In this work, an evaluation of an intense biomass burning event observed over Ny-angstrom lesund (Spitsbergen, European Arctic) in July 2015 is presented. Data from the multi-wavelengths Raman-lidar KARL, a sun photometer and radiosonde measurements are used to derive some microphysical properties of the biomass burning aerosol as size distribution, refractive index and single scattering albedo at different relative humidities. Predominantly particles in the accumulation mode have been found with a bi-modal distribution and dominance of the smaller mode. Above 80% relative humidity, hygroscopic growth in terms of an increase of particle diameter and a slight decrease of the index of refraction (real and imaginary part) has been found. Values of the single scattering albedo around 0.9 both at 355nm and 532nm indicate some absorption by the aerosol. Values of the lidar ratio are around 26sr for 355nm and around 50sr for 532nm, almost independent of the relative humidity. Further, data from the photometer and surface radiation values from the local baseline surface radiation network (BSRN) have been applied to derive the radiative impact of the biomass burning event purely from observational data by comparison with a clear background day. We found a strong cooling for the visible radiation and a slight warming in the infra-red. The net aerosol forcing, derived by comparison with a clear background day purely from observational data, obtained a value of -95 W/m(2) per unit AOD500.

BACKGROUND: Platinum-based drugs such as Cisplatin are commonly employed for cancer treatment. Despite an initial therapeutic response, Cisplatin treatment often results in the development of chemoresistance. To identify novel approaches to overcome Cisplatin resistance, we tested Cisplatin in combination with K+ channel modulators on colorectal cancer (CRC) cells. METHODS: The functional expression of Ca2+-activated (KCa3.1, also known as KCNN4) and voltage-dependent (Kv11.1, also known as KCNH2 or hERG1) K+ channels was determined in two CRC cell lines (HCT-116 and HCT-8) by molecular and electrophysiological techniques. Cisplatin and several K+ channel modulators were tested in vitro for their action on K+ currents, cell vitality, apoptosis, cell cycle, proliferation, intracellular signalling and Platinum uptake. These effects were also analysed in a mouse model mimicking Cisplatin resistance. RESULTS: Cisplatin-resistant CRC cells expressed higher levels of KCa3.1 and Kv11.1 channels, compared with Cisplatin-sensitive CRC cells. In resistant cells, KCa3.1 activators (SKA-31) and Kv11.1 inhibitors (E4031) had a synergistic action with Cisplatin in triggering apoptosis and inhibiting proliferation. The effect was maximal when KCa3.1 activation and Kv11.1 inhibition were combined. In fact, similar results were produced by Riluzole, which is able to both activate KCa3.1 and inhibit Kv11.1. Cisplatin uptake into resistant cells depended on KCa3.1 channel activity, as it was potentiated by KCa3.1 activators. Kv11.1 blockade led to increased KCa3.1 expression and thereby stimulated Cisplatin uptake. Finally, the combined administration of a KCa3.1 activator and a Kv11.1 inhibitor also overcame Cisplatin resistance in vivo. CONCLUSIONS: As Riluzole, an activator of KCa3.1 and inhibitor of Kv11.1 channels, is in clinical use, our results suggest that this compound may be useful in the clinic to improve Cisplatin efficacy and overcome Cisplatin resistance in CRC

Size-segregated particle samples were collected in the Arctic (Ny-Ålesund, Svalbard) in April 2011 both at ground level and in the free atmosphere exploiting a tethered balloon equipped also with an optical particle counter (OPC) and meteorological sensors. Individual particle properties were investigated by scanning electron microscopy coupled with energy dispersive microanalysis (SEM-EDS). Results of the SEM-EDS were integrated with particle size and optical measurements of the aerosols properties at ground level and along the vertical profiles. Detailed analysis of two case studies reveals significant differences in composition despite the similar structure (layering) and the comparable texture (grain size distribution) of particles in the air column. Differences in the mineral chemistry of samples point at both local (plutonic/metamorphic complexes in Svalbard) and remote (basic/ultrabasic magmatic complexes in Greenland and/or Iceland) geological source regions for dust. Differences in the particle size and shape are put into relationship with the mechanism of particle formation, that is, primary (well sorted, small) or secondary (idiomorphic, fine to coarse grained) origin for chloride and sulfate crystals and transport/settling for soil (silicate, carbonate and metal oxide) particles. The influence of size, shape, and mixing state of particles on ice nucleation and radiative properties is also discussed.

Atmospheric new particle formation (NPF) and growth significantly influences climate by supplying new seeds for cloud condensation and brightness. Currently, there is a lack of understanding of whether and how marine biota emissions affect aerosol-cloud-climate interactions in the Arctic. Here, the aerosol population was categorised via cluster analysis of aerosol size distributions taken at Mt Zeppelin (Svalbard) during a 11 year record. The daily temporal occurrence of NPF events likely caused by nucleation in the polar marine boundary layer was quantified annually as 18%, with a peak of 51% during summer months. Air mass trajectory analysis and atmospheric nitrogen and sulphur tracers link these frequent nucleation events to biogenic precursors released by open water and melting sea ice regions. The occurrence of such events across a full decade was anti-correlated with sea ice extent. New particles originating from open water and open pack ice increased the cloud condensation nuclei concentration background by at least ca. 20%, supporting a marine biosphere-climate link through sea ice melt and low altitude clouds that may have contributed to accelerate Arctic warming. Our results prompt a better representation of biogenic aerosol sources in Arctic climate models.

Ice-core records of climate from Greenland and Antarctica show asynchronous temperature variations on millennial timescales during the last glacial period. The warming during the transition from glacial to interglacial conditions was markedly different between the hemispheres, a pattern attributed to the thermal bipolar see-saw. However, a record from the Ross Sea sector of East Antarctica has been suggested to be synchronous with Northern Hemisphere climate change. Here we present a temperature record from the Talos Dome ice core, also located in the Ross Sea sector. We compare our record with ice-core analyses from Greenland, based on methane synchronization, and find clearly asynchronous temperature changes during the deglaciation. We also find distinct differences in Antarctic records, pointing to differences in the climate evolution of the Indo-Pacific and Atlantic sectors of Antarctica. In the Atlantic sector, we find that the rate of warming slowed between 16,000 and 14,500 years ago, parallel with the deceleration of the rise in atmospheric carbon dioxide concentrations and with a slight cooling over Greenland. In addition, our chronology supports the hypothesis that the cooling of the Antarctic Cold Reversal is synchronous with the Bølling-Allerød warming in the northern hemisphere 14,700 years ago. © 2011 Macmillan Publishers Limited. In this paper impact of intensive biomass burning (BB) in North America in July 2015, on aerosol optical and microphysical properties measured in the European Arctic is discussed. This study was made within the framework of the Impact of Absorbing Aerosols on radiating forcing in the European Arctic (iAREA) project. During the BB event aerosol optical depth (AOD) at 500 nm exceeded 1.2 in Spitsbergen and 0.7 in Andenes (Norway). Ångström Exponent (AE) exceeded 1.4 while the absorbing Ångström Exponent (AAE) varied between 1 and 1.25. BB aerosols were observed in humid atmosphere with a total water vapor column between 2 and 2.5 cm. In such conditions aerosols are activated and may produce clouds at different altitudes. Vertical structure of aerosol plumes over Svalbard, obtained from ceilometers and lidars, shows variability of range corrected signal between surface and middle and upper troposphere. Aerosol backscattering coefficients show values up to 10 -5m-1sr-1at 532 nm. Aerosol surface observations indicate chemical composition typical for biomass burning particles and very high single scattering properties. Scattering and absorption coefficients at 530 nm were up to 130 and 15 Mm-1, respectively. Single scattering albedo at the surface varied from 0.9 to 0.94. The averaged values over the entire atmospheric column, ranged from 0.93 to 0.99. Preliminary statistics of model and sunphotometer data as well as previous studies indicate that this event, in the Arctic region, must be considered extreme (such AOD was not observed in Svalbard since 2005) with a significant impact on energy budget.

Daily PM10 aerosol samples were collected at the Gruvebadet observatory, Ny-Ålesund (Svalbard Islands), during the spring-summer 2014 Italian Arctic Campaign. A total of 136 samples were analysed for ion (inorganic anions and cations, selected organic anions) composition aiming to evaluate the seasonal pattern of sulfate, as a key component of the Arctic haze. Ionic balances indicated a strong sulfate seasonality with mean spring concentration about 1.5 times higher than that measured in summer. The spring and summer aerosol was almost neutral, indicating that ammonia was the major neutralizing agent for atmospheric acidic species. The linear regression between sulfate from potential acidic sources (non-sea salt sulfate and non-crustal sulfate) and ammonium indicated that the mean sulfate/ammonium ratio was intermediate between semi-(NH4HSO4) and complete ((NH4)2SO4) neutralization. Using sea-salt sodium as sea-spray marker, non-sea-salt calcium as crustal marker and methanesulfonic acid as biogenic marker, a detailed source apportionment for sulfate was carried out. The anthropogenic input (calculated as the differences between total sulfate and the sum of sea-salt, crustal and biogenic contributes) was found to be the most relevant contribution to the sulfate budget in the Ny-Ålesund aerosol in summer and, especially, in spring. In this last season, crustal, sea-salt, biogenic and anthropogenic sources accounted for 3.3, 12.0, 11.5 and 74.8 %, respectively.

A Flow Injection Analysis (FIA) spectrofluorimetric method for calcium determination in ice cores was optimised in order to achieve better analytical performances which would make it suitable for reliable calcium measurements at ppt level. The method here optimised is based on the formation of a fluorescent compound between Ca and Quin-2 in buffered environment. A careful evaluation of operative parameters (reagent concentration, buffer composition and concentration, pH), influence of interfering species possibly present in real samples and potential favourable effect of surfactant addition was carried out. The obtained detection limit is around 15 ppt, which is one order of magnitude lower than the most sensitive Flow Analysis method for Ca determination currently available in literature and reproducibility is better than 4% for Ca concentrations of 0.2 ppb. The method was validated through measurements performed in parallel with Ion Chromatography on 200 samples from an alpine ice core (Lys Glacier) revealing an excellent fit between the two chemical series. Two Holocene ice core records from East Antarctica (Vostok and EPICA-Dome C) were analysed for dust concentration and size distribution at a temporal resolution of 1 sample per similar to 50 years. A series of volcanic markers randomly distributed over the common part of the ice cores (from 9.8 to 3.5 kyear BP) ensures accurate relative dating (+/-33 years). Dust-size records from the two sites display oscillations structured in cycles with sub-millennial and secular scale frequencies that are apparently asynchronous. The power spectra of the composite sum (1) of the two dust-size records display spectral energy mostly for 150- to 500-year periodicities. On the other hand, the 200-year band is common to both records and the 200 year components of the two sites are out-of-phase (100-year lead or lag) over similar to 5.5 kyear, a phenomenon also reflected by a significant ( > 99 % conf. lev.) band in the power spectra of the composite difference (A) of the two size records. During long-range transport, mineral dust originating from the Southern Hemisphere continents is graded to a variable extent depending on the altitude and duration of atmospheric transport. Relatively coarse dust is associated with air mass penetration from the middle-lower troposphere and conversely relatively fine dust with upper troposphere air masses or the influence of subsidence over the Antarctic plateau, a hypothesis already proposed for the changes that occurred during the Last Glacial Maximum to Holocene transition (Delmonte et al. 2004b). Moreover, we assume that the overall fluctuation of air mass advection over Antarctica depends on the meridional pressure gradient with respect to low latitudes, i.e. the Antarctic Oscillation (AAO). We therefore suggest a regional variability in atmospheric circulation over East Antarctica.
