Tomato plants were subjected to three fertilisation treatments (M: mineral fertiliser; DMPP: mineral fertiliser + 3,4- dimethylpyrazole phosphate; OM: NKP + organic animal manure) in combination with two water regimes (100% and 50% evapotranspiration). Plant biomass, fruit production, nitrogen use efficiency (NUE) and N uptake, maximal PSII photochemical efficiency, Fv/Fm and cumulative soil N2O emission were determined. Well-watered OM plants showed higher values of biomass, fruit production, NUE and N uptake than M and DMPP plants; cumulative N2O fluxes were lower in DMPP plots than in M and OM plots. The reduced water supply determined a drop in crop biomass, fruit production, NUE and N uptake, and cumulative N2O fluxes in M and OM treatments that were higher in OM plots, whereas it determined a significant rise in cumulative N2O fluxes in DMPP plots that was lower in absolute term compared to M and OM plots recorded under well-water irrigation. It can be concluded that DMPP added-fertiliser has a good performance in semiarid environment resulting a better nitrogen source compared to conventional and organo-mineral fertilisers under reduced water supply, able to preserve crop yield and to determine soil N2O emissions (as expressed in CO2 eq) not dangerous for global environment. Background: The thymidine phosphorylase (TP) enzyme is expressed in higher levels in cancer tissue when compared with normal tissue. It is involved in the intratumoral activation of widely prescribed pyrimidine-derived antimetabolites such as 5'-deoxy-5-fluorouridine and capecitabine (Xeloda®). The purpose of this study was to determine the clinical correlation between TP expression in tumor tissue and the clinical outcome of capecitabine-based therapy in patients with locally advanced (stage III) or metastatic breast cancer (stage IV). Methods: The following variables were analyzed as potential determinants of benefit from a capecitabine-based therapy: TP expression, estrogen receptor (ER) and progesterone receptor (PR) status, human epidermal growth factor receptor-2 status, and Ki67 status. This was accomplished by immunohistochemical analysis of paraffin-embedded cancer tissues from 18 patients with breast cancer treated with at least one cycle of capecitabine. Clinical outcome was measured as time to progression. Results: TP staining intensities in both the invasive and in situ components in patients with lobular and ductal carcinomas were reported. Higher levels of TP in the invasive component were expressed in ER-negative tumors when compared with ER-positive tumors (P<0.05). The ER-positive group expressing lower levels of TP had a median time to progression of 13 months compared with the ER-negative group expressing higher levels of TP which had a median time to progression of 7.5 months (P=0.14). Conclusion: Patients with ER-positive tumors expressing lower levels of TP exhibit a longer time to progression when compared with patients with ER-negative tumors. Using irrigation water with a salinity level of 0.8, 2, and 5 g L-1, the impact of saline water irrigation on the yield and quality of 2 Chinese Cucurbit species, melon (Cucumis melo cv. Huanghe) and watermelon (Citrullus lanatus. convar megulaspemus) were studied in Minqin Oasis, northwestern China. Our results show that melon yields decreased as water salinity increased, just as concentrations of glutamic acid content rose, although the concentration of most amino acids did not differ significantly. In contrast, watermelon yields decreased significantly as water salinity rose, and fruit number, fruit firmness, fruit crude protein, and essential amino acid (EAA) levels of the watermelon showed a significant increase as water salinity rose. Salt stress on both Cucurbit species resulted in an increase in total soluble solids (TSS) and Na+ concentrations, while Ca2+ and Cl- concentrations were not affected significantly. In conclusion, our results showed that, although melon and watermelon yields were restrained by saline water irrigation, fruit quality was not influenced, such as for the fruit's amino acids. The fact that saline water irrigation is feasible for melon and watermelon species shows that saline water is a potential irrigation water resource in arid areas of northwest China. This study combines the use of corn starch and Tetradesmus obliquus microalgae for the production of antioxidant starch films as flexible packaging material. Starch was plasticized with glycerol and blended with 1 w% polyallylamine chosen as an agent to modify the film physical properties. The addition of polyallylamine improved film water stability and water vapor transmission rate as well as mechanical stiffness and tenacity. The dried Tetradesmus obliquus microalgae, which showed an EC50 value of 2.8 mg/mg DPPH (2.2-Diphenyl-1-picrylhydrazyl radical), was then used as antioxidant filler. The addition of microalgae provided the films with good antioxidant activity, which increased with microalgae content increasing. To our knowledge, this is the first study reporting the development of sustainable bioactive packaging films composed of almost 100% starch, and follows the European union's goals on plastics strategy concerning the promotion of bio-based, compostable plastics and the setting up of approaches to prevent food waste with a simple plastic packaging. Fertilizer inputs, crop yields, the composition of technological operations and intensity of treatment with different types of pesticides in both basic approaches were evaluated. A comprehensive comparison of impacts showed that all crops, except sugar beet, achieved better economic and emission parameters of production based on the evaluation of GHG production by using reduced tillage compared to ploughing. The total reduction of GHG emissions based on CO2eq on average of all crops per ton as a result of the technological processes was 6% using reduced tillage. The most significant CO2eq reductions were achieved for rye and oat (13%), and spring barley (8%). The reduction of crop yields ranges from about 1% (spring barley) to 4% (grain maize). Cost reduction per tone was in the range of 14% (rye) to 2% (silage maize). The energy gain was at reduced tillage improved at poppy (8%), rape (4%), oat (3%), rye (3%) and spring and winter barley (2%). From the evaluation of the number of chemical protections, a lower number of total protections was found at the no-till system for most crops. In most cases, there was no difference between ploughing and reduced tillage. There was an increase in specific nitrogen consumption per tonne of production in marginal areas, reduced tillage led to an increase in soil compaction. [ABSTRACT FROM AUTHOR]

Peat substrates are well known to become hydrophobic during desiccation, thus degrading their water retention properties. Synthetic wetting agents are commonly incorporated to limit the risk of hydrophobicity, but substrates companies are searching for more sustainable alternatives. To that end, the effect of wood fiber addition in peat-based mixes was measured using contact angles and hydration curves. The study was carried out on two raw materials (white milled peat and wood fiber) and binary mixes. The results showed a shift from hydrophilic to more hydrophobic character with a decrease in the ability to rewet of peat-based substrates in relation to the intensity of drying, whereas wood fiber remained hydrophilic. Increasing wood fiber content in peat-based mixes improved the rehydration efficiency, but with a lower intensity of that measured with synthetic wetting agent addition. Our results highlighted the hydrophilic nature of wood fiber and demonstrated an additional benefit of wood fiber use in peat-based growing media. [ABSTRACT FROM AUTHOR]

Management zones (MZs) are delineated areas within an agricultural field with relatively homogenous soil properties, and therefore similar crop fertility requirements. Consequently, such MZs can often be used for site-specific management of crop production inputs. This study evaluated the effectiveness of four classification methods for delineating MZs in an 8-ha commercial potato field located in Prince Edward Island, Canada. The apparent electrical conductivity (ECa) at two depths from a commercial Veris sensor were used to delineate MZs using three classification methods without spatial constraints (i.e., fuzzy k-means, ISODATA and hierarchical) and one with spatial constraints (i.e., spatial segmentation method). Soil samples (0.0–0.15 m depth) from 104 sampling points was used to measure soil physical and chemical properties and their spatial variation in the field were used as reference data to evaluate four delineation methods. Significant Pearson correlations between ECa and soil properties were obtained (0.22 < r < 0.85). The variance reduction indicated that two to three MZs were optimal for representing the field's spatial variability of soil properties. For two MZs, most soil physical and chemical properties differed significantly between MZs for all four delineation methods. For three MZs, there was greater discrimination among MZs for several soil properties for the spatial segmentation-based method compared with other delineation methods. Moreover, consideration of the spatial coordinates of the data improved the delineation of MZs and thereby increased the number of significant differences among MZs for individual soil properties. Agricultural sites contribute extensively to atmospheric emissions of climate-altering gases such as nitrous oxide. Several strategies have been considered to mitigate the impact of agriculture on climate, among these the utilization of fertilizers added with nitrification inhibitors such as DMPP (3,4-dimethylpyrazole phosphate) may represent a suitable solution. DMPP inhibits the growth and activity of ammonia-oxidizing microorganisms, particularly the ammonia-oxidizing bacteria, which are involved in N2O production. At present, little information is available on the effects of DMPP on the catabolic diversity of soil microbial community. In this study, the N2O emission by soil was performed by using the static chamber technique. The biological determinations of the microbial biomass carbon and the catabolic profile were assessed by measuring the substrate-induced respiration during the entire growing season of a potato crop under two nitrogen treatments: fertilization with and without DMPP. Our results did not show a clear mitigation of N2O emission by DMPP, even if a tendency to lower N2O fluxes in DMPP plots occurred when soil temperatures were lower than 20 °C. Conversely, DMPP deeply affected the microbial biomass and the catabolism of soil microorganisms, exerting a negative effect when it accumulated in excessive doses in the soil, limiting the growth and the capacity of soil microorganism communities to use different substrates. Diabetic Foot Syndrome is a complex and challenging clinical condition associated with high risk of mortality and lower limb amputation. The distal lesions represent the epiphenomenon of this syndrome and request a multidisciplinary care and an appropriate therapeutic path to ensure their healing. This case report describes the management of burns in a patient with type 2 diabetes mellitus, end stage renal disease and Diabetic Foot Syndrome. The lesions were treated with autologous epidermal skin graft until healing. The Difoprev system constituted by a sock loaded with nanocapsules containing a hydrating agent in the diabetic foot is tested. A total of 30 neuropathic outpatients with foot anhydrosis were randomized into group A, treated with the application of the sock with the nanocapsules, and group B wearing only the socks without the nanocapsules. Patients were blindly evaluated with a clinical score, hygrometry, transepidermal water loss, skin temperature, and skin hardness at baseline and after 6 weeks. No difference between the groups emerged at baseline. Although group B showed no changes at the end of the treatment, group A significantly (P< .05) improved in all the parameters evaluated. No adverse events were recorded in both groups during the study. The use of hydrating agents carried by nanocapsules-loaded socks is safe and effective for the neuropathic diabetic foot.

RNA interference (RNAi) has emerged recently as an efficient mechanism for specific gene silencing. Short double-stranded small interfering RNAs (siRNAs) are now widely used for cellular or drug target validation; however, their use for silencing clinically relevant genes in a therapeutic setting remains problematic because of their unfavourable metabolic stability and pharmacokinetic properties. To address some of these concerns, we have investigated the properties of siRNA modified with 2'-deoxy-2'-fluoro-beta-d-arabinonucleotide units (araF-N or FANA units). Here we provide evidence that these modified siRNAs are compatible with the intracellular RNAi machinery and can mediate specific degradation of target mRNA. We also show that the incorporation of FANA units into siRNA duplexes increases activity and substantially enhances serum stability of the siRNA. A fully modified sense 2'-deoxy-2'-fluoro-beta-D-arabinonucleic acid (FANA) strand when hybridized to an antisense RNA (i.e. FANA/RNA hybrid) was shown to be 4-fold more potent and had longer half-life in serum (approximately 6 h) compared with an unmodified siRNA (<15 min). While incorporation of FANA units is well tolerated throughout the sense strand of the duplex, modifications can also be included at the 5' or 3' ends of the antisense strand, in striking contrast to other commonly used chemical modifications. Taken together, these results offer preliminary evidence of the therapeutic potential of FANA modified siRNAs.
