Abstract

Schild analysis of the determination of relative potencies of muscarinic receptor antagonists helps identify the subtypes of muscarinic receptors mediating the carbachol-induced stimulation of phosphoinositide (PI) hydrolysis and inhibition of adenosine 3',5'-cyclic monophosphate (cAMP) formation in guinea pig gallbladder smooth muscle. Muscarinic M3 receptors stimulate PI hydrolysis and inhibit cAMP accumulation, while cholecystokinin receptors are associated only with PI hydrolysis.

Abstract

Are human beings genetically predisposed to prosocial and cooperative behavior? Are human beings inherently virtuous or evil? Is love self-sacrifice? This book was written to answer these questions from the view point of the neuroendocrinology regulating oxytocin (OXT) system in our brain. OXT is synthesized mainly in a section of brain called “hypothalamus”. While OXT plays a crucial role in childbirth by facilitating uterus contraction and milk ejection, OXT is called as “love hormone” because it also plays a key role in regulating affectionate behaviors. OXT is linked to health-promoting cardiovascular, analgesic, and anti-stress effects. It is hightly recommened to consider how to upregulate the internal OXT system, rather than depending on OXT administered from outside the body. Manual therapies that involve sensory nerve stimulation of the skin and muscle increase hypothalamic OXT expression, and help alleviate symptoms associated with stress. Social interaction also upregulates OXT expression by which affection, empathy, friendship and love among humans can be transmitted across generations. Once this interaction is persistently maintained, the positive loop of love won’t be terminated.

Abstract

Complementary and alternative medicine (CAM) is a diverse group of health care systems, practices, and products that are not presently considered a part of conventional Western medicine (CWM). CAM mainly includes mind-body therapies (meditation, Yoga, Tai-chi, Qi-gong, music therapy, etc.) and manual therapies (chiropractic, massage, acupuncture, touch therapy, etc.). The beneficial effects of CAM are mainly due to a reduction of stress responses and pain pathways. Mind-body therapies promote these effects by stimulating brain activity throughout its network (top-down pathway). Manual therapies promote these effects through the stimulation of peripheral sensory nerves (bottom-up pathway). Both pathways activate various neuropeptides in the brain, such as opioid and oxytocin, which mediate anti-stress and analgesic effects. Public interest in CAM has dramatically increased over the past decade, and a great deal of research is backing up that trend with evidence supporting the benefits of CAM therapies. A national health survey in 2007 revealed that more than $34 billion was spent on CAM annually in the US. As CAM therapies are proven to be safe and effective, CAM is destined to claim an important role in conventional health care.

Am J Physiol Gastrointest Liver Physiol 279: G740-G747, 2000.--The role of nitric oxide (NO) and ATP in the regulation of nonadrenergic, noncholinergic (NANC) inhibitory transmission in the pylorus remains unclear. In the presence of atropine and guanethidine, electric field stimulation induced NANC relaxations in a frequency-dependent manner (1-20 Hz) in the rat pylorus. NANC relaxations were significantly inhibited by [N.sup.G]-nitro-L-arginine methyl ester (L-NAME; [10.sup.-4] M). [P.sub.2X] purinoceptor antagonist pyridoxal phosphate-6-azophenyl-2',4'-disulfonic acid (PPADS; 3 x [10.sup.-5] M) and [P.sub.2Y] purinoceptor antagonist reactive blue 2 (2 x [10.sup.-5] M) had no effect on NANC relaxations. However, the combined administration of L-NAME and PPADS, but not reactive blue 2, evoked greater inhibitory effects on NANC relaxation than that evoked by L-NAME alone. [Alpha]-Chymotrypsin and vasoactive intestinal polypeptide antagonist did not affect NANC relaxations. ATP ([10.sup.-5]-[10.sup.-3] M) and [P.sub.2X] purinoceptor agonist [Alpha], [Beta]-methyleneadenosine 5'-triphosphate ([10.sup.-7]-[10.sup.-5] M), but not [P.sub.2Y] purinoceptor agonist 2-methylthioadenosine 5'-triphosphate ([10.sup.-7]-[10.sup.-5] M), induced muscle relaxations in a dose-dependent manner, and relaxations were significantly reduced by PPADS and unaffected by TTX. These studies suggest that NO and ATP act in concert to mediate NANC relaxation of the rat pylorus. ATP-induced relaxation appears to be mediated by [P.sub.2X] purinoceptors located on smooth muscle cells. [P.sub.2X] purinoceptors; [P.sub.2Y] purinoceptors
