Abstract

We have recently shown that fluoxetine (FX) suppressed polyinosinic-polycytidylic acid-induced inflammatory response and endothelin release in human epidermal keratinocytes, via the indirect inhibition of the phosphoinositide 3-kinase (PI3K)-pathway. Because PI3K-signaling is a positive regulator of the proliferation, in the current, highly focused follow-up study, we assessed the effects of FX (14 µM) on the proliferation and differentiation of human epidermal keratinocytes. We found that FX exerted anti-proliferative actions in 2D cultures (HaCaT and primary human epidermal keratinocytes [NHEKs]; 48- and 72-h; CyQUANT-assay) as well as in 3D reconstructed epidermal equivalents (48-h; Ki-67 immunohistochemistry). Importantly, FX did not influence epidermal thickness (hematoxylin-eosin staining), and it did not have a major impact on the differentiation-associated alteration of the gene expression pattern (24-h treatments; RNA-Seq). Moreover, neither keratin (K)-1, nor K10 expression was altered by FX in NHEKs (RT-qPCR) or in 3D epidermal equivalents (semi-quantitative immunohistomorphometry). FX did not influence differentiation-induced up-regulation of occludin (RT-qPCR; NHEKs), and did not alter differentiation-associated barrier forming capacity of epidermal keratinocytes (electrical impedance; Lucifer Yellow penetration assay). Our data indicate that, besides the previously reported combined anti-inflammatory and putative anti-pruritic effects, FX may also suppress proliferation of human epidermal keratinocytes without impairing their differentiation and barrier-forming capacity. Abstract

The signature of human serum IgG glycosylation is critical in the defense against pathogens. Alterations of IgG N-glycome were associated with COVID-19 (Coronavirus disease 2019) severity, although knowledge on the response to vaccination is limited. IgG N-glycome was analyzed in this study in post-COVID-19 and post-vaccination patients to reveal potential glycosylation-based alterations using hydrophilic interaction liquid chromatography (HILIC-UPLC) with fluorescence (FLR) and mass-spectrometric (MS) detection. IgG antibodies were purified from serum samples through protein G affinity chromatography followed by PNGase F digestion-based deglycosylation. The released glycans were fluorescently derivatized by procainamide labeling and purified via solid-phase extraction. Higher levels of sialylation and afucosylation were identified in post-COVID-19 patients, which was further expanded by vaccination, but only in those who were previously SARS-CoV-2 (Severe acute respiratory syndrome coronavirus 2) infected. Abstract

Background: Subarachnoid hemorrhage is a serious condition caused by ruptured intracranial aneurysms, resulting in severe disability mainly in young adults. Cerebral vasospasm is one of the most common complication of subarachnoid hemorrhage; thus, active prevention is key to improve the prognosis. The glycosylation of proteins is a critical quality attribute which is reportedly altered in patients diagnosed with acute ischemic stroke. In this study, we examined the N-glycosylation profile of serum glycoproteins in patients with subarachnoid hemorrhage without vasospasm compared to patients with vasospasm. Methods: The serum N-glycans were released by PNGase F (Peptide: N-glycosidase F) digestion and subsequently labeled by procainamide via reductive amination. The samples were analyzed by hydrophilic-interaction liquid chromatography after solid-phase extraction-based sample purification. Results: Besides the glycosylation pattern, we also investigated the biomarkers following subarachnoid hemorrhage. Multiple statistical analyses were performed in order to find significant differences and identify potential prediction factors of cerebral vasospasm. Significant differences were identified such as higher sialylation on bi-, tri-, and tetra-antennary structures in patients with subarachnoid hemorrhage and cerebral vasospasm. Abstract

Background and purpose: MicroRNA (miRNA) therapy is a promising approach to induce cardioprotection. We have previously identified cardiac microRNA‐125b* (microRNA‐125b‐2‐3p; miR‐125b*) as a potential cardioprotective miRNA, termed ProtectomiR. We aimed to characterize the pharmacokinetics and pharmacodynamics, and the effect of miR‐125b* mimic on infarct size using an in vivo mouse model. Experimental approach: To characterize the pharmacokinetics properties of miR‐125b* mimic, a single injection of 10‐μg miR‐125b* mimic or its scramble miRNA control, or vehicle i.v. was given to C57BL/6 mice. MiR‐125b* expression was measured from plasma, heart, kidney and liver samples. Effect of miR‐125b* on area at risk and infarct size was assessed after 45‐min coronary occlusion, followed by 24‐h reperfusion; 10‐μg miR‐125b* mimic or 10‐μg non‐targeting miRNA mimic control or vehicle were administered via the right jugular vein at 10th mins of coronary occlusion. To assess molecular mechanism involved in cardioprotection, expression of mRNA targets of miR‐125b* were measured from ventricular myocardium at 1, 2, 4, 8 or 24 h post‐treatment using quantitative real time polymerase chain reaction. Key results: MiR‐125b* expression was markedly increased in plasma and myocardium 1 h, and in the liver 2h after treatment. Infarct size was significantly reduced after miR‐125b* mimic treatment when compared to the vehicle. The expression of Ccna2, Eef2k and Cacnb2 target mRNAs was significantly reduced 8 h after injection of miR‐125b* mimic. Conclusion and implications: This is the first demonstration of pharmacokinetic and molecular pharmacodynamic properties as well as the cardioprotective effect of miR‐125b* mimic in vivo. Abstract

Parallaxes for 331 classical Cepheids, 31 Type II Cepheids and 364 RR Lyrae stars in common between Gaia and the Hipparcos and Tycho-2 catalogues are published in Gaia Data Release 1 (DR1) as part of the Tycho-Gaia Astrometric Solution (TGAS). In order to test these first parallax measurements of the primary standard candles of the cosmological distance ladder, that involve astrometry collected by Gaia during the initial 14 months of science operation, we compared them with literature estimates and derived new period-luminosity ($PL$), period-Wesenheit ($PW$) relations for classical and Type II Cepheids and infrared $PL$, $PL$-metallicity ($PLZ$) and optical luminosity-metallicity ($M_V$-[Fe/H]) relations for the RR Lyrae stars, with zero points based on TGAS. The new relations were computed using multi-band ($V,I,J,K_{\mathrm{s}},W_{1}$) photometry and spectroscopic metal abundances available in the literature, and applying three alternative approaches: (i) by linear least squares fitting the absolute magnitudes inferred from direct transformation of the TGAS parallaxes, (ii) by adopting astrometric-based luminosities, and (iii) using a Bayesian fitting approach. TGAS parallaxes bring a significant added value to the previous Hipparcos estimates. The relations presented in this paper represent first Gaia-calibrated relations and form a "work-in-progress" milestone report in the wait for Gaia-only parallaxes of which a first solution will become available with Gaia's Data Release 2 (DR2) in 2018., Comment: 29 pages, 25 figures. Abstract

Context. The first Gaia Data Release contains the Tycho-Gaia Astrometric Solution (TGAS). This is a subset of about 2 million stars for which, besides the position and photometry, the proper motion and parallax are calculated using Hipparcos and Tycho-2 positions in 1991.25 as prior information. Aims. We investigate the scientific potential and limitations of the TGAS component by means of the astrometric data for open clusters. Methods. Mean cluster parallax and proper motion values are derived taking into account the error correlations within the astrometric solutions for individual stars, an estimate of the internal velocity dispersion in the cluster, and, where relevant, the effects of the depth of the cluster along the line of sight. Internal consistency of the TGAS data is assessed. Results. Values given for standard uncertainties are still inaccurate and may lead to unrealistic unit-weight standard deviations of least squares solutions for cluster parameters. Reconstructed mean cluster parallax and proper motion values are generally in very good agreement with earlier Hipparcos-based determination, although the Gaia mean parallax for the Pleiades is a significant exception. We have no current explanation for that discrepancy. Most clusters are observed to extend to nearly 15 pc from the cluster centre, and it will be up to future Gaia releases to establish whether those potential cluster-member stars are still dynamically bound to the clusters. Conclusions. The Gaia DR1 provides the means to examine open clusters far beyond their more easily visible cores, and can provide membership assessments based on proper motions and parallaxes. 69. Investigation of Host–Guest Interactions in 2-Ureido-4-ferrocenylpyrimidine Derivatives.

Abstract

In the present study, synthesis, conformational behavior, host–guest complex formation, and electrochemical properties of novel 6-substituted-2-ureido-4-ferrocenylpyrimidines were explored. A comprehensive NMR spectroscopic investigation was carried out to confirm the structure and conformational equilibrium of the ureidopyrimidines through studying the temperature- and concentration dependence of NMR spectra. Low-temperature NMR measurements were used to clarify structural changes inflicted by a 2,6-diaminopyridine guest. Association constant (Kassoc) values of host–guest complexes were calculated based on low-temperature titrations. It was shown that the introduction of a pyridin-2-yl substituent in the pyrimidine ring in host 10 induced a considerable change not only in the conformational equilibrium of the host itself but also in that of the host–guest complex. Geometries and relative stabilities of the conformers of host 10 as well as its host–guest complexes were determined by quantum chemical calculations. Electrochemical behavior of ureidopyrimidine hosts and host–guest complexes was investigated by cyclic voltammetry (CV) and linear sweep voltammetry (LSV) measurements. Two ureidopyrimidine derivatives were immobilized on the surface of spectral graphite electrodes, and their electrochemical response on the addition of 2,6-diaminopyridine was compared. These results also supported the importance of the pyridin-2-yl substituent in the efficient sensing of the guest. [ABSTRACT FROM AUTHOR]

Abstract

This paper aims to analyze comprehensively the climate exposure, sensitivity, perception, adaptive capacity, vulnerability, and resilience of the Hungarian agricultural sector, particularly focusing on fruit, vegetable, and grape producers. Four distinct Hungarian case studies were examined, representing different regions with diverse environmental and socioeconomic conditions. The research combined quantitative and qualitative methods, including statistical and GIS analysis of climate, agricultural, and socioeconomic data, as well as field research and semi-structured interviews. The study investigated exposure, sensitivity, perception, and adaptation, leading to the identification of key components and influencing factors. Qualitative research revealed that farms operating in geographically close proximity, in the same regulatory and support environment, can have different adaptive capacities. In the current state of the adaptation process, the extent to which farmers can rely on supportive professional networks and seek out and adopt new practices is crucial. Field experience suggests that without a strong and supportive producer organization (extension network), farmers may prefer to resort to extensification (afforestation) to mitigate production risks. From a development policy perspective, it is worthwhile to present good practices and provide information on possible adaptation techniques through existing local sectoral organizations.

Abstract

Calixarene 0118 (OTX008) and chrysin (CHR) are promising molecules for the treatment of fibrosis and diabetes complications but require an effective delivery system to overcome their low solubility and bioavailability. Sulfobutylated β-cyclodextrin (SBECD) was evaluated for its ability to increase the solubility of CHR by forming a ternary complex with OTX008. The resulting increase in solubility and the mechanisms of complex formation were identified through phase-solubility studies, while dynamic light-scattering assessed the molecular associations within the CHR-OTX008-SBECD system. Nuclear magnetic resonance, differential scanning calorimetry, and computational studies elucidated the interactions at the molecular level, and cellular assays confirmed the system’s biocompatibility. Combining SBECD with OTX008 enhances CHR solubility more than using SBECD alone by forming water-soluble molecular associates in a ternary complex. This aids in the solubilization and delivery of CHR and OTX008. Structural investigations revealed non-covalent interactions essential to complex formation, which showed no cytotoxicity in hyperglycemic in vitro conditions. A new ternary complex has been formulated to deliver promising antifibrotic agents for diabetic complications, featuring OTX008 as a key structural and pharmacological component.

Abstract

Background: Our previous studies demonstrated that sour cherry anthocyanins (AC) reduce the salivary count of Streptococcus mutans and inhibit salivary amylase activity within 30 minutes after chewing AC gum. AC gum and changing toothbrushes after scaling reduced the Gram-negative species in the unstimulated salivary microbiota. The present study examined the effect of AC gums on salivary factors, including changes in microbiome. Methods: The study was conducted over three weeks with two groups; young adults (18–30) and adults (30–45). Ten participants changed their toothbrushes, while the other 10 participants did not change after the control period. After scaling, all participants received three doses of AC gum daily. The salivary mRNA and protein levels of cytokines, mucins, melatonin, and the microbiota of unstimulated and stimulated saliva were determined by polymerase chain reaction, enzyme-linked immunosorbent assay, and 16S rRNA gene sequencing. Results: Significantly higher levels of tumor necrosis factor α (TNFα), interleukin-1β (IL-1β), mucin5B (MUC5B), mucin7 (MUC7), and melatonin were detected in stimulated saliva. Correlation analysis of these factors with the microbiota showed positive correlations with the genera Lachnospiraceae, Eikenella, Saccharibacteria_(TM7), Streptococcus, Prevotella, and Haemophilus. Conclusions: AC chewing gum has a beneficial effect on the composition of the oral microbiome, and toothbrush replacement leads to changes in the levels of salivary pro-inflammatory cytokines.

Abstract

The ESA Gaia mission provides a unique time-domain survey for more than one billion sources brighter than G=20.7 mag. Gaia offers the unprecedented opportunity to study variability phenomena in the Universe thanks to multi-epoch G-magnitude photometry in addition to astrometry, blue and red spectro-photometry, and spectroscopy. Within the Gaia Consortium, Coordination Unit 7 has the responsibility to detect variable objects, classify them, derive characteristic parameters for specific variability classes, and provide global descriptions of variable phenomena. We describe the variability processing and analysis that we plan to apply to the successive data releases, and we present its application to the G-band photometry results of the first 14 months of Gaia operations that comprises 28 days of Ecliptic Pole Scanning Law and 13 months of Nominal Scanning Law. Out of the 694 million, all-sky, sources that have calibrated G-band photometry in this first stage of the mission, about 2.3 million sources that have at least 20 observations are located within 38 degrees from the South Ecliptic Pole. We detect about 14% of them as variable candidates, among which the automated classification identified 9347 Cepheid and RR Lyrae candidates. Additional visual inspections and selection criteria led to the publication of 3194 Cepheid and RR Lyrae stars, described in Clementini et al. (2016). Under the restrictive conditions for DR1, the completenesses of Cepheids and RR Lyrae stars are estimated at 67% and 58%, respectively, numbers that will significantly increase with subsequent Gaia data releases. Data processing within the Gaia Consortium is iterative, the quality of the data and the results being improved at each iteration. Abstract

Ureido-heterocycles exhibiting different triple- and quadruple H-bonding patterns are useful building blocks in the construction of supramolecular polymers, self-healing materials, stimuli-responsive devices, catalysts and sensors. The heterocyclic group may provide hydrogen bond donor/acceptor sites to supplement those in the urea core, and they can also bind metals and can be modified by pH, redox reactions or irradiation. In the present review, the main structural features of these derivatives are discussed, including the effect of tautomerization and conformational isomerism on self-assembly and complex formation. Some examples of their use as building blocks in different molecular architectures and supramolecular polymers, with special emphasis on biomedical applications, are presented. The role of the heterocyclic functionality in catalytic and sensory applications is also outlined.

96. Factors Affecting Attendance of and Attitudes towards Artistic Events among Primary School Children

Abstract

In this study, the aim is twofold; on the one hand, we investigate extracurricular artistic activities, and on the other hand, regularity of visits to ten different artistic and cultural events, such as theatres, puppet theatres, dance and folk-dance performances, exhibitions, and musical performances for children, among 9–10 year old primary school children (N = 974). We are looking for an answer to the question of whether there is a connection between active artistic activity and the attitude towards cultural and artistic events. Additionally, we plan to identify differences in the family backgrounds of individuals who participate in artistic activities compared to those who do not. Furthermore, we will examine the influence of social, cultural, and economic capital on children’s cultural consumption and related attitudes. We tested our hypotheses using SPSS; the methods included logistic regression and factor analysis. In addition to highlighting the role of school in equalising cultural capital, the results confirm the powerful effect of generational transfer. When parents have actively engaged in artistic activities, they tend to offer enhanced support for their children’s artistic pursuits. Those who engage in certain types of extracurricular artistic activities during their childhood are more likely to exhibit more positive attitudes towards cultural events as adult consumers. However, parents’ high educational attainment and favourable financial circumstances are not the sole determinants of openness to art.
