Abstract

Direct β-arylation of thiophene derivatives with bromide as directing group is disclosed. The reaction is conducted with PdCl2 /(p -tolyl)3 P as catalyst, silver carbonate as additive, and aryl iodide as coupling partner, affording brominated biaryl compounds as product. Control experiments indicated that the presence of bromide group enhances the reactivity of the C–H bond, enabling β-arylation. Furthermore, the C–Br bond can be easily converted into many useful functional groups through a wide range of methodologies. 77. Overexpression of TEM8 promotes ovarian cancer progression via Rac1/Cdc42/JNK and MEK/ERK/STAT3 signaling pathways

Abstract

Tumor endothelial cell marker 8 (TEM8) is a type I transmembrane protein, that has been widely studied in the areas of anthrax toxin infection and tumor angiogenesis. However, the role of TEM8 in the progression of epithelial ovarian cancer (EOC) remains unclear. In this study, we determined that TEM8 was highly expressed in ovarian cancer and associated with poor prognosis in EOC patients. In vitro experiments showed that TEM8 overexpression significantly promoted ovarian cancer proliferation. TEM8 overexpression also promoted the G0/G1 phase transition, migration, and invasion of ovarian cancer cells but suppressed apoptosis. Moreover, experimental verification confirmed that TEM8 overexpression increased the expression of Ki-67, cyclin D1, Bcl2/Bax, MMP2, MMP9, and VEGFA and the phosphorylation of Rac1/Cdc42, JNK, MEK, ERK, and STAT3 (Ser727). Subsequently, the addition of RAC1 (EHop-016) and MEK (PD98059) pathway inhibitors suppressed malignant behaviors in the TEM8 overexpression group, which robustly indicated that TEM8 activated Rac1/Cdc42/JNK and MEK/ERK/STAT3 signaling pathways. In addition, we also revealed that the transcription factor GATA2 bound to the TATTAGTTATCTTT site of the TEM8 promoter region and regulated its expression. In conclusion, our study may provide a new theoretical basis for TEM8 application as a clinical biomarker and potential target in EOC patients.

87. Involvement of the autophagy-related gene BdATG8in development and pathogenicity in Botryosphaeria dothidea

Abstract

Botryosphaeria dothideais a destructive fungal pathogen that causes Botryosphaeria canker and fruit ring rot on apple worldwide. Autophagy is a process of self-degradation that maintains intracellular homeostasis vialysosomal pathway. To date, the biological role of autophagy in B. dothidearemains unknown. In this study, we identified and characterized the autophagy-related gene BdATG8in B. dothidea. BdATG8was able to restore the defect in nitrogen starvation tolerance of Saccharomyces cerevisiae ATG8deletion mutant. GFP-BdAtg8 was shown to be a useful marker for monitoring autophagy in B. dothidea. Target deletion of BdATG8(ΔBdAtg8) blocked autophagy and significantly impaired mycelial growth, conidiation and perithecium formation. In addition, ΔBdAtg8showed significantly increased sensitivity to phytoalexin and oxidative stress, suggesting that BdATG8plays critical roles in overcoming phytoalexin and reactive oxygen species (ROS)-mediated plant immunity. Pathogenicity assays revealed that ΔBdAtg8almost lost ability to infect hosts. Overall, our results indicate that BdATG8plays an important role in fungal development, stress responses and pathogenesis in B. dothidea.

Abstract

To find new lead compounds with high antifungal activity, a series of new thiourea derivatives containing 1,3,4-thiadiazole and thioether skeleton was designed via linking the active sub-structures. The target compounds were prepared via three steps from the commercially available thiosemicarbazide. Their structures were characterized by means of HRMS, 1H NMR, 13C NMR and IR spectroscopy. The preliminary results indicate that the title compounds show various antifungal activity against the tested fungi. Compounds 4c, 4g, 4h, 4k, 4n, 4o, 4p, 4q and 4r display excellent antifungal activities against one or more tested fungi with inhibitory efficiencies of 90%―100% at 200 μg/mL. Especially, compound 4o shows the best inhibitory effect against Curvularia lunata, Cotton Fusarium Wilt, P. P. var nicotianae and Fusarium spp. with the EC50 values of 28.12, 30.41, 15.2 and 6.22 μg/mL, respectively, which are even superior to those of triadimefon(98.73, 96.58, 105.37 and 102.18 μg/mL). The preliminary structure-activity relationship indicates that allyl and aromatic groups are favorable to their antifungal activities.

96. SOFT SEDIMENT DEFORMATION AND GENESIS OF THE EARLY-MIDDLE JURASSIC YANAN FORMATION, ORDOS BASIN.

Abstract

The paleoearthquake-related soft sediment deformation structure plays a role in revealing the tectonic activity of basin and periphery in basin evolution. Based on the core description and field survey of Yanan Formation in Ordos Basin, the soft sediment liquefaction deformation layers are identified in DT3522 well in the southwest of Dingbian and the Yanhe section in Ansai, including the liquefaction-related pillow beds, liquefied sandstone vein, liquefied breccia, mud volcano and 9 deformation structures such as load structure and ball-and-pillow structure. Through study on the correlation between soft sediment deformation layers and deformation characteristics, combined with regional tectonic setting, it is believed that there are 3 stages of paleoseismic activities with the earthquake from weak to intense in the late sedimentary period of Y7 oil layer in Yanan Formation, Ordos Basin. 97. Biotinylated and fluorophore-incorporated polymeric mixed micelles for tumor cell-specific turn-on fluorescence imaging of Al3+.

Abstract

Excessive amounts of Al3+ in the human body can cause adverse effects on immune function and induce several neurodegenerative disorders. So far, most of the reported fluorescent probes for Al3+ present some common drawbacks, such as low sensitivity and poor water solubility. In addition, a number of traditional fluorescent probes failed to image Al3+ in tumor cells due to the lack of tumor cell targeting capacity and cell penetrating abilities. To overcome these shortcomings, we constructed tumor-targeting fluorescent mixed nano-micelles (mPEG-Dye–Biotin) with an average particle size of 21 nm from an amphiphilic polymer containing a Schiff-base fluorescent unit (mPEG-Dye) and another amphiphilic polymer containing a tumor cell recognition ligand (DSPE–PEG–Biotin), through the co-self-assembly of both amphiphilic polymers in water using the film rehydration method. The as-prepared nanoprobe showed a highly sensitive and selective turn-on fluorescence response to Al3+ in aqueous solution with a low detection limit. MTT assay revealed the negligible cytotoxicity of the mPEG-Dye–Biotin nanoprobe to both HeLa cells and COS-7 cells, indicating the safety of mPEG-Dye–Biotin for biological applications. More importantly, the biotinylated nanoprobe showed better ability to enter biotin receptor-positive HeLa cells than that of the non-biotinylated micelle mPEG-Dye, which made it more suitable for imaging Al3+ in biotin receptor-positive tumor cells. This work provides a simple and general strategy to design a highly sensitive and tumor cell-specific metal ion nanoprobe, which can not only be applied in Al3+ imaging, but can also be extended to other ions or biomolecules by changing the incorporated fluorescent unit in the amphiphilic polymer.
