Abstract

The thermochemical process of gasification is widely utilized for bioenergy production and is particularly attractive due to its high conversion efficiency. However, a gasification byproduct, known as char, is commonly treated as industrial waste despite its attractive qualities. Indeed, due to its high carbon content (up to 91%) and well-developed porosity (specific surface areas up to 1253 m2 g−1), gasification char could be considered a cost-effective substitute for activated carbon in various applications, such as catalysis and adsorption. However, its properties are highly dependent on the gasification parameters and the initial feedstock selected, and thus a careful characterization of the material is needed to find the most suitable applications. This review can act as a preliminary assessment of the gasification along with the expected char properties, aiding in the selection of the most appropriate valorization pathways. In particular, different application areas, their relation to the gasification process, and the char properties are extensively discussed.

60. Electronic structure theory on modeling short-range noncovalent interactions between amino acids.

Abstract

While short-range noncovalent interactions (NCIs) are proving to be of importance in many chemical and biological systems, these atypical bindings happen within the so-called van der Waals envelope and pose an enormous challenge for current computational methods. We introduce SNCIAA, a database of 723 benchmark interaction energies of short-range noncovalent interactions between neutral/charged amino acids originated from protein x-ray crystal structures at the "gold standard" coupled-cluster with singles, doubles, and perturbative triples/complete basis set [CCSD(T)/CBS] level of theory with a mean absolute binding uncertainty less than 0.1 kcal/mol. Subsequently, a systematic assessment of commonly used computational methods, such as the second-order Møller−Plesset theory (MP2), density functional theory (DFT), symmetry-adapted perturbation theory (SAPT), composite electronic-structure methods, semiempirical approaches, and the physical-based potentials with machine learning (IPML) on SNCIAA is carried out. It is shown that the inclusion of dispersion corrections is essential even though these dimers are dominated by electrostatics, such as hydrogen bondings and salt bridges. Overall, MP2, ωB97M-V, and B3LYP+D4 turned out to be the most reliable methods for the description of short-range NCIs even in strongly attractive/repulsive complexes. SAPT is also recommended in describing short-range NCIs only if the δMP2 correction has been included. The good performance of IPML for dimers at close-equilibrium and long-range conditions is not transferable to the short-range. We expect that SNCIAA will assist the development/improvement/validation of computational methods, such as DFT, force-fields, and ML models, in describing NCIs across entire potential energy surfaces (short-, intermediate-, and long-range NCIs) on the same footing. Abstract

We recently identified Zinc-finger protein 768 (ZNF768) as a novel transcription factor controlling cell fate decision downstream of Rat sarcoma virus (RAS). We showed that ZNF768 depletion impairs cell cycle progression and triggers cellular senescence, while its overexpression allows cells to bypass oncogene-induced senescence. Elevated ZNF768 levels is common in tumors, suggesting that ZNF768 may help to escape cellular senescence, sustain proliferation and promote malignant transformation. Here, we discuss these recent findings and highlight key questions emerging from our work.

Abstract

Structure-borne sound sources affect the acoustic quality inside buildings. Based on many years of research, methods have been standardised to characterise structure-borne sound sources in the laboratory (EN 15657) and to predict the sound transmission from these sources when installed in buildings (EN 12354-5). This contribution is about the uncertainty of the source descriptors used in EN 15657 and the predicted installed sound power which is the input quantity for EN 12354-5. Recently an interlaboratory test with 7 participating laboratories has been performed. The task was to measure the blocked force, free velocity and mobility of an artificial source and the blocked force of the ISO tapping machine. From the evaluation of the measurement results estimates for the uncertainties of the source quantities and the installed power could be deduced.

Abstract

A low immission due to structure-borne sound sources is a major component of the acoustic quality inside buildings. After many years of research, methods have been standardised to characterise such sources (EN 15657) and to predict their impact in buildings (EN 12354-5). This contribution is dedicated to the question what the uncertainty of the source descriptors and the predicted installed sound power is. To answer this question, an interlaboratory test with an artificial source was performed. Altogether seven laboratories participated, and estimates for the uncertainties of the source quantities could be deduced from the measurement results. Additionally, measurements were performed with a standardised structure-borne sound source, the ISO tapping machine, by all participating laboratories. The measured source quantity for this source turned out to be in good agreement with the theoretically predicted values thereby validating this theoretical prediction.

Abstract

Objective: The mechanistic target of rapamycin (mTOR) is a serine/threonine kinase that functions into distinct protein complexes (mTORC1 and mTORC2) that regulates growth and metabolism. DEP-domain containing mTOR-interacting protein (DEPTOR) is part of these complexes and is known to reduce their activity. Whether DEPTOR loss affects metabolism and organismal growth in vivo has never been tested. Methods: We have generated a conditional transgenic mouse allowing the tissue-specific deletion of DEPTOR. This model was crossed with CMV-cre mice or Albumin-cre mice to generate either whole-body or liver-specific DEPTOR knockout (KO) mice. Results: Whole-body DEPTOR KO mice are viable, fertile, normal in size, and do not display any gross physical and metabolic abnormalities. To circumvent possible compensatory mechanisms linked to the early and systemic loss of DEPTOR, we have deleted DEPTOR specifically in the liver, a tissue in which DEPTOR protein is expressed and affected in response to mTOR activation. Liver-specific DEPTOR null mice showed a reduction in circulating glucose upon fasting versus control mice. This effect was not associated with change in hepatic gluconeogenesis potential but was linked to a sustained reduction in circulating glucose during insulin tolerance tests. In addition to the reduction in glycemia, liver-specific DEPTOR KO mice had reduced hepatic glycogen content when fasted. We showed that loss of DEPTOR cell-autonomously increased oxidative metabolism in hepatocytes, an effect associated with increased cytochrome c expression but independent of changes in mitochondrial content or in the expression of genes controlling oxidative metabolism. We found that liver-specific DEPTOR KO mice showed sustained mTORC1 activation upon fasting, and that acute treatment with rapamycin was sufficient to normalize glycemia in these mice. Conclusion: We propose a model in which hepatic DEPTOR accelerates the inhibition of mTORC1 during the transition to fasting to adjust metabolism to the nutritional status. Abstract

International audience; Planning the energy transition requires decision makers to have an in-depth knowledge about a given territory. To achieve this, data is collected from multiple sources, at multiple scales, with constraints such as privacy policies. Resulting data informs about given areas of space without a specific point location. Such is the case of Energy Performance Certificate (EPC). EPC databases are released under specific constraints: anonymization, geo-localization with postal address, missing details. This paper shows that learning the observed EPCs to predict missing ones can also be seen as a spatial interpolation problem. It presents a way to treat EPC as a geo-localized information and predict its value at building level. Kriging methodology is applied to random fields observed at random locations to find a Best Linear Unbiased Predictor (BLUP). This new model is referred to as Mixture Kriging. While the usual Gaussian setting is lost, we show that conditional mean, variance and covariance can be derived. This new model gives interesting results in EPC prediction at building level which is a prerequisite for decision maker to target renovation efforts. The specific case of a city in France is taken as an example. The presented model includes Mixture co-Kriging so that covariates can be used to improve the result. It is also suggested that Mixture Kriging can be usefully implemented to control uncertainty propagation. We present potential applications on simulated data.; Pour planifier la transition énergétique, les décideurs ont besoin d’une connaissance approfondie de leur territoire. Pour cela, des données sont collectées de sources multiples, à plusieurs échelles, avec des contraintes comme les politiques de confidentialité. De telles données fournissent des informations sur des zones spatiales mais sans localisation spécifique. C’est le cas du Diagnostic de Performance Energétique (DPE). Les bases de données des DPE réalisés sont publiées sous des contraintes spécifiques : anonymisation, géolocalisation avec adresse postale, détails manquants. Ce document montre que l’apprentissage des DPE observés pour prédire les DPE manquants peut être considéré comme un problème d’interpolation spatiale. Il présente une manière de traiter le DPE en tant qu’information géolocalisée et de prédire sa valeur au niveau du bâtiment. La méthodologie du krigeage est appliquée à des champs aléatoires observés à des emplacements aléatoires pour trouver le meilleur prédicteur linéaire non biaisé (BLUP). Ce nouveau modèle est appelé krigeage de mixtures. Bien que le cadre gaussien habituel soit perdu, nous montrons que la moyenne conditionnelle, la variance et la covariance peuvent être calculées. Ce nouveau modèle donne des résultats intéressants dans la prédiction du DPE au niveau du bâtiment, ce qui est une condition préalable pour que les décideurs ciblent les efforts de rénovation. Le cas spécifique d’une ville française est pris comme exemple. Le modèle présenté inclut également le co-krigeage de mixtures de sorte que les covariables puissent être utilisées pour améliorer le résultat. Il est également suggéré que le krigeage de mixtures puisse être utilement mis en oeuvre pour contrôler la propagation de l’incertitude. Nous présentons des applications en ce sens sur des données simulées.
