Abstract

RAF kinase inhibitor protein (RKIP) is a seminal regulator of intracellular signaling and exhibits both antimetastatic and antitumorigenic properties. Decreased expression of RKIP has been described in several human malignancies, including acute myelogenous leukemia (AML). As the mechanisms leading to RKIP loss in AML are still unclear, we aimed to analyze the potential involvement of miRNAs within this study. miRNA microarray and qPCR data of more than 400 AML patient specimens revealed correlation between decreased expression of RKIP and increased expression of miR-23a, a member of the miR-23a/27a/24-2 cluster. In functional experiments, overexpression of miR-23a decreased RKIP mRNA and protein expression, whereas miR-23a inhibition caused the opposite effect. By using an RKIP 3′-untranslated region luciferase reporter construct with and without mutation or deletion of the putative miR-23a-binding site, we could show that RKIP modulation by miR-23a is mediated via direct binding to this region. Importantly, miR-23a overexpression induced a significant increase of proliferation in hematopoietic cells. Simultaneous transfection of an RKIP expression construct lacking the miR- 23a-binding sites reversed this phenotype, indicating that this effect is truly mediated via downregulation of RKIP. Finally, by analyzing more than 4,300 primary patient specimens via database retrieval from The Cancer Genome Atlas, we could highlight the importance of the miR-23a/RKIP axis in a broad range of human cancer entities. Abstract

Two opposing microtribometry approaches have been developed over the past decade to help connect the dots between fundamental and practical tribology measurements: spring-based (e.g., AFM) approaches use low speed, low stiffness, and long relative slip length to quantify friction, while quartz crystal microbalance (QCM)-based approaches use high speed, high stiffness, and short relative slip length. Because the friction forces generated in these experiments are attributed to entirely different phenomena, it is unclear if or how the resulting friction forces are related. This study aims to resolve this uncertainty by integrating these distinct techniques into a single apparatus that allows two independent measurements of friction at a single interface. Alumina microspheres were tested against single-crystal MoS2, a model nominally wear-free solid lubricant, and gold, a model metal control, at loads between 0.01 and 1 mN. The combined results from both measurement approaches gave friction coefficients (mean ± standard error) of 0.087 ± 0.007 and 0.27 ± 0.02 for alumina-MoS2and alumina-gold, respectively. The observed agreement between these methods for two different material systems suggests that friction in microscale contacts can be far less sensitive to external effects from compliance and slip speed than currently thought. Perhaps more importantly, this Article describes and validates a novel approach to closing the “tribology gap” while demonstrating how integration creates new opportunities for fundamental studies of practical friction.

Abstract

Spin chains are the most elementary entities in the study of magnetic ordering behavior. The authors describe a route toward the realization of artificial XY spin chains. Using thin-film deposition and e-beam lithog., 300 and 550 nm diam. pillars consisting of alternating disks of permalloy and Al2O3 were made. Shape anisotropy forces the magnetization in plane, creating a chain of two-dimensional magnetic moments →i. The disks couple to each other by their dipolar stray fields, resulting in an interaction Jij→i·→j, with Jij the coupling between disks i and j. This form closely resembles a chain of xy spins, coupled by effective interactions Jij. The authors present a magnetic characterization of the pillars, and compare the results with model calcns. Abstract

The status of the search for NN-decoupled resonances in the πNN-system is reviewed. After a short survey on the history of dibaryon searches reactions suitable for detecting such resonances are discussed. Special emphasis is put on the pionic double charge exchange, where recently evidence for the signature of a narrow πNN resonance, called d′, with I(J P) = even (0 -) and m = 2.06 GeV has been quoted. Further reactions discussed in this context are the pion photoproduction on the deuteron and the two-pion production in nucleon-nucleon collisions., Source: Scopus
