Abstract

A total of 10 ciprofloxacin-sensitive (ciprofloxacin minimum inhibitory concentration, MIC < 0.5 μ g/ml) and 10 ciprofloxacin-resistant (MIC 16 to 32 μ g/ml) presumptive C. jejuni were further characterized and evaluated for their inhibition by natural orange oil fractions. Partial species identification was performed by using a hippuricase gene-based polymerase chain reaction (PCR) assay. One of the isolates appeared to be atypical and failed to hydrolyze hippurate. Of the ciprofloxacin-resistant C. jejuni isolates tested, six were found to have their quinolone resistance determined by a C → T mutation in codon 86 of gyrA. Both groups of ciprofloxacin-sensitive and -resistant C. jejuni isolates were most susceptible to cold-pressed terpeneless Valencia orange oil (C4) which yielded inhibition zones from 44.0 ± 1.4 to 80 ± 0.0 mm. Less inhibitory responses were recorded for 5-fold concentrated Valencia orange oil (C3) and distilled d-limonene (C7) which exerted similar effects on both ciprofloxacin-sensitive and -resistant C. jejuni isolates. Abstract

Beef trimmings, inoculated with Escherichia coli (EC) and Salmonella typhimurium (ST), were treated with 3% potassium lactate (K-L), 4% sodium metasilicate (NMS), 0.1% acidified sodium chlorite (ASC) or 0.02% peroxyacetic acid (PAA) prior to grinding. The ground beef packages were sampled at 0, 1, 2, 3 and 7 days of simulated retail display. All treatments reduced (P < 0.05) EC, coliforms and aerobic plate counts up to and some in excess of 1 log and ST count ≥ 1.5 logs. Sensory panelists found ground beef from all treatments to generally be similar (P > 0.05) to the control in odor and similar (P > 0.05) in color for the K-L and NMS treatments during initial display. Instrumental color results indicated that K-L, NMS and PAA were similar (P > 0.05) in redness (a* ) to the control. These findings indicated that use of tested antimicrobial agents can reduce microbial numbers with little impact on sensory odor and color characteristics. PRACTICAL APPLICATIONS Although antimicrobial interventions have shown promising results in decontaminating meat products, there are only a limited number of antimicrobial products that can be used without causing adverse effects on quality characteristics of the final product. Consumers often discriminate against discolored meat products, and thus any deleterious effects on color and other quality attributes in decontaminated products will lead to a negative economic impact. The outcome of this research will provide information on selection of antimicrobial agents that will maximize ground beef safety without causing deleterious effects on quality attributes. Application of suitable antimicrobial interventions at the end of a production line will be advantageous to supply ground beef with increased shelf-life and decreased or no risk for illnesses caused by pathogens. 96. Landsat patterns considered in relation to Australian resources surveys

Abstract

In 1956-7 an area in arid central Australia was surveyed by conventional methods (aerial photography and field work) to produce a map at 1:1,000,000 scale showing types of country with characteristic topography, soils and vegetation. Working independently and without field checks or reference to the literature, three workers mapped part of this area according to the patterns they could discern on 5th generation Landsat I imagery. They then compared their patterns with the types of country as shown in the survey map, this by calculating the area common to patterns and types of country, with a threshold mutual involvement of 50%. Their mapping established about half as many patterns and occurrences of patterns as were found on the same area of the survey map. Correlations were achieved by all three workers, broadest for the least experienced and closest for the most experienced interpreter. Their subjective evaluation is that suitably processed early generation imagery could probably be used on its own as a practical means of producing similar maps in arid regions if the normal supporting programme were carried out.

98. Pasture patterns and associated soil water in partially cleared woodland

Abstract

Aerial photographs of parts of the Hunter Valley show many of the trees in the centre of a circle which is paler than the surroundings. The circles have been investigated through the aerial photographs and in the field, and appear to be related to the influence of tree roots upon the grasses in certain soils in regions of fairly low rainfall. No evidence of competition for soil water could be established in the relationship.
