an effect of time of day on bite mass. Research to quantify interactions between sward structure, supplementation and grazing activity, focusing on bite mass, should enable development of strategies to exploit the potential of grazed grass and provide appropriate supplementation, which will ultimately contribute to improved profitability of milk production.

Abstract

Shocklets and short large-amplitude magnetic structures (SLAMS) are steepened magnetic fluctuations commonly found in Earth's upstream foreshock. Here we present Venus Express observations from the 26th of February 2009 establishing their existence in the steady-state foreshock of Venus, building on a past study which found SLAMS during a substantial disturbance of the induced magnetosphere. The Venusian structures were comparable to those reported near Earth. The 2 Shocklets had magnetic compression ratios of 1.23 and 1.34 with linear polarization in the spacecraft frame. The 3 SLAMS had ratios between 3.22 and 4.03, two of which with elliptical polarization in the spacecraft frame. Statistical analysis suggests SLAMS coincide with unusually high solar wind Alfvén mach-number at Venus (12.5, this event). Thus, while we establish Shocklets and SLAMS can form in the stable Venusian foreshock, they may be rarer than at Earth. We estimate a lower limit of their occurrence rate of ≳14%., QC 20231020

Abstract

Interplanetary shocks are large-scale heliospheric structures often caused by eruptive phenomena at the Sun, and represent one of the main sources of energetic particles. Several interplanetary shock crossings by spacecraft at $1$ AU have revealed enhanced energetic-ion fluxes that extend far upstream of the shock. Surprisingly, in some shock events, ion fluxes with energies between $100$ keV and about $2$ MeV acquire similar values (which we refer to as ``overlapped'' fluxes), corresponding to flat energy spectra in that range. In contrast, closer to the shock, the fluxes are observed to depend on energy. In this work, we analyze three interplanetary shock-related energetic particle events observed by the Advanced Composition Explorer spacecraft where flat ion energy spectra were observed upstream of the shock. We interpret these observations via a velocity filter mechanism for particles in a given energy range. This reveals that low energy particles tend to be confined to the shock front and cannot easily propagate upstream, while high energy particles can. The velocity filter mechanism has been corroborated from observations of particle flux anisotropy by the Solid-State Telescope of Wind/3DP.

We present recent high time resolution observations from an oblique (43 deg) shock crossing from the Magnetospheric Multiscale mission. Short-duration bursts between 10 and 100 ms of ion acoustic waves are observed in this event alongside a persistent reflected ion population. High time resolution (150 ms) particle measurements show strongly varying ion distributions between successive measurements, implying that they are bursty and impulsive by nature. Such signatures are consistent with ion bursts that are impulsively reflected at various points within the shock. We find that, after instability analysis using a Fried-Conte dispersion solver, the insertion of dispersive ion bursts into an already stable ion distribution can lead to wave growth in the ion acoustic mode for short durations of time. We find that impulsively reflected ions are a plausible mechanism for ion acoustic wave growth in the terrestrial bow shock and, furthermore, suggest that wave growth can lead to a small but measurable momentum exchange between the solar wind ions and the reflected population.

83. Expression of functional human growth factors in insect cells

Peptide growth factors are important in a wide variety of physiological and pathological processes as a consequence of their ability to generate a mitogenic signal via their interaction with and activation of a specific membrane-associated receptor. The subversion of the normal growth factor signalling pathway, albeit by the enhanced expression of the growth factor or receptor, the unregulated activity of the receptor protein-tyrosine kinase or the uncoupling of intracellular pathways can lead to uncontrolled cellular proliferation resulting in cellular transformation and the formation of tumours in vivo. The major part of this thesis was devoted to the development of baculoviral expression systems producing large amounts of biologically active EGF and TGFa for the assessment of the structural and functional basis of ligand-receptor interactions. Insect cells infected with the recombinant baculoviruses expressed growth factors with the correct putative molecular weights which were immunologically indistinguishable from their authentic counterparts. The recombinant growth factors were able to bind to the EGF receptor expressed in insect cells and NR6+ fibroblasts and activate the receptor protein-tyrosine kinase activity, Furthermore, they induced a mitogenic response in NR6+ and Swiss 3T3 fibroblasts. It is anticipated that milligram quantities of these biologically important growth factors will be available for crystallisation. The elucidation of the three-dimensional structures of the growth factors by diffractional analysis of the crystals and structural mutational analysis will establish the nature of the receptor binding face. This information will lead to the design of clinically important EGF and TGFa antagonists. The recombinant ligands will allow the continued biophysical characterisation of the conformation changes occurring in the extracellular domain of the EGF receptor in response to ligand binding. Cocrystallisation will ultimately lead to the determination of the ligand binding site of the receptor. The biosynthetic precursors for EGF and TGFa are integral membrane glycoproteins suggesting that they may function to mediate physiological cell-cell recognition events. The TGFa precursor expressed in insect cells was able to stimulate the EGF receptor protein-tyrosine kinase activity. Characterisation of the precursor in insect cells will determine the relevance of this mechanism for the production of secreted growth factors and investigate further potential roles for their membrane- associated precursors. The expression of c-erbB-2 in insect cells has allowed the evaluation of a putative growth factor receptor which is highly homologous to the EGF receptor. Furthermore, an in vivo association of the EGF receptor and p185c-erbB-2 been demonstrated. This system will provide a valuable tool for the determination of the mechanism responsible for the transmembrane translocation of the mitogenic signal. It is anticipated that baculoviral coexpression of growth factor receptors and their cognate ligands, in conjunction with intracellular substrates will play an important role in the identification of their intracellular signalling pathways. The elucidation of the growth factor signalling pathway is crucial to the understanding of how oncogenic activation of anyone of the components can result in cellular transformation.

Abstract

High-resolution particle and wave measurements taken during an oblique bow shock crossing by the Magnetospheric Multiscale (MMS) mission are analyzed. Two regions of differing magnetic behavior are identified within the shock, one with active magnetic fluctuations and one with laminar interplanetary magnetic field topology. A prominent reflected ion population is observed in both regions. The active magnetic region is characterized by large-amplitude (>100 mV/m) electrostatic solitary waves, electron Bernstein waves, and ion acoustic waves, along with intermittent current activity and localized electron heating. In the region of laminar magnetic field, ion acoustic waves are prominently observed. Solar wind ion deceleration is observed in both regions of active and laminar magnetic field. All observations suggest that solar wind deceleration can occur as a result of multiple independent processes, in this case current and ion-ion instabilities.

Abstract

Lion roars are narrowband whistler wave emissions that have been observed in several environments, such as planetary magnetosheaths, the Earth's magnetosphere, the solar wind, downstream of interplanetary shocks, and the cusp region. We present measurements of more than 30,000 such emissions observed by the Magnetospheric Multiscale spacecraft with high‐cadence (8,192 samples/s) search coil magnetometer data. A semiautomatic algorithm was used to identify the emissions, and an adaptive interval algorithm in conjunction with minimum variance analysis was used to determine their wave vector. The properties of the waves are determined in both the spacecraft and plasma rest frame. The mean wave normal angle, with respect to the background magnetic field (B(sub 0)), plasma bulk flow velocity (V(sub b)), and the coplanarity plane (V(sub b) × B(sub 0)) are 23°, 56°, and 0°, respectively. The average peak frequencies were ∼31% of the electron gyrofrequency (ω(sub ce)) observed in the spacecraft frame and ∼18% of ω(sub ce) in the plasma rest frame. In the spacecraft frame, ∼99% of the emissions had a frequency <ω(sub ce), while 98% had a peak frequency <0.72 ω(sub ce) in the plasma rest frame. None of the waves had frequencies lower than the lower hybrid frequency, ω. From the probability density function of the electron plasma β(sub e), the ratio between the electron thermal and magnetic pressure, ∼99.6% of the waves were observed with β(sub e)<4 with a large narrow peak at 0.07 and two smaller, but wider, peaks at 1.26 and 2.28, while the average value was ∼1.25.

Abstract

We present a long-duration (approx. 10 yr) statistical analysis of the temperatures, plasma betas, and temperature ratios for the electron, proton, and alpha-particle populations observed by the Wind spacecraft near 1 au. The mean(median) scalar temperatures are T(sub e)(sub tot) = 12.2(11.9) eV, T(sub p)(sub tot) = 12.7(8.6) eV, and T(sub a)(sub tot) = 23.9(10.8) eV. The mean(median) total plasma betas are Beta(sub e)(sub tot) = 2.31(1.09), Beta(sub p)(sub tot) = 1.79(1.05), and Beta(sub a)(sub tot) = 0.17(0.05). The mean(median) temperature ratios are (T(sub e)/T(sub p))(sub tot) = 1.64(1.27), (T(sub e)/T(sub a))(sub tot) = 1.24(0.82), and (T(sub a)/T(sub p))(sub tot) = 2.50(1.94). We also examined these parameters during time intervals that exclude interplanetary (IP) shocks, times within the magnetic obstacles (MOs) of interplanetary coronal mass ejections (ICMEs), and times that exclude MOs. The only times that show significant alterations to any of the parameters examined are those during MOs. In fact, the only parameter that does not show a significant change during MOs is the electron temperature. Although each parameter shows a broad range of values, the vast majority are near the median. We also compute particle–particle collision rates and compare to effective wave–particle collision rates. We find that, for reasonable assumptions of wave amplitude and occurrence rates, the effect of wave–particle interactions on the plasma is equal to or greater than the effect of Coulomb collisions. Thus, wave–particle interactions should not be neglected when modeling the solar wind.

98. Kinetic Properties of an Interplanetary Shock Propagating Inside a Coronal Mass Ejection

We investigate the kinetic properties of a typical fast-mode shock inside an interplanetary coronal mass ejection (ICME) observed on 1998 August 6 at 1 au, including particle distributions and wave analysis with the in situ measurements from Wind. Key results are obtained concerning the shock and the shock-ICME interaction at kinetic scales: (1) gyrating ions, which may provide energy dissipation at the shock in addition to wave-particle interactions, are observed around the shock ramp; (2) despite the enhanced proton temperature anisotropy of the shocked plasma, the low plasma beta inside the ICME constrains the shocked plasma under the thresholds of the ion cyclotron and mirror-mode instabilities; (3) whistler heat flux instabilities, which can pitch-angle scatter halo electrons through a cyclotron resonance, are observed around the shock, and can explain the disappearance of bi-directional electrons (BDEs) inside the ICME together with normal betatron acceleration; (4) whistler waves near the shock are likely associated with the whistler heat flux instabilities excited at the shock ramp, which is consistent with the result that the waves may originate from the shock ramp; (5) the whistlers share a similar characteristic with the shocklet whistlers observed by Wilson et al., providing possible evidence that the shock is decaying because of the strong magnetic field inside the ICME.

99. Subcritical Growth of Electron Phase-Space Holes in Planetary Radiation Belts

The discovery of long-lived electrostatic coherent structures with large-amplitude electric fields (1 less than or equal to E less than or equal to 500 mV/m) by the Van Allen Probes has revealed alternative routes through which planetary radiation belts' acceleration can take place. Following previous reports showing that small phase-space holes, with q(phi)/T (exp c)(sub e) approximately minus 10 (exp -2) - 10 (exp -3), could result from electron interaction with large-amplitude whistlers, we demonstrate one possible mechanism through which holes can grow nonlinearly (i.e., Gamma alpha square root of phi) and subcritically as a result of momentum exchange between hot and cold electron populations. Our results provide an explanation for the common occurrence and fast growth of large-amplitude electron phase-space holes in the Earth's radiation belts.
