Davidia involucrata Baill. is a kind of tertiary paleotropical plant floristic relic species unique to China. This rare plant is disappearing due to poor adaptability and serious poaching. We first assembled the complete chloroplast (cp) genome of Davidia involucrata Baill. by Illumina paired-end reads data. The whole genome was 169,085 bp, consisting of a pair of inverted repeats of 169,379 bp, large single copy region and a small single copy region (96,712 and 67,667 bp in length, respectively). The cp genome contained 90 genes, including 64 protein-coding genes, 22 trRNA genes and 4 rRNA genes. The overall GC content of the whole genome was 38.04%. A neighbor-joining phylogenetic analysis demonstrated a close relationship between Davidia involucrata Baill. and Nyssa yunnanensis. [ABSTRACT FROM AUTHOR]

As an essential grain, oil, and feed crop worldwide, soybean plays a crucial role. Developing high-yielding and high-quality soybean varieties is a critical goal for breeders. The grain weight per plant and 100-grain weight directly impact the soybean yield. This study combined genotypic data from the population with phenotypic data. Based on genome-wide association analysis (GWAS), GLM and MLM analysis models were used to locate the Gm04_21489088, Gm04_15703616, and Gm04_46466250 are loci related to soybean grain weight per plant, and find the Gm09_20334173, Gm04_39518612 and Gm04_39518624 are loci related to 100-seed weight. After performing a reference comparison, we conducted gene annotation and identified candidate genes Glyma.04G203400 and Glyma.04G125600 , potentially associated with grain weight per plant in soybeans. These genes are primarily involved in protein synthesis and cell differentiation processes. The candidate gene Glyma.09G109100 , associated with the 100-grain weight trait, was successfully annotated. The analysis revealed that the gene primarily involves enzyme activity, suggesting its potential role in regulating grain weight. These findings offer valuable insights into the mechanism of soybean yield and serve as a critical theoretical foundation and genetic resource for cultivating new soybean germplasm with high yield. To verify the function of chalcone reductase gene (CHR1) in soybean Daidzein synthesis, CHR1 gene in soybean was cloned, and an RNAi expression vector pCPB-CHR1-RNAi was constructed. Four transformed plants in T0 generation and thirteen transformed plants in T1 generation of soybean "Jinong28" were obtained by agrobacterium-mediated genetic transformation, in which transcriptions of CHR1 gene were depressed. Southern blotting showed the functional fragment of pCPB-CHR1-RNAi was integrated into the genome of recipient soybean in the form of a single copy. Detection of the transcription of CHR1 gene using quantitative real-time PCR (qRT-PCR) showed that the expression of CHR1 gene in transformed plants decreased 60%-99% compared to the recipient soybean, while the content of isoliquiritigenin, the precursors of daidzein, decreased 38.7%. These results indicate that RNA interference can suppress the transcription of CHR1 gene expression successfully.
