Abstract

Vegetative propagation of economically important woody, horticultural and agricultural species rely on an efficient adventitious root (AR) formation. The formation of ARs is a complex genetic trait regulated by the interaction of environmental and endogenous factors among which the phytohormone auxin plays an essential role. This article summarizes the current knowledge related to the intricate network through which auxin controls adventitious rooting. How auxin and recently identified auxin-related compounds affect AR formation in different plant species is discussed. Particular attention is addressed to illustrate how auxin has a central role in the hormone cross-talk leading to AR development. In parallel, we describe the molecular players involved in the control of auxin homeostasis, transport and signaling, for a better understanding of the auxin action during adventitious rooting.

Abstract

Premise of research. Well-preserved Triassic plant fossils from Antarctica yield insights into the physiology of plant growth under the seasonal light regimes of warm polar forests, a type of ecosystem without any modern analogue. Among the many well-known Triassic plants from Antarctica is the enigmatic Petriellaea triangulata, a dispersed seedpod structure that is considered a possible homologue of the angiosperm carpel. However, the morphology and physiology of the plants that produced these seedpods have so far remained largely elusive. Methodology. Here, we describe petriellalean stems and leaves in compression and anatomical preservation that enable a detailed interpretation of the physiology and ecology of these plants. Pivotal results. Our results indicate that the Petriellales were diminutive, evergreen, shade-adapted perennial shrubs that colonized the understory of the deciduous forest biome of polar Gondwana. This life form is very unlike that of any other known seed-plant group of that time. By contrast, it fi ts remarkably well into the "dark and disturbed" niche that some authors considered to have sheltered the rise of the fl owering plants some 100 Myr later. Conclusions. The hitherto enigmatic Petriellales are now among the most comprehensively reconstructed groups of extinct seed plants and emerge as promising candidates for elucidating the mysterious origin of the angiosperms. Abstract

Summary Auxin is a major regulator of adventitious rooting and, to better understand its role, we identified suppressor mutants of superroot2-1. This provides new resources for the discovery of genetic players involved in auxin signalling or auxin crosstalk with other hormones., The plant hormone auxin plays a central role in adventitious rooting and is routinely used with many economically important, vegetatively propagated plant species to promote adventitious root initiation and development on cuttings. Nevertheless the molecular mechanisms through which it acts are only starting to emerge. The Arabidopsis superroot2-1 (sur2-1) mutant overproduces auxin and, as a consequence, develops excessive adventitious roots in the hypocotyl. In order to increase the knowledge of adventitious rooting and of auxin signalling pathways and crosstalk, this study performed a screen for suppressors of superroot2-1 phenotype. These suppressors provide a new resource for discovery of genetic players involved in auxin signalling pathways or at the crosstalk of auxin and other hormones or environmental signals. This study reports the identification and characterization of 26 sur2-1 suppressor mutants, several of which were identified as mutations in candidate genes involved in either auxin biosynthesis or signalling. In addition to confirming the role of auxin as a central regulator of adventitious rooting, superroot2 suppressors indicated possible crosstalk with ethylene signalling in this process.

Abstract

International audience; Plant volatiles are important cues for many herbivorous insects when choosing a suitable host plant and finding a mating partner. An appropriate behavioural response to sensory cues from plants and other insects is crucial for survival and fitness. As the natural environment can show both large spatial and temporal variability, herbivores may need to show behavioural plasticity to the available cues. By using earlier experiences, insects can adapt to local variation of resources. Experience is well known to affect sensory-guided behaviour in parasitoids and social insects, but there is also increasing evidence that it influences host plant choice and the probability of finding a mating partner in herbivorous insects. In this review, we will focus upon behavioural changes in holometabolous insect herbivores during host plant choice and localization of mating partners, modulated by experience to sensory cues. The experience can be acquired during both the larval and the adult stage and can influence later responses to plant volatiles and other sensory cues not only within the developmental stage but also after metamorphosis. Furthermore, we will address the neurophysiological mechanisms underlying the experience-dependent behavioural adaptations and discuss ecological and evolutionary aspects of insect behavioural plasticity based upon experience

Abstract

When D-branes are inserted in flux backgrounds of opposite charge, the resulting solution has a certain singularity in the fluxes. Recently it has been argued, using numerical solutions, that for anti-D3 branes in the Klebanov-Strassler background, these singularities cannot be cloaked by a horizon, which strongly suggests they are not physical. In this paper we provide an analytic proof that the singularity of all codimension-three antibrane solutions (such as anti-D6 branes in massive type IIA supergravity or anti-D3 branes smeared on the T3 of R3×T3 with fluxes) cannot be hidden behind a horizon and that the charge of black branes with smooth event horizons must have the same sign as the charge of the flux background. Abstract

Comparison of simulated and reconstructed past climate variability within the last millennium provides an opportunity to aid the understanding and interpretation of palaeoclimate proxy data and to test hypotheses regarding external forcings, feedback mechanisms and internal climate variability under conditions close to those of the present day. Most such comparisons have been made at the Northern Hemispheric scale, of which a selection of recent results is briefly discussed here. Uncertainties in climate and forcing reconstructions, along with the simplified representations of the true climate system represented by climate models, limit our possibility to draw certain conclusions regarding the nature of forced and unforced climate variability. Additionally, hemispheric-scale temperature variations have been comparatively small, wherefore the last millennium is apparently not a particularly useful period for estimating climate sensitivity. Nevertheless, several investigators have concluded that Northern Hemispheric-scale decadal-mean temperatures in the last millennium show a significant influence from natural external forcing, where volcanic forcing is significantly detectable while solar forcing is less robustly detected. The amplitude of centennial-scale variations in solar forcing has been a subject for much debate, but current understanding of solar physics implies that these variations have been small – similar in magnitude to those within recent sunspot cycles – and thus they have not been a main driver of climate in the last millennium. This interpretation is supported by various comparisons between forced climate model simulations and temperature proxy data. Anthropogenic greenhouse gas and aerosol forcing has been detected by the end of Northern Hemispheric temperature reconstructions. Abstract

International audience; Exchange of nuclear magnetization between spin pools, either by chemical exchange or by cross-relaxation or both, has a significant influence on the signal attenuation in stimulated-echo-type pulsed field gradient experiments. Hence, in such cases the obtained molecular self-diffusion coefficients can carry a large systematic error. We propose a modified stimulated echo pulse sequence that contains T2-filters during the z-magnetization store period. We demonstrate, using a common theoretical description for chemical exchange and cross-relaxation, that these filters suppress the effects of exchange on the diffusional decay in that frequent case where one of the participating spin pools is immobile and exhibits a short T2. We demonstrate the performance of this experiment in an agarose/water gel. We posit that this new experiment has advantages over other approaches hitherto used, such as that consisting of measuring separately the magnetization exchange rate, if suitable by Goldman–Shen type experiments, and then correcting for exchange effects within the framework of a two-site exchange model. We also propose experiments based on selective decoupling and applicable in systems with no large T2 difference between the different spin pools.

Abstract

International audience; We have made creep experiments on columnar grained ice and characterised the microstructure and intragranular misorientations over a range of length scales. A FFT full-field model was used to predict the deformation behaviour, using the experimentally characterised microstructure as the starting material. This is the first time this combination of techniques has been used to study the deformation of ice. The microstructure was characterised at the cm scale using an optical technique, the automatic ice texture analyser AITA and at the micron scale using electron backscattered diffraction EBSD. The crystallographic texture and intragranular misorientations were fully characterised by EBSD (3 angles). The deformed microstructure frequently showed straight subgrain boundaries often originating at triple points. These were identified as kink bands, and for the first time we have measured the precise misorientation of the kink bands and deduced the nature of the dislocations responsible for them. These dislocations have a basal edge nature and align in contiguous prismatic planes enabling deformation along the c-axis. In addition, non-uniform grain boundaries and regions of recrystallization were seen. We present coupling between fine scale characterization of intragranular misorientations, from experiments, and prediction of internal stresses that cause it. The model predicts the morphology of the observed local misorientations within the grains, however it over predicts the misorientation values. This is because the annealing and recrystallization mechanisms are not taken into account in the model. Ice is excellent as a model material for measuring, predicting and understanding deformation behaviour for polycrystalline materials. Specifically for ice this knowledge is needed to improve models of ice sheet dynamics that are important for climatic signal interpretation.

Abstract

International audience; Crohn disease (CD) is a multifactorial disease in which an abnormal immune response in the gastrointestinal (GI) tract leads to chronic inflammation. The small intestine, particularly the ileum, of patients with CD is colonized by adherent-invasive E. coil (AIEC) a pathogenic group of E. coil able to adhere to and invade intestinal epithelial cells. As the earliest inflammatory lesions are microscopic erosions of the epithelium lining the Peyer's patches (PPs), we investigated the ability of AIEC bacteria to interact with PPs and the virulence factors involved. We found that AIEC bacteria could interact with mouse and human PPs via long polar fimbriae (LPF). An LPF-negative AIEC mutant was highly impaired in its ability to interact with mouse and human PPs and to translocate across monolayers of M cells, specialized epithelial cells at the surface of PPs. The prevalence of AIEC strains harboring the lpf operon was markedly higher in CD patients compared with controls. In addition, increased numbers of AIEC, but not LPF-deficient AIEC, bacteria were found interacting with PPs from Nod2(-/-) mice compared with WT mice. In conclusion, we have identified LPF as a key factor for AIEC to target PPs. This could be the missing link between AIEC colonization and the presence of early lesions in the PPs of CD patients.

Abstract

Lipid vesicles can be connected by membrane nanotubes to build networks with promising bioanalytical properties. Here we characterize electrophoretic transport in such membrane tubes, with a particular eye to how their soft-material nature influences the intratube migration. In the absence of field, the tube radius is 110 +/- 26 nm, and it remains in this range during electrophoresis even though the applied electric field causes a slight decrease in the tube radius (approximately 6-11%). The electrophoretic velocity of the membrane wall (labeled with quantum dots) varies linearly with the field strength. Intratube migration is studied with latex spheres of radii 15, 50, 100, and 250 nm. The largest particle size does not enter the tube at fields strengths lower than 1250 V/m because the energy cost for expanding the tube around the particles is too high. The smaller particles migrate with essentially the same velocity as the membrane at low fields. Above 250 V/cm, the 15 nm particles exhibit an upward deviation from linear behavior and in fact migrate faster than in free solution whereas the 100 nm particles deviate downward. We propose that these nonlinear effects arise because of lipid adsorption to the particles (dominating for 15 nm particles) and a pistonlike compression of the solvent in front of the particles (dominating for 100 nm). As expected from such complexities, existing theories for a sphere migrating in a rigid-wall cylinder cannot explain our velocity results in lipid nanotubes.

Abstract

Introduction : The genus Brachyspira contains well-known enteric pathogens of veterinary significance, suggested agents of colonic disease in humans, and one potentially zoonotic agent. There are recent studies showing that Brachyspira are more widespread in the wildlife community than previously thought. There are no records of this genus in wildlife from the southern Atlantic region and Antarctica. Our aim was therefore, to determine whether intestinal spirochaetes of genus Brachyspira colonise marine and coastal birds in this region. Method : Faecal samples were collected from marine and coastal birds in the southern Atlantic region, including sub-Antarctic islands and Antarctica, in 2002, 2009, and 2012, with the aim to isolate and characterise zoonotic agents. In total, 205 samples from 11 bird species were selectively cultured for intestinal spirochaetes of genus Brachyspira . To identify isolates to species level, they were subjected to phenotyping, species-specific polymerase chain reactions, sequencing of partial 16S rRNA, NADH oxidase ( nox ), and tly A genes, and phylogenetic analysis. Antimicrobial susceptibility tests were performed. Results : Fourteen unique strains were obtained from 10 birds of three species: four snowy sheathbills ( Chionis albus ), three kelp geese ( Chloephaga hybrida subsp. malvinarum ), and three brown skua ( Stercorarius antarcticus subsp. lonnbergi ) sampled on the Falkland Islands, Tierra del Fuego in Argentina, South Georgia, South Shetland Islands, and the Antarctic Peninsula. Five Brachyspira strains were closely related to potentially enteropathogenic Brachyspira sp. of chickens: B . intermedia ( n= 2, from snowy sheathbills), and B . alvinipulli ( n= 3, from a kelp goose and two snowy sheathbills). Three strains from kelp geese were most similar to the presumed non-pathogenic species ‘ B. pulli ’ and B . murdochii , whereas the remaining six strains could not be attributed to currently known species. No isolates related to human strains were found. None of the tested strains showed decreased susceptibility to tiamulin, valnemulin, doxycycline, tylvalosin, lincomycin, or tylosin. Conclusions : This is the first report of intestinal spirochaetes from this region. Despite limitations of current diagnostic methods, our results, together with earlier studies, show that Brachyspira spp., including potentially pathogenic strains, occur globally among free-living avian hosts, and that this genus encompasses a higher degree of biodiversity than previously recognised. Abstract

Boys’ (under) achievement is frequently on the agenda in the Nordic countries, like in other parts of the Western world. Theoretical explanations for their lower achievement than girls typically emphasise the dissociation between hegemonic young masculinities and school work. In this paper, we focus on boys’ strategies for managing schools conflicting demands, using data from a recently completed Swedish ethnographic study of the discourses of gender and pupil achievement at school in various local settings. The analyses show that boys engage in a strategy of complex parallel positioning to master school demands and peer-group expectations; they appear to distance themselves from swotting yet, at the same time, devote themselves to schoolwork. This dual positioning needs to be accomplished over a short period of time at the beginning of the semester. The analyses point to the critical time sequencing, and reveal what seemed to be a carefully self-monitored process where boys’ academic participation had to appear convincing to teachers but neither too long nor too intense to interfere with their peer-group interactions and positioning. The analyses also show teachers’ appreciation of the boys who manage to position themselves well academically. Abstract

Mn-doped GaAs nanowires were grown in the self-catalytic growth mode on oxidized Si(100) surface by molecular beam epitaxy and characterized by scanning and transmission electron microscopy, Raman scattering, photoluminescence, cathodoluminescence, and electron transport measurements. The transmission electron microscopy studies evidenced the substantial accumulation of Mn inside the catalyzing Ga droplets at the top of the nanowires. Optical and transport measurements revealed that the limit of Mn content for self-catalysed growth of GaAs nanowires corresponds to the doping level, i.e., is much lower than the Mn/Ga flux ratio (about 3%) used during the MBE growth. The resistivity measurements of individual nanowires confirmed that they are conductive, in accordance with the photoluminescence measurements which showed the presence of Mn2+ acceptors located at Ga sites of the GaAs host lattice of the nanowires. An anomalous temperature dependence of the photoluminescence related to excitons was demonstrated for Mn-doped GaAs nanowires., 19 pages, 11 figures

Abstract

Background and Aims Patients with Crohn’s disease [CD] harbour an increased number of adherent-invasive E. coli [AIEC]. The strain LF82, identified in the ileal mucosa of CD patients, has been extensively studied for pathogenic mechanisms. However, understanding of the interaction of LF82 with the intestinal mucosa of CD patients is lacking. Methods Here, we investigated the importance of long polar fimbriae [LPF] type 1 pili and the carcinoembryonic antigen-related cell-adhesion molecule 6 [CEACAM6] for translocation of LF82 in an in vitro model of follicle-associated epithelium [FAE], and in the FAE and villus epithelium [VE] of patients with CD and controls, using Ussing chambers. Results Significantly greater LF82 passage occurred in the FAE model compared with in the VE Caco-2cl1 mono-culture. Moreover, bacterial translocation was inhibited by either LPF disruption or pre-incubation with anti-CEACAM6 antibody. Tissue mounted in Ussing chambers showed significantly higher LF82 passage in FAE from patients with CD compared with control FAE, that was diminished in LF82 lacking LPF and by blocking host CEACAM6. Interestingly, addition of LF82 to the CD FAE tissues significantly increased paracellular permeability [of 51Chromium-EDTA] compared with baseline, and the increase was inhibited by anti-CEACAM6. Immunofluorescence and immunoblots showed higher expression of CEACAM6 in FAE of patients with CD compared with in FAE from controls. Conclusions These data suggest that the FAE of CD patients is a site of vulnerability for invasion by LF82 via a mechanism that requires both bacterial LPF and host CEACAM6. Further, LF82 has the ability to increase paracellular passage through the FAE of patients with CD. These data can help define novel therapeutic targets in CD for the prevention of clinical recurrence.

Abstract

The ASDEX Upgrade (AUG) programme is directed towards physics input to critical elements of the ITER design and the preparation of ITER operation, as well as addressing physics issues for a future DEMO design. Since 2015, AUG is equipped with a new pair of 3-strap ICRF antennas, which were designed for a reduction of tungsten release during ICRF operation. As predicted, a factor two reduction on the ICRF-induced W plasma content could be achieved by the reduction of the sheath voltage at the antenna limiters via the compensation of the image currents of the central and side straps in the antenna frame. There are two main operational scenario lines in AUG. Experiments with low collisionality, which comprise current drive, ELM mitigation/suppression and fast ion physics, are mainly done with freshly boronized walls to reduce the tungsten influx at these high edge temperature conditions. Full ELM suppression and non-inductive operation up to a plasma current of I p = 0.8 MA could be obtained at low plasma density. Plasma exhaust is studied under conditions of high neutral divertor pressure and separatrix electron density, where a fresh boronization is not required. Substantial progress could be achieved for the understanding of the confinement degradation by strong D puffing and the improvement with nitrogen or carbon seeding. Inward/outward shifts of the electron density profile relative to the temperature profile effect the edge stability via the pressure profile changes and lead to improved/decreased pedestal performance. Seeding and D gas puffing are found to effect the core fueling via changes in a region of high density on the high field side (HFSHD). The integration of all above mentioned operational scenarios will be feasible and naturally obtained in a large device where the edge is more opaque for neutrals and higher plasma temperatures provide a lower collisionality. The combination of exhaust control with pellet fueling has been successfully demonstrated. High divertor enrichment values of nitrogen E N ⩾ 10 have been obtained during pellet injection, which is a prerequisite for the simultaneous achievement of good core plasma purity and high divertor radiation levels. Impurity accumulation observed in the all-metal AUG device caused by the strong neoclassical inward transport of tungsten in the pedestal is expected to be relieved by the higher neoclassical temperature screening in larger devices.
