Abstract

Globally there’s an astounding and often underestimated number of ponds (small stagnant water bodies). Ponds, whilst directly providing habitats for a multitude of animals and plants, also contribute to habitat connectivity by enabling organisms to cross the landscape, acting as “blue stepping stones”. The diverse community of insects emerging from such ponds acts as a vector of dietary energy to the surrounding terrestrial environment. By providing highly unsaturated fatty acids, emerging insects constitute a high nutritional food resource for terrestrial (e.g. spiders, lizards) and avian (e.g. bats, birds) consumers. One important dietary resource these insects export is essential fatty acids. These are introduced initially into the local food web and then on into the main terrestrial cycle. However, knowledge about the exact ecological role of ponds, their contribution to beta- and gamma-biodiversity, and dietary energy transfer is limited. Herein, we present preliminary results for the first seasonal sampling (Autumn 2020) of our ongoing “EUROPONDS” project. EUROPONDS aims to fill this gap of knowledge on ponds by analyzing 55 ponds distributed across Europe from Sweden in the north to remote Mediterranean islands in the south, from rural areas to large cities, and including both artificial and natural ponds. EUROPONDS brings together around 80 European early-career limnologists and covers autumn, winter, spring, and summer sampling campaigns through 2020/21. Preliminary results from autumn 2020 indicate differences in emerging insect biomass, trophic status of ponds, and between artificial and natural ponds.

Abstract

Different outcomes may be expected when a particular host is exposed to an infectious disease, depending on its immunity status. For example, because of a previous encounter with the disease, the host may not become infected as consequence of the presence of antibodies. While sometimes it is difficult to establish when this disease avoidance occurred, the recognition of the existence and duration of such immunity is critical for the development of vaccines and accurate epidemiological models. In this paper, we propose a generalized computer model that can examine a wide range of epidemiological data and generate expected patterns of strain-specific infections and reinfection. Abstract

Patient-derived xenografts (PDXs) are resected human tumors engrafted into mice for preclinical studies and therapeutic testing. It has been proposed that the mouse host affects tumor evolution during PDX engraftment and propagation, affecting the accuracy of PDX modeling of human cancer. Here, we exhaustively analyze copy number alterations (CNAs) in 1,451 PDX and matched patient tumor (PT) samples from 509 PDX models. CNA inferences based on DNA sequencing and microarray data displayed substantially higher resolution and dynamic range than gene expression-based inferences, and they also showed strong CNA conservation from PTs through late-passage PDXs. CNA recurrence analysis of 130 colorectal and breast PT/PDX-early/PDX-late trios confirmed high-resolution CNA retention. We observed no significant enrichment of cancer-related genes in PDX-specific CNAs across models. Moreover, CNA differences between patient and PDX tumors were comparable to variations in multiregion samples within patients. Our study demonstrates the lack of systematic copy number evolution driven by the PDX mouse host., Analysis of copy number alterations in 1,451 patient-derived xenografts (PDXs) and matched patient tumor samples shows strong conservation from patient tumors through late-passage PDXs and a lack of systematic copy number evolution driven by the mouse host.

Abstract

Whilst being small and shallow, permanent ponds provide valuable habitats for a multitude of organisms as well as provide habitats for animals like aquatic insects and others, including many rare species. These ponds, which were often neglected in limnological studies, were the core of our joint project, the 3rd freshwater project - EUROPONDS in order to evaluate the often overlooked ecological ecosystem services provided by permanent ponds across Europe. As a part of this project, two Hungarian ponds were surveyed for a full year (2020/21). One of the project aims was to assess the freshwater macroinvertebrate assemblages by dip netting over the four annual seasons. Here, we present preliminary results examining the data on larval Odonata of two Hungarian ponds in relation to the physicochemical variables measured, as well as compare the results to a previous national survey. Altogether the examination of 24 waterbodies (22 from a national survey and two from the EUROPONDS project) was carried out in this study. All of the involved water bodies were clustered using a Ward analysis including the water physicochemical parameters into three groups, and then the dragonfly fauna was investigated within the groups found. Diversity metrics and the presence or absence of different species were also included and the results were compared between the different clusters. Herein, we aimed to assess differences between the three main clusters, identify key factors driving the presence of the studied species and the relationship between the diversity indexes and the physicochemical parameters influencing the Odonata communities in the studied ponds.

Abstract

Whilst being generally small and shallow lentic water bodies, ponds provide valuable habitats for a multitude of groups and contribute to habitat connectivity. Semi-aquatic insects emerging from such ponds act as one of the most important vectors of dietary energy to the adjacent terrestrial environment. By providing essential fatty acids, emerging insects constitute an important nutritional food resource for terrestrial (e.g. spiders, lizards, ants) and aerial (e.g. bats, birds) consumers. Broad-scale studies are needed to explore differences in dietary exports encompassing diverse pond types and different landscapes or regions, with the final aim of drawing general patterns. Herein we present the first results of "EUROPONDS". In this ongoing project we quantify the seasonal and spatial variation in insect emergence. In a total of 55 ponds distributed across Europe we installed emergence traps for one week per season over a full year. This broad scale set-up allows us to quantify differences in biomass export between different geographical regions, between rural and urban systems as well as between natural and artificial ponds. The project brings together the knowledge of 80 early-career limnologists from all over Europe and therefore not only provides important insight in the aquatic-terrestrial linkages of ponds, but also provides an important exchange among the future generation of European limnologists.

Abstract

Background: Opioid agonist therapy is strongly recommended for pregnant persons with opioid use disorder. Buprenorphine may be associated with more favorable neonatal and maternal outcomes than methadone, but existing data are limited.Methods: We conducted a cohort study involving pregnant persons who were enrolled in public insurance programs in the United States during the period from 2000 through 2018 in which we examined outcomes among those who received buprenorphine as compared with those who received methadone. Exposure to the two medications was assessed in early pregnancy (through gestational week 19), late pregnancy (gestational week 20 through the day before delivery), and the 30 days before delivery. Risk ratios for neonatal and maternal outcomes were adjusted for confounders with the use of propensity-score overlap weights.Results: The data source for the study consisted of 2,548,372 pregnancies that ended in live births. In early pregnancy, 10,704 pregnant persons were exposed to buprenorphine and 4387 to methadone. In late pregnancy, 11,272 were exposed to buprenorphine and 5056 to methadone (9976 and 4597, respectively, in the 30 days before delivery). Neonatal abstinence syndrome occurred in 52.0% of the infants who were exposed to buprenorphine in the 30 days before delivery as compared with 69.2% of those exposed to methadone (adjusted relative risk, 0.73; 95% confidence interval [CI], 0.71 to 0.75). Preterm birth occurred in 14.4% of infants exposed to buprenorphine in early pregnancy and in 24.9% of those exposed to methadone (adjusted relative risk, 0.58; 95% CI, 0.53 to 0.62); small size for gestational age in 12.1% and 15.3%, respectively (adjusted relative risk, 0.72; 95% CI, 0.66 to 0.80); and low birth weight in 8.3% and 14.9% (adjusted relative risk, 0.56; 95% CI, 0.50 to 0.63). Delivery by cesarean section occurred in 33.6% of pregnant persons exposed to buprenorphine in early pregnancy and 33.1% of those exposed to methadone (adjusted relative risk, 1.02; 95% CI, 0.97 to 1.08), and severe maternal complications developed in 3.3% and 3.5%, respectively (adjusted relative risk, 0.91; 95% CI, 0.74 to 1.13). Results of exposure in late pregnancy were consistent with results of exposure in early pregnancy.Conclusions: The use of buprenorphine in pregnancy was associated with a lower risk of adverse neonatal outcomes than methadone use; however, the risk of adverse maternal outcomes was similar among persons who received buprenorphine and those who received methadone.
