Abstract Developing stable perovskite nanocrystals (NCs) with enhancing luminescent properties holds great importance for future potential applications in optoelectronics. Here, we engaged perovskite NCs in Cu2+ ion-based metal–organic framework (MOF) Cu-BTC (BTC = 1,3,5-benzene tricarboxylate) by physical mixing of MOF with CsPbBr3 NCs in toluene solution. MOF-protected perovskite NCs achieved high photoluminescence quantum yield 96.51% than pristine state CsPbBr3 NCs (51.66%). Along with the improvement in optical properties, the long-term stability of CsPbBr3 NCs in the solution phase also increases considerably upon loading in Cu-BTC MOF. Moreover, the changes in the luminescent intensity of the samples have been observed for 3 months in the solution. After 1 month, pristine CsPbBr3 NCs lose their emission intensity 68% from the initial, while the MOF-protected CsPbBr3 NCs show only a 10% reduction from the initial. These results indicate that the effective passivation of Cu-BTC MOF inhibits the aggregation of NCs, protecting them from the defective atmosphere. The excellent photoluminescence findings provide a new pathway for future optoelectronic applications.

Abstract Primary challenges for current and future precision neutrino experiments using liquid argon time projection chambers (LArTPCs) include understanding detector effects and quantifying the associated systematic uncertainties. This paper presents a novel technique for assessing and propagating LArTPC detector-related systematic uncertainties. The technique makes modifications to simulation waveforms based on a parameterization of observed differences in ionization signals from the TPC between data and simulation, while remaining insensitive to the details of the detector model. The modifications are then used to quantify the systematic differences in low- and high-level reconstructed quantities. This approach could be applied to future LArTPC detectors, such as those used in SBN and DUNE.

Floating offshore wind turbines may experience large surge motions, which can cause blade–vortex interaction if they are similar to or faster than the local wind speed. Previous research hypothesized that this blade–vortex interaction phenomenon represented a turbulent wake state or even a vortex ring state, rendering the actuator disc momentum theory and the blade element momentum theory invalid. This hypothesis is challenged, and we show that the actuator disc momentum theory is valid and accurate in predicting the induction at the actuator in surge, even for large and fast motions. To accomplish this, we develop a dynamic inflow model that simulates the vorticity–velocity system and the effect of motion. The model's predictions are compared to other authors' results, a semi-free-wake vortex ring model, other dynamic inflow models, and CFD simulations of an actuator disc in surge. The results show that surge motion and rotor–wake interaction do not result in a turbulent wake or vortex ring state and that the application of actuator disc momentum theory and blade element momentum theory is valid and accurate when applied correctly in an inertial reference frame. In all cases, the results show excellent agreement with the higher-fidelity simulations. The proposed dynamic inflow model includes a modified Glauert correction for highly loaded streamtubes and is accurate and simple enough to be easily implemented in most blade element momentum models.

Invertebrate-derived eDNA (iDNA) is an emerging tool for taxonomic and spatial biodiversity monitoring. Here, the authors use metabarcoding of leech-derived iDNA to estimate vertebrate occupancy over an entire protected area, the Ailaoshan Nature Reserve, China.

The streptothricin natural product mixture (also known as nourseothricin) was discovered in the early 1940s, generating intense initial interest because of excellent gram-negative activity. Here, we establish the activity spectrum of nourseothricin and its main components, streptothricin F (S-F, 1 lysine) and streptothricin D (S-D, 3 lysines), purified to homogeneity, against highly drug-resistant, carbapenem-resistant Enterobacterales (CRE) and Acinetobacter baumannii. For CRE, the MIC50 and MIC90 for S-F and S-D were 2 and 4 μM, and 0.25 and 0.5 μM, respectively. S-F and nourseothricin showed rapid, bactericidal activity. S-F and S-D both showed approximately 40-fold greater selectivity for prokaryotic than eukaryotic ribosomes in in vitro translation assays. In vivo, delayed renal toxicity occurred at >10-fold higher doses of S-F compared with S-D. Substantial treatment effect of S-F in the murine thigh model was observed against the otherwise pandrug-resistant, NDM-1-expressing Klebsiella pneumoniae Nevada strain with minimal or no toxicity. Cryo-EM characterization of S-F bound to the A. baumannii 70S ribosome defines extensive hydrogen bonding of the S-F steptolidine moiety, as a guanine mimetic, to the 16S rRNA C1054 nucleobase (Escherichia coli numbering) in helix 34, and the carbamoylated gulosamine moiety of S-F with A1196, explaining the high-level resistance conferred by corresponding mutations at the residues identified in single rrn operon E. coli. Structural analysis suggests that S-F probes the A-decoding site, which potentially may account for its miscoding activity. Based on unique and promising activity, we suggest that the streptothricin scaffold deserves further preclinical exploration as a potential therapeutic for drug-resistant, gram-negative pathogens.

ABSTRACT Pathogen inactivation is a strategy to improve the safety of transfusion products. The only pathogen reduction technology for blood products currently approved in the US utilizes a psoralen compound, called amotosalen, in combination with UVA light to inactivate bacteria, viruses, and protozoa. Psoralens have structural similarity to bacterial multidrug efflux pump substrates. As these efflux pumps are often overexpressed in multidrug-resistant pathogens, we tested whether contemporary drug-resistant pathogens might show resistance to amotosalen and other psoralens based on multidrug efflux mechanisms through genetic, biophysical, and molecular modeling analysis. The main efflux systems in Enterobacterales, Acinetobacter baumannii, and Pseudomonas aeruginosa are tripartite resistance-nodulation-cell division (RND) systems, which span the inner and outer membranes of Gram-negative pathogens, and expel antibiotics from the bacterial cytoplasm into the extracellular space. We provide evidence that amotosalen is an efflux substrate for the E. coli AcrAB, Acinetobacter baumannii AdeABC, and P. aeruginosa MexXY RND efflux pumps. Furthermore, we show that the MICs for contemporary Gram-negative bacterial isolates for these species and others in vitro approached and exceeded the concentration of amotosalen used in the approved platelet and plasma inactivation procedures. These findings suggest that otherwise safe and effective inactivation methods should be further studied to identify possible gaps in their ability to inactivate contemporary, multidrug-resistant bacterial pathogens. IMPORTANCE Pathogen inactivation is a strategy to enhance the safety of transfused blood products. We identify the compound, amotosalen, widely used for pathogen inactivation, as a bacterial multidrug efflux substrate. Specifically, experiments suggest that amotosalen is pumped out of bacteria by major efflux pumps in E. coli, Acinetobacter baumannii, and Pseudomonas aeruginosa. Such efflux pumps are often overexpressed in multidrug-resistant pathogens. Importantly, the MICs for contemporary multidrug-resistant Enterobacterales, Acinetobacter baumannii, Pseudomonas aeruginosa, Burkholderia spp., and Stenotrophomonas maltophilia isolates approached or exceeded the amotosalen concentration used in approved platelet and plasma inactivation procedures, potentially as a result of efflux pump activity. Although there are important differences in methodology between our experiments and blood product pathogen inactivation, these findings suggest that otherwise safe and effective inactivation methods should be further studied to identify possible gaps in their ability to inactivate contemporary, multidrug-resistant bacterial pathogens.

Skull base metastasis from renal cell carcinoma are exceedingly rare. Though renal cell carcinoma often presents at an advanced stage and with metastasis, signs and symptoms of skull base involvement would rarely suggest renal cell carcinoma. We described skull base metastasis as the initial presentation of metastatic renal cell carcinoma in a middle aged male. Symptoms of skull base involvement that persist should prompt immediate investigation for rare causes such as renal cell carcinoma.

Sire et al. examine the effects of climate-induced forest dieback and salvage logging on insect diversity in silver fir Pyrenean forests using metabarcoding. Although no consistent variation in species richness was found, forest dieback cause massive changes in community composition particularly affecting rare species.

Abstract Decorating single atoms of transition metals on MXenes to enhance the electrocatalytic properties of the resulting composites is a useful strategy for developing efficient electrocatalysts, and the mechanisms behind this enhancement are under intense scrutiny. Herein, we anchored Co single atoms onto several commonly used MXene substrates (V2CTx, Nb2CTx and Ti3C2Tx) and systematically studied the electrocatalytic behavior and the mechanisms of oxygen and hydrogen evolution reactions (OER and HER, respectively) of the resulting composites. Co@V2CTx composite displays an OER overpotential of 242 mV and an HER overpotential of 35 mV at 10 mA cm−2 in 1.0 M KOH electrolyte, which is much lower than for Co@Nb2CTx and Co@Ti3C2Tx, making it comparable to the commercial noble metal Pt/C and RuO2/C electrocatalysts. The experimental and theoretical results point out that the enhanced bifunctional catalytic performance of Co@V2CTx benefits from the stronger hybridization between Co 3d and surface terminated O 2p orbitals which optimized the electronic structure of Co single atoms in the composite. This, in turn, results in lowering the OER and HER energy barriers and acceleration of the catalytic kinetics in case of the Co@V2CTx composite. The advantage of Co@V2CTx was further validated by its high overall water splitting performance (1.60 V to deliver 10 mA cm−2). Our study sheds light on the origins of the catalytic activity of single transition metals atoms on MXene substrates, and provides guidelines for designing efficient bifunctional MXene‐based electrocatalysts.

Background Immunotherapy has revolutionized clinical outcomes for patients suffering from lung cancer, yet relatively few patients sustain long-term durable responses. Recent studies have demonstrated that the tumor immune microenvironment fosters tumorous heterogeneity and mediates both disease progression and response to immune checkpoint inhibitors (ICI). As such, there is an unmet need to elucidate the spatially defined single-cell landscape of the lung cancer microenvironment to understand the mechanisms of disease progression and identify biomarkers of response to ICI.Methods Here, in this study, we applied imaging mass cytometry to characterize the tumor and immunological landscape of immunotherapy response in non-small cell lung cancer by describing activated cell states, cellular interactions and neighborhoods associated with improved efficacy. We functionally validated our findings using preclinical mouse models of cancer treated with anti-programmed cell death protein-1 (PD-1) immune checkpoint blockade.Results We resolved 114,524 single cells in 27 patients treated with ICI, enabling spatial resolution of immune lineages and activation states with distinct clinical outcomes. We demonstrated that CXCL13 expression is associated with ICI efficacy in patients, and that recombinant CXCL13 potentiates anti-PD-1 response in vivo in association with increased antigen experienced T cell subsets and reduced CCR2+ monocytes.Discussion Our results provide a high-resolution molecular resource and illustrate the importance of major immune lineages as well as their functional substates in understanding the role of the tumor immune microenvironment in response to ICIs.

Abstract On 15 January 2022, the Hunga Tonga‐Hunga Ha'apai (HT) eruption injected SO2 and water into the middle stratosphere. Shortly after the eruption, the water vapor anomaly moved northward toward and across the equator. This northward movement appears to be due to equatorial Rossby waves forced by the excessive infrared water vapor cooling. Following the early eruption stage, persistent mid‐stratospheric water vapor and aerosol layers were mostly confined to Southern Hemisphere tropics (Eq. to 30°S). However, during the spring of 2022, the westerly phase of the tropical quasi‐biennial oscillation (QBO) descended through the tropics. The HT water vapor and aerosol anomalies were observed to again move across the equator coincident with the shift in the Brewer‐Dobson circulation and the descent of the QBO shear zone.

ABSTRACT The Escherichia coli acriflavine resistance protein D (AcrD) is an efflux pump that belongs to the resistance-nodulation-cell division (RND) superfamily. Its primary function is to provide resistance to aminoglycoside-based drugs by actively extruding these noxious compounds out of E. coli cells. AcrD can also mediate resistance to a limited range of other amphiphilic agents, including bile acids, novobiocin, and fusidic acids. As there is no structural information available for any aminoglycoside-specific RND pump, here we describe cryo-electron microscopy (cryo-EM) structures of AcrD in the absence and presence of bound gentamicin. These structures provide new information about the RND superfamily of efflux pumps, specifically, that three negatively charged residues central to the aminoglycoside-binding site are located within the ceiling of the central cavity of the AcrD trimer. Thus, it is likely that AcrD is capable of picking up aminoglycosides via this central cavity. Through the combination of cryo-EM structural determination, mutagenesis analysis, and molecular simulation, we show that charged residues are critically important for this pump to shuttle drugs directly from the central cavity to the funnel of the AcrD trimer for extrusion. IMPORTANCE Here, we report cryo-EM structures of the AcrD aminoglycoside efflux pump in the absence and presence of bound gentamicin, posing the possibility that this pump is capable of capturing aminoglycosides from the central cavity of the AcrD trimer. The results indicate that AcrD utilizes charged residues to bind and export drugs, mediating resistance to these antibiotics.

ObjectiveThe purpose of the study was to determine trends in the prevalence of individual health risk factors across time and to examine if their associations with mortality have changed over time.MethodsData from the National Health and Nutrition Examination Surveys (NHANES III- 1988-1994 and NHANES 1999-2014; age ≥20 years) was used to examine differences in the odds ratio (OR) of 5-year mortality risk associated with various common health risk factors over the two survey periods using weighted logistic regression analysis adjusting for age, sex, obesity category and white ethnicity (n = 28,279).ResultsOver 97% of individuals had at least one of the 19 risk factors examined with no difference in the prevalence over time (P>0.34). The prevalence of lifestyle, social/mental and physical risk factors (2.2 to 19.1%) increased over time (P0.1), but were significantly associated with a higher 5-year OR for mortality in 1999-2014 (PConclusionHealth risk factors have both increased and decreased in prevalence over time, along with changes in the association between many of the risk factors and mortality risk. Taken together, these changes complicate interpretation of temporal trends and warrant cautious interpretation of population health patterns based on surveillance data.

BackgroundAs a type of welfare technology, care robotics is now widely seen as a potential aide to rehabilitation, increasing independence and enhancing the wellbeing of people with disabilities and older adults. Research into and development of care robots have both been vigorously promoted in North America, Europe and Asia, and the competition for technological advancement in robotics is becoming fierce. AI ethics and policy guidelines are being established. However, there are still differences in attitudes and perceptions, as well as national policies regarding this type of welfare technology. Moreover, despite the anticipated usefulness, it is believed that progress has been slow in the diffusion of care robots.PurposeIn order to explore how public discourses support technological innovation, such as care robots, while preparing society for potential risks and impact, we sought to ascertain whether public discourse on care robots varies from region to region. For example, what are the hopes and promises associated with care robots and what are the concerns?MethodsTo address these questions, this article explored how care robots have been portrayed in five major broadsheet newspapers in five jurisdictions in Asia and Europe (France, Great Britain, Hong Kong SAR, Ireland and Japan). We obtained 545 articles for the period between January 2001 and September 2020, more than half of which originated in Japan. A thematic analysis was conducted of these articles written in four languages (Chinese, English, French and Japanese).ResultsPositive and negative narratives were teased out, alongside other key prominent themes identified, such as Japan as the land of robots, the pandemic, and the impact of robots on the economy. As the number of robot-related articles grew from the year 2012 onwards, narratives became more nuanced in European newspapers, but not in Asian ones. Furthermore, recent articles began to address the social and relational impact of care robots, while providing concrete examples of improvements in the quality of life for users. Further careful examination will be necessary in the future in order to establish the impact of robotics use in rehabilitation for people with disabilities, older adults, their carers and society at large.

Abstract Background The cuticular microbiomes of Acromyrmex leaf-cutting ants pose a conundrum in microbiome biology because they are freely colonisable, and yet the prevalence of the vertically transmitted bacteria Pseudonocardia, which contributes to the control of Escovopsis fungus garden disease, is never compromised by the secondary acquisition of other bacterial strains. Game theory suggests that competition-based screening can allow the selective recruitment of antibiotic-producing bacteria from the environment, by providing abundant resources to foment interference competition between bacterial species and by using Pseudonocardia to bias the outcome of competition in favour of antibiotic producers. Results Here, we use RNA-stable isotope probing (RNA-SIP) to confirm that Acromyrmex ants can maintain a range of microbial symbionts on their cuticle by supplying public resources. We then used RNA sequencing, bioassays, and competition experiments to show that vertically transmitted Pseudonocardia strains produce antibacterials that differentially reduce the growth rates of other microbes, ultimately biassing the bacterial competition to allow the selective establishment of secondary antibiotic-producing strains while excluding non-antibiotic-producing strains that would parasitise the symbiosis. Conclusions Our findings are consistent with the hypothesis that competition-based screening is a plausible mechanism for maintaining the integrity of the co-adapted mutualism between the leaf-cutting ant farming symbiosis and its defensive microbiome. Our results have broader implications for explaining the stability of other complex symbioses involving horizontal acquisition.

Abstract Zinc-anode-based electrochromic devices (ZECDs) are emerging as the next-generation energy-efficient transparent electronics. We report anatase W-doped TiO2 nanocrystals (NCs) as a Zn2+ active electrochromic material. It demonstrates that the W doping in TiO2 highly reduces the Zn2+ intercalation energy, thus triggering the electrochromism. The prototype ZECDs based on W-doped TiO2 NCs deliver a high optical modulation (66% at 550 nm), fast spectral response times (9/2.7 s at 550 nm for coloration/bleaching), and good electrochemical stability (8.2% optical modulation loss after 1000 cycles).

ABSTRACT Bacterial efflux pumps in the resistance-nodulation-cell division (RND) family of Gram-negative bacteria contribute significantly to the development of antimicrobial resistance by many pathogens. In this study, we selected the MtrD transporter protein of Neisseria gonorrhoeae as it is the sole RND pump possessed by this strictly human pathogen and can export multiple antimicrobials, including antibiotics, bile salts, detergents, dyes, and antimicrobial peptides. Using knowledge from our previously published structures of MtrD in the presence or absence of bound antibiotics as a model and the known ability of MtrCDE to export cationic antimicrobial peptides, we hypothesized that cationic peptides could be accommodated within MtrD binding sites. Furthermore, we thought that MtrD-bound peptides lacking antibacterial action could sensitize bacteria to an antibiotic normally exported by the MtrCDE efflux pump or other similar RND-type pumps possessed by different Gram-negative bacteria. We now report the identification of a novel nonantimicrobial cyclic cationic antimicrobial peptide, which we termed CASP (cationic antibiotic-sensitizing peptide). By single-particle cryo-electron microscopy, we found that CASP binds within the periplasmic cleft region of MtrD using overlapping and distinct amino acid contact sites that interact with another cyclic peptide (colistin) or a linear human cationic antimicrobial peptide derived from human LL-37. While CASP could not sensitize Neisseria gonorrhoeae to an antibiotic (novobiocin) that is a substrate for RND pumps, it could do so against multiple Gram-negative, rod-shaped bacteria. We propose that CASP (or future derivatives) could serve as an adjuvant for the antibiotic treatment of certain Gram-negative infections previously thwarted by RND transporters. IMPORTANCE RND efflux pumps can export numerous antimicrobials that enter Gram-negative bacteria, and their action can reduce the efficacy of antibiotics and provide decreased susceptibility to various host antimicrobials. Here, we identified a cationic antibiotic-sensitizing peptide (CASP) that binds within the periplasmic cleft of an RND transporter protein (MtrD) produced by Neisseria gonorrhoeae. Surprisingly, CASP was able to render rod-shaped Gram-negative bacteria, but not gonococci, susceptible to an antibiotic that is a substrate for the gonococcal MtrCDE efflux pump. CASP (or its future derivatives) could be used as an adjuvant to treat infections for which RND efflux contributes to multidrug resistance.

Summary: The SARS-CoV-2 virus has triggered a worldwide pandemic. According to the BioGrid database, CLN7 (MFSD8) is thought to interact with several viral proteins. The aim of this work was to investigate a possible involvement of CLN7 in the infection process. Experiments on a CLN7-deficient HEK293T cell line exhibited a 90% reduced viral load compared to wild-type cells. This observation may be linked to the finding that CLN7 ko cells have a significantly reduced GM1 content in their cell membrane. GM1 is found highly enriched in lipid rafts, which are thought to play an important role in SARS-CoV-2 infection. In contrast, overexpression of CLN7 led to an increase in viral load. This study provides evidence that CLN7 is involved in SARS-CoV-2 infection. This makes it a potential pharmacological target for drug development against COVID-19. Furthermore, it provides insights into the physiological function of CLN7 where still only little is known about.

Monocular visual field defects generally localize at or anterior to the optic chiasm, while homonymous hemianopias localize to the retrochiasmal visual pathway. Highly incongruous visual field defects may be difficult to identify on 24-2 Humphrey visual field testing, and this case demonstrates the value of optical coherence tomography (OCT) ganglion cell-inner plexiform layer (GCIPL) in rapidly localizing the lesion. A 54-year-old woman was found on routine examination to have an isolated superonasal quadrant visual field defect respecting the vertical meridian in the left eye only on Humphrey 24-2 SITA-Fast testing. She had a remote history of significant head trauma. Visual acuity, anterior segment, and fundus examination were normal. OCT revealed a bow-tie atrophy of the retinal nerve fiber layer in the right eye (OD), and binocular homonymous hemi-macular atrophy of OCT GCIPL, confirming the localization was the left retrochiasmal visual pathway. A repeat Humphrey 30-2 SITA-Fast visual field demonstrated that the visual field defect was also present in the OD in a highly incongruous manner. Magnetic resonance imaging of the brain with contrast showed mild atrophy of the left optic tract. This case demonstrates that highly incongruous visual field defects may be difficult to identify on Humphrey 24-2 SITA-Fast visual fields, and OCT GCIPL serves as a rapid way to localize the lesion. More detailed visual field testing including 30-2 programs should be considered in these cases.

The variation in the concentrations of ambient PM2.5 (particles with an aerodynamic diameter less than 2.5 µm) generally forms a continuous sawtooth cycle with a recurring smooth increase followed by a sharp decrease. The episode of abrupt decay of pollution is mostly meteorological in origin and is controlled by the passage of synoptic systems. One affordable and effective measure for quickly reducing PM2.5 concentrations in northern China is to wait for a strong wind to arrive. However, it is still unclear how strong the wind needs to be and exactly what kind of synoptic system most effectively results in the rapid decay of air pollution episodes. PM2.5 variations over the 28 pollution channel cities of the Beijing region are investigated to determine the mechanisms by which synoptic patterns affect the decay processes of pollution episodes. This work shows more obvious day-to-day variations in PM2.5 concentration in winter than in summer, which implies that wintertime PM2.5 variations are more sensitive to meteorological factors. There were 365 decay processes from January 2014 to March 2020, and 97 of them were related to the effective wet deposition. In total, 26 %–43 % of PM2.5 pollutant is removed by the wet deposition in different seasons. Two dominant circulation patterns are identified in summer. All the other three seasons have three circulation types (CTs), respectively. The three CTs in spring show the same patterns as those in autumn and winter. The circulation patterns beneficial to the decay processes all exhibit a higher-than-normal surface wind speed, a negative relative humidity anomaly and net outflow of PM2.5 from the domain. In addition, CT1 in spring, autumn and winter is controlled by northeasterly wind and features the most significant horizontal net outflow of air pollutants and effective upward spread of air pollutants to the free atmosphere. CT2 is the most frequent CT in autumn and winter, with the highest wind speed from the northwest, highest boundary layer height (BLH) and lowest relative humidity among the three CTs, all of which are favorable for the reduction of PM2.5 concentrations. In CT3, strong vertical wind shear within the boundary layer enhances the mixing of surface air pollutants, which is the extra cleaning mechanism besides dry and clean air mass inflow. PM2.5 concentrations show significant decreases of more than 37 %, 41 % and 27 % after the passage of CT1, CT2 and CT3, respectively. A dry airflow with a positive BLH anomaly and the effective horizontal outflow of air pollutants are the main reasons for the abrupt decay phase in summer. PM2.5 concentrations after the decay process show a significant decreasing trend from 2014 to 2020, reflecting successful emission mitigation. Emission reductions have led to a 4.3–5.7 µgm-3yr-1 decrease in PM2.5 concentrations in the 28 pollution channel cities of the Beijing region.

Flood disasters frequently threaten people and property all over the world. Therefore, an effective numerical model is required to predict the impacts of floods. In this study, a dynamic bidirectional coupled hydrologic–hydrodynamic model (DBCM) is developed with the implementation of characteristic wave theory, in which the boundary between these two models can dynamically adapt according to local flow conditions. The proposed model accounts for both mass and momentum transfer on the coupling boundary and was validated via several benchmark tests. The results show that the DBCM can effectively reproduce the process of flood propagation and also account for surface flow interaction between non-inundation and inundation regions. The DBCM was implemented for the floods simulation that occurred at Helin Town located in Chongqing, China, which shows the capability of the model for flood risk early warning and future management.

Constructing the transmission tower from LiDAR point clouds is a fundamental step for smart grid. However, currently the transmission tower construction method relies heavily on manual editing, which is far from the practical industrial application. This paper proposes a model-driven based method to realize 3D construction of transmission tower fast and accurately. This method first generates different types of 3D tower models. Then, it calculates the direction characteristic of point clouds distribution using the obtained transmission towers point clouds. While finding the principal direction of transmission towers, the local coordinates of the transmission towers are settled. And then the key points are captured in a semi-automatically way. According to these key points, the transmission tower model that best matches the point clouds is selected using the model matching algorithm. Comparing with the existing traditional manual editing methods, the method proposed in this paper can ensure the integrity and accuracy of the reconstructed tower model using the model-driven based strategy. The proposed method makes a trade-off between manual editing and efficiency, which guarantees the quality of tower modelling. And the feasibility and practicability of the proposed method are verified by the experiments on real-world point clouds data.

Smart grid construction puts higher demands on the construction of 3D models of substations. However, duo to the complex and diverse structures of substation facilities, it is still a challenge to extract the fine three-dimensional structure of the substation facilities from the massive laser point clouds. To solve this problem, this paper proposes a method for extracting substation equipment from laser scanning point clouds. Firstly, in order to improve the processing efficiency and reduce the noises, the regular voxel grid sampling method is used to down-sample the input point cloud. Furthermore, the multi-scale morphological filtering algorithm is used to segment the point cloud into ground points and non-ground points. Based on the non-ground point cloud data, the substation region is extracted using plane detection in point clouds. Then, for the filtered substation point cloud data, a three-dimensional polygon prism segmentation algorithm based on point dimension feature is proposed to extract the substation equipment. Finally, the substation LiDAR point cloud data collected by the UAV laser scanning system is used to verify the algorithm, and the qualitative and quantitative comparison analysis between the detected results and the manually extracted results are carried out. The experimental results show that the proposed method can accurately extract the substation equipment structure from the laser point cloud data. The results are consistent with the manually extracted results, which demonstrate the great potential of the proposed method in substation extraction and power system 3D modelling applications.

We quantify the connection between deep convective occurrence and summertime 100 hPa water vapor anomaly over the North American (NA) region and find substantial consistency between their interannual variations and also that the water vapor mixing ratio over the NA region is up to ∼1 ppmv higher when deep convection occurs. We use a Lagrangian trajectory model to demonstrate that the structure and the location of the NA anticyclone, as well as the tropical upper tropospheric temperature, mediate the moistening impact of convection. The deep convection mainly occurs over the Central Plains region. Most of the convectively moistened air is then transported to the center of the NA anticyclone, and the anticyclonic structure helps maintain high water vapor content there. This explains both the summer seasonal cycle and interannual variability of the convective moistening efficiency in the NA region and can provide valuable insight into modeling stratospheric water vapor.

To monitor the large area land subsidence in Dezhou city, Shandong province, China. This paper uses 23 scenes of Sentinel-1A radar images from August 2017 to February 2019, and based on small baseline subset (SBAS) technology to obtain the subsidence information in Dezhou urban area. The monitoring results show that: Dezhou city has a serious subsidence phenomenon in large coverage area. A subsidence funnel with Chenzhuang as the center was formed. The average annual subsidence rate (along the vertical direction) of the subsidence center exceeded 45 mm/yr. There was also a serious subsidence phenomenon in the eastern and northeastern parts of the urban area, and there was a tendency of forming a whole area. After detailed data analysis, it is found that the ground subsidence presents seasonal characteristics closely related to the groundwater level and is affected by large-scale engineering construction on the surface.

With the accelerated development of the global economy, the scale of UHV (Ultra High Voltage) large-capacity transmission lines has increased dramatically. Nevertheless, existing transmission line inspection methods rely on human inspectors detecting abnormal facilities from ground level or helicopters. Those methods not only require extremely high labor and equipment costs, but also are time-consuming. In this paper, a thin object 3D reconstruction method is proposed for the extraction of 3D structures of power lines from sequential oblique UAV images. The proposed method recovers the 3D structure of the power lines from three sequential images that depict the structure of power lines in 2D. Proposed method was tested in a typical transmission line corridor and the reconstructed results show the effectiveness of the proposed method.

The solution stability of river models using the one-dimensional (1D) Saint-Venant equations can be easily undermined when source terms in the discrete equations do not satisfy the Lipschitz smoothness condition for partial differential equations. Although instability issues have been previously noted, they are typically treated as model implementation issues rather than as underlying problems associated with the form of the governing equations. This study proposes a new reference slope form of the Saint-Venant equations to ensure smooth slope source terms and eliminate one source of potential numerical oscillations. It is shown that a simple algebraic transformation of channel geometry provides a smooth reference slope while preserving the correct cross-section flow area and the total Piezometric pressure gradient that drives the flow. The reference slope method ensures the slope source term in the governing equations is Lipschitz continuous while maintaining all the underlying complexity of the real-world geometry. The validity of the mathematical concept is demonstrated with the open-source Simulation Program for River Networks (SPRNT) model in a series of artificial test cases and a simulation of a small urban creek. Validation comparisons are made with analytical solutions and the Hydrologic Engineering Center's River Analysis System (HEC-RAS) model. The new method reduces numerical oscillations and instabilities without requiring ad hoc smoothing algorithms.

The medial sural artery perforator (MSAP) flap is a versatile fasciocutaneous flap that has been used successfully in the reconstruction of defects across the body. In specific cases, it may prove superior to more commonly used options (e.g., anterolateral thigh flap and radial forearm free flap). Historically, a disadvantage of the MSAP flap is the relatively small surface area it provides for reconstruction. We recently encountered a patient with extensive pelvic injuries from prior trauma resulting in significant scarring and contracture of the groin, tethering of the penis, and loss of the scrotum and one testicle. The patient was unable to achieve erection from tethering and his remaining testicle had been buried in the thigh. In considering the reconstructive options, he was not a suitable candidate for a thigh-based or forearmbased flap. An extended MSAP flap measuring 25 cm×10 cm was used for resurfacing of the groin and pelvis as well as for the formation of a neoscrotum. This report is the first to document an MSAP flap utilized for simultaneous groin resurfacing and scrotoplasty. Additionally, the dimensions of this flap make it the largest recorded MSAP flap to date.

Abstract Background Socioeconomic inequalities in access to, and utilization of medical care have been shown in many jurisdictions. However, the extent to which they exist at end-of-life (EOL) remains unclear. Methods Studies in MEDLINE, EMBASE, CINAHL, ProQuest, Web of Science, Web of Knowledge, and OpenGrey databases were searched through December 2019 with hand-searching of in-text citations. No publication date or language limitations were set. Studies assessing SES (e.g. income) in adults, correlated to EOL costs in last year(s) or month(s) of life were selected. Two independent reviewers performed data abstraction and quality assessment, with inconsistencies resolved by consensus. Results A total of twenty articles met eligibility criteria. Two meta-analyses were performed on studies that examined total costs in last year of life – the first examined costs without adjustments for confounders (n = 4), the second examined costs that adjusted for confounders, including comorbidities (n = 2). Among studies which did not adjust for comorbidities, SES was positively correlated with EOL costs (standardized mean difference, 0.13 [95% confidence interval, 0.03 to 0.24]). However, among studies adjusting for comorbidities, SES was inversely correlated with EOL expenditures (regression coefficient, −$150.94 [95% confidence interval, −$177.69 to -$124.19], 2015 United States Dollars (USD)). Higher ambulatory care and drug expenditure were consistently found among higher SES patients irrespective of whether or not comorbidity adjustment was employed. Conclusion Overall, an inequality leading to higher end-of-life expenditure for higher SES patients existed to varying extents, even within countries providing universal health care, with greatest differences seen for outpatient and prescription drug costs. The magnitude and directionality of the relationship in part depended on whether comorbidity risk-adjustment methodology was employed.

Abstract The dimension is a leading parameter in customization of unique optoelectronic properties of halide perovskite, while it is still challenging to achieve dimension control beyond the well‐developed composition regulation‐based strategy considering their intrinsic feature of ionic crystal. In this paper, ligands‐directed confinement effect is emphasized in rendering the reduced‐dimensional engineering of halide perovskite, and quasi‐2D (OAm)2CsPb2Br7 (n = 2) nanosheets are obtained by introducing a large amount of metal ions. As a representation, with Mg2+ participation, the initial adsorption of organic ligands (i.e., OA and OAm) to inorganic components (i.e., [PbBr6]4−) is promoted, conferring a ligands‐directed surface reconstruction to form the lamellar structure composed of alternate organic and inorganic layers. Namely, the inorganic–organic lamellar ensemble will play as a soft template, which effectively restricts the growth of [PbBr6]4− in the c‐axis direction even after the Cs‐OA injection, thus producing the anisotropic layered perovskite of (OAm)2CsPb2Br7 from CsPbBr3 scenario. Notably, such reduced‐dimensional engineering enables a remarkable luminescence tailoring, of which the deep blue emission centered at 439 nm is achieved. In addition, benefiting from such universal strategy, the luminescence is also dependent on the species of introduced metal ions (e.g., Ca2+, Co2+, Sr2+, and Mn2+).

Several neurological disorders may be amenable to treatment with gene-targeting therapies such as antisense oligonucleotides (ASOs) or viral vector-based gene therapy. The US FDA has approved several of these treatments; many others are in clinical trials. Preclinical toxicity studies of ASO candidates have identified dose-dependent neurotoxicity patterns. These include degeneration of dorsal root ganglia, the cell bodies of peripheral sensory neurons. Quantitative sensory testing (QST) refers to a series of standardized mechanical and/or thermal measures that complement clinical neurologic examination in detecting sensory dysfunction. QST primarily relies on patient self-report or task performance (i.e., button-pushing). This brief report illustrates individualized pragmatic approaches to QST in non-verbal subjects receiving early phase investigational intrathecal drug therapies as a component of clinical trial safety protocols. Three children with neurodevelopmental disorders that include Neuronal Ceroid Lipofuscinosis Type 7, Ataxia-Telangiectasia, and Epilepsy of Infancy with Migrating Focal Seizures are presented. These case studies discuss individualized testing protocols, accounting for disease presentation, cognitive and motor function. We outline specific considerations for developing assessments for detecting changes in sensory processing in diverse patient groups and safety monitoring trials of early phase investigational intrathecal drug therapies. QST may complement information obtained from the standard neurologic examination, electrophysiologic studies, skin biopsies, and imaging. QST has limitations and challenges, especially in non-verbal subjects, as shown in the three cases discussed in this report. Future directions call for collaborative efforts to generate sensory datasets and share data registries in the pediatric neurology field.

ABSTRACT Antibiotic resistance among bacterial pathogens continues to pose a serious global health threat. Multidrug-resistant (MDR) strains of the Gram-negative organism Acinetobacter baumannii utilize a number of resistance determinants to evade current antibiotics. One of the major resistance mechanisms employed by these pathogens is the use of multidrug efflux pumps. These pumps extrude xenobiotics directly out of bacterial cells, resulting in treatment failures when common antibiotics are administered. Here, the structure of the novel tetracycline antibiotic TP-6076, bound to both the Acinetobacter drug efflux pump AdeJ and the ribosome from Acinetobacter baumannii, using single-particle cryo-electron microscopy (cryo-EM), is elucidated. In this work, the structure of the AdeJ–TP-6076 complex is solved, and we show that AdeJ utilizes a network of hydrophobic interactions to recognize this fluorocycline. Concomitant with this, we elucidate three structures of TP-6076 bound to the A. baumannii ribosome and determine that its binding is stabilized largely by electrostatic interactions. We then compare the differences in binding modes between TP-6076 and the related tetracycline antibiotic eravacycline in both targets. These differences suggest that modifications to the tetracycline core may be able to alter AdeJ binding while maintaining interactions with the ribosome. Together, this work highlights how different mechanisms are used to stabilize the binding of tetracycline-based compounds to unique bacterial targets and provides guidance for the future clinical development of tetracycline antibiotics. IMPORTANCE Treatment of antibiotic-resistant organisms such as A. baumannii represents an ongoing issue for modern medicine. The multidrug efflux pump AdeJ serves as a major resistance determinant in A. baumannii through its action of extruding antibiotics from the cell. In this work, we use cryo-EM to show how AdeJ recognizes the experimental tetracycline antibiotic TP-6076 and prevents this drug from interacting with the A. baumannii ribosome. Since AdeJ and the ribosome use different binding modes to stabilize interactions with TP-6076, exploiting these differences may guide future drug development for combating antibiotic-resistant A. baumannii and potentially other strains of MDR bacteria.

The development of electrochromic materials has opened the door to the development of numerous devices including smart windows, color displays, optical filters, wearable camouflages, among others. Although the current electrochromic devices do not consume energy while maintaining their colored or colorless states, their bistable operation requires external electrical energy to be consumed during switching. To reduce the energy consumption of an electrochromic device, an emerging Zn anode‐based electrochromic device concept was recently introduced to partially retrieve the consumed electrical energy. In this Review, key technological developments and scientific challenges are presented for a broad range of Zn anode‐based electrochromic device configurations with emphasis on the inherent distinctions between the Zn anode‐based and conventional electrochromic devices. Specifically, a comprehensive comparison of the two classes of electrochromic devices in the high‐performance device design is provided. For the electrochromic layer, the methods for obtaining high‐quality electrochromic materials and thin films are reviewed. For the electrolytes, the effect of the dual ion system on the electrochromic performance is discussed. Also, some critical but unresolved issues in the device design and fabrication are discussed. The perspectives and outlook at the end of this Review provide recommendations to improve performance for future electrochromic studies.

Abstract 0D perovskites have gained much attention in recent years due to their fascinating properties derived from their peculiar structure with isolated metal halide octahedra or metal halide clusters. However, the systematic discussion on the crystal and electronic structure of 0D perovskites to further understand their photophysical characteristics and the comprehensive overview of 0D perovskites for their further applications are still lacking. In this review, the unique crystal and electronic structure of 0D perovskites and their diverse properties are comprehensively analyzed, including large bandgaps, high exciton binding energy, and largely Stokes‐shifted broadband emissions from self‐trapped excitons. Furthermore, the photoluminescence regulation are discussed. Then, the various synthetic methods for 0D perovskite single crystals, nanocrystals, and thin films are comprehensively summarized. Finally, the emerging applications of 0D perovskites to light‐emitting diodes, solar cells, detectors, and some others are illustrated, and the outlook on future research in the field is also provided.

Energy consumption of buildings during heating and cooling accounts for about 15% of global total energy consumption. Advanced dynamic switchable windows that enable independent control of solar heat will contribute to optimal energy efficiency in heating, cooling, and artificial lighting systems throughout buildings. Recently, energy‐efficient plasmonic electrochromic smart windows (PESWs) based on metal oxide nanocrystals (NCs) have been gaining increasing attention due to their effective and controllable regulation over the near‐infrared region of the solar spectrum without affecting the dynamic visible transmittance of the smart windows. Herein, the current state‐of‐the‐art results of colloidal metal oxide NCs for PESWs are highlighted, along with their design strategies and working principles. The recent research status of PESWs in typical colloidal metal oxide NCs is reviewed in detail, and the challenges and corresponding countermeasures in this field are discussed. Furthermore, an outlook into novel opportunities in PESW‐related academic research and how to accelerate the pace of real‐world applications is presented.

In situ measurements reveal that proton temperature anisotropy is ubiquitous in the solar wind. Various plasma instabilities have been proposed to regulate the distribution of the proton temperature anisotropy in the solar wind; detailed constraint processes are still unclear. In this paper, we study the effects of alpha beams on both the forward and backward proton temperature anisotropy instabilities at parallel and oblique propagation with the Vlasov theory, and compare the theoretical results with the Wind observation. As the alpha-beam drift velocity v _α / v _A increases, the growth rates of forward Alfvén/ion-cyclotron (FA/IC) and backward magnetosonic/whistler (BM/W) instabilities increase, those of backward Alfvén/ion-cyclotron (BA/IC) and forward magnetosonic/whistler (FM/W) instabilities decrease, and those of the mirror and forward Alfvén wave (FAW) instabilities are nearly constant. In particular, there are different constraining mechanisms on the distribution of proton temperature anisotropy for different values of the alpha-beam drift velocity. The proton temperature anisotropy instability together with the alpha beam can provide a potential explanation for the distribution of the proton temperature anisotropy in the solar wind.

We report the X-ray timing results of the black hole candidate MAXI J1820+070 during its 2018 outburst using the Hard X-ray Modulation Telescope (Insight-HXMT) and Neutron Star Interior Composition Explorer Mission (NICER) observations. Low-frequency quasi-periodic oscillations (LFQPOs) are detected in the low/hard state and the hard intermediate state, which lasted for ∼90 days. Thanks to the large effective area of Insight-HXMT at high energies and NICER at low energies, we are able to present the energy dependence of the LFQPO characteristics and phase lags from 0.2 to 200 keV, which has never been explored by previous missions. We find that the centroid frequency of the LFQPOs does not change significantly with energy, while the full width at half maximum and fractional rms show a complex evolution with energy. The LFQPO phase lags at high energies and low energies show consistent energy-dependence relations taking the ∼2 keV as reference. Our results suggest that the LFQPOs from high energy come from the LT precession of the relativistic jet, while the low-energy radiation is mainly from the perpendicular innermost regions of the accretion disk.

On 2020 April 19–20, a solar ejection was seen by spacecraft in a radial alignment that included Solar Orbiter and Wind. The ejection contained a magnetic flux rope where magnetic field and plasma parameters were well correlated between spacecraft. This structure is called an “unperturbed magnetic flux rope” (UMFR). Ahead of the UMFR is a portion of the ejection (not sheath) that is referred to as “upstream” (US). We focus on the US and inquire why the correlation is so much weaker there. Specifically, we analyze data collected by Solar Orbiter at 0.81 au and Wind at L1. We show that a plausible cause for the lack of coherence in the US is a combination of front erosion and internal reconnection occurring there. Front erosion is inferred from an analysis of azimuthal magnetic flux balance in the UMFR. In the present case, we contend that the US, rather than the UMFR, is the source of the eroded field lines. The presence of erosion is supported further by a direct comparison of the magnetic field data at both spacecraft that shows, in particular, a massive shrinkage of the front portion of the US. Internal reconnection is also happening at thin current sheets inside the US. Strong nonradial flows are reconfiguring the structure. As a result of these reconnection processes, a whole section of the US is disrupted and field lines move down the flanks of the ejection and out of view of Wind.

Black widows are millisecond pulsars ablating their companions. The material blown from the companion blocks the radio emission, resulting in radio eclipses. The properties of the eclipse medium are poorly understood. Here, we present direct evidence of the existence of magnetic fields in the eclipse medium of the black widow PSR J2051−0827 using observations made with the Five-hundred-meter Aperture Spherical radio Telescope. We detect a regular decrease in rotation measure (RM) in the egress of the eclipse, changing from 60 to −28.7 rad m ^−2 . The RM gradually changes back to normal when the line of sight moves away from the eclipse. The estimated line-of-sight magnetic field strength in the eclipse medium is ∼0.1 G. The RM reversal could be caused by a change in the magnetic field strength along the line of sight due to binary orbital motion. The RM reversal phenomenon has also been observed in some repeating fast radio bursts (FRBs), and the study of spider pulsars may provide additional information about the origin of FRBs.

In situ measurements from spacecraft typically provide a time series at a single location through coronal mass ejections (CMEs), and they have been one of the main methods to investigate CMEs. The CME properties derived from these in situ measurements are affected by temporal changes that occur as the CME passes over the spacecraft, such as radial expansion and aging, as well as spatial variations within a CME. This study uses multispacecraft measurements of the same CME at close separations to investigate both the spatial variability (how different a CME profile is when probed by two spacecraft close to each other) and the so-called aging effect (the effect of the time evolution on in situ properties). Although low-frequency quasiperiodic oscillations (LFQPOs) are commonly detected in the X-ray light curves of accreting black hole X-ray binaries, their origin still remains elusive. In this study, we conduct phase-resolved spectroscopy in a broad energy band for LFQPOs in MAXI J1820+070 during its 2018 outburst, utilizing Insight-HXMT observations. By employing the Hilbert–Huang transform method, we extract the intrinsic quasiperiodic oscillation (QPO) variability, and obtain the corresponding instantaneous amplitude, phase, and frequency functions for each data point. With well-defined phases, we construct QPO waveforms and phase-resolved spectra. By comparing the phase-folded waveform with that obtained from the Fourier method, we find that phase folding on the phase of the QPO fundamental frequency leads to a slight reduction in the contribution of the harmonic component. This suggests that the phase difference between QPO harmonics exhibits time variability. Phase-resolved spectral analysis reveals strong concurrent modulations of the spectral index and flux across the bright hard state. The modulation of the spectral index could potentially be explained by both the corona and jet precession models, with the latter requiring efficient acceleration within the jet. Furthermore, significant modulations in the reflection fraction are detected exclusively during the later stages of the bright hard state. These findings provide support for the geometric origin of LFQPOs and offer valuable insights into the evolution of the accretion geometry during the outburst in MAXI J1820+070.
